RESUMO

Microplastics (MPs) are present across the global ocean and can be encountered by many species, including zooplankton. Although they fall within the size range of zooplankton prey, there are few studies on MPs ingestion carried out in situ. In this study, we analyzed MPs ingestion during two seasons (rainy and dry) of organisms from 5 taxonomic groups of zooplankton from two bays of the Mexican central Pacific: Manzanillo and Navidad. In total, 2643 individuals were analyzed, and of those 23 individuals contained MPs. The ingestion rate by taxonomic group was 1 MP/36 copepods (0.02), 1 MP/30 decapods-mysis (0.03), 1 MP/29 decapods-megalopa (0.03), and 1 MP/200 fish larvae (0.005). No plastics were found in chaetognaths, amphipods, or decapods-zoea. The average length of the ingested particles was 468.1 ± 113.8 µm, with a minimum of 15.6 and a maximum of 647.6 µm. All MPs >300 µm were fibers, with diameters <50 µm. Fragments were the most abundant MPs (54.2%), followed by fibers (34.2%) and spheres (11.4%). Statistical analyses showed no significant differences (p > 0.05) between the bays or seasons. Using RAMAN spectroscopy, it was possible to identify 6 different types of polymers, with poly (ethylene:propylene) being the most abundant (42.8%). This polymer is commonly used to manufacture plastic bags, ropes and fishing nets. The results confirm that certain zooplankton groups are consuming MPs and suggest that omnivorous species are more likely to ingest MPs, possibly due to their capacity for foraging flexibility and opportunistic feeding strategies. However, the ingestion of MPs cannot be attributed to a single factor; it is necessary to consider variables such as the sampling area, feeding strategy, size, and seasonality to understand the dynamics of MPs ingestion by zooplankton groups.

RESUMO

This study deals with the effect of plasminogen/plasmin on the in vitro maturation (IVM) of bovine cumulus-oocyte complexes (COCs). Exogenous plasminogen activator streptokinase (SK) added to the IVM medium revealed similar values of cumulus expansion and oocyte nuclear maturation compared to controls (standard IVM medium). However, a decrease in both determinations was observed in COCs matured with the supplementation of ɛ-aminocaproic acid (ɛ-ACA), a specific plasmin inhibitor. After in vitro fertilization, no differences were observed in either cleavage or blastocyst rates between SK and control groups; however, ε-ACA treatment caused a decrease in both developmental rates. Zona pellucida (ZP) digestion time decreased in the SK group while it increased in the ε-ACA group. Raman microspectroscopy revealed an increase in the intensity of the band corresponding to the glycerol group of sialic acid in the ZP of oocytes matured with SK, whereas ZP spectra of oocytes treated with ɛ-ACA presented similarities with immature oocytes. The results indicate that although treatment with SK did not alter oocyte developmental competence, it induced modifications in the ZP of oocytes that could modify the folding of glycoproteins. Plasmin inhibition impairs oocyte maturation and has an impact on embryo development, thus evidencing the importance of this protease during IVM.

RESUMO

This study presents for the first time results about the microplastic concentration and their origin in a stream of the Pampas region in Argentina, receiving wastewater of an intermediate city. The most appropriate procedure to analyze and quantify the microplastics in the studied system is the use of an oxidative digestion process using a 30% H2O2 solution to eliminate the organic matter in the samples. A high quantity of MPs, on the order of millions of microplastics per m3 of water, was estimated in the Langueyú stream. 56% of the microplastics correspond to fibers with diameters between 10 and 15 µm and lengths less than 500 µm, while 44% are fragments with sizes of tens of micrometers. Raman microspectroscopy was used to identify the type of fibers. The characteristics of the microplastic fibers released in a wash load test are comparable with those observed in the Langueyú stream, in particular, the average sizes and the distribution of the diameters of the MPFs are similar. The processes in the sewage treatment plant, prior to their discharge in the stream, would affect the color of the fibers and their length.

RESUMO

The success of the onychomycosis treatment is directly associated with factors such as the choice of the medication, the administration route, and the pharmaceutical formulation. Photodynamic therapy (PDT) is an emerging and promising technique indicated for onychomycosis treatment. For this application, the main challenge is the efficient delivery of the photosensitizer (PS). Curcumin is widely used as a PS, however it is an unstable molecule and it is a challenge to develop a formulation with good penetration into the nail plate, maintaining the stability of curcumin. In this study, the molecular mechanisms underlying the efficacy of two topical formulations containing curcumin used in a clinical trial for onychomycosis treatment were analyzed by Raman microspectroscopy. It is shown that curcumin is present in both formulations in aggregated and non-aggregated states, and in aggregates it is present in different conformations, depending on the interaction with the solvent. This proves to be critical for efficient and uniform PS delivery to the nail and its complete use during the treatment. These analyses are showing how promising Raman microspectroscopy is in understanding the molecular mechanisms of the efficiency of photosensitizers and are helping to improve the development of pharmaceutical formulations.
