RESUMO

Retinoblastoma protein is central in signaling networks of fundamental cell decisions such as proliferation and differentiation in all metazoans and cancer development. Immunostaining and biochemical evidence demonstrated that during interphase retinoblastoma protein is in the nucleus and is hypophosphorylated, and during mitosis is in the cytoplasm and is hyperphosphorylated. The purpose of this study was to visualize in vivo in a non-diseased tissue, the dynamic spatial and temporal nuclear exit toward the cytoplasm of this protein during mitosis and its return to the nucleus to obtain insights into its potential cytosolic functions. Using high-resolution time-lapse images from confocal microscopy, we tracked in vivo the ortholog in plants the RETINOBLASTOMA RELATED (RBR) protein tagged with Green Fluorescent Protein (GFP) in Arabidopsis thaliana's root. RBR protein exits from dense aggregates in the nucleus before chromosomes are in prophase in less than 2 min, spreading outwards as smaller particles projected throughout the cytosol during mitosis like a diffusive yet controlled event until telophase, when the daughter's nuclei form; RBR returns to the nuclei in coordination with decondensing chromosomal DNA forming new aggregates again in punctuated larger structures in each corresponding nuclei. We propose RBR diffused particles in the cytoplasm may function as a cytosolic sensor of incoming signals, thus coordinating re-aggregation with DNA is a mechanism by which any new incoming signals encountered by RBR may lead to a reconfiguration of the nuclear transcriptomic context. The small RBR diffused particles in the cytoplasm may preserve topologic-like properties allowing them to aggregate and restore their nuclear location, they may also be part of transient cytoplasmic storage of the cellular pre-mitotic transcriptional context, that once inside the nuclei may execute both the pre mitosis transcriptional context as well as new transcriptional instructions.

OBJECTIVE: To compare the effect of two programs of Food and Nutrition Education (FNE) on the Nutrition knowledge of elderly women, Brazil. METHODS: Randomized field trial. Community-based elderly women over 60 years (n = 36) participated in a twelve-week intervention in either of two FNE programs: Psychopedagogical Program (PP) or Culinary Workshop Program (CWP). There were three moments of data collection in the study; pre-intervention knowledge data); post intervention and follow up (after 6 months upon intervention completion). A questionnaire about knowledge of food and Nutrition was administered at every data collection moment. RESULTS: In the analysis performed by the Generalized Estimation Equation Models (GEE) there was a significant interaction effect (P = 0.033) in the diabetes block in which the percentage of correct answers in the follow-up moment was higher compared to the pre-intervention moment in the CWP group (65.65% and 45.66%, respectively). In the knowledge blocks: total percentage of correct answers, hypertension, food processing, the evaluation effect was observed, regardless of the intervention performed.Three blocks of knowledge, including the block of the total percentage of correct answers in knowledge in Nutrition, presented a significant evaluation effect, which suggests that the interventions were important for the gain of knowledge, regardless of the type of intervention performed. Elderly-focused FNE programs should include a methodology that encompasses recreational education activities, and practical activities, such as culinary workshops.

Millet is largely explored for a diversity of applications. The use of millet high-quality seeds is essential to increase grain yield. Accordingly, the purposeof this studywas to evaluate the yield variability of milletplants from higher and lower-quality seeds in the expression of dry matter yield parameters. The studywas conducted in the experimental and didactic area of IFRS -campusIbirubá, Rio Grande do Sul, Brazil, with a completely randomized design. The treatments were plants from seeds of higher (HQS) and lower (LQS) quality and two season crops (2015/16 and 2017/18) in eight replications. Theemergence speed was appliedto identify plants from seeds of different qualities. The phyllochron, leaf number, height, stem diameter, number of nodes, and dry mass yield were the agronomic components evaluatedfor this study. Meteorological information was used to calculate phyllochron, water deficit, and average temperature. The collected data were submitted for analysis of variance and the Tukey test at a 5% probability of error. The HQS expressed lower phyllochron and most significant agronomic components. Furthermore, the 2015/16 crop had the lowest water deficit and obtained the best results.(AU)


O milheto é amplamente explorado para uma diversidade de aplicações. O uso de sementes de milheto de alta qualidade é essencial para aumentar a produtividade de grãos. Nesse sentido, o propósito deste estudo foi avaliar a variabilidade da produtividade de plantas de milheto provenientes de sementes de melhor e de menor qualidade na expressão dos parâmetros de produção de matéria seca. O estudo foi realizado na área experimental e didática do IFRS - campus Ibirubá, Rio Grande do Sul, Brasil, com delineamento inteiramente casualizado. Os tratamentos foram plantas provenientes de sementes de qualidade superior (SAS) e inferior (SQI) e duas safras (2015/16 e 2017/18) em oito repetições. A velocidade de emergência foi aplicada para identificar plantas a partir de sementes de diferentes qualidades. O filocrono, número de folhas, altura, diâmetro do caule, número de nós e rendimento de massa seca foram os componentes agronômicos avaliados para este estudo. As informações meteorológicas foram utilizadas para calcular o filocrono, o déficit hídrico e a temperatura média. Os dados coletados foram submetidos à análise de variância e ao teste de Tukey a 5% de probabilidade de erro. O SQS expressou menor filocrono e componentes agronômicos mais significativos. Ainda, a safra 2015/16 apresentou o menor déficit hídrico e obteve os melhores resultados.(AU)

For seed germination, it is necessary to restart the cell cycle, a process regulated at multiple levels including transcriptional control, that is executed by the E2F family of transcription factors. We identified 12 genes of the E2F family in maize that are expressed differentially during the first 28 h post imbibition (HAI). E2Fa/b1;1 and E2Fc proteins were characterized as an activator and a putative repressor respectively, both forming heterodimers with DPb2 that bind differentially to consensus E2F response elements in promoters of E2F target genes. Transcripts of target genes for these transcription factors accumulate during germination; in dry seeds E2Fc protein is enriched in the target promoters and is replaced by E2Fa/b1;1 as germination advances. RBR1 is found in the same promoters in non-imbibed and 28 HAI seeds, when DNA replication has concluded, and transcription of the E2F targets should stop. During germination promoters of these target genes seem to be decorated with histone marks related to relaxed chromatin structure. Therefore, E2Fs appear to occupy their target genes in a context of open chromatin, with RBR1 fine tuning the progression between the phases.

Abstract AIMS the purpose of this study was to assess the effects of a supervised dance intervention based on video game choreography on isokinetic concentric and eccentric knee peak of torque (PT), quadriceps and hamstrings cross sectional area (CSA) and functionality in community-dwelling older women. METHODS Forty-seven older women were allocated in Control Group (CG, n=25) and Intervention Group (IG, n=22). The IG performed dance based on video game choreography (Dance Central, XBOX 360®, Kinect), during 12 weeks, ~40 minutes, 3x/week, without foam (1-6week) and with foam and visual disturbances (7-12 week). The pretest-training-posttest assessments included: isokinetic concentric and eccentric knee PT and CSA (Magnetic resonance imaging) of quadriceps and hamstrings and functional tests. RESULTS The light-to-moderate-intensity 12-weeks training increased 8.5% the eccentric PT of quadriceps at 60°/s (p=0.04) and 1.3% quadriceps CSA (p=0.02). CONCLUSIONS These findings suggest that dance training based on video game choreography can enhance PT and induce hypertrophy in community-dwelling older women.

Leaf rust, caused by the foliar pathogen Puccinia triticina is a major disease of wheat in the southern region of Brazil and invariably impacts on production, being responsible for high yield losses. The Brazilian wheat cultivar Toropi has proven, durable adult plant resistance (APR) to leaf rust, which uniquely shows a pre-haustorial resistance phenotype. In this study we aimed to understand the interaction between P. triticina and the pre-haustorial APR in Toropi by quantitatively evaluating the temporal transcription profiles of selected genes known to be related to infection and defense in wheat. The expression profiles of 15 selected genes varied over time, grouping into six expression profile groups. The expression profiles indicated the induction of classical defence pathways in response to pathogen development, but also the potential modification of Toropi's cellular status for the benefit of the pathogen. Classical defence genes, including peroxidases, ß-1,3-glucanases and an endochitinase were expressed both early (pre-haustorial) and late (post-haustorial) over the 72 h infection time course, while induction of transcription of other infection-related genes with a potential role in defence, although variable was maintained through-out. These genes directly or indirectly had a role in plant lignification, oxidative stress, the regulation of energy supply, water and lipid transport, and cell cycle regulation. The early induction of transcription of defence-related genes supports the pre-haustorial resistance phenotype in Toropi, providing a valuable source of genes controlling leaf rust resistance for wheat breeding.
