BACKGROUND: The Eastern Tropical Pacific (ETP) harbors a great diversity of Porifera. In particular, the Aplysina genus has acquired biotechnological and pharmacological importance. Nevertheless, the ecological aspects of their species and populations have been poorly studied. Aplysina gerardogreeni is the most conspicuous verongid sponge from the ETP, where it is usually found on rocky-coralline ecosystems. We evaluated the polymorphism levels of 18 microsatellites obtained from next-generation sequencing technologies. Furthermore, we tested the null hypothesis of panmixia in A. gerardogreeni population from two Mexican-Pacific localities. METHODS AND RESULTS: A total of 6,128,000 paired reads were processed of which primer sets of 18 microsatellites were designed. The loci were tested in 64 specimens from Mazatlan, Sinaloa (N = 32) and Isabel Island, Nayarit (N = 32). The microsatellites developed were moderately polymorphic with a range of alleles between 2 and 11, and Ho between 0.069 and 0.785. Fifteen loci displayed significant deviation from the Hardy-Weinberg equilibrium. No linkage disequilibrium was detected. A strong genetic structure was confirmed between localities using hierarchical Bayesian analyses, principal coordinates analyses, and fixation indices (FST = 0.108*). All the samples were assigned to their locality; however, there was a small sign of mixing between localities. CONCLUSIONS: Despite the moderate values of diversity in microsatellites, they showed a strong signal of genetic structure between populations. We suggest that these molecular markers can be a relevant tool to evaluate all populations across the ETP. In addition, 17 of these microsatellites were successfully amplified in the species A. fistularis and A. lacunosa, meaning they could also be applied in congeneric sponges from the Caribbean Sea. The use of these molecular markers in population genetic studies will allow assessment of the connectivity patterns in species of the Aplysina genus.

Despite the important roles that marine sponges play in ecosystem functioning and structuring, little is known about how the sponge holobiont responds to local anthropogenic impacts. Here we assess the influence of an impacted environment (Praia Preta) on the microbial community associated with the endemic sponge Aplysina caissara in comparison to a less-impacted area (Praia do Guaecá) from the coast of São Paulo state (Brazil, southwestern Atlantic coast). We hypothesized that the local anthropogenic impacts will change the microbiome of A. caissara and that the community assembly will be driven by a different process (i.e. deterministic versus stochastic) under distinct levels of impact. The microbiome at the amplicon sequence variants level was found to be statistically distinct between sponges from the different sites, and this was also seen for the microbial communities of the surrounding seawater and sediments. Microbial communities of A. caissara from both sites were found to be assembled by deterministic processes, even though the sites presented distinct anthropogenic impacts, showing a pivotal role of the sponge host in selecting its own microbiome. Overall, this study revealed that local anthropogenic impacts altered the microbiome of A. caissara; however, assembly processes are largely determined by the sponge host.

Staphylococci are one of the most common causes of biofilm-related infections. Such infections are hard to treat with conventional antimicrobials, which often lead to bacterial resistance, thus being associated with higher mortality rates while imposing a heavy economic burden on the healthcare system. Investigating antibiofilm strategies is an area of interest in the fight against biofilm-associated infections. Previously, a cell-free supernatant from marine-sponge-associated Enterobacter sp. inhibited staphylococcal biofilm formation and dissociated the mature biofilm. This study aimed to identify the chemical components responsible for the antibiofilm activity of Enterobacter sp. Scanning electron microscopy confirmed that the aqueous extract at the concentration of 32 µg/mL could dissociate the mature biofilm. Liquid chromatography coupled with high-resolution mass spectrometry revealed seven potential compounds in the aqueous extract, including alkaloids, macrolides, steroids, and triterpenes. This study also suggests a possible mode of action on staphylococcal biofilms and supports the potential of sponge-derived Enterobacter as a source of antibiofilm compounds.

BACKGROUND: Mycale cecilia is an abundant Eastern Tropical Pacific sponge living in a wide variety of habitats, including coral reefs where it may directly interact with corals. It is also known to possess secondary metabolites of pharmacological value. These aspects highlight the importance of having a better understanding of its biology, and genetic and population diversity. METHODS AND RESULTS: In the present study, we isolated and characterized twelve novel microsatellite loci by Illumina MiSeq sequencing. The loci were tested in 30 specimens collected from two coral reef localities (La Paz, Baja California Sur and Isabel Island, Nayarit) from the Mexican Pacific using M13(-21) labeling. All loci were polymorphic, with two to nine alleles per locus. Expected heterozygosities varied from 0.616 to 0.901. Eleven loci were tested and successfully amplified in M. microsigmatosa from the Gulf of Mexico. CONCLUSION: Here we report the first microsatellite loci developed for a sponge species from the Eastern Pacific coast. These molecular markers will be used for population genetic studies of M. cecilia, and potentially in other congeneric species; particularly in vulnerable marine areas that require protection, such as coral reefs.

Sponges-associated microorganisms play important roles in their health and ecology; consequently, they may be crucial in the successful adaptation of exotic species to novel environments. However, few studies have focused on the microbial diversity of exotic sponges, especially those with calcium carbonate spicules (class Calcarea). Therefore, this is the first in situ characterization of the microbiota of the exotic calcareous sponges Sycettusa hastifera and Paraleucilla magna. Our results suggest that S. hastifera has a more stable microbiota than P. magna, as there were no differences in its beta diversity among sampling sites. Conversely, P. magna showed significant differences in its microbial communities, perhaps related to its adhesion to artificial substrate and/or shellfish mariculture activities. Each sponge species presented a single dominant proteobacterial OTU potentially active in the nitrogen cycle, which could help sponge detoxification, especially in polluted areas where exotic species usually establish. Our results show the importance of assessing the microbial diversity to unveil host-microorganism relationships and suggest that these associated nitrogen-cycling microorganisms could favor the success of exotic sponges in new environments.

Many studies investigated the presence and effects of microplastics in marine species, but data about sponges are still incipient. We quantified these pollutants in a population of the tropical sponge Cinachyrella alloclada, comparing museum specimens sampled in 1981 with specimens sampled in 2017. The mean number of microplastics in specimens collected decades ago was one order of magnitude lower (0.13 ± 0.40/g of sponge tissue) than those sampled more recently (1.37 ± 0.94/g). We observed microplastics in only 10 % of the specimens collected in 1981 but in 80 % of those from 2017. According to Raman spectroscopy, fibers in C. alloclada consisted of polypropylene. Our results reinforce the importance of natural history collections to reduce the gap of knowledge on the interaction between marine sponges and microplastics.

The survival of a species largely depends on the ability of individuals to reproduce, thus perpetuating their life history. The advent of metazoans (i.e. pluricellular animals) brought about the evolution of specialized tissues and organs, which in turn led to the development of complex protein regulatory pathways. This study sought to elucidate the evolutionary relationships between female reproduction-associated proteins by analyzing the transcriptomes of representative species from a selection of marine invertebrate phyla. Our study identified more than 50 reproduction-related genes across a wide evolutionary spectrum, from Porifera to Vertebrata. Among these, a total of 19 sequences had not been previously reported in at least one phylum, particularly in Porifera. Moreover, most of the structural differences between these proteins did not appear to be determined by environmental pressures or reproductive strategies, but largely obeyed a distinguishable evolutionary pattern from sponges to mammals.

Sponges are fundamental components of coral reef communities and, unfortunately, like other major benthic members, they too have been impacted by epizootic and panzootic events. We report on the prevalence of disease-like conditions affecting populations of the giant barrel sponge Xestospongia muta across shallow and mesophotic coral reefs off La Parguera Natural Reserve (LPNR) and Mona Island Marine Reserve (MIMR) in Puerto Rico. Four different conditions affecting X. muta were observed during our surveys, of which 3 have been previously reported: cyclic spotted bleaching (CSB; apparently non-lethal), Xestospongia-tissue wasting disease (X-TWD; apparently lethal), and sponge orange band disease (SOB; sparsely associated with X-TWD infected individuals). Additionally, we describe a fourth condition, Xestospongia-tissue hardening condition (X-THC), a previously unreported disease recently observed along the insular shelf margin off LPNR and MIMR. Within LPNR, a total of 764 specimens of X. muta were inspected and measured. Of these, 590 sponges (72.2%) had CSB, 25 (3.27%) had signs of X-TWD, 7 (0.92%) had SOB, and the remaining 142 (18.6%) were apparently healthy. Three colonies inhabiting upper mesophotic depths on the LPNR insular shelf showed signs of CSB and X-TWD. At MIMR, video-transect surveys revealed a total of 514 colonies, of which 40 (7.78%) had signs of CSB and/or XTWD, 14 (2.72%) were affected by X-THC, while the remaining 460 (89.5%) showed no external signs of disease and appeared healthy. The presence of 4 concomitant disease-like conditions in barrel sponges of Puerto Rico is alarming, and indicative of the deteriorating status of Caribbean coral reefs.

Marine sponges are known to harbor a diverse and complex microbiota; however, a vast majority of surveys have been investigating the prokaryotic communities in the north hemisphere and Australia. In addition, the mechanisms of microbial community assembly are poorly understood in this pivotal player of the ecosystem. Thus, this survey addressed the holobiome of the sponge species in the São Paulo region (Brazil) for the first time and investigated the contribution of neutral and niche processes of prokaryotic, fungal, and unicellular eukaryotic assemblage in three sympatric species Aplysina caissara, Aplysina fulva, and Tedania ignis along with environmental samples. The compositions of the holobiome associated with the sponges and detected in environmental samples were strikingly different. Remarkably, between 47 and 88% of the assigned operational taxonomic units (OTUs) were specifically associated with sponge species. Moreover, around 77, 69, and 53% of the unclassified OTUs from prokaryotic, fungal, and unicellular eukaryotic communities, respectively, showed less than 97% similarity with well-known databases, suggesting that sponges from the southwestern Atlantic coast are an important source of microbial novelty. These values are even higher, around 80 and 61% of the unclassified OTUs, when excluding low abundance samples from fungal and unicellular eukaryotic datasets, respectively. Host species were the major driver shaping the sponge-associated microbial community. Deterministic processes were primarily responsible for the assembly of microbial communities in all sponge species, while neutral processes of prokaryotic and fungal community assembly were also detected in the sympatric A. caissara and T. ignis replicates, respectively. Most of the species-rich sponge-associated lineages from this region are also found in the Northern seas and many of them might play essential roles in the symbioses, such as biosynthesis of secondary metabolites that exhibit antimicrobial and antiviral activities, as well as provide protection against host predation. Overall, in this study the microbiota was assembled by interactions with the host sponge in a deterministic-based manner; closely related sponge species shared a strong phylogenetic signal in their associated prokaryotic and fungal community traits and Brazilian sponges were a reservoir of novel microbial species.

This paper provides a comprehensive review of the environments where freshwater sponges occur and evaluates the use of sponge spicules as a proxy in paleoenvironmental studies in the Neotropical region. The paper aims to: I) review the information about the ecology of inland sponges to facilitate the use of spicules as a paleoenvironmental tool; and II) identify possible incongruities in the use of this information in paleoenvironmental reconstructions that have been conducted in Neotropical regions. The study compiled data on 77 sponge species, specialist or generalist that occur under certain environmental conditions, such as: substrate type for growth, hydrodynamic types, as well as salinity and acidity concentrations. In addition, it provides a comparison of the paleoenvironmental conditions applied to reconstruction studies that have been carried out within this biogeographic region, highlighting incongruities regarding the current ecology of the sponges.

Despite the ecological importance and the potential pharmacological application of the sponge Mycale (Carmia) cecilia, it is uncertain whether the body-color variation, even in individuals coexisting in the same area, is due to intraspecific phenotypic plasticity or corresponds to taxonomic divergence. This uncertainty is relatively common in several Porifera groups, which lack the resolution of morphological diagnostic characters and slow-evolving mitochondrial genomes as occurs in early splitting lineages. We sequenced the RNA of six individuals with two different body-color (green-morphotype and red-morphotype) collected at the same time side by side. High-throughput sequencing of cDNA libraries produced ~ 129 million reads with a length of 150 bp. Each morphotype was assembled separately owing to the low overlapping in the global assembly. Metatranscriptome de novo assembly of the trimmed and normalized reads produced 461 thousand transcripts for the green-morphotype and 342 thousand for the red-morphotype (respectively). Over 30% of the transcripts contained Open Reading Frames (ORFs) with functional significance. BUSCO analysis of the ORFs of putative poriferan origin (31.3% green or 30.4% red) indicated that our assemblies are 60% complete. This is the first attempt to evaluate the morphological diversity in the species M. (C.) cecilia and the phylum Porifera at the transcriptomic level. Due to the minimum overlap of the assembly and that, the red-morphotype diverged significantly from the green-morphotype (original color of M. (C.) cecilia). Therefore, we suggest that the red-morphotype should undergo a complete taxonomic investigation and its taxonomic status be reviewed. We expect that the transcriptome assembly metrics can be useful for comparing other transcriptome assemblies of non-model organisms.

The complete mitogenome of Thoosa mismalolli Carballo, Cruz-Barraza & Gómez, 2004 (Tetractinellida, Thoosidae) was sequenced. This is the first complete mitogenome of the suborden Thoosina and the third Tetractinellid so far. The mitochondrial genome of T. mismalolli was assembled based on reads obtained with the Illumina HiSeq platform. The length of complete mitogenome is 19,019 bp long and contained 14 protein-coding genes and 23 tRNA, with two tRNA genes. Phylogenetic reconstruction (maximum-likelihood) based on mitogenome of Tetractinellids, supports T. mismalolli as a sister group. This result is congruent with those obtained with molecular markers (CO1, 18S, and 28S), supporting the monophyletic status of Thoosa and providing additional molecular data in favor of the suborder Thoosina.

Cryptopontius is the most species-rich genus in the family Artotrogidae with 25 species, being recorded in almost all oceans, except in the Arctic. However only three species of this genus have been recorded from the Brazilian coast, all of them along the coast of Pernambuco state. This study describes two new species of the genus. Cryptopontius phyllogorgius sp. nov. differs from other species by having 9-segmented antennule, and nine elements on the third exopodal segment of leg 4. Cryptopontius pentadikos sp. nov. differs from its congeners by having five expressed antennulary segments proximal to fused ancestral IX-XII, two setae on second endopodal segment of leg 1, eight elements on third exopodal segment of leg 1 and the free segment of leg 5 with two setae. A key to species of the genus Cryptopontius is provided.

Currently 26 sponge species are known for the Peruvian coast, but so far no raspailiids had been recorded from this region. Raspailiidae are distributed worldwide and its species are characterized by encrusting, massive, lobate, fan-shaped or branching growth forms, usually with a very hispid surface. In the present study, three new species of Raspailiidae are described from the Peruvian coast. Two new Eurypon spp. were collected at Islas Lobos de Afuera (Lambayeque). Eurypon lacertus sp. nov. is a thinly encrusting orange sponge with choanosomal skeleton composed of large tylostyles and small acanthostyles, and ectosomal skeleton with anisoxeas. Eurypon hookeri sp. nov. is a crustose, ruby red sponge, with choanosomal skeleton composed of large (subtylo)styles and acanthostyles, and ectosomal skeleton pierced by acanthostyles and (subtylo)styles, often surrounded by bouquets of smaller styles. The third new species, Plocamione matarani sp. nov., was collected at Matarani (Arequipa). It is a thinly encrusting orange sponge, the only Plocamione with two categories of choanosomal styles, and a single category of ectosomal styles, acanthostyles and anisoacanthostrongyles. These are the first records of Eurypon and Plocamione for the Peruvian coast, and the entire Southeastern Pacific, in the case of the latter.

Porifera has been relatively well studied from underwater caves worldwide. However, sponges in Mexico are only known from two anchialine caves: La Quebrada and El Aerolito, both in Cozumel Island. An ecological study found that sponges have the second highest species richness and density in La Quebrada. The aim of the present work is to describe the sponge biodiversity inhabiting the dark zone of La Quebrada anchialine cave. A total of ten species were found and are described herein, six are new species to science vis. Calyx maya n. sp. a thinly branched sponge; Haliclona (Reniera) stygobia n. sp. globe-shaped with the thinnest diameter of oxeas in its group; Haliclona (Halichoclona) chankanaabiis n. sp. tubular habit with the smallest oxeas in its group; Neosiphonia microtriaeneae n. sp. characterized by very small dichotriaenes; Svenzea germanyanezi n. sp. a lone conical shape; and Diplastrella cozumella n. sp. for its tri to multilobed tylostyles. The other four species are previously known from open reefs or deep-sea: Discodermia adhaerens, Siphonidium ramosum, Cinachyrella kuekenthali, and Plakinastrella onkodes. This is the only inventory of Porifera in Mexican caves up to now, thus the species here described are the first stygobiont sponges from Mexico with an endemism rate of Porifera in this anchialine cave of 60%. Conservation programs are needed to preserve this unique and special ecosystem.

Ciocalypta includes species with finger-shaped fistules emerging from a basal mass and skeleton composed by a central axis and secondary tracts supporting the ectosome. Although worldwide distributed, no species of this genus has been reported from the Eastern Pacific. In this study, a new Ciocalypta is described from shallow waters of the southern coast of Peru. Ciocalypta magnastyla sp. nov. is greenish white, possesses the typical fistules with oscula located at their tips, and it is characterized by the presence of large styles (370-1000/5.0-32.5 µm) in combination with oxeas of intermediate size (160-500/5.0-12.5 µm). This new species constitutes the first record of the genus for the Peruvian coast as well as for the entire Eastern Pacific.

Knowledge of homoscleromorph sponge biodiversity has greatly improved during the last decade thanks to the increasing use of integrative taxonomy and extensive exploration of remote ecosystems. Indeed, recently described species have mostly been small sponges living in dark and near-impenetrable habitats. This work integrates morphological, cytological, ecological and molecular data to describe a new species belonging to the Plakina genus. Plakina doudou sp. nov. was found first during close inspection of photographs taken previously in a submarine cave on Martinique Island, where several new species had already been revealed. The new species lives in syntopy with P. arletensis. It is thinly encrusting, whitish in vivo, and its skeleton harbors a unique composition of diods, triods, monolophose triods and monolophose, dilophose and trilophose calthrops. Sequencing of a portion of the mitochondrial gene cox-1 indicates that the new species belongs to a well-supported clade containing the Mediterranean P. crypta and P. trilopha. However, at the time of publication of this work, we have not yet managed to identify synapomorphies that would support the different clades of Plakina. This genus includes a total of 39 species to date, of which 10 have been recorded in the Western Tropical Atlantic, and 4 in Caribbean submarine caves.

Although the French Polynesian reefs are among the most well studied reefs of the world, sponges are still poorly known, with only 199 species or OTUs of sponges having been described from French Polynesia, 167 at an OTU level and 32 at a species level. From those 199 species, just five are calcareous sponges. As it is possible that this number is underestimated, the aim of the present work was to study the diversity of calcareous sponges from French Polynesia. Hence, different French Polynesian archipelagos were surveyed by SCUBA from 3 to 60 m of depth. Identifications were performed using morphological and molecular (ITS and C-LSU) tools. We found a total of nine species of Calcarea, comprising five different genera. Five species are new to science: Clathrina fakaravae sp. nov., Clathrina huahineae sp. nov., Ernstia variabilis sp. nov., Leucascus digitiformis sp. nov., and Leucandra tahuatae sp. nov. With the present work, the number of identified sponges from French Polynesia at a species level increased from 32 to 41. The only calcareous sponge previously known from French Polynesia that was recollected by our group was Leucetta chagosensis. Our results suggest that the Eastern Indo-Pacific Realm shows more affinity with the Central and the Western Indo-Pacific Realms. Four species supported these affinities: Ascandra cf. crewsi, previously known only from Papua New Guinea, Leucascus simplex from South Australia, and Leucetta chagosensis and L. microraphis, both widespread species in the Indo-Pacific. These two Leucetta species, however, most likely represent species complexes. Once again the molecular markers ITS and C-LSU helped in the identification of calcareous sponges, showing how important is an integrative taxonomy. Although our work has increased in 250% (6 spp to 15 spp) the diversity of calcareous sponges in French Polynesia, it is most possible that this number is still underestimated.

Geographic distributions of freshwater sponges are related to the geological and climatic history of the continents, on the presence of gemmules and the morphological complexity of this resistant body to withstand change. Gemmules are characteristic of the freshwater Families Spongillidae, Metaniidae and Potamolepidae. However, Acanthotylotra alvarengai, Echinospongilla brichardi and a number of other species within the genus Potamolepis do not produce gemmules. Potamolepis is endemic to the Afrotropical region with seven valid species. African freshwater sponges however, are mostly known from a single specimen (the holotype), due to the scarcity of material from these freshwater systems. In the present study, we describe two new species of non-gemmule bearing freshwater sponges from the Neotropical and Afrotropical Regions.

Antarctic sponges were mainly studied from trawling or dredging (shallow and deep water) during pioneering oceanographic expeditions carried out since the late 19th century. More recently, sponge collections by SCUBA diving have allowed the detection of species in more cryptic habitats such as rocky walls. In this study, we analyzed Antarctic sponges collected by SCUBA (in 2016 to 2018; shallower than 25 m) around Doumer Island (Palmer Archipelago, Western Antarctic Peninsula-WAP), where only five sponge species have been known. We gathered over 215 specimens, most part identified; 18 known species and one new species. Clathria (Axosuberites) retamalesi sp. nov., is set apart from its congeners on account of the combination of its habit, categories and dimensions of spicules. The East Antarctic material named as Hymeniacidon spec. (3397 m depth) from the Gauss scientific collection has been related to the shallow species H. torquata Topsent, 1916. We described H. torquata based in several specimens (n= 51) from Doumer Island (WAP), only ca. 41 km from Petermann Island (the type locality). Spicules of H. torquata are smaller than the ones present in the Hymeniacidon spec. material, which is here named Hymeniacidon hentscheli sp. nov., since it does not fit into any known cold water species of Hymeniacidon from Antarctica or the Southern Hemisphere, due to a combination of habit, oscula shape, and spicule dimensions. Only five sponge species were previously known from Doumer Island, also collected by SCUBA. Our findings suggest that the ongoing study of collections of sponges assembled at Doumer Island will still yield new taxonomic findings.
