RESUMO

The tribe Phaseoleae includes several legume crops with assembled genomes. Comparative genomic studies have evidenced the preservation of large genomic blocks among legumes, although chromosome dynamics during Phaseoleae evolution has not been investigated. We conducted a comparative genomic analysis to define an informative genomic block (GB) system and to reconstruct the ancestral Phaseoleae karyotype (APK). We identified GBs based on the orthologous genes between Phaseolus vulgaris and Vigna unguiculata and searched for GBs in different genomes of the Phaseolinae (P. lunatus) and Glycininae (Amphicarpaea edgeworthii) subtribes and Spatholobus suberectus (sister to Phaseolinae and Glycininae), using Medicago truncatula as the outgroup. We also used oligo-FISH probes of two P. vulgaris chromosomes to paint the orthologous chromosomes of two non-sequenced Phaseolinae species. We inferred the APK as having n = 11 and 19 GBs (A to S), hypothesizing five chromosome fusions that reduced the ancestral legume karyotype to n = 11. We identified the rearrangements among the APK and the subtribes and species, with extensive centromere repositioning in Phaseolus. We also reconstructed the chromosome number reduction in S. suberectus. The development of the GB system and the proposed APK provide useful approaches for future comparative genomic analyses of legume species.

Repetitive sequences are ubiquitous and fast-evolving elements responsible for size variation and large-scale organization of plant genomes. Within tribe Phaseoleae (Fabaceae), some genera, such as Phaseolus, Vigna, and Cajanus, show small genome and mostly stable chromosome number. Here, we applied a combined computational and cytological approach to study the organization and diversification of repetitive elements in some species of these genera. Sequences were classified in terms of type and repetitiveness and the most abundant were mapped to chromosomes. We identified long terminal repeat (LTR) retrotransposons, especially Ogre and Chromovirus elements, making up most of genomes, other than P. acutifolius and Vigna species. Satellite DNAs (SatDNAs) were less representative, but highly diverse among species, showing a clear phylogenetic relationship. In situ localization revealed preferential location at pericentromeres and centromeres for both types of sequences, suggesting a heterogeneous composition, especially for centromeres. Few elements showed subterminal accumulation. Copy number variation among chromosomes within and among species was observed for all nine identified SatDNAs. Altogether, our data pointed two main elements (Ty3/Gypsy retrotransponsons and SatDNAs) to the diversification on the repetitive landscape in Phaseoleae, with a typical set of repeats in each species. The high turnover of these sequences, however, did not affect total genome size.

Comparative studies on the structure of foliar anatomy in four species of Rhynchosia from Roraima state (northern Brazilian Amazon) were carried out to identify additional morphological characters to support the definition of the systematic boundaries among the species. Fully expanded leaves, including the petioles, were collected from the upper nodes of a plants stem. Anatomical characters were observed in cross-sections of the middle portion of leaflets and petiole segments. Presence and distribution of glandular trichomes, continuity of sclerenchyma around the vascular bundles in leaflet blades and the conformation of the area between the two ridges of petioles were important characters to distinguish among the species of Rhynchosia. For the first time we report a distinct multicellular gland-like structure which we found in the petioles of three species.(AU)


Estudos comparativos da estrutura anatômica foliar foram realizados para identificar caracteres morfológicos adicionais para a caracterização sistemática de quatro espécies de Rhynchosia ocorrentes em Roraima. Folhas completamente expandidas, incluindo o pecíolo, foram coletadas nos nós superiores da planta. Os caracteres anatômicos foram estudados a partir de observações de cortes tranversais da região mediana de folíolos e pecíolos. A presença e a distribuição de tricomas glandulares, a continuidade do esclerênquima em torno dos feixes vasculares nos folíolos e a conformação da região entre as duas alas nos pecíolos se mostraram importantes caracteres para a distinção das espécies amazônicas de Rhynchosia. Também foi registrada pela primeira vez a ocorrência de uma diferenciada estrutura similar a glândula nos pecíolos de três espécies.(AU)

The EF1α is a multifunctional protein with additional unrelated activities to its primary function in translation. This protein is encoded by a multigene family and few studies are still available in plants. Expression of six EF1α genes in Glycine max was performed using RT-qPCR and RNA-seq data to advance in the function of each gene during plant development, stress conditions and phytohormone treatments. A phylogenetic classification in Phaseoleae tribe was used to identify the G. max EF1α genes (EF1α 1a1, 1a2, 1b, 2a, 2b and 3). Three EF1α types (1-3) were found in Phaseoleae revealing duplications in G. max types 1 and 2. EF1α genes were expressed in all studied tissues, however, specific amount of each transcript was detected. In plant development, all EF1α transcripts were generally more expressed in younger tissues, however, in unifoliolate leaves and cotyledons a higher expression occurred in older tissues. Five EF1α genes (except 2a) were up-regulated under stress in a response tissue/stress/cultivar-dependent. EF1α 3 was the most stress-induced gene linked to cultivar stress tolerance mainly in aerial tissues. Auxin, salicylate and ethylene induced differentially the EF1α expression. Overall, this study provides a consistent EF1α classification in Phaseoleae tribe to better understand their functional evolution. The RT-qPCR and RNA-seq EF1α expression profiles were consistent, both exhibiting expression diversification of each gene (spatio-temporal, stress and phytohormone stimuli). Our results point out the EF1α genes, especially EF1α 3, as candidate for developing a useful tool for future G. max breeding.

Rhynchosia, gênero pantropical, é constituído por 230 espécies distribuídas principalmente nos neotrópicos, considerado o centro de diversidade do grupo. Está relacionado filogeneticamente com o gênero Eriosema (DC.) Rchb., do qual se diferencia principalmente por características morfológicas. Em Mato Grosso do Sul está representado por oito táxons: Rhynchosia balansae Micheli var. balansae, R. balansae var. psilantha Fortunato, R. clausseni Benth., R. corylifolia Mart. ex Benth., R. edulis Griseb., R. leucophylla Benth., R. melanocarpa Grear e R. minima (L.) DC. O presente estudo apresenta chave de identificação, descrições, comentários, ilustrações, dados de distribuição geográfica e dos ambientes preferenciais de cada táxon.


Rhynchosia Lour. is a pantropical genus with 230 species, mainly distributed in the neotropics considered the center of diversity of the group. It is philogenetically related to the genus Eriosema (DC.) Rchb., from which it differs mainly through morphological characters. In Mato Grosso do Sul it is represented by eight taxa: Rhynchosia balansae Micheli var. balansae, R. balansae var. psilantha Fortunato, R. clausseni Benth., R. corylifolia Mart. ex Benth., R. edulis Griseb., R. leucophylla Benth., R. melanocarpa Grear and R. minima (L.) DC. The current study presents identification key, descriptions, comments, illustrations, data on geographic distribution and habitat preference of each taxon.

The aim of this work was to describe the morphology of seedlings and saplings of Pachyrhizus ahipa, a cultivated species of Phaseoleae (Fabaceae), analysing the seedling type and characterizing the structure of cotyledons, eophylls and metaphylls. It was observed that the seedling was semi-hypogeal, a germination type not yet recorded for the Phaseoleae tribe. It formed two opposite and unifoliated eophylls with an evident pair of stipels. Metaphylls were trifoliolate and had alternate phyllotaxis. Both eophylls and metaphylls were pinnate, camptodromous, and brochidodromous.


É de conhecimento geral que existe grande escassez de estudos referentes às fases juvenis das plantas, importantes como subsídio para diversos trabalhos ecológicos, taxonômicos e filogenéticos. Com base nesse pressuposto, descreveram-se morfologicamente plântulas e plantas jovens de Pachyrhizus ahipa, espécie cultivada de Phaseoleae (Fabaceae), analisando-se o tipo de plântula e caracterizando estruturalmente os cotilédones, eofilos e metafilos. Verificou-se que a plântula é semi-hipógea, tipo de germinação até então não registrado para a tribo Phaseoleae. A plântula forma dois eofilos unifoliolados e opostos, acompanhados por evidente par de estipelas. Os metafilos são trifoliolados, de filotaxia alterna. Tanto eofilos quanto metafilos são pinados, camptódromos e broquidódromos.
