RESUMO

The iron ore mining activity results in considerable waste production and impacts on surrounding ecosystems. Natural recovery of impacted areas is absent or occurs slowly, especially when associated with prolonged dry seasons in tropical regions. The objective of this work was to unveil the mechanisms of Paspalum densum (Poir.) grass to overcome the periods of seasonal drought and its metal accumulation in areas impacted by iron mining, a tailings storage facilities and surrounding ferruginous grassland in Brazil. Lower mortality was observed among individuals in the tailings storage facilities, with a 74.3% survival rate. In contrast, after the beginning of the dry season, all individuals died in the ferruginous grassland. The plants in the tailings deposits showed better nutrition, with a higher concentration of P, Cu, Fe, Zn, Mn and greater capacity to accumulate Pb and Cd over time. Pb was the element with highest bioconcetration factor (BCF) and bioaccumulation coefficient (BAC), while Mn was the one with the highest translocation factor (TF). The dry season resulted in reduced chlorophyll a, b and total and effective quantum yield of photosystem II (PSII) of P. densum individuals. However, the plants in the tailings storage facilities showed adjustments to overcome the effects of drought, with an increase in the concentration of proline in leaves and reduction of oxidative damage (MDA and H2O2) at the end of the dry season. The grass P. densum showed rapid acclimatization in the tailings storage facilities and resistance to drought through antioxidant and photosynthetic adjustments and was still able to bioaccumulate and translocate in plant tissues some metals present in the iron ore impacted sites.

RESUMO

(1) Background: Cadmium is a toxic heavy metal that is widely distributed in water, soil, and air. It is present in agrochemicals, wastewater, battery waste, and volcanic eruptions. Thus, it can be absorbed by plants and enter the trophic chain. P. fasciculatum is a plant with phytoremediation capacity that can tolerate Cd stress, but changes in its proteome related to this tolerance have not yet been identified. (2) Methods: We conducted a quantitative analysis of the proteins present in P. fasciculatum leaves cultivated under greenhouse conditions in mining soils doped with 0 mg kg-1 (control), 30 mg kg-1, or 50 mg kg-1. This was carried out using the label-free shotgun proteomics technique. In this way, we determined the changes in the proteomes of the leaves of these plants, which allowed us to propose some tolerance mechanisms involved in the response to Cd stress. (3) Results: In total, 329 variable proteins were identified between treatments, which were classified into those associated with carbohydrate and energy metabolism; photosynthesis; structure, transport, and metabolism of proteins; antioxidant stress and defense; RNA and DNA processing; and signal transduction. (4) Conclusions: Based on changes in the differences in the leaf protein profiles between treatments, we hypothesize that some proteins associated with signal transduction (Ras-related protein RABA1e), HSPs (heat shock cognate 70 kDa protein 2), growth (actin-7), and cellular development (actin-1) are part of the tolerance response to Cd stress.

RESUMO

Most apomictic plants are facultative, maintaining the ability to reproduce sexually at different frequencies depending on the taxa, ploidy, and reproductive stage. In this context, Paspalum species are good model systems for studies evaluating the varying levels of apomixis expression. We aimed to identify, in apomictic tetraploid Paspalum species, the degree of apomixis and residual sexuality in three stages of reproductive development, and if their expression varies along them in order to predict their realized impact on the genetic diversity of future generations. Three main stages in the reproductive development (i.e., ovule, seed, and progeny) were studied in tetraploids from populations of P. cromyorhizon and P. maculosum. Mature ovules were studied using cytoembryological analysis, seeds by flow cytometry, and progeny tests with molecular markers. The expression of sexuality and apomixis was compared in each stage. We observed a decline in expression of sexual reproduction through the consecutive stages, jointly with an increase of apomixis expression. Both species showed at least one tetraploid plant capable of producing progeny by sexual means. These small rates of sexually originated progeny prove the ability of apomictic plants to produce low levels of genetic variation through rare events of sexuality. This study also demonstrates the importance of analyzing different reproductive stages in order to get a whole picture of the reproductive outcomes in plant evolution.

RESUMO

The capacity for apomixis in Paspalum notatum is controlled by a single-dominant genomic region, which shows strong synteny to a portion of rice chromosome 12 long arm. The locus LOC_Os12g40890, encoding the Auxin/Indole-3-Acetic Acid (Aux/IAA) family member OsIAA30, is located in this rice genomic segment. The objectives of this work were to identify transcripts coding for Aux/IAA proteins expressed in reproductive tissues of P. notatum, detect the OsIAA30 putative ortholog and analyze its temporal and spatial expression pattern in reproductive organs of sexual and apomictic plants. Thirty-three transcripts coding for AUX/IAA proteins were identified. Predicted protein alignment and phylogenetic analysis detected a highly similar sequence to OsIAA30 (named as PnIAA30) present in both sexual and apomictic samples. The expression assays of PnIAA30 showed a significant down-regulation in apomictic spikelets compared to sexual ones at the stages of anthesis and post-anthesis, representation levels negatively correlated with apospory expressivity and different localizations in sexual and apomictic ovules. Several PnIAA30 predicted interactors also appeared differentially regulated in the sexual and apomictic floral transcriptomes. Our results showed that an auxin-response repressor similar to OsIAA30 is down-regulated in apomictic spikelets of P. notatum and suggests a contrasting regulation of auxin signaling during sexual and asexual seed formation.

RESUMO

BACKGROUND: The in vitro gas production technique has been used to evaluate forage fermentation kinetics. However, individual and animal species variation can change fermentation patterns due to differences in ruminal environment and microbiota. The aim was to verify whether rumen inoculum (RI) of llama had superior intrinsic digestion capacity and reduced methane (CH4 ) production compared to sheep RI using fescue and paspalum hay as substrates. RESULTS: Dry and organic matter (OM) digestibility produced with llama RI tended to exceed that of sheep (P = 0.099 and 0.074, respectively) at 24 h of incubation. However, at 48 h, the sheep RI presented higher substrate digestibility and asymptotic value of gas production than that of llama (P < 0.010). CH4 production showed no differences between RI or substrates (P > 0.050). The NH3 -N and total volatile fatty acid concentrations were greater in the RI of llamas compared to those of sheep (P < 0.050). Acetate and butyrate proportions and acetate-to-propionate ratio were greater in the RI of llamas compared to those of sheep (P < 0.001) at 24 and 48 h. However, propionate proportion was greater in sheep compared with llama (P < 0.001). CONCLUSION: Llama RI tended to surpass that of sheep in dry and OM digestibility at 24 h of incubation, but sheep RI at 48 h presented a higher digestibility and gas production value than llama RI. No differences between the two species were detected for CH4 production. This study showed that llama RI did not perform better than sheep RI in digesting low-quality forages. © 2022 Society of Chemical Industry.

RESUMO

This study quantified K uptake kinetic parameters in grass species Paspalum notatum, Paspalum urvillei and Axonopus affinis to associate them with growth. Ten milliliters (10 ml) of nutrient solution were collected in two-liter pots - with five repetitions in 24 hours - in order to determine K concentrations in the samples. Shoot and root dry matter, root length, volume, diameter and surface area, as well as kinetic parameters associated with nutrient uptake (Vmax, Km, Cmin and I) were determined. Species P. notatum was the one presenting the highest root, shoot and total dry matter production, as well as the highest root volume. P. urvillei recorded the highest K content, which may be associated with thinner roots and greater root surface area. A. affinis recorded the highest Vmax value, which led to greater K uptake efficiency.

In angiosperms, gametophytic apomixis (clonal reproduction through seeds) is strongly associated with polyploidy and hybridization. The trait is facultative and its expressivity is highly variable between genotypes. Here, we used an F1 progeny derived from diploid apomictic (aposporic) genotypes of Paspalum rufum and two F2 families, derived from F1 hybrids with different apospory expressivity (%AES), to analyze the influence of the environment and the transgenerational transmission of the trait. In addition, AFLP markers were developed in the F1 population to identify genomic regions associated with the %AES. Cytoembryological analyses showed that the %AES was significantly influenced by different environments, but remained stable across the years. F1 and F2 progenies showed a wide range of %AES variation, but most hybrids were not significantly different from the parental genotypes. Maternal and paternal genetic linkage maps were built covering the ten expected linkage groups (LG). A single-marker analysis detected at least one region of 5.7 cM on LG3 that was significantly associated with apospory expressivity. Our results underline the importance of environmental influence in modulating apospory expressivity and identified a genomic region associated with apospory expressivity at the diploid level.

RESUMO

Native species may have adaptive traits that are advantageous for overcoming the adverse environmental conditions faced during the early stages of mine land rehabilitation. Here, we examined the nitrogen (N) growth responses of two native perennial grasses (Axonopus longispicus and Paspalum cinerascens) from canga in nutrient-poor iron mining substrates. We carried out vegetative propagation and recovered substantial healthy tillers from field-collected tussocks of both species. These tillers were cultivated in mining substrates at increasing N levels. The tillering rates of both species increased with the N application. Nonetheless, only in P. cinerascens did the N application result in significant biomass increase. Such growth gain was a result of changes in leaf pigment, stomatal morphology, gas exchanges, and nutrients absorption that occurred mainly under the low N additions. Reaching optimum growth at 80 mg N dm-3, these plants showed no differences from those in the field. Our study demonstrates that an input of N as fertilizer can differentially improve the growth of native grasses and that P. cinerascens plants are able to deposit high quantities of carbon and protect soil over the seasons, thus, making them promising candidates for restoring nutrient cycling, accelerating the return of other species and ecosystem services.

RESUMO

Apomixis seems to emerge from the deregulation of preexisting genes involved in sexuality by genetic and/or epigenetic mechanisms. The trait is associated with polyploidy, but diploid individuals of Paspalum rufum can form aposporous embryo sacs and develop clonal seeds. Moreover, diploid hybrid families presented a wide apospory expressivity variation. To locate methylation changes associated with apomixis expressivity, we compare relative DNA methylation levels, at CG, CHG, and CHH contexts, between full-sib P. rufum diploid genotypes presenting differential apospory expressivity. The survey was performed using a methylation content-sensitive enzyme ddRAD (MCSeEd) strategy on samples at premeiosis/meiosis and postmeiosis stages. Based on the relative methylation level, principal component analysis and heatmaps, clearly discriminate samples with contrasting apospory expressivity. Differential methylated contigs (DMCs) showed 14% of homology to known transcripts of Paspalum notatum reproductive transcriptome, and almost half of them were also differentially expressed between apomictic and sexual samples. DMCs showed homologies to genes involved in flower growth, development, and apomixis. Moreover, a high proportion of DMCs aligned on genomic regions associated with apomixis in Setaria italica. Several stage-specific differential methylated sequences were identified as associated with apospory expressivity, which could guide future functional gene characterization in relation to apomixis success at diploid and tetraploid levels.

RESUMO

Successive mowing are the major maintenance costs of lawns. Thus, both the expenditure with mowing and the visual and physiological aspect of the lawn have led to the search for alternatives to mechanical management. Thus, this work aimed to study the effects of different rates of imazapic herbicide applied alone or combined with imazapyr as a growth regulator of Bahiagrass (Paspalum notatum) and St. Augustine grass (Stenotaphrum secundatum). The experimental design was a randomized block with four replicates, and the treatments consisted of six rates of imazapic herbicide (35; 70; 105; 140; 175 and 210 g a.i. ha-1) for both species, three rates of imazapic + imazapyr in tank mix (15.57 + 5.25; 23.625 + 7.875; 32.5 + 10.5 g a.i. ha-1) for Bahiagrass and four rates of imazapic + imazapyr mixture (7.875 + 2.625; 15.57 + 5.25; 23.625 + 7.875; 32.5 + 10.5 g a.i. ha-1) for St. Augustine grass. The effect of the treatments was evaluated by observing visible injury symptoms, canopy height, height and number of inflorescences and total dry matter of clippings. Applications of imazapic alone or combined with imazapyr were effective in reducing plant height, number and height of inflorescences and total amount of dry matter of clippings produced by Bahiagrass plants. Imazapic provided satisfactory control of St. Augustine growth, but its utilization caused an increase in the number of inflorescences present in the lawns.

RESUMO

The revegetation of areas degraded by iron ore mining is a difficult challenge mainly due to water availability and impoverished metal-rich substrates. We sought to understand the photosynthetic responses to drought of native tropical grasses Paspalum densum (Poir.) and Setaria parviflora (Poir.) grown in iron ore tailing. The grass P. densum presented better photosynthetic adjustments when grown in the iron ore tailing and S. paviflora in response to water stress. Both species accumulated iron above the phytotoxic threshold when grown in an iron ore tailing. The net photosynthesis, stomatal conductance, transpiration, and water use efficiency decreased followed by a reduction in leaf relative water content in response to water stress for both species. The photochemical efficiency of photosystem II only decreased at the point of maximum drought. At this point, the water-stressed grass grown in the iron ore tailing presented higher H2O2 concentrations, particularly S. parviflora. After rehydration, full recovery of photosynthetic variables was achieved with decreased malondialdehyde concentrations, increased catalase activity, and, consequently, decreased H2O2 concentrations in leaves for both species. The fast recovery of the native grasses P. densum and S. parviflora to drought in the iron ore tailing substrate is indicative of their resistance and potential use in the revegetation of impoverished mined areas with high iron content and seasonal water deficit.

RESUMO

The objective of this work was to determine Paspalum notatum genotypes appropriate for the establishment of mixtures with temperate legumes and compare the productivity of mixtures with mineral-fertilized systems along two years. Four hybrids of P. notatum, ecotype Bagual and cv. Pensacola, were either subjected to mixtures with white clover (Trifolium repens) and birdsfoot trefoil (Lotus corniculatus) or fertilized with 0, 60, 120, 240, and 480 kg N ha−1 year−1. P. notatum leaf blades and stems and herbage accumulation denoted a management × year interaction. Herbage accumulation varied between the mixtures and fertilized systems in first year, while in the second year, herbage accumulation of mixed systems employing the B26, B43, C22, C9, and Bagual genotypes were similar to that of systems fertilized with 120 and 240 kg N ha−1. Hybrids B26 and C9 and ecotype Bagual, once made available to producers, could be indicated for mixtures with temperate legumes because they are superior to cv. Pensacola. Herbage accumulation in mixtures involving white clover plus birdsfoot trefoil and P. notatum genotypes is similar to that of mineral nitrogen-fertilized systems with 240 kg N ha−1, emphasizing the viability of mixed between these species.

RESUMO

This study investigated the water parameters and the levels of several heavy metals in leaves and roots of the aquatic macrophyte Paspalum repens and in bed sediments of one unpolluted lagoon and two polluted lagoons of the Orinoco River floodplain to know the impact of the alumina refining wastes on these ecosystems. The sampling was performed during four hydrological periods, covering one hydrological year. Chemical parameters in water (pH, conductivity, and total dissolved solids) and the levels of Al, Pb, Cr, and Cu in sediments were the highest in Macanillal (LMaca) lagoon, whereas sediments of Punta Cuchillo (LPC) lagoon also had elevated concentrations of Al and Fe in comparison with Las Aruhacas (LArh) lagoon (unpolluted lagoon). Chemical parameters in waters returned almost to natural values during high water stage because of the huge inundation of the Orinoco River during the high discharge periods. The solid/liquid discharges of industrial effluents over these systems also affected the grain size distribution of sediments in the lagoons. In all the studied lagoons, the distribution of elements in leaves of P. repens was in the order K ≥ Cr > Ca > Mg > Al > Fe > Mn > Cu > Pb, whereas Al was found to be the more abundant element in roots. The high abundance of Cr in P. repens was related with the high Cr accumulation potential shown by aquatic macrophytes. Despite this, the concentration of elements in P. repens did not show differences between the plants collected in the polluted lagoons versus the plants collected in the unpolluted lagoon. This suggests that P. repens is unsuitable for bio-monitoring studies of heavy metals in these aquatic environments impacted by alkaline residues.

RESUMO

The objective of the following research was to investigate the physiological responses of two bahiagrass genotypes (Paspalum notatum Flügge) submitted to water restriction, induced by polyethylene glycol (PEG6000), during seed germination. Seeds of cv. Pensacola (diploid) and Bagual ecotype (tetraploid) were placed on paper moistened with osmotic solutions at potentials of 0.0; -0.1; -0.2 and -0.3 MPa. Composed of two subsamples of 100 seeds and four experimental units per treatment, the germination test was conducted in a germination chamber under alternating temperatures of 30 °C (8 hours) and 20 °C (16 hours), with presence of light during the higher temperature. Final germination percentage and germination speed index of the seeds, as well as early growth, activity and expression of antioxidant enzymes of seedlings were evaluated. The water restriction induced by PEG6000 reduced the percentage of germination in Pensacola (from 74% to 59%) and Bagual (34% to 10%). For the latter, the evaluation of seedlings showed higher dry mass in relation to cv. Pensacola (1.7 times higher in root and 1.6 times in shoot). In response to moderate water restriction, the genotypes showed increases in root growth (1.7 times higher in -0.1 compared to 0.0 MPa), both in expression and enzymatic activity (superoxide dismutase and catalase).(AU)

BACKGROUND: Paspalum notatum exhibits both sexual and apomictic cytotypes and, thus, is considered a good model for studies of apomixis because it facilitates comparative approaches. In this work, transcriptome sequencing was used to compare contrasting P. notatum cytotypes to identify differential expression patterns and candidate genes involved in the regulation of expression of this trait. RESULTS: We built a comprehensive transcriptome using leaf and inflorescence from apomictic tetraploids and sexual diploids/tetraploids and a coexpression network based on pairwise correlations between transcript expression profiles. We identified genes exclusively expressed in each cytotype and genes differentially expressed between pairs of cytotypes. Gene Ontology enrichment analyses were performed to better interpret the data. We de novo assembled 114,306 reference transcripts. In total, 536 candidate genes possibly associated with apomixis were detected through statistical analyses of the differential expression data, and several interacting genes potentially linked to the apomixis-controlling region, genes that have already been reported in the literature, and their neighbors were transcriptionally related in the coexpression network. CONCLUSIONS: Apomixis is a highly desirable trait in modern agriculture due to the maintenance of the characteristics of the mother plant in the progeny. The reference transcriptome, candidate genes and their coexpression network identified in this work represent rich resources for future grass breeding programs.

RESUMO

Claviceps species affecting Paspalum spp. are a serious problem, as they infect forage grasses such as Paspalum dilatatum and P. plicatulum, producing the ergot disease. The ascomycete C. paspali is known to be the pathogen responsible for this disease in both grasses. This fungus produces alkaloids, including ergot alkaloids and indole-diterpenes, that have potent neurotropic activities in mammals. A total of 32 isolates from Uruguay were obtained from infected P. dilatatum and P. plicatulum. Isolates were phylogenetically identified using partial sequences of the genes coding for the second largest subunit of RNA polymerase subunit II (RPB2), translation elongation factor 1-α (TEF1), ß-tubulin (TUB2), and the nuc rDNA 28S subunit (28S). Isolates were also genotyped by randomly amplified polymorphic DNA (RAPD) and presence of genes within the ergot alkaloid (EAS) and indole-diterpene (IDT) biosynthetic gene clusters. This study represents the first genetic characterization of several isolates of C. paspali. The results from this study provide insight into the genetic and genotypic diversity of Claviceps paspali present in P. dilatatum and suggest that isolates from P. plicatulum could be considered an ecological subspecies or specialized variant of C. paspali. Some of these isolates show hypothetical alkaloid genotypes never reported before.

RESUMO

The phytoremediation capacity of Paspalum fasciculatum Willd. ex Flüggé, was evaluated in soils from a gold mine contaminated with cadmium (Cd) and lead (Pb), using three concentration levels of each metal (15, 30, and 50 mg kg-1). Their ability to assimilate Cd and Pb in its different tissues was evaluated during 90 days of exposure. Plant growth behavior, accumulation of Cd and Pb, and translocation (TF) and bioaccumulation (BAF) factors were also determined. During the first 60 days of exposure, Cd had an inductive effect on the growth of P. fasciculatum; however, after 90 days, this metal had begun to show toxic effects. Plants showed a similar pattern of accumulating Cd and Pb in their tissues with concentrations decreasing in the order roots > leaves > stem. However, the accumulated concentrations of Cd were generally higher than those of Pb with the highest metal uptakes being observed during the first 30 days of exposure. P. fasciculatum was shown to have a phytostabilization effect with regard to Cd, high concentrations of metals in tissues and little translocation, whereas it showed phytoextraction capacity for Pb. In addition, it can increase pH and organic matter in the soil rhizosphere.

The Paspalum genus forms the natural pastures of the tropical and subtropical regions of America and includes several species of recognized forage potential. The aim of this study was to evaluate the influence of different levels of nitrogen fertilization on the agronomic traits and nitrogen use efficiency in superior genotypes of Paspalum notatum Flügge. Four nitrogen fertilization levels (0, 60, 180 and 360 kg N ha-1) and nine genotypes were evaluated, six from the United States Department of Agriculture (USDA) (30N, 36N, 48N, 70N, 83N and 95N), collected in South America, two native genotypes of Rio Grande do Sul, Brazil (André da Rocha and Bagual) and cv. Pensacola. The experimental design was a randomized complete block in a 4 x 9 factorial, with four field replications. From the variability of the responses found for the agronomic traits, the genotypes 48N, André da Rocha and Bagual are indicated to continue in the breeding program because they reach the maximum productive efficiencies according to the doses of N tested. The genotype 36N obtained the highest efficiency of nitrogen utilization, being biologically viable with the lowest N dose, equivalent of 60 kg of N ha-1.(AU)
