BACKGROUND: The integration of molecular data from hosts, parasites, and microbiota can enhance our understanding of the complex biological interactions underlying the resistance of hosts to parasites. Haemonchus contortus, the predominant sheep gastrointestinal parasite species in the tropics, causes significant production and economic losses, which are further compounded by the diminishing efficiency of chemical control owing to anthelmintic resistance. Knowledge of how the host responds to infection and how the parasite, in combination with microbiota, modulates host immunity can guide selection decisions to breed animals with improved parasite resistance. This understanding will help refine management practices and advance the development of new therapeutics for long-term helminth control. METHODS: Eggs per gram (EPG) of feces were obtained from Morada Nova sheep subjected to two artificial infections with H. contortus and used as a proxy to select animals with high resistance or susceptibility for transcriptome sequencing (RNA-seq) of the abomasum and 50 K single-nucleotide genotyping. Additionally, RNA-seq data for H. contortus were generated, and amplicon sequence variants (ASV) were obtained using polymerase chain reaction amplification and sequencing of bacterial and archaeal 16S ribosomal RNA genes from sheep feces and rumen content. RESULTS: The heritability estimate for EPG was 0.12. GAST, GNLY, IL13, MGRN1, FGF14, and RORC genes and transcripts were differentially expressed between resistant and susceptible animals. A genome-wide association study identified regions on chromosomes 2 and 11 that harbor candidate genes for resistance, immune response, body weight, and adaptation. Trans-expression quantitative trait loci were found between significant variants and differentially expressed transcripts. Functional co-expression modules based on sheep genes and ASVs correlated with resistance to H. contortus, showing enrichment in pathways of response to bacteria, immune and inflammatory responses, and hub features of the Christensenellaceae, Bacteroides, and Methanobrevibacter genera; Prevotellaceae family; and Verrucomicrobiota phylum. In H. contortus, some mitochondrial, collagen-, and cuticle-related genes were expressed only in parasites isolated from susceptible sheep. CONCLUSIONS: The present study identified chromosome regions, genes, transcripts, and pathways involved in the elaborate interactions between the sheep host, its gastrointestinal microbiota, and the H. contortus parasite. These findings will assist in the development of animal selection strategies for parasite resistance and interdisciplinary approaches to control H. contortus infection in sheep.

ABSTRACT Natural products are safe environmentally friendly agents and have no negative impact on the environment, they can be used to combat parasitic diseases. Helminthiasis and coccidiosis are parasitic diseases that harm both health and the economy. This research aimed to see how Anethum graveolens leaves extract (AGLE) worked as an anti-parasitic modulator during oocyst sporulation of an Eimeria papillata infection. FT-IR phytochemical analysis revealed the presence of eight compounds. The time required to induce paralysis and death in worms at the highest concentration (200 mg/mL) was 4.57±0.26 and 5.22±0.10 min, respectively. In an in vitro study, AGLE (300 mg/ml) inhibited sporulation by approximately 100% after 72 and 96 hr. AGLE (200, 100, and 50 mg/ml), amprolium, DettolTM, and phenol induced variable inhibition levels at 96 hr of 5.54%, 1.01%, 37.33%, 81.33%, and 89.33%, respectively. Our findings suggest that AGLE has potent anthelmintic and anticoccidial properties that could be further developed into a novel therapeutic agent.
RESUMO Os produtos naturais são agentes ecologicamente corretos, seguros e não têm impacto negativo sobre o meio ambiente, podendo ser usados para combater doenças parasitárias. A helmintíase e a coccidiose são doenças parasitárias que prejudicam a saúde e a economia. O objetivo desta pesquisa foi verificar como o extrato das folhas de Anethum graveolens (AGLE) funcionou como modulador antiparasitário durante a esporulação de oocistos de uma infecção por Eimeria papillata. A análise fitoquímica FT-IR revelou a presença de oito compostos. O tempo necessário para induzir a paralisia e a morte dos vermes na concentração mais alta (200mg/mL) foi de 4,57±0,26 e 5,22±0,10 minutos, respectivamente. Em um estudo in vitro, o AGLE (300 mg/ml) inibiu a esporulação em aproximadamente 100% após 72 e 96 horas. O AGLE (200, 100 e 50 mg/ml), o amprólio, o DettolTM e o fenol induziram níveis de inibição variáveis de 5,54%, 1,01%, 37,33%, 81,33% e 89,33%, respectivamente, após 96 horas. Nossos resultados sugerem que o AGLE tem propriedades anti-helmínticas e anticoccidianas potentes que poderiam ser desenvolvidas em um novo agente terapêutico.

Among the gastrointestinal nematodes affecting sheep, Haemonchus contortus is the most prevalent and virulent, resulting in health problems and production losses. Therefore, selecting sheep resistant to H. contortus is a suitable and sustainable strategy for controlling endoparasites in flocks. Here, 287 lambs of the native Brazilian Morada Nova hair sheep breed were subjected to two consecutive artificial infections with H. contortus and assessed for fecal egg count (FEC), packed cell volume (PCV), and live weight (LW). Forty-four animals ranked as having extreme resistance phenotypes were genotyped using the Illumina OvineSNP50v3 chip. A case−control genome-wide association study (GWAS) detected 37 significant (p < 0.001) markers in 12 ovine chromosomes in regions harboring quantitative trait loci (QTL) for FEC, Trichostrongylus spp. adults and larvae, weight, and fat; and candidate genes for immune responses, mucins, hematological parameters, homeostasis, and growth. Four single-nucleotide polymorphisms (SNP; OAR1_rs427671974, OAR2_rs419988472, OAR5_rs424070217, and OAR17_rs401006318) genotyped by qPCR followed by high-resolution melting (HRM) were associated with FEC and LW. Therefore, molecular markers detected by GWAS for H. contortus resistance in Morada Nova sheep may support animal selection programs aimed at controlling gastrointestinal nematode infections in flocks. Furthermore, genotyping of candidate genes using HRM qPCR may provide a rapid and efficient tool for animal identification.
RESUMO
Climate change is causing detrimental changes in living organisms, including pathogens. This review aimed to determine how climate change has impacted livestock system management, and consequently, what factors influenced the gastrointestinal nematodes epidemiology in small ruminants under tropical conditions. The latter is orientated to find out the possible solutions responding to climate change adverse effects. Climate factors that affect the patterns of transmission of gastrointestinal parasites of domesticated ruminants are reviewed. Climate change has modified the behavior of several animal species, including parasites. For this reason, new control methods are required for controlling parasitic infections in livestock animals. After a pertinent literature analysis, conclusions and perspectives of control are given.

In this study, we evaluated the therapeutic efficacy of diminazene diaceturate at a dose of 7 mg/kg (DA), imidocarb dipropionate at 4.8 mg/kg (IMD), isometamidium chloride at 0.5 and 1.0 mg/kg (ISM 0.5 and ISM 1.0) and combinations applied through different methods to treat Trypanosoma vivax in experimentally infected calves. Thirty male Girolando calves were kept indoors and infected intravenously with T. vivax trypomastigotes (approximately 1 × 106). On D-1, the calves were randomized based on the quantity of infecting parasites per animal, yielding six groups of five animals each: G1: positive control group without treatment; G2 animals treated with DA on Day 0 intramuscularly (IM); G3 animals treated with IMD on Day 0 and D + 14 subcutaneously; G4 animals treated with ISM 0.5 on Day 0 IM; G5 animals treated with ISM 1.0 on Day 0 IM; G6 animals received DA on Day 0 and ISM 1.0 on D + 14, both IM. Throughout 180 days, blood samples were collected for the evaluation of T. vivax using the Woo, Brener and PCR methods. The results indicated that the treatment protocols with DA and/or ISM 0.5 and ISM 1.0 had high efficacy (100 %) against T. vivax. Interestingly, cattle that received ISM remained free of parasites until D + 180. In contrast, animals treated with IMD had relapsed T. vivax detected on the 10th and 14th days post-treatment (DPT). Cattle that received ISM 1.0 did not exhibit relapsed T. vivax in the blood, even after reinfection performed on the 50th DPT. However, treatment with DA on Day 0 failed to prevent a new infection of T. vivax on the 50th DPT. The animals that received ISM 1.0 had a transient decrease in packed cell volume similar to that found in the control group. The reappearance of T. vivax in herds in Brazil treated with DA likely occurred due to the short half-life of the drug and not necessarily due to T. vivax resistance to DA.

The aim of this study was to compare the economic revenue related to the use of low- or high-efficacy anthelmintic drugs within suppressive or strategic schemes of treatment in growing heifers. Heifers raised in a semi-intensive grazing system in southern Brazil were used. Levamisole and ivermectin were selected as the high- and the low-efficacy drugs, respectively, based on a previous efficacy test. Subsequently, these drugs were used within strategic (Strat; four times per year) or suppressive (Supp; once a month) treatment regimens in the heifers, and their liveweight and eggs per gram of feces counts were monthly evaluated during a 13-month period. The total costs of the treatments and their cost-benefit ratio in regard to liveweight gain were calculated. Final mean liveweight gains (kg) observed were 126.7 (Strat-Low), 133.6 (Supp-Low), 141.3 (Strat-High), 142.9 (Supp-High), and 125.8 (Control). Treatments with a high-efficacy drug resulted in monetary gains of US$ 19.56 (Strat-High) and US$ 14.98 (Supp-High), but Supp-Low and Strat-Low treatments caused economic losses. Total cost of the efficacy test (US$ 374.79) could be paid by the additional liveweight gain of 20 heifers from the Strat-High group. These results showed that it would be preferable not to treat the heifers against GIN if compared with treating them with a low-efficacy drug. In addition, we showed that the use of four treatments per year with a high-efficacy drug-selected by efficacy test-resulted in a profitable management to control GIN in growing heifers raised in a semi-intensive gazing system in southern Brazil.

Infection with Haemonchus contortus is the most economically important gastrointestinal nematode parasitosis and the most important cause of mortality in sheep production. The aim of this study was to estimate variance components of gastrointestinal parasite resistance traits, maternal efficiency (ME) and ewe adult weight (AW) in Santa Inês breed in tropical conditions. The phenotypic records were collected from 700 animals, belonging to four flocks located south-east and north-east in Brazil. The evaluated traits were as follows: degree of anaemia assessed using the FAMACHA chart (FMC), haematocrit (HCT), white blood cell (WBC), red blood cell (RBC), haemoglobin (HGB), platelets (PLT), faecal egg count (EPGlog ), ME, metabolic maternal efficiency (MME), AW and metabolic ewe adult weight (MAW). From the 700 animals, 576 (82% of the evaluated population) were genotyped with the Ovine SNP12k BeadChip (Illumina, Inc.). Markers with unknown genomic position, located on sex chromosomes, monomorphic, with minor allele frequency <0.05, call rate <90% and with excess heterozygosity were excluded. The variance components were estimated using a single-trait animal model with ssGBLUP procedure. The correlation between the parasite's resistance indicators and the ME suggested that selecting animals with both higher adult weight and ME will also favour the selection of animals with better resistance to gastrointestinal nematodes parasites, specially H. contortus. Therefore, since there are few or no studies with Santa Inês breed in this area, it is important to study those traits to better manage selection programs.

The objective of this study was to determine if the mean faecal egg count (FEC) from a first experimental Haemonchus contortus infection could be used to classify parasite-naïve Pelibuey hair sheep as parasite-resistant high responders and parasite-susceptible low responders. Twenty 6- to 7-month-old Pelibuey male sheep raised free of gastrointestinal nematodes were challenged with 7500±1412 H. contortus L3 larvae administrated orally on day 0 of the study. Faecal samples from each lamb were obtained daily from 21 to 41days post-infection (Stage I). Lambs received a second artificial infection of 8420±1545L3 larvae on day 42, with faecal samples collected from day 65 to day 78 (Stage III). The mean FEC for each lamb in Stage I was used to classify 8 lambs with means for FEC that were more than two standard errors (SE) below the overall mean (i.e., <4764 eggs per gram of feces; epg) as high responders. The remaining 12 lambs were classified as low responders. Means for FEC in Stage I were 2449±194 epg for high responders and 14,461±1044 epg for low responders (P<0.05). High responders also had lower FEC than low responders in Stage III (actual means of 650±220 vs. 5933±1990 epg; P<0.05 following log transformation to normalize the FEC distribution). Lambs were then reclassified as high and low responders based on their mean FEC in Stage III. Fourteen lambs with means for FEC that were more than one standard error (SE) below the overall mean (i.e., below 1537 epg) were classified as high responders. The remaining six lambs were classified as low responders. Use of the Stage I responder class to predict the Stage III responder class resulted in an 83.3% sensitivity but only a 50% specificity. The positive predictive value was 41.7% and the negative predictive value was 87.5%. The poor positive predictive value was caused by 5 animals with high FEC in Stage I, but low FEC in Stage III. The first infection thus identified most high-responder lambs, but a second infection may improve accuracy by separating lambs with an intermediate level of resistance from truly susceptible lambs. This protocol now requires additional validation under more practical conditions involving natural parasite infections and larger lamb numbers.

Tests with acaricides followed standard protocols, recommended for in vitro testing. The effectiveness of theproduct was calculated based on egg laying and hatching. The drug tested is considered effective if the result was morethan 95%. The number of farms with populations of resistant ticks were 13 (41.9% - group A), 2 (6.45% - group B), 15(48.38% - group C), 6 (19.35% - group D) and 12 (38.7% - group E). Since horn fly populations became established throughout Brazil, complaints regarding control failure have increased around the country. A broad survey to evaluate the susceptibility of horn flies to both organophosphate (OP) and pyrethroid insecticides was conducted from October 2000 to April 2003. Bioassays using filter papers impregnated with cypermethrin, permethrin or diazinon were conducted on 154 horn fly populations in 14 states and 78 municipalities. Resistance to cypermethrin, the active ingredient present in most insecticide products for horn fly control in Brazil, was detected in 98.46% of the populations, with resistance ratios (RR) ranging from 2.5 to 719.9. Resistance to permethrin (RRs < 6.3) was found in 96.67% of the populations, despite its lack of use. In general, pyrethroid resistance was detected in 97.18% of the horn fly populations, with frequencies greater than 87% in all regions of the country. The status of susceptibility of horn fly populations in Brazil to insecticides can be characterized by high susceptibility to OPs and widespread resistance to pyrethroids, potentially compromising the efficacy of pyrethroid products in most cases. Although some partial results have previously been presented, a general picture of horn fly susceptibility in Brazil is presented here for the first time.(AU)

Bioensaios com papéis de filtro impregnados com cipermetrina, permetrina ou diazinon foram conduzidos em 154 populações da mosca-dos-chifres de 14 Estados e 78 municípios. Resistência à cipermetrina, ingrediente ativo presente na maioria dos produtos para controle da mosca-dos-chifres no Brasil, foi detectada em 98,46% das populações, com fatores de resistência (FR) variando de 2,5 a 719,9. Resistência à permetrina (FR < 6,3) foi encontrada em 96,67% das populações testadas, apesar da falta de uso de produtos contendo este princípio ativo. Em geral, resistência a piretróides foi detectada em 97,18% das populações, com frequências acima de 87% em todas as regiões do país. A situação da suscetibilidade da mosca-dos-chifres a inseticidas, no Brasil, pode ser caracterizada por uma elevada suscetibilidade aos organofosforados e ocorrência generalizada de resistência aos piretróides, potencialmente comprometendo a eficácia desses produtos na maioria dos casos. Apesar de sua conhecida resistência, os ovinos estão sujeitos as endoparasitoses, sendo este o principal fator limitante para a sua produção em todo o mundo, especialmente em regiões tropicais. Este trabalho objetiva avaliar a carga parasitária, o perfil hematológico e bioquímico sérico, e o grau Famacha de ovinos sem padrão racial definido infectados e tratados com ivermectina e closantel. O trabalho foi realizado no mês de julho de 2010, numa propriedade rural no município de Mossoró-RN. Foram selecionados 41 animais, os quais foram divididos em três grupos: grupo I- controle (sem tratamento), grupo II- tratado com ivermectina (0,2mg/kg) e Grupo III- tratado com closantel (5mg/kg). As amostras de fezes foram coletadas nos dias 0, 7, 14 e 21, pós-tratamento para análise quantita tiva (OPG) e para análise qualitativa (coprocultura), foram utilizadas amostras dos dias 0, 14 e 21. A primeira coleta de sangue se deu no dia 0, a segunda e a terceira 24 e 72 horas após a primeira, respectivamente, a quarta e a quinta 7 e 14 dias após a primeira, dos quais foram analisados o hemograma, os níveis séricos de proteínas totais, albumina e globulinas, e a relação albumina/globulinas. Foi determinado o grau Famacha de cada animal em todos os momentos de coleta de amostras. O tratamento com closantel foi eficaz no tratamento das helmintoses, principalmente Haemonchus contortus, nos ovinos avaliados. Por outro lado, houve resistência dos parasitas à ivermectina. Os dados de Famacha apresentaram correlação negativa com hematócrito, leucócitos, hemoglobina, albumina, proteínas totais, globulinas e relação albumina/globulinas. O hematócrito apresentou uma correlação forte e positiva com Hemoglobina, Albumina e Proteínas totais. Os tratamentos com closantel e ivermectina não foram responsáveis por alterações consideráveis nos parâmetros hematológicos e bioquímicos avaliados.
Despite its known resistance, the sheep are subject to endoparasitoses, which are the main limiting factor for its production worldwide, especially in tropical regions. This study aims to evaluate the changes in parasitic load, serum biochemical and hematological panel, and Famacha scores of mixed-bred sheep naturally infected with nematodes and treated with ivermectin and closantel. The study was conducted at July 2010, in a farm in the municipality of Mossoró, Rio Grande do Norte state, Brazil. We selected 41 animals were divided into three groups: group I- control (without treatment), group II- treated with ivermectin (0.2 mg/kg), and Group III- treated with closantel (5mg/kg). Stool samples were collected on days 0, 7, 14 and 21 after treatment for quantitative analysis (EPG) and qualitative analysis (stool culture), we used samples from days 0, 14 and 21. The first blood sample was given on day 0, the second and third 24 and 72 hours after the first, respectively the fourth and fifth 7 and 14 days after the first of which analyzed the blood count, serum total protein, albumin and globulins, and albumin/globulins ratio. Famacha scores were determined the degree of each animal at all times of sampling. Closantel administration was effective in the treatment of helminthiasis, especially Haemonchus contortus in evaluated sheep. On the other hand, there was parasite resistance to ivermectin. Famacha data showed negative correlation with packed cell volume, leukocytes, hemoglobin, albumin, total protein, globulin and albumin/globulin ratio. The packed cell volume showed a strong and positive correlation with hemoglobin, albumin and total protein. Treatment with ivermectin and closantel were not responsible for considerable changes in hematological and biochemical parameters evaluated.

Apesar de sua conhecida resistência, os ovinos estão sujeitos as endoparasitoses, sendo este o principal fator limitante para a sua produção em todo o mundo, especialmente em regiões tropicais. As lactonas macrocíclicas (LMs) (avermectinas e milbemicinas) são endectocidas amplamente utilizados em animais e em algumas parasitoses humanas. Em bovinos, a resistência parasitária às LMs é emergente, e o surgimento de formulações que diferem nas suas propriedades farmacológicas tornou complexa a escolha da droga mais indicada a cada caso. Com o objetivo de avaliar possíveis alternativas para recuperar a eficácia de LMs sobre cepas resistentes de nematódeos gastrintestinais, testaram-se, neste estudo, dez diferentes tratamentos a base de LMs sobre uma população de nematódeos gastrintestinais de bovinos a qual, sabidamente, sofrera pressão de seleção por avermectinas a 1 por cento. Adicionalmente, testou-se um benzimidazol. A eficácia das drogas foi calculada com base na redução de ovos por grama de fezes (OPG) dos bovinos. A resistência de cada gênero foi avaliada por meio de identificação de larvas, obtidas de cultivos nas fezes, pré- e pós-tratamentos. Não se obteve a eficácia desejada com o emprego de avermectinas de longa ação - com alta concentração e em associação - ou mesmo, com a aplicação de superdoses. Os gêneros Cooperia spp., Haemonchus spp. e Trichostrongylus spp. foram resistentes às avermectinas, e Ostertagia spp. à ivermectina. Observou-se que, uma vez estabelecida a resistência parasitária a LMs a 1 por cento, a aplicação de fármacos, deste mesmo grupo químico, ainda que em formulações mais concentradas, asso-ciações ou superdoses, pode não resultar na eficácia esperada.
The macrocyclic lactones (MLs) (avermectins and milbemycins) are endectocides broadly used in livestock and in some parasitic diseases of humans. In cattle, parasite resistance to MLs is emerging, and the appearance of formulations that differ in their pharmacological properties become complex the choice of the most appropriate drug to each case. In order to evaluate possible alternatives to restore the effectiveness of MLs on resistant strains of gastrointestinal nematodes, were tested, in this study, ten different treatments based on the MLs on a population of gastrointestinal nematodes of cattle which, known, was under pressure of selection by 1 percent avermectins. Additionally, was tested a benzimidazole. The efficacy of the drugs was calculated with basis on the reduction of eggs per gram of feces (EPG) of cattle. The resistance of each genus was evaluated by identification of the larvae, obtained from culture in the feces, pre- and post-treatments. The desired efficacy was not obtained using long action avermectins - with high concentration and in association - even with the application of high doses. The genera Cooperia spp., Haemonchus spp. and Trichostrongylus spp. were resistant to avermectins, and Ostertagia spp. to ivermectin. It was observed that, once established parasite resistance to the 1 percent MLs, the application of drugs, of this same chemical group, even in formulations of high concentration, association or in high doses, may not result in the expected efficacy.
