RESUMO

A population pharmacokinetic model was developed to describe alterations in ceftaroline brain disposition caused by meningitis in healthy and methicillin-resistant Staphylococcus aureus (MRSA)-infected rats. Blood and brain microdialysate samples were obtained after a single bolus dose of ceftaroline fosamil (20 mg/kg) administered intravenously. Plasma data were modeled as one compartment, and brain data were added to the model as a second compartment, with bidirectional drug transport between plasma and brain (Qin and Qout). The cardiac output (CO) of the animals showed a significant correlation with the relative recovery (RR) of plasma microdialysis probes, with animals with greater CO presenting lower RR values. The Qin was approximately 60% higher in infected animals, leading to greater brain exposure to ceftaroline. Ceftaroline brain penetration was influenced by MRSA infection, increasing from 17% (Qin/Qout) in healthy animals to 27% in infected animals. Simulations of a 2-h intravenous infusion of 50 mg/kg every 8 h achieved >90% probability of target attainment (PTA) in plasma and brain for the modal MRSA MIC (0.25 mg/L), suggesting that the drug should be considered an option for treating central nervous system infections.

Ceftaroline, approved to treat skin infections and pneumonia due to methicillin-resistant Staphylococcus aureus (MRSA), has been considered for the treatment of central nervous system (CNS) infections. A population pharmacokinetic (popPK) model was developed to describe ceftaroline soft tissue and cerebrospinal fluid (CSF) distributions and investigate the probability of target attainment (PTA) of the percentage of the dosing interval that the unbound drug concentration exceeded the MIC (%fT>MIC) to treat MRSA infections. Healthy subjects' plasma and microdialysate concentrations from muscle and subcutaneous tissue following 600 mg every 12 h (q12h) and q8h and neurosurgical patients' plasma and CSF concentrations following single 600-mg dosing were used. Plasma concentrations were described by a two-compartment model, and tissue concentrations were incorporated as three independent compartments linked to the central compartment by bidirectional transport (clearance in [CLin] and CLout). Apparent volumes were fixed to physiological interstitial values. Healthy status and body weight were identified as covariates for the volume of the central compartment, and creatinine clearance was identified for clearance. The CSF glucose concentration (GLUC) was inversely correlated with CLin,CSF. Simulations showed a PTA of >90% in plasma and soft tissues for both regimens assuming an MIC of 1 mg/L and a %fT>MIC of 28.8%. Using the same target, patients with inflamed meninges (0.5 < GLUC ≤ 2 mmol/L) would reach PTAs of 99.8% and 97.2% for 600 mg q8h and q12h, respectively. For brain infection with mild inflammation (2 < GLUC ≤ 3.5 mmol/L), the PTAs would be reduced to 34.3% and 9.1%, respectively. Ceftaroline's penetration enhanced by meningeal inflammation suggests that the drug could be a candidate to treat MRSA CNS infections.

Biofilms and infectious process may alter free antimicrobial concentrations at the site of infection. Tobramycin (TOB), an aminoglycoside used to treat lung infections caused by Pseudomonas aeruginosa, binds to alginate present in biofilm extracellular matrix increasing its minimum inhibitory concentration (MIC). This work aimed to investigate the impact of biofilm-forming P. aeruginosa infection on TOB lung and epithelial lining fluid (ELF) penetration, using microdialysis, and to develop a population pharmacokinetic (popPK) model to evaluate the probability of therapeutic target attainment of current dosing regimens employed in fibrocystic and non-fibrocystic patients. The popPK model developed has three compartments including the lung. The ELF concentrations were described by a penetration factor derived from the lung compartment. Infection was a covariate in lung volume (V3) and only chronic infection was a covariate in central volume (V1) and total clearance (CL). Simulations of the recommended treatments for acute and chronic infection achieved >90% probability of target attainment (PTA) in the lung with 4.5 mg/kg q24h and 11 mg/kg q24h, respectively, for the most prevalent P. aeruginosa MIC (0.5 mg/mL). The popPK model was successfully applied to evaluate the PTA of current TOB dosing regimens used in the clinic, indicating the need to investigate alternative posology.

Bacterial and fungal infections are challenging due to their low susceptibility and resistance to antimicrobial drugs. For this reason, antimicrobial peptides (AMP) emerge as excellent alternatives to overcome these problems. At the same time, their active insertion into the cell wall of microorganisms can be availed for biorecognition applications in biosensing platforms. Temporin-PTA (T-PTA) is an AMP found in the skin secretions of the Malaysian fire frog Hylarana picturata, which presents antibacterial activity against MRSA, Escherichia coli, and Bacillus subtilis. In this work, T-PTA was explored as an innovative sensing layer aiming for the electrochemical differentiation of Klebsiella pneumoniae, Acinetobacter baumannii, Bacillus subtilis, Enterococcus faecalis, Candida albicans, and C. tropicalis based on the structural differences of their membranes. The biosensor was analyzed through electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). In this approach, the different structural features of each microorganism resulted in different adherence degrees and, therefore, different electrochemical responses. The transducing layer was fabricated by the self-assembling of a 4-mercaptobenzoic acid (MBA) monolayer and gold-capped magnetic nanoparticles (Fe3O4@Au) implemented to improve the electrical signal of the biointeraction. We found that each interaction, expressed in variations of electron transfer resistance and anodic peak current, demonstrated a singular response from which the platform can discriminate all different microorganisms. We found expressive sensitivity towards Gram-negative species, especially K. pneumoniae. A detection limit of 101 CFU.mL-1 and a linear range of 101 to 105 CFU.mL-1 were obtained. The T-PTA biosensor platform is a promising and effective tool for microbial identification.

RESUMO

This paper aims to make a proposal to govern the Peruvian State under the umbrella of management cybernetics, following the paths of the viable system model (VSM), proposed by Prof. Stafford Beer, enriched with other soft and hard systemic methodologies and technologies, to cover the soft and hard issues that are part of the complex Peruvian reality at different levels of recursion. For doing this, four defined perspectives were adopted to understand the complexity of Peru: the sectoral view, the regions view, the river basins view and the macroregions view. Peru is seen as a system in focus, defining, for each of these four perspectives, the five systems that VSM has. The application of the VSM in each perspective serves to apply it in two modes: diagnosis and design, according to the respective perspective. Then an integrative analysis and reflection is done considering the four perspectives, to analyze the viability of the VSM approach in the governance of the Peruvian State to establish some conclusions and recommendations in relation to the proposal, appearing at the end of the paper.

A new RuII-ReII complex salt, µ-cyanido-κ2 C:N-bis-[(η5-cyclo-penta-dien-yl)bis(3,5,7-tri-aza-phosphaadamantane-κP)ruthenium(II)] tetra-bromido-(ethanol/methanol-κO)nitrosylrhenate(II), [Ru(CN)(C5H5)2(C6H12N3P)4][ReBr4(NO)(CH4O)0.5(C2H6O)0.5] or [RuCp(PTA)2-µ-CN-1κC:2κ2 N-RuCp(PTA)2][Re(NO)Br4(EtOH)0.5(MeOH)0.5] (PTA = 3,5,7-tri-aza-phosphaadaman-tane) was obtained and characterized by single-crystal X-ray diffraction, elemental analysis and infrared spectroscopy. The title salt was obtained by liquid-liquid diffusion of methanol/DMSO solutions of (NBu4)[Re(NO)Br4(EtOH)] and [(PTA)2CpRu-µ-CN-1κC:2κ2 N-RuCp(PTA)2](CF3SO3). The RuII and ReII independent moieties correspond to a binuclear and mononuclear complex ion, respectively. A deep geometrical parameter analysis was performed, and no significant differences were found with earlier reports containing similar mol-ecules. The magnetic properties were investigated in the temperature range 2.0-300 K, and the complex behaves as a quasi-magnetically isolated spin doublet with weak anti-ferromagnetic inter-actions.

Records of 234 lactations from F1 Holstein x Gyr daughters of proven bulls were used in this study for the evaluation of productive and reproductive characteristics of cows. Data were collected from a private farm located in the municipality of Santo Antônio do Monte, MG, Brazil. The objective of this study was to evaluate the effects of Holstein bull's PTA milk (predicted transmitting ability) calving year, season, and parity on the productive and reproductive characteristics of F! cows. The productive characteristics analyzed were total milk production (TMP), average daily production (ADP), days in milk (DIM), and milk yield per day of calving interval (MYDCI). The studied reproductive characteristics were service interval (SI), calving interval (CI), and gestation length (GL). The results were subjected to statistical analysis and means for significant effects were compared using Duncan's test at a significance level of 5%. F1 animals efficiently expressed their genetic potential for milk production with an average total milk production of 7842.61 kg and average daily production of 24.53 kg. Means for days in milk, milk yield per day of calving interval, service interval, calving interval, and gestation length were 322.52 days, 20.73 kg, 106.0 days, 383.23 days, and 276.18 days, respectively. Properly managed F1 Holstein x Gyr daughters of proven bulls fed a balanced diet efficiently expressed their genetic potential for milk production without adverse effects on reproduction. Moreover, Holstein x Gyr cows should be selected based on parents' PTA for milk traits and other factors such as environmental conditions and management.

Amazonia harbors the largest and most diverse tropical forest in the world, but knowledge about the species diversity of the region is still far from ideal. Given this low level of faunal and floral knowledge, we present an annotated list of the species of amphibians and reptiles found in Floresta Nacional do Pau-Rosa (FNPR), along the Rio Paraconi, municipality of Maués, state of Amazonas, Brazil. Herpetofauna of the FNPR was sampled with pitfall traps, active search and occasional encounters from February 18 to March 28, 2009. A total of 270 specimens were collected, representing 39 species of amphibians and 24 species of reptiles. At least seven of the species collected at FNPR represented, at the time, unnamed taxa (four of which have now been named). The number of taxa collected and the high number of unnamed taxa highlight the importance of this area in terms of biodiversity and as a priority for conservation. We also discuss about the amphibian diversity in Amazonia.(AU)


A Amazônia engloba a maior e mais diversa floresta tropical do mundo, mas o conhecimento sobre a diversidade de espécies da região ainda está longe do ideal. Apresentamos aqui uma lista comentada das espécies de anfíbios e répteis encontradas na Floresta Nacional de Pau-Rosa (FNPR), ao longo do Rio Paraconi, município de Maués, estado do Amazonas, Brasil. Espécimes da FNPR foram coletados com armadilhas de queda, busca ativa e encontros ocasionais de 18 fevereiro a 28 março de 2009. Um total de 270 espécimes foram coletados, o que representou 39 espécies de anfíbios e 24 espécies de répteis. Pelo menos sete das espécies coletadas na FNPR representaram, na época, táxons sem nome (quatro dos quais já foram nomeados). O número de exemplares recolhidos e o elevado número de táxons destaca a importância desta área em termos de biodiversidade e como uma área prioritária para a conservação. Nós também discutimos a diversidade de anfíbios na Amazônia.(AU)

Estudos divulgados recentemente, alguns com até 5 décadas de observações, concluíram com alto grau de evidências, que a exposição involuntária e sistemática à fumaça do cigarro tem implicações genéticas em gerações. Vai muito além do risco do "exposto primário". Ficaram evidentes riscos de doenças tabaco relacionadas em descendentes até a 3ª. geração, isto é, de avós para netos, mesmo que estes não sejam expostos à Poluição Tabagística Ambiental (PTA). As leis que controlam o tabagismo em locais públicos ou privados com circulação de pessoas trouxeram grandes benefícios para toda a coletividade, porém, no ambiente domiciliar, no interior do carro ou a exposição de "gestante/feto" a realidade é outra

It goes far beyond the risk of the "primary exposed". There have been evident risks of tobacco-related diseases in descendants to the third generation, even if they had never been exposed to "Environmental Tobacco Pollution". Laws that control smoking in public or private places, with circulation of people, have brought great benefits to the whole community, However, regarding places such as the home environment, in the car or the exposure of fetus during pregnancy, have shown different results

This review outlines risk factors of post-laser in situ keratomileusis (LASIK) ectasia that can be detected preoperatively and presents a new metric to be considered in the detection of ectasia risk. Relevant factors in refractive surgery screening include the analysis of intrinsic biomechanical properties (information obtained from corneal topography/tomography and patient's age), as well as the analysis of alterable biomechanical properties (information obtained from the amount of tissue altered by surgery and the remaining load-bearing tissue). Corneal topography patterns of placido disk seem to play a pivotal role as a surrogate of corneal strength, and abnormal corneal topography remains to be the most important identifiable risk factor for ectasia. Information derived from tomography, such as pachymetric and epithelial maps as well as computational strategies, to help in the detection of keratoconus is additional and relevant. High percentage of tissue altered (PTA) is the most robust risk factor for ectasia after LASIK in patients with normal preoperative corneal topography. Compared to specific residual stromal bed (RSB) or central corneal thickness values, percentage of tissue altered likely provides a more individualized measure of biomechanical alteration because it considers the relationship between thickness, tissue altered through ablation and flap creation, and ultimate RSB thickness. Other recognized risk factors include low RSB, thin cornea, and high myopia. Age is also a very important risk factor and still remains as one of the most overlooked ones. A comprehensive screening approach with the Ectasia Risk Score System, which evaluates multiple risk factors simultaneously, is also a helpful tool in the screening strategy.

They were evaluated the effects of order and season of calving, of Gir and Holstein sires grandfathers and fathers of the1/2 Holstein x Nelogir matrices, respectively. Total milk production, with average lactation length of 288 days, was 3,426.9kg of milk. The order of calving influenced the daily average production which was higher in the second calving and lactation length was superior in the first calving. The beginning of rain period (November to January) showed lower lactation length and production a day of calving interval. Total milk production did not vary according to Gir sire, Nelogir’s father, and was higher in the matrices daughters of Holstein sires with positive PTA for milk. In the first calving the production peak was of 13.03kg at 45 days of lactation, and from the second calving the production was descending one. In tropical conditions, productive and reproductive traits support the initiative of farm animals 1/2 Holstein x Nelogir from matrices composed Nelogir and that the choice of Holstein bull to produce F1 HNG should be based on the genetic value milk production (milk PTA).

Avaliou-se os efeitos de ordem e época de parto, do reprodutor da raça Gir e do reprodutor da raça Holandesa, avô materno e pais das matrizes 1/2 Holandês x Nelogir, respectivamente. A produção de leite total, com duração da lactação média de 288 dias, foi de 3.426,9kg de leite. A ordem de parto influenciou a produção média diária que foi maior no segundo parto e a duração da lactação, superior nos animais de primeiro parto. O período de início das águas apresentou menor duração da lactação e produção por dia de intervalo de parto. Conclui-se que, em condições tropicais, as características produtivas e reprodutivas sustentam a iniciativa de produção de animais 1/2 Holandês x Nelogir a partir de matrizes compostas Nelogir e que a escolha do touro da raça Holandesa para produção de F1 HNG deve ser baseada no valor genético aditivo para produção de leite (PTA leite).

In the first calving the production peak was of 13.03kg at 45 days of lactation, and from the second calving the production was descending one. In tropical conditions, productive and reproductive traits support the initiative of farm animals 1/2 Holstein x Nelogir from matrices composed Nelogir and that the choice of Holstein bull to produce F1 HNG should be based on the genetic value milk production (milk PTA).


ResumoObjetivou-se avaliar as características produtivas e reprodutivas de fêmeas 1/2 Holandês x » Nelore, » Gir (Nelogir). Avaliou-se os efeitos de ordem e época de parto, do reprodutor da raça Gir e do reprodutor da raça Holandesa, avô materno e pais das matrizes 1/2 Holandês x Nelogir, respectivamente. A produção de leite total, com duração da lactação média de 288 dias, foi de 3.426,9kg de leite. A ordem de parto influenciou a produção média diária que foi maior no segundo parto e a duração da lactação, superior nos animais de primeiro parto. O período de início das águas apresentou menor duração da lactação e produção por dia de intervalo de parto. A produção de leite total não variou de acordo com o reprodutor Gir, pai da Nelogir, e foi superior nas matrizes filhas de reprodutores Holandeses com PTA positivo para leite. Ao primeiro parto o pico de produção foi de 13,03kg aos 45 dias de lactação, e a partir do segundo parto a produção foi decrescente desde o parto. Conclui-se que, em condições tropicais, as características produtivas e reprodutivas sustentam a iniciativa de produção de animais 1/2 Holandês x Nelogir a partir de matrizes compostas Nelogir e que a escolha do touro da raça Holandesa para produção de F1 HNG deve ser baseada no valor genético aditivo para produção de leite (PTA leite).

The species snakes Boa constrictor (red-tailed boa) is representatives of the family Boidae. Boa constrictors feed on small or medium mammals, birds and even lizards, and their food is directly related to biochemical differences of the animal. This aim of this review is address data that highlight the importance of understanding the biochemical profile of wild species, aiming reviews for possible treatment and breed in captivity, as well as rehabilitate wild animals. Among the laboratorial exams, serum biochemical profiles reflect cellular integrity and organ function. These patterns reflect both the leakage of cellular constituents to the serum, and regulation of damaged absorption, production or excretion of various serum components. In this context, the study of hematology it is essential to determine the health status of reptiles.


As serpentes da espécie Boa constrictor (jiboias) são representantes da família Boidae. As jiboias alimentam-se de mamíferos pequenos a médios, aves e até mesmo lagartos e sua alimentação está diretamente relacionada com diferenças nos valores bioquímicos do animal. Esta revisão teve por objetivo abordar dados que destacam a relevância de se conhecer o perfil bioquímico de espécies silvestres, visando possíveis avaliações para tratamento e criação destes animais em cativeiro, assim como reabilitar animais de vida livre. Dentre os exames laboratoriais, os perfis bioquímicos séricos refletem a integridade celular e a função orgânica. Esses padrões refletem tanto o extravasamento de constituintes celulares para o soro, quanto à regulação prejudicada da absorção, produção ou excreção dos vários componentes séricos. Neste contexto, o estudo da hematologia torna se fundamental para a determinação do estado de saúde dos répteis.

The flagellar cytoskeleton of Leishmania promastigotes contains the canonical 9+2 microtubular axoneme and a filamentous structure, the paraflagellar rod (PFR), which is present alongside the axoneme. In contrast to promastigotes, which contain a long and motile flagellum, the amastigote form of Leishmania displays a short flagellum without a PFR that is limited to the flagellar pocket domain. Here, we investigated the biogenesis of the Leishmania flagellum at 0, 4, 6 and 24h of differentiation. Light and electron microscopy observations of the early stages of L. amazonensis differentiation showed that the intermediate forms presented a short and wider flagellum that did not contain a PFR and presented reduced motion. 3D-reconstruction analysis of electron tomograms revealed the presence of vesicles and electron-dense aggregates at the tip of the short flagellum. In the course of differentiation, cells were able to adhere and proliferate with a doubling time of about 6h. The new flagellum emerged from the flagellar pocket around 4h after initiation of cell cycle. Close contact between the flagellar membrane and the flagellar pocket membrane was evident in the intermediate forms. At a later stage of differentiation, intermediate cells exhibited a longer flagellum (shorter than in promastigotes) that contained a PFR and electron dense aggregates in the flagellar matrix. In some cells, PFR profiles were observed inside the flagellar pocket. Taken together, these data contribute to the understanding of flagellum biogenesis and organisation during L. amazonensis differentiation.

Climatic efficiency (EC) is an index that shows the yield loss caused by water deficiency. The objective of this study was to evaluate the EC of soybean and wheat crops for the state of Rio Grande do Sul, Brazil. The EC was obtained by the ratio between the attainable yield (PA) and potential yield (PPf) of these crops, estimated by crop simulation models. The PA was obtained by estimating the potential yield (PPf), which was determined by the FAO Agroecological Zone method, depreciated by the water deficit in each crop phenological phase for each sowing date, between 1979 and 2008. The results showed that sowing dates influenced the EC for the soybean crop, ranging from 0.31 to 0.61. Based on the results from EC and PA, there is an increase of crop yield when sowings are delayed from 01/10 to 21/12. For the wheat crop, the average EC was greater than 0.81, which allows to conclude that the best sowing dates should be chosen considering the highest PPf. The highest potential yields for the wheat crop were obtained for sowings from mid-June to end July.


Eficiência climática (EC) é um índice que demonstra em termos quantitativos a redução produtiva ocasionada pelo déficit hídrico. Assim, este trabalho teve como objetivo avaliar a EC das culturas da soja e do trigo para o estado do Rio Grande do Sul, considerando-se diferentes locais e datas de semeaduras. A EC foi obtida pela relação entre as produtividades atingível (PA) e potencial (PPf), estimadas por meio de modelos de simulação. A PA foi obtida pela estimativa da PPf, a qual foi determinada pelo método de Zona Agroecológica da FAO, deflacionada pelo déficit hídrico em cada uma das fases das culturas para cada data de semeadura, entre os anos de 1979 e 2008. Os resultados evidenciaram que as datas de semeadura influenciam a EC para a cultura da soja, a qual variou de 0,31 a 0,61. Com base nos resultados de EC e PA, há um ganho de produtividade com o atraso da semeadura de 01/10 para 21/12. Para a cultura do trigo, a EC média foi superior a 0,81, o que leva a concluir que se deve optar por datas de semeadura em que há maior PPf. As maiores PPf para o trigo são obtidas nas semeaduras a partir de meados do mês de junho ao final do mês de julho.

This study evaluated the effects of isotonic electrolyte solutions administered via small-bore nasogastric intubation on a continuous flow rate on the hematocrit, total protein, albumin, sodium, potassium, chloride, total magnesium, urea, creatinine, glucose and lactate in cattle experimentally dehydrated. Six crossbred adult cattle, two males and four females were used for the study. Enteral fluid therapy was performed by using 15mL kg-1 h-1 for 12 hours. The animals were divided in two treatments in a "crossover" experimental design and subjected to the following treatments: SEGlic: 4g NaCl, 1g KCl, 4g NaHCO3, 5g dextrose in 1.000mL of water; SEProp: 8g NaCl, 0.5g KCl, 0.5g CaCl2, 15mL propylene glycol in 1.000mL of water. Both treatments demonstrated expansion of blood volume, reduction of hematocrit and total protein. In the SEGlic treatment, hypernatremia and increase of lactate were observed. On the other hand, the SEProp treatment caused an increase in the level of plasma glucose, while the other parameters remained within the reference values.

Seis bovinos adultos, mestiços, dois machos e quatro fêmeas foram avaliados. A hidratação enteral foi administrada na dose de 15mL kg-1 h-1 durante 12 horas. Os animais foram separados em dois tratamentos num delineamento experimental crossover e submetidos a cada um dos seguintes tratamentos: SEGlic: NaCl 4g, KCl 1g, NaHCO3 4g, dextrose 5g em 1.000mL de água; SEProp: NaCl 8g, KCl 0,5g, CaCl2 0,5g, propilenoglicol 15mL em 1.000mL de água. Após a administração das soluções eletrolíticas, ocorreu expansão da volemia, em ambos os tratamentos, ocasionando diminuição do hematócrito e proteína total. A SEGli ocasionou o aparecimento de hipernatremia e aumento do lactato plasmático. Por sua vez, a SEProp aumentou a taxa de glicose plasmática, enquanto os demais parâmetros avaliados permaneceram na faixa de referência.
