Morganella morganii is a bacterium belonging to the normal intestinal microbiota and the environment; however, in immunocompromised individuals, this bacterium can become an opportunistic pathogen, causing a series of diseases, both in hospitals and in the community, being urinary tract infections more prevalent. Therefore, the objective of this study was to evaluate the prevalence, virulence profile, and resistance to antimicrobials and the clonal relationship of isolates of urinary tract infections (UTI) caused by M. morganii, both in the hospital environment and in the community of the municipality of Londrina-PR, in southern Brazil, in order to better understand the mechanisms for the establishment of the disease caused by this bacterium. Our study showed that M. morganii presents a variety of virulence factors in the studied isolates. Hospital strains showed a higher prevalence for the virulence genes zapA, iutA, and fimH, while community strains showed a higher prevalence for the ireA and iutA genes. Hospital isolates showed greater resistance compared to community isolates, as well as a higher prevalence of multidrug-resistant (MDR) and extended-spectrum beta lactamase (ESBL)-producing isolates. Several M. morganii isolates from both sources showed high genetic similarity. The most prevalent plasmid incompatibility groups detected were FIB and I1, regardless of the isolation source. Thus, M. morganii isolates can accumulate virulence factors and antimicrobial resistance, making them a neglected opportunistic pathogen.

Mortality of chicken embryos and first-week chickens was reported in a commercial incubator company in Costa Rica. Six 1-day-old Cobb chickens and twenty-four embryonated chicken eggs were examined in the Laboratory of Avian Pathology and the Laboratory of Bacteriology of the National University of Costa Rica. Twelve dead-in-shell embryos showed maceration and were immersed in a putrid, turbid, slightly thick brown liquid. Additionally, the other 12 embryonated eggs had milky yellow-orange content. The livers of those embryos had congestion, haemorrhages and multifocal cream foci of necrosis. Granulocytic infiltration was observed in the bursa of Fabricius, myocardium, liver, lung and kidney. Livers and egg yolks from six embryonated chickens and all 1-day-old chickens were aseptically collected and cultured. In addition, tissues from six better conserved embryos and all 1-day-old chickens were fixed in buffered formalin and embedded in paraffin. Biochemical and molecular tests identified Comamonas testosteroni as the cause of the early, middle and late embryo mortality. As all the eggshells from the sampled embryonated eggs were dirty with soiled a fecal matter, contamination after manipulating the eggs was considered the source of infection. C. testosteroni is an environmental microorganism that has rarely been reported to cause human disease. To our knowledge, this is the first report of C. testosteroni causing mortality in a hatchery. Cleaning and disinfection using ozone were implemented in the hatchery to eliminate the embryo mortality associated with C. testosteroni.

Acinetobacter species have the potential to invade and colonize immunocompromised patients, therefore being well-known as opportunistic pathogens. Among these bacteria, the species of the Acinetobacter calcoaceticus-Acinetobacter baumannii "complex" (Acb members) emerge as the main often isolated bacteria in clinical specimens. The unequivocal taxonomy is crucial to correctly identify these species and associated with comparative genomic analyses aids to understand their life-styles as well. In this study, all publicly available Acinetobacter species at the date of this study preparation were analyzed. The results revealed that the Acb members are in fact a complex when phenotypic methods are confronted, while for comparative and phylogenomics analyses this term is misleading, since they composed a monophyletic group instead. Nine best gene markers (response regulator, recJ, recG, phosphomannomutase, pepSY, monovalent cation/H + antiporter subunit D, mnmE, glnE, and bamA) were selected for identification of Acinetobacter species. Moreover, representative strains of each species were split according their isolation sources in the categories: environmental, human, insect and non-human vertebrate. Neither niche-specific genome signature nor niche-associated functional and pathogenic potential were associated with their isolation source, meaning it is not the main force acting on Acinetobacter adaptation in a given niche and corroborating that their ubiquitous distribution is a reflex of their generalist life-styles.

Pregnancy outcomes and women's health are directly affected by vaginal microbiota. This microbiota consists of a dynamic ecosystem of various microbes in different ratios, which in healthy conditions protect the vaginal epithelium from infections. However, cases of vaginal infection are regularly diagnosed in women of reproductive age, contributing to more severe outcomes. Therefore, our main goal was to determine the prevalence of bacterial vaginosis (BV), aerobic vaginitis (AV), and vulvovaginal candidiasis (VVC) among Ecuadorian pregnant and non-pregnant women. A cross-sectional study was conducted among 217 women between 13 and 40 years old seeking primary healthcare in Carlos Andrade Marin Hospital (HCAM), Gynecological-Obstetric Hospital Isidro Ayora (HGOIA) and Center for Teaching Health Cipriana Dueñas during October 2018 to February 2019. The classical characterization of the vaginal microbiota was performed through microscopy by the Nugent criteria to evaluate the presence of BV, healthy and intermediate microbiota, by the criteria of Donders to determine the presence of AV and by the Marot-Leblond criteria to diagnose VVC. DNA extraction from vaginal samples and Polymerase Chain Reaction (PCR) analysis was performed to characterize the presence of Gardnerella spp., Mobiluncus mulieris, Escherichia coli, Enterococcus spp., and Lactobacillus spp. Finally, quantification of the lactobacilli was performed by quantitative real-time PCR (qPCR) for samples from women with normal vaginal microbiota and women with AV. Our results showed 52% of women with healthy microbiota, 7% with intermediate microbiota, and 41% with vaginal dysbiosis, comprising 27% with AV, 8% with BV and 4% with VVC and 2% with co-infections or co-dysbiosis. Additionally, a higher amount of lactobacilli were found in pregnant women when compared to non-pregnant women, while AV cases were characterized by a significant drop of Lactobacillus spp., more precisely, between 1E3 and 1E5 colony forming units (CFU)/ml. Finally, women with normal vaginal microbiota showed an average load of lactobacilli between 1E6 and 1E7 CFU/ml. This pilot study showed no statistically significant differences between pregnant and non-pregnant women, pointing to the possibility to use lactobacilli quantification for the prevention of future vaginal infections.

Acinetobacter haemolyticus is a Gammaproteobacterium that has been involved in serious diseases frequently linked to the nosocomial environment. Most of the strains causing such infections are sensitive to a wide variety of antibiotics, but recent reports indicate that this pathogen is acquiring very efficiently carbapenem-resistance determinants like the blaNDM-1 gene, all over the world. With this work we contribute with a collection set of 31 newly sequenced nosocomial A. haemolyticus isolates. Genome analysis of these sequences and others collected from RefSeq indicates that their chromosomes are organized in 12 syntenic blocks that contain most of the core genome genes. These blocks are separated by hypervariable regions that are rich in unique gene families, but also have signals of horizontal gene transfer. Genes involved in virulence or encoding different secretion systems are located inside syntenic regions and have recombination signals. The relative order of the synthetic blocks along the A. haemolyticus chromosome can change, indicating that they have been subject to several kinds of inversions. Genomes of this microorganism show large differences in gene content even if they are in the same clade. Here we also show that A. haemolyticus has an open pan-genome.

El complejo Burkholderia cepacia está formado por 22 especies conocidas como patógenos oportunistas en personas inmunocomprometidas, especialmente en aquellas con fibrosis quística. También se aíslan de infecciones nosocomiales y son difíciles de erradicar debido a su capacidad intrínseca para resistir una gran variedad de antibióticos. En general, estas especies presentan genomas de gran tamaño (hasta 9 Mpb) divididos en 2-5 replicones. Esta característica aporta una gran versatilidad metabólica, que se considera importante para habitar el suelo, el agua, las plantas, incluso los nódulos en leguminosas. Algunas especies del complejo B. cepacia exhiben actividades benéficas, como biorremediación, biocontrol y promoción del crecimiento vegetal. No obstante, debido a su papel en infecciones de humanos, su uso en la agricultura está restringido. El complejo B. cepacia es un tema constante de estudio debido a su impacto en el sector salud y su potencial en la agricultura. En este trabajo se examina la historia del complejo B. cepacia y se revisa la información reciente relacionada con este grupo de bacterias.

Mycobacterium simiae (Karassova V, Weissfeiler J, Kraszanay E, Acta Microbiol Acad Sci Hung 12:275-82, 1965) is a slow-growing nontuberculous Mycobacterium species found in environmental niches, and recently evidenced as an opportunistic Human pathogen. We report here the genome of a clinical isolate of M. simiae (MsiGto) obtained from a patient in Guanajuato, Mexico. With a size of 6,684,413 bp, the genomic sequence of strain MsiGto is the largest of the three M. simiae genomes reported to date. Gene prediction revealed 6409 CDSs in total, including 6354 protein-coding genes and 52 RNA genes. Comparative genomic analysis identified shared features between strain MsiGto and the other two reported M. simiae genomes, as well as unique genes. Our data reveals that M. simiae MsiGto harbors virulence-related genes, such as arcD, ESAT-6, and those belonging to the antigen 85 complex and mce clusters, which may explain its successful transition to the human host. We expect the genome information of strain MsiGto will provide a better understanding of infective mechanisms and virulence of this emergent pathogen.

Las especies del género Aspergillus Link constituyen importantes agentes biodeteriorantes de objetos de valor constructivo, cultural y patrimonial. En ambientes laborales, sus propágulos y metabolitos pueden provocar afecciones a la salud humana. Los estudios aeromicológicos pueden contribuir a minimizar el impacto de Aspergillus spp. en el biodeterioro y la calidad de vida. Se han caracterizado de acuerdo a su potencial biodeterió- geno y patogénico representantes de Aspergillus spp. en el aire de depósitos de materiales en tres instituciones patrimoniales cubanas. Las muestras se tomaron empleando un biocolector SAS súper 100. Se evaluó cualitativamente la actividad celulolítica, amilolítica y proteolítica de los aislados; así como la excreción de pigmentos y ácidos orgánicos. Se evaluó el crecimiento a 37°C y la secreción de hemolisinas. Aspergillus spp. constituyó parte considerable de la micobiota aérea viable en los depósitos con 22 especies correspondientes a 12 secciones, las de mayor variabilidad fueron Flavi y Restricti, mientras la primera y la sección Aspergillus fueron las mejor representadas. El 74 por ciento de las cepas mostraron alta capacidad para degradar los compuestos orgánicos evaluados y secretar ácidos, mientras el 34 por ciento excretó pigmentos de amarillo a pardo oscuro, se evidenció un amplio potencial biodeteriorante en general. Más del 30 por ciento de los aislados mostraron ser patógenos oportunistas peligrosos para el hombre al crecer a 37°C, secretar hemolisinas, y presentar un tamaño conidial que posibilita la depositación en alvéolos pulmonares. Las cepas con mayor impacto biodeteriorante y patogénico fueron las correspondientes a las Secciones Flavi, Nigri y la especie Aspergillus fumigatus Fresen.
Species of the genus Aspergillus Link cause damage to objects constructive, cultural and heritage value. In work environments, their propagules and metabolites may cause disorders to human health. Aerobiological studies can help minimize the impact of Aspergillus spp. in the biodeterioration and quality of life. Aspergillus spp. strains present in the air of the repositories in three Cuban heritage institutions were characterized according to their biodeteriogenic and pathogenic potential. Each isolated was classified taxonomically, cellulolytic, amylolytic and proteolytic activity as well as excretion of pigments and organic acids was evaluated. The capacity to grow at 37°C and secrete hemolysins was qualitatively evaluated. Aspergillus spp. was significant portion of viable air micobiota in the three repositories with 22 species corresponding to 12 sections, most variability were Flavi and Restricti, while the first section and Aspergillus were the best represented. The 74 percent of the strains showed high ability to degrade organic compounds evaluated and secrete acids, while 34 percent excreted pigments from yellow to dark brown, evidenced a broad biodeteriogenic potential in general. More than 30 percent of the isolates were shown to be dangerous opportunistic pathogens for humans to grow at 37°C, secrete hemolysin, and present a conidial size which enables the deposition pulmonary alveoli. Strains higher biodeteriogenic and pathogenic impact were those corresponding to the Flavi, Nigri sections and the species Aspergillus fumigatus Fresen.

Desde 1981 a aids tem estado presente em nossa sociedade. Como consequência, doenças infectocontagiosas e novos patógenos oportunistas têm desenvolvido novos comportamentos e adquirido novas virulências. Mesmo com a aquisição de novas tecnologias como técnicas imunológicas, ressonância magnética (RM), tomografia computadorizada (TC), tomografia por emissão de pósitron (PET) e espectrometria , existem desafios para o diagnóstico que incluem falsos negativos e positivos. Reportamos nossa experiência com paciente imunocomprometido e suas múltiplas infecções oportunistas, e juntamos casos com diferentes patógenos que mimetizam tumores de estirpe glial com resultados falsos positivos e negativos nas provas imunológicas, seja por comprometimento imunológico do hospedeiro, seja por provas cruzadas de colagenoses.
Since 1981 AIDS has been present in our society. Following it, controlled infection diseases and new opportunistic pathogen have raised different behavior and virulence. Even with the important technical advance such as immunologic assay, magnetic resonance (MRI), computed tomography CT, positron emission tomography (PET) and spectrometry there still exists a real challenge for diagnosis, including high levels of false negatives. We report in this work our experience with imunocompromised patients and their multiple opportunistic diseases and attach it different pathogens that may mimic tumoral glial diseases with false positive and negatives results in immunological assay, developed by host imunocompromised reactivity or by cross reaction of collagenosis desease.

The objectives of this study were to assess the effect of the intake of aflatoxin on the development of tilapia and to evaluate the impact of inoculation with Aeromonas hydrophila on performance parameters, so these two individual tests were performed. One hundred and twenty fingerlings aged 35 days old, with mean weight of 1.55 ± 0.005 g and mean length of 5 cm were used in each test, distributed in 20 tanks. Each experimental unit consisted of a 60 L tank with six fingerlings. In the first experiment, increasing levels of aflatoxin (0.350, 0.757, 1.177 mg.kg feed-1) were used as treatments and, for the control group, a diet without aflatoxin was used in a completely randomized design with four treatments and five replicates. In the second experiment, a control group of Nile tilapia fingerlings was used and received a diet without aflatoxin, inoculated with saline solution (group 1) and Aeromonas hydrophila (group 2), as well as groups of animals fed on diets containing 0.350 mg.kg feed-1of aflatoxin (group 3), 0.757 mg.kg feed-1of aflatoxin (group 4) and 1.177 mg.kg feed-1of aflatoxin (group 5), and these groups were inoculated with Aeromonas hydrophila. The survival rate and the total length of fingerlings were influenced by the treatments (p 0.05). The synergistic action of aflatoxins and Aeromonas hydrophila was effective and caused the death of experimental fish, thus affecting feed conversion and length.
Os objetivos deste estudo foram analisar o consumo de aflatoxinas sobre o desenvolvimento de alevinos de tilápia e avaliar o efeito da inoculação de Aeromonas hydrophilasobre os parâmetros de desempenho produtivo dos animais. Para isso, dois testes individuais foram realizados. Foram utilizados 120 alevinos com 35 dias e 1,55 ± 0,005 g de peso médio em cada ensaio, distribuídos em 20 aquários. Cada unidade experimental foi constituída por um aquário de 60 L com seis alevinos. No primeiro ensaio, como tratamentos foram utilizados níveis crescentes de inclusão de aflatoxina (0,350; 0,757; 1,177 mg.kg ração-1) e um grupo controle com ração sem aflatoxina, em um delineamento inteiramente randomizado com quatro tratamentos e cinco repetições.
