The expansion of the agricultural frontier in the eastern llanos region of Colombia has endangered the moriche palm (Mauritia flexuosa) which has an important ecological function and provides various ecosystem services. In particular, the moriche that grows in this region is wild and has been little studied; therefore, there are no reports of its potential as a source of monounsaturated and polyunsaturated fatty acids, information that could be useful for the conservation of the species. This study performed a physicochemical characterization of the oil extracted from the dried pulp of moriche and identified the fatty acids present in the oil, pulp and peel, seed, and flour of this fruit from the Bita River Basin, Vichada, Colombia. The fatty acid composition was characterized by gas chromatography, including physicochemical tests of interest in the oil according to AOCS protocols. The results showed that the highest fatty acid content was found in the extracted oil, with a distribution of 81.64% unsaturated fat and 18.36% saturated fat. These fats included 79.20% oleic acid (omega-9), 0.26% palmitoleic acid (omega-7), 1.01% linoleic acid (omega-6), 1% linolenic acid (omega-3), 16.91% palmitic acid, and 1.33% stearic acid. We conclude that moriche from Bita Basin is an oleaginous fruit due to its high nutritional value in terms of unsaturated fatty acids and that both the flour and the oil obtained are bioproducts with potential industrial application.

We tested the hypothesis that administration of omega (ω)-9, ω-3, and ω-6 to mice can prevent oxidative alterations responsible for behavioral and cognitive alterations related with aging. Twenty-eight-day-old mice received skim milk (SM group), SM enriched with omega oil mixture (EM group), or water (control group) for 10 and 14 months, equivalent to middle age. Mice were evaluated for behavioral alterations related to depression and memory and oxidative status [brain levels of thiobarbituric acid reactive substances (TBARS), reduced glutathione (GSH), and myeloperoxidase (MPO)]. The 10-month EM group increased immobility time during the forced swimming test compared with control, indicating increased stress response. The 14-month SM- and EM-treated groups increased sucrose consumption compared with control, showing an expanded motivational state. The 14-month SM group decreased the number of rearings compared with the 14-month control and EM groups. The number of entries and time spent in the central square of the open field was higher in the 10-month EM group than in the control, revealing an anxiolytic-like behavior. TBARS decreased in the hippocampus and striatum of the 10-month EM group compared with the control. A similar decrease was observed in the striatum of the 10-month SM group. GSH levels were higher in all 14-month treated groups compared with 10-month groups. MPO activity was higher in the 14-month EM group compared with the 14-month control and SM groups, revealing a possible pro-inflammatory status. In conclusion, omega oils induced conflicting alterations in middle-aged mice, contributing to enhanced behavior and anxiolytic and expanded motivational state, but also to increased stress response and pro-inflammatory alterations.

The Mediterranean diet (MedDiet) consists of consumption of vegetables and healthy oils and have beneficial effects on metabolic and inflammatory diseases. Our goal here is to discuss the role of fatty acid content in MedDiet, mostly omega-3, omega-6, and omega-9 on malaria. Malaria affects millions of people around the globe. The parasite Plasmodium causes the disease. The metabolic and inflammatory alterations in the severe forms have damaging consequences to the host. The lipid content in the MedDiet holds anti-inflammatory and pro-resolutive features in the host and have detrimental effects on the Plasmodium. The lipids from the diet impact the balance of pro- and anti-inflammation, thus, lipids intake from the diet is critical to parasite elimination and host tissue damage caused by an immune response. Herein, we go into the cellular and molecular mechanisms and targets of the MedDiet fatty acids in the host and the parasite, reviewing potential benefits of the MedDiet, on inflammation, malaria infection progression, and clinical outcome.

The treatment of cutaneous inflammation with topical corticosteroids may cause adverse effects reinforcing the need for therapeutic alternatives to treat inflammatory skin disorders. We investigated the anti-inflammatory effect of oleic acid (OA), a fatty acid of the omega-9 (ω-9) family, and we point out it as an alternative to treat inflammatory skin disorders. OA was incorporated into Lanette®- or Pemulen® TR2-based semisolid preparations and the pH, spreadability, rheological behavior and in vivo anti-inflammatory performance in a UVB radiation-induced skin inflammation model in mice were assessed. The anti-inflammatory activity was verified after single or repeated treatment of the mouse ear following the UVB. The OA action on glucocorticoid receptors was investigated. Both semisolids presented pH values compatible with the deeper skin layers, appropriate spreadability factors, and non-Newtonian pseudoplastic rheological behavior. Pemulen® 3% OA inhibited ear edema with superior efficacy than Lanette® 3% OA and dexamethasone after a single treatment. Pemulen® 3% OA and dexamethasone also reduced inflammatory cell infiltration. After repeated treatments, all formulations decreased the ear edema at 24 h, 48 h and 72 h after UVB. OA in semisolids, especially Pemulen® TR2-based ones, presented suitable characteristics for cutaneous administration and its anti-inflammatory activity seems to occur via glucocorticoid receptors. OA was also capable to reduce croton oil-induced skin inflammation. Besides, the ex vivo skin permeation study indicated that OA reaches the receptor medium, which correlates with a systemic absorption in vivo. The natural compound OA could represent a promising alternative to those available to treat inflammatory skin disorders.

This paper describes a bioprocess to obtain omegas-6 and 9 from the hydrolysis of Açaí (Euterpe oleracea Martius) and Buriti (Mauritia flexuosa) oils by lipases immobilized on octyl-sepharose. For this, oils and butters were initially selected as the carbon source which resulted in higher production of lipases in Beauveria bassiana and Fusarium oxysporum cultures. The carbon source that provided secretion of lipase by B. bassiana was Açaí oil, and for F. oxysporum, Bacuri butter. Lipases obtained under these conditions were immobilized on octyl-sepharose, and both, the derivatives and the crude extracts were biochemically characterized. It was observed that the immobilization promoted an increase of stability in B. bassiana and F. oxysporum lipase activities at the given temperatures and pH. In addition, the immobilization promoted hyperactivation of B. bassiana and F. oxysporum lipase activities being 23.5 and 11.0 higher than free enzyme, respectively. The hydrolysis of Açaí and Buriti oils by the derivatives was done in a biphasic (organic/aqueous) system, and the products were quantified in RP-HPLC. The results showed the potential of these immobilized lipases to obtain omegas-6 and 9 from Brazilian natural oils. This work may improve the enzymatic methodologies for obtaining foods and drugs enriched with fatty acids.

Avaliou-se a produção de oócitos e embriões de vacas Nelore in vitro e a resistência à vitrificação. Foram utilizadas 12 vacas Nelore, distribuídas aleatoriamente em dois tratamentos: T1-tratados com canola em grão (2,0kg/animal/dia) e T2-controle. Cada animal foi aspirado quatro vezes para obtenção de óocitos para fecundação in vitro. Os oócitos foram quantificados e classificados em viáveis ou inviáveis. Os zigotos foram cultivados in vitro e, sete dias após, os embriões foram avaliados quanto à qualidade e grau de desenvolvimento e vitrificados em hastes próprias. Na sequência, foram descongelados e cultivados em 6, 12 e 24 horas, observando-se a taxa de expansão e eclosão. Não houve diferenças (P>0,05) no número total de oócitos viáveis, T1=12,7% e T2=11,0%, na taxa de clivagem, T1=60,6% e T2=61,4%, e taxa de blastocistos, T1=23,7% e T2=27,0%, em função do tratamento. Também não houve influência na taxa de re-expansão, T1=70,5 e T2=59,6%, após a vitrificação e descongelamento. Todavia houve diferença (P<0,06) para a taxa de eclosão, T1=69,2 e T2=35,7. Conclui-se que a adição de canola na dieta de vacas não alterou a produção de embriões; entretanto, os embriões resultantes de oócitos coletados de vacas alimentadas com canola são mais resistentes à vitrificação.(AU)


It was evaluated the production of oocytes and embryos from Nellore cows in vitro, as well as its resistance to vitrification, when the animals were supplemented with canola grains. Twelve Nellore cows were randomly divided into two treatments: T1-treated with Canola grains (2.0kg/animal/day) and T2-control. Each animal was submitted to other four aspirations, to obtain oocytes for the in vitro fertilization. The oocytes were quantified and classified as viable or not viable. The embryos were cultivated in vitro, seven days after the quality and the level of development of embryos was evaluated and they were vitrified in vitrification straws. Then, the embryos were thawed and grown during 6, 12 and 24 hours, and the rate of expansion and hatching were recorded. There were no differences (P> 0.05) in the whole number of viable oocytes T1: 12.7±1.71 and T2: 11.0±1.77, cleavage rate T1= 60.6±4,72 and T2= 61.4±4.88 or blastocysts rate T1=23,7±5.12 and T2=27,0±5.30 due to the treatment. The treatments did not influence the rate of re-expansion T1= 70,5±6.99 and T2: 59.6±7.09 after vitrification and thawing. However, there was a significant difference (P <0.06) in the hatching rate (T1: 69.17±7.43; T2: 35.66±6.86). Thus, we conclude that supplementation with canola grains did not change embryos production, but the embryos yielded from oocytes of cows fed canola grains are more resistant to vitrification.(AU)

A resposta imunológica é um processo complexo que envolve a interação e sinalização de diferentes tipos celulares e moleculares, conhecidos como biomarcadores, os quais atuam de modo coordenado para promover a defesa do organismo. Essa resposta inicia-se com a instalação do processo inflamatório, que culmina com eliminação de agentes microbianos e cicatrização do tecido afetado, podendo evoluir para uma resposta mais específica. Muitos mediadores produzidos durante essas reações, sobretudo aqueles de natureza lipídica e peptídica, podem ser originados da imunomodulação promovida por ácidos graxos insaturados fornecidos pela dieta ou sintetizados endogenamente. Ácidos graxos insaturados são constituintes das membranas celulares e reguladores da expressão gênica, atuando nas vias de sinalização, transdução de sinais e proliferação celular e gerando produtos que regulam os mecanismos da imunidade humoral e celular. Esses lipídios apresentam várias propriedades biológicas, incluindo importantes efeitos imunomoduladores sobre a inflamação e a cicatrização. Ácidos graxos insaturados regulam a ativação de diversas células envolvidas nesses processos, tais como macrófagos, neutrófilos, linfócitos, queratinócitos e células dendríticas, bem como a secreção de seus produtos, exercem efeitos sobre enzimas e fatores de transcrição gênica, estimulando ou inibindo a produção de eicosanóides, como as prostaglandinas e leucotrienos, citocinas, moléculas de adesão, fatores de crescimento e outras moléculas específicas envolvidas nas diferentes fases do reparo tecidual, além de modular o estresse oxidativo. Neste contexto, esse artigo tem como objetivo revisar o papel dos principais biomarcadores celulares e moleculares envolvidos na resposta imune-inflamatória modulada por ácidos graxos insaturados.(AU)


Immune response is a complex process that involves interaction and signaling of different cell and molecular types, known as biomarkers, which are organized to promote the defense of organism. This response begins with installation of the inflammatory process and culminates with elimination of the microbial agents, damage tissue and repair. The inflammatory response can progress to a more specific process. Many mediators are produced during inflammation, especially lipid and peptide mediators. These substances can be generated from immunomodulation promoted by unsaturated fatty acids provided by dietary or endogenously synthesized. Unsaturated fatty acids are constituents of cell membranes and regulators of gene expression and act on signaling pathways, signal transduction and cell proliferation, generating products that regulate the mechanisms of humoral and cellular immunity. These lipids have multiple biological properties, including important immunomodulatory effects on inflammation and wound healing. Unsaturated fatty acids regulate the activation of various cells involved in these processes, such as macrophages, neutrophils, lymphocytes, keratinocytes and dendritic cells, as well as secretion of their products, exert effects on enzymes and gene transcription factors by stimulating or inhibiting the production of eicosanoids as prostaglandins and leukotrienes, cytokines, adhesion molecules, growth factors and other specific molecules involved in different phases of tissue repair, and modulate oxidative stress. In this context, the aim of this paper is to review the role of main cellular and molecular biomarkers involved in immune-inflammatory response modulated by unsaturated fatty acids.(AU)

This article describes the possibility of modifying the composition of fat tissue in broiler chickens fed canola oil, which is high in monounsaturated fatty acids. 128 one-day old broiler chickens, randomly assigned into 4 groups of 32 chicks each, received one of four diets containing 15% oil with different percentages of canola oil (diet 1: 0% canola oil, diet 2: 5% canola oil, diet 3: 10% canola oil and diet 4: 15% canola oil), for 31 days. Each group was divided into 4 subgroups of 8 chicks. The birds were sacrificed at day 45 to obtain tissue samples. The fatty acid composition was measured in meat (legs and breasts), fat (abdominal and subcutaneous) and plasma. An increase in oleic acid (p<0.01) was detected, as well as a decrease in linoleic acid (p<0.01), together with a slight increase in α-linolenic acid (p<0.05) with a higher percentage of canola oil. The composition of fat tissue was more representative of the dietary fatty acids than muscle tissue. In conclusion, canola oil increased the content of omega 9 and omega 3 fatty acids and decreased the content of omega 6 fatty acids in meat, fat and plasma in broiler chickens.
