RESUMO

Paracetamol, a contaminant of emerging concern, has been detected in different bodies of water, where it can impact ecological and human health. To quantify this paracetamol, electroanalytical methods have gained support. Thus, the present study developed a simple, inexpensive, and environmentally friendly method for paracetamol quantification using a carbon fiber microelectrode based on commercial carbon fiber. To improve the carbon fiber microelectrode's paracetamol sensitivity and selectivity, it was subjected to an activation process via electrochemical oxidation in an acid medium (H2SO4 or HNO3), using 20 consecutive cycles of cyclic voltammetry. The treated (activated) carbon fiber microelectrode was characterized using scanning electron microscopy and electrochemical techniques, including chronoamperometry and electrochemical impedance spectroscopy. The H2SO4-activated carbon fiber microelectrode exhibited enhanced figures of merit, with a linear dynamic range of paracetamol detection from 0.5 to 11 µmol L-1 and a limit of detection of 0.21 µmol L-1 under optimized conditions. The method was optimized by quantifying paracetamol in commercial pharmaceutical tablets, spiked running tap water, and river water (Pita River, Quito, Ecuador, latitude -0.364955°, longitude -78.404538°); the respective recovery values were 102.89, 103.93, and 112.40%. The results demonstrated an acceptable level of accuracy and the promising applicability of this carbon fiber microelectrode as a sensor to detect paracetamol.

RESUMO

Seafood consumption is the primary exposure route for trace metals like mercury. Accordingly, canned tuna meat has been focused on by researchers because of the potential bioaccumulation of high amounts of mercury. This study aimed to test a novel and reliable electroanalytical method employing a working electrode consisting of gold-nanoparticle-modified carbon microfibers to quantify total mercury in canned tuna samples. Determination was achieved via differential pulse anodic stripping voltammetry. The proposed method had a limit of detection of 3.9781 ± 0.0001 µg L-1 and a limit of quantification of 33.6634 ± 0.0001 µg L-1, with a sensitivity of 0.3275 nA µg L-1. The modified electrode was evaluated in samples taken from three canned tuna brands sold in the Sangolquí parish in Rumiñahui, Ecuador. These brands, coded A, B, and C, represent 47.92%, 27.08%, and 11.98% of all canned tuna sold in the Ecuadorian market, respectively. The resulting respective total mercury concentrations were 0.5999 ± 0.0001 mg kg-1; 0.9387 ± 0.0001 mg kg-1; and 0.3442 ± 0.0001 mg kg-1 for A, B, and C. Method accuracy was determined through the recovery percentages of ≥98%, which indicated acceptable accuracy for the final optimized method. Mean mercury concentrations for all samples did not represent a carcinogenic risk for consumers. However, the values obtained for potential no-carcinogenic risk and daily consumption rate suggest that consumers of tuna canned in water, particularly brand C, may be at risk.

Loop-mediated isothermal amplification (LAMP) has been recently studied as an alternative method for cost-effective diagnostics in the context of the current COVID-19 pandemic. Recent reports document that LAMP-based diagnostic methods have a comparable sensitivity and specificity to that of RT-qPCR. We report the use of a portable Arduino-based LAMP-based amplification system assisted by pH microelectrodes for the accurate and reliable diagnosis of SARS-CoV-2 during the first 3 min of the amplification reaction. We show that this simple system enables a straightforward discrimination between samples containing or not containing artificial SARS-CoV-2 genetic material in the range of 10 to 10,000 copies per 50 µL of reaction mix. We also spiked saliva samples with SARS-CoV-2 synthetic material and corroborated that the LAMP reaction can be successfully monitored in real time using microelectrodes in saliva samples as well. These results may have profound implications for the design of real-time and portable quantitative systems for the reliable detection of viral pathogens including SARS-CoV-2.

INTRODUCTION: In 2006 and 2009, we reported the levels of acute and chronic tissue damage after cordectomy associated with use of the microlectrodes using high frequency energy. In 2010, we shifted to radiofrequency rather than high frequency electrogenerators. OBJECTIVE: The aim of this study is to evaluate acute tissue damage in the larynx after cordectomy using microelectrodes coupled to a radiofrequencygenerator. METHODS: We studied 22 patients with a stage T1 glottic squamous cell carcinoma. The patients were randomly assigned to the two operating mode: cutting or coagulation (11 patients each mode). The strength of the study is that there are no previous studies on the effect of radiofrequency in human vocal cord. RESULTS: Tissue damage was milder when microelectrodes were coupled to a 4 MHz generator operating in the cutting mode. Thus, when using microelectrodes and radiofrequency, we recommend that the cutting mode be used for epithelial incision and the coagulation mode to treat the stroma and muscle and for final hemostasis. CONCLUSION: Microelectrodes and radiofrequency in transoral laryngeal surgery produced mild tissue damage and offer an excellent alternative to the use of high frequency energy.

Speciation and partitioning of trace metals, from solid to solution phases of sediments, control their bioavailability and thus their potential ecological risk to organisms. Therefore, in order to obtain a broad evaluation of their risk, it is necessary to couple methodologies that are able to assess metal mobility in sediment. In this study, the Diffusive Gradients in Thin Films (DGT) technique and the application of 0.1 M HCl acid extraction methods, together with solid-state voltammetric sensors, were used with the objective of assessing mobility and potential availability of Cr, Cu, Ni, Pb, V and Zn in sediment porewaters and solid sediments in southeastern and southern Brazilian shipyard areas. The highest labile metal concentrations were found in shipyards with the longest histories of operations. Trace metal distributions in porewater and in the solid phase of sediments (labile metals) and significant correlations among metals enabled to distinguish the contribution of anti-fouling paint components. The diffusive flux of every metal measured at the surface of the sediment indicated that CuDGT had the highest flux (3.66E-03 mmol·m-2 d-1) in the shipyard with the longest operating time. Therefore, enrichment was observed for Cu, Pb and Zn in sediments, indicating a possible ecological risk level of 'Effects Range Median' to 'Apparent Effects Threshold' for oyster larvae (Mollusca) (Cu), bivalves (Pb) and the infaunal community (Zn). Probable Effect Concentrations (PEC) to sediment-dwelling biota can be expected as well, related to high concentrations of Cu and Zn in sediment. This study allowed a comprehensive evaluation of potential bioavailability and ecological risk of trace metals in aquatic systems where there is continuous and specific input of these elements. The use of the DGT technique with solid-state voltammetry in the sediment of distinct Brazilian estuarine systems demonstrated its potential to be applied in future environmental network programs.

The plasma membrane functions both as a natural insulator and a diffusion barrier to the movement of ions. A wide variety of proteins transport and pump ions to generate concentration gradients that result in voltage differences, while ion channels allow ions to move across the membrane down those gradients. Plasma membrane potential is the difference in voltage between the inside and the outside of a biological cell, and it ranges from ~- 3 to ~- 90 mV. Most of the most significant discoveries in this field have been made in excitable cells, such as nerve and muscle cells. Nevertheless, special attention has been paid to some events controlled by changes in membrane potential in non-excitable cells. The origins of several blood disorders, for instance, are related to disturbances at the level of plasma membrane in erythrocytes, the structurally simplest red blood cells. The high simplicity of erythrocytes, in particular, made them perfect candidates for the electrophysiological studies that laid the foundations for understanding the generation, maintenance, and roles of membrane potential. This article summarizes the methodologies that have been used during the past decades to determine Δψ in red blood cells, from seminal microelectrodes, through the use of nuclear magnetic resonance or lipophilic radioactive ions to quantify intra and extracellular ions, to continuously renewed fluorescent potentiometric dyes. We have attempted to highlight the advantages and disadvantages of each methodology, as well as to provide a description of the technical aspects involved.

Aroma is closely related to the food product acceptability and an important product quality indicator. Electronic-nose (E-nose) systems are an interesting alternative to traditional methods of aroma analyses. A lab-made E-nose system equipped with an array of sensing units comprised by gold interdigitated microelectrodes (IDEs) using polyaniline (Pani) as sensitive layers deposited by the in situ and Layer-by-layer (LbL) methods was used to analyze aromas in gummy candies. Different concentrations from artificial aromas (apple, strawberry and grape), added to the gummy candies were evaluated. Our system presented 21.6 mV.ppb-1 sensitivity, ppb range detection limit, and good reversibility, around 97.6%. The sensitive layers of Pani films was adequate deposited on IDEs observed by the Attenuated Total Reflection/Fourier-transform infrared spectroscopy (ATR/FTIR). Linear Discriminant Analysis (LDA) was able to classify apple, strawberry, and grape aromas added to gummy candies using saturation potential values from the E-nose system, demonstrating its applicability in food matrices.

El láser como alternativa a la cirugía abierta de la vía aérea superior ha venido a modificar la forma de abordaje de las patologías en esta área, pero no deja de ser un procedimiento costoso que no está al alcance de todos los servicios. Por este motivo se han reinventado nuevas formas de abordaje que cumplan los mismos requisitos tanto de la cirugía abierta como con láser pero con un menor coste. Presentamos una serie de 30 casos realizados en un período de 6 años por motivos tanto tumorales como no, en los que se realizaron abordajes cerrados a través de microcirugía con disección mediante microelectrodos. Obteniendo pocas complicaciones y una disminución de la estancia hospitalaria significativa. Con lo cual nos parece una técnica eficiente para abordajes de este tipo.


The laser as an alternative to open surgery of the upper airway has come to change the form of approaching the disease in this area, but it is still an expensive procedure that is not available to all services. For this reason a new ways of approach to meet the same requirements both open as laser but at a lower cost surgery. We present a series of 30 cases performed over a period of 6 years for reasons as much tumor, which closed approaches through microsurgical dissection were performed using microelectrodes. Obtaining few complications and significant decreased hospital stay. Our considerations is it seems an efficient technique for such approaches.

We have developed an anodic stripping voltammetry method that employs carbon fiber ultra-microelectrodes modified with gold nanoparticles to determine arsenic in natural waters. Gold nanoparticles were potentiostatically deposited on carbon fiber ultra-microelectrodes at -0.90V (vs SCE) for a time of 15s, to form the carbon fiber ultra-microelectrodes modified with gold nanoparticles. Cyclic voltammetry, electrochemical impedance spectroscopy and scanning electron microscopy coupled to an X-ray microanalysis system were used to check and confirm the presence of gold nanoparticles on the carbon fiber ultra-microelectrodes. Arsenic detection parameters such as deposition potential and deposition time were optimized allowing a detection range between 5 to 60µgL-1. The developed modified electrodes allowed rapid As determination with improved analytical characteristics including better repeatability, higher selectivity, lower detection limit (0.9µgL-1) and higher sensitivity (0.0176nAµgL-1) as compared to the standard carbon electrodes. The analytical capability of the optimized method was demonstrated by determination of arsenic in certified reference materials (trace elements in water (NIST SRM 1643d)) and by comparison of results with those obtained by hydride generation atomic absorption spectrometry (HG-AAS) in the determination of the analyte in tap and well waters.

In recent years, new recording technologies have advanced such that oscillations of neuronal networks can be identified from simultaneous, multisite recordings at high temporal and spatial resolutions. However, because of the deluge of multichannel data generated by these experiments, achieving the full potential of parallel neuronal recordings also depends on the development of new mathematical methods capable of extracting meaningful information related to time, frequency and space. In this review, we aim to bridge this gap by focusing on the new analysis tools developed for the automated detection of high-frequency oscillations (HFOs, >40Hz) in local field potentials. For this, we provide a revision of different aspects associated with physiological and pathological HFOs as well as the several stages involved in their automatic detection including preprocessing, selection, rejection and analysis through time-frequency processes. Beyond basic research, the automatic detection of HFOs would greatly assist diagnosis of epilepsy disorders based on the recognition of these typical pathological patterns in the electroencephalogram (EEG). Also, we emphasize how these HFO detection methods can be applied and the properties that might be inferred from neuronal signals, indicating potential future directions.

The α-carbonic anhydrases (CAs) are zinc-containing enzymes that catalyze the interconversion of CO2 and HCO3 (-). Here, we focus on human CA II (CA II), a ubiquitous cytoplasmic enzyme. In the second paper in this series, we examine CA IV at the extracellular surface. After microinjecting recombinant CA II in a Tris solution (or just Tris) into oocytes, we expose oocytes to 1.5% CO2/10 mM HCO3 (-)/pH 7.50 while using microelectrodes to monitor intracellular pH (pHi) and surface pH (pHS). CO2 influx causes the familiar sustained pHi fall as well as a transient pHS rise; CO2 efflux does the opposite. Both during CO2 addition and removal, CA II increases the magnitudes of the maximal rate of pHi change, (dpHi/dt)max, and the maximal change in pHS, ΔpHS. Preincubating oocytes with the inhibitor ethoxzolamide eliminates the effects of CA II. Compared with pHS, pHi begins to change only after a delay of ~9 s and its relaxation has a larger (i.e., slower) time constant (τpHi > τpHS ). Simultaneous measurements with two pHi electrodes, one superficial and one deep, suggest that impalement depth contributes to pHi delay and higher τpHi . Using higher CO2/HCO3 (-) levels, i.e., 5%/33 mM HCO3 (-) or 10%/66 mM HCO3 (-), increases (dpHi/dt)max and ΔpHS, though not in proportion to the increase in [CO2]. A reaction-diffusion mathematical model (described in the third paper in this series) accounts for the above general features and supports the conclusion that cytosolic CA-consuming entering CO2 or replenishing exiting CO2-increases CO2 fluxes across the cell membrane.

Human carbonic anhydrase IV (CA IV) is GPI-anchored to the outer membrane surface, catalyzing CO2/HCO3 (-) hydration-dehydration. We examined effects of heterologously expressed CA IV on intracellular-pH (pHi) and surface-pH (pHS) transients caused by exposing oocytes to CO2/HCO3 (-)/pH 7.50. CO2 influx causes a sustained pHi fall and a transient pHS rise; CO2 efflux does the opposite. Both during CO2 addition and removal, CA IV increases magnitudes of maximal rate of pHi change (dpHi/dt)max, and maximal pHS change (ΔpHS) and decreases time constants for pHi changes (τpHi ) and pHS relaxations (τpHS ). Decreases in time constants indicate that CA IV enhances CO2 fluxes. Extracellular acetazolamide blocks all CA IV effects, but not those of injected CA II. Injected acetazolamide partially reduces CA IV effects. Thus, extracellular CA is required for, and the equivalent of cytosol-accessible CA augments, the effects of CA IV. Increasing the concentration of the extracellular non-CO2/HCO3 (-) buffer (i.e., HEPES), in the presence of extracellular CA or at high [CO2], accelerates CO2 influx. Simultaneous measurements with two pHS electrodes, one on the oocyte meridian perpendicular to the axis of flow and one downstream from the direction of extracellular-solution flow, reveal that the downstream electrode has a larger (i.e., slower) τpHS , indicating [CO2] asymmetry over the oocyte surface. A reaction-diffusion mathematical model (third paper in series) accounts for the above general features, and supports the conclusion that extracellular CA, which replenishes entering CO2 or consumes exiting CO2 at the extracellular surface, enhances the gradient driving CO2 influx across the cell membrane.
