RESUMEN Objetivo : Comparar la microdureza de la resina compuesta con técnica índex convencional y modificada con cubeta impresa en dos fotopolimerizaciones. Materiales y métodos : Se contó con seis grupos de estudio de acuerdo a la técnica y a la cantidad de fotopolimerizaciones: técnica directa con una fotopolimerización (D1F), directa con dos fotopolimerizaciones (D2F), índex convencional con una fotopolimerización (IC1F), índex convencional con dos fotopolimerizaciones (IC2F), índex modificada con una fotopolimerización (IM1F), índex modificada con dos fotopolimerizaciones (IM2F). Se utilizaron quince muestras por cada grupo. Las muestras tuvieron dimensiones de 2 mm de altura por 5 mm de diámetro. Se realizó la fotopolimerización siguiendo las indicaciones del grupo al que corresponde, para luego someter a la prueba de dureza de Vickers a tres indentaciones por cada cara con carga de 200 g por 15 segundos. Se realizaron dos registros de microdureza, una superficial y otra a 2 mm. Se recolectaron los datos en el instrumento confeccionado y fueron procesados mediante SPSS v. 26. Resultados : Existe una diferencia significativa entre los grupos de microdureza superficial de primera fotopolimerización (p < 0,001); y también existe diferencia significativa entre los grupos de microdureza a 2 mm de primera fotopolimerización (p < 0,001). Asimismo, no existen diferencias significativas entre los grupos de microdureza superficial de segunda fotopolimerización (p = 0,519) ni en los grupos de microdureza a 2 mm de segunda fotopolimerización (p = 0,279). Conclusiones: No existen diferencias significativas en la microdureza superficial y a 2 mm de profundidad con técnica índex convencional y modificada en cubeta impresa en dos fotopolimerizaciones.
ABSTRACT Objective : To compare the microhardness of composite resin with conventional and modified index techniques with a printed tray in two light cures. Materials and methods : There were six study groups according to the technique and the number of photopolymerizations: direct technique with one photopolymerization (D1F), direct with two photopolymerizations (D2F), conventional index with one photopolymerization (IC1F), conventional index with two photopolymerizations (IC2F), modified index with one photopolymerization (IM1F), modified index with two photopolymerizations (IM2F). Fifteen samples were used for each group. The samples had dimensions of 2 mm in height by 5 mm in diameter. The photopolymerization was performed following the indications of the group to which it corresponds and then subjected to the Vickers hardness test with three indentations on each side with a load of 200 g for 15 seconds. Two microhardness recordings were made, one superficial and the other at 2 mm. Results : There is a significant difference between the first light-curing surface microhardness groups (p < 0.001), and there is also a significant difference between the first light-curing 2 mm microhardness groups (p < 0.001). Likewise, there are no significant differences between the second light-curing surface microhardness groups (p = 0.519) or the second light-curing 2 mm microhardness groups (p = 0.279). Conclusions : There are no significant differences in surface microhardness and microhardness at 2 mm depth with conventional and modified index techniques in printed trays in two photopolymerizations.

Da mesma forma, não há diferença significativa entre os grupos de microdureza da superfície do segundo fotopolimerizador (p = 0,519) e nos grupos de microdureza de 2 mm do segundo fotopolimerizador (p = 0,279). Conclusões : Não existem diferenças significativas na microdureza da superfície e na microdureza a 2 mm de profundidade com a técnica de índice convencional e modificada em duas fotopolimerizações.
RESUMO
Objective:To analyze the effect of methylene blue and 10% curcumin in fungi and bacteria through an in vitrostudy using photodynamic therapy (PDT). Methods:Curcumin and methylene blue were photosensitized by a Photon Lase III laser applied for 90 s in a dark environment within a laminar flow chamber. Enterococcus faecalisand Candida albicans strains were cultured and standardized.Then, a minimum inhibitoryconcentration (MIC) assay was conducted for these photosensitizers, with concentration variations and incubation to evaluate their antimicrobial activity. Results:With PDT, Curcumin had significant antibacterial activity against E. faecalis (MIC = 250 µg/mL).In contrast, methylene blue had antibacterial activity against E. faecalis (MIC < 12.5 µg/mL with PDT) and antifungal activity against C. albicans (MIC <12.5 µg/mL with or without PDT).Both agents showed greater efficacy in the presence of the laser.The results suggest that curcumin and methylene blue associated with laser may effectively treat microbial infections. Conclusion:Further research is needed to evaluate the efficacy and safety of using these agents in animal and human models and theireffectiveness against different bacterial and fungal strains.

OBJECTIVE: This study is to evaluate fracture resistance, failure mode, and gap formation at the restorative interface of unrestored or restored non-carious cervical lesions (NCCLs) submitted to a short-term erosive environment. MATERIALS AND METHODS: Artificial NCCLs were produced in vitro in bovine incisors, and were randomly divided into four restorative resins groups (n = 22): nanohybrid-NR; bulk-fill-BR; flow with a nanohybrid layer-FNR; bulk-fill with a nanohybrid layer-BNR; and a group unrestored-UR (n = 16). Half of the specimens were submitted to an erosive challenge (per 5 min, 3 × a day for 7 days, before and after restoration), and the other half, was immersed in artificial saliva. After, all teeth undergone thermal (5 ºC, 37 ºC, and 55 ºC, 3600 cycles) and mechanical (50 N, 2 Hz, 300,000 cycles) aging. Eighty teeth were subjected to compressive loading, and resistance and failures were analyzed, while 24 teeth were evaluated for gaps by microcomputed tomography. Statistical tests were performed (p < 0.05). RESULTS: The restorative approaches affected fracture resistance (η2p = 0.14, p = 0.023), and gap formation (η2 = 0.18, p = 0.012) and so did the immersion medium (fracture η2p = 0.09, p = 0.008; gap η2 = 0.09, p = 0.017). BNR showed the highest resistance, while UR the lowest. FNR showed the highest gaps in both immersion media. Neither the resin groups nor the immersion media were associated with failure mode. CONCLUSIONS: The erosive immersion medium based in acid beverages has been shown to affect NCCLs with or without restoration, but when Bulk-Fill resin is covered by nanohybrid resin, the performance is good. CLINICAL RELEVANCE: Erosion negatively affects restorations, but unrestored NCCL shows worse biomechanical performance in stress-bearing situations.

PURPOSE: This study compared the mechanical behavior of two fixation techniques used in three sections representing the sagittal split ramus osteotomy (SSRO) in polyurethane replicas that were divided into groups, according to type of section, and sub-groups according to type of fixation, simulating 11-mm advancement and 6º clockwise mandibular rotation. METHODS: Loads were applied in two regions, aiming at progressive application and consequent strength value, measured in kilogram-force in displacements of 1, 3, 5, and 7 mm, from the load application tip. Shapiro-Wilk test was performed, followed by two-way analysis of variance (ANOVA-2 way), and Bonferroni's multiple comparison. RESULTS: The results showed no statistically significant difference in the type of section and type of fixation used when load was applied to the inter-incisor region. However, when load was applied to the first molar region, statistically significant difference was observed in 1-mm displacement, in which section described by Epker with two modifications showed greater strength, regardless of type of fixation used (p = 0.007). CONCLUSION: In the application of load in the inter-incisor region, there was no statistical difference between the type of osteotomy and the type of fixation used. When applying loads to molars, there was a difference for the type of osteotomy, where the Epker osteotomy with 2 modifications presented greater resistance, regardless of the type of fixation used.

PURPOSE: Compare canal transportation and dentin removal using micro computed tomography (micro-CT) and measure instrumentation time in the root canal preparation of primary molars using K-files, ProDesign Logic and Reciproc files. METHODS: A representative sample of 81 root canals of primary molars was randomised into three groups: K-files (#15-40), ProDesign Logic (#25) and Reciproc (R25). A single-trained operator prepared all root canals. A standardised micro-CT evaluation was performed before and after instrumentation using the CTan® program. Instrumentation time was also recorded. Statistical analysis involved one-way ANOVA, Tukey's post hoc test, Kruskal-Wallis test and Dunn's test, considering a 5% level of significance. RESULTS: Reciproc led to more canal transportation than K-files and ProDesign Logic (p < 0.001) in middle third, whereas K-files led to the most canal transportation in the apical third (p = 0.017). ProDesign Logic and Reciproc removed less dentin on the opposite side of the root curvature in the apical third compared to K-files (p = 0.008). Instrumentation time was better with ProDesign Logic and Reciproc than K-files (p < 0.001). CONCLUSION: ProDesign Logic demonstrated good results with regard to canal transportation and required less instrumentation time in primary molars. Reciproc and K-files led to the most canal transportation in the middle and apical third, respectively.

The objective of this study was to evaluate the effects of alkaloid extracts of Prosopis juliflora (Sw.) D.C. pods obtained by two extraction methods as compared with sodium monensin on the gas production kinetic, mitigation of methane, and rumen fermentation products using wheat bran or Tifton 85 hay as substrates, by the semi-automatic in vitro gas production technique. A completely randomized design was adopted, and two natural additives were tested made from mesquite pod (alkaloid extract I and alkaloid extract II) at three levels (3.9, 7.9, and 12 µg), sodium monensin 5 µM (positive control), and no inclusion of additives (negative control). The volume of gases produced by the degradation of the fibrous fraction of wheat bran was influenced by the concentration of the extract I added to the medium, and the amounts of 7.9 and 12 µg were equal to monensin at the lowest value. The degradation rate of the fibrous carbohydrates with additive extract I at 12 µg was lower in relation to monensin. When Tifton 85 hay was utilized, alkaloid extract I provided a shorter colonization time as compared with monensin at the added amounts of 7.9 and 12 µg and higher production of gases from the fibrous fraction but without interfering with the total volume of gases produced during 96 h of fermentation of carbohydrates. In the periods of 12 and 24 h of incubation, utilizing alkaloid extract I, the mean values of methane production with wheat bran and Tifton 85 hay were lower than monensin (p < 0.05) when the respective amounts of 7.9 and 12 µg were added. Alkaloid extract I has similar potential to sodium in reducing production of total gases, methane, and the acetate/propionate ratio.

Objetivo: O presente estudo teve como objetivo analisar a influência do modelo experimental e substrato nas alterações do esmalte dental decorrentes do tratamento clareador de consultório em diferentes tempos de análise. Materiais e Métodos: Um total de 140 discos contendo esmalte e dentina foram confeccionados, a dentina planificada e o esmalte polido e submetido ao teste de microdureza de superfície em KHN (MS) para padronização e seleção inicial dos espécimes (n= 80). A seguir, foram divididos em 8 grupos (n=10): G1- Controle in vitro em dentes humanos (VHC); G2- Clareamento in vitro utilizando agente clareador Pola Office a base de peróxido de hidrogênio a 35% (Pola PH 35%) em dentes humanos (VHP); G3- Controle in situ em dentes humanos (SHC); G4- Clareamento in situ com Pola PH 35% em dentes humanos (SHP); G5- Controle in vitro em dentes bovinos (VBC); G6- Clareamento in vitro com Pola PH 35% em dentes bovinos (VBP); G7- Controle in situ dentes bovinos (SBC); G8- Clareamento in situ com Pola PH 35% em dentes bovinos (SBP). O esmalte dental foi avaliado quantitativamente pelas análises de rugosidade, MS e microdureza longitudinal do esmalte em KHN (ML) e qualitativamente por microscopia eletrônica de varredura (MEV). As análises de rugosidade e MS foram realizadas antes do início do tratamento (T0), após a 3° semana de tratamento (T1) e 15 dias após o término do tratamento (T2). Já a análise de ML e as imagens de MEV foram realizadas apenas em T2. Os dados foram submetidos ao teste de homocedasticidade Shapiro-wilk, em seguida, foi realizada à análise de variância (ANOVA) dois fatores medidas-repetidas e pós-teste de Tukey ou Sidak (p<0,05) ou teste de Friedman, seguido pelo teste post hoc de Wilcoxon com correção de Bonferroni. Resultados: Na comparação entre os modelos in vitro e in situ, em todas as análises realisadas, pode-se notar diferença estatisticamente significante 15 dias após o término do tratamento clareador (p< 0,05), sendo evidente a recuperação do esmalte dentário no modelo in situ. Com relação aos substratos, estes apresentaram diferença estatística nas análises de MS e ML (p> 0,05). No tocante a análise dos grupos ao longo do tempo, nas variáveis rugosidade e MS, observou-se diferenças estatísticas (p<0,05), comprovando a ação negativa do tratamento clareador em T1, seguida em T2 pela manutenção dessas alterações no grupo in vitro e recuperação dessas superfícies nos grupos in situ. Conclui-se que: O modelo experimental foi decisivo para o estudo das alterações do esmalte dentário clareado, pois, o modelo in situ permite redução ou recomposição das alterações dentárias promovidas pelo tratamento clareador. - Em pesquisas sobre o clareamento dental, ambos os substratos empregados neste estudo podem ser utilizados, desde que, as diferenças existentes entre eles sejam consideradas no momento da interpretação dos dados. - Os diferentes tempos de análise foram determinantes para a observação da ação do agente clareador sobre a estrutura dental(AU)
Objective: The objective of the present study was to analyze, influence, model, experimental, substrate, birth, treatment, repair, surgery, different times, analysis. Materials and Methods: a total of 140 discotheques containing enamel and dentin were made, a planned dentin and polished omalmalte and submitted to the surface microhardness test in KHN (MS) for standardization and initial selection of spagos (n = 80). They were divided into 8 groups (n = 10): G1- In vitro control in human teeth (VHC); G2- In vitro bleaching, for example, Pola Office whitening agent, 35% hydrogen peroxide base (Pola PH 35%) in human teeth (VHP); G3-In situ control in human teeth (SHC); G4- In situ bleaching with Pola PH 35% in human teeth (SHP); G5- In vitro control in bovine teeth (VBC); G6- In vitro whitening with PH Pola 35% in bovine teeth (VBP); G7- In situ control of bovine teeth (SBC); G8- In situ bleaching with Pola PH 35% in bovine teeth (SBP). The enamel was evaluated quantitatively in the analysis of roughness, MS and longitudinal microhardness of the enamel in KHN (ML) and qualitatively by scanning electron microscopy (SEM). The roughness and MS analyzes were performed before the start of treatment (T0), after one week of treatment (T1) and 15 days after treatment (T2). Already an ML analysis and as SEM images were performed only in T2. The data were submitted to the Shapiro-wilk homoscedasticity test, then performed in analysis of variance (ANOVA) of measures and Tukey or Sidak test repetitions (p <0.05) or Friedman's test, by the post test Hoc analysis of Wilcoxon with Bonferroni correction. Results: In the relativity of in vitro and in situ models, a statistically significant difference can be observed 15 days after the end of the bleaching treatment (p <0.05) in all analyzes, In situ. Regarding the substrates, these presented a statistical difference in the MS and ML analyzes (p> 0.05). Regarding the analysis of the groups over time, in the roughness and MS variables, statistical statistics (p <0.05) were observed, proving a negative action of the bleaching treatment in T1, instead of T2 through maintenance, there were no groups In vitro and surface recovery in in situ groups. It is concluded that: The experimental model was decisive for the study of the author's keywords, the in situ model allows to reduce or to reward the dental alternatives promoted by the bleaching treatment. - In research on dental bleaching of the substrates used in the study can be used, since, as residues between them, are not considered in the interpretation version of the data. - The different times of analysis were determinant for an observation of the action of the bleaching agent on a dental structure(AU)

A homeopatia apresentase como uma excelente opção de baixo custo e toxicidade para uso na prática da reprodução tanto humana quanto animal. Entretanto, desperta um alto nível de ceticismo em relação a sua real eficácia, notadamente devido ao possível efeito placebo. O uso de modelos in vitro, como a tecnologia do ovário artificial, apresentase como uma ferramenta de grande precisão para dirimir tal controvérsia. Diante disso, esta revisão tem como objetivo fornecer algumas bases sobre a foliculogênese e sua regulação, relatar a importância do cultivo in vitro, com ênfase no hormônio folículo estimulante (FSH), na avaliação do papel dos medicamentos homeopáticos no tratamento de distúrbios reprodutivos ovarianos e no seu uso para melhorar as biotécnicas reprodutivas.
The homeopathy is a lowcost and toxicity alternative to using in the human and animal reproduction. However, the effect homeopathy awakens a skepticism about its effectiveness, because of the possible placebo effect. The in vitro models, as artificial ovary, is a excellent tool to resolve this controversy. Therefore, the aim of this review to provide some basis on folliculogenesis and its regulation, to report the importance of in vitro culture, with an emphasis on follicle stimulating hormone (FSH), in assessing the role of homeopathic medicines in treating ovarian reproductive disorders and its use to improve reproductive biotechnologies.

The objective of this work was to estimate the intestinal digestibility of rumen-undegradable protein (RUDP) of feeds by a three-steps procedure. The evaluated feeds were the soybean meal, wheat meal, soybean peel, meat flour and fish flour. Firstly, the feeds were incubated in rumen during 16 hours to determine the rumen-undegradable protein. The residue was submitted to the digestion with pepsin solution during 1 hour, and pancreatic solution during 24 hours at 38ºC. Soon after, those residues were analyzed for total nitrogen (TN). The estimate of RUDP ranged of 22.07% to 91.30% and the intestinal digestibility of RUDP ranged from 35.13% to 80.67%. The soybean peel and the meat flour presented better intestinal digestibility, and the wheat meal presented the lowest digestibility. Although some formulation systems of diets for ruminant consider the intestinal digestibility of dietary protein as being constant, the data obtained in this work suggest that there are variation among the different feeds.
O objetivo deste trabalho foi estimar a digestibilidade intestinal da proteína não-degradada no rúmen (PNDR), de alimentos, por intermédio da técnica de três estágios. Os alimentos avaliados foram o farelo de soja, farelo de trigo, casca de soja, farinha de carne e farinha de peixe. Os alimentos foram inicialmente incubados no rúmen por 16 horas, para determinação da PNDR. O resíduo foi submetido à digestão com solução de pepsina, durante 1 hora, e solução de pancreatina a 38ºC, durante 24 horas. Em seguida, esses resíduos foram analisados para nitrogênio total (NT). A estimativa da PNDR variou de 22,07% a 91,30% e a digestibilidade intestinal da PNDR de 35,13% a 80,67%. A casca de soja e a farinha de carne apresentaram melhor digestibilidade intestinal e o farelo de trigo apresentou a menor digestibilidade. Embora alguns sistemas de adequação de dietas para ruminantes considerem a digestibilidade intestinal da proteína dietética como sendo constante, os resultados encontrados neste trabalho sugerem que a mesma possui variação entre os diferentes alimentos.
