RESUMO

This systematic review evaluated the efficacy of doxycycline in MMP inhibition, its antibacterial action, and other properties relevant to dental materials testing. The study protocol was registered at the Open Science Framework ( https://doi.org/10.17605/OSF.IO/ZVK2T ). Reporting was based on PRISMA statement. The search was carried out in the databases: PubMed, Scopus, Web of Science, Embase, Lilacs, and Google Scholar. Articles were restricted to Portuguese, English, and Spanish, with no date limit. In vitro studies were selected based on the following outcomes: DOX antibacterial and anti-metalloproteinase activity and its influence in physico-chemical properties. Two researchers independently selected the articles and collected the data. Of 1507 documents, 82 were fully evaluated and 21 were included. Different forms of doxycycline incorporation were found, both as free form and incorporated into carrier agents. The drug was tested as primers, incorporated in adhesive or glass ionomer cement. No studies were found that evaluated its incorporation in resin composite or resin cement. The results confirmed the therapeutic properties of the medication, with more significant results when incorporated in an adhesive. However, although promising, the use of this substance requires standardization in application methods and adopted concentrations, allowing for more direct comparisons between studies. Furthermore, long-term studies are interesting to conduct, ensuring biocompatibility and complete understanding of long-term effects on dental materials.

OBJECTIVE: The aim of this study was to evaluate the effect of an adhesive loaded with 0.2 % copper (Cu) and 5 % zinc oxide (ZnO) nanoparticles (Nps) on its adhesive properties and enzymatic activity at the hybrid layer ex vivo in a randomized clinical model. METHODS: Fifteen patients participated in this study, and a total of 30 third molars were used. Occlusal cavities (4 × 4 × 2 mm) were made in each tooth, and randomly divided into 2 groups: (i) Experimental group: commercial adhesive loaded with 0.2wt % CuNps and 5wt % ZnONps; and (ii) Control Group: non-loaded commercial adhesive. Teeth were restored with resin composite. Thirty days later, extractions were performed. Extracted teeth were longitudinally sectioned. Nps in powder were characterized by field emission scanning electron microscope (FE-SEM) and energy dispersive X-ray (EDX) analysis. Microtensile bond strength (µTBS), degree of conversion (DC), and nanoleakeage (NL) tests were executed. In situ zymography (Zym) was performed to evaluate the gelatinolytic activity at the hybrid layer. Student's t-test (α = 0.05) was applied for all tests. RESULTS: µTBS and DC did not show significant differences (p > 0.05) between both groups. However, NL and gelatinolytic activity at the hybrid layer showed significant values (p < 0.05) for experimental group in comparison with control group. CONCLUSION: The addition of 0.2 % CuNps and 5 % ZnONps to a universal adhesive decreases NL and gelatinolytic activity at the hybrid layer, without jeopardizing its adhesive properties. SIGNIFICANCE: This randomized clinical trial with ex vivo analysis demonstrate that a commercial adhesive modified with 0.2wt % Cu and 5wt % ZnO Nps that does not affect its adhesive properties, reducing gelatinolytic activity and nanoleakage at the hybrid layer, which should contribute to an improvement of long term bonding-dentine clinical performance.

This study evaluated the effect of different concentrations of alpha-tocopherol in gel form on fracture strength, hybrid layer formation, and microtensile bond strength of endodontically treated teeth bleached with 40% hydrogen peroxide (H2 O2 ). Sixty bovine incisors were randomized into one of six groups (n = 10 incisors per group) defined by the interventions carried out after endodontic treatment. In the control group, no additional intervention was carried out, while all teeth in the five intervention groups were bleached with 40% H2 O2 and subsequently treated with alpha-tocopherol at concentrations of 15% (15AT), 20% (20AT), or 25% (25AT), with 10% sodium ascorbate (10SA), or with nothing (40HP). Fracture strength was evaluated in a mechanical testing machine, hybrid layer formation was assessed using scanning electron microscopy, and bond strength was determined using microtensile bond-strength testing. Data were analyzed using Kruskal-Wallis and Dunn's tests. No statistically significant difference regarding fracture strength was observed among groups. Hybrid layer formation was greater in the 15AT group than in groups 40HP and 10SA. Teeth in groups 15AT, 20AT, and 25AT demonstrated higher bond strength than teeth in groups 40HP and 10SA. Alpha-tocopherol, preferably at 15%, effectively reverses the deleterious effects, of bleaching, on hybrid layer formation and bond strength to dentin.

OBJECTIVES: This study aimed at evaluating the microtensile bond strength (µTBS) and the resin-dentine ultramorphology (24 h and 10 months ageing) of contemporary universal adhesives applied in self-etch (SE) or etch-and-rinse (ER) mode. MATERIALS AND METHODS: Sixty-four sound human molars were collected and randomly allocated in 4 main experimental groups (n = 16) according to the adhesive system employed and subsequently divided into two subgroups depending on their application mode SE or ER (n = 8): ZipBond X (ZBX-SE; ZBX-ER), Prime and Bond Active (PBA-SE; PBA-ER), Clearfil Universal Bond Quick (CBQ-SE; CBQ-ER) or Scotchbond Universal (SCH-SE; SCH-ER). The specimens were cut into sticks with a cross-sectional area of approximately 0.9 mm2 and subjected to µTBS testing at 24 h or after 10 months of ageing in artificial saliva (AS). Five representative fractured specimens from each group were analysed using field-emission scanning electron microscopy (FE-SEM). Resin-dentine slabs (Ø 0.9mm2) from each experimental group were immersed in Rhodamine B and subsequently analysed using confocal microscopy analysis (CLSM). The µTBS results were analysed using a two-way ANOVA and Newman-Keuls multiple-comparison test (α = 0.05). RESULTS: ZBX, PBA and SCH exhibited greater µTBS values than CQB at 24 h in both SE and ER modes (p < 0.05). CQB showed a significant decrease in µTBS values after ageing both when used in SE and ER mode (p < 0.05). ZBX-ER exhibited no significant differences in the µTBS test after ageing (p > 0.05), while a significant drop in µTBS was seen in SCH-ER and APB-ER after 10-month ageing (p < 0.05). Clear signs of degradation were evident in the resin-dentine interface created with CQB regardless of the application mode or the ageing time. In APB-ER and SCH-ER groups, such signs of degradation were evident after ageing in AS. ZBX showed slight dye infiltration both when used in ER and SE mode. CONCLUSIONS: The long-term bonding performance of modern universal adhesives is usually influenced by the adhesive strategy employed; self-etching application should be prioritised during dentine bonding. Moreover, the use of shortened bonding protocols may compromise the quality of the resin-dentine interface and the bonding performance of most modern universal adhesives. CLINICAL RELEVANCE: The use of etch-and-rinse bonding procedures, as well as "shortened" application protocols should be eluded when using modern universal adhesives in dentine. However, new generation universal adhesives based on innovative chemical formulations may probably allow clinicians to achieve long-term bonding performance with such simplified system also when employed in ER mode.

The objective of this in vitro study was to assess the effects of two antioxidants (sodium ascorbate [SA] and alpha-tocopherol [AT]) on the adhesive interface and dentin bond strength immediately after bleaching with 38 % hydrogen peroxide (38HP) in endodontically-treated teeth. Two stages of experimentation were carried out. Bovine incisors were allocated into four groups (n = 10/group for each experiment): NB, non-bleached restored crowns; 38HP, bleached and immediately restored crowns; 38HP-SA, bleached crowns and SA use; and 38HP-AT, bleached crowns and AT use. Hybrid layer length in dentin (μm) and bond strength (MPa) were assessed with confocal microscopy laser and micro-shear bond strength (μSBS) test, respectively. Failure mode was determined by stereomicroscope. Data analysis was performed with analysis of variance (ANOVA) and Kruskal-Wallis, Dunn, Tukey, and Fisher-exact tests (a = 0.05). Higher values of hybrid layer length were observed similarly in the NB and 38HP-SA groups. The highest μSBS mean values were observed in the NB group (18 .51 ? 1.33), whereas the SBS values for 38HP-AT (1.68 ? 0.32) were similar to the 38HP group (1.61 ? 0.51) (p > 0.05) and significantly lower than the 38HP-SA group (5.78 ? 0.71). Adhesive failures were predominant in the 38HP and 38HP-AT groups. Cohesive and mixed failures were mostly observed in the NB and 38HP-SA groups, respectively. In conclusion, AT has no immediate effect on the hybrid layer formation and μSBS of dentin. Although SA promotes an increase in hybrid layer formation, it was not reflected in the μSBS values.


El objetivo de este estudio in vitro fue evaluar los efectos de dos antioxidantes (ascorbato de sodio [AS] y alfa-tocoferol [AT]) sobre la interfaz adhesiva y la fuerza de unión de la dentina inmediatamente después del blanqueamiento con peróxido de hidrógeno al 38 % (38HP) en endodoncia. -Dientes tratados. Se llevaron a cabo dos etapas de experimentación. Los incisivos bovinos se dividieron en cuatro grupos (n = 10 / grupo para cada experimento): NB, coronas restauradas no blanqueadas; 38HP, coronas blanqueadas y restauradas inmediatamente; 38HP- AS, coronas blanqueadas y uso SA; y 38HP-AT, coronas blanqueadas y uso de AT. La longitud de la capa híbrida en dentina (μm) y la fuerza de unión (MPa) se evaluaron con láser de microscopía confocal y la prueba de fuerza de unión por micro-cizallamiento (μSBS), respectivamente. El modo de falla se determinó mediante estereomicroscopio. El análisis de los datos se realizó con análisis de varianza (ANOVA) y pruebas de Kruskal-Wallis, Dunn, Tukey y Fisher (α = 0,05). De manera similar, se observaron valores más altos de longitud de capa híbrida en los grupos NB y 38HP-AS. Los valores medios más altos de μSBS se observaron en el grupo NB (18,51 ? 1,33), mientras que los valores de SBS para 38HP-AT (1,68 ? 0,32) fueron similares a los del grupo 38HP (1,61 ? 0,51) (p> 0,05) y significativamente más bajos que el grupo 38HP-AS (5,78 ? 0,71). Las fallas adhesivas fueron predominantes en los grupos de 38HP y 38HP-AT. Las fallas cohesivas y mixtas se observaron principalmente en los grupos NB y 38HP-AS, respectivamente. En conclusión, la AT no tiene un efecto inmediato sobre la formación de la capa híbrida y el μSBS de dentina. Aunque AS promueve un aumento en la formación de capas híbridas, no se refleja en los valores de μSBS.

OBJECTIVE: A silica-based nanofilm has been successfully deposited via Room-Temperature Atomic Layer Deposition (RT-ALD) on the surface of a glass. The purpose of this study was to evaluate the mechanical performance of a hybrid interface created between yttria-stabilized zirconia (Y-PSZ) transformed layer and silica-based nanofilm via RT-ALD. MATERIAL AND METHODS: Fully-sintered Y-PSZ (14 × 4.0 × 1.5 mm) specimens in different translucencies (MO, MT, LT; IPS e.max Zircad, Ivoclar Vivadent) were distributed in 5 groups: control (C - no treatment); hydrothermal treatment (HT- 15h, 134°C, 2 bar); alumina blasting (B - 50 µm Al2O3); RT-ALD silica deposition (S); HT followed by silica deposition (HTS). RT-ALD cycles consisted of the sequential exposure of specimens to tetramethoxysilane orthosilicate (TMOS - 60s) and ammonium hydroxide (NH4OH - 10 min) vapors in 40 cycles. Mechanical performance was analyzed by flexural strength (FS) (n = 10) and fatigue failure load (staircase method; n = 20) tests. Surface hardness (H) and Young's modulus (YM) were analyzed by nanoindentation. For surface chemical and topographical characterization, X-ray photoelectron spectroscopy (XPS) and scanning electron microscopy (SEM) were performed. Data from surface H, YM, FS, and fatigue limit (FL) were analyzed by two-way analysis of variance (ANOVA). RESULTS: The interaction between material and treatment had a significant effect on FS (p < 0.001). The FS values ranged from 436.23 MPa to 856.65 MPa. HT resulted in the highest FS (856.65 MPa) for LT and the lowest FS (436.23 MPa) for MO zirconia. For all materials, S and B treatments resulted in similar FS values (p > 0.410). S did not affect FL when compared to the C group (p > 0.277) for any material investigated. HTS resulted in higher FL than S for LT and MO materials (p < 0.001). Surface hardness and modulus were similar between control and S-treated specimens for all materials analyzed. XPS analysis showed homogeneous silica content after 20 and 40 RT-ALD cycles, and SEM did not show significant changes in surface morphology between C and S-treated specimens. CONCLUSION: RT-ALD resulted in effective silica deposition without any deleterious effect on zirconia-based materials mechanical properties. Alumina blasting promoted higher alteration on surface topography. HT prior to S resulted in superior FL (for MO and MT) and flexural strength (MO) for some of the materials investigated.

OBJECTIVES: To evaluate the effect of electric current application on the resin composite-tooth bond strength and hybrid layer of three adhesive systems light-cured by two light-curing units (LCUs). MATERIALS AND METHODS: Human molar teeth were distributed into 12 groups (n=6). Three adhesive systems were used: two-step etch-and-rinse (SB2; Adper Single Bond 2, 3M ESPE); two-step self-etch (CSE; Clearfil SE Bond, Kuraray); and one-step self-etch (SBU; Single Bond Universal, 3M ESPE) applied with (50µA) and without (control; conventional application) electric current, and light-cured with different LCUs. Resin composite blocks (Filtek Z350XT, 3M ESPE) were produced and cut into sticks (~1mm2) for microtensile bond strength (µTBS). Fracture patterns were analyzed on stereomicroscope and classified as cohesive-dentin, cohesive-resin, adhesive, or mixed. Specimens were prepared for scanning electron microscope observation. The hybrid layer analysis was carried out using a confocal laser scanning microscopy (n=2). Data were submitted to three-way ANOVA followed by Tukey's post hoc test (α=0.05). RESULTS: The electric current increased the µTBS for all adhesive systems light-cured with single-emission peak and multiple-emission peak LCUs. Both LCUs presented similar µTBS values. CSE applied under electric current showed the highest µTBS mean values. The adhesive failure pattern was more frequently observed in all groups. The electric current formed long resin tags for all adhesive systems. CONCLUSIONS: The adhesive systems applied under electric current increased the bond strength using single-emission peak and multiple-emission peak LCUs. CLINICAL RELEVANCE: Electric current at 50µA applied throughout the dentin is a safe mode and results in better impregnation of the adhesive systems.

Matrix metalloproteinases (MMPs) are enzymes that can degrade collagen in hybrid layer and reduce the longevity of adhesive restorations. As scientific understanding of the MMPs has advanced, useful strategies focusing on preventing these enzymes' actions by MMP inhibitors have quickly developed in many medical fields. However, in restorative dentistry, it is still not well established. This paper is an overview of the strategies to inhibit MMPs that can achieve a long-lasting material-tooth adhesion. Literature search was performed comprehensively using the electronic databases: PubMed, ScienceDirect and Scopus including articles from May 2007 to December 2019 and the main search terms were "matrix metalloproteinases", "collagen", and "dentin" and "hybrid layer". MMPs typical structure consists of several distinct domains. MMP inhibitors can be divided into 2 main groups: synthetic (synthetic-peptides, non-peptide molecules and compounds, tetracyclines, metallic ions, and others) and natural bioactive inhibitors mainly flavonoids. Selective inhibitors of MMPs promise to be the future for specific targeting of preventing dentin proteolysis. The knowledge about MMPs functionality should be considered to synthesize drugs capable to efficiently and selectively block MMPs chemical routes targeting their inactivation in order to overcome the current limitations of the therapeutic use of MMPs inhibitors, i.e., easy clinical application and long-lasting effect.

Objetivos: Evaluar la resistencia de unión a la microtracción en dentina humana de un sistema adhesivo universal con clorhexidina en su composición, en modo de grabado y lavado en 2 pasos, y en modo de autograbado. Metodología: 20 terceros molares divididos aleatoriamente en 4 grupos según el tipo de sistema adhesivo utilizado (Single Bond Universal®, 3MESPE y Peak Universal Bond®, Ultradent Products) y modo de uso (grabado total y autograbado). Se confeccionaron cuerpos de prueba sometidos al ensayo de microtracción utilizando una máquina de ensayos universales. Resultados: No se encontraron diferencias estadísticamente significativas entre los cuatro grupos estudiados. Conclusiones: La resistencia de unión de una resina compuesta a dentina humana no fue afectada por el uso de un sistema adhesivo universal que contiene clorhexidina en su composición aplicado en los modos de grabado total y autograbado


Objectives: To evaluate the microtensile bond strength to human dentin of chlorhexidine containing universal adhesive system applied in the total-etch and selfetch modes. Methods: Twenty third molars were randomly divided into four groups according to the universal adhesive system (Single Bond Universal ®, 3M ESPE and Peak Universal Bond®, Ultradent) and application mode used (total-etch and self-etch). Specimens were prepared and subjected to a microtensile bond strength using a MTS SANS universal testing machine. Results: No statistically significant differences were found in the microtensile bond strength between the four groups studied. Conclusions: The bond strength of a composite resin to human dentin was not affected by the use of a universal adhesive system with chlorhexidine in its composition applied in the total-etch and self-etch modes.


Objetivos: Avaliar a resistência de união á microtração na dentina humana de um sistema adesivo universal com clorexidina na sua composição, no modo de condicionamento total em dois passos e no modo autocondicionante. Metodologia: 20 terceiros molares foram divididos aleatoriamente em 4 grupos, de acordo com o tipo de sistema adesivo utilizado (Single Bond Universal®, 3MESPE e Peak Universal Bond®, Ultradent Products) e modo de uso (condicionamento total e autocondicionante). Os corpos de prova criados foram submetidos ao teste de microtração utilizando uma máquina de ensaios universal. Resultados: Não foram encontradas diferenças estatisticamente significativas entre os quatro grupos estudados. Conclusões: A resistência de união da resina composta à dentina humana não foi afetada pelo uso de um sistema adesivo universal contendo clorexidina em sua composição, aplicado nos modos de condicionamento total ou autocondicionamento.

This study evaluated the effect of 10% sodium hypochlorite (NaOCl) as deproteinizing agent and storage media on bond strength (BS) of two etch-and-rinse adhesive systems to dentin. Twenty-eight sound extracted human third molars were divided in four groups (n = 7), according to dentin treatment (conventional etching or etching followed by 10% NaOCl application) and adhesive systems (GB-Gluma 2Bond and OS-One-Step). After dentin treatments and adhesive application, a composite block was built-up on dentin surface and teeth were serially sectioned to obtain bonded sticks specimens. The sticks were submitted to three aging conditions: (24H) 24 hr in water (immediate), (SH) 3 hr of NaOCl accelerated-aging or (1Y) 1 year of water storage. Afterward, submitted to microtensile bond strength test (µTBS), failure modes and adhesive interfaces analyzes. Data were analyzed by two-way analysis of variance (ANOVA) and Tukey's test (α = .05). Dentin deproteinization before bonding significantly reduced µTBS for GB-treated group (p < .05), regardless the aging conditions. Water storage for 1 year (1Y) and NaOCl accelerated-aging (SH) decreased µTBS for both adhesives. Yet, the groups stored in NaOCl (SH) exhibited the lowest BS results (p < .05). Bond strength of deproteinized dentin was dependent on the adhesive system composition and NaOCl accelerated-aging promoted decreased bond strength and further degradation than water storage for 1 year.

OBJECTIVE: To evaluate the influence of the degree of dentin moisture on interfacial ultramorphology and bond strength (µTBS) of universal adhesives. MATERIALS AND METHODS: Futurabond U (FBU), Scotchbond Universal (SBU), Adhese Universal (ADU), and Prime&Bond active (PBA) were used. After acid-etching, moist or over-dried dentin surfaces were tested. Teeth were restored for scanning and transmission electron microscopy (n = 3) and µTBS evaluation (n = 5). µTBS results were analyzed by two-way ANOVA and Tukey. RESULTS: For moist dentin, a well-formed hybrid layer (HL) was observed. However, when applied to over-dried dentin, remarkable differences were observed. Defects, gaps, and reduced HL thickness were observed mainly for ADU and FBU. When applied to wet dentin, µTBS values were similar for all adhesives, except for FBU, which was significantly lower. When applied to over-dried dentin, PBA presented the highest µTBS values, followed by SBU, ADU, and FBU. ADU presented significantly lower µTBS when applied to over-dried dentin. CONCLUSION: PBA, SBU, and FBU µTBS values were not sensitive to the degree of moisture. Even though application to over-dried dentin revealed defects, gaps and reduced HL thickness for SBU, ADU, and FBU, µTBS analysis only revealed a significant reduction for ADU. CLINICAL SIGNIFICANCE: Universal adhesives can be applied in either self-etching or etch-and-rinse mode. However, clinicians are not aware which universal adhesives should be strictly applied on a moist dentin for bonding in the etch-and-rinse mode.

Restorative procedures associated with bioglasses have shown to be a strategy to satisfy the contemporary concept of minimally invasive dentistry. Thus, the aim of this study was to evaluate bond strength to dentin treated by two different methods of biosilicate microparticle application. Dentin surfaces from 30 sound human molars were exposed and randomly assigned into three groups (n = 10) according to the surface treatment: (1) blasting with biosilicate microparticles (distance = 1 cm/pressure = 5 bar/time = 1 min); (2) 10% biosilicate microparticles paste; and (3) control (no treatment). After, dentin surfaces were restored with self-etch adhesive (Adper Easy Bond) and nanofilled composite (Filtek Z350). Specimens were sectioned perpendicularly to the adhesive interface to obtain sticks (cross-section area = 1 mm²), which were submitted to microtensile test (0.5 mm/min; 50 kgf). Data were analyzed by ANOVA and Tukey's test (α = 5%). Dentin/adhesive interfaces were morphologically analyzed by scanning electron microscopy (SEM). Data analysis showed that biosilicate-treated groups reached similar results (p > 0.05) and both of them demonstrated higher values (p < 0.05) than control group. SEM micrographs revealed hybridization with clear resin tags and no separation between resin-dentin adhesive interfaces. Within the limitations of this study, surface treatment with biosilicate positively influenced the adhesion to dentin and does not alter the morphology of the adhesive interface.

OBJECTIVES: To evaluate the effect of desensitizing agent containing calcium phosphate nanoparticles on the bond strength of etch-and-rinse adhesive system (Scotchbond Multi-Purpose), presence of precipitate, dentinal tubule obliteration and hybrid layer formation in dentin in comparison with potassium nitrate plus sodium fluoride or strontium chloride compounds. METHODS: 150 bovine incisors were treated with (n = 10): G1, Desensibilize Nano P (Ca3(PO4)2+5%KNO3+0.9%NaF); G2, Desensibilize (10%SrCl2+5%KNO3); G3, Desensibilize KF2% (5%KNO3+0.2%NaF); G4, Ultra EZ (3%KNO3+0.25%NaF) and G5, no treated (control). Scanning electron microscopy was used to assess the incidence of precipitates (500×) and obliterated dentinal tubule counts (1.000×). The adhesive system was used after all desensitization treatments. The bond strength (n = 40) and the fracture pattern were evaluated. Confocal laser microscopy was used to quantify the hybrid layer formation in dentin. RESULTS: G1 and G2 presented higher adhesive system bond strength (MPa) than G4 and G5, however no significant differences were observed in comparison with G3. Cohesive fracture was frequently found: G1 (58.5%), G2 (51.3%) and G3 (43.8%). G1 showed the highest incidence of precipitates and the highest number of blocked dentinal tubules. G1 and G2 presented similar hybrid layer formation and the highest hybrid layer formation values. CONCLUSIONS: Desensibilize Nano P (G1) favored the bond strength of the adhesive system to dentin, increased the precipitation of residues, obliteration of dentinal tubules, and hybrid layer formation in comparison with other agents. CLINICAL RELEVANCE: Desensitizers promote dentin obliteration, however, may affect dentin bonding.

Abstract The aim was to evaluate the influence of different dentin preparation mode in the smear layer characteristics (SL), hybrid layer (HL), and microtensile bond strength (µTBS) to dentin with two resin cements. The occlusal dentin of 120 third molars was exposed. The teeth were divided into 4 groups (n=30) according to the dentin preparation mode: 1- fine grain diamond bur; 2- coarse grain diamond bur; 3- multi laminate carbide steel bur; and, 4- ultrasonic CV Dentus diamond bur. Each treated dentin group was divided into 2 sub-groups (n=15) according to the resin cement: (1) RelyX U200 and (2) RelyX ARC. Resin composite blocks were cemented on dentin. After storage at 37o C for 24 h, beams with a cross section area of 1.0 mm2 were obtained, and tested in a universal testing machine at a crosshead speed of 0.5 mm/min. Two additional teeth for each sub-group were prepared to analyze the SL and HL on a scanning electron microscopy. According to Kruskal-Wallis test and Dunn tests, there was no significant difference in µTBS among the rotary instruments within each resin cement group. RelyX ARC obtained higher µTBS values compared to RelyX U200 (p<0.05). RelyX ARC formed evident HL, which was not observed for RelyX U200. The dentin mode preparation did not influence the µTBS of the resin cements. The SL was different for all instruments. The cementing agent is more determinant in the hybrid layer formation and bond strength to dentin than the instruments applied on dentin.


Resumo O objetivo deste estudo foi avaliar a influência de diferentes modos de preparação da dentina nas características da smear layer (SL), da camada híbrida (CH) e na resistência de união à microtração (RUµT) à dentina com dois cimentos resinosos. A dentina oclusal de 120 terceiros molares humanos foi exposta. Os dentes foram divididos em 4 grupos (n=30) de acordo com o modo de preparação da dentina: 1 - ponta diamantada de granulação fina; 2 - ponta diamantada de granulação grossa; 3 - ponta de carboneto de aço multilaminada; 4 - ponta diamantada ultrassônica. Cada grupo foi subdividido em dois subgrupos (n=15) de acordo com o cimento resinoso: (1) RelyX U200 e (2) RelyX ARC. Blocos de resina composta foram cimentados na dentina. Após armazenamento a 37 °C por 24 h, as amostras foram cortadas para obter palitos com área de 1,0 mm2, os quais foram testados em máquina de ensaio universal com velocidade de 0,5 mm/min. Dois dentes adicionais por subgrupo foram preparados para analisar as características da SL e da CH em microscópio eletrônico de varredura. De acordo com o teste de Kruskal-Wallis e o teste de Dunn, não houve diferença significativa na RUµT entre os instrumentos rotatórios para cada cimento resinoso. RelyX ARC obteve maiores valores de RUµT em comparação ao RelyX U200 (p<0,05). RelyX ARC formou uma CH evidente, o que não foi observado para o RelyX U200. O modo de preparação da dentina não influenciou na RUµT dos cimentos resinosos. A SL foi diferente para todos os instrumentos. O agente de cimentação foi mais determinante na formação da camada híbrida e na resistência de união do que os instrumentos utilizados para preparar a dentina.

Antecedentes: La formación de la capa híbrida debe ser proporcional a la capacidad de los cementos para retener con éxito postes de fibra de vidrio en el interior del conducto preparado endodónticamente. Objetivo: Medir el espesor de la capa híbrida en los tercios coronal y apical as usar cementos autograbadores de uno y dos pasos. Métodos: Se realizó un estudio experimental con 30 premolares humanos que se trataron endodónticamente y se asignaron a tres grupos. Se cementaron postes de fibra de vidrio con tres cementos siguiendo las instrucciones de los fabricantes. Se realizaron cortes coronales y apicales y se cuantificó el grosor de la capa híbrida por microscopía electrónica de barrido. Los datos se analizaron con las pruebas estadísticas H de Kruskal-Wallis y U de Mann-Whitney (p<0,05). Resultados: El promedio del grosor de la capa híbrida fue de 0,74 mm con Multilink N® (grupo 1), 0,24 mm con RelyX Ultimate® (grupo 2) y 0,03 mm con RelyX U200® (grupo 3) (p = 0,0092). Los promedios en los tercios coronal y apical fueron: 1,29 mm y 0,19 mm para el grupo 1; 0,33 mm y 0,14 mm para el grupo 2 y 0,26 mm y 0,23 mm para el grupo 3, diferencias que fueron estadísticamente significativas (p = 0,043). Conclusiones: El espesor de la capa híbrida formada con el uso de los cementos de dos pasos fue mayor que con el cemento de un solo paso, especialmente en la porción coronal.


Background: Hybrid layer formation should be proportional to cement properties to successfully retain fiberglass posts into root canals. Purpose: To measure of hybrid layer thickness in coronal and apical thirds after using one-step and two-step self-etching cements. Methods: This experimental study used 30 human premolars with root-canal treatments, which were assigned into three groups. Fiberglass posts were cemented with three types of self-etching cements following manufacturers' instructions. Roots were cut in the coronal and apical thirds and hybrid layer thickness was quantified through scanning electron microscopy. Data were analyzed with Kruskal-Wallis and Mann-Whitney tests (p < 0.05). Results: Average thickness of the hybrid layer was 0.74 µm with Multilink N™ (group 1), 0.24 µm with RelyX Ultimate™ (group 2), and 0.03 µm with RelyX U200™ (group 3) (p = 0.0092). Average thickness in coronal and apical thirds were: 1.29 µm and 0.19 µm for group 1; 0.33 µm and 0.14 µm for group 2; and 0.26 µm and 0.23 µm for group 3, differences that were statistically significant (p = 0.043). Conclusions: The thickness of the hybrid layer formed after suing two-step cements was greater than that of one-step cement, especially in the tooth coronal third.

OBJECTIVES: To evaluate the effect of intrinsic water permeation on the microtensile bond strengths of different adhesive systems to dentine and the quality of resin-dentine interfaces. METHODS: Ninety-six non-carious human third molars were divided into 4 groups: Clearfil S3 Bond Plus (CSBP; Kuraray); Clearfil S3 Bond (C3S; Kuraray); iBond Self-Etch (IB; Heraeus-Kulzer) and Prime&Bond NT (PB, control etch-and-rinse adhesive, Dentply-Sirona). For each adhesive, specimens from one subgroup (N=10) were bonded using zero pulpal pressure, while specimens from the other subgroup (N=10) were bonded using 15cm water pressure (PP). Each bonded tooth was sectioned into 1×1mm sticks and stressed to failure. Data were analysed using two-way ANOVA and Holm-Sidak pairwise comparisons to examine the effects of "adhesive", "pulpal pressure" and their interaction on bond strength (α=0.05). Representative fractured sticks were examined by SEM. The remaining tooth slabs in each subgroup were used for TEM and CLSM. RESULTS: Microtensile bond strengths (mean±SD; in MPa) were: 33.4±6.9 (CSBP), 33.2±4.7 (CSBP-PP), 35.0±8.6 (C3S), 25.5±7.3 (C3S-PP), 18.4±4.0 (IB), 16.5±6.9 (IB-PP), 28.2±5.5 (PB), 20.5±7.2 (PB-PP). "Adhesive-type" (P<0.001), "pulpal-pressure" (P<0.001) and their interactions (P<0.001) significantly affected bond strength results. No difference between no-PP and PP subgroups was found for CSBP and IB (P>0.05). Water droplets were identified along the resin-dentine interface for IB, IB-PP and C3S-PP. CONCLUSION: IB exhibits water sensitivity when bonding is performed with/without pulpal pressure. C3S exhibits water sensitivity when bonding is performed with pulpal pressure. CSBP does not exhibit water sensitivity when bonding is performed with/without pulpal pressure. CLINICAL SIGNIFICANCE: Intrinsic water permeation during bonding procedures significantly affects bond strength results and the resin-dentine interface of contemporary single-bottle self-etch dentine adhesive systems.

Degradation of the hybrid layer created in dentin by dentin adhesives is caused by enzyme activities present within the dentin matrix that destroy unprotected collagen fibrils. The aim of the present study was to evaluate the effect of a one-step self-etch adhesive system on dentinal matrix metalloproteinases 2 and 4 (MMP-2 and MMP-9, respectively) using in situ zymography and an enzymatic activity assay. The null hypothesis tested was that there are no differences in the activities of dentinal MMPs before and after treatment with a one-step adhesive system. The MMP-2 and MMP-9 activities in dentin treated with the one-step adhesive, Adper Easy Bond, were quantified using an enzymatic activity assay system. The MMP activities within the hybrid layer created by the one-step adhesive tested were also evaluated using in situ zymography. The enzymatic assay revealed an increase in MMP-2 and MMP-9 activities after treatment with adhesive. In situ zymography indicated that gelatinolytic activity is present within the hybrid layer created with the one-step self-etch adhesive. The host-derived gelatinases were localized within the hybrid layer and remained active after the bonding procedure. It is concluded that the one-step self-etch adhesive investigated activates endogenous MMP-2 and MMP-9 with the dentin matrix, which may cause collagen degradation over time.

AIM: This study evaluated the influence of acid-etching time on collagen exposure in adhesive interfaces established on primary and permanent dentin. MATERIALS AND METHODS: Flat dentin surfaces were produced on sound primary molars and premolars (n = 8). The surfaces were divided into mesial and distal halves, and each half was etched with phosphoric acid for 5 or 15 seconds. The teeth were randomly allocated into two groups according to the adhesive system applied: Prime & Bond NT or Prime & Bond 2.1. After the adhesive application, the specimens were processed for Goldner's trichrome staining. The thickness of the uninfiltrated collagen zone (UCZ) in the hybrid layer was measured under optical microscopy. Data were analyzed by analysis of variance and Tukey tests (α = 0.05). RESULTS: The thickness of UCZ was adhesive dependent. Within the same substrate, the specimens treated with Prime & Bond 2.1 presented thicker UCZ when etched for 15 seconds. Collagen exposure was significantly higher for the primary teeth etched for 5 seconds and treated with Prime & Bond 2.1. CONCLUSION: The thickness of UCZ in hybrid layers is directly affected by acid-etching time and by the adhesive system applied. Primary dentin seems to be more susceptible to collagen exposure than is permanent dentin. CLINICAL SIGNIFICANCE: Both acid-etching time and adhesive system can influence the amount of exposed collagen interfering on resin-dentin bond quality, especially on primary dentin.

OBJECTIVES: This study evaluated the bond strength of a two-step conventional adhesive system to dentin pretreated with 2.5 % titanium tetrafluoride (TiF(4)) according to application timing (before or after acid conditioning) and primer/adhesive application method (active or passive). MATERIALS AND METHODS: Dentin surfaces were randomly treated with different adhesive procedures (n = 6): etching with phosphoric acid (PA) + primer/adhesive (Adper Single Bond 2/3M ESPE) actively applied; PA + primer/adhesive passively applied; TiF(4) before PA + primer/adhesive actively applied; TiF(4) before PA + primer/adhesive passively applied; TiF(4) after PA + primer/adhesive actively applied; and TiF(4) after PA + primer/adhesive passively applied. A composite block was built onto the tooth, which was sectioned into sticks (adhesive area of approximately 1 mm(2)). Microtensile bond strength tests and the failure mode were determined. RESULTS: Two-way ANOVA revealed no significant interaction between TiF(4) application timing and primer/adhesive application method (p = 0.184). The use of TiF(4), before or after PA significantly increased bond strength values (p < 0.001). There was no significant difference in bond strength values when using TiF4 before or after PA. The primer/adhesive application method had no influence on bond strength, regardless of whether TiF(4) was used (p = 0.906). Failure mode was predominantly adhesive. CONCLUSIONS: The use of TiF(4) promoted higher immediate bond strength to dentin. The conventional adhesive system may be applied either actively or passively, regardless of TiF(4) application timing. CLINICAL SIGNIFICANCE: Pretreatment with TiF(4) increased bond strength in a conventional two-step adhesive system to dentin, regardless of the primer/adhesive application method and the timing of dentin pretreatment.

The aim of this study was to evaluate the effect of 2% chlorhexidine (CHX) application during the bonding protocol on microshear bond strength of two adhesive systems, after storage in different media. Seventy-two human molars had their crowns cut in half and embedded in PVC cylinders with acrylic resin. The specimens were randomly divided into experimental groups (n=12) according to the adhesive system (Ambar and Single Bond 2), use of CHX in the bonding protocol, and time interval (24 h and 15 days) in the storage media (distilled water, mineral oil and 1% sodium hypochlorite - NaOCl). Adhesive systems were applied in accordance to manufacturers' recommendations, with or without the use of CHX, and resin composite (Z350 XT) cylinders were placed on the hybridized dentin. After photoactivation, the specimens were stored in distilled water, mineral oil and 1% NaOCl for 24 h and 15 days. Microshear bond strength was determined at a crosshead speed of 0.5 mm/min until fracture. The bond strength data were analyzed statistically by 4-way ANOVA and Tukey's test (α=5%). Use of CHX in the bonding protocol did not cause loss of bond strength in any of the evaluated situations, irrespective of time and storage medium. The storage medium had no influence on bond strength values after 15 days when the bond protocol without CHX application was used. However, the use of CHX in the protocol influenced negatively the bond strength values for Single Bond 2 after 15 days storage in distilled water and 1% NaOCl.


Resumo O objetivo deste trabalho foi avaliar o efeito da aplicação de clorexidina 2% (CHX) na resistência de união (RU) ao microcisalhamento de dois sistemas adesivos após armazenagem em diferentes meios. Setenta e duas coroas de molares humanos foram cortadas e incluídas em cilindros de PVC. Os espécimes foram distribuídos aleatoriamente nos grupos (n=12), segundo o sistema adesivo (Ambar e Single Bond 2), uso ou não de CHX e o tempo (24 h e 15 dias) e meio de armazenamento (água destilada, óleo mineral e hipoclorito de sódio 1% - NaOCl). Os sistemas adesivos foram aplicados de acordo com as recomendações do fabricante, com e sem o uso de CHX, e cilindros de resina composta (Z350 XT) foram colocados sobre a dentina já hibridizada. Após a fotoativação, os espécimes foram armazenados em um dos três meios (água destilada, óleo mineral e NaOCl a 1%) a 37 ºC por 24 h e 15 dias. RU foi determinada pelo ensaio de microcisalhamento, com velocidade de 0,5 mm/min. Os dados foram analisados por ANOVA a quatro fatores e teste de Tukey (α=5%). O uso da CHX não provocou perda de RU em nenhuma das situações avaliadas, independentemente do tempo e meio de armazenamento. Não houve influência do meio de armazenamento na RU após 15 dias quando foi utilizado o protocolo adesivo sem a aplicação de CHX. Porém, a utilização de CHX influenciou negativamente os valores de RU para o Single Bond 2 após 15 dias de armazenamento em água destilada e NaOCl a 1%.
