In vitro tissue culture can be an alternative method for endangered species propagation, biodiversity conservation and secondary metabolite studies. Paratecoma peroba (Record) Kuhlm. (Bignoniaceae) is an endemic and endangered Brazilian species. This work aimed to establish in vitro morphogenesis and callus induction and to perform a phytochemical analysis of P. peroba callus extract. Higher seed germination (43%) was obtained in Wood Plant Medium culture without activated charcoal (AC). Combination of 5 µM benzyladenine + 10 µM gibberellic acid, without AC, resulted in a higher number of shoots (2 shoots/explant). A callus culture was stabilised from zygotic embryos using 2,4-dichlorophenoxyacetic acid. A callus methanolic extract was used for phytochemical analysis. The isolated substance was identified as tiliroside (kaempferol 3-O-ß-D-(6''-O-E-p-coumaroyl)-glucopyranoside) by NMR and quantified in callus and leaf extracts by HPLC. This study adds to the chemical knowledge of this species and it is the first report of a flavonol in Paratecoma.

Information from Pereskia aculeata Miller (Cactaceae) responses regarding the portion of the stem cuttings branch parental plants and the shading level need to be investigated in order to establish techniques for their producing seedlings. We aimed was to evaluate the effect of cutting types and shading levels in the production of P. aculeata seedlings. We studied three cutting types, collected from different portions of the stem cuttings branch: herbaceous, semi-hardwood, and hardwood, which were arranged under two shading levels: 0% (full sun) or 50% (shade). The selected parent plants had an adequate and vigorous phytosanitary aspect. After 90 days after the cuttings, the seedlings were evaluated regarding survival, growth indicators, biomass production and partitioning, and allometric indices. Seedlings from hardwood cuttings and produced under 0% shading showed higher survival. The largest number of sprouts occurred in seedlings of semi-hardwood and hardwood cuttings. The highest leaf area values ​​occurred in seedlings of semi-hardwood and hardwood cuttings under 0% shading. Seedlings from hardwood cuttings had higher biomass allocation in the roots under 50% shading. Seedlings of herbaceous and semi-hardwood portion allocated values ≥ 70% of the biomass in the aerial part. Seedlings of species are able to adjust to different shading levels as a plasticity mechanism. For the production of P. aculeata seedlings, the hardwood portion of the stem cuttings branch under full sun cultivation is recommended. In addition, semi-hardwood cuttings grown under 50% shading can also be used for seedlings production.


Informações das respostas da Pereskia aculeata Miller. (Cactaceae) quanto à porção da estaca do ramo da planta matriz e o nível de sombreamento precisam ser investigadas com o intuito de estabelecer protocolos para sua produção de mudas. Objetivamos avaliar o efeito do tipo de estaca e níveis de sombreamento na produção de mudas de P. aculeata. Estudamos três tipos de estacas baseados na porção de coleta do ramo: herbácea, semi-lenhosa e lenhosa, acondicionadas sob dois níveis de sombreamento: 0% (pleno sol) ou 50% (sombra). As plantas matrizes selecionadas tiveram aspecto fitossanitário adequado e eram vigorosas. Após 90 dias da estaquia, as mudas foram avaliadas quanto a sobrevivência, indicadores de crescimento, produção e particionamento de biomassa e índices alométricos. Mudas provenientes de estacas lenhosas e produzidas sob 0% de sombreamento tiveram maior sobrevivência. Os maiores números de brotos ocorreram nas mudas de estacas da porção semi-lenhosas e lenhosa. As maiores áreas foliares e radiculares ocorreram nas mudas de estacas semi-lenhosas e lenhosas sob 0% de sombreamento. Mudas provenientes de estacas da porção lenhosa tiveram maior alocação de biomassa nas raízes sob 50% de sombreamento. Mudas de estacas da porção herbáceas e semilenhosas alocaram valores ≥ 70% de biomassa da parte aérea. As mudas da espécie são capazes de se ajustar aos diferentes níveis de sombreamento como mecanismo de plasticidade. Para a produção de mudas de P. aculeata¸ recomenda-se utilizar a porção lenhosa do ramo do caule sob pleno sol. Além disso, estacas semi-lenhosas crescidas sob 50% de sombreamento podem ser utilizadas para a produção de mudas.

African mahogany species (Khaya spp.) have proven to be promising in the Brazilian forestry scenario, replacing native mahogany owing to their medium-fast growth and relevant timber value. This study aimed to carry out forest inventory and assessments of a Khaya grandifoliola plantation in the first years after planting, test hypsometric models to describe tree growth, and identify the maximum commercial stem yield (i.e., greater than 6 m in height). The stand was located in the municipality of Piracanjuba (GO), where seedlings of seed origin were used. Twenty random plots with a 15 m radius were allocated, and the total height (HT), stem height (HS), diameter at breast height (DBH), crown area, and forest canopy were measured. Four hypsometric models were employed in this study. The best equation was selected based on determination coefficients and standard errors. Further, the models were cross-validated to evaluate predictability and bias. At four years of planting, the largest class of HS was found to range from 3.1 to 4.1 m, and most trees had a DBH ranging from 0.084 to 0.126 m. The percentage of trees with stems > 6 m was 8.35%. The linear model ensured more consistent results for estimating HT, while the quadratic and Weibull models led to more consistent results for HS. By using models, stem measurements can be measured based on DBH, ultimately aiding the selection of stem management strategies for the growth of forests with greater commercial value.

Lignin is a complex phenolic biopolymer present in plant cell walls and a by-product of the cellulose pulping industry. Lignin has functional properties, such as antioxidant activity, that make it a potential natural active ingredient for health-care products. However, not all safety aspects of lignin fractions have been adequately investigated. Herein, we evaluated the antioxidant and genotoxic potential of two hardwood kraft lignins (F3 and F5). The chemical characterization of F3 and F5 demonstrated their thermal stability and the presence of different phenolic units, while the DPPH assay confirmed the antioxidant activity of these lignin fractions. Despite being antioxidants in the DPPH assay, F3 and F5 were capable of generating intracellular reactive oxygen species (ROS) and subsequently causing oxidative DNA damage (Comet assay) in HepG2 cells. The biological relevance of the DPPH assay might be uncertain in some cases; therefore, we suggest combining in chemico tests with biological system-based tests to determine efficacy and safety levels of lignins and define appropriate applications of lignins for consumer products. Moreover, kraft lignins obtained by acid precipitation may pose risks to human health; however, as genotoxicity is not the sole endpoint of toxicity required in hazard assessments, additional toxicological evaluations are needed.

BACKGROUND: Myracrodruon urundeuva is a hardwood tree, and its bark, heartwood and leaf contain lectins (MuBL, MuHL and MuLL respectively) with termiticidal activity against Nasutitermes corniger. In this work, the effects of these lectins on the midgut of N. corniger workers were evaluated. RESULTS: The insects were supplied with an artificial diet containing the lectins at their respective LC50 (previously determined). At 48 h after treatment, the midguts were dissected and fixed for histopathology analyses. Toluidine-blue-stained midguts from lectin-treated workers showed disorganisation, with the presence of debris in the lumen and the absence of brush border. Fluorescence microscopy revealed that the numbers of digestive and proliferating cells were lower in lectin-treated individuals than in the control, and caspase-3 staining confirmed the occurrence of cell apoptosis. Enteroendocrine cells were not seen in the treated individuals. The midguts from treated insects showed greater staining for peroxidase than the control, suggesting that the lectins caused oxidative stress. Staining with wheat germ agglutinin conjugated to FITC revealed that the lectins interfered with the integrity of the peritrophic matrix. CONCLUSION: This study showed that termiticidal lectins from M. urundeuva cause severe injuries, oxidative stress and cell death in the midgut of N. corniger workers. © 2016 Society of Chemical Industry.

The larvicidal activities of extracts of three hardwood species (Hymenaea stigonorcapa, Anadenanthera colubrina and Bowdichia virgilioides) against 4th instar larvae of Aedes aegypti were evaluated using WHO guidelines. Extracts of H. stignocarpa and A. colubrina showed weak activity. The highest larvicidal effect was obtained with the cyclohexane extract of the heartwood of B. virgilioides, which caused 100% mortality at concentrations at 50 and 100 µg/mL. Fraction toluene/EtOAc (8:2) from this extract showed larvicidal activity (LC50 = 34.90 ± 1.27 µg/mL). A mixture of two compounds identified as medicarpin and maackiain exhibited a very good larvicidal activity (sub-fraction 2, LC50 = 17.5 ± 1.87 µg/mL) and maackiain showed to be a strong larvicidal compound (LC50 = 21.95 ± 1.34 µg/mL). This result can be of value in the search for new natural larvicidal compounds from other hardwood plant extracts and presents the first report of B. virgilioides being used to control a mosquito vector.
