TRPM4 is a calcium activated non-selective cation channel, impermeable to Ca2+, in neurons it has been implicated in the regulation of the excitability and in the persistent firing. Cholinergic stimulation is also implicated in changes in excitability that leads neurons to an increased firing frequency, however it is not clear whether TRPM4 is involved in the cholinergic-induced increase in firing frequency. Here using a combination of patch clamp electrophysiology, Ca2+ imaging, immunofluorescence, fluorescence recovery after photobleaching (FRAP) and pharmacological approach, we demonstrate that carbachol (Cch) increases firing frequency, intracellular Ca2+ and that TRPM4 inhibition using 9-Ph and CBA reduces firing frequency and decreases the peak in intracellular Ca2+ induced by Cch in cortical pyramidal neurons in culture. Moreover, we determined that cholinergic stimulation reduces TRPM4 recycling and stabilizes TRPM4 in the plasma membrane. Together our results indicate that cholinergic stimulation increases firing in a TRPM4 dependent manner, and also increases the TRPM4 stability in the membrane, suggesting that TRPM4 is locked in microdomains in the membrane, possibly signaling or cytoskeleton proteins complexes.

The pequi (Caryocar brasiliense) fruit peel, despite being frequently discarded, has a high content of bioactive compounds, and therefore has a high nutritional value. The present study aimed to explore the bioactivities in the pequi peel, particularly their potential health benefits at the level of antioxidant activity. The exploitation of this fruit could also present significant economic benefits and applications of pequi by-products would represent a reduction in waste, having a positive impact on the environment. Phenolic compounds present in the pequi exocarp and external mesocarp were identified by paper spray mass spectrometry (PS-MS) and quantified by HPLC. The total phenolic content (TPC) along with the amount of 2,2-diphenyl-1-picrylhydrazyl (DPPH), Ferric Reducing Antioxidant Power (FRAP), and the amount of 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) were also determined in peel extracts. Epicatechin was the most abundant phenolic compound found, followed by the caffeic, salicylic, and gallic acids. In addition, fingerprinting revealed compounds related to several beneficial health effects. In short, the results obtained were encouraging for potential applications of pequi peel in the field of functional foods.

One of the most used methods to measure antioxidant capacity in food is the ferric reducing antioxidant power (FRAP) test, which is simple, sensitive, and economical, nevertheless has long analysis times, causing measurement errors due to the instability of the FRAP reagent due to its precipitation sequential injection analysis (SIA) is a flow technique that can correct these disadvantages because it is more quickly. So, a novel FRAP-SIA method was developed to evaluate the antioxidant capacity. The system was optimized using a central composite design for hydrodynamic and chemical factors, resulting in a flow rate of 35 µL s-1, and aspirate volumes of 33 µL-38 µL-33 µL for the sequence (FRAP-Antioxidant-FRAP). FRAP reagent was prepared with an HCl solution at 0.005 mol L-1, improving its stability 24 times, concerning when it is in acetate buffer at pH 3.6. The method showed excellent accuracy (RSD <3%) with a LOD of 1.0 µmol L-1 of Trolox for a linear range of 5-120 µmol L-1. The reaction time was diminished by 96% concerning the FRAP-microplate assay (from 30 min to 1.2 min). The method was applied in beverages and extracts, obtaining recovery values ranging from 91.24 to 114.22%.

Fluorescence recovery after photobleaching (FRAP) is a laser method of light microscopy to evaluate the rapid movement of fluorescent molecules. To have a more reliable approach to analyze data from FRAP, we designed Fraping, a free access R library to data analysis obtained from FRAP. Unlike other programs, Fraping has a new form of analyzing curves of FRAP using statistical analysis based on the average curve difference. To evaluate our library, we analyzed the differences of actin polymerization in real time between dendrites and secondary neurites of cultured neuron transfected with LifeAct to track F-actin changes of neurites. We found that Fraping provided greater sensitivity than the conventional model using mobile fraction analysis. Likewise, this approach allowed us to normalize the fluorescence to the size area of interest and adjust data curves choosing the best parametric model. In addition, this library was supplemented with data simulation to have a more significant enrichment for the analysis behavior. We concluded that Fraping is a method that reduces bias when analyzing two data groups as compared with the conventional methods. This method also allows the users to choose a more suitable analysis approach according to their requirements. RESEARCH HIGHLIGHTS: Fraping is a new programming tool to analyze FRAP data to normalize fluorescence recovery curves. The conventional method uses one-point analysis, and the new one compares all the points to define the similarity of the fluorescence recovery.

Handroanthus impetiginosus, popularly known as "ipê-roxo", is used in folk medicine to treat skin inflammations, infections, stomach diseases, and cancer. Fatty acid methyl esters (FAMEs) obtained from the esterification reaction of fatty acids (FA) found in the hexane extract (HE) of seeds of H. impetiginosus were identified by gas chromatography-mass spectrometry (GC-MS). The antioxidant and cytotoxic activities of the HE and FAMEs were evaluated. Methyl palmitate, methyl linoleate, methyl oleate, and methyl stearate were the major FAMEs obtained from the HE. The samples, especially the HE, exhibited a significant antioxidant potential analyzed by ferric reducing ability power (FRAP) assay. In the A. salina larvae bioassay, the HE showed no cytotoxic effects, but the FAMEs exhibited a high toxicity. This study reported, for the first time, the antioxidant and cytotoxic activities of the HE and FAMEs obtained from H. impetiginosus seeds.

RESUMO

Selenium is an essential trace element in living organisms, and is present in selenoenzymes with antioxidant activity, like glutathione peroxidase (GPx) and thioredoxin reductase (TrxR). The search for small selenium-containing molecules that mimic selenoenzymes is a strong field of research in organic and medicinal chemistry. In this review, we review the synthesis and bioassays of new and known organoselenium compounds with antioxidant activity, covering the last five years. A detailed description of the synthetic procedures and the performed in vitro and in vivo bioassays is presented, highlighting the most active compounds in each series.

Knowledge is limited about the level of bioactive compounds and antioxidant activity of seeds from bred lines of common beans developed from interspecific crosses using four different Phaseolus species (P. vulgaris L., P. coccineus L., P. acutifolius A. Gray. Gray., and P. dumosus). In this study, differences in the nutritional quality of seeds among 112 bean genotypes were evaluated by measuring the levels of phenolic compounds, pigments, antioxidant activity, and sugars. The bean genotypes were grown under high temperatures and acid soil conditions in the Amazon region of Colombia. Five typology groups of bean genotypes were identified based on the level of bioactive compounds and their functional capacity: (1) highly bioactive and functional (HBF); (2) moderately bioactive and functional (MBF); (3) moderate antioxidant content with pigment influence (MACP); (4) moderately antinutritional with limited antioxidant potential (MALAP); and (5) antinutritional, low bioactive, and functional (ALBF). We developed a nutritional quality index (NQI) with values ranging from 0 to 1 based on the nutritional and anti-nutritional balance of each genotype and the higher values of the NQI of a genotype indicating greater nutritional quality. We found three interspecific bred lines (SER 212, SER 213, and RRA 81), with NQI values higher than 0.8. These three lines belong to the typology group of HBF. The superior nutritional quality of these three interspecific bred lines is attributed to a greater level of bioactive compounds and antioxidant capacity. These three bred lines may serve as useful parents to develop nutritionally superior and stress-resilient beans from bean breeding programs. Further research is needed to explore the role of testa color in improving the nutritional quality of seeds of common bean genotypes grown under different climatic conditions.

Grape pomaces have a wide and diverse antioxidant phenolics composition. Six Uruguayan red grape pomaces were evaluated in their phenolics composition, antioxidant capacity, and anti-inflammatory properties. Not only radical scavenging methods as DPPH· and ABTS·+ were employed but also ORAC and FRAP analyses were applied to assess the antioxidant potency of the extracts. The antioxidant reactivity of all extracts against hydroxyl radicals was assessed with ESR. The phenol profile of the most bioactive extract was analyzed by HPLC-MS, and a set of 57 structures were determined. To investigate the potential anti-inflammatory activity of the extracts, Nuclear Factor kappa-B (NF-κB) modulation was evaluated in the human colon cancer reporter cell line (HT-29-NF-κB-hrGFP). Our results suggest that Tannat grapes pomaces have higher phenolic content and antioxidant capacity compared to Cabernet Franc. These extracts inhibited TNF-alpha mediated NF-κB activation and IL-8 production when added to reporter cells. A molecular docking study was carried out to rationalize the experimental results allowing us to propose the proactive interaction between the NF-κB, the grape extracts phenols, and their putative anti-inflammatory bioactivity. The present findings show that red grape pomace constitutes a sustainable source of phenolic compounds, which may be valuable for pharmaceutical, cosmetic, and food industry applications.

Smilax brasiliensis Sprengel is a monocotyledon of the Smilacaceae family, native to the Brazilian Cerrado, popularly known as "salsaparrilha" or "japecanga". In this study, the ethanol extract (EE) and the hexane (HEXF), dichloromethane (DCMF), ethyl acetate (ACF), and hydroethanol (HEF) fractions of the stems were obtained. The chemical composition was determined, the contents of phenolic compounds and flavonoids were quantified, and the antioxidant potential and the cytotoxic effect on Artemia salina were evaluated. Fatty acid esters, hydrocarbons, and phytosterols were identified in the HEXF analyzed by gas chromatography - mass spectrometry (GC-MS). The EE and DCMF, ACF, and HEF were analyzed by liquid chromatography coupled to a diode array detector and mass spectrometer (LC-DAD-MS), and the identified constituents included glycosylated (rutin, 3-O-ß-galactopyranosyl quercetin, 3-O-ß-glucopyranosyl quercetin, O-deoxyhexosyl-hexosyl quercetin, O-deoxyhexosyl-hexosyl kaempferol, O-deoxyhexosyl-hexosyl O-methyl quercetin, and others), and non-glycosylated (quercetin) flavonoids, phenylpropanoids (3-O-E-caffeoyl quinic acid, 5-O-E-caffeoyl quinic acid, O-caffeoyl shikimic acid, and others), neolignan, steroidal saponin (dioscin), and N-feruloyltyramine. The EE, DCMF, and ACF showed high total contents of phenolic compounds (112.99, 175.71, and 524.02 µg of GAE/mg, respectively), and in the ACF and DCMF a great content of flavonoids was also quantified (50.08 and 31.49 µg of QE/mg, respectively). The EE, DCMF, ACF, and HEF exhibited great antioxidant potential by DPPH (IC50 1.71 - 32.83 µg/mL) and FRAP (IC50 0.63 - 6,71 µg/mL) assays. A maximum cytotoxic activity on A. salina of 60% was observed for the DCMF (LC50 = 856.17 µg/mL). This study contributes to the phytochemical study of S. brasiliensis since these compounds were identified for the first time in the stems of this species. The S. brasiliensis stems demonstrated to be a rich source of polyphenols compounds and exhibited high antioxidant potential without toxicity. Thus, extract and fractions obtained from the S. brasiliensis stems can be used in food supplements or as natural antioxidants in the food industry.

The increased production of mechanically separated poultry meat has led to its use in the production of foods of higher commercial value, such as hamburgers. However, hamburgers are more susceptible to oxidation, therefore, the goal of this study was to evaluate the antioxidant and antibacterial properties of hamburgers made with mechanically separated meat (MSM) from broilers and broiler breeder hens supplemented with oregano (Origanum vulgare) or peppermint (Mentha piperita). Hamburgers with a synthetic antioxidant and three levels of oregano and peppermint essential oils were evaluated using a sensory acceptance test. An inclusion level of 0.04% for both essentials oils was chosen for this test to evaluate the physical-chemical compositions as well as the microbiological and antioxidant activities because an inclusion level of 0.08% received a lower score from the evaluators. Higher pH values were found in hamburgers made with broiler MSM (control (6.94) and peppermint essential oil (6.93) and in broiler breeder hens MSM supplemented with peppermint essential oil (6.89). When comparing the origin of MSM, lower pH values were found in the broiler hamburgers supplemented with oregano essential oils (6.83). The total coliform counts at day zero were significantly higher for the hamburgers made with broiler breeder hens MSM with oregano essential oil, however, broilers MSM supplemented with a synthetic antioxidant showed the lowest count when compared to that of the others. For Staphylococcus coagulase-negative bacteria, growth was observed only on day zero, with a low count in hamburgers made with broiler breeder hens MSM and broiler MSM supplemented with synthetic antioxidant or peppermint essential oil. Hamburgers made with MSM from broilers and broiler breeder hens supplemented with synthetic antioxidants showed higher antioxidant capacity with an average of 2378.75 and 2265.43 µmols equivalent Trolox/mg, respectively. Our results suggest that the essential oils of oregano or peppermint can be used in the formulation of hamburgers since they presented high sensory acceptability and reduced contamination.(AU)


O aumento da produção de carne mecanicamente separada (CMS) de aves levou a indústria a buscar seu uso na produção de produtos de maior valor comercial, como os hambúrgueres. No entanto, os hambúrgueres são mais susceptíveis a oxidação, em vista disso, o objetivo desse trabalho foi avaliar hambúrgueres elaborados com CMS de frangos e galinhas pesadas e adicionados de óleos essenciais de orégano (Origanum vulgare) ou hortelã-pimenta (Mentha piperita) como antioxidantes e antibacterianos. Para o teste de aceitação sensorial, foi avaliado um antioxidante sintético e mais três níveis de óleos essenciais de orégano e hortelã-pimenta. A partir desse teste, foi escolhido o nível de inclusão de 0,04% de ambos os óleos essenciais para realizar as análises físico-químicas, microbiológica e capacidade antioxidante, uma vez que o nível de 0,08% recebeu a menor nota pelos avaliadores. Em relação ao pH, maior valor foi encontrado nos hambúrgueres elaborados com CMS de frango (controle 6,94) e óleo essencial de hortelã-pimenta (6,93) e CMS de galinhas pesadas e adicionados de óleo essencial de hortelã-pimenta (6,89). Ao comparar individualmente a origem da CMS, menor valor de pH foi encontrado para o hambúrguer de frango adicionado de óleo essencial de orégano (6,83). As contagens de coliformes totais, no dia zero, foi significativamente maior para os hambúrgueres elaborados com CMS de galinhas pesadas adicionados de óleo essencial de orégano, porém, a CMS de frangos acrescida de antioxidante sintético apresentou a menor contagem quando comparado aos demais. Já para Staphylococcus coagulase negativa, houve crescimento apenas no dia zero, com baixas contagens nos hambúrgueres elaborados com CMS de galinhas pesadas e de frangos adicionados de antioxidante sintético ou óleo essencial de hortelã-pimenta. Hambúrgueres elaborados com CMS de frangos e galinhas pesadas adicionados de antioxidante sintético apresentaram maior capacidade antioxidante, uma vez que apresentaram média de 2378,75 e 2265,43 µmols equivalente Trolox/mg respectivamente para frangos e galinhas pesadas. Conclui-se que os óleos essenciais de orégano ou hortelã-pimenta podem ser utilizados na formulação dos hambúrgueres, uma vez que apresentam boa aceitabilidade e reduzem as contaminações.(AU)

This study aimed to investigate the effects of delivery type (normal or caesarean) on the antioxidant and oxidative capacity of colostrum collected shortly after delivery. A total of 61 parturients were included in the study and divided into two groups: those who underwent vaginal delivery (n = 36) and those who underwent elective caesarean section (n = 25). Colostrum samples were collected by manual milking up to 48 h post parturition and analysed for thiol groups (-SH), vitamin C, ferric reducing ability (FRAP), nitrate/nitrite oxides (NOx), and advanced oxidation protein products (AOPP). Colostrum levels of -SH (p = 0.0042), vitamin C (p = 0.0455), and FRAP (p = 0.0374) were significantly lower in the vaginal delivery group. The results suggest that vaginal delivery, compared to caesarean section, is associated with lower levels of antioxidants in colostrum and the mode of delivery plays an important role in the composition of antioxidants in maternal colostrum that help protect newborns from oxidative damage.IMPACT STATEMENTWhat is already known on this subject? Colostrum is the first biological fluid produced by the mother after delivery and is responsible for a child's growth, cognitive development and health. It is known that childbirth can cause oxidative imbalance, and its effects have already been evaluated in maternal and foetal blood, however, there are few studies evaluating the effects of childbirth on colostrum composition.What do the results of this study add? Previously, a study showed that caesarean section caused greater oxidation of colostrum compared to vaginal delivery. Thus, we sought to evaluate other markers (thiol groups, vitamin C, ferric reducing ability, nitrate/nitrite oxides, and advanced oxidation protein products), in a short period of time after delivery, in order to elucidate this still little discussed issue. Unlike the previous one, our study suggests that vaginal delivery, compared to caesarean section, is associated with lower levels of antioxidants in colostrum, which may make it difficult to protect newborns from oxidative damage.What are the implications of these findings for clinical practice and/or further research? Our study suggests that normal delivery can influence the antioxidant composition of maternal colostrum, and it is debateable for future clinical practice to improve eating habits during pregnancy and lactation, in order to strengthen the antioxidant capacity of colostrum and reduce oxidative damage to newborns.

The overexpression of HER2 in breast cancer (BC) can contribute to redox imbalance, which is related to damage and structural modification in many biomolecules. To the best of our knowledge, this is the first study that has investigated the infrared spectrum wavenumbers obtained by ATR-FTIR and their relationship with the levels of redox status markers such as reduced glutathione, superoxide dismutase (SOD), catalase, Ferric Reducing Antioxidant Power (FRAP), and protein carbonyl among women with HER2+ BC, HER2- BC, and benign breast disease (BBD). The study was conducted with 25 women, 17 of whom were diagnosed with BC (6 HER2+ and 11 HER2-) and 8 with BBD. Our results indicate HER2+ BC cases could be distinguished from HER2- BC and BBD cases by their serum's antioxidant capacity [HER2+ BC vs. HER2- BC (AUC = 0.818; specificity = 81.82%; sensitivity = 66.67%); HER2+ BC vs. BBD (AUC = 0.875; specificity = 75%; sensitivity = 83.33%)]. The changes in biochemical terms that occur in serum as a result of the scarcity of antioxidants are related to a peculiar fingerprint in the infrared spectrum obtained by ATR-FTIR. In the serum of women with BBD, the SOD enzyme level is the highest, and this characteristic allowed us to distinguish them from HER2- BC. Finally, data regarding the serological antioxidant capacity of FRAP and the infrared spectrum by ATR-FTIR will allow us to assess biochemical changes that occur before clinical signs, indicating whether changes in therapy or interventions are necessary.

SUMMARY Aims: This study aimed to characterize and compare the antioxidative potential of extracts obtained by infusion, decoction, tincture, aqueous extraction, and hydro-ethanolic maceration from the aerial parts of Polygonum acre H.B.K., which has been traditionally used in herbal preparations, for different purposes. The therapeutic benefits are attributed to phenolic compounds and their antioxidant properties. Methods: All extracts were characterized considering their quantitative content of the total phenolics, flavonoids, condensed and, hydrolysable tannins, by colori-metric methods. EHW-PA was selected for HPLC analysis as it showed a higher yield (10.58 % w/m) and a phenolic content > 200 mg GAE/g dry extract. The phenolic profile showed a chromatogram with 20 peaks, and the presence of gallic acid, rutin, and quercetin was verified by comparison with the retention times of standard compounds. The antioxidant activities were determined by ABTS capture test, ferric reducing antioxidant power test (FRAP), and the superoxide anion scavenging test. Results: Tinctures showed a higher average content of phenolic compounds, present mainly as flavonoid content. A significant correlation coefficient was observed between the total phenolic content and its antioxidant activity, determining by ABTS and FRAP assays. Differently, a low to moderate correlation between the flavonoid content and antioxidant activity was verified. Conclusion: This study reinforces the ethnopharmacological relevance of the Polygonum genus and could contribute to the scientific basis for the use of P. acre preparations.


Objetivos: Este estudio tuvo como objetivo caracterizar y comparar el potencial antioxidante de extractos obtenidos por infusión, decocción, tintura, extracción acuosa e hidroetanólica de las partes aéreas de Polygonum acre H.B.K., que se ha utilizado tradicionalmente en preparaciones a base de hierbas, para diferentes fines. Los beneficios terapéuticos se atribuyen a los compuestos fenólicos y sus propiedades antioxidantes. Métodos: Todos los extractos se caracterizaron considerando su contenido cuantitativo de fenólicos totales, flavonoides, taninos condensados e hidrolizables, por métodos colorimétricos. Se seleccionó EHW-PA para el análisis de HPLC ya que mostró un rendimiento más alto (10,58% m/m) y un contenido fenólico > 200 mg GAE/g de extracto seco. El perfil fenólico mostró un cromatograma con 20 picos y se verificó la presencia de ácido gálico, rutina y quercetina por comparación con los tiempos de retención de los compuestos estándar. Las actividades antioxidantes se determinaron mediante la prueba de captura ABTS, la prueba del poder antioxidante reductor férrico (FRAP) y la prueba de eliminación del anión superóxido. Resultados: Las tinturas mostraron un mayor contenido promedio de compuestos fenólicos, presentes principalmente como contenido de flavonoides. Se observó un coeficiente de correlación significativo entre el contenido fenólico total y su actividad antioxidante, determinado por ensayos ABTS y FRAP. De manera diferente, se verificó una correlación de baja a moderada entre el contenido de flavonoides y la actividad antioxidante. Conclusión: Este estudio refuerza la relevancia etnofarma-cológica del género Polygonum y podría contribuir a la base científica para el uso de preparaciones de P. acre.


Objetivos: O presente estudo teve como objetivo caracterizar e comparar o potencial antioxidante de extratos obtidos por infusão, decocção, tintura, extração aquosa e maceração hidroetanólica da parte aérea de Polygonum acre H.B.K., tradicional- mente utilizado em preparações fitoterápicas, para diversos fins. Os benefícios terapêuticos são atribuídos aos compostos fenólicos e suas propriedades antioxidantes. Métodos: Todos os extratos foram caracterizados quanto ao teor quantitativo de fenólicos totais, flavonoides, taninos condensados e hidrolisáveis, por métodos colorimétricos. O EHW-PA foi selecionado para análise por HPLC por apresentar maior rendimento (10,58% m/v) e conteúdo fenólico > 200 mg GAE/g de extrato seco. O perfil fenólico apresentou cromatograma com 20 picos, e a presença de ácido gálico, rutina e quercetina foi verificada pela comparação com os tempos de retenção dos compostos padrão. As atividades antioxidantes foram determinadas pelo teste de captura do radical ABTS, ensaio do potencial antioxidante por redução férrica (FRAP) e teste de desativação do ânion superóxido. Resultados: As tinturas apresentaram maior teor médio de compostos fenólicos, presentes, principalmente, na forma de flavonoides. Foi observado um coeficiente de correlação significativo entre o conteúdo fenólico total e sua atividade antioxidante, determinado pelos ensaios de ABTS e FRAP. Diferentemente, verificou-se uma correlação baixa a moderada entre o conteúdo de flavonoides e a atividade antioxidante. Conclusão: Este estudo reforça a relevância etnofarmacológica do gênero Polygonum e pode contribuir para a fundamentação científica do uso de preparações de P. acre.

The current trend of lowering 5-hydroxymethylfurfural (5-HMF) dietary exposure is challenging since its formation is parallel with the development of food color, flavor and aroma. We aimed to investigate the effect of gallic acid (GA) addition on 5-HMF formation, color development and antioxidant activity (AA) in a series of Maillard Reaction (MR) model systems. The effects of GA addition on browning and AA development were not uniform for all model systems, but always occurred in the same direction, indicating that these phenomena were interconnected. GA mitigated 5-HMF development in four of the nine tested systems, possibly by preventing the oxidation of MR intermediates. Correlation analysis indicated that when GA addition mitigated 5-HMF formation, browning was either promoted or not affected. The proposed strategy was effective for glucose/arginine and sucrose/arginine systems, since GA mitigated 5-HMF formation (49% and 54%, respectively) in addition to increasing color development and antioxidant activity.

Gum arabic, modified corn starch (EMCAP), modified malt (MALT), either blended or isolated, were assessed as encapsulating agents for Capsicum oleoresin. Capsicum oleoresin microparticles were obtained by spray drying and analysed for physicochemical properties and in vivo. Obtained powders were adequate for storage, given their low water activity (<0.150), hygroscopicity (<11.43 g/100 g), moisture (<4.76%) and high glass transition temperature (<98.3 °C). FT-IR analysis concluded that carbohydrates matrices were loaded after spray drying, with peaks around 2850 cm -1 for aromatic compounds, and bands around 1760 cm-1, pointing to the presence of capsaicin inside the microparticles. All formulations exhibited high antioxidant activity, low contact angles and great solubility in water. Any adverse effect was observed in the experimental assay, neither change on the level of hepatic aminotransferases. The intake of a High-Fat Diet (HFD) supplemented with Capsicum oleoresin microparticles decreased weight gain when compared to the HFD control.

The aim of this study was to evaluate the follicular development, morphological integrity, and oxidative stress of preantral ovarian follicles from Bos taurus indicus females grown in vitro with ascorbic acid. Ovaries (n = 20) from Bos taurus indicus females were collected, fragmented, and were cultured in vitro for 6 or 12 days in minimum essential medium (MEM), or MEM supplemented with 50 or 100 ng/ml ascorbic acid, with an extracellular matrix of agarose gel, in an incubator at 38.5°C; every 2 days, 100% of the culture medium was replaced. The data were analyzed using the chi-squared test and/or Fisher's exact test. In the event of a significant effect, the proportions were compared using a 2 × 2 proportion test. The oxidative stress analysis data were submitted to analysis of variance followed by the Bonferroni test. Values were considered significant when P ≤ 0.05. The addition of 100 ng/ml of ascorbic acid to the in vitro culture medium of preantral ovarian follicles from bovine females promoted follicular development, was efficient in maintaining morphological integrity, as well as the stability of reactive oxygen species, after 6 days of in vitro culture.

The use of guayusa (Ilex guayusa Loes.) leaves as functional food has increase recently. This work discusses the antioxidant activity and volatile compounds of guayusa leaves extract and fractions. The methanol crude extract was obtained by maceration, subsequently hexane, chloroform, ethyl acetate, and aqueous fractions were collected by solvent-solvent partition. Total phenolic content (TPC), total flavonol/flavone content (TFC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and ferric reducing antioxidant power (FRAP) were measured by ultraviolet-visible (UV-Vis) spectrophotometry. The results revealed that ethyl acetate fraction showed highest inhibition against DPPH radical (93.86 ± 0.95%) at 500 µg/mL, and reduce the ferric-tripyridyltriazine complex (Fe3+-TPTZ) at 1619.81 mg trolox equivalent (TE)/g, followed by aqueous fraction. This bioactivity could be related to phenolic acids, flavones and flavonols content, as well as the caffeine, dodecanoic acid isopropyl ester, caffeic acid, and malic acid identified by gas chromatography-mass spectrometry (GC-MS). These findings support the antioxidant properties of this plant material.

Abstract The aim of this paper is to study the chemical composition of alkaloids present in Haloxylon scoparium Pomel extracts and to evaluate their antioxidant capacity. The alkaloids were isolated from two parts of Haloxylon scoparium plant by two extraction protocols. and The quantitative study made it possible to propose the best protocol for the extraction of the alkaloids. Moreover, GC-MS analysis of alkaloid extracts allowed us to determine their chemical composition. Haloxylon scoparium contains four types of alkaloids: tetraisoquinolines, phenylethylamines, tryptolines and tryptamines. The main compounds are the tetraisoquinolines type, the predominant product of which was N-methylsalsoline. These compounds present a great interest for the researchers due to their various pharmacological and biological activities. The antioxidant effect of the different plant extracts was studied by two methods: the ferric reducing antioxidant power (FRAP) and 1,1-diphenyl-2-picryl hydrazyl free radical (DPPH·) scavenging tests. The results show that extracts of root part are more active than those from aerial part; the acetone/water extract is the most powerful. The interesting results obtained in this study will be supplemented by other analyses and biological tests in order to better valorize this plant.

The objective of the work was to determine the antioxidant potential in vitro of freeze-dried peel extracts of 20 fruits from the northern region of Peru through five tests (Folin-Ciocalteu, DPPH., ABTS+., FRAP and CUPRAC). According to multivariate statistical analyzes, five groups were found: (i.) peel extracts with the highest values of antioxidant capacity (AC) from custard apple, and star fruit; (ii.) rind extracts with high AC values from quince, sweet granadilla, guava, and black grape; (iii.) husk extracts with middle values of AC from passion fruit, and red mombin; (iv.) shell extracts with low AC values from tangerine, mandarine, and bitter orange; and, (v.) coating extracts with the lowest AC values from pawpaw, red pawpaw, muskmelon, dragon fruit, yellow and red indian figs, pear, apple, and green grape. To conclude, the fruit lyophilized-husk extracts of custard apple, star fruit, quince, sweet granadilla, guava, and black grape obtained the best AC.


El objetivo del trabajo fue determinar el potencial antioxidante in vitro de extractos de cáscara liofilizada de 20 frutos de la región norte del Perú mediante cinco pruebas (Folin-Ciocalteu, DPPH., ABTS+., FRAP y CUPRAC). Según análisis estadísticos multivariados, se encontraron cinco grupos: (i.) Extractos de piel con los valores más altos de capacidad antioxidante (CA) de chirimoya y carambola; (ii.) extractos de cáscara con altos valores de CA de membrillo, granadilla dulce, guayaba y uva negra; (iii.) extractos de cáscara con valores medios de CA de maracuyá y mombina roja; (iv.) extractos de cáscara con valores bajos de CA de mandarina, mandarina y naranja amarga; y (v.) recubrir extractos con los valores de CA más bajos de papaya, papaya roja, melón, fruta del dragón, higos indios amarillos y rojos, pera, manzana y uva verde. Para concluir, los extractos de cáscara liofilizada de chirimoya, carambola, membrillo, granadilla dulce, guayaba y uva negra obtuvieron el mejor CA.

Navicula incerta is a marine microalga distributed in Baja California, México, commonly used in aquaculture nutrition, and has been extended to human food, biomedical, and pharmaceutical industries due to its high biological activity. Therefore, the study aimed to optimize culture conditions to produce antioxidant pigments. A central composite experimental design and response surface methodology (RSM) was employed to analyze the best culture conditions. The medium (nitrogen-deficient concentrations), salinity (PSU = Practical Salinity Unity [g/kg]), age of culture (days), and solvent extraction (ethanol, methanol, and acetone) were the factors used for the experiment. Chlorophyll a (Chl a) and total carotenoids (T-Car), determined spectroscopically, were used as the response variables. The antioxidant capacity was evaluated by DPPH• and ABTS•+ radical inhibition, FRAP, and anti-hemolytic activity. According to the overlay plots, the optimum growth conditions for Chl a and T-Car production were the following conditions: medium = 0.44 mol·L-1 of NaNO3, salinity = 40 PSU, age of culture: 3.5 days, and solvent = methanol. The pigment extracts obtained in these optimized conditions had high antioxidant activity in ABTS•+ (86.2-92.1% of inhibition) and anti-hemolytic activity (81.8-96.7% of hemolysis inhibition). Low inhibition (33-35%) was observed in DPPH•. The highest value of FRAP (766.03 ± 16.62 µmol TE/g) was observed in the acetonic extract. The results demonstrated that RSM could obtain an extract with high antioxidant capacity with potential applications in the biomedical and pharmaceutical industry, which encourages the use of natural resources for chemoprevention of chronic-degenerative pathologies.
