A greater understanding of factors influencing fertility is essential to improve pregnancy rates and reduce the occurrence of embryonic mortality in beef herds. The objective of the current study was to evaluate retrospective data of pregnancy per artificial insemination (P/AI) and pregnancy loss in Nelore females subjected to timed-AI (TAI) in Brazil. Data from 40,104 TAI collected from six breeding seasons (2016-2022) were analyzed, and the effects of animal category (e.g., classification based on age and parity), farm, month of parturition, sire, sire breed (Nelore vs Angus), estrus expression at TAI, animal temperament, and body condition scores (BCS) were evaluated. P/AI and pregnancy loss were affected (P < 0.001) by animal category. There was also an effect of farm (P = 0.0013) on P/AI and pregnancy loss (P = 0.001), as P/AI ranged from 49.28% and 55.58% and pregnancy loss from 3.37% to 6.89% across the herds evaluated. Month of parturition also affected (P < 0.001) P/AI and was higher for cows that became pregnant at the beginning of the previous breeding season. Calmer animals, presenting lower velocity scores while exiting the chute following TAI, achieved higher P/AI (P < 0.001). Lower BCS at TAI was associated (P < 0.001) with increased pregnancy loss, and BCS gain following AI was associated (P < 0.001) with reduced rates of embryonic mortality. There was a major effect (P < 0.001) of sire on P/AI and pregnancy loss, as P/AI ranged from 11% to 79%, and embryonic mortality from 0% to 40% for the bulls used in the study, highlighting the importance of the sire fertility on overall pregnancy success. Results from the current study reinforce the idea that animal age and parity at the beginning of the breeding season, BCS at the onset of estrous synchronization, BCS gain following AI, estrus expression at TAI, sire, and month of parturition are important factors influencing P/AI and rates of embryonic mortality in beef herds.

The objectives of this study were: 1- to evaluate the association of Bovine Viral Diarrhea Virus (BVDV), Bovine Herpes Virus 1 (BoHV-1), and Neospora caninum (N. caninum) with the risk for Late Embryonic Loss (LEL) in grazing dairy cows, 2- to evaluate blood progesterone concentration at the time of LEL occurrence, and 3- to describe a novel ultrasound-guided technique for conceptus sampling. We run a prospective cohort study involving 92 cows (46 LEL and 46 NLEL). An LEL cow was that having an embryo with no heartbeat, detached membranes, or floating structures, including embryo remnants detected at pregnancy check by ultrasonography (US) 28-42 days post-AI, whereas an NLEL cow was that with embryo heartbeats detectable by US at pregnancy check 28-42 d post-IA. We took two blood samples from every cow at pregnancy check by US (the day of LEL detection) and 28 d later to perform serological diagnosis of BVDV, BoHV-1, and N. caninum; and to measure blood progesterone concentration at pregnancy check (28-42 d post-AI). We also sampled the conceptus from all the LEL cows. We performed PCR to detect BVDV, BoHV-1, and N. caninum in sampled conceptuses from LEL cows. Finally, we evaluated the associations of risk factors (serological titers, seroconversion, and progesterone) with LEL odds with logistic models. The risk for LEL was associated with serological titers to BVDV (P = 0.03) and tended to be associated with seroconversion to BVDV, given that 19.6% (9/46) in LEL and 6.5% (3/46) in NLEL cows seroconverted to BVDV (P = 0.09). In addition, BVDV was detected in conceptuses from LEL cows that seroconverted to BVDV but not in LEL cows that did not seroconvert. Conversely, the risk for LEL was not associated with the titers or seroconversion to BoHV-1 and N. caninum. BoHV-1 and N. caninum were not identified in any of the conceptuses. Finally, blood progesterone concentration was similar in LEL and NLEL cows, and it was not associated with the risk for LEL (P = 0.54). In conclusion, BVDV infection is a risk factor for LEL in dairy cows.

Background: Hormonal and physiological changes during pregnancy period have significant effects on animals' metabolisms. The purpose of the present study is to evaluate the variations of aspartate aminotransferase (AST) and gamma-glutamyl transferase (GGT) levels according to different gestation trimesters of the pregnant mares to assess changes of embryonic losses and abortions. Materials, Methods & Results: Blood samples from 49 thoroughbred mares were analyzed during this study. Age ratio of the mares is 5-18 years and their body condition score (BCS; score 1 to 9) varies between 5 and 6. A total of 28 mares had given birth to healthy foals at the end of a healthy gestation period (Group C), whereas 11 mares had embryonic losses (Group E) and 10 mares had late period abortions (Group A). Following the confirmation of pregnancy, one blood sample per gestation trimester were taken (14-16 days of pregnancy for 1st trimester; 180 days for 2nd trimester, 270 days for 3rd trimester). Early embryonic losses (loss of a 16-25 days embryo) were observed in 6 of 11 mares in Group E and the 5 of 11 mares had late period embryonic losses (loss of a 35-40 days embryo). In Group A, 6 mares had the abortion within 7th month and the remaining 4 mares had the abortion within the 8th month of pregnancy. Repeated Measures ANOVA, Kruskal-Wallis test and Mann-Whitney test were performed for statistical analysis, the mean AST level in Group C (n = 28) was higher during the 1st trimester in comparison to 2 following trimesters (respectively: P = 0.011 ; P = 0.01). Besides, no statistical difference was observed between 2nd and 3rd trimester regarding AST activation (P > 0.05). The mean GGT level in Group C was significantly decreased lower in the 3rd trimester compared to 1st and 2nd trimester (respectively: P = 0.007 ; P = 0.009). No statistical difference was observed between 2nd and 3rd trimester regarding GGT activation (P > 0.05). Among all groups (C, E, A) no significant difference was observed on AST levels (P > 0.05), nonetheless GGT levels had a significant rise (P = 0.039) in Group A in comparison to the 1st trimester levels of Group C. In Group A, there was a statistical decrease of AST during the 2nd trimester in comparison to the 1st trimester (P = 0.001), accompanied by a decrease of GGT activation during the 2nd trimester compared to the 1st trimester (P = 0.009). Discussion: The aminotransferases are catalisors that play an important role on the metabolism of amino acids and carbohydrates. Although serum AST levels were within the reference ranges during 3 trimesters in this study, serum AST levels were determined to be decreased in the 2nd and 3rd trimesters in healthy pregnant mares compared to the 1st trimester. It is thought that these results can be obtained due to the increase in metabolic needs during pregnancy. In this study, serum GGT levels remained within physiological limits with a tendency to decrease as the pregnancy advances, in accordance with previous study results. Serum GGT levels that are used as an indicator to liver damage may vary during the pregnancy as a result of increased metabolic load. It is thought that the increase of serum GGT levels in Group A might be related to the fetal (chromosome-related) issues that may result to pregnancy losses. As a result, it is considered that serum AST and GGT levels in mares might be valuable parameters that predict embryonic loss or abortus cases.

This study aims to evaluate the association between reproductive disorders and the main serogroups of Leptospira spp. in dairy herds. Blood samples from 216 cows from nine herds were collected for a Microscopic Agglutination Test (MAT), and a reproductive follow-up with ultrasonography was carried out monthly for at least 12 months. A total of 75 (16.5%) cows were reactive, while seroreactivity to a herd level ranged from 16.0% to 52.8%. Out of the nine herds, in five (55.0%) Sejroe was found to be the predominant serogroup and in four (45.0%) it was Icterohaemorrhagiae. The major reproductive problems identified were embryonic death (ED) and abortions. All herds presented ED and abortion was observed in three (33.3%). Herds with high seroreactivity (>20%) had more reproductive problems. Those with a predominance of Sejroe were associated with ED (P < 0.05), while those with Icterohaemorrhagiae were associated with abortion. The results show that different leptospiral strains lead to different reproductive problems, as well as showing that the main problem - ED - is caused by leptospirosis, particularly for those determined by strains of the serogroup Sejroe.

Following the induction of oestrus out of season in small ruminants, low fertility and variations in fertility rates are associated with embryonic losses. One of the main causes of embryonic loss is luteal dysfunction. Gonadotropin Releasing Hormone (GnRH) supports the luteal structure, and increasing progesterone levels may be beneficial in terms of promoting embryonic life. The main objective of the present study was to evaluate the efficacy of GnRH administration following an oestrus induction protocol in the anoestrus season for preventing embryonic loss in goats having failure to conceive during the season. In the study, 106 Damascus goats aged 3-5 years and weighing 45-60 kg were used. The oestrus of 106 goats in the anoestrous group was stimulated with progesterone and pregnant mare serum gonadotropin (PMSG) treatment. Out of breeding season, goats were divided into the 4 following groups: GnRH0 (n = 27), GnRH7 (n = 26), GnRH0+7 (n = 27) and control (n = 26). In each goat, an intravaginal sponge (IS) containing 20 mg of fluorogestone acetate (FGA) was placed into the vagina and left for 9 days. With the withdrawal of the sponge, 550IU PMSG and 125 µg of d-cloprostenol were injected intramuscularly. Oestrus detection was made via teaser bucks for 3 days starting 24 h after withdrawal of the IS. Eighteen bucks known to be fertile were used for breeding. Goats in the oestrus period were mated via natural breeding. The GnRH analogue lecirelin was injected intramuscularly at breeding in the GnRH0 group, on day 7 post-breeding in the GnRH7 group, and both at breeding and on day 7 post-breeding in the GnRH0+7 group. No injections were given to the control group. Blood samples for progesterone measurement were taken by jugular vena puncturing on days 3, 6, 7, 10, 13, 16, and 19 after breeding from 10 randomly chosen goats in all groups. The goats with a level of > 3.5 ng/mL of progesterone on day 21 post-breeding were evaluated as pregnant. Pregnancy was also viewed on day 50 after breeding by real-time ultrasonography (USG) with a 5-7.5 MHz convex probe. The oestrus rate was 96.23% (102/106) in the goats. The rates of onset of oestrus between 36-48 h, 48-60 h and 60 h and beyond were 38.7% (41/106), 21.7% (23/106) and 35.8% (38/106), respectively. The total pregnancy rate was 35.8% (38/106). There were no statistically significant differences (P > 0.05) found for the pregnancy rate, embryonic death rate or progesterone concentration of the groups. However, serum progesterone levels were statistically different in the GnRH7 group compared with the control group (P < 0.05). After synchronisation, various anti-luteolytic strategies can be used to support corpus luteum development and elevate progesterone concentration in the luteal phase to decrease embryonic loss and increase reproductive performance. Therefore, application of GnRH to support the luteal structure and to increase progesterone levels may be beneficial in terms of supporting embryonic life. The results showed that GnRH treatment on the day 7 post-breeding following oestrus induction, including FGA and PMSG, can increase serum progesterone levels in Damascus goats in the anoestrus period. However, following oestrus induction in the anoestrus period, it was seen that GnRH treatment at breeding or on day 7 after breeding did not have any positive effect on embryonic loss or reproductive performance. In conclusion, it was considered that this protocol could be implemented successfully, yielding a 35% pregnancy rate in Damascus goats in the anoestrus period, but embryonic loss must be deeply studied in detail.(AU)

Determining if reproductive failures in ewes at the semiarid region in the state of Bahia are related to the consumption of the species Cenostigma pyramidale (Tul.) Gagnon & G.P. Lewis, and this study was developed using pregnant ewes divided into six groups: G1, G2, G3, G4 with six animals each, G5 and G6 with ten animals. Each group received fence leaves in the proportion of 1%, 2%, 0.5%, and 0.25% of live weight (LW) respectively; G5 and G6, with ten animals each, receiving 0.25% and 0.5% of the LW, respectively, and the Control Group, comprising 16 ewes, were grass feeding (Cynodon dactylon). Ewes from G1 to G4 were the same, except for two, and started ingestion of the plant four days after ending of natural mating on the 80th day of gestation, while those regarding from G5 to G6 groups started ingestion on the 26th day of gestation ending on the 98 day. The ultrasonographic test was performed weekly. In G1 ewes (1%), there was an embryonic loss on the 32nd and 39th days of gestation and abortion on the 46th day. In G2 (2%), the embryo loss was earlier (on the 26th day of gestation), and abortion on the 46th day of gestation. In G3 group (0.5%), there was an embryonic loss around the 40th day of gestation. In G4 group (0.25%), it was observed the occurrence of one death lamb with bone malformations. In G6 (0.5%), abortion occurred later (108 days), followed by retained placenta. This was also verified in G5 group (0.25%). The presence of fetal malformation was found in death lambs born in G4 group, born alive from G5 and G6 groups, and one aborted from G6. In G5 and G6 groups, there were also genetic alterations on surviving lambs. In addition to these results, recurrent estrus was observed without gestation in G1, G2, G3, and G4 ewes. From the Control Group, 13 normal lambs were born without genetic alterations; furthermore, concerning a quadruple birth, three lambs were born dead. The results infer that species of C. pyramidale in low doses causes reproductive losses in pregnant ewes, therefore it is not recommended for sheep diet over the first 60 days of gestation.(AU)

Nos do G2 (2%) a perda embrionária foi mais precoce (26 dias), e aborto aos 46 dias. No G3 (0,5%), houve perda embrionária em torno dos 40 dias. No G4 (0,25%), verificou-se ocorrência de natimorto com malformações aos 150 dias de gestação. No G6 (0,5%) o aborto ocorreu mais tardiamente (108 dias), seguido de retenção de placenta. Essa ocorrência também foi verificada no G5 (0,25%). A presença de malformação fetal foi encontrada em fetos natimorto do G4, nascidos vivos do G5 e G6, e um abortado do G6. No G5 e G6 também foram observadas alterações de aprumos em cordeiros sobreviventes. Do Grupo Controle nasceram 13 borregos normais, porém uma ovelha apresentou gestação quádrupla com três natimortos. Os resultados inferem que C. pyramidale fenada em baixas doses causa perdas reprodutivas em ovelhas gestantes, não sendo por isso recomendada para a dieta de ovelhas durante os primeiros 60 dias de gestação.(AU)

The main objective of this study was to evaluate the risk factors for late embryonic loss (LEL) in supplemented grazing dairy cows. Additional objectives were to assess the incidence of LEL and its association with the reproductive performance of cows. A data set containing productive, reproductive, and health records of 13,551 lactations was used. A retrospective case-control study involving 631 cows with LEL (cases) and 2,524 controls (4 controls per case within each study year) was run. A case of LEL was defined when the embryo had no heartbeat or there was evidence of detached membranes or floating structures including embryo remnants by ultrasonography (US) at 28 to 42 d post-artificial insemination (AI), whereas a non-case was defined as a cow diagnosed with positive pregnancy by US 28 to 42 d post-AI and reconfirmed as pregnant 90 ± 7 d post-AI. Four controls per case were randomly selected from the non-cases with a temporal matching criterion (±3 d around the date of the fecundating AI of the case). Multivariable logistic models were offered with the following predictors: year of LEL (2011 through 2015), season of LEL (summer vs. fall vs. winter vs. spring), parity (1 vs. 2 vs. ≥3), uterine disease (UD), non-uterine disease (NUD), body condition score at parturition, body condition score at 28 to 42 d post-AI (BCS-LEL), days in milk (DIM), and daily milk yield (MY). Statistical significance was set at P < 0.05 and a tendency was set at P ≤ 0.10. We found that 4.7, 22, and 23% of cows had LEL, UD, and NUD, respectively. Cases tended to have higher daily MY than controls (32.5 vs. 31.8 kg); also, cases had much longer calving to pregnancy interval (226 vs. 118 d), lower hazard of pregnancy [hazard ratio = 0.39, 95% confidence interval (CI) = 0.35-0.43], and higher odds for non-pregnancy [odds ratio (OR) = 2.89, 95% CI = 2.37-3.54] than controls. We found that the odds for LEL increased with parity number (OR = 2.48, 95% CI = 1.99-3.08 for parity ≥3) and with BCS-LEL <2.50 (OR = 1.81, 95% CI = 1.33-2.47). Conversely, the odds for LEL decreased with BCS-LEL >3.00 (OR = 0.70, 95% CI = 0.53-0.91). The odds for LEL increased with UD (OR = 1.23, 95% CI = 1.01-1.49), NUD (OR = 1.24, 95% CI = 1.01-1.54), DIM (OR = 1.03, 95% CI = 1.00-1.05), and daily MY (OR = 1.14, 95% CI = 1.04-1.25) in univariable models only. Finally, the odds for LEL were not associated with year, season, DIM, and body condition score at parturition. In conclusion, LEL is associated with extended calving to pregnancy interval, and among its risk factors are parity number and BCS-LEL.

The feeding of diets with greater energy content than that needed for body maintenance following mating is believed to reduce embryonic survival in pigs. In swine operations, therefore, feed intake is often restricted during the first and second week of pregnancy to reduce embryo mortality. There is thought to be a relationship between feeding diets that result in energy intake that is greater than that needed for body maintenance and embryonic death. This relationship is associated with lesser than typical progesterone (P4) concentrations when feeding diets with greater energy content due to increased hepatic clearance. There is no consensus, however, as to whether feeding should be restricted during early pregnancy to avert this possible detrimental effect. Thus, the aim of this systematic review is to assess the effect in sows and gilts of feeding diets with different energy contents post-mating on embryonic survival, evaluating when possible, the relationship of a greater energy intake and P4 concentrations on embryonic survival. An electronic search was conducted of the PubMed, Science Direct, Scopus, Web of science, and Scielo databases during June 2018. A total of 109 articles were retrieved, and of these, only 16 articles were selected after applying the selection criteria. There was no negative effect of a greater feed intake than that needed for body maintenance after breeding in 75% of the experiments. Results from 35% of the experiments indicated feeding early pregnant sows a diet with greater energy content than that needed for body maintenance resulted in augmented embryonic death. In 66.7% of the experiments, in which there was assessment of P4 concentration, there was no negative effect of feeding after farrowing a diet with greater energy than that needed for body maintenance. In conclusion, it appears that restricted feed intake in early pregnancy is no longer relevant when there are modern prolific dam lines utilized in swine production enterprises because dietary energy of as great as 54 MJ ME/day had no detrimental effect on embryo survival.

High reproductive performance is an element factor for profitability in dairy herds; although, pregnancy loss of non-infectious nature during early and mid-to-late embryonic period is increasing in dairy cattle. Based on that, the present study aimed to determine the incidence of pregnancy loss before and after 60 days of insemination, and to identify some non-infectious causes that could enhance it. The herd was composed by 600 crossbred dairy cows and those with a corpus luteum (CL) were treated with prostaglandinF2α, then inseminated, on the other hand, those without a CL were submitted to a timed artificial insemination protocol (TAI). Pregnancy losses rates were analyzed by logistic regression by SAS, and differences were considered significant when P < 0.05. The overall pregnancy loss and mid-to-late pregnancy loss were not affected by animal category (P > 0.05); although, early pregnancy loss was higher in cows than in heifers (11.90 vs. 3.39%). The early pregnancy loss was higher in those cows that calved on spring/summer when compared to those calved on fall/winter (9.22% vs. 16.11%), moreover, those inseminated during spring/summer tended to have higher early pregnancy loss when compared to those inseminated on fall/winter (13.35% vs. 8.57%). In conclusion, when some of non-infectious causes were evaluated, it was observed that cows that calved on spring/summer had higher pregnancy loss. At this point, strategies should be developing to minimize pregnancy loss in dairy herds, as it could be considered an important reproductive problem.

The objectives were to evaluate the effects of the administration of either eCG or progesterone (P4) alone or combined on endogenous P4 concentrations and pregnancy per AI in lactating dairy cows. Cows received a P4-releasing intravaginal device (PRID) and estradiol benzoate on D-8. The PRID was removed and a PGF2α injection was given on D-3. An estradiol cypionate was given on D-2 and TAI was performed on D0. On D-2, cows were randomly allocated to treatments in a 2×2 factorial design: Control-saline solution on the D-2 and D+3 (n=104), eCG - 400IU eCG on D-2 (n=93), P4 - 600mg of P4 on D+3 (n=106), and eCG+P4 - 400IU eCG on D-2 and 600mg of P4 on D+3 (n=95). Blood samples were collected on days three, four, and thirteen and pregnancy diagnoses were performed at 32 and 46 days after AI. There was no interaction between eCG and P4 injection. Cows treated with eCG and with P4 injection had higher serum P4 on Day +4. On Day +13 serum P4 was lower in eCG-untreated primiparous cows (Interaction eCG×parity). Cows with serum P4<4.57ng/mL on Day +13 had lower probability to be pregnant on day 32. P/AI on days 32 and 46 and embryonic losses were not influenced by eCG and P4 injection. In conclusion, the addition of 400IU of eCG on D-2 and/or 600mg of P4 on D+3 to the present TAI protocol did not increase P/AI.

Estudou-se o efeito da idade sobre a fertilidade de éguas inseminadas com sêmen asinino diluído, resfriado e armazenado. Os ciclos foram acompanhados por palpação transretal e rufiação, sendo as inseminações realizadas às terças, quintas e sábados, a partir da detecção de um folículo de 3,0 a 3,5cm de diâmetro, em um dos ovários, até a ovulação. O sêmen de cinco jumentos da raça Pêga foi diluído nos diluidores de leite em pó desnatado-glicose ou glicina-gema de ovo, resfriado a 5ºC e armazenado por 12 horas, sendo a dose inseminante de 400 x 106 espermatozoides móveis (no momento da diluição final, pré-resfriamento). Os resultados de 195 ciclos estrais, referentes a 141 éguas, foram agrupados em classes, de acordo com a idade das éguas: 2,5 a 6 anos, 6,5 a 10 anos, 10,5 a 14 anos e 14,5 a 19 anos. As taxas de concepção, ao primeiro ciclo, foram de 68,42%, 50,75%, 46,88% e 52,17% e, após quatro ciclos, de 69,57%, 47,92%, 46,34% e 45,71% para as faixas etárias de 2,5 a 6, 6,5 a 10, 10,5 a 14 e 14,5 a 19 anos, respectivamente (P>0,05). The effect of the mare age on fertility of mares inseminated with diluted, cooled and stored jackass semen was studied. The females were controlled by transrectal palpation and teasing, and inseminated every Tuesday, Thursday and Saturday, since the detection of a 3.0 to 3.5cm follicle diameter, in one of the ovaries, until ovulation. The semen of five Pêga jackasses was diluted in skim milk-glucose or in egg yolk-glicine extender and cooled at 5ºC for 12 hours, with the inseminate dose of 400 x 106 motile spermatozoa (at the moment of the final dilution, before cooling). The results of 195 cycles of 141 mares were grouped, in accordance with the age: 2.5 to 6 years, 6.5 to 10 years, 10.5 to 14 years and 14.5 to 19 years. The pregnancy rates for the first cycle were 68.42%, 50.75%, 46.88% and 52.17%, and after four cycles, the pregnancy rates/cycle were 69.57%, 47.92%, 46.34% and 45.71%, respectively for 2.5 to 6, 6.5 to 10, 10.5 to 14 and 14.5 to 19 years (P>0,05).
