RESUMO

Cyclodextrins are nanometric cyclic oligosaccharides with amphiphilic characteristics that increase the stability of drugs in pharmaceutical forms and bioavailability, in addition to protecting them against oxidation and UV radiation. Some of their characteristics are low toxicity, biodegradability, and biocompatibility. They are divided into α-, ß-, and γ-cyclodextrins, each with its own particularities. They can undergo surface modifications to improve their performances. Furthermore, their drug inclusion complexes can be made by various methods, including lyophilization, spray drying, magnetic stirring, kneading, and others. Cyclodextrins can solve several problems in drug stability when incorporated into dosage forms (including tablets, gels, films, nanoparticles, and suppositories) and allow better topical biological effects of drugs at administration sites such as skin, eyeballs, and oral, nasal, vaginal, and rectal cavities. However, as they are nanostructured systems and some of them can cause mild toxicity depending on the application site, they must be evaluated for their nanotoxicology and nanosafety aspects. Moreover, there is evidence that they can cause severe ototoxicity, killing cells from the ear canal even when applied by other administration routes. Therefore, they should be avoided in otologic administration and should have their permeation/penetration profiles and the in vivo hearing system integrity evaluated to certify that they will be safe and will not cause hearing loss.

Abstract Introduction: the blue-eyed pleco {Panaque cochliodon) is a species endemic to the Cauca and Magdalena basin. However, not much information is available on this species, including the absence of an adequate sedation protocol for captive work with this species. Objetive: this research standardizes a sedation protocol for P. cochliodon. Materials and methods: for this purpose, different dosages and concentrations of the sedative Eugenol ranging from 20 to 40 mg/L were evaluated and 4 types of sedation and 5 types of recovery were adopted, thus finding the appropriate degrees of sedation for the different confinement management procedures that were performed with this species. Results: all specimens reached type 4 sedation and type 5 recovery without mortality. Significantly, the concentration at which the specimens took the longest time to achieve type 4 sedation and type 5 recovery was 20 mg / L, the other concentrations did not show differences between them, but indicate an inverse trend with respect to the concentration and experimental time. Conclusions: all the concentrations evaluated are safe for the management of P. cochliodon of 314 ± 11.6 gr, there was no mortality; concentrations of 30 to 35 mg / L, offer 2.6 minutes of work, without apparent deleterious effects on the species.

RESUMO

SUMMARY OBJECTIVES: This study aimed to develop artificial intelligence and machine learning-based models to predict alterations in liver enzymes from the exposure of low annual average effective doses in radiology and nuclear medicine personnel of Institute of Nuclear Medicine and Oncology Hospital. METHODS: Ninety workers from the Radiology and Nuclear Medicine departments were included. A high-capacity thermoluminescent was used for annual average effective radiation dose measurements. The liver function tests were conducted for all subjects and controls. Three supervised learning models (multilayer precentron; logistic regression; and random forest) were applied and cross-validated to predict any alteration in liver enzymes. The t-test was applied to see if subjects and controls were significantly different in liver function tests. RESULTS: The annual average effective doses were in the range of 0.07-1.15 mSv. Alanine transaminase was 50% high and aspartate transaminase was 20% high in radiation workers. There existed a significant difference (p=0.0008) in Alanine-aminotransferase between radiation-exposed and radiation-unexposed workers. Random forest model achieved 90-96.6% accuracies in Alanine-aminotransferase and Aspartate-aminotransferase predictions. The second best classifier model was the Multilayer perceptron (65.5-80% accuracies). CONCLUSION: As there is a need of regular monitoring of hepatic function in radiation-exposed people, our artificial intelligence-based predicting model random forest is proved accurate in prediagnosing alterations in liver enzymes.

As variáveis analisadas foram: hormônios testosterona e cortisol, leucócitos totais, neutrófilos, linfócitos, monócitos e as enzimas creatina kinase (CK) e lactato desidrogenase (LDH).

The objective of the present study was to analyze the acute hormonal, immunological and enzymatic responses of professional basketball players to a basketball game. The sample was composed of eight basketball athletes, with a minimum of 4 years’ experience in basketball. A real game was simulated with a total duration of 40 minutes, divided into two halves of 20 minutes each and an interval of 10 minutes between halves. Blood samples were collected before and immediately after the game (20 ml, vacuum tube system). The variables analyzed were: testosterone and cortisol hormones, total leukocytes, neutrophils, lymphocytes, monocytes and the enzymes creatine kinase (CK) and lactate dehydrogenase (LDH). Statistical analysis was with descriptive statistics and the Student’s t test for paired samples to p≤0.05. The pre (13.34 nmol/L and 301.97 nmol/L) and post game (17.34 nmol/L and 395.91 nmol/L) levels of testosterone and cortisol were statistically different, with higher levels after the game for both hormones. The immune cell counts exhibited significant differences for total leukocytes (6393.75 nmol/L and 9158.75 nmol/L) and neutrophils (3532.5 nmol/L and 6392.62 nmol/L), with levels being higher after the game. No statistical differences were observed for the enzymatic variables. Therefore, based on the markers analyzed, testosterone and cortisol exhibited pronounced increases after the game and the same behavior was observed for total leukocytes and neutrophils.
