Sperm quality is defined as the sperm cell ability to successfully fertilize eggs and allow normal embryo development⁠. Few studies explore sperm quality using aquatic invertebrates. Parhyale hawaiensis is a marine amphipod with a circumtropical distribution and considered a model for evolution, development, and ecotoxicological studies. We aimed to develop a methodology to collect sperm cells of P. hawaiensis and evaluate their viability and DNA damage (comet assay). We directly exposed the sperm cells to different mutagenic agents to optimize/develop the protocols. Then, as a proof of concept, we exposed the males to mutagenic compounds (EMS, benzo[a]pyrene (BaP), azo and anthraquinone dyes) at non-lethal concentrations verified by the proposed viability test and analyzed their sperm cells for DNA damage (comet assay). Organisms exposed to EMS presented a clear concentration response in the DNA damage response. We also showed that BaP was able to induce a statistically significant increase in DNA damage of the sperm cells. For the two dyes, although DNA damage increased, statistically differences were not observed. We believe we successfully developed a test to detect genotoxicity of chemicals in sperm cells using an invertebrate model. The protocol for sperm cell viability needs to be further explored with different chemicals to verify its utility as a toxicity endpoint. The developed genotoxicity test has the advantages to employ organisms that are easily cultivated in reduced space, use simple laboratory resources and reduced amount of material and reagents. Positive responses with this model could be used to disclose new germ cell mutagen candidates which could be further confirmed in vertebrates' systems.

Uca maracoani is a fiddler crab found in estuaries along the western Atlantic coast, with a notable preference for euhaline environments. This study aimed to analyze the population structure and dynamics of this species in an estuary on the North Coast of Brazil, specifically in an area of the upper estuary where seasonal rainfall fluctuations result in significant changes in salinity. Monthly crab samples were taken from December 2013 to November 2015, together with measurements of environmental variables, such as water and climate parameters. The population maintains a balanced sex ratio; however, males are generally larger, with lower mortality rates and longer lifespans than females. Reproduction is continuous but mainly takes place in the dry season when salinity levels are higher (above 12‰). Higher crab densities have been observed during the rainy season when, despite lower salinity levels (below 10‰), the conditions for survival (food availability and milder climate) seem to be more favorable. The estimated average annual biomass and production for the population were 2.62 g AFDM m-2 and 5.43 g AFDM m-2 year-1, respectively, characterized by a high turnover rate (P/B = 2.10 year-1). Our results suggest that U. maracoani has thriving populations in the Amazon coast's mangroves, benefiting from the vast muddy intertidal zone and the high organic content delivered by the estuaries.

RESUMO

The neuroendocrine system of crustaceans is complex and regulates many processes, such as development, growth, reproduction, osmoregulation, behavior, and metabolism. Once stimulated, crustaceans' neuroendocrine tissues modulate the release of monoamines, ecdysteroids, and neuropeptides that can act as hormones or neurotransmitters. Over a few decades, research has unraveled some mechanisms governing these processes, substantially contributing to understanding crustacean physiology. More aspects of crustacean neuroendocrinology are being comprehended with molecular biology, transcriptome, and genomics analyses. Hence, these studies will also significantly enhance the ability to cultivate decapods, such as crabs and shrimps, used as human food sources. In this review, current knowledge on crustacean endocrinology is updated with new findings about crustacean hormones, focusing mainly on the main neuroendocrine organs and their hormones and the effects of these molecules regulating metabolism, growth, reproduction, and color adaptation. New evidence about vertebrate-type hormones found in crustaceans is included and discussed. Finally, this review may assist in understanding how the emerging chemicals of environmental concern can potentially impair and disrupt crustacean's endocrine functions and their physiology.

Host-microbe interactions are ubiquitous and play important roles in host biology, ecology, and evolution. Yet, host-microbe research has focused on inland species, whereas marine hosts and their associated microbes remain largely unexplored, especially in developing countries in the Southern Hemisphere. Here, we review the current knowledge of marine host microbiomes in the Southern Hemisphere. Our results revealed important biases in marine host species sampling for studies conducted in the Southern Hemisphere, where sponges and marine mammals have received the greatest attention. Sponge-associated microbes vary greatly across geographic regions and species. Nevertheless, besides taxonomic heterogeneity, sponge microbiomes have functional consistency, whereas geography and aging are important drivers of marine mammal microbiomes. Seabird and macroalgal microbiomes in the Southern Hemisphere were also common. Most seabird microbiome has focused on feces, whereas macroalgal microbiome has focused on the epibiotic community. Important drivers of seabird fecal microbiome are aging, sex, and species-specific factors. In contrast, host-derived deterministic factors drive the macroalgal epibiotic microbiome, in a process known as "microbial gardening". In turn, marine invertebrates (especially crustaceans) and fish microbiomes have received less attention in the Southern Hemisphere. In general, the predominant approach to study host marine microbiomes has been the sequencing of the 16S rRNA gene. Interestingly, there are some marine holobiont studies (i.e., studies that simultaneously analyze host (e.g., genomics, transcriptomics) and microbiome (e.g., 16S rRNA gene, metagenome) traits), but only in some marine invertebrates and macroalgae from Africa and Australia. Finally, we introduce an ongoing project on the surface microbiome of key species in the Strait of Magellan. This is an international project that will provide novel microbiome information of several species in the Strait of Magellan. In the short-term, the project will improve our knowledge about microbial diversity in the region, while long-term potential benefits include the use of these data to assess host-microbial responses to the Anthropocene derived climate change.

The crayfish plague is an emerging infectious disease caused by the pathogen Aphanomyces astaci (Oomycota), which is responsible for the decimation of Eurasian freshwater crayfish. This pathogen can coexist with the North American crayfish. These are chronic carriers of the disease as consequence of an immune response that can contain the growth of the pathogen without killing it. The origin of A. astaci locates in the southeastern United States and coincides with the origin of the family Cambaridae. This diverse family of decapods is distributed in North America from southern Canada to Honduras. However, only the native crayfish species from Canada and the USA have been examined for the presence of A. astaci. In this study, we describe for the first time the presence of A. astaci in Mexico in a population of the native species Cambarellus montezumae. By analyzing the small (rrnS) and large (rrnL) mitochondrial ribosomal regions, we showed the presence of two haplotypes of A. astaci within the same population (d1-haplotype and, a novel haplotype that was named, mex1-haplotype). The finding of A. astaci in Mexico confirms the occurrence of this pathogen within the range of the family Cambaridae. The individuals of C. montezumae appear to be chronic carriers of A. astaci, indicated by the lack of documented crayfish plague outbreaks in this population, similar to the pattern observed in other North American species. Thus, the results are of special concern to susceptible species of southern regions of America, i.e., Parastacidae. Therefore, this work emphasizes the need to better understand the distribution and genetic diversity of A. astaci within the distribution range of the natural carriers, i.e., North American species, especially the unexplored area of the family Cambaridae.

Considerable efforts have been made by modern aquaculture to mitigate the environmental damages caused by its practices while also attempting to improve the quality of the aquatic organisms by promoting alternatives, such as the use of natural products, like garlic (Allium sativum), and instead of chemical agents. Garlic has multiple properties, including antifungal, antibacterial, antiviral, antitoxic, and anticancer effects. In fish, the antiparasitic activity of garlic is one of the most reported effects in the literature, mainly using immersion baths for aquatic organisms. Using garlic also has an antimicrobial effect on the culture of aquatic organisms. Therefore, this review focuses on the impact of garlic on the health and production of aquatic organisms.

Dilocarcinus pagei is a South American crab commonly found in fishponds. As crabs are a source of astaxanthin (AST) and food inputs, this preliminary research aimed at studying the composition of females and males of this species to assess its potential for commercial applications, and at optimising the extraction of AST with edible oils to promote its use as an ingredient for the nutraceutical, pharmaceutical and feed industries. The chemical composition differed between males and females only in moisture, and the values obtained were: 65.4 ± 1.0% and 72.5 ± 3.1% moisture, 45.7-40.3% d.m. minerals, 22.0-24.1% d.m. fibres, 18.2-17.4% d.m. proteins, and 10.4-11.1% d.m. lipids. The Box-Behnken design was applied and validated for extraction with soya bean and sunflower oils, adjusting the oil:crab ratio, temperature, and extraction time. The optimal conditions found consisted of 140 mL/g, 90 °C and 170 min for soya bean oil, reaching an accumulation of AST of 50 ± 5 µg/g crab d.m. For sunflower oil, the conditions were 60 mL/g, 90 °C and 161 min, reaching 31 ± 3 µg/g crab d.m. Finally, the amounts of AST obtained using soya bean oil were higher than those obtained using sunflower oil; thus, soya bean oil would be recommended as a solvent to extract the pigment.

The echinoderm Loxechinus albus has a symbiotic relationship with the pinnotherid crustacean Pinnaxodes chilensis. Females of the crustacean develop in the terminal section of the sea urchin's digestive system, remaining there for life. This relationship has been suggested as commensalism. However, a potential negative impact on gonadal development and on the morphology of the sea urchin's digestive system suggest that it is instead parasitic. To study if there is a negative impact of the crustacean symbiont on the host, specimens of L. albus of all sizes were collected from a rocky shore in southern Chile. The gonadal and somatic tissues of sea urchins that were and were not harboring the pinnotherid were weighed and compared. Our results show that the presence of the pinnotherid was related to sea urchin gonads of lower biomass, decreased gonadosomatic index levels, and alterations in the morphology of the terminal portion of the host digestive system. The lower gonadal biomass suggests a negative impact on gamete production as well as a diversion of energy due to changes of the digestive system tissues and the potential consumption of algal food by the resident crustacean. These results suggest that the prolonged relationship between these two species is one of parasitism rather than one of commensalism.

N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine-quinone (6PPD-quinone), an oxidation product of the tire additive, 6PPD, has been associated with high mortality of salmonids (0.1 µg/L). The objective of this study was to determine the acute toxicity using neonates and mutagenicity (micronuclei in hemolymph of exposed adults) of 6PPD-quinone in the marine amphipod Parhyale hawaiensis. Also, we studied its mutagenicity in the Salmonella/microsome assay using five strains of Salmonella with and without metabolic system (rat liver S9, 5%). 6PPD-quinone did not present acute toxicity to P. hawaiensis from 31.25 to 500 µg/L. Micronuclei frequency increased after 96 h-exposure to 6PPD-quinone (250 and 500 µg/L) when compared to the negative control. 6PPD-quinone also showed a weak mutagenic effect for TA100 only in the presence of S9. We conclude that 6PPD-quinone is mutagenic to P. hawaiensis and weakly mutagenic to bacteria. Our work provides information for future risk assessment of the presence of 6PPD-quinone in the aquatic environment.

Ocypode quadrata, a Ghost crab species found along the western Atlantic coast, is considered a bioindicator of anthropogenic impact on sandy beaches. Ghost Crabbing, a touristic activity in which ghost crabs are chased just for fun, is a potentially threatening activity for this crab. In crustaceans, metabolites such as glucose and lactate, and the gene expression of crustacean hyperglycemic hormone (CHH) and heat shock proteins (HSPs) increase when the animals are exposed to several types of stress, including alterations in temperature, salinity, or exposure to xenobiotics. This work was developed to identify if being chased by humans would affect these markers of stress in this species of crab. The effects of chasing stress on hemolymph and tissue metabolites and the gene expression levels of CHH and HSP70 were investigated. The levels of lactate in the hemolymph of stressed crabs were six times higher than those of control crabs immediately after chasing and decreased progressively during recovery, indicating an active anaerobic metabolism during the stress. On the contrary, glucose levels in the hemolymph of the stressed crabs increased progressively from 30 to 60 min after chasing, indicating an inverse correlation between glucose and lactate and the conversion of lactate to glucose by gluconeogenesis. In stressed crabs, the levels of triglycerides in the hemolymph decreased 30 min after chasing, while the opposite tended to occur in the hepatopancreas, indicating that during recovery, the crabs use triglycerides as energy source to sustain aerobic metabolism. Finally, this study demonstrates that ghost crabs are stressed by minimum human contact and that "ghost crabbing" must not be encouraged as a tourist activity.

Introduction: Neohelice granulata crabs live in mudflats where they prey upon smaller crabs. Predatory behavior can be elicited in the laboratory by a dummy moving at ground level in an artificial arena. Previous research found that crabs do not use apparent dummy size nor its retinal speed as a criterion to initiate attacks, relying instead on actual size and distance to the target. To estimate the distance to an object on the ground, Neohelice could rely on angular declination below the horizon or, since they are broad-fronted with eye stalks far apart, on stereopsis. Unlike other animals, binocular vision does not widen the visual field of crabs since they already cover 360° monocularly. There exist nonetheless areas of the eye with increased resolution. Methods: We tested how predatory responses towards the dummy changed when animals' vision was monocular (one eye occluded by opaque black paint) compared to binocular. Results: Even though monocular crabs could still perform predatory behaviors, we found a steep reduction in the number of attacks. Predatory performance defined by the probability of completing the attacks and the success rate (the probability of making contact with the dummy once the attack was initiated) was impaired too. Monocular crabs tended to use frontal, ballistic jumps (lunge behavior) less, and the accuracy of those attacks was reduced. Monocular crabs used prey interception (moving toward the dummy while it approached the crab) more frequently, favoring attacks when the dummy was ipsilateral to the viewing eye. Instead, binocular crabs' responses were balanced in the right and left hemifields. Both groups mainly approached the dummy using the lateral field of view, securing speed of response. Conclusion: Although two eyes are not strictly necessary for eliciting predatory responses, binocularity is associated with more frequent and precise attacks.

The biochemical effects of sublethal exposure to polyethylene microplastics (PEM) of 40-48 µm particle size and the flame retardant tetrabromobisphenol A (TBBPA), a plastic additive, on the freshwater shrimp Palaemonetes argentinus were assessed. Here, we postulate that the use of enzyme and thyroid hormones as biomarkers contributes to the knowledge of the effects of microplastics and plastic additives on freshwater crustaceans. To address this, we evaluated the activities of acetylcholinesterase (AChE), glutathione S-transferase (GST), and carboxilesterase (CbE, using 1-naphthyl acetate (NA) as substrate) and levels of the thyroid hormones thyroxine (T4) and triiodothyronine (T3) after shrimp were exposed (for 96 h) to these xenobiotics at environmentally realistic concentrations. The results showed that the mixture of both xenobiotics led to a decrease in AChE and GST activities and increased T4 levels. We suggest that physiological processes could be compromised in freshwater organisms when exposed to microplastics and TBBPA together, and this could ultimately affect upper levels of the food web.

The red crab, Pleuroncodes planipes, is a decapod crustacean abundant off the Pacific coast of the Baja California Peninsula. This species is caught and used in preparing animal feed, such as flour, particularly for aquaculture. Levels of calcium (Ca), cadmium (Cd), copper (Cu), iron (Fe), lead (Pb), magnesium (Mg), manganese (Mn), nickel (Ni), phosphorus (P), and zinc (Zn) were measured in red crabs collected from three geographic zones during three cruises in different seasons. Significant differences were found in the levels of Ca, Cd, Cu, Fe, Mg, Ni, P, and Zn between the two El Niño years (cruises C1 and C3, based on a threshold of ±0.5 °C for the Oceanic Niño Index). The highest concentrations of most elements were observed in the south of the Baja California Peninsula, a highly productive area influenced by upwelling events. Our findings suggest that while environmental temperature plays a central role in the benthic or pelagic distribution of red crabs, their content and variability of trace and macro elements appear to be associated with the presence of oceanic conditions, such as upwelling and potential changes in the composition of their diet associated with the depth in which these crustaceans are collected.

Shrimp antilipopolysaccharide factors (ALFs) form a multifunctional and diverse family of antimicrobial host defense peptides (AMPs) composed of seven members (groups A to G), which differ in terms of their primary structure and biochemical properties. They are amphipathic peptides with two conserved cysteine residues stabilizing a central ß-hairpin that is understood to be the core region for their biological activities. In this study, we synthetized three linear (cysteine-free) peptides based on the amino acid sequence of the central ß-hairpin of the newly identified shrimp (Litopenaeus vannamei) ALFs from groups E to G. Unlike whole mature ALFs, the ALF-derived peptides exhibited an α-helix secondary structure. In vitro assays revealed that the synthetic peptides display a broad spectrum of activity against both Gram-positive and Gram-negative bacteria and fungi but not against the protozoan parasites Trypanosoma cruzi and Leishmania (L.) infantum. Remarkably, they displayed synergistic effects and showed the ability to permeabilize bacterial membranes, a mechanism of action of classical AMPs. Having shown low cytotoxicity to THP-1 human cells and being active against clinical multiresistant bacterial isolates, these nature-inspired peptides represent an interesting class of bioactive molecules with biotechnological potential for the development of novel therapeutics in medical sciences.

PURPOSE: The present study deals with a redescription of the copepod Lernaeenicus longiventris Wilson, 1917 (Copepoda: Siphonostomatoida: Pennellidae) found on the Pacific crevalle jack Caranx caninus Günther, using morphological and molecular analyses. METHODS: Fish were collected off Mazatlán Port (23° 12' N, 106° 26' W), in the State of Sinaloa, Mexico (southeastern Gulf of California). The copepods were morphologically analyzed by light microscopy. Sequences of the COI mtDNA gene were generated for the first time for this species. These sequences were compared to COI sequences from six species of Lernaeenicus available in GenBank. RESULTS: The specimens of the present study exhibited a cephalosome without apparent lateral processes, which were originally described for L. longiventris. No remarkable differences were observed with previous descriptions regarding appendages and body proportions. The phylogenetic tree based on COI sequences showed that L. longiventris was closer to L. radiatus although with low bootstrap values support in ML tree, both species formed a sister clade of L. sprattae. CONCLUSIONS: Lernaeenicus longiventris is the unique species of the genus in the Mexican Pacific and the Gulf of California, and also the unique species of Lernaeenicus infecting C. caninus. Molecular data of L. longiventris from host and locality type are required to avoid misidentification of this species.

We present the novel reference genome of the Versatile Fairy Shrimp, Branchinecta lindahli. The Versatile Fairy Shrimp is a freshwater anostracan crustacean found across the western United States from Iowa to Oregon and from Alberta to Baja California. It is an ephemeral pool specialist, living in prairie potholes, irrigation ditches, tire treads, vernal pools, and other temporary freshwater wetlands. Anostracan fairy shrimp are facing global declines with 3 species in California on the Endangered Species list. This species was included in the California Conservation Genomics Project to provide an easily accessible reference genome, and to provide whole-genome resources for a generalist species, which may lead to new insights into Anostracan resiliency in the face of climate change. The final gapped genome comprises 15 chromosome-length scaffolds covering 98.63% of the 384.8 Mb sequence length, and an additional 55 unscaffolded contigs.

A new species of parasitic copepod, Lepeophtheirus mondacola sp. nov. (Siphonostomatoida; Caligidae), is described based on female and male specimens obtained from the shortjaw leatherjacket Oligoplites refulgens (Actinopterygii; Perciformes; Carangidae), captured in the southeastern Gulf of California off northwestern Mexico. The new species can be separated from its congeners by a combination of characters that includes: adult female with a subquadrate genital complex bearing slightly protruded posterolateral corners, two indistinct somites on the abdomen which, when combined together, is about two times longer than wide, a caudal ramus that is twice as long as it is wide, a postantennal process comprising a stout base and short claw, a dentiform process of the maxillule with two unequal tines, a maxilliped with a stout protopod and subchela, a sternal furca with a pair of bifurcated tines, a leg 3 exopod composed of 2 segments, five setae on the distal endopodal segment of leg 3 and a leg 4 exopod composed of three segments and armed with one long and two short apical spines on the distal exopodal segment; adult male with a suborbicular genital complex, an abdomen composed of one short and one long, indistinctly separated somites, a caudal ramus that is twice as long as it is wide, a stout postantennal process, a small triangular process at the base of the inner tine of the maxillulary dentiform process and a 3-segmented exopod on leg 4. Lepeophtheirus mondacola sp. nov. represents the first record of a species of Lepeophtheirus from a member of Oligoplites and the second caligid species reported from O. refulgens.

Edible brachyurans are recognized as a popular source of food due to their delicious taste and nutritional quality. This study investigated the nutrient compositional variation and interrelationship in the muscle of wild and plastic-reared Cardisoma armatum. The plastic-reared crab had 27.81 ± 2.29 g 100 g-1protein while crab from the wild contained 22.45 ± 2.65 g 100 g-1protein. The difference in protein content of plastic-reared and its counterpart from the mangrove swamp was not significantly different(p > 0.05). This is also true of other proximate composition except that wild crabs were slightly higher in both crude fat (2.68 ± 0.35 g 100 g-1) and carbohydrate (5.89 ± 3.05 g 100 g-1). Generally, the total energy contributed due to protein, carbohydrate and fat in the tissues of both wild and plastic-reared C. armatumare similar. In the wild crab, contents of calcium (16083.27 ± 2127.90 mg 100 g-1) and phosphorus (1191.42 ± 199.21) were relatively higher, while contents of magnesium (368.69 ± 111.05 mg 100 g-1), sodium (125.30 ± 11.18 mg 100 g-1) and potassium (87.36 ± 7.27 mg 100 g-1) were relatively higher inplastic-reared crabs. All significant mineral ratios in wild and plastic-reared C. amartumfall within acceptable range. The positive relationship in the nutritional quality indicates that changes in proximate composition are associated with changes in mineral contents of the crab tissue.(AU)

Crustins represent the largest and most diverse family of antimicrobial peptides (AMPs) found in crustaceans. They are classically defined as disulfide-rich peptides/polypeptides holding a typical Whey Acidic Protein (WAP) domain at the C-terminal end. This WAP domain has eight cysteine residues forming a tightly packed structure, the four-disulfide core (4DSC) motif, that is also found in other proteins displaying protease inhibitory properties. Crustins are highly diverse in terms of primary structure, size and biochemical features, thus exhibiting a series of biological functions beyond their antimicrobial properties. In order to better categorize the distinct crustin members, different classification systems have been proposed. In this review, we discuss the current classification systems and explore the biological implication of the impressive molecular diversity of this unique AMP family. We also summarize the recent findings on the role of these effectors in crustacean immunity and homeostasis as well as in host-microbe interactions.

In ecdysozoan animals, moulting entails the production of a new exoskeleton and shedding of the old one during ecdysis. It is induced by a pulse of ecdysone that regulates the expression of different hormonal receptors and activates a peptide-mediated signalling cascade. In Holometabola, the peptidergic cascade regulating ecdysis has been well described. However, very little functional information regarding the neuroendocrine regulation of ecdysis is available for Hemimetabola, which display an incomplete metamorphosis. We use Rhodnius prolixus as a convenient experimental model to test two hypotheses: (1) the role of neuropeptides that regulate ecdysis in Holometabola is conserved in hemimetabolous insects; and (2) the neuropeptides regulating ecdysis play a role in the regulation of female reproduction during the adult stage. The RNA interference-mediated reduction of ecdysis triggering hormone (ETH) mRNA levels in fourth-instar nymphs resulted in lethality at the expected time of ecdysis. Unlike in holometabolous insects, knockdown of eth and orcokinin isoform A (oka) did not affect oviposition in adult females, pointing to a different endocrine regulation of ovary maturation. However, eth knockdown prevented egg hatching. The blockage of egg hatching appears to be a consequence of embryonic ecdysis failure. Most of the first-instar nymphs hatched from the eggs laid by females injected with dsRNA for eclosion hormone (dsEH), crustacean cardioactive peptide (dsCCAP) and dsOKA died at the expected time of ecdysis, indicating the crucial involvement of these genes in post-embryonic development. No phenotypes were observed upon corazonin (cz) knockdown in nymphs or adult females. The results are relevant for evolutionary entomology and could reveal targets for neuropeptide-based pest control tools.
