RESUMO

In mouse embryos, inside cells are allocated in 16-cell embryos through a well-orchestrated sequence of events involving compaction and polarization. The emergence of inside cells is of great importance as itl later gives rise to the inner cell mass and epiblast. In this study, we report the sequence of critical events in embryology (compaction, inside cells allocation and fragmentation) in bovine 72 h.p.i. 9-16 cell embryos, while also investigating the effects of X-sorted semen on these events. We found a wide distribution of total cell numbers among embryos, attributed to an asynchronous cleavage pattern and blastomere death. Additionally, 13% of embryos displayed irregular shapes. The establishment of the inside cell compartment increased (p < 0.01) in embryos with more cells. However, only 53.8% of 16-cell embryos presented inside cells. Compaction was present in 32.4% embryos and was positively correlated (p = 0.03, OR 3.02) with the establishment of inside cells, occurring independently of cell number. Fragmentation was present in 36% embryos, being more frequent (p = 0.01) in embryos with lower cell numbers. A possible association between irregular shape and fragmentation was considered (p = 0.06). The use of X-sorted semen had no effect on most evaluated parameters. However, it did have a marked effect on cleavage rate (p < 0.01) and the arrest of 2- and 4- cell embryos. In conclusion, bovine embryos exhibit an asynchronous cleavage pattern, high levels of fragmentation, and demonstrate compaction and inside cell allocation later in development compared to mouse embryos. Semen X-sorting has major effects on cleavage and embryo arrest. Further studies are needed to elucidate the association between irregularly shaped embryos and fragmentation, as well as the effects of sex on inside cell allocation.

RESUMO

The dimensions of mechanized agricultural systems depend on the edaphoclimatic conditions, crops, and work regimes. This study aimed to geographically estimate the monthly available time and number of favorable hours for agricultural field spraying in the state of Mato Grosso do Sul, Brazil. The meteorological restrictions imposed during unfavorable hours were as follows: ambient temperature above 32 ºC, relative humidity below 50 %, wind speed above 15 km h-1, and volumetric soil humidity above 39 % (equivalent to 90 % of the available water capacity). Mathematical models were then developed considering a period of ten years, which used historical data from the ground monitoring stations of the National Institute of Meteorology within the region. The subsequent algorithm was programmed and installed in a web server to simulate the time required for agricultural field spraying. During the cropping period in the region, there were climatic restrictions on performing agricultural spraying, with relative humidity being the variable with the most significant impact. However, soil moisture conditions restricted the available time for agricultural spraying more than the wind speed, relative air humidity, or ambient temperature.

The bulk density values indicative of compaction in clayey soils correspond to a wide range of values (1.30-1.47 Mg m-3), due to the diverse types of soil management. Our hypothesis is that if we consider bulk density values within similar management groups (those that fall to the ground and those that do not fall), the values will be more accurate within each management group. Our objective in this work was to analyze using the concepts of (Yates & Cochran, 1938; Whitehead, 2002) what is a suitable statistical way of grouping these results to reach a consensus regarding a tested hypothesis, the greatest number of density values surveyed between 1977 and 2021, considered for the study of compaction in clayey Oxisols under different managements, to verify whether our hypothesis will be confirmed. We selected 1,521 studies with bulk densities in the literature, corresponding to 44 years (1977 and 2021), which were analyzed by a statistical technique that integrates the results of two or more independent studies on the same subject and combines them into a summary measure (portion forestry). From the data collected in the literature, it was possible to separate two groups of bulk density and management in clayey soils indicative of compaction: the managements that perform little or no manipulation of the soil, such as pasture and no-tillage, densities between 1.41-1.45 Mg.m-3 (average 1.43 Mg m-3), and for the others, such as conventional plowing and minimum tillage, soil densities between 1.31-1.38 Mg.m-3 (average 1.35 Mg m-3). Finally, we conclude that compaction must be analyzed considering the texture and type of soil management.(AU)

Pre-implantation embryos release extracellular vesicles (EVs) to extracellular environment. In this work it is hypothesized that the EVs miRNA cargo will vary during pre-implantation development due to the constant changes in gene expression that take place through this period. The concentration, size and miRNA cargo of EVs secreted by competent bovine embryos during the period from compaction to blastulation (Day 3-7) were analyzed. For this analysis tow developmental windows were defined: W2 from 8-cells (D3) to morula (D5) and W3 from morula (D5) to blastocyst (D7). For W2, in vitro produced embryos were individually cultured in EVs-depleted medium from D3 to D5; culture media were collected and assigned to Group W2. Morulae were kept in culture up to blastocyst stage to determine the developmental competence. For W3, D5 morulae were collected and cultured individually in EVs-depleted medium up to blastocyst stage; culture media were assigned to Group W3, and blastocysts were kept in culture up to day 11 to define their competence. The mean size of EVs was similar between groups, however, EVs concentration was lower in W2. A total of 140 miRNAs were identified. From them, 79 were differentially expressed between the groups, 28 upregulated and 51 downregulated. miRNAs differentially detected between both developmental windows participate in the regulation of signaling pathways which crucial for embryonic development. It was concluded that the secretion of EVs is regulated by the developmental progress of the embryo during the pre-implantation period.

RESUMO

Livestock production in drylands requires consideration of the ecological applications of ecohydrological redistribution of water. Intensive cattle trampling and the associated increase of surface runoff are common concerns for rangeland productivity and sustainability. Here, we highlight a regional livestock production system in which cattle trails and trampling surrounding an artificial impoundment are purposely managed to enhance redistribution and availability of water for cattle drinking. Based on literature synthesis and field measurements, we first describe cattle production systems and surface water redistribution in the Dry Chaco rangelands of South America, and then develop a conceptual framework to synthesize the ecohydrological impacts of livestock production on these ecosystems. Critical to this framework is the pioshere-a degraded overgrazed and overtrampled area where vegetation has difficulties growing, usually close to the water points. The Dry Chaco rangelands have three key distinctive characteristics associated with the flat sedimentary environment lacking fresh groundwater and the very extensive ranching conditions: (1) cattle drinking water is provided by artificial impoundments filled by runoff, (2) heavy trampling around the impoundment and its adjacent areas generates a piosphere that favors runoff toward the impoundment, and (3) the impoundment, piosphere, and extensive forage areas are hydrologically connected with a network of cattle trails. We propose an ecohydrological framework where cattle transit and trampling alter the natural water circulation of these ecosystems, affecting small fractions of the landscape through increased runoff (compaction in piosphere and trails), surface connectivity (convergence of trails to piosphere to impoundment), and ponding (compaction of the impoundment floor) that operate together making water harvesting and storage possible. These effects have likely generated a positive water feedback on the expansion of livestock in the region with a relatively low impact on forage production. We highlight the role of livestock transit as a geomorphological agent capable of reshaping the hydrology of flat sedimentary rangelands in ways that can be managed positively for sustainable ranching systems. We suggest that the Dry Chaco offers an alternative paradigm for rangelands in which cattle trampling may contribute to sustainable seminatural production systems with implications for other dry and flat rangelands of the world.

Introduction: Despite the increase in knowledge of human morphogenesis, especially cardiogenesis and the processes by which the morphology of the ventricular myocardium is defined, little is known about the quantitative details in it. Objectives: To quantitatively describe the development of compact and non-compact ventricular myocardium and its correlation with cranio-rump length. Methods: descriptive, cross-sectional study in 18 human embryos from Carnegie stages 17 to 23, belonging to the Embryoteca of the Villa Clara University of Medical Sciences. The length of the embryo, the thickness of the compact, trabecular and total myocardium were measured in the lateral wall of both ventricles and the cardiac apex. Results: The thickness of the lateral wall of the compact myocardium increases in both ventricles from Carnegie stages 17 to 23, from 0.06 mm to 0.17 mm in the right and from 0.09 to 0.23 mm in the left ventricles. The thickness of the trabeculated lateral wall decreases with the advancement of the stages, from 0.43 mm to 0.34 mm in the right and from 0.45 mm to 0.37 mm in the left. The total lateral wall thickness increases from 0.48 mm to 0.51 mm in the right ventricle and from 0.52 mm to 0.62 mm in the left. The wall thickness of the compact vertex increases from 0.19 mm to 0.25 mm. Conclusions: Ventricular wall compaction increases with development; the cranio-spinal length is related to the thickness of the ventricular myocardium(AU)

RESUMO

Objectives: This study aimed to investigate the influence of different obturation techniques compared to cold lateral compaction on the success rate of primary non-surgical endodontic treatments. Materials and Methods: Systematic searches were performed for studies published up to May 17th, 2022 in MEDLINE/PubMed, Cochrane Library, Web of Science, Scopus, EMBASE, and Grey Literature Reports. Randomized clinical trials and nonrandomized (nonrandomized clinical trials, prospective or retrospective) studies that evaluated the success rate of primary non-surgical endodontic treatments obturated with the cold lateral compaction (control) and other obturation techniques were included. The revised Cochrane risk of bias tools for randomized trials (RoB 2) and nonrandomized studies of interventions (ROBINS-I) were used to evaluate the risk of bias. The Grading of Recommendations Assessment, Development, and Evaluation (GRADE) tool was used to evaluate the certainty of evidence. Results: Eleven studies (4 randomized clinical trials (RCTs), 4 prospective, and 3 retrospectives) were included. Two RCTs were classified as having some concerns risk of bias and 2 as a low risk of bias. Two nonrandomized studies were classified as having a critical risk of bias and 5 as having a moderate risk of bias. The GRADE analysis demonstrated a very low to moderate certainty of evidence. Conclusions: This systematic review generally evidenced no differences in the success rate of primary non-surgical endodontic treatments when the cold lateral compaction technique and other obturation techniques are performed. Further well-designed studies are still necessary.

RESUMO

Non-compaction cardiomyopathy (NCCM) is associated with neuromuscular disorders; however, there has been little investigation on its association with other neurological diseases, such as multiple sclerosis. We present the case of a 46-year-old woman with a history of multiple sclerosis who developed heart failure and was diagnosed with non-compaction cardiomyopathy.

RESUMO

The objective of the present study was to evaluate losses, production and polluting potential of the effluent, nutritional value and aerobic stability of silages of Brachiaria brizantha cv. Paiaguás grass, in different particle sizes and compaction density in silage. Three theoretical particle sizes (TTP 5; 8 and 12mm) and three compaction densities (DC 550; 600 and 650kg/m3) were evaluated, distributed in a factorial design (3 x 3), with four repetitions. The highest volume of effluent was found in silages with higher compaction densities (600 and 650kg/m3) and lower TTP (5 and 8mm). The highest chemical oxygen demand and biochemical oxygen demand were registered in the treatment with TTP of 5mm and higher DC (600 and 650kg/m3). Greater in vitro digestibility of DM was verified in the silage chopped at 5 and 8mm. There was no break in aerobic stability for 216 hours. Silage with a low compaction density 550kg/m3 and processing with a theoretical particle size of 12mm reduces effluent losses. In general, the nutritional value of Paiaguás grass was not influenced by the treatments. Different particle sizes and compaction density did not change the aerobic stability of silages.(AU)

Fractal and polarization analysis of diffusively scattered light is applied to determine the complex relationship between fractal dimension of structural morphology and concentration of chemically active ingredients in two pharmaceutical mixture systems including a series of binary mixtures of acetaminophen in lactose and three multicomponent blends with a proprietary active ingredient. A robust approach is proposed to identify and filter out multiple- and single-scattering components of scattering indicatrix. The fractal dimension extracted from scattering field reveals complex structural details of the sample, showing strong dependence on low-dose drug concentration in the blend. Low-angle diffraction shows optical "halo" patterns near the angle of specular reflection caused by light refraction in microcrystalline aggregates. Angular measurements of diffuse reflection demonstrate noticeable dependence of Brewster's angle on drug concentration. It is shown that the acetaminophen microcrystals produce scattered light depolarization due to their optical birefringence. The light scattering measurement protocol developed for diffusively scattered light by microcrystalline pharmaceutical compositions provides a novel approach for the pattern recognition, analysis and classification of materials with a low concentration of active chemical ingredients.

RESUMO

Our goal was to analyze the impact of a multipractice soil conservationist system on coffee production in tropical soils (LATOSSOLO VERMELHO-AMARELO caulinítico/kaolinitic Haplustox and LATOSSOLO VERMELHO-gibbsítico/gibbsitic Acrustox) in Minas Gerais, Brazil. In the experimental area, the soil management involved a set of multiple conservationist practices during cultivation for more than 3.5 yrs. Soil samples were collected at 0-5, 10-15, and 20-25 cm depths at two positions: planting rows and between rows. The soil physical and mechanical properties were determined, with a focus mainly on the preconsolidation stress model (preconsolidation stress versus matric tension). For the purpose of analysis, a split-split plot experimental design was implemented. A linear mixed regression model (LMR) was fitted, and Wald's test was performed (P < 0.05). An improvement of soil physical quality for both Oxisols was observed at the planting row position. The effects of multipractice soil management after 3.5 yrs of cultivation in association with increased gibbsite content could promote new soil structure organization, as revealed by LMR, resulting in more resilient soils (between rows) and increasing the resistance to external pressures on gibbsitic Acrustox. Overall, the findings herein are in concordance with global trends towards conservationist practices that may alleviate soil compaction in agricultural systems to maintain environmental sustainability.(AU)

Land use and tillage practices may change soil structure and undermine sustainable agriculture; however, such changes are hardly identified in the short term. In this sense, agroforestry systems have been used to reduce soil degradation and promote sustainable production in coffee plantations. These areas are expected to have well-structured soils and hence improved root distribution. This study aimed to evaluate soil quality by the morphostructural and root distribution analyses comparing open-grown coffee and coffee in agroforestry systems with rubber trees for 19 years, in an Oxisol in northern Paraná State (Brazil). Treatments consisted of open-grown coffee (OG), coffee partially shaded by rubber trees (PSH), and coffee fully shaded by rubber trees (FSH). The mapping of morphostructural features and soil resistance to penetration in cultural profile walls identified changes in soil structure resulting from different tillage systems. Root distribution was better in coffee plants grown in PSH and FSH systems. At greater depths, cultural profiles of FSH and PSH showed a larger numbers of roots compared to OG. Among the three systems, PSH provided a better environment for root growth and distribution. This result could be attributed to the high biological activity and interaction between roots and aggregates in that profile. The FSH agroforestry system provided less compact morphological structures and more roots throughout the soil profile. The agroforestry systems presented fewer soil structural changes by tillage operations and lower values of soil penetration resistance. Coffee root distribution was an effective indicator of soil quality and consistent with the morphostructural characterization of cultural profile.

RESUMO

This study determined the spatial variability of soil penetration resistance and yield of the soybean crop in lowland areas. The soil resistance to penetration at four different depths (0 to 0.10 m; 0.11 to 0.20 m; 0.21 to 0.30 m and 0.31 to 0.40 m), volumetric humidity of the soil at two depths (0 to 0.20 m and 0.21 to 0.40 m) and soybean yield were determined in an area of 1.13 hectares, using a sample mesh of 10 x 10 m. The corresponding data were subjected to descriptive statistical analysis. Pearsons simple linear correlation analysis (p0.05) was conducted and the spatial dependence was assessed by analyzing the isotropic semivariograms using spherical, exponential, linear, and Gaussian models. The results showed that the soil penetration resistance increased with depth, with restrictive values to root growth between 0.05 and 0.35 m. There was no correlation between yield and soil penetration resistance, and the semivariograms did not show a defined ascending region regarding the soil penetration resistance data. For the conditions under which the experiment was conducted, the sample 10 x 10 m mesh was suitable for assessing the spatial variability of soil resistance to penetration in depths exceeding 0.10 m.(AU)

Digestive disorders in cattle are associated with the breeding system and feed provided to the animals. Abomasal compaction is primarily related to the ingestion of forage with elevated levels of lignin, low quality, and difficult digestibility. In addition, the excess of fibrous food in the diet can lead to phytobezoars that may be responsible for intestinal obstruction disorders. This study aimed to describe pathological and clinical aspects of an outbreak of digestive disorders associated with the consumption of palm fiber (Elaeis guineensis). The outbreak struck a herd of 499 animals raised in a feedlot system after a change in diet that included an increase in the amount of palm fiber. Forty (8.01%) animals showed clinical signs such as fattening and regurgitation during rumination, and 21 (4.2%) animals died later. The cattle affected presented with apathy, emaciation, dehydration, distended abdomen, incomplete or absent ruminal movements, and congestive mucosa. Three animals were submitted to necropsy, and distended rumen and reticulum has a large amount of brownish liquid, long and tangled vegetable fibers with sand and stones. In two animals, the omasum had many rounded structures measuring approximately 5cm in diameter, made of vegetable fiber (phytobezoars). Abomasum of animals had similar material to the rumen, and one animal had compressed content. In two animals, dilatation was observed in the small intestine, and in the opening, the total obstruction of the lumen by phytobezoar was observed. During the follow-up of the slaughter of 76 cattle, 15 (19.7%) had phytobezoars of different sizes in the omasum and abomasum. The increased amount of oil palm fiber in animal feeding favored the occurrence of compression abomasum and intestinal obstruction phytobezoa, causing significant economic losses.(AU)

ABSTRACT: This study determined the spatial variability of soil penetration resistance and yield of the soybean crop in lowland areas. Pearson's simple linear correlation analysis (p≤0.05) was conducted and the spatial dependence was assessed by analyzing the isotropic semivariograms using spherical, exponential, linear, and Gaussian models. The results showed that the soil penetration resistance increased with depth, with restrictive values to root growth between 0.05 and 0.35 m. There was no correlation between yield and soil penetration resistance, and the semivariograms did not show a defined ascending region regarding the soil penetration resistance data. For the conditions under which the experiment was conducted, the sample 10 x 10 m mesh was suitable for assessing the spatial variability of soil resistance to penetration in depths exceeding 0.10 m.

In this paper a study on the process of densification of oil palm empty fruit bunches (OPEFB) is presented. An empirical-statistical model that allows the evaluation of densification process is obtained through an experimental factorial design. The main purpose of the experimental arrangement is to find the appropriate reference values for the experimental factors - moisture content, fiber length and compaction time-with which optimal performance responses of briquettes are achieved. Statistical models are obtained that explain in an acceptable way the influence of independent experimental factors on the mechanical properties of briquettes, such as briquettes density, durability index and compressive strength. It is possible to conclude that briquettes manufactured with the appropriate reference values, moisture content of 8% w.b., fiber length of 73.6 mm and a compaction time of 26.6 s respectively, meet mechanical and thermal requirements that are required in the most representative standards for biomass briquettes for energy purposes. Results obtained in the current investigation can be used as reference for the design of an industrial pilot plant destined to the densification of EFB of African oil palm.

ABSTRACT Introduction: Controversies still persist regarding the events of cardiovascular morphogenesis and an almost total absence of morphometric parameters in the initial phases of its development. Objectives: To determine the non-compacted to compacted (NC/C) myocardium ratio in both ventricles and the chronological progression of this ratio in the period studied. Method: A descriptive, cross-sectional study was carried out with 18 human embryos belonging to the Embryoteca of the Universidad de Ciencias Médicas de Villa Clara (Cuba) classified between Carnegie stages 17 and 23. The NC/C ratio -which is simply the mathematical calculation of the ratio between the non-compacted and compacted portions per specimen and per stage- was determined. Results: The application of this ratio in the right ventricle of the embryos obtained the following results: 7.17; 4.26; 3.12; 2.79; 2.36; 2.84 and 2.10 in Carnegie's stages 17, 18, 19, 20, 21, 22 and 23, respectively. In these same specimens, the left ventricle yielded the following results: 5.0; 3.80; 2.68; 2.18; 2.50; 2.01 and 1.56, also organized by stages. Conclusions: NC/C ratios obtained quantitatively support a progression of the ventricular myocardial compaction in the evaluated stages; their higher values at the apex denote that it may still be incomplete in this zone.

RESUMO

PURPOSE: To analyze the evolution post-cardiac resynchronization therapy (CRT) in left ventricular non-compaction (LVNC) cardiomyopathy (CM) patients compared to other types of CM, according to clinical and functional variables, by using gated-SPECT myocardial perfusion imaging (MPI). METHODS: Ninety-three patients (60 ± 11 years, 28% women) referred for pre-CRT assessment were studied and divided into three groups: 1 (non-ischemic CM with LVNC, 11 patients), 2 (ischemic CM, 28 patients), and 3 (non-ischemic CM, 53 patients). All were studied by a 99mTc-MIBI gated-SPECT MPI at rest pre-CRT implantation and 6 ± 1 months after, including intraventricular dyssynchrony assessment by phase analysis. Quality of life was measured by the Minnesota Living with Heart Failure Questionnaire (MLHFQ). RESULTS: No differences in sex, atherosclerotic risk factors other than smoking habit, and MLHFQ results were found among groups. LVNC CM patients were younger, with greater QRS width and lower left ventricular ejection fraction (LVEF) at baseline, but the differences were not significant. No significant differences were found at baseline regarding ventricular function, although end-systolic volume was slightly higher in LVNC CM patients. Mean SRS was significantly higher (p < 0.0001) in ischemic patients (14.9) versus non-ischemic ones (8.7 in group 1 and 9 in group 2). At baseline, LVNC CM patients were significantly more dyssynchronous: Their phase standard deviation (PSD) was higher (89.5° ± 14.2°) versus groups 2 (65.2° ± 23.3°) and 3 (69.7° ± 21.7°), p = 0.007. Although the quality of life significantly improved in all groups, non-ischemic patients (with or without LVNC) showed a higher LVEF increase and volumes reduction at 6 months post-CRT. Dyssynchrony reduced post-CRT in all groups. Nevertheless, those more dyssynchronous at baseline (LVNC CM) exhibited the most significant intraventricular synchronism improvement: PSD was reduced from 89.5° ± 14.2° at baseline to 63.7° ± 20.5° post-CRT (p = 0.028). Six months post-CRT, 89% of patients were responders: 11 (100%) of those with LVNC CM, 25 (86%) of those with ischemic CM, and 47 (89%) of patients with non-ischemic CM. No patient with LVNC CM had adverse events during the follow-up. CONCLUSION: CRT contributes to a marked improvement in non-ischemic CM patients with non-compaction myocardium. Phase analysis in gated-SPECT MPI is a valuable tool to assess the response to CRT.
