RESUMO

The proliferation of rearing systems providing opportunities for birds to engage in natural behaviors can trigger behavioral repertoires that when not manageable compromise animal welfare and the economic viability of the flock. Smothering in laying hens has long been perceived as "natural" or the result of hysteria among birds in the flock. However, the current literature has recognized smothering as an abnormal outcome with the potential to result in significant losses in cage-free poultry systems. Recent studies have specifically aimed to categorize the organization of smothering behavior and highlight its potential causes and consequences. In this study, literature review and bibliographic mapping, drawing on published articles and engagement with poultry farmers through extension and rural technical assistance, were employed. The findings indicate that smothering is a behavior triggered by factors related to the environment in which the laying hens are kept. This study concludes that there is a critical need for more rigorous and detailed research to elucidate the nuances of avian behavioral physiology and assess the impact of production systems on animal welfare and the economic impacts on the flock. This research contributes to a deeper understanding of bird behavior in high-production environments and provides practical insights for the poultry industry.

RESUMO

Introduction: Medical students are vulnerable to psychological distress and face a number of stressors such as academic pressures, economic concerns, and difficulties in interpersonal relationships. The quality of their educational training may also impact their mental health and subjective well-being. Objective: to describe their perception of subjective well-being and the impact of medical training on their own mental health. Methods: We surveyed 119 medical students and administered the "Perception survey on vocation, living and recreational habits, training and professional attitudes," the CAGE questionnaire, the Generalized Anxiety Disorder-7 (GAD-7) and the Patient Health Questionnaire-2 (PHQ-2). Results: Students' mean age was 22.5±2.28 years old and 59.7 % of participants were women, their lifestyle habits were scored as acceptable, and the employment of cell-phones and social networks ranged 76-99%; 24.1 % recognized a problematic alcohol, 67.2 % reported anxiety according to the GAD-7, and 49.6 % depression according to PHQ-2. Symptoms of anxiety were frequent among females and were higher when the perceived quality of training and student/teacher ratio were lower. Discussion: Students' perceptions of lower quality of training and resources seemed to be associated with higher levels of anxiety, which may suggest that educational policies and resources should be carefully revised and implemented to improve students' health and well-being.

BACKGROUND: Breast cancer is the most common malignant tumor disease and the leading cause of female mortality. The evolution of nanomaterials science opens the opportunity to improve traditional cancer therapies, enhancing therapy efficiency and reducing side effects. METHODS AND MAJOR RESULTS: Herein, protein cages conceived as enzymatic nanoreactors were designed and produced by using virus-like nanoparticles (VLPs) from Brome mosaic virus (BMV) and containing the catalytic activity of glucose oxidase (GOx) enzyme. The GOx enzyme was encapsulated into the BMV capsid (VLP-GOx), and the resulting enzymatic nanoreactors were coated with human serum albumin (VLP-GOx@HSA) for breast tumor cell targeting. The effect of the synthesized GOx nanoreactors on breast tumor cell lines was studied in vitro. Both nanoreactor preparations VLP-GOx and VLP-GOx@HSA showed to be highly cytotoxic for breast tumor cell cultures. Cytotoxicity for human embryonic kidney cells was also found. The monitoring of nanoreactor treatment on triple-negative breast cancer cells showed an evident production of oxygen by the catalase antioxidant enzyme induced by the high production of hydrogen peroxide from GOx activity. CONCLUSIONS AND IMPLICATIONS: The nanoreactors containing GOx activity are entirely suitable for cytotoxicity generation in tumor cells. The HSA functionalization of the VLP-GOx nanoreactors, a strategy designed for selective cancer targeting, showed no improvement in the cytotoxic effect. The GOx containing enzymatic nanoreactors seems to be an interesting alternative to improve the current cancer therapy. In vivo studies are ongoing to reinforce the effectiveness of this treatment strategy.

RESUMO

Due to the high density of chickens housed within the same space in aviaries, a large volume of organic matter accumulates on the floor. This large supply of available resources allows the development of arthropods, including organisms that affect poultry productivity and health. This study aimed to evaluate the composition of the mite community associated with organic matter in poultry houses with different rearing systems and the verification of potential predators to be evaluated in clean management techniques, such as biological control. The study was carried out in the state of Rio Grande do Sul (Brazil) in a total of six poultry houses, two of the 'Californian' system (SIG) and four of the 'cage free' system (SIF). In total, 839,459 specimens were collected, of which 358,044 were collected from SIF and 481,415 specimens from SIG. Fifty-two species/morphospecies were found and we observed a grouping of aviaries from the same rearing model. Species of medical veterinary importance were found, such as Dermanyssus gallinae, Megninia ginglymura, Ornithonyssus sylviarum and Tyrophagus putrescentiae. The results demonstrate that the organic matter present in the aviaries harbors a high diversity of mites of medical-veterinary importance and predators, which are influenced by the rearing system.

RESUMO

Glyphosate (GLP) is the herbicide with the highest level of global commercialization and historical use. Even though numerous studies have found this substance to be harmless, current research demonstrates that GLP might affect human health. For this reason, researcher efforts are concentrating on alternatives for analytical quantification, such as Surface Enhanced Raman Spectroscopy (SERS). In this work, a DVD-R@AgNPs SERS substrate was produced by the Cathodic Cage Plasma Deposition (CCPD) technique, which allowed a thin film layer deposition of silver nanoparticles (AgNPs) on the PC grating structure from Digital Video/Versatile Disc Recordable (DVD-R). Scanning Electron Microscopy with energy-dispersive X-ray spectroscopy was used to characterize the substrate and chemical changes on the surface after AgNPs deposition. The DVD-R@AgNPs substrate was used to detect standard crystal violet (CV), GLP, and RoundupTM GLP (GLP-RU) using Raman Spectroscopy. The CV was used as a control sample for SERS measurement, allowing the calculation of the substrate enhancement factor, which was in the order of âˆ¼ 105. To evaluate the efficiency of the SERS substrate, the limit of detection was calculated and showed values of âˆ¼ 10-10 mol/L for CV, 10-7 and 10-8 mol/L for GLP, and 10-6 mol/L for GLP-RU. Thus, the DVD-R@AgNPs SERS sensor is a low-cost substrate that analyzes traces of pesticides such as commercial GLP, demonstrating high SERS sensitivities and many applications.

RESUMO

Attractants are a powerful tool for pest management. The lack of specific attractants for the South American fruit fly, Anastrepha fraterculus, a complex of cryptic species of great economic importance in South America, makes it difficult to monitor the pest in the field. The γ-lactone male sex and aggregation pheromones of several Anastrepha species, naturally released in a 7:3 epianastrephin to anastrephin ratio, and a structurally related naturally occurring γ-lactone ((±)-trans-tetrahydroactinidiolide) with gem-dimethyl groups (dimethyl) at C(4), were evaluated as potential attractants of this species. Different age and mating conditions of A. fraterculus males and females were evaluated during electroantennography (EAG) and field cage experiments in which polymeric lures were deployed to contain 100 mg of attractant. Epianastrephin and dimethyl were EAG+ for all fly conditions, with epianastrephin eliciting the highest response for both sexes and immature flies showing greater responsiveness than mature flies. In the field cage experiments, immature flies were only attracted to leks; virgin females were attracted to leks, dimethyl, and both epianastrephin-anastrephin formulations (95 and 70 wt.% epianastrephin); mature-mated males were attracted to leks, dimethyl and 70 wt.% epianastrephin; and mature-mated females were only attracted to leks. Our bioassays showed a promising performance of the analog dimethyl since it elicited the same response as epianastrephin, requires fewer steps to synthesize, and contains one less chiral center than the natural pheromones. The attraction to leks was recorded for all mating conditions and ages of flies and suggests that air-borne volatiles of calling males contain cues that could act as sensory traps. The addition of any of these compounds in the synthetic attractants may result in a greater attraction and thus deserves further evaluation. Dose-response experiments will provide additional information to move a step forward and validate the results obtained in open-field conditions.

RESUMO

Anthropogenic pollution by trace metals in aquatic environments in semiarid zones is a critical area of investigation. The objective of this study was to investigate the concentration and spatial distribution of trace metals in surface sediments in the Rosário reservoir, which is affected by the intensive aquaculture of Tilápia-do-Nilo (Oreochromis niloticus). Sediment samples were collected in three different areas, postculture (PCTV), cultivation (CTV) and control (CTRL) in the dry season in 2019. The granulometric composition, organic matter and concentrations of Fe, Mn, Zn, Cu, Cr, Cd, Pb and Ni metals were determined. Multivariate statistics were used. Geochemical and ecotoxicological indices and a comparison with sediment quality guidelines (SQG) were used. The sediment was characterized by silty clay loam with an average organic matter of 18.76 ± 4.27. The analytical merit figures demonstrated accuracy (metal recoveries in certified standards) between 89 to 99% and high precision (RSD < 5%). The concentration ranges for the metals were Fe: 0.11-0.85 (%), Mn: 14.46-86.91, Zn: 2.6-220.56, Cu: 26.89-98.75, Cr: 60.18-76.06, Cd: 0.38-0.59, Pb: 18.13-43.13, and Ni: 34.4-46.75, all in (mg/kg-1). The highest concentration values were found in the CTV areas (Fe: 40 ± 0.22, Mn: 66.48 ± 19.11, Zn: 114.83 ± 59.75 and Cr: 70.85 ± 2.62) and PCTV (Cd: 0.53 ± 0.04, Cu: 71.83 ± 21.20, Pb: 33.71 ± 4.34 and Ni: 44.60 ± 1.79). Pearson's correlation, hierarchical cluster analysis and principal component analysis confirmed the influence of fish farming on metals. Only Ni presented concentration values higher than the reference value established in the SQG. Thus, considering the probable geochemical and ecotoxicological effects, they comprise the two lowest levels of impact.

RESUMO

The Salmonella enterica bacteriophage P22 is one of the most promising models for the development of virus-like particle (VLP) nanocages. It possesses an icosahedral T = 7 capsid, assembled by the combination of two structural proteins: the coat protein (gp5) and the scaffold protein (gp8). The P22 capsid has the remarkable capability of undergoing structural transition into three morphologies with differing diameters and wall-pore sizes. These varied morphologies can be explored for the design of nanoplatforms, such as for the development of cargo internalization strategies. The capsid proteic nature allows for the extensive modification of its structure, enabling the addition of non-native structures to alter the VLP properties or confer them to diverse ends. Various molecules were added to the P22 VLP through genetic, chemical, and other means to both the capsid and the scaffold protein, permitting the encapsulation or the presentation of cargo. This allows the particle to be exploited for numerous purposes-for example, as a nanocarrier, nanoreactor, and vaccine model, among other applications. Therefore, the present review intends to give an overview of the literature on this amazing particle.

RESUMO

In the rapid development of organic light-emitting diodes (OLEDs), phosphorescent transition metal complexes have played a crucial role as the most promising candidates for next generation display and lighting applications. However, most devices are fabricated using iridium and platinum-based complexes which are expensive and available in very limited quantities, whereas using relatively abundant organometallic complexes for fabrication results mostly in inefficient performance results. To overcome these issues, we have synthesized tetra copper iodide with tetra triphenyl cage like structure (denoted as CIPh) as an emerging class of luminescent material by mechanochemical grinding followed by thermal treatment for application in white OLED. The CIPh complex exhibits considerable quantum yield and a millisecond decay lifetime. Phosphorescent OLEDs were fabricated using CIPh complex as emitter shows a remarkable performance with external quantum efficiency and current efficiency of 5.28 % and 22.76 cd/A, with a high brightness of 4200 cd m-2, respectively. White OLEDs were also fabricated with a fluorescent blue and phosphorescent red emitted with (CIPh) as green emitter and achieved an impressive CRI of 82 with an EQE of over 3 %. This is the first ever attempt at fabricating WOLEDs using organocopper complex.

RESUMO

Low-cost adjuvants are urgently needed for the development of veterinary vaccines able to trigger strong immune responses. In this work, we describe a method to obtain a low-cost cage-like particles (ISCOMATRIX-like) adjuvant useful to formulate veterinary vaccines candidates. The main components to form the particles are lipids and saponins, which were obtained from egg yolk by ethanolic extraction and by dialyzing a non-refined saponins extract, respectively. Lipids were fully characterized by thin layer chromatography (TLC) and gas-chromatography (GC) and enzymatic methods, and saponins were characterized by TLC, HPLC and MALDI-TOF. Cage-like particles were prepared with these components or with commercial inputs. Both particles and the traditional Alum used in veterinary vaccines were compared by immunizing mice with Ovalbumin (OVA) formulated with these adjuvants and assessing IgG1, IgG2a anti OVA antibodies and specific Delayed-type Hypersensitivity (DTH). In the yolk extract, a mixture of phospholipids, cholesterol and minor components of the extract (e.g. lyso-phospholipids) with suitable proportions to generate cage-like particles was obtained. Also, semi-purified saponins with similar features to those of the QuilA® were obtained. Cage-like particles prepared with these components have 40-50 nm and triggers similar levels of Anti-OVA IgG1 and DTH than with commercial inputs but higher specific-IgG2a. Both adjuvants largely increased the levels of IgG1, IgG2a and DTH in relation to the formulation with Alum. The methods described to extract lipids from egg yolk and saponins from non-refined extract allowed us to obtain an inexpensive and highly effective adjuvant.

RESUMO

INTRODUCTION: Tuberculosis (TB) is an infectious, transmissible and immune disease caused by the Mycobacterium tuberculosis-complex (MTBC). Although osteoarticular tuberculosis (OATB) has been widely described, the ribcage variety remains a rare form. CASE REPORT: A thirteen-month-old male and a twenty-month-old female, both with pain and increased volume of anterolateral left rib cage were described. Physical examination revealed the presence of a soft consistent mass at the level of the 9th and 5th costal arches in the male and female patients respectively. Upon clinical evaluation, tuberculosis was suspected, which was confirmed by X-ray and histopathological studies. After confirmation, the management, based on anti-tuberculosis therapy was started as follows: nine months of anti-tuberculosis therapy for the male patient and fourteen months for the female. The outcomes were favorable for both patients. However, further interventions, consisting of abscess drainage in the male patient and excisional biopsy in the female patient were necessary. With these therapeutic interventions, to date, the patients are without any evidence of active TB.

RESUMO

The search for improvements in the poultry production chain and for increased egg productivity have grown significantly. However, intensive production with or by using of confinement systems negatively affect the welfare of animals and is favoring the appearance of harmful ectoparasites in production. Due to differences in the existing management systems, (e.g. Californian and cage-free systems) and given the importance of the poultry industry in the international market, the aim of the present study was: (1) to compare mite community between SIG (Intensive system - cage: Californian) and SIF (Intensive system - free: cage free); (2) egg-laying systems to analyze how the abundances of the ectoparasites Dermanyssus gallinae, Megninia ginlgymura, and Ornithonyssus sylviarum are affected by these egg-laying models; (3) to evaluate the association between the functional diversity found in traps, organic matter, and feathers in both systems; and finally, (4) to analyze how abiotic factors (e.g. humidity, temperature at sampling) affect mite community composition inside each model. A total of 860,571 mites were sampled, belonging to 59 taxa from six functional groups. Mite composition in the SIF system was different from the SIG system, while their variations in dispersion homogeneity were quite similar to each other. Although taxonomic composition is different between them, they are quite similar when clustered according to functional diversity. The variables 'humidity' and 'temperature' were not determining factors for the appearance of species. The species that most contributed for distinguishing the models were: M. ginlgymura, Dermatophagoides farinae, D. gallinae, Tyrophagus putrescentiae, O. sylviarum, Blattisocius keegani, Glycyphagus sp., Acotyledon sp., and Cheyletus malaccensis, respectively. Dermanyssus gallinae was strongly associated with the Californian model. Two taxa were highly correlated with cloaca, neck, dorsum, and inner wings; variations in relative air humidity also affected M. ginlgymura and O. sylviarum populations. Organic matter had a high abundance of predatory mites with potential for controlling mites of sanitary importance in the poultry chain.

ABSTRACT Defective management of secretions is one of the most frequent complications in invasive mechanically ventilated patients. Clearance of secretions through chest physiotherapy is a critical aspect of the treatment of these patients. Manual rib cage compression is one of the most practiced chest physiotherapy techniques in ventilated patients; however, its impact on clinical outcomes remains controversial due to methodological issues and poor understanding of its action. In this review, we present a detailed analysis of the physical principles involved in rib cage compression technique performance, as well as the physiological effects observed in experimental and clinical studies, which show that the use of brief and vigorous rib cage compression, based on increased expiratory flows (expiratory-inspiratory airflow difference of > 33L/minute), can improve mucus movement toward the glottis. On the other hand, the use of soft and gradual rib cage compression throughout the whole expiratory phase does not impact the expiratory flows, resulting in ineffective or undesired effects in some cases. More physiological studies are needed to understand the principles of the rib cage compression technique in ventilated humans. However, according to the evidence, rib cage compression has more potential benefits than risks, so its implementation should be promoted.

RESUMO

Palm kernel cake (PKC) is an abundant side stream that can only be added to non-ruminant feed in small concentrations due to its content of antinutritional factors, mainly galactomannan, which cannot be digested by non-ruminants. ß-mannanases can be added to partially hydrolyze galactomannan to form mannose oligosaccharides, which are known to be prebiotic. We here investigate the action of a ß-mannanase from B. subtilis on PKC by colorimetry, NMR and fluorescence microscopy. The amount of mannan oligosaccharides in solution was significantly increased by the ß-mannanase and their degree of polymerization (DP) was significantly reduced. There was a dose-response behavior in that larger ß-mannanase concentrations increased the amount of soluble mannose oligosaccharides while reducing their average DP. Using confocal immunofluorescence microscopy, solubilization of galactomannan in PKC was clearly visualized. Images show a clear disruption of the cellulose and galactomannan structures of the PKC cell walls. We thus show in this study that using commercial dosages of ß-mannanase on PKC can lead to formation of prebiotic compounds. Thus, this study suggests that utilization of PKC in poultry feed formulation might be increased by addition of a ß-mannanase and would improve the return on investment.

RESUMO

Anterior cage migration is the most infrequent and dangerous complication seen in posterior lumbar interbody fusion (PLIF) procedures. We report the case of a 74-year-old woman who underwent PLIF at the L5-S1 level. During the surgery, one of the PLIF-cages dislodged anteriorly into the abdominal cavity without vascular injury. An anterior retroperitoneal approach to remove the cage and complete the fusion was made. The patient was discharged 2 weeks later with encouraging clinical results. In a patient hemodynamically stable, removing the cage by a vascular surgeon, and complete the Anterior Lumbar Interbody Fusion could be a feasible option at L5-S1.

Background: Genetics influence the vulnerability to alcohol use disorders, and among the implicated genes, three previous studies have provided evidences for the involvement of LRRK2 in alcohol dependence (AD). LRRK2 expression is broadly dysregulated in postmortem brain from AD humans, as well as in the brain of mice with alcohol dependent-like behaviors and in a zebrafish model of alcohol preference. The aim of the present study was to evaluate the association of variants in the LRRK2 gene with AD in multiethnic populations from South and North America. Methods: Alcohol-screening questionnaires [such as CAGE and Alcohol Use Disorders Identification Test (AUDIT)] were used to determine individual risk of AD. Multivariate logistic regression analyses were done in three independent populations (898 individuals from Bambuí, Brazil; 3,015 individuals from Pelotas, Brazil; and 1,316 from the United States). Linkage disequilibrium and conditional analyses, as well as in silico functional analyses, were also conducted. Results: Four LRRK2 variants were significantly associated with AD in our discovery cohort (Bambuí): rs4768231, rs4767971, rs7307310, and rs1465527. Two of these variants (rs4768231 and rs4767971) were replicated in both Pelotas and US cohorts. The consistent association signal (at the LRRK2 locus) found in populations with different genetic backgrounds reinforces the relevance of our findings. Conclusion: Taken together, these results support the notion that genetic variants in the LRRK2 locus are risk factors for AD in humans.

RESUMO

Tyrophagus putrescentiae (Schrank) (Acaridae) is one of the most allergenic mite species worldwide, and it particularly infests household environments, animal feed factories, and food factories. It has been the aim of several studies recently in the healthcare area due to its allergenic potential. However, there had been no records of this species attacking laying birds until this report. This is the first record of adverse reactions in animals triggered by infestations of this species. Therefore, the present study aims at describing the damages and symptomatology of T. putrescentiae infestations on commercial laying hens. Alopecia and dermatitis were symptoms exacerbated by T. putrescentiae infestation starting at 40 weeks old. The presence of this species on hens, as observed in the present study, might be directly related to the accumulation of food and poultry litter on these birds, in their cages, or in the vicinities. Hence, the results might be related to their feeding behavior, which consists of continuous supply of animal feed in these management systems. The present study provides an unprecedented record that reveals the importance and potential for emergency pest status of T. putrescentiae in the poultry industry. Its infestations in poultry houses probably required constant sanitary care and monitoring are required in order to prevent outbreaks of this mite.

Piscirickettsia salmonis, the etiological agent of the Salmon Rickettsial Septicemia (SRS), is one the most serious health problems for the Chilean salmon industry. Typical antimicrobial strategies used against P. salmonis include antibiotics and vaccines, but these applications have largely failed. A few years ago, the first attenuated-live vaccine against SRS (ALPHA JECT LiVac® SRS vaccine) was released to the market. However, there is no data about the agents involved in the activation of the immune response induced under field conditions. Therefore, in this study we evaluated the expression profile of a set of gene markers related to innate and adaptive immunity in the context of a cellular response in Atlantic salmon (Salmo salar) reared under productive farm conditions and immunized with a live-attenuated vaccine against P. salmonis. We analyzed the expression at zero, 5-, 15- and 45-days post-vaccination (dpv). Our results reveal that the administration of the attenuated live SRS LiVac vaccine induces a short-term upregulation of the cellular-mediated immune response at 5 dpv modulated by the upregulation of ifnα, ifnγ, and the cd4 and cd8α T cell surface markers. In addition, we also registered the upregulation of il-10 and tgfß. Altogether, the results suggest that a balanced activation of the immune response took place only at early times post-vaccination (5 dpv). The scope of this short-term upregulation of the cellular-mediated immune response against a natural outbreak in fish subjected to productive farm conditions deserves further research.

RESUMO

STUDY DESIGN: Systematic review. OBJECTIVES: The objective of this study was to compare clinical and radiological outcomes following discectomy and anterior cervical fusion for the treatment of cervical degenerative disorder performed with stand-alone cages and anterior cervical plates. METHODS: Electronic searches were performed in the MEDLINE, LILACS, and Cochrane Systematic Reviews databases, according to PRISMA guidelines, with no language or date restriction. The review was registered in PROSPERO under number CRD42018109180. RESULTS: Six randomized clinical trials were selected, which evaluated at least one of the objectives of this work, such as pain control, bone consolidation, neurological symptoms, and cervical lordosis, thus satisfying the inclusion criteria. Articles that did not directly compare the 2 surgical techniques were excluded. A total of 309 patients were included and the results showed no significant difference in clinical (visual analogue scale and neck disability index) or radiological (cervical lordosis and fusion) outcome between the 2 groups. The operative time was shorter in the group with stand-alone cages (mean difference = -18.40; 95% CI = [-24.89, -11.92]; P < .66). CONCLUSION: The stand-alone cages and anterior cervical plate techniques have similar clinical and radiological outcomes. Despite the significantly shorter operative time for one group, other randomized clinical trials are needed to establish conclusive evidence in favor of one of the comparative treatments.
