Despite its crucial location, the western side of Amazonia between the Andes and the source(s) of the Amazon River is still understudied from a genomic and archaeogenomic point of view, albeit possibly harboring essential information to clarify the complex genetic history of local Indigenous groups and their interactions with nearby regions,1,2,3,4,5,6,7,8 including central America and the Caribbean.9,10,11,12 Focusing on this key region, we analyzed the genome-wide profiles of 51 Ashaninka individuals from Amazonian Peru, observing an unexpected extent of genomic variation. We identified at least two Ashaninka subgroups with distinctive genomic makeups, which were differentially shaped by the degree and timing of external admixtures, especially with the Indigenous groups from the Andes and the Pacific coast. On a continental scale, Ashaninka ancestors probably derived from a south-north migration of Indigenous groups moving into the Amazonian rainforest from a southeastern area with contributions from the Southern Cone and the Atlantic coast. These ancestral populations diversified in the variegated geographic regions of interior South America, on the eastern side of the Andes, differentially interacting with surrounding coastal groups. In this complex scenario, we also revealed strict connections between the ancestors of present-day Ashaninka, who belong to the Arawakan language family,13 and those Indigenous groups that moved further north into the Caribbean, contributing to the early Ceramic (Saladoid) tradition in the islands.14,15.

The Upper Rio Negro regional social system is made up of more than 30 languages belonging to six linguistic families. This results from socio-historical processes stretching back at least two millennia, which have built a system with different levels of autonomy and hierarchy associated with a mythical and ritual complex, and with social and linguistic exchanges. The analysis of these processes require an interdisciplinary outlook to understand the ways in which people from different linguistic families interacted and created it. More specifically, we ask how linguistic and cultural diversity have been created in the context of intense relations of multilingualism and inter-ethnic contact. To this end, we integrate perspectives from historical linguistics (regarding languages from the Tukanoan, Arawakan and Naduhup families) with archaeological data from the Amazonian past. Through this multidisciplinary approach, we seek to develop a linguistic-anthropological understanding of the dynamics shaping the region's diversity and inter-ethnic relations. We show that processes creating diversity are interrelated with changes in social histories, and are especially tied to the establishment of new forms of social organization as a result of pre-colonial inter-ethnic relations. This has led to the construction of various local multilingual ecologies connected to macro-regional processes in Amazonia.

Moving back in time from the early colonial to the late pre-colonial period we evaluate the hypothesis asserting the migratory movement of Cariban-speaking groups from the Middle Orinoco River area towards north-central Venezuela. The explanation in vogue maintains that the migration followed fluvial routes and occurred between 1350 and 1150 BP (AD 600-800). We examine archaeological, linguistic, ethnohistorical, genetic, and ecological data seeking similarities between the Orinoco emigrants and their north-central Venezuelan descendants. As a result, we propose an alternative terrestrial/fluvial route and suggest these events occurred between 1150 and 1050 BP (AD 800-900). The route first proceeded upstream along rivers of the central llanos and later followed a natural terrestrial geomorphological corridor into the Lake Valencia Basin. We argue that, while future interdisciplinary (especially archaeo-linguistic and bioarchaeological) research is needed to further assess the results of these analyses, the Orinocan descendants in north-central Venezuela emerge as one of the most dynamic sociopolitical Cariban-speaking entities in all northeastern South America and the insular Caribbean on the eve of the European Conquest.
