RESUMO

Xylitol was biotechnologically produced by Kluyveromyces marxianus ATCC36907 using the hemicellulosic hydrolysate of the cashew apple bagasse (CABHH). Sequentially, the present study investigated the recovery and purification of xylitol evaluating different antisolvents [ethanol, isopropanol and the ionic liquid 2-hydroxyl-ethylammonium acetate (2-HEAA)], their proportion in the medium (10-90% v/v), and their cooling rate (VC 0.25-0.50 °C/min). These processes were contrasted with the crystallization process of commercial xylitol. This study is the first to assess xylitol crystallization using a protic ionic liquid. The hydrolysate obtained from a mild treatment with sulfuric acid contained mainly glucose and xylose at concentrations of 15.7 g/L and 11.9 g/L, respectively. With this bioprocess, a maximum xylitol production of 4.5 g/L was achieved. The performance of the investigated antisolvents was similar in all conditions evaluated in the crystallization process of the commercial xylitol, with no significant difference in yields. For the crystallization processes of the produced xylitol, the best conditions were: 50% (v/v) isopropanol as antisolvent, cooling rate of 0.5 °C/min, with a secondary nucleation of yield and purity of 69.7% and 84.8%, respectively. Under the same linear cooling rate, using ethanol, isopropanol or the protic ionic liquid 2-hydroxyl-ethylammonium acetate (2-HEAA), crystallization did not occur, probably due to the presence of carbohydrates not metabolized by the yeast in the broth, which influences the solubility curve of xylitol. With the results of this work, a possible economical and environmentally friendly process of recovery and purification of xylitol from CABHH could be proposed.

Solid-state fermentation (SSF) is used in enzyme and antibiotic production, bioethanol and biodiesel as an alternative energy source, biosurfactants with environmental goals, and the production of organic acids and bioactive compounds. The present project determined the quantity of secondary metabolites and the antioxidant activity of the extracts obtained by the solid-state fermentation of apple and agave mezcalero bagasse over 28 days, inoculated with the Pleurotus ostreatus strain. The extraction was carried out with three solvents: acetone and water (80:20 v/v), 100% methanol and 100% water. The results showed a higher presence of phenolic compounds, flavonoids, total triterpenes and antioxidant activity in the apple bagasse from the SSF on day 21 in the extract of acetone and water (80:20 v/v), 100% methanol and aqueous; while the agave bagasse showed a significant presence of phenolic compounds and flavonoids only in the aqueous extract. In conclusion, the presence of secondary metabolites exhibiting antioxidant activities from the solid-state fermentation in the residues of the cider and mezcal industry is an alternative use for wasted raw material, plus, it reduces the pollution generated from the agroindustrial residues.

AIMS: A culture medium based on apple bagasse was designed and tested as a substrate for biomass production of conventional and unconventional native wine yeasts. METHODS AND RESULTS: The physicochemical characterization of the apple bagasse was carried out and its potential utility as a constituent of a complete culture medium for the production of yeast biomass was analysed using the experimental statistical designs. Growth parameters of conventional and nonconventional Patagonian wine yeasts were analysed with Placket-Burman designs and response surface methodology, comparing in each assay the apple bagasse substrate with the commonly used substrate for biomass development, cane molasses. Culture media composition was optimized and models were validated. CONCLUSIONS: This study demonstrates that, both from a nutritional and from an economic point of view, apple bagasse constitutes a more advantageous substrate than cane molasses for the propagation of native yeasts from Patagonia. SIGNIFICANCE AND IMPACT OF THE STUDY: We used an alternate carbon-rich material, generously available in our region, originally generated as fruit industrial waste, to transform it into a source of sustainable, economically profitable and environmentally friendly energy resource.

To enhance the enzymatic digestibility of cashew apple bagasse (CAB) feedstock in order to produce sugar fermentation-derived bioproducts, the CAB was subjected to three different pretreatments with the ionic liquid 2-hydroxyl-ethylammonium acetate (2-HEAA) and characterized by FTIR, NMR and chemical methods. All conditions were able to delignify CAB, however the best lignin removal (95.8%) was achieved through the method performed with 8.7% w/w of CAB/2-HEAA ratio at 130°C for 24h. Although the cellulose crystallinity has been increased in CAB treated with the ionic liquid, but this fact did not influence its digestibility. Nevertheless, the pretreatment with 2-HEAA enhanced significantly the cellulose digestibility, increasing the glucose yield from 48 to 747.72mgglucose/gCAB. Furthermore, 2-HEAA pretreatment was efficient even with reused ionic liquid, obtaining high glucose concentration.

Abstract Solid-state fermentation can be used to produce feeds for ruminants, which can provide an enriched population of yeasts to improve ruminal fermentation. Fermentation of apple bagasse was performed to obtain a yeast-rich product, with the objective of isolating, identifying, and characterizing yeast strains and testing their capability to enhance in vitro ruminal fermentation of fibrous feeds. Yeasts were isolated from apple bagasse fermented under in vitro conditions, using rumen liquor obtained from cannulated cows and alfalfa as a fibrous substrate. A total of 16 new yeast strains were isolated and identified by biochemical and molecular methods. The strains were designated Levazot, followed by the isolate number. Their fermentative capacity was assessed using an in vitro gas production method. Strain Levazot 15 (Candida norvegensis) showed the greatest increase in gas production (p < 0.05) compared with the yeast-free control and positively affected in vitro ruminal fermentation parameters of alfalfa and oat straw. Based on these results, it was concluded that the Levazot 15 yeast strain could be potentially used as an additive for ruminants consuming high-fiber diets. However, further studies of effects of these additives on rumen digestion, metabolism, and productive performance of ruminants are required.

Ethanol production from acidic-alkaline pretreated cashew apple bagasse (CAB-OH) was investigated using separated hydrolysis and fermentation (SHF) and simultaneous saccharification and fermentation (SSF) processes. First, a screening of Kluyveromyces strains was conducted by SHF and a maximum ethanol concentration of 24.1 g L(-1) was obtained using Kluyveromyces marxianus ATCC36907, which presented similar profiles when compared to results obtained by a Saccharomyces strain. The effect of temperature on ethanol production conducted by SHF using K. marxianus ATCC36907 was investigated, and the maximum ethanol yield (YE/G) was obtained at 40 °C (0.46 g g(-1)) using a synthetic medium. In the SHF using CAB-OH hydrolysate, the maximum ethanol concentration obtained was 24.9 g L(-1), 5.92 g L(-1) h(-1) of productivity, and ethanol yield of 0.43 g g(-1) at 40 °C. Afterwards, K. marxianus ATCC36907 was used in the bioconversion of CAB-OH by SSF, and an ethanol concentration of 41.41 ± 0.2 g L(-1) was obtained using 10 % CAB-OH at 40 °C, 150 rpm and 24 h, resulting in a Y'E/G of 0.50 gE gG (-1) and an efficiency of 98.4 %, in the process conducted with cellobiase supplementation. SHF and SSF processes using CAB-OH and K. marxianus ATCC36907 can be used to ethanol production, but the SSF process required only one step to achieve the same production.

Avaliou-se o efeito da adição de bagaço de caju desidratado (BCD), durante o pós-parto, em 41 ovelhas, alojadas com suas crias em baias, onde recebiam, à vontade, 75 por cento de capim-elefante + 25 por cento de concentrado (DI; n=17), ou 50 por cento de BCD + 25 por cento de capim-elefante + 25 por cento de concentrado (DII; n=24). Cinquenta dias pós-parto, o estro foi sincronizado, e as ovelhas submetidas à monta natural. O grupo DI perdeu mais peso (P<0,05), e o consumo de proteína bruta e de fibra em detergente neutro foi maior nesse grupo (P<0,001). A inclusão de BCD na dieta II induziu ao aumento do consumo de matéria seca, extrato etéreo e fibra em detergente ácido (P<0,001) e redução significativa das concentrações de colesterol, lipídios totais, albumina e proteína total (P<0,001). Não houve diferença entre os grupos (P>0,05) no tempo de retorno do primeiro corpo lúteo funcional, na resposta à sincronização do estro e nas taxas de gestação e prolificidade. A inclusão de 50 por cento de BCD amenizou as perdas de peso, foi bem consumida e não influenciou nas respostas reprodutivas após a sincronização do estro.


The effect of the addition of dehydrated cashew apple bagasse (DCB) to the diet on ewe postpartum response was evaluated on 41 ewes, housed with their kids in pens, where they received two diets containing 75 percent of Elephant Grass plus 25 percent of concentrate (Diet I; n=17), or 50 percent of DCB plus 25 percent of Elephant Grass plus 25 percent of concentrate (Diet II; n=24). On the 50th day postpartum, estrus was synchronized and ewes mated. Diet Igroup exhibited a greater loss of live weight (P<0.05) and higher intakes of crude protein and neutral detergent fiber (P<0.001). By contrast, animals from diet II showed a significantly increase in dry matter, ether extract, and acid detergent fiber intakes (P<0.001), and lower plasmatic concentrations of cholesterol, total lipids, albumin, and total protein (P<0.001). No differences were found between groups (P>0.05) for the presence of first functionally CL, estrus synchronization response, and gestation or prolificity rates. Thus, the addition of DCB to the diet was well accepted, induced a reduction of weight loss, and did not affect the reproductive response of ewes.
