Aeromonas hydrophila is a cause of infectious disease outbreaks in carp species cultured in South Asian countries including Pakistan. This bacterium has gained resistance to a wide range of antibiotics and robust preventive measures are necessary to control its spread. No prior use of fish vaccines has been reported in Pakistan. The present study aims to develop and evaluate inactivated vaccines against local strain of A. hydrophila in Pakistan with alum-precipitate as adjuvant. The immunogenic potential of vaccine was evaluated in two Indian major carps (Rohu: Labeo rohita, Mori: Cirrhinus mrigala) and a Chinese carp (Grass carp: Ctenopharyngodon idella). Fish were vaccinated intraperitoneally followed by a challenge through immersion. Fish with an average age of 4-5 months were randomly distributed in three vaccinated groups with three vaccine concentrations of 108, 109 and 1010 colony forming unit (CFU)/ml and a control group. Fixed dose of 0.1ml was applied to each fish on 1st day and a booster dose at 15 days post-vaccination (DPV). Blood samples were collected on 14, 28, 35, 48 and 60 DPV to determine antibody titers in blood serum using compliment fixation test (CFT). Fish were challenged at 60 DPV with infectious A. hydrophila with 108 CFU/ml through immersion. Significantly higher levels of antibody titers were observed from 28 DPV in all vaccinated groups as compared to those in the control group. In challenge experiment the average RPS (relative percent survivability) was 71% for groups vaccinated with 109 and 1010 CFU/ml and 86% for 108 CFU/ml. Vaccine with 108 CFU/ml induced highest immune response followed by 109 and 1010 CFU/ml. The immune response of L. rohita and C. idella was better than that of C. mrigala. ABSTRACT The study aimed to explore the effect of dietary supplementation of synbiotics on growth performance, carcass characteristics and nutrient digestibility in broiler chicken. For this purpose, three hundred 1-day-old Cobb-500 broiler chicks were purchased from the hatchery and randomly distributed into five dietary treatment groups. Each treatment had six replicates, each containing 10 chicks. The experimental diets were supplemented with 0, 700, 1200, 1700 or 2200 g/ton of feed synbiotics and respectively designated as A, B, C, D and E, with A being the control diet. Feed intake, weight gain and feed conversion ratio data were recorded on a weekly basis. At the research trials end, two birds from each pen were randomly selected and slaughtered to get data on carcass characteristics. Results showed that group Cs feed intake was reduced (p 0.05) when compared to control. Body weight was similar (p>0.05) among all treatments. However, feed conversion ratio was significantly improved (p 0.05) in group C as compared to other dietary treatments. Nutrient digestibility was improved (p 0.05) in group B and C, as compared to control. Carcass characteristics were not significantly affected and remained the same across all treatments. However, liver weight decreased in birds fed diet C. Meat quality and antibody titer were not affected in any of the dietary treatments. It is concluded that synbiotics can be safely used up to 1200 g/ton of feed, improving bird performance without harmful effects on bird health.

The aim of this study was to assess the effects of vegetable oils on the antioxidant capacity, blood lipid components and immune responses in chickens raised in hot climate. In a completely randomized design, 300 chicks were assigned to four treatments and five replicates with 15 chicks per each. The highest concentration of malondialdehyde was found in broilers fed palm oil (p < 0.05). Chickens fed corn oil had the lowest and those fed flaxseed oil had the highest antioxidant enzymes activities (p < 0.05). The highest hepatic Alanine-transferase and Aspartate-transferase was found in chickens fed corn oil and the lowest levels in those received flaxseed and olive oils (p < 0.05). The highest hypersensitivity was in chickens fed palm oil and the lowest was for olive oil. The highest antibody titer against sheep red blood cells was found in chickens fed flaxseed oil and the lowest titer was for those received palm oil (p < 0.05). The highest triglyceride, cholesterol and very low density lipoprotein and the lowest high density lipoprotein was found in chickens fed palm oil and vice versa for flaxseed and olive oils. Olive and flaxseed oils had beneficial effects on antioxidant activity and lipid attributes and palm oil had detrimental effects on these parameters that related to the poultry health.

BACKGROUND: The epidemiology of the pandemic A(H1N1) virus has been changing as population immunity continues to co-evolve with the virus. The impact of genetic changes in the virus on human's susceptibility is an outstanding important question in vaccine design. In a community-based study, we aim to (1) determine the genetic characteristics of 2009-2015 pandemic H1N1 viruses, (2) assess antibody response following natural infections and (3) assess the correlation of A/California/07/09 antibody titers to protection in the 2013 and 2015 epidemics. METHODS: In a household transmission study, serum specimens from 253 individuals in Managua, Nicaragua were analyzed. Combined nose and throat swabs were collected to detect RT-PCR confirmed influenza infection and virus sequencing. Hemagglutination inhibition assays were performed and the protective titer for circulating H1N1pdm was determined. RESULTS: Clade 6B pandemic H1N1 viruses predominated in Nicaragua during the 2013 and 2015 seasons. Our household transmission study detected a household secondary attack rate of 17% in 2013 and 33% in 2015. Infected individuals, including vaccinees, showed an apparent antibody response to A/California/07/09. Baseline titers of A/California/07/09 antibodies were found to associate with protection in both seasons. A titer of ≥1:40 correlated to a 44% protection in children, a 29% protection in adults 15-49years old and a 51% protection in adults 50-85years old. CONCLUSION: In 2013 and 2015, antibody titers to A/California/07/09 associated with an infection risk reduction amongst exposed household contacts. This is consistent with a detectable vaccine effectiveness reported in a number of studies. Genetic changes in clade 6B viruses might have led to a reduced immunity in some whereas others might have been less affected. The use of human serologic data is important in virus characterization and if performed in a timely manner, could assist in vaccine strain selection.

BACKGROUND: Major ABO mismatch between donor and recipient in bone marrow transplantation (BMT) may cause hemolysis, delayed red blood cell (RBC) engraftment and pure red cell aplasia (PRCA), which result in increased transfusion needs. High pretransplant anti-A/B antibody titers have been associated with increased risk of PRCA. Herein, we studied the impact of anti-A/B titers on transfusion needs after BMT with major ABO mismatch. METHODS: We reviewed the medical charts of 27 patients who underwent to BMT with major ABO mismatch and categorized them into two groups according to anti-A/B titers of IgG (≤16 and ≥32). We recorded the number of RBC and platelet units transfused in the first 180 days after transplantation. We also evaluated the impact of anti-A/B titers on overall survival. RESULTS: Patients with anti-A/B titer ≥32 of IgG class required more RBC transfusion than patients with titer ≤16 (6.60±4.55 vs 21.29±14.68; P=.03). Anti-A/B of IgM class had no impact on both RBC and platelet transfusion needs. Anti-A/B titers had no impact on overall survival. CONCLUSION: Higher titers of anti-A/B antibodies of IgG class, but not of IgM, are associated with a higher demand for RBC transfusion.

In order to investigate the immune enhancement effects of Ophiopogon japonicus polysaccharide Ophiopogon japonicus (OJPS) on Newcastle disease (ND) live vaccine, chickens vaccinated against ND live vaccine was orally administered with the OJPS at high, medium and low concentrations respectively. In negative control group, chickens were given orally equal volume of physiological saline. On day 14, 21 and 28, the serum antibody titer, erythrocyte-C3b receptor rosette rate (E-C3bRR), erythrocyte-C3b immune complex rosette rate (E-ICRR) and peripheral lymphocyte proliferation were measured. The results showed that at most time points, the antibody titer, peripheral lymphocyte proliferation, E-C3bRR and elimination rate of immune complex of three OJPS administrating groups were significantly higher (P < 0.05) than those in negative control group. It indicated that OJPS could significantly improve the immune efficacy of Newcastle disease live vaccine, Ophiopogon japonicus polysaccharide possessed synergistical immunoenhancement.(AU)

Com o objetivo de avaliar a utilização do extrato de orégano nas dietas de codornas japonesas (Coturnix coturnix japonica) e desafiadas com cepas de Escherichia coli, sobre as características de desempenho, a incidência de celulite aviária e titulação de anticorpos específicos contra antígenos de E. coli, foram utilizadas 360 codornas japonesas, com 90 dias de idade, distribuídas em gaiolas de arame galvanizado em galpão convencional. O delineamento experimental foi inteiramente casualizado, em esquema fatorial 5x2 (extrato de orégano x desafiado ou não com E. oli), totalizando dez tratamentos com seis repetições de seis aves por gaiola. Os níveis do extrato de orégano (EO) avaliados foram: 0,00; 0,025; 0,050; 0,100 e 0,150%. Foram avaliadas características de desempenho produtivo, lesões macroscópicas da celulite após períodos pós-inoculação das cepas e amostras de soro foram colhidas para verificar a titulação de anticorpos nas aves. Os dados obtidos foram submetidos à análise de variância e as médias comparadas pelo Teste T. Foi observado efeito de E. coli sobre todas as características produtivas, independentemente dos níveis de EO avaliados, onde grupos desafiados apresentaram piores resultados de desempenho. As lesões macroscópicas, características da celulite, observadas somente nas aves desafiadas com E. coli foram classificadas como grau leve e sem presença de hemorragias. Para a titulação de anticorpos específicos, houve maior quantificação para aves desafiadas com as cepas de E. coli em relação às não desafiadas. Pode-se concluir que o extrato de orégano suplementado nas rações não se mostrou eficaz frente ao desafio com E. coli em codornas na fase de postura e as aves desafiadas com E. coli apresentaram maiores respostas imunes humoral e celular, em relação às não desafiadas, caracterizadas pelo aumento na titulação de anticorpos e pela lesão macroscópica peitoral, independentemente dos níveis de extrato de orégano avaliados.(AU)

It can be concluded that oregano extract supplemented in the diet was not effective against the challenge with E. coli in laying quails, and challenged birds with E. coli showed higher humoral and cellular immune response, compared with unchallenged birds, characterized by increased antibody titer and pectoral macroscopic lesion, regardless of the oregano extract levels evaluated.(AU)

Background: Newcastle disease (ND), caused by avian paramyxovirus serotype 1 (APMV-1), also known as Newcastle disease virus (NDV), is regarded as one of the two most devastating diseases of poultry with the characteristics of serious disease and high flock mortality. It causes severe economic losses in domestic poultry, especially in chickens. At present, there were no effective drugs to treat this disease and the main method to control ND was vaccination. Moreover, new strains of virus resistant to chemotherapy continue to emerge, so does the need for a safe and effective vaccine. The immune adjuvant can make vaccine generate a strong immune response providing long-term protection against infection. With commonly usage of some immune adjuvants (e.g. mineral oil and aluminium hydroxide), many problems were occurred, such as side effects, strong local stimulation and carcinogenesis, together with complicated preparation, or failure to increase immunogenicity of weak antigen and so on. Botanical polysaccharides, as a new type of adjuvant or immunopotentiator, had become the hot research area because of their less side effects and no toxicity. The purpose of this research was to observe whether Chuanminshen violaceum polysaccharide (CVPS) possessed synergistical immunoenhancement, and offer the theoretical evidence for developing potential new-type adjuvant. Materials, Methods & Results: 200 three-yellow chickens at one day of age were randomly divided into four groups and reared in separated pens. On 7 days old, the average maternal serum hemaglutination inhibition (HI) antibody titer was less than 3 log2, all chickens of each group (the average body weight (BW) was 120 g) were vaccinated with ND live vaccine through nasal drip and eye-drop. At the same time, the chickens in three CVPS groups (high, medium and low doses of CVPS) were orally administered with 0.5 mL of CVPS at concentrations of 100 mg/kg BW, 50 mg/kg BW, 25 mg/kg BW respectively, once a day for fi ve successive day, in negative control group (NC), with 0.5 mL of physiological saline, once a day for fi ve successive day. On days 14, 21 and 28, the serum antibody titer, erythrocyte-C3b receptor rosette rate (E-C3bRR), erythrocyte-C3b immune complex rosette rate (E-ICRR), peripheral lymphocyte proliferation and peripheral CD4+/CD8+ ratio were measured. The results showed that the antibody titer, E-C3bRR, elimination rate of immune complex and peripheral lymphocyte proliferation in three CVPS groups and peripheral CD4+/CD8+ ratio in medium dosage of CVPS group were significantly higher (P < 0.05) than those in control group throughout the process of whole experiment almost. Discussion: The CVPS not only improved the E-C3bRR and accelerated the elimination rate of CIC, but also induced higher antibody titer, peripheral lymphocyte proliferation and peripheral CD4+ /CD8+ ratio in chickens vaccinated against ND live vaccine. This indicated that CVPS possessed immune-enhancement efficacy of ND live vaccine and might be expected as a candidate of new-type adjuvant.

Marigold flower extract, a natural pigment, was used to determine its effect on carcass and skin pigmentation, immunity and growth performance of broiler chickens. Two hundred and forty 1-day-old Arbor Acres broiler chicks were randomly distributed into four treatment groups with six replicates in a randomized block design. Birds were fed basal diet for 42 d with or without supplementation of marigold flower extract at various concentrations, i.e., 0 (MG0, control), 100 (MG100), 150 (MG150) and 200 (MG200) mg/kg of feed, respectively. Feed intake and live body weight were weekly recorded. Carcass and shank color, and antibody titers against Newcastle and Influenza viruses were measured. Results showed that marigold flower extract significantly (p 0.05) improved live body weight and relative thymus weight. However, feed intake, feed conversion ratio (FCR), and spleen and bursa weights were not significantly affected. Yellowness (b*) of breast and thigh muscles increased by the dietary supplementation of marigold flower extract compared with the control diet. However, lightness (L*), redness (a*) and redness to yellowness ratio (a/b) were not influenced by the treatments. Moreover, Roche color fan scores of the shank skin were increased at market age (d 42). The results revealed that marigold extract enhanced antibody titers against Newcastle and influenza viruses. It was possible to conclude that the dietary supplementation with marigold flower extract at the rate of 200 mg/kg of feed enhanced carcass and shank color, antibody titers against ND and AI, and growth performance of broiler chickens.

Neospora caninum has been described as an important cause of abortion in bovine worldwide. The objective of the present study was to characterize patterns of antibody dynamics during gestation in dairy cows naturally infected with N. caninum. Twelve N. caninum naturally infected cows were selected from four dairy herds from Brazil and blood samples were monthly collected during pregnancy. Serum were tested for antibodies against N. caninum by Indirect Fluorescent Antibody Test (IFAT). During this period, all cows remained clinically normal and gave birth to healthy calves. The cows remained seropositives during the study and N. caninum IFAT titers ranged from 100 to 12,800; only animal 234 presented one negative result in the first month of pregnancy. Significant differences of N. caninum IFAT titers were found between months from 1 to 9 of pregnancy bythe Friedman Test (P<0.001). The statistical analysis showed an increase of N. caninum antibody titers from second and third trimester of pregnancy in relation to first trimester. High titers were observed in few cows after month fifth of pregnancy. This study showed a variation of specific antibody levels in seropositive cows during different gestational periods. The highest values were observed during the second and third trimester. The antibody increase after the fifth month of gestation was not associated to abortion.(AU)


Neospora caninum é descrito como uma importante causa de abortamento em bovinos por todo o mundo. O objetivo do presente estudo foi caracterizar o padrão da dinâmica de anticorpos durante a gestação em vacas leiteiras infectadas naturalmente por N. caninum. Doze vacas gestantes infectadas naturalmente com N.caninum foram selecionadas de quatro rebanhos leiteiros do Brasil e amostras de sangue foram mensalmente colhidas da concepção até o parto das vacas. Soros foram testados para anticorpos contra N. caninum pela reação de imunofluorescência indireta (RIFI). Durante a gestação, todas as vacas permaneceram clinicamente normais e geraram bezerros saudáveis. As vacas permaneceram soropositivas durante o estudo e títulos de anticorpos anti-N.caninum variaram de 100 a 12.800; somente o animal 234 apresentou um resultado negativo no primeiro mês de gestação. Diferenças significativas dos títulos da RIFI para N. caninum foram encontradas entre os meses de 1 a 9 de gestação pelo Teste de Friedman (P<0,001). As análises estatísticas mostraram um aumento dos títulos de anticorpos anti-N. caninum no segundo e terceiro trimestre de gestação em relação ao primeiro trimestre. Altos títulos de anticorpos foram observados em algumas vacas após o mês cinco de gestação. Este estudo mostrou variação dos níveis de anticorpos em vacas soropositivas durante diferentes períodos gestacionais. Altos títulos foram observados durante o segundo e terceiro trimestre e o aumento dos títulos de anticorpos após o quinto mês de gestação não foi associado a abortamentos.(AU)
