RESUMO

AIM: To analyze the influencing factors of postoperative dry eye complication after corneal transplantation and to build a nomogram prediction model.METHODS: Clinical data were collected on 117 patients who underwent corneal transplantation at our hospital from March 2021 to February 2023. They were divided into dry eye group(n=96)and non-dry eye group(n=21)according to whether there was a postoperative dry eye. The risk factors of postoperative complication of dry eye after corneal transplantation were analyzed, the nomogram prediction model for predicting postoperative complication of dry eye after corneal transplantation was constructed, and the internal validation of the model and the prediction efficacy were assessed by calibration curves and decision curves, respectively.RESULTS: Comorbid diabetes mellitus, comorbid sleep disorders, comorbid meibomian gland dysfunction, chronic eye drop abuse, chronic corneal contact lens wear, interleukin-6(IL-6), and tumor necrosis factor-α(TNF-α)were the risk factors for the complication of dry eye after corneal transplantation(P<0.05). The nomogram model predicted a C-index of 0.890(95% CI 0.877-0.903). The nomogram model had a threshold >0.07, and the nomogram model provided higher net clinical benefit than the single index in all cases.CONCLUSION: The nomogram model built in this study based on the factors affecting the complication of dry eye after corneal transplantation has a good predictive value for the complication of dry eye after corneal transplantation.

Graphene quantum dots (GQDs), which have high photostability, anti-photobleaching and scintillation, good biocompatibility and low toxicity, are important member of the fluorescent material family, and have attracted extensive research interest. In this paper, a fluorescence resonance energy transfer (FRET) biosensor based on protoporphyrin IX (PpIX) and GQDs was developed for melamine detection. PpIX was bound to the surface of GQDs to produce self-assembled nanosensors, and a FRET process occurred between GQDs and PpIX. However, due to the combination of melamine and PpIX, the FRET process was shut down in the presence of melamine. The FRET system could quickly and accurately detect melamine with a detection range of 1.0 × 10-8 to 2.0 × 10-6 mol/L based on the fluorescence intensity ratio of PpIX and GQDs, and the detection limit was 3.6 × 10-9 mol/L. This method obtained satisfactory results when it was employed to the determination of melamine in milk samples.

Objective:To explore the effect of Yifei-Huoxue Decoction combined with conventional western medicine on patients with oxidant-antioxidant imbalance with COPD. Methods:A total of 102 patients who met the inclusion criteria from January 2016 to December 2018 were divided into two groups, according to the random table method, 51 cases in each group. The control group was treated with conventional western medicine, while the research group was treated with Yifei-Huoxue Decoction based on control group. Both the two groups were treated for 14 days. The traditional Chinese medicine symptom scores of the two groups were compared before and after the treatment. The forced expiratory volume in one second/predicted value ratio (FEV1%pred) was measured by pulmonary function instrument, FEV1/forced vital capacity (FVC) was calculated. Partial pressure of oxygen (PaO 2) and partial pressure of carbon dioxide (PaCO 2) were measured by blood gas analyzer. The levels of MDA, SOD and Glutathioneperoxidase (GSH-Px) were detected by ELISA, and the clinical effect was evaluated. Results:The total effective rate of research group was 98.0% (50/51), the control group was 80.4% (41/51), and the difference between the two groups was statistically significant ( χ2=8.254, P=0.004). After the treatment, the scores of cough, expectoration, asthma, panic, chest distress and fatigue in the research group were significantly lower than those of the control group ( t values were 3.716, 6.524, 5.032, 2.516, 4.804 and 2.882, respectively, all Ps<0.05). After the treatment, the FEV1%pred (68.05% ± 6.11% vs. 59.02% ± 5.45%, t=7.876), FEV1/FVC (60.10 ± 5.83 vs. 54.05 ± 5.07, t=5.592), PaO 2 (67.79 ± 3.82 mmHg vs. 60.63 ± 3.55 mmHg, t=9.805) in the research group were significantly higher than those of the control group ( P<0.05), the PaCO 2 (56.08 ± 2.25 mmHg vs. 61.14 ± 2.13 mmHg, t=11.663) was significantly lower than that of the control group ( P<0.01), the levels of serum MDA (3.21 ± 0.63 μmol/L vs. 4.95 ± 0.87 μmol/L, t=11.568) was significantly lower than that of the control group ( P<0.01), the levels of SOD (73.35 ± 8.97 μU/ml vs. 61.68 ± 7.82 μU/ml, t=7.003), GSH-Px (170.35 ± 17.72 U/ml vs. 149.14 ± 15.04 U/ml, t=6.517) were significantly higher than those of the control group ( P<0.01). Conclusions:The Yifei-Huoxue Decoction combined with conventional western medicine could improve the lung function of children with COPD , adjust the imbalance condition of oxidation/antioxidation, so as to improve the clinical effect.

Objective:To observe the efficacy of tedrogesterone tablets combined with clomiphene on polycystic ovary syndrome(PCOS) with infertility, and investigate its influence on serum levels of apolipoprotein B/apolipoprotein A1 ratio(ApoB/ApoA1), anti-mullerian hormone(AMH), advanced glycation end products(AGEs).Methods:From January 2017 to December 2018, 80 PCOS patients with infertility who met the criteria were screened and randomly divided into control group and observation group according to the digital table, with 40 cases in each group.The control group received clomiphene 50 mg/d, qd, and for 5 consecutive days.In the observation group, on the basis of the control group, tedrosterone tablet was added, 10 mg/time and qd, and continuously administered for 21 days.The two groups were treated for 4 menstrual cycles.The ovarian volume, follicular development, pregnancy rate, hormone levels, the endometrial receptivity, and serum ApoB/ApoA1, AMH, AGEs levels were compared between the two groups.Results:After treatment, the ovarian volume[(5.62±0.87)cm 3]of the observation group was significantly smaller than that of the control group[(7.94±1.03)cm 3]( t=7.722, P=0.000), and the maximum diameter of follicles in the observation group[(13.30±2.04)mm] was significantly larger than that in the control group[(11.12±1.83)mm]( t=5.530, P=0.0001). The pregnancy rate in the observation group was significantly higher than that in the control group(χ 2=4.990, P=0.025). The serum levels of follicle stimulating hormone(FSH)[(4.80±0.56)IU/L], luteinizing hormone(LH)[(7.35±0.11)IU/L], uterine pulse index(PI)(2.77±0.41), and uterine artery resistance index(RI)(0.61±0.09) in the observation group were significantly lower than those in the control group( t=5.003, P=0.000; t=6.449, P=0.000; t=5.304, P=0.000; t=5.008, P=0.000), and the estradiol(E 2)[(190.51±23.11)ng/L] and endometrial thickness[(10.11±1.57)mm] were significantly higher than those in the control group( t=38.521, P=0.000; t=6.001, P=0.000). The serum levels of ApoB/ApoA1(0.62±0.08), AMH[(4.13±0.61)μg/L], and AGEs[(118.04±13.35)ng/L] in the observation group were significantly lower than those in the control group( t=5.553, P=0.000; t=5.790, P=0.000; t=18.043, P=0.000). Conclusion:The efficacy of tedrogesterone tablets combined with clomiphene on PCOS with infertility is significant, and can down-regulate serum levels of ApoB/ApoA1, AMH, and AGEs.

Objective To investigate the effect of single antiepileptic drugs(AEDs) and combination therapy on the effect of seizure after stroke,prognosis and recurrent seizures.Methods From November 2013 to December 2017,280 patients with epilepsy and epilepsy that had been included in the epilepsy course and AEDs course for >6 months in Shaoxing Central Hospital were divided into early-onset epilepsy group and late-onset epilepsy group according to the patients' condition,with 140 cases in each group.According to different AEDs treatment regimens,they were further divided into monotherapy group(valproate) and combination therapy group(valproate,oxcarbazepine combined with levetiracetam).Results The proportion of monotherapy in the early-onset epilepsy group was significantly higher than that of the late-onset epilepsy group,and the proportion of the late-onset epilepsy group was significantly higher than that of the early-onset epilepsy group(x2 =22.857,P < 0.0001).The incidence and effectiveness of epilepsy patients after single-agent and combination therapy in the early-onset epilepsy group had statistically significant differences [onset rate:40.0% (36/90) vs.16.0% (8/50),x2 =8.591,P =0.003;effective rate:88.9%(80/90) vs.100.0% (50/50),x2 =5.983,P =0.014].The incidence and effectiveness of single-agent and combination therapy in the late-onset epilepsy group had statistically significant differences [seizure rate:40.0%(20/50) vs.82.2% (74/90),x2 =25.974,P =0.000;effective rate:64.0% (32/50) vs.87.8 % (79/90),x2 =1 1.065,P =0.000].Conclusion The time of post-stroke epilepsy is not related with the site,type and risk factors of stroke;early epilepsy usually manifested partial seizures and had better curative effect of AEDs,and late epilepsy usually manifested overall tonic clonic seizure;there were no obvious difference in curative effect and recurrence rate of early and late epilepsy on single and combined AEDs treatment.

Cotton is one of the most economically important crops, but most cultivated varieties lack adequate innate immunity or resistance to Verticillium wilt. This results in serious losses to both yield and fiber quality. To identify the genetic resources for innate immunity and understand the pathways for pathogen defenses in this crop, here we focus on orthologs of the central Arabidopsis thaliana defense regulator Enhanced Disease Susceptibility 1 (EDS1). The full-length cDNA of GbEDS1 was obtained by screening the full-length cDNA library of Gossypium barbadense combining with RACE strategy. Its open reading frame is 1848 bp long, encoding 615 amino acid residues. Sequence analysis showed that GbEDS1 contains a conserved N-terminal lipase domain and an EDS1-specific KNEDT motif. Expression profiling indicated that the gene is induced by Verticillium dahliae as well as salicylic acid (SA) treatment. Subcellular localization assays revealed that GbEDS1 is located in the cell cytoplasm and nucleus. Overexpression of GbEDS1 in Arabidopsis dramatically up-regulated SA and H2O2 production, resulting in enhanced disease resistance to V. dahliae. Silencing of GbEDS1 in G. barbadense significantly decreased SA and H2O2 accumulation, leading to the cotton more susceptibility. Moreover, combining the gene expression results from transgenic Arabidopsis and silenced-GbEDS1 cotton, it indicated that GbEDS1 could activate GbNDR1 and GbBAK1 expression. These findings not only broaden our knowledge about the biological role of GbEDS1, but also provide new insights into the defense mechanisms of GbEDS1 against V. dahliae in cotton.

AIM:To investigate the effect of cigarette smoking condensate on histone deacetylase 2 (HDAC2) and inflammatory mediators in mouse myoblast C 2C12 cells.METHODS:C2C12 cells were treated with cigarette smoke extract (CSE).HDAC2 siRNA was transfected into the cells using Lipofectamine TM 2000.The levels of interleukin-8 (IL-8) and tumor necrosis factor-α( TNF-α) in the culture supernatants were measured by ELISA , and the expression of HDAC2 at mRNA and protein levels was determined by real-time PCR and Western blotting .RESULTS:The expression of HDAC2 at mRNA and protein levels in CSE group was lower than that in control group (P<0.05).The supernatant levels of IL-8 and TNF-αin CSE group were significantly higher than those in control group ( P<0.05 ) .When the cells were transfected with HDAC2 siRNA followed by CSE stimulation , the expression of HDAC2 at mRNA and protein levels was de-creased , and the supernatant levels of IL-8 and TNF-αwere significantly increased as compared with CSE group and control group (P<0.05).CONCLUSION: Under the oxidative stress condition , C2C12 cells generate high levels of IL-8 and TNF-αby down-regulating the expression of HDAC2.

RESUMO

By using a self-developed IF power and a ASTM contact material experimental system of small-capacity and variable frequency, the value of arcing characteristics and the welding force of the silver-based contact material are acquired under low voltage, resistive load and small current at 400 Hz and 50 Hz. By means of an electricity-ray analytical balance, SEM and EDAX, the weighing values of the contact materials and the changes of AgCdO, AgNi, AgC and AgW contact material surface profile and micro-area constituent are obtained and analyzed. The arc erosion causes of silver-based alloy contact materials at 400 Hz and 50 Hz are also discussed.
