RESUMO

Chromosome studies were done in 8 males and 18 females who were accidentally exposed for 4 work days (i.e., 8 hr/day) to vapors of ortho dichlorobenzene. The clinical symptoms in 10 individuals included headache, general malaise, dizziness, and nausea. All persons had variable degrees of mucosal irritations. Of the 1345 cells studied, 120 disclosed chromosomal aberrations (mean = 8.92%), whereas a control group of 11 healthy individuals revealed 19 cells with aberrations in 942 cells examined (mean = 2.02%). The main chromosomal alterations were 84 single breaks (6.25%) and 86 double breaks (6.39%). In the control group there were 2 single breaks (0.92%) and 10 double breaks (1.06%), with significant statistic values of P less than .001 for the exposed group. Other chromosomal aberrations were poliploidy and ring formation but these were not statistically significant. Chromosome studies conducted 6 months later in 15 persons of the exposed group disclosed a significant reduction of chromosomal aberrations, but these were still present as compared with the control group. Although definite chromosomal changes occurred, these alterations seemed to be reversible after several months, at least with an original 4-day exposure to the clastogen chemical.

RESUMO

Este estudio se diseno para conocer la frecuenccia de aparicion del cromosoma Philadelphia y otras anomalias cromosomicas em 74 pacientes adultos (40 mujeres y 34 varones) con leucemia granulocitica cronica diagnosticada en los Servicios de Hematologia del Hospital de Oncologia y del Hospital General del Centro Medico Nacional del Instituto Mexicano del Seguro Social. Todos los pacientes, excepto uno, mostraron cromosoma Philadelphia. En 20 pacientes habia, ademas, otras anormalidades cromosomicas tales como aneuploidias y poliploidias, duplicacion del cromosoma Philadelphia, deleciones y traslocaciones, todos estos marcadores en forma aislada o en combinaciones. Llama la atención que solamente en un caso no hubo cromosoma Philadelphia (1.36 por ciento), lo que contrasta con otras series de la literatura extranjera, en las que se senala 15 por ciento de ausencia de este marcador, lo que en general se acompana de peor pronosticos. No se encontro correlacion entre las anormalidades cromosomicas y el tiempo de supervivencia, en contraste con lo que ocurre en otros tipos de leucemia. La baja frecuencia de aparicion de cromosoma Philadelphia sugiere que existen factores ambientales diferentes a los que ocurren en otros paises, o bien que el diagnostico de leucemia se basa casi exclusivamente en el hallazgo de este marcador cromosomico

A study was carried out with the purpose of establishing the frequency of female sex chromatin (Barr corpuscle) and male sex chromatin or ("F") fluorescent corpuscles in the Purkinje cerebellar neurones, that are tetraploid cells. Two Barr corpuscles were observed in 18 per cent of Purkinje cells in hematoxylin-eosin stained histological sections in five females and none in a similar number of male sex individuals. In the cerebellar smears stained according to Klinger's method, Barr corpuscles were observed in Purkinje cells in 30 per cent of females different to what was observed in male sex individuals. Smears stained with quinacrine dihydrochloride showed two "F" corpuscles in Purkinje cells of male individuals and only one fluorescent corpuscle in a lower percentage of glial cells and of the granule cell layer in this same material. "F" corpuscles were not observed in females. This study shows that in tetraploidy, as the case of Purkinje neurones, an X gonosome is expressed for each set of chromosomes in female individuals and an "F" corpuscle, corresponding to the Y gonosome of each chromosomic set is found in male sex individuals.
