Indirect somatic embryogenesis (ISE) is a morphogenetic pathway in which somatic cells form callus and, later, somatic embryos (SE). 2,4-dichlorophenoxyacetic acid (2,4-D) is a synthetic auxin that promotes the proliferation and dedifferentiation of somatic cells, inducing the ISE. However, 2,4-D can cause genetic, epigenetic, physiological and morphological disorders, preventing the regeneration and/or resulting abnormal somatic embryos (ASE). We aimed to evaluate the toxic 2,4-D effect during the Coffea arabica and C. canephora ISE, assessing the SE morphology, global 5-methylcytosine levels (5-mC%) and DNA damage. Leaf explants were inoculated in media with different 2,4-D concentrations. After 90 days, the friable calli were transferred to the regeneration medium, and the number of normal and abnormal SE was monthly counted. The increase of the 2,4-D concentration increased the number of responsive explants in both Coffea. At 9.06, 18.08 and 36.24 µM 2,4-D, C. arabica presented the highest values of responsive explants, differing from C. canephora. Normal and abnormal SE regeneration increased in relation to the time and 2,4-D concentration. Global 5-mC% varied at different stages of the ISE in both Coffea. Furthermore, the 2,4-D concentration positively correlated with global 5-mC%, and with the mean number of ASE. All ASE of C. arabica and C. canephora exhibited DNA damage and showed higher global 5-mC%. The allotetraploid C. arabica exhibited greater tolerance to the toxic effect of 2,4-D than the diploid C. canephora. We conclude that synthetic 2,4-D auxin promotes genotoxic and phytotoxic disorders and promotes epigenetic changes during Coffea ISE.

The spermatic membrane is rich in polyunsaturated fatty acids, which makes it sensitive to the action of reative species of oxygen, which can damage the seminal quality of the scraps. The purpose of this study was to evaluate the effect of the supplementation of two selenium sources at different doses. Third five scraps were allocated in four groups: (INOR30) 0.30 ppm sodium selenite; (COMP30) 0.30 ppm selenium metal-amino acid; (MIXED15+15) 0.15 ppm sodium selenite + 0.15 ppm selenium metal-amino acid and (COMP15) 0.15 ppm selenium metal-amino acid. The ejaculates of the scraps were evaluated over 22 weeks, resulting in 210 samples evaluated for volume, motility, pH, presence of agglutination and morphological changes, and 140 samples for spermatic concentration. The data was analyzed with repeated measures in time in a mixed model with type of selenium supplementation, periods of evaluation (one period of two weeks + five periods of four weeks) and their interactions as fixed effects, and animal and the worker that collected the ejaculates as random effects. Results showed no difference in selenium supplementation with the sources and doses used. In this way, it was possible to verify that the metal amino acid of selenium at the dose of 0.15 ppm promotes the same effect as the diets formulated with 0.30 ppm of sodium selenite.(AU)
A membrana espermática é rica em ácidos graxos poliinsaturados, o que a torna sensível à ação de espécies reativas de oxigênio, que podem prejudicar a qualidade seminal dos cachaços. O objetivo do presente estudo foi avaliar o efeito da suplementação de duas fontes de selênio em diferentes doses. Trinta e cinco cachaços foram distribuídos em quatro grupos: (INOR30) 0,30 ppm de selenito de sódio; (COMP30) 0,30 ppm de metal-aminoácido de selênio; (MISTO15+15) 0,15 ppm de selenito de sódio + 0,15 ppm de metal-aminoácido de selênio e (COMP15) 0,15 ppm de metal-aminoácido de selênio. Os ejaculados dos cachaços foram avaliados durante 22 semanas, resultando em 210 amostras avaliadas para volume, motilidade, pH, presença de aglutinação e alterações morfológicas, e 140 amostras para concentração espermática. Os dados foram analisados com medidas repetidas no tempo em modelo misto, em que o tipo de suplementação de selênio, os períodos de avaliação (um período de duas semanas + cinco períodos de quatro semanas) e suas interações foram os efeitos fixos, e o animal e o funcionário que coletou os ejaculados foram os efeitos aleatórios. Os resultados obtidos demonstraram não haver diferença na suplementação de selênio com as fontes e doses utilizadas. Com isso, foi possível verificar que o metal-aminoácido de selênio na dose de 0,15 ppm promove o mesmo efeito das dietas formuladas com 0,30 ppm de selenito de sódio.(AU)

Some forest trees have been polyploidized to improve their traits and to supply new germplasms for breeding programs. As trees have a long juvenile stage, the early characterization of the chromosome set doubling effects is crucial for previous selection. Thus, we aimed to characterize the chemical variability of essential oils from diploid and autotetraploid germplasms (autotetraploid A and B) of Eucalyptus benthamii, as well as to evaluate their larvicidal and allelopathic effects. Autotetraploid A showed a higher essential oil yield than diploid and autotetraploid B, which did not differ quantitatively. Aromadendrene, viridiflorol and α-pinene were the major compounds in the diploid essential oil. In contrast, compounds were present in autotetraploids, such as 1,8-cineole, limonene, α-terpineol, and α-terpinyl-acetate. Essential oils from the diploid at 50-200 ppm were twice as larvicidal than those from autotetraploids against Aedes aegypti larvae. Considering the phytotoxicity bioassays using Lactuca sativa, essential oils from both ploidy levels affected root growth. Moreover, the essential oils inhibited shoot growth at all concentrations tested (187.5; 375; 750; 1500; and 3000 ppm). Autotetraploid A and B had the same effect on shoot growth as glyphosate. The essential oils had no cytogenotoxic effect on root meristematic cells of L. sativa, whereas phytotoxic potential was identified mainly in shoot growth. This work demonstrated a dramatic change in secondary metabolism (terpene composition) related to an increase in the ploidy level in Eucalyptus germplasms. In addition, we report the novelty of the chemical composition of essential oils among germplasms and their potential use as larvicidal and post-emergence weed control agents.

O objetivo deste trabalho foi avaliar os efeitos de manejo de uniformização, ordem de nascimento (ON), peso ao nascimento (PN) e suas interações no desempenho de leitões na fase de maternidade. Porcas (n = 25) e suas leitegadas (n = 388 leitões) foram distribuídas em dois tratamentos: UNIF - uniformização de leitões entre leitegadas distintas logo após o nascimento; MBIO - leitões mantidos até 12 horas pós-parto com a mãe biológica. Nos dois tratamentos o efeito da ON foi avaliado em três grupos caracterizados pela ON de 1-6, 7-12 e ≥ 13; o efeito de PN foi avaliado em quatro grupos definidos como Muito leve, Leve, Médio e Pesado. O delineamento foi inteiramente casualizado em esquema fatorial 2 × 3 × 4, totalizando 24 tratamentos com 16 leitões/tratamento, em média. O ganho de peso do nascimento até 72 horas de vida (GP72h) foi maior em MBIO. A quantidade de colostro ingerida (QCI) foi menor em leitões de ON ≥ 13. Em geral, o desempenho dos leitões aumentou em resposta ao aumento do PN. Houve interação entre manejo de uniformização e PN para QCI em relação ao peso corporal (QCIPC), que foi maior em leitões Muito leves em UNIF (27,48% PC) e menor em leitões Pesados em MBIO (16,82% PC). É possível obter bom desempenho de leitões mantendo a leitegada com a mãe biológica até 12 horas pós-parto. A quantidade de colostro ingerida em base absoluta (g) é maior, mas relativa ao peso corporal (% PC) é menor em leitões pesados.
The goal of this study was to assess the effects of uniformization management, birth order (BO), birth weight (BiW) and their interactions on the performance of piglets in the maternity phase. Sows (n = 25) and their litters (n = 388 piglets) were distributed into two treatments, namely: UNIF - uniformization of piglets between different litters right after birth; and BIOM - piglets kept up to 12 hours after birth with their biological mothers. In both treatments, the effect of BO was assessed in three groups characterised by BO of 1-6, 7-12, and ≥13. The effect of BiW was assessed in four groups defined as very light, light, medium, and heavy. The design was completely randomised in a 2×3×4 factorial scheme, totalling 24 treatments with 16 piglets/treatment, on average. Weight gain from birth to 72 hours after birth (WG72h) was greater in the BIOM treatment. The colostrum intake (CI) was lower in piglets with BO ≥13. In general, piglet performance increased in response to the increase in BiW. There was interaction between uniformization management and BiW with respect to CI related to body weight (CIBW), which was greater in very light piglets in the UNIF treatment (27.48% BW), and lower in heavy piglets in the BIOM treatment (16.82% BW). It was possible to obtain satisfactory piglet performance by keeping the litters with their biological mothers until 12 hours after birth. The CI expressed on an absolute basis (g) was greater; however, CI expressed on relative basis with respect to body weight (% BW) was lower in heavy piglets.

Painting plant chromosomes through chromosomal in situ suppression (CISS) hybridization has long been considered impracticable. Seeking to build specific and complex probes from a single microdissected chromosome, we employed human chromosomes as models to standardize all the necessary steps for application in plants. Human metaphases were used to define the adequate conditions for microdissection, chromosome DNA amplification and labeling through degenerate oligonucleotide-primed PCR, and in situ hybridization stringency. Subsequently, these methodologies were applied in the plant species Zea mays (chromosome 1) and Capsicum annuum (chromosome 7 or 8). The high quality of human and plant cytogenetic preparations and the meticulous standardization of each step, especially the most critical ones - microdissection and first round of DNA amplification - were crucial to eliminate the signs of non-specific hybridization and for direct application in plants. By overcoming these challenges, we obtained chromosome-specific probes, which allowed to achieve a clear and uniform painting of the entire target chromosomes with little or no background, evidencing their complexity and specificity. Despite the high amount of ubiquitous repetitive sequences in plant genomes, the main drawback for chromosome painting, we successfully employed our methodology on two plant species. Both have more than 80% repetitive sequences, which is compared to the human genome (66-69%). This is the first time that plant chromosome-specific probes were successfully obtained from a single A mitotic or meiotic microdissected chromosome. Thereby, we assume that chromosome painting through microdissection and CISS hybridization can now be considered a reality in the field of plant cytogenetics.

RESUMO Objetivo: caracterizar o perfil clínico epidemiológico e a distribuição espacial da incidência da hanseníase em territórios fronteiriços da América do Sul. Método: trata-se de um estudo ecológico. O estudo compreendeu a Província de Misiones na Argentina e a Região Sul do Brasil. A população foi composta por 10.319 casos novos de hanseníase, diagnosticados entre 2010 e 2016. Resultados: o estado do Paraná foi o mais endêmico, representando 70,2% (n=7,247) dos casos. Houve predomínio da classificação operacional multibacilar (79,8%, n=8.233) e Grau 0 de incapacidade física (50,6%, n=5.223). Em Misiones 18,9% utilizaram esquema de tratamento substitutivo. No período, notou-se uma situação hiperendêmica em 780 (65,5%) dos municípios/ departamentos estudados. Conclusão: o estudo mostrou que as regiões estudadas apresentam alta endemicidade, transmissão ativa e diagnóstico tardio da hanseníase. Essas tendências entrelaçadas à força de morbidade e de transmissão recente e persistente da doença, ampliam a relevância da hanseníase como problema de saúde pública na região.

Metodología: se trata de un estudio ecológico. El estudio incluyó la Provincia de Misiones en Argentina y la Región Sur de Brasil. La población estaba compuesta por 10.319 nuevos casos de lepra diagnosticados entre 2010 y 2016. Resultados: el estado de Paraná fue el más endémico, representando el 70,2% (n=7.247) de los casos. Predominaron la clasificación operacional multibacilar (79,8%, n=8.233) y el Grado 0 de discapacidad física (50,6%, n=5.223). En Misiones, el 18,9% utilizó un régimen de tratamiento de sustitución. En el periodo se observó una situación hiperendémica en 780 (65,5%) de las ciudades/departamentos estudiados. Conclusión: El estudio mostró que las regiones estudiadas tienen alta endemicidad, transmisión activa y diagnóstico tardío de la lepra. Estas tendencias entrelazadas en cuanto a la fuerza de la morbilidad y la transmisión reciente y persistente de la enfermedad aumentan la importancia de la lepra como un problema de salud pública en la región.
ABSTRACT Objective: to characterize the epidemiological clinical profile and spatial distribution of the incidence of leprosy in border territories of South America. Method: this is an ecological study. The study included the Province of Misiones in Argentina and the Southern Region of Brazil. The population consisted of 10,319 new leprosy cases diagnosed between 2010 and 2016. Results: the state of Paraná was the most endemic, representing 70.2% (n=7.247) of the cases. There was a predominance of multibacillary operational classification (79.8%, n=8,233) and Grade 0 of physical disability (50.6%, n=5,223). In Misiones, 18.9% used a substitution treatment regimen. In the period, a hyperendemic situation was observed in 780 (65.5%) of the studied cities/departments. Conclusion: the study revealed that the regions studied have high endemicity, active transmission and late diagnosis of leprosy. These intertwined trends in the strength of morbidity and recent and persistent transmission of the disease increase the relevance of leprosy as a public health problem in the region.
