RESUMO

Resident macrophages are tissue-specific innate immune cells acting as sentinels, constantly patrolling their assigned tissue to maintain homeostasis, and quickly responding to pathogenic invaders or molecular danger signals molecules when necessary. Adenosine triphosphate (ATP), when released to the extracellular medium, acts as a danger signal through specific purinergic receptors. Interaction of ATP with the purinergic receptor P2X7 activates macrophages and microglial cells in different pathological conditions, triggering inflammation. The highly expressed P2X7 receptor in these cells induces cell membrane permeabilization, inflammasome activation, cell death, and the production of inflammatory mediators, including cytokines and nitrogen and oxygen-reactive species. This review explores the techniques to evaluate the functional and molecular aspects of the P2X7 receptor, particularly in macrophages and microglial cells. Polymerase chain reaction (PCR), Western blotting, and immunocytochemistry or immunohistochemistry are essential for assessing gene and protein expression in these cell types. Evaluation of P2X7 receptor function involves the use of ATP and selective agonists and antagonists and diverse techniques, including electrophysiology, intracellular calcium measurements, ethidium bromide uptake, and propidium iodide cell viability assays. These techniques are crucial for studying the role of P2X7 receptors in immune responses, neuroinflammation, and various pathological conditions. Therefore, a comprehensive understanding of the functional and molecular aspects of the P2X7 receptor in macrophages and microglia is vital for unraveling its involvement in immune modulation and its potential as a therapeutic target. The methodologies presented and discussed herein offer valuable tools for researchers investigating the complexities of P2X7 receptor signaling in innate immune cells in health and disease.

To assess the ideal time for caffeine administration in preterms, identifying its effects and safety. Study Design: Meta-analysis conducted including preterms <32 weeks GA or BW < 1500 g, comparing caffeine administration time: <24 x ≥24HOL, <48 x ≥48HOL, <72 x ≥72HOL. 18 studies included 76.998 patients. The median age of starting caffeine was the first 24 HOL. In the overall comparisons, there was reduction in patent ductus arteriosus (OR 0.71 [0.55, 0. 92]; low evidence), retinopathy of prematurity (OR 0.71 [0.54, 0.93]; moderate evidence), severe brain injury (OR 0.79 [0.70, 0.91]; moderate evidence), bronchopulmonary dysplasia (BPD) (OR 0.69 [0.59, 0.81]; moderate evidence), composite outcome of BPD or death (OR 0.76 [0.66, 0.88]; moderate evidence). Mortality increase was found (OR 1.20 [1.12, 1.29], very low evidence).Caffeine in the first 24 HOL has benefits in reducing morbidities associated with prematurity. Mortality finding is potentially due to survival bias.

BACKGROUND: In general, patients are referred for rheumatological evaluation due to isolated laboratory abnormalities, especially antinuclear antibody (ANA) positivity, with the risk of more severe patients remaining on the waiting list for longer than desired. The aim of this study was to analyze the demographic, clinical, and laboratory information of patients referred to a specialized rheumatological care unit because of positive antinuclear antibody. METHODS: This is a retrospective study of 99 out of 1670 patients seen by the same rheumatologist between 01/01/2011 and 01/01/2019. Patients whose referrals were exclusively due to the ANA test result and the specialist's final diagnosis being "abnormal finding of serum immunological test" (ICD-10 R769) were included. Sociodemographic, clinical, and laboratory information were extracted from the consulting rheumatologist's chart. Descriptive statistics were used for data analysis. RESULTS: A total of 99 patients were included, most of whom were female (84.8%) with a median age of 49 years. At the moment of specialist's appointment, 97 patients (97.9%) repeated the ANA test, and 77 patients remained positive. Of these, only 35 (35.35%) were in a high titer range (greater than or equal to 1:320). Complete blood count for cytopenia's investigation was not performed in a high percentage of patients (22.2%), as well as urinalysis (31.3%). In addition, more than 70% of patients score 0 to 1 classification criteria for Systemic Lupus Erythematosus, according to SLE - ACR 1987 (American College of Rheumatology) and SLICC 2012 (Systemic Lupus International Collaborating Clinics). CONCLUSIONS: Most patients are still referred for specialized evaluation due to the misinterpretation of laboratory tests that were inappropriately requested in patients without clinical evidence of autoimmune rheumatic disease.

OBJECTIVE: To determine intraocular pressure (IOP) and tear production, as well as to compare the IOP obtained with the TonoVet Plus® (rebound) with the Tono-Pen Avia® (applanation) tonometers. ANIMALS: Twenty-five Mini Lionhead rabbits (n = 50 eyes). PROCEDURE: Tear production was measured at 6:00 a.m. and 6:00 p.m. by using the STT. The IOP reading was performed with the rebound tonometer, followed by the applanation tonometer, at 6:00 a.m., 9:00 a.m., 12:00 p.m., 3:00 p.m., and 6:00 p.m. Regression analysis, analysis of variance (anova) and Bland-Altman statistics were used. RESULTS: Daily tear production was 10.25 ± 3.75 mm/min, with no differences among the moments evaluated. Average daily IOP was 17.7 ± 3.08 mmHg with the TonoVet Plus® and 11.5 ± 4.56 mmHg with the Tono-Pen Avia®. IOP values were higher at the beginning and end of the day with both tonometers. CONCLUSION: The IOP values are higher with the TonoVet Plus® tonometer. The reference values of IOP and tear production obtained in this work may support the diagnosis, treatment, and monitoring of ocular disorders in pet Mini Lionhead rabbits.

Among non-communicable diseases, cardiovascular diseases are the most prevalent, accounting for approximately 17 million deaths per year. Despite conventional treatment, cardiac tissue engineering emerges as a potential alternative for the advancement and treatment of these patients, using biomaterials to replace or repair cardiac tissues. Among these materials, gelatin in its methacrylated form (GelMA) is a biodegradable and biocompatible polymer with adjustable biophysical properties. Furthermore, gelatin has the ability to replace and perform collagen-like functions for cell development in vitro. The interest in using GelMA hydrogels combined with nanomaterials is increasingly growing to promote the responsiveness to external stimuli and improve certain properties of these hydrogels by exploring the incorporation of nanomaterials into these hydrogels to serve as electrical signaling conductive elements. This review highlights the applications of electrically conductive nanomaterials associated with GelMA hydrogels for the development of structures for cardiac tissue engineering, by focusing on studies that report the combination of GelMA with nanomaterials, such as gold and carbon derivatives (carbon nanotubes and graphene), in addition to the possibility of applying these materials in 3D tissue engineering, developing new possibilities for cardiac studies.

The present study evaluated the role of heme oxygenase 1 (HO-1)/carbon monoxide (CO) pathway in the cholera toxin-induced diarrhea and its possible action mechanism. The pharmacological modulation with CORM-2 (a CO donor) or Hemin (a HO-1 inducer) decreased the intestinal fluid secretion and Cl- efflux, altered by cholera toxin. In contrast, ZnPP (a HO-1 inhibitor) reversed the antisecretory effect of Hemin and potentiated cholera toxin-induced intestinal secretion. Moreover, CORM-2 also prevented the alteration of intestinal epithelial architecture and local vascular permeability promoted by cholera toxin. The intestinal absorption was not altered by any of the pharmacological modulators. Cholera toxin inoculation also increased HO-1 immunoreactivity and bilirubin levels, a possible protective physiological response. Finally, using fluorometric technique, ELISA assay and molecular docking simulations, we show evidence that CO directly interacts with cholera toxin, forming a complex that affects its binding to GM1 receptor, which help explain the antisecretory effect. Thus, CO is an essential molecule for protection against choleric diarrhea and suggests its use as a possible therapeutic tool.

Objetivo: Relatar um caso de hiperplasia fibrosa inflamatória em lábio inferior, decorrente de um transtorno de comportamento repetitivo (mordedura) e o seu devido tratamento. Relato de caso: Paciente do gênero masculino, 27 anos de idade, apresentou aumento de volume em região de lábio inferior, indolor, com evolução clínica de seis a sete meses, com relatos de práticas de mordedura em região de lábio inferior. Ao exame clínico observou-se lesão exofítica em coloração de mucosa, pediculada, medindo cerca de 8mm, no seu maior diâmetro, com aspecto circunferencial, consistente à palpação e não sangrante. O paciente foi submetido à biópsia excisional, mantendo-se uma distância mínima de 5mm do pedículo. Conclusão: No caso relato, o paciente foi submetido a uma biópsia excisional com a completa remoção da lesão e obteve um prognóstico favorável... (AU)


Objective: To report a case of inflammatory fibrous hyperplasia in the lower lip, resulting from a repetitive be havior disorder (biting) and its proper treatment. Case report: Male patient, 27 year sold, presented pain less swelling in the lowerl ipregion, with clinical evolution of sixt o seven months, with report sof biting practices in the lower lip region. Clinical examination revealed an exophytic lesion in mucosal color, peduncula ted, mea suring about 8 mm in its largest diameter, with a circumferent ial appearance, consisten ton palpation and not bleeding. The patient underwent excisional biopsy, keeping a minimum distance of 5 mm from the pedicle. Reporte de caso: Paciente masculino, de 27 años, presentó un aumento de volumen em la región del labio inferior, indoloro, com evolución clínica de seis a siete meses, con reporte de prácticas de mordedura em la región del labio inferior. Theranostics combines therapeutic and imaging diagnostic techniques that are extremely dependent on the action of imaging agent, transporter of therapeutic molecules, and specific target ligand, in which fluorescent probes can act as diagnostic agents. In particular, naphthoimidazoles are potential bioactive heterocycle compounds to be used in several biomedical applications. With this aim, a group of seven naphth[1,2-d]imidazole compounds were synthesized from ß-lapachone. Their optical properties and their cytotoxic activity against cancer cells and their compounds were evaluated and confirmed promising values for molar absorptivity coefficients (on the order of 103 to 104), intense fluorescence emissions in the blue region, and large Stokes shifts (20-103 nm). Furthermore, the probes were also selective for analyzed cancer cells (leukemic cells (HL-60). The naphth[1,2-d]imidazoles showed IC50 between 8.71 and 29.92 µM against HL-60 cells. For HCT-116 cells, values for IC50 between 21.12 and 62.11 µM were observed. The selective cytotoxicity towards cancer cells and the fluorescence of the synthesized naphth[1,2-d]imidazoles are promising responses that make possible the application of these components in antitumor theranostic systems.

Background: Malignant pleural mesothelioma (MPM) is a neoplasm with low incidence in small animals, and the possible causes are poorly elucidated but may be related to contact with asbestos. In the thoracic cavity, MMP can be localized or generalized to all cavity structures, and its clinical signs depend on this localization. Although some alternative therapies are being discussed, few studies are conclusive, with surgical intervention as the leading therapeutic option. Given this context, this report aimed to describe a case of MMP located in the mediastinum of a bitch treated with radical excision through mediated sternotomy. Case: A 7-year-old bitch of the Shar-pei breed was referred for care due to progressive weight loss and intense dyspnea. During the physical examination, dyspnea and muffled lung sounds were noted. The patient underwent hemodialysis, which showed neutrophilic leukocytosis. An abdominal ultrasound was also performed and revealed mild abdominal effusion, and chest radiography revealed an extensive tumor covering the entire chest cavity. Thoracocentesis was performed, and the material analyzed was a malignant exudate; the patient was referred to median sternotomy for exploratory purposes, and afterward, total macroscopic extirpation of the tumor was performed. A sample was sent for histopathology, and malignant mesothelioma was confirmed. The patient was discharged after 8 days of hospitalization with home treatment and did not return to the hospital. Upon contacting the guardian, we were informed that the animal had died 154 days after the procedure due to unknown causes. Discussion: Malignant pleural mesothelioma affects humans and animals; it is associated with the risk factor of contact with asbestos and the use of flea antiparasitic drugs. In small animals, its incidence is rare, albeit mesotheliomas have been reported in wild and large animals. The clinical signs are related to the location of the neoplasm. When it is located in the thoracic region, dyspnea, muffled lung sounds, cyanosis, and pleural effusion are observed in most cases. Diagnosis is usually late and incidental, although some tests, such as ultrasonography, magnetic resonance imaging, tomography, radiography, and needle biopsy, can help in the diagnosis. Histopathology is the exam of choice for definitive diagnosis, as it helps one observe the proliferation of neoplastic mesothelial cells, atypical mitosis figures, and marked cellular pleomorphism. Many therapeutic options have been discussed, including chemotherapy, immunotherapy, and anti-tumor immunization, although there is little scientific proof of their efficacy in animals. The current treatment of choice is tumor excision by surgical procedure with a palliative objective since the prognosis of the disease is unfavorable. Minimally invasive video surgery has been gaining more and more space in veterinary medicine and has proven successful in numerous cases of thoracic masses. In the present report, we chose to perform median sternotomy for total excision due to the extension of the mass that occupied the thoracic cavity practically in its entirety. Further research should be conducted to help in palliative treatments and increase the survival of patients with mesotheliomas, given that most studies are done in humans and not animals. We conclude that median sternotomy is still the therapeutic option of choice for the palliative treatment of patients with extensive thoracic pleural mesotheliomas.

Esta revisão integrativa teve como objetivo identificar na literatura a produção científica que estabeleça uma relação entre a vacinação contra o HPV e a prevenção do câncer do colo uterino. No período entre 24 de março e 28 de abril de 2023, foi realizada a busca bibliográfica seguindo os critérios de inclusão: artigos publicados nos últimos cinco anos, em português, inglês ou espanhol e Qualis CAPES mínimo B2. Para alcançar os objetivos dessa revisão, foram selecionados artigos a partir da análise dos títulos, dos resumos e da leitura integral, excluindo em cada etapa os que não se enquadravam nos critérios pré-estabelecidos. Após busca na base de dados, foram localizados um total de 973 artigos, 58 na PubMed e 915 na Biblioteca Virtual em Saúde. Destes, apenas oito artigos atenderam a todos os critérios e foram incluídos na presente revisão. Os trabalhos incluídos foram publicados, entre os anos de 2018 e 2022, em periódicos internacionais. Existem diversas evidências da relação entre a vacinação contra o HPV e a prevenção do câncer de colo uterino. Porém, a taxa de adesão à vacina ainda é muito baixa, por mais que esta tenha um papel fundamental na prevenção do câncer cervical, bem como na prevenção do câncer de pênis, anal e orofaríngeo e outras infecções pelo HPV.


This integrative review aimed to identify in the literature the scientific production that establishes a relationship between the immunization against HPV and the prevention of cervical cancer. In the period between 24 March and 28 April 2023 it was made a bibliography research following the inclusion criteria: articles published in the last five years, in Portuguese, English or Spanish, and Qualis CAPES minimum as B2. To reach this review goals, there were selected articles based on title analysis, summary and unabridged reading, excluding, in every stage, those that did not enter the preestablished criteria. After the research in the data base, there was found the total of 973 articles, 58 in PubMed and 915 in Virtual Health Library. Only eight articles met all criteria and were included in this review. All the articles included were published, between 2018 and 2022, in international periodicals. There is evidence of a link between HPV vaccination and the prevention to cervical cancer. However, the adhesion to the vaccine is still very low, even if it has a fundamental role in the prevention to cervical, penile, anal and oropharyngeal cancer, as well as in the prevention of other HPV infections.


Esta revisión integradora tuvo como objetivo identificar en la literatura la producción científica que establece una relación entre la inmunización contra el VPH y la prevención del cáncer de cuello uterino. En el período comprendido entre el 24 de marzo y el 28 de abril de 2023 se realizó una investigación bibliográfica siguiendo los criterios de inclusión: artículos publicados en los últimos cinco años, en portugués, inglés o español, y Qualis CAPES mínimo como B2. Para alcanzar los objetivos de esta revisión, se seleccionaron los artículos a partir del análisis del título, resumen y lectura íntegra, excluyendo, en cada etapa, aquellos que no entraban en los criterios preestablecidos. Después de la pesquisa en la base de datos, se encontró el total de 973 artículos, 58 en PubMed y 915 en la Biblioteca Virtual de Salud. Sólo ocho artículos cumplieron todos los criterios y fueron incluidos en esta revisión. Todos los artículos incluidos fueron publicados, entre 2018 y 2022, en publicaciones periódicas internacionales. Existen evidencias de la relación entre la vacunación frente al VPH y la prevención del cáncer de cuello de útero. Sin embargo, la adhesión a la vacuna es todavía muy baja, aunque tenga un papel fundamental en la prevención al cáncer de cuello uterino, pene, ano y orofaringe, así como en la prevención de otras infecciones por VPH.

Resumo Este artigo objetiva analisar como as psicólogas atuantes na atenção básica percebem a influência dos efeitos psicossociais do racismo e do sexismo em suas práticas e no processo de saúde e cuidado prestado às mulheres negras. Trata-se de pesquisa qualitativa de caráter descritivo e exploratório, seguindo a perspectiva da psicologia social crítica. Foi realizado um levantamento teórico-metodológico sobre os temas relevantes, assim como a produção do instrumento de coleta de dados e entrevistas semiestruturadas. Participaram do estudo oito psicólogas que atendem o público feminino e trabalham na rede de atenção básica da cidade de Salvador (BA). Os dados foram analisados pelo método hermenêutico-dialético. Nota-se que a maioria das percepções foi associada à questão biologizante da raça/cor e à noção de cuidado voltado à visão universalista do sujeito, o que implicou a ausência de ações direcionadas à promoção da saúde de mulheres negras. Contudo, observou-se a percepção interseccional e o letramento racial nos relatos de três das participantes, o que possibilitou o desenvolvimento de tentativas de práticas antirracistas e antissexistas. Conclui-se que ainda se faz necessário explorar ações da psicologia que visem à promoção da saúde de mulheres negras e que possam associar a teoria com a prática.


Abstract This article aims to analyze how psychologists who work in primary care, perceive the influence of the psychosocial effects of racism and sexism in their practices and in the health and care process provided to black women. This is a qualitative research with a descriptive and exploratory character, following the critical social psychology framework. A theoretical-methodological survey was carried out about the main subjects, as well as the production of the data collection instrument and the semi-structured interviews. The study included eight psychologists who serve the female public and work in the primary care network in the municipality of Salvador (BA). Data were analyzed by the hermeneutic-dialectical method. Most perceptions were associated with the biologizing issue of race/color and the notion of care aimed at the universalist view of the subject, which implied the absence of actions aimed at promoting the health of black women. However, the intersectional perception and racial literacy were observed in the reports of three of the participants, which enabled the development of attempts at anti-racist and anti-sexist practices. In conclusion, exploring psychology actions aimed at promoting the health of black women and that can associate theory with practice is still necessary.

The objective of this study was to determine the best level of wine making by-product meal (WBM) as a natural antioxidant to replace butylhydroxytoluene (BHT) in beef burger stored at -20 °C for up to 120 days. The treatments consisted of control (basic formulation - BF, without antioxidant); BF with BHT; and BF with WBM0.5, WBM1.0, WBM1.5, and WBM2.0, with 0.5, 1.0, 1.5 and 2.0 g WBM/100 g BF, respectively. Up to 60 days of storage, the lipid oxidation value between BHT and WBM0.5 treatments did not differ and were lower than the values presented by the other treatments. On day 90 and 120, the lipid oxidation values of treatments BHT, WBM0.5, and WBM1.0 did not differ and were lower than the values presented by WBM1.5 and WBM2.0 treatments. Burgers from all treatments with WBM inclusion had crude fiber values above 3 g/100 g. WBM1.5 and WBM2.0 treatments had the worst scores for appearance, aroma, juiciness and tenderness, in addition to the highest cooking losses. WBM can be used at up to 1 g/100 g to replace BHT in frozen beef burgers. Higher levels of WBM inclusion increased lipid oxidation and negatively affected the sensory quality of burgers.

RESUMO

The P2X7 receptor is a critical purinergic receptor in immune cells. Its activation was associated with cathepsin release into macrophage cytosol, suggesting its involvement in lysosomal membrane permeabilization (LMP) and leakage. Nevertheless, the mechanisms by which P2X7 receptor activation induces LMP and leakage are unclear. This study investigated cellular mechanisms associated with endosomal and lysosomal leakage triggered by P2X7 receptor activation. We found that ATP at 500 µM and 5 mM (but not 50 µM) induced LMP in non-stimulated peritoneal macrophages. This effect was not observed in P2X7-deficient or A740003-pretreated macrophages. We found that the P2X7 receptor and pannexin-1 channels mediate calcium influx that might be important for activating specific ion channels (TRPM2 and two-pore channels) on the membranes of late endosomes and lysosomes leading to LMP leakage and consequent cathepsin release. These findings suggest the critical role of the P2X7 receptor in inflammatory and infectious diseases via lysosomal dysfunction.

Introdução: o climatério pode ser influenciado por mudanças psicossociais simultâneas, que associadas às alterações hormonais, como o hipoestrogenismo e a diminuição dos níveis de progesterona, podem intensificar os sintomas e agravos na mulher. Objetivo: identificar os perfis sociodemográficos, obstétricos, ginecológicos, de saúde e hábitos de vida das mulheres climatéricas atendidas na rede básica de saúde, por meio da aplicação de questionário, escalas de Hamilton de Ansiedade e Depressão, e Índice Menopausal de Kupperman e Blatt. Material e Método: estudo transversal descritivo exploratório de relato de experiência, desenvolvido por graduandas em Medicina e Enfermagem. As participantes apresentam idade entre 45 e 60 anos, atendidas em quatro unidades de Estratégia de Saúde da Família. Resultados: participaram 41 mulheres, com idade média de 52,3 anos. Prevaleceram brancas, casadas, com filhos, ensino médio completo, sem plano de saúde, escore de depressão leve, índice menopausal leve, ansiedade normal e uso regular de medicamentos. Conclusão: as mulheres climatéricas necessitam de um espaço para verbalizar seus sentimentos e dúvidas em relação ao climatério e ter acesso à escuta atenta por profissionais capacitados, visando o atendimento integral de saúde com resolutividade.


Introduction: climacteric can be influenced by simultaneous psychosocial changes, which associated with hormonal changes, such as hypoestrogenism and decreased progesterone levels, can intensify symptoms and health problems in women. Objective: to identify the health profile of climacteric women in primary care regarding their obstetrics, gynecological, social, and demographic aspects, and their lifestyle as well, through a questionnaire and the application of the Hamilton Anxiety Scale, Depression Scale, and the Blatt-Kupperman Menopausal Index. Materials and Methods: it is an exploratory-descriptive cross-sectional study presented as an experience report, developed by medical and nursing students. The participants had 45 to 60 years-old and visited four units of the Family Health Strategy. Results: 41 women participated in the research, with an average age of 52.3-year-old. The majority were white, married, high school graduates, with no health insurance; had children, normal anxiety levels, mild depression, mild menopausal index, and reported regular use of medication. Conclusion: the climacteric women need some time to verbalize their feelings and doubts about the climacteric to trained professionals, aiming for a resolutive and comprehensive health care.

Toxoplasma gondii is a protozoan parasite with worldwide distribution. Dogs are considered sentinel animals, useful for the detection of environmental infections and outbreaks of disease through serological surveys. Considering this, we aimed to assess the seroprevalence of toxoplasmosis in dogs from rural areas of the hinterland of the state of Sergipe, Northeastern Brazil. A total of 77 blood samples were collected from dogs and analyzed by the Indirect Hemagglutination test for the detection of anti-T. gondii antibodies Samples were considered positive when titers greater than or equal to 1:16 were observed. In addition, an epidemiological questionnaire designed to identify characteristics of the animal was performed to owners regarding sex, breed, age, sterilization, access to areas outside the house, environmental hygiene, contact with other animals and dog food. A total of 49.3% of dogs (n = 38) were positive for anti-T. gondii antibodies, most with high titers (44.7%). A higher relative frequency of seropositivity was observed in male dogs, not sterilized, mixed breed, which were fed exclusively on homemade food and had access to the street and contact with other animals. Taken together, our data reinforce the importance of developing health education actions within the scope of public health aimed at preventing this zoonosis.


Toxoplasma gondii é um protozoário com distribuição mundial. Os cães são considerados animais sentinela, úteis para a detecção de infecções ambientais e surtos da doença por meio de levantamentos sorológicos. Portanto, o presente estudo teve como objetivo determinar a soroprevalência da toxoplasmose em cães da zona rural da região rural do Alto Sertão Sergipano, Brasil. Um total de 77 amostras de sangue de cães foram coletadas no município de Nossa Senhora da Glória, Sergipe e submetidas ao teste de Hemaglutinação Indireta para detecção de anticorpos anti-T. gondii, considerando títulos maiores ou iguais a 1:16 como positivos. Além disso, foi aplicado um questionário epidemiológico aos proprietários solicitando informações sobre sexo, raça, idade, esterilização, acesso às ruas, limpeza do ambiente, contato com outros animais e ração, além de dados do proprietário, como renda familiar e nível de educação. 49,3% dos cães (38/77) apresentaram anticorpos anti-T. gondii, a maioria com títulos elevados (44,7%). Foi observado maior frequência relativa de soropositividade em cães macho, não esterilizados, sem raça definida, que se alimentam exclusivamente de comida caseira e tem acesso a rua e contato com outros animais. Os dados obtidos reforçam a importância do desenvolvimento de ações de educação em saúde no âmbito da saúde pública voltadas à prevenção dessa zoonose.

A biocomposite system was developed and tested for the removal of the azo dye Reactive Red (RR195) from wastewater. The biocomposite was synthesized using ceramic particles containing 75% alumina which were coated using chitosan cross-linked with oxalic acid. The biocomposite showed high performance at low pH (maximum adsorption capacity = 345.3mg.g-1 at pH=2.0). The physicochemical and structure characteristics of the matrix were evaluated by Z-potential, FTIR-ATR, SEM-EDS, XRD, and porosity. Langmuir sorption isotherm and pseudosecond-order model gave the best fit. The electrostatic interaction between RR195 (due to the sulfonate groups) and the free amino groups of chitosan, enabled successive desorption/regeneration cycles. The maximum removal percentage (>80%) occurred at pH=2.0 due to the cross-linking effect. Experiments at different temperatures allowed the calculation of thermodynamic parameters (ΔG, ΔS, ΔH); adsorption was spontaneous, exothermic, and enthalpy controlled. The presence of inorganic ions ([Formula: see text] ) was analyzed during the adsorption process. This novel biocomposite can be applied as a cost-effective and environmentally friendly adsorbent for anionic azo dye removal from wastewater. The application of chitosan cross-linked with oxalic acid as a coating of the ceramic support enhanced the adsorption capacity and enabled its use under acidic conditions without solubilization.

This study evaluated the multivariate relationship among the crude protein (CP) intake and digestibility, the neutral detergent fiber (NDF) intake and digestibility, and the nitrogen excretion in hair sheep fed Mombasa grass silage mixed with açai palm seeds using canonical correlation analysis (CCA). For this purpose, a dataset was constructed using the information derived from a metabolic experiment with Santa Ines sheep (BW ± SE: 33.9 ± 1.37 kg) allocated into a 5 × 5 Latin square. The following two multivariate relationships were explored using CCA: (1) the N in both feces (Nfeces) and urine (Nurine) (canonical variate U) versus the intake of both CP (CPI) and NDF (NDFI) (canonical variate V). (2) the Nfeces and Nurine (canonical variate W) versus the digestibility of both CP (CPD) and NDF (NDFD) (canonical variate Z). Canonical loadings for relationship No. 1 revealed that Nfeces (0.971) and Nurine (- 0.249) had a positive and negative correlation, respectively, with U. Both CPI (0.960) and NDFI (0.997) had a positive and similar correlation with V. Regarding relationship No. 2, both Nfeces (0.860) and Nurine (0.500) had a positive correlation with W. Conversely, both CPD (- 0.910) and NDFD (- 0.889) had a negative and similar association with Z. In conclusion, our data showed that when a multivariate approach is used, an increase in both CPI and NDFI produces an increase in Nfeces, as well as a decrease in Nurine. Additionally, a decrease in both CPD and NDFD produces an increase in both Nfeces and Nurine.

SUMMARY Introduction: Recent research has reported the cytotoxic potential of hydrazones against various strains of cancer cells. Aim: To evaluate the anticancer activity in vitro and the pharmacokinetic profile of six synthesized hydrazonic compounds, identified as vanillin 1-phthalazinylhydrazone (VAN-1); 2,4-dinitrophenylhydra-zone vanillin (VAN-2); phenylhydrazone cinnamaldehyde (CIN-1); isonicotinoyl hydrazone cinnamaldehyde (CIN-2); cinnamaldehyde 1-phthalazinylhydrazone (CIN-3); and 2,4-dinitrophenylhydrazone cinnamaldehyde (CIN-4). The cytotoxic activity was evaluated against four strains of cancer cells. Methodology: The pharmacokinetic parameters of absorption, distribution, metabolism, excretion, and toxicity (ADME/T) of the hydrazones were evaluated using the PreADMET program. Results: Hydrazones derived from cinnamaldehyde (CIN-1 and CIN-2) showed high cytotoxic activity against leukemic (HL-60) and glioblastomas (SF-295) cell lines. The pharmacokinetic profile of the hydrazones showed that, in general, the hydrazones presented satisfactory characteristics of ADME/T. In addition, it was observed that CIN-2 presented the most promising in silico profile, showing high intestinal absorption, desirable distribution profile related to plasma protein binding, adequate renal excretion, and low toxicity. The ADME/T profile of the CIN-1 compound highlighted its potential as a promising antineoplastic agent with action of the CNS, more specifically against glioblastomas.


RESUMEN Introducción: Investigaciones recientes han informado del potencial citotóxico de las hidrazonas contra varias líneas de células cancerosas. Objetivo: Evaluar la actividad anticancerígena in vitro y el perfil farmacocinético de seis compuestos hidrazónicos sintetizados, identificados como vainillina 1-ftalazinilhidrazona (VAN-1); vainillina 2,4-dinitrofenilhidrazona (VAN-2); fenilhidrazona cinamal-dehído (CIN-1); cinamaldehído de isonicotinoil hidrazona (CIN-2); cinamalde-hído 1-ftalazinilhidrazona (CIN-3); y 2,4-dinitrofenilhidrazona cinamaldehído (CIN-4). Se evaluó la actividad citotóxica frente a cuatro líneas celulares cancerosas. Metodología: Los parámetros farmacocinéticos de absorción, distribución, metabolismo, excreción y toxicidad (ADME/T) de las hidrazonas se evaluaron mediante el programa PreADMET. Resultados: Las hidrazonas derivadas del cinamaldehído (CIN-1 y CIN-2) mostraron una alta actividad citotóxica contra las líneas celulares leucémicas (HL-60) y glioblastomas (SF-295). El perfil farmacocinético de las hidrazonas mostró que, en general, las hidrazonas mostraban características satisfactorias de ADME/T. Además, se observó que CIN-2 presentó el perfil in silico más prometedor, presentando alta absorción intestinal, perfil de distribución deseable relacionado con la unión a proteínas plasmáticas, excreción renal adecuada y baja toxicidad. El perfil ADME/T del compuesto CIN-1 destacó su potencial como agente antineoplásico prometedor con acción sobre el SNC, más específicamente contra los glioblastomas.

A atividade citotóxica foi avaliada contra quatro linhagens de células cancerígenas. Metodologia: Os parâmetros farmacocinéticos de absorção, distribuição, metabolismo, excreção e toxicidade (ADME/T) das hidrazonas foram avaliados utilizando o programa PreADMET. Genome annotation conceptually consists of inferring and assigning biological information to gene products. Over the years, numerous pipelines and computational tools have been developed aiming to automate this task and assist researchers in gaining knowledge about target genes of study. However, even with these technological advances, manual annotation or manual curation is necessary, where the information attributed to the gene products is verified and enriched. Despite being called the gold standard process for depositing data in a biological database, the task of manual curation requires significant time and effort from researchers who sometimes have to parse through numerous products in various public databases. To assist with this problem, we present CODON, a tool for manual curation of genomic data, capable of performing the prediction and annotation process. This software makes use of a finite state machine in the prediction process and automatically annotates products based on information obtained from the Uniprot database. CODON is equipped with a simple and intuitive graphic interface that assists on manual curation, enabling the user to decide about the analysis based on information as to identity, length of the alignment, and name of the organism in which the product obtained a match. Further, visual analysis of all matches found in the database is possible, impacting significantly in the curation task considering that the user has at his disposal all the information available for a given product. An analysis performed on eleven organisms was used to test the efficiency of this tool by comparing the results of prediction and annotation through CODON to ones from the NCBI and RAST platforms.

BACKGROUND: Natural naphthoquinones have shown diversified biological activities including antibacterial, antifungal, antimalarial, and cytotoxic activities. However, they are also compounds with acute cytotoxicity, immunotoxicity, carcinogenesis, and cardio- and hepatotoxicity, and the modification at their redox center is an interesting strategy to overcome such harmful activity. OBJECTIVE: In this study, four novel semisynthetic hydrazones, derived from the isomers α- and ß- lapachones (α and ß, respectively) and coupled with the drugs hydralazine (HDZ) and isoniazid (ACIL), were prepared, evaluated by electrochemical methods and assayed for anticancer activity. METHODS: The semisynthetic hydrazones were obtained and had their molecular structures established by NMR, IR, and MS. Anticancer activity was evaluated by cell viability determined by reduction of 3-(4,5-dimethyl-2-thiazol)-2,5-diphenyl-2H-tetrazolium bromide (MTT). The electrochemical studies, mainly cyclic voltammetry, were performed, in aprotic and protic media. RESULTS: The study showed that the compounds 2, 3, and 4 were active against at least one of the cancer cell lines evaluated, compounds 3 and 4 being the most cytotoxic. Toward HL-60 cells, compound 3 was 20x more active than ß-lapachone, and 3x more cytotoxic than doxorubicin. Furthermore, 3 showed an SI value of 39.62 for HL-60 cells. Compound 4 was active against all cancer cells tested, with IC50 values in the range 2.90-12.40 µM. Electrochemical studies revealed a profile typical of self-protonation and reductive cleavage, dependent on the supporting electrolyte. CONCLUSION: These results therefore indicate that compounds 3 and 4 are strong candidates as prototypes of new antineoplastic drugs.
