RESUMO

The aim of this study was to evaluate the effect of diets containing whole cottonseed (WC) on blood biochemical parameters of bulls. Thirty bulls with 30±6 months and 382.7±28.4kg were kept in feedlot (85 days) and fed the following WC levels: 0, 2.22, 4.44, 6.66, 8.88 and 11.11%. In comparing biochemical indicators from the beginning to the end of the experiment, the control group (CG) and those fed diets containing 2.22, 8.88 and 11.11% of WC had an increase (P<0.05) in serum Ca concentrations (8.34±0.65−9.56±0.92mEqL-1). In relation to Fe (202.79±69.04−300.04±79.88µg/dL), the CG and those treated with 6.66% WC showed an increase (P<0.05) in serum concentrations. As to the Mg (1.92±0.18−2.40±0.27mEqL-1), groups treated with diet containing 2.22, 4.44 and 6.66% of WC had higher (p<0.05) concentrations at the end of the study. Regarding blood lipids, groups with diets containing 2.22; 4.44; 6.66 and 8.88% of WC increased (p<0.05), respectively, in concentrations of HDL, TG, VLDL and COL. The group that received 11.11% of WC increased (P<0.05) in the concentrations of COL and HDL. It can be concluded that diets containing WC, caused no alterations in the concentrations of blood parameters analyzed in this study, with the exception of Mg.(AU)


Objetivou-se avaliar o efeito de dietas contendo caroço de algodão (CA) sobre os parâmetros sanguíneos de bovinos. Trinta touros com 30±6 meses e 382,7±28,4kg foram confinados (85 dias) e receberam dietas com as seguintes proporções de CA: 0; 2,22; 4,44; 6,66; 8,88, e 11,11%. Ao se compararem as concentrações dos indicadores bioquímicos do início com as do final do experimento, foi observado que o grupo controle e aqueles com dietas contendo 2,22; 8,88 e 11,11% de CA apresentaram aumento (P<0,05) na concentração sérica de Ca (8,34±0,65−9,56±0,92 mEqL-1). Já em relação ao Fe (202,79±69,04−300,04±79,88µg/dL), observou-se que apenas o grupo controle e aquele tratado com 6,66% apresentaram aumento (P<0,05) em sua concentração. Quanto ao Mg (1,92±0,18−2,40±0,27 mEqL-1), os grupos tratados com 2,22; 4,44 e 6,66% de CA apresentaram maiores concentrações (P<0,05) ao final do estudo. Com relação aos lipídeos sanguíneos, os grupos com dietas contendo 2,22; 4,44; 6,66 e 8,88% de CA tiveram aumento (P<0,05), respectivamente, nas concentrações de HDL, TG, VLDL e COL. O grupo que recebeu 11,11% de CA teve aumento (P<0,05) nas concentrações de COL e HDL. Conclui-se que as dietas contendo CA não causaram alterações (P>0,05) nas concentrações dos parâmetros sanguíneos analisados, com exceção do Mg.(AU)

Abstract Crepuscular period is one of the factors that may influence the biting activity of mosquitoes. Many of these insects have a peak activity in this period. The purpose of this study was to investigate the afternoon crepuscular activity of Culicidae in a remaining area of Atlantic Forest in western Santa Catarina, southern Brazil. Moreover, the possible influence of abiotic factors, the abundance and species richness were verified. In order to better analyze the influence of crepuscular period in specific composition and abundance of mosquitoes, the dusk was divided into three periods: pre-sunset, sunset and post-sunset. At the end of the study, nine hundred and eight four specimens distributed in 12 genera and 23 species were collected. Trichoprosopon pallidiventer (Lutz, 1905) (59.76%), Aedes crinifer (Theobald, 1903) (8.13%), Ae. scapularis (Rondani, 1848) (5.89%) were the most abundant species. Spring time presented the greatest abundance and species richness. During the study, among the three periods evaluated, pre-sunset had the greatest abundance and post-sunset the lowest. Pre-sunset and sunset had the greatest similarity between species. Regarding to the abiotic factors evaluated seven and 15 days before sampling, they did not present significant correlation for the three most abundant species. However, temperature had a positive correlation to these species. Moreover, the correlation between collected species and its possible role as vectors of etiological agents of diseases was discussed.


Resumo O período crepuscular é um dos fatores que pode influenciar na atividade hematofágica dos mosquitos. Muitos desses insetos iniciam ou terminam suas atividades nesse período. O objetivo deste trabalho foi estudar os Culicidae que ocorrem no crepúsculo vespertino em uma área de Floresta Atlântica no oeste de Santa Catarina, sul do Brasil. Além disso, foi analisada a possível influência de fatores abióticos, bem como abundância e riqueza de espécies. Para melhor avaliar a influência do período crepuscular na composição das espécies e na abundância destas, o crepúsculo foi dividido em três períodos: pré-crepúsculo, crepúsculo e pós-crepúsculo. Ao final do estudo foram coletados 984 exemplares distribuídos em 12 gêneros e 23 espécies. Trichoprosopon pallidiventer (Lutz, 1905) (59,76%), Aedes crinifer (Theobald, 1903) (8,13%) e Ae. scapularis (Rondani, 1848) (5,89%) foram as espécies mais abundantes. A maior abundância e riqueza de espécies se deram na primavera. Dentre os três períodos estudados, o pré-crepúsculo apresentou a maior abundância de mosquitos, em contrapartida, o pós-crepúsculo apresentou a menor abundância. Os períodos pré-crepuscular e crepuscular apresentaram maior similaridade entre si com relação à composição das espécies. Relacionando os fatores abióticos e as três espécies mais abundantes, não foi observada correlação significativa nos dados avaliados nos sete e 15 dias anteriores às coletas. Entretanto, a temperatura apresentou uma correlação positiva para estas três espécies. A relação entre as espécies coletadas e a potencial transmissão de agentes etiológicos causadores de doenças foi comentada.

Sensory and instrumental analyses evaluated the meat and fat characteristics of feedlot-finished steers fed a diet containing cottonseed. Global impression, texture, meat color, and fat color were assessed. Thirty Nellore bulls with an average age of 30 ± 6 months and initial body weight of 382.7 ± 28.4kg were kept in feedlot stalls and fed the following cottonseed levels: 0; 2.22 %; 4.44 %; 6.66 %; 8.88 %; 11.11 % of the dietary dry matter. The cottonseed used in this experiment had an average free gossypol content of 4.5g/kg of cottonseed. The overall impression of the samples, assessed by the triangle test for difference, did not differ for more than 62 % of the panelists (P>0.01). The shear strength of roasted meat varied from 6.00 to 6.54kg. According to texture profile analysis (TPA), the hardness, springiness, and chewiness of roast meat ranged from 24.15 to 28.01 N, 0.52 to 0.56, 8.42 to 11.01 N, respectively; of raw meat, 9.51 to 13.86 N, 0.26 to 0.29, and 1.38 to 1.81 N, respectively. The different treatments did not affect meat texture, meat color, or fat color (P>0.05). Meat color, luminosity, and red intensity ranged from 37.71 to 42.85, 20.68 to 25.25, and 6.74 to 8.61, respectively; fat color, luminosity, and yellow intensity ranged from 62.26 to 63.78, 11.13 to 11.62, and 10.53 to 10.86, respectively. Cottonseed intake of up to 1.13kg/animal/day, equivalent to a free gossypol intake of 5.05g/animal/day, in place of soybean meal and ground corn, did not significantly change the global sensory impression, texture, and color of the meat and fat.(AU)


Avaliou-se, por meio de análise sensorial e instrumental, o efeito da adição de caroço de algodão à dieta de bovinos confinados sobre as características da carne e da gordura. Foram avaliadas a impressão global, a textura, a cor da carne e a cor da gordura. Trinta touros da raça Nelore, com médias de idade e peso vivo inicial de 30 ± 6 meses e 382,7 ± 28,4kg, foram confinados e receberam dietas com os seguintes teores de caroço de algodão: 0; 2,22%; 4,44%; 6,66%; 8,88%; 11,11% na matéria seca da dieta. O caroço de algodão utilizado neste experimento apresentou conteúdo médio de 4,5g de gossipol livre/kg de caroço de algodão. A impressão global da carne, avaliada por meio de teste triangular de diferença, mostrou que mais de 62% dos provadores não perceberam diferença significativa (P>0,01) entre as amostras. A textura da carne assada avaliada por meio do teste de força de cisalhamento variou de 6,00 a 6,54kg. Na análise do perfil de textura (TPA) da carne assada, a dureza, a elasticidade e a mastigabilidade variaram, respectivamente, de 24,15 a 28,01N, de 0,52 a 0,56N e de 8,42 a 11,01N. Na TPA da carne crua, variaram, respectivamente, de 9,51 a 13,86N, de 0,26 a 0,29N e de 1,38 a 1,81N. Na avaliação da cor da carne, a luminosidade, a intensidade de vermelho e a intensidade de amarelo da cor da carne variaram, respectivamente, de 37,71 a 42,85, de 20,68 a 25,25 e de 6,74 a 8,61. E para a cor da gordura, variaram, respectivamente, de 62,26 a 63,78, de 11,13 a 11,62 e de 10,53 a 10,86. A textura, a cor da carne e a cor da gordura não apresentaram diferenças significativas (P>0,05) entre os diferentes tratamentos. O consumo de caroço de algodão em até 1,13kg/animal/dia, que resultou no consumo de 5,05g de gossipol livre/animal/dia, em substituição ao farelo de soja e ao grão de milho triturado, não causou alterações significativas nas características da carne quanto à impressão sensorial global, à textura, à cor da carne e à cor da gordura.(AU)

RESUMO

Os objetivos deste trabalho foram estimar componentes de variância genética aditiva, fenotípica e residual e a herdabilidade para características relacionadas com a produção de mel e com a estrutura do ninho de abelhas Melipona quadrifasciata anthidioides. Sessenta colônias de diferentes regiões da Bahia foram transladadas para caixas padronizadas modelo INPA e divididas, originando as gerações parentais G1 e G2. Foram medidas as características: estimativa da produção de mel; número, largura, volume e altura dos potes de mel; número, altura e diâmetro dos potes de pólen; peso; número, largura e diâmetro dos discos de cria e estimativa da população da colônia. As medidas foram corrigidas para o efeito fixo de mês de mensuração. Os componentes de variância e herdabilidade foram estimados por meio do método de semelhança entre parentes, utilizando-se abordagem Bayesiana. As médias e os desvios padrão variaram de 2,01±0,70cm para diâmetro de potes de pólen a 2.333,0±384,1kg para o peso das caixas. Houve indicação de convergência para todas as cadeias obtidas. As estimativas de variância genética aditiva variaram de 0,02cm para as características largura dos potes de mel a 38.587,72kg para o peso. Para as estimativas de variâncias fenotípicas, os valores variaram de 0,05 para a altura dos potes de pólen a 95.136,43kg para o peso; e para as variâncias residuais, os valores encontrados variaram de 0,02 para a variável largura dos potes de mel a 56.548,71kg para o peso. As estimativas de herdabilidade variaram de 0,35 a 0,53. Os resultados demonstraram que as características avaliadas possuem variação genética aditiva que garante boa resposta à seleção.


The aim of this study was to estimate components of genetic variance, phenotypic and residual and heritability for traits related to the production of honey and the nest structure of bee Melipona quadrifasciata anthidioides. Sixty colonies from different regions of Bahia were transferred to standard INPA model boxes and divided, creating the parental generations G1 and G2. The following characteristics were measured: estimated production of honey, number, width, height and volume of the honey pots, number, height and diameter of pollen storage pots, weight, number, length and diameter of the brood combs and estimate the population of the colony. The measurements were corrected for the fixed effects of month of measurement. The variance components and heritability were estimated by the method of similarity between relatives using the Bayesian approach. The mean and standard deviations ranged from 2.01±0.70cm diameter pots for pollen to 2333.0±384.1kg to the weight of the boxes. There was indication of convergence for all the chains obtained. Estimates of the additive genetic variance ranged from 0.02cm to the width characteristics of honey pots to 38587.72kg for weight. For the estimates of phenotypic variance the values ranged from 0.05 for the height of the pollen pots to 95136.43kg for weight; and the residual variance the values varied from 0.02 for the variable width of the honey pots to 56548.71kg for weight. The results showed that the characteristics assessed have additive genetic variation that ensures good response to selection.

Os objetivos deste trabalho foram estimar componentes de variância genética aditiva, fenotípica e residual e a herdabilidade para características relacionadas com a produção de mel e com a estrutura do ninho de abelhas Melipona quadrifasciata anthidioides. Sessenta colônias de diferentes regiões da Bahia foram transladadas para caixas padronizadas modelo INPA e divididas, originando as gerações parentais G1 e G2. Foram medidas as características: estimativa da produção de mel; número, largura, volume e altura dos potes de mel; número, altura e diâmetro dos potes de pólen; peso; número, largura e diâmetro dos discos de cria e estimativa da população da colônia. As medidas foram corrigidas para o efeito fixo de mês de mensuração. Os componentes de variância e herdabilidade foram estimados por meio do método de semelhança entre parentes, utilizando-se abordagem Bayesiana. As médias e os desvios padrão variaram de 2,01±0,70cm para diâmetro de potes de pólen a 2.333,0±384,1kg para o peso das caixas. Houve indicação de convergência para todas as cadeias obtidas. As estimativas de variância genética aditiva variaram de 0,02cm para as características largura dos potes de mel a 38.587,72kg para o peso. Para as estimativas de variâncias fenotípicas, os valores variaram de 0,05 para a altura dos potes de pólen a 95.136,43kg para o peso; e para as variâncias residuais, os valores encontrados variaram de 0,02 para a variável largura dos potes de mel a 56.548,71kg para o peso. AIM: To evaluate the ex vivo efficacy of the EndoVac system and photodynamic treatment (PDT) as adjuncts to chemomechanical debridement associated with calcium hydroxide (CaOH2 ) in reducing the levels of intracanal Enterococcus faecalis. METHODOLOGY: One hundred and twenty-five sterile premolar teeth were conventionally accessed, prepared and then contaminated with E. faecalis (ATCC 29212) for 30 days. Teeth were randomly divided into 4 groups: Control (chemomechanical debridement with conventional irrigation); Endovac (chemomechanical debridement with EndoVac system); PDT (chemomechanical debridement with conventional irrigation and PDT) and Endovac+PDT (chemomechanical debridement with EndoVac and PDT). The irrigants used in all groups were 5.25% sodium hypochlorite and 17% EDTA. After treatment, an intracanal dressing (CaOH2 ) was applied in all canals for 7 days. Samples were obtained before (T1) and after the therapeutic procedures (T2) and, after intracanal medication (T3), plated onto BHI media and incubated (37 °C, 48 h) to determine the colony-forming units (CFU mL(-1) ). RESULTS: The overall mean cell counts (CFU mL(-1) ) of E. faecalis were high at the initial contamination (T1). A significant reduction (P < 0.05) of E. faecalis mean counts was observed in all groups from baseline (T1) to both post-therapy samplings (T2 and T3); no differences amongst the groups were detected. No significant change in bacterial counts from T2 to T3 was detected. CONCLUSION: The adjunctive use of the EndoVac system and the photodynamic treatment, in combination or not, was as effective as the conventional chemomechanical debridement associated with CaOH2 on reducing the counts of intracanal E. faecalis.

AIMS: To isolate and to characterize the diversity of Chromobacterium violaceum from the Brazilian Amazon region. METHODS AND RESULTS: Twenty-two isolates were obtained from the waters and banks of the river Negro, in the Brazilian Amazon. All isolates were able to grow in vitro at 44 degrees C and pH 4.0, but were adversely affected by temperatures below 15 degrees C, and unable to survive at 4 degrees C, properties that may be related to the adaptation to the ecosystem. The isolates were joined at a final level of similarity of only 13% in the rep-PCR analysis. The analysis of 16S rRNA genes resulted in three main groups clustered at a final level of similarity of 97% and only three isolates were clustered with the type strain. Similar data were obtained for the 23S rRNA gene. CONCLUSIONS: A high level of genetic diversity was verified with indications that the Brazilian isolates would fit into at least two new clusters besides C. violaceum species. SIGNIFICANCE AND IMPACT OF THE STUDY: The results show remarkable bacterial adaptability and genetic diversity of C. violaceum in the Amazon region.

The effects of metabolic inhibition of cholesterol biosynthesis on the trafficking of the nicotinic acetylcholine receptor (AChR) to the cell membrane were studied in living CHO-K1/A5, a Chinese hamster ovary clonal line that heterologously expresses adult alpha2betadeltaepsilon mouse AChR. To this end, we submitted CHO-K1/A5 cells to long-term cholesterol deprivation, elicited by Mevinolin, a potent inhibitor of 3-hydroxy-3-methyl-glutaryl-CoA reductase and applied a combination of biochemical, pharmacological and fluorescence microscopy techniques to follow the fate of the AChR. When CHO-K1/A5 cells were grown for 48 h in lipid-deficient medium supplemented with 0.5 microM Mevinolin, total cholesterol was significantly reduced (40%). Concomitantly, the maximum number of binding sites (Bmax) of the cell-surface AChR for the competitive antagonist alpha-bungarotoxin was reduced from 647+/-30 to 352+/-34 fmol/mg protein, i.e. by 46%. The apparent dissociation constant (Kdapp) for alpha-bungarotoxin of the AChRs remaining at the cell surface was not modified by cholesterol depletion. Similarly, the half-concentration inhibiting the specific binding of the radioligand (IC50) for another competitive antagonist, d-tubocurarine, did not differ from that in control cells. The decrease in cell-surface AChR was paralleled by an increase in intracellular AChR levels, which rose from 44+/-2.1% in control cells to 74+/-3.3% in Mevinolin-treated cells. When analyzed by wide-field fluorescence microscopy, the fluorescence signal arising from alpha-bungarotoxin labeled cell-surface AChRs was reduced by approximately 70% in Mevinolin-treated cells. The distribution of intracellular AChR also changed: Alexa594-alpha-bungarotoxin-labeled AChR exhibited a highly compartmentalized pattern, concentrating at the perinuclear and Golgi-like regions. Temperature-arrest of protein trafficking magnified this effect, emphasizing the Golgi localization of the AChR. Colocalization studies using the transiently expressed fluorescent trans-Golgi/trans-Golgi network marker pEYFP/human beta1,4-galactosyltransferase and the trans-Golgi network marker syntaxin 6 provided additional support for the Golgi localization of intracellular AChRs. The low AChR cell-surface expression and the increase in intracellular AChR pools in cholesterol-depleted cells raise the possibility that cholesterol participates in the trafficking of the receptor protein to the plasmalemma and its stability at this surface location.

Denture stomatitis (DS) is frequently associated with high levels of Candida in saliva and deficient denture hygiene. In order to analyse the incidence of DS and its pre-disposing factors, we evaluated 77 edentulous patients before and 6 months after the placement of new complete dentures. Denture stomatitis was observed in 50.6 and 18.2% of the patients at the first and second evaluation, respectively. Salivary flow, Candida counts in saliva and Candida species were similar in both evaluations. Denture stomatitis and Candida in saliva were more common in females. Our results showed that denture replacement and denture hygiene improvement were useful for DS resolution, particularly types I and II. However, oral and denture hygiene must be continuous, inasmuch as salivary Candida counts remained high and it is considered an important pre-disposing factor for DS.

The group-specific component (GC) system is of interest in anthropological genetic studies because the distribution of its subtypes distinguishes among major ethnic groups. The GC system was analyzed in Curiaú and Pacoval, two remnant Quilombo populations (African-derived populations) from the Brazilian Amazon. There was no significant statistical difference in allelic frequencies between the two populations or between them and three other African-derived Brazilian populations (Mimbó, Sítio Velho, and Gaucinha in Northeastern Brazil). These populations share similarities among themselves and with African populations (high frequencies of GC*1F and lower frequencies of GC*1S), which may reflect the influence of a high level of African contribution to their formation, but there is a clear difference between them and Europeans and South American Indians. It is suggested that the GC system is a useful marker for studying relationships between single populations and major ethnic groups, but does not discriminate between populations which share the same parental stock.

The carriage of five Candida species in the mouths of normal and sialoadenectomised rats was determined for periods up to 30 days after inoculation into the oral cavity. In both test and control animals, Candida albicans was the species recovered in greatest quantities at all periods, followed by C. parapsilosis and C. tropicalis. In contrast, C. guilliermondii and C. krusei were isolatable only in small numbers and only from the 1st up to the 5th day; they were not present thereafter. Sialoadenectomy favoured oral colonisation only by C. albicans (P < 0.05) and did not influence the carriage of the other species.

Several features of the clonal cell line BC3H-1 resemble those of embryonic muscle cells at their early stage of development (Patrick et al.: J Biol Chem 252:2143-2153, 1977). Under normal culture conditions, fully differentiated BC3H-1 cells possess a spindle-shaped, muscle-like morphology with a single nucleus. Like embryonic muscle cell counterparts, they neither express the epsilon subunit nor exhibit the clustered organization of the nicotinic acetylcholine receptor (AChR) characteristic of mature myocytes. Instead, AChRs are evenly distributed upon the sarcolemma upon differentiation. Here we report that BC3H-1 cells can be induced to express AChR clusters by co-culturing the cell line with heterologous retinal neurons, which establish contacts with former cells. Clustering was also triggered by pretreating the culture substratum with several cell adhesion molecules. Polynucleation, a phenomenon observed in mature myotubes in vivo, was also observed in BC3H-1 cells under the two sets of experimental conditions. AChR clustering and polynucleation, both characteristic of the mature postsynaptic region, can thus be induced in BC3H-1 cells by at least two quite distinct pathways: neuronal cell contact and/or cell adhesion molecules. It is suggested that heterologous retinal neurons can elicit only an incomplete differentiation of BC3H-1 cells, failing to concentrate the clusters beneath the neuronal endings, and failing to induce expression of the epsilon subunit, characteristic of the mature AChR.

Phospholipid and neutral lipid composition was studied in the course of myogenic differentiation of the clonal cell line BC3H-1. Total phospholipid content increased during differentiation, predominantly in the major classes of choline and ethanolamine glycerophospholipids. The contents of other lipids, such as triacylglycerols, diminished more than 50% during this period. The content and distribution of fatty acids also underwent marked differentiation-dependent changes. The polyunsaturated (tetrapenta- and hexaenoic) fatty acid species of several phospholipid classes diminished during differentiation, especially those in choline, serine and inositol glycerophospholipids. Most noticeable were the changes in phosphatidylserine; long-chain fatty acids having 20 to 22 carbon atoms and 4 to 6 double bonds decreased from about 30 to about 10 mol%. Although increased levels of saturation in other phospholipid fatty acyl chains appear to accompany the myogenic changes of BC3H-1 cells, some unsaturated fatty acids, such as oleic acid (18:1), increased by as much as 80% during the same period, suggesting the activation of a delta 9 desaturase. Sphingomyelin contained only saturated and monoenoic fatty acids and exhibited a four- to five-fold decrease in its content of monoenoic acyl groups. Diacylglycerols became enriched in arachidonate and docosahexaenoate. The amount of cholesterol and its esters increased slightly during differentiation of BC3H-1 cells. The data show that several metabolic pathways change during myogenic differentiation of the BC3H-1 clonal cell line, particularly de novo biosynthetic pathways, elongation/desaturation reactions, and acyl chain turnover.(ABSTRACT TRUNCATED AT 250 WORDS)

Attempts to develop a satisfactory vaccine against New World cutaneous leishmaniasis have been made with varying success. We found that in mice, pretreated subcutaneously with 2 X 10(6) BCG organisms 2 weeks prior to immunization at the same site with as low as 10 micrograms of a microsomal preparation (Pol-F) of Leishmania mexicana amazonensis, a profound alteration was observed in the course of infection produced by inoculation of virulent amastigotes of the same strain. The BCG-Pol-F vaccine preparation was consistently shown to have a protective capacity associated with larger classical cellular immune response (skin test) and higher specific antibody titres in response to leishmanial challenge dose (1 X 10(6) amastigotes/mouse).
