RESUMO

Our proposal was to develop a vaccine based on total Leishmania antigens (TLA) adjuvanted with polyinosinic-polycytidylic acid [Poly(I:C)] able to induce a Th1 response which can provide protection against Leishmania infection. Mice were vaccinated with two doses of TLA-Poly(I:C) administered by subcutaneous route at 3-week interval. Humoral and cellular immune responses induced by the immunization were measured. The protective efficacy of the vaccine was evaluated by challenging mice with infective promastigotes of Leishmania (Leishmania) amazonensis into the footpad. Mice vaccinated with TLA-Poly(I:C) showed a high anti-Leishmania IgG titre, as well as increased IgG1 and IgG2a subclass titres compared with mice vaccinated with the TLA alone. The high IgG2a indicated a Th1 bias response induced by the TLA-Poly(I:C) immunization. Accordingly, the cellular immune response elicited by the formulation was characterized by an increased production of IFN-γ and no significant production of IL-4. The TLA-Poly(I:C) immunization elicited good protection, which was associated with decreased footpad swelling, a lower parasite load and a reduced histopathological alteration in the footpad. Our findings demonstrate a promising vaccine against cutaneous leishmaniasis that is relatively economic and easy to develop and which should be taken into account for preventing leishmaniasis in developing countries.

The toxicity of cadmium and copper were assayed in the estuarine crab Chasmagnathus granulata. Acute assays were made on the first larvae stage, and both acute and chronic assays were made on juvenile crabs. The acute lethal toxicity of the assayed heavy metals was three orders of magnitude higher in larvae than in juveniles. Cadmium proved to be more toxic than copper in most cases; this difference was more evident in the chronic assays on juveniles, according to the nonphysiological feature of cadmium and its persistent accumulation in organisms. During these chronic assays, cadmium produced both a significant mortality and a clear retardation of molting, in accordance with its inhibitory effect on the molting process as previously reported. Copper only caused a molting acceleration during chronic assays, perhaps as a detoxifying mechanism. Heavy metal concentrations having effects in the chronic assays have been reported in some sectors of the estuary where the assayed species lives.

Plant species associated with commercial melon crops and surrounding areas were examined to identity the natural host plants of Aphis gossypii Glover. The study was conducted in two farms located in different melon production areas and plant life zones of Costa Rica. Plant species diversity, percent coverage and distribution over time were recorded during one year. Differences between locations were observed. A total of 86 plant species (49 families) and 72 plant species (40 families) were identified associated to the crop in farms A and B, respectively. In both farms a total of 24 species plants (16 families) were colonized by A. gossypii and 16 (10 families) are new reports of host plant species for this aphid. The new reports are: Justicia comata, Tetramerium nervosum, Alternanthera pubiflora, Cassia massoni, C. reticulata, Cleome viscosa, C. spinosa, Croton argenteus, Caperonia palustris, Chamaesyce gyssopilopia, Phyllantus amarus, Sida decumbens, Ludwigia erecta, Passiflora foetida, Guazuma ulmifolia and Corchorus orinocensis.

Two exploratory investigations found an increased risk of intussusception after oral polio vaccine (OPV). A large, national, population-based study was undertaken in Cuba to investigate a possible association. Three hundred and thirty-five cases of intussusception in children under 2 years of age occurring in 1995-2000 were identified and their OPV records retrieved. The relative incidence (RI) of intussusception in defined periods up to 42 days after OPV in children under 1 year was estimated using the self-controlled case series method, controlling for age and season. The RI was not significantly raised in any of the time intervals examined within the 0-42 day period after OPV. For the period 0-42 days as a whole the RI was 1.11, 95% CI 0.74-1.67. This study does not support the hypothesis that OPV causes intussusception.
