PURPOSE: Patients submitted to radiotherapy (RT) may present in their healthy tissues surrounding the treated tumor, some typical acute inflammatory reactions induced by ionizing radiation (IR). The manifestation of inflammatory processes is a result of exacerbation of the immune system, as a response to radiation exposure, and this can be a limiting factor for RT protocols. To counteract this, some thiazolidinediones, such as LPSF/GQ-16, may be useful for modulating the patient's radioinduced inflammatory response in normal tissues. In this context, the present work aims to evaluate the activity of LPSF/GQ-16 on the levels of cytokines and the expression of the gene PPARγ in mononuclear cells irradiated in vitro, to analyze the immunomodulatory activity of the molecule and its action on radiomitigation. MATERIALS AND METHODS: For this, blood samples from eight donors were collected and irradiated with 2 Gy, then the PBMC (peripheral blood mononuclear cells) were cultured and treated with LPSF/GQ-16. The levels of cytokines TNF-α, IFN-γ, IL-2 and IL-4 were quantified by CBA, while the genes of TNF-α, IFN-γ and PPARγ were analyzed by RT-PCR. RESULTS: LPSF/GQ-16 significantly reduced the expression of proinflammatory cytokines (IFN-γ and TNF-α) in irradiated and nonirradiated groups. There was no significant reduction of anti-inflammatory cytokines (IL-2 and IL-4) by LPSF/GQ-16. The mRNA expression of PPAR-γ, IFN-γ and TNF-α in the presence of LPSF/GQ-16 was higher in the nonirradiated sample. CONCLUSION: LPSF/GQ-16 showed effective activity after irradiation, with an important immunomodulatory activity in irradiated PBMCs.

A gemini surfactant imidazoline type, namely N-(3-(2-fatty-4,5-dihydro-1H-imidazol-1-yl) propyl) fatty amide, has been obtained from the fatty acids contained in the mango seed and used as a CO2 corrosion inhibitor for API X-120 pipeline steel. Employed techniques involved potentiodynamic polarization curves, linear polarization resistance, and electrochemical impedance spectroscopy. These tests were supported by detailed scanning electronic microscopy (SEM) and Raman spectroscopy studies. It was found that obtained gemini surfactant greatly decreases the steel corrosion rate by retarding both anodic and cathodic electrochemical reactions, with an efficiency that increases with an increase in its concentration. Gemini surfactant inhibits the corrosion of steel by the adsorption mechanism, and it is adsorbed on to the steel surface according to a Langmuir model in a chemical type of adsorption. SEM and Raman results shown the presence of the inhibitor on the steel surface.

Radiotherapy (RT) is an important treatment for cervical cancer. The quality of life of patients undergoing RT may be compromised during and following treatment by nausea, diarrhea, vomiting, burns, erythema and fistula. Cytokinesis-block micronucleus (CBMN) assays may be useful for predicting adverse effects of RT for cancer. The CBMN test is easy to perform and is reproducible for screening subjects exposed to ionizing radiation. We investigated the use of the frequency of micronuclei (MN) from peripheral blood samples, irradiated in vitro, as a possible biomarker to predict the side effects of RT in patients with cervical cancer. We used 10 patients with cervical cancer receiving RT and chemotherapy. We found a strong relation between the frequency of MN and the appearance of acute side effects of RT for cervical cancer. We suggest that the methodology presented here may be useful for predicting side effects of RT for patients affected by cervical cancer and who have undergone chemotherapy.

We recently reported that Tregs from long-term Belatacept-treated kidney transplant patients displayed an altered phenotype and impaired suppressive function compared to Tregs from healthy controls. However, it remains unknown whether ex vivo expansion of Tregs from patients who underwent long-term immunosuppression may be feasible to be used in their treatment. In this work, Tregs from Belatacept-treated patients were polyclonally expanded in vitro in the presence of rapamycin and IL-2. After four weeks of expansion, Tregs from patients expressed high levels of FOXP3, CD25, CTLA-4, Helios and CCR7, and showed strong suppressive activity, even in the presence of pro-inflammatory cytokines. However, FOXP3 TSDR demethylation remained lower in expanded Tregs from Belatacept-treated patients compared to healthy control Tregs. These data suggest that ex vivo expansion of Tregs from patients undergoing long-term immunosuppression may require the use of epigenetic modifying agents to stabilize FOXP3 expression to be considered as treatment in kidney transplant patients.

PURPOSE: Vitamin D deficiency (VDD) and polymorphisms in the group-specific component (GC) gene are known to be associated in different populations. However, the effects of such genetic variants may vary across different populations. Thus, the objective of this study was to estimate the association between Vitamin D-Binding Protein (VDBP) haplotypes and VDD in mestizo postmenopausal women and Mexican Amerindian ethnic groups. METHODS: This was a cross-sectional study of 726 postmenopausal Mexican women from the Health Workers Cohort Study (HWCS) and 166 postmenopausal women from the Metabolic Analysis in an Indigenous Sample (MAIS) cohort in Mexico. GC polymorphisms (rs7045 and rs4588) were analyzed by TaqMan probes. Serum 25-hydroxyvitamin D [25(OH)D] levels were measured by Chemiluminescent Microparticle Immuno Assay. RESULTS: The prevalence of VDD serum 25(OH)D < 20 ng/mL was 43.7% in mestizo women and 44.6% in indigenous women. In HWCS, the single nucleotide polymorphisms (SNPs) rs7041 and rs4588 were associated with VDD. In addition, women from the HWCS, carrying the haplotypes GC2/2 and GC1f/2 had higher odds of VDD (OR = 2.83, 95% CI 1.14, 7.02; and OR = 2.30, 95% CI 1.40, 3.78, respectively) compared to women with haplotype 1f/1 s. These associations were not statistically significant in the MAIS cohort. CONCLUSIONS: Our results show genetic association of the analyzed SNPs and related haplotypes, on the GC gene, with VDD in mestizo Mexican postmenopausal women. Moreover, a high prevalence of VDD with high genetic variability within the country was observed. Our results support the need for national policies for preventing VDD.

Resumen Introducción El Modelo de Aceptación Tecnológica (TAM), ha sido ampliamente utilizado para predecir el uso de bases de datos científicas (BDC). No obstante, es preciso indagar el peso de otras variables identificadas en la revisión de la literatura. Objetivo: Identificar los factores influyentes en la aceptación y uso de las bases de datos científicas por parte de los estudiantes de la carrera de enfermería de una universidad privada. Método El estudio tiene un diseño no experimental, transversal de tipo descriptivo y correlacional. Se aplicó un cuestionario a estudiantes de la carrera de Enfermería (Chile). Se realizó un análisis basado en regresión de mínimos cuadrados parciales en SmartPLS. Resultados Se corroboran como variables predictoras del uso de BDC a las variables TAM (intención de uso, utilidad y facilidad de uso) y a otras variables externas al modelo (obligatoriedad, las habilidades informacionales y el entrenamiento en tecnologías). Discusión La evidencia identifica que una de las principales barreras en el uso de las BDC son las habilidades informacionales y digitales para su utilización. Conclusiones Se hace necesario incorporar nuevas metodologías para aumentar la competencia de los estudiantes con el uso de estas bases, para este efecto se consideraron las variables que resultaron predictoras.


Abstract Introduction The Technology Acceptance Model (TAM) has been extensively utilized to predict the use of scientific databases; nevertheless, it is necessary to further inquire on the weight of other literature review identified variables. Objective To identify factors influencing the acceptance and use of scientific databases by nursing students in a private university. Method This is a correlational, descriptive, transversal, and non-experimental study. A corresponding questionnaire was distributed among nursing students in Chile. A partial least squares regression analysis was performed using SmartPLS. Results TAM predicting variables to the usage of scientific databases, including intention to use, usefulness, and ease of use, were corroborated. Other TAM-external predicting variables such as obligatoriness, information-handling skills, and technology training, were also found. Discussion Evidence suggested that a main barrier to the use of scientific databases refers to the skills to handle digital information. Conclusions Considering the related predicting variables, it is necessary to incorporate new methodologies aimed at enhancing the competence of students to use scientific databases.

Studies made in the Madin-Darby canine kidney (MDCK) epithelial cell line showed that ouabain regulates cell adhesion and cell-adhesion-related biological processes, such as migration. Here, we demonstrated that 10 nM ouabain accelerates collective cell migration and heals wounds in cultured MDCK cell monolayers. Ouabain-induced acceleration of cell migration depends on activation of the cSrc-ERK1/2 signaling cascade, as it was inhibited by the kinase inhibitors PP2 and PD98059. Activation of the cSrc-ERK1/2 signaling cascade increased expression and activation of the extracellular matrix metalloproteinase-2 (MMP-2). Inhibition of MMP activity using the generic inhibitor GM6001 or the potent iMMP-2 inhibitor prevented the accelerative effect of ouabain. Likewise, Focal Adhesion Kinase (FAK) inhibition with the transfection of dominant negative peptide FRNK impaired the effect of ouabain. These results suggest that ouabain binding to the Na+,K+-ATPase accelerates collective migration of MDCK cells through activation of the cSrc-ERK1/2-FAK signaling cascade and promoting secretion and MMP activity.

OBJECTIVE: This study aimed to investigate the association of seven single nucleotide polymorphisms (SNPs) on the RMND1, CCDC170, and ESR1 genes with osteoporosis or hip fracture in a postmenopausal Mexican population. METHODS: We included a group of 400 postmenopausal women from the Health Workers Cohort Study from the Mexican Institute of Social Security. As a replication sample, we recruited 423 postmenopausal women from the National Institute of Rehabilitation. Demographic data were collected through a structured questionnaire. Bone mineral density was assessed using dual X-ray absorptiometry. Individuals were classified as normal, osteopenia, osteoporosis, and fracture, according to World Health Organization criteria. Genotyping was performed using predesigned TaqMan Probes. Linear regression analysis was used to investigate association. RESULTS: All of the analyzed SNPs showed association with at least one of the phenotypes of the study groups. In addition, we observed a region with linkage disequilibrium within the ESR1 gene in all groups. CONCLUSION: This study shows that an association of the SNPs can exist with osteopenia, osteoporosis, or fragility fracture. Our results agree with data published elsewhere, supporting the potential of these loci for the identification of the population at risk. However, additional studies are required to determine the extent of this association for other geographic regions of Mexico.

Several substances of synthetic and natural origin have been studied to determine their ability to protect the body from damage caused by ionizing radiation. Among these substances, quercetin has been shown to be a naturally occurring molecule with high radioprotective and radiomitigator potential due to its antioxidant properties. The objective of this work was to ascertain the potential radiomitigator effect of quercetin on chromosome aberration yield in lymphocytes of in vitro-irradiated human peripheral blood. At first, the DPPH (2,2-diphenyl-1-picryl-hydrazyl) radical capture test was performed to determine the antioxidant activity of quercetin and to select the concentrations to be tested. The blood was irradiated at doses of 2.5, 3.5, and 4.5 Gy and lymphocytes were cultured with quercetin at preselected concentrations of 37.5 and 75 µM. Then, the slides were prepared for scoring unstable chromosome aberrations (dicentrics, rings, and fragments). The results showed that the lymphocytes irradiated and later exposed to quercetin presented a lower frequency of chromosomal alterations compared to the control sample which was irradiated and not exposed to quercetin. The results suggest a potential radiomitigator effect of the flavonoid quercetin on human lymphocytes exposed, in vitro, to ionizing radiation. This effect may be related to decrease in the release of cytokines (INF-γ, PGE2, IL-1ß, IL6, IL-8) involved in the proinflammatory processes as well as downregulation of NF-kB and reduction of expression TGF-ß.

KEY POINTS: Light at night is essential to a 24/7 society, but it has negative consequences on health. Basically, light at night induces an alteration of our biological clocks, known as chronodisruption, with effects even when this occurs during pregnancy. Here we explored the developmental impact of gestational chronodisruption (chronic photoperiod shift, CPS) on adult and fetal adrenal biorhythms and function. We found that gestational chronodisruption altered fetal and adult adrenal function, at the molecular, morphological and physiological levels. The differences between control and CPS offspring suggest desynchronization of the adrenal circadian clock and steroidogenic pathway, leading to abnormal stress responses and metabolic adaptation, potentially increasing the risk of developing chronic diseases. ABSTRACT: Light at night is essential to a 24/7 society, but it has negative consequences on health. Basically, light at night induces an alteration of our biological clocks, known as chronodisruption, with effects even when this occurs during pregnancy. Indeed, an abnormal photoperiod during gestation alters fetal development, inducing long-term effects on the offspring. Accordingly, we carried out a longitudinal study in rats, exploring the impact of gestational chronodisruption on the adrenal biorhythms and function of the offspring. Adult rats (90 days old) gestated under chronic photoperiod shift (CPS) decrease the time spent in the open arm zone of an elevated plus maze to 62% and increase the rearing time to 170%. CPS adults maintained individual daily changes in corticosterone, but their acrophases were distributed from 12.00 h to 06.00 h. CPS offspring maintained clock gene expression and oscillation, nevertheless no daily rhythm was observed in genes involved in the regulation and synthesis of steroids. Consistent with adult adrenal gland being programmed during fetal life, blunted daily rhythms of corticosterone, core clock gene machinery, and steroidogenic genes were observed in CPS fetal adrenal glands. Comparisons of the global transcriptome of CPS versus control fetal adrenal gland revealed that 1078 genes were differentially expressed (641 down-regulated and 437 up-regulated). In silico analysis revealed significant changes in Lipid Metabolism, Small Molecule Biochemistry, Cellular Development and the Inflammatory Response pathway (z score: 48-20). Altogether, the present results demonstrate that gestational chronodisruption changed fetal and adult adrenal function. This could translate to long-term abnormal stress responses and metabolic adaptation, increasing the risk of developing chronic diseases.

Animal by-product rendering establishments are still relevant industries worldwide. Animal by-product meal safety is paramount to protect feed, animals, and the rest of the food chain from unwanted contamination. As microbiological contamination may arise from inadequate processing of slaughterhouse waste and deficiencies in good manufacturing practices within the rendering facilities, we conducted an overall establishment's inspection, including the product in several parts of the process.An evaluation of the Good Manufacturing Practices (GMP) was carried out, which included the location and access (i.e., admission) to the facilities, integrated pest management programs, physical condition of the facilities (e.g., infrastructure), equipments, vehicles and transportation, as well as critical control points (i.e., particle size and temperature set at 50 mm, 133°C at atmospheric pressure for 20 min, respectively) recommended by the OIE and the European Commission. The most sensitive points according to the evaluation are physical structure of the facilities (avg 42.2%), access to the facilities (avg 48.6%), and cleaning procedures (avg 51.4%).Also, indicator microorganisms (Salmonella spp., Clostridium spp., total coliforms, E. coli, E. coli O157:H7) were used to evaluate the safety in different parts of the animal meal production process. There was a prevalence of Salmonella spp. of 12.9, 14.3, and 33.3% in Meat and Bone Meal (MBM), poultry by-products, and fish meal, respectively. However, there were no significant differences (P = 0.73) in the prevalence between the different animal meals, according to the data collected.It was also observed that renderings associated with the poultry industry (i.e., 92.0%) obtained the best ratings overall, which reflects a satisfactory development of this sector and the integration of its production system as a whole.

Sleep loss induces a low-grade inflammatory status characterized by a subtle but sustained increase of pro-inflammatory mediators, which are key regulators of blood-brain barrier function. To investigate the influence of inflammatory status on blood-brain barrier dysfunction induced by sleep restriction we performed an experiment using two strains of mice with different immunological backgrounds, C57BL/6 mice that have a predominant pro-inflammatory response and BALB/c mice that have a predominant anti-inflammatory response. Mice were sleep-restricted during 10 days using the flowerpot technique during 20 h per day with 4 h of daily sleep opportunity. The systemic inflammatory status, blood-brain barrier permeability, and the hippocampal expression of neuroinflammatory markers were characterized at the 10th day. Serum levels of TNF and IFN-γ increased in sleep-restricted C57BL/6 but not in BALB/c mice; no changes in other cytokines were found. Sleep restriction increased blood-brain barrier permeability in C57BL/6 strain but not in BALB/c. The hippocampus of sleep-restricted C57BL/6 mice exhibited an increase in the expression of the neuroinflammatory markers Iba-1, A2A adenosine receptor, and MMP-9; meanwhile in sleep-restricted BALB/c mice the expression of this markers was lesser than the control group. These data suggest that cytokines may be playing a key role in modulating blood-brain barrier function during sleep restriction, and probably the effects are related to Iba-1, MMP-9 and A2A adenosine receptor overexpression.

Sleep is a vital phenomenon related to immunomodulation at the central and peripheral level. Sleep deficient in duration and/or quality is a common problem in the modern society and is considered a risk factor to develop neurodegenerative diseases. Sleep loss in rodents induces blood-brain barrier disruption and the underlying mechanism is still unknown. Several reports indicate that sleep loss induces a systemic low-grade inflammation characterized by the release of several molecules, such as cytokines, chemokines, and acute-phase proteins; all of them may promote changes in cellular components of the blood-brain barrier, particularly on brain endothelial cells. In the present review we discuss the role of inflammatory mediators that increase during sleep loss and their association with general disturbances in peripheral endothelium and epithelium and how those inflammatory mediators may alter the blood-brain barrier. Finally, this manuscript proposes a hypothetical mechanism by which sleep loss may induce blood-brain barrier disruption, emphasizing the regulatory effect of inflammatory molecules on tight junction proteins.

Efforts to elucidate the doubtful character of the static magnetic field (SMF) influence on living cells have been made, although the topic still faces controversies because confusing reports in the scientific literature. This study intended to collect the most relevant issues separated by different topics (relating the SMF to its action on cellular systems) and analyze how the many field intensities, cell types and exposure time would affect the cell or intracellular structures. The analysis was based in the search in online databases aiming to give a general view of how the data can show conformity. It is proposed that scientists have been searching for linearity in what is actually a well characterized nonlinear system and two outputs are considered: the high sensitivity of parameters in which specific cell responses are generated and also the complexity and particularity of each cellular system. It is possible to trigger effects from a SMF, however in a stochastic way and depending on the cell system.

The aim of this work was to determine the existence of a functional Wnt/ß-catenin signaling pathway in boar spermatozoa, which would be linked with the already well-known GSK-3 signaling pathway. This was first confirmed by detecting the presence of the specific Frizzled 3 receptor in these cells. Furthermore, this signaling pathway was activated in boar spermatozoa subjected to 'in vitro' capacitation (IVC) and subsequent progesterone-induced 'in vitro' acrosome exocytosis (IVAE) by incubating cells with separate concentrations of Wnt1 ligand, the Wnt/ß-catenin signaling pathway-specific effector. Incubation with the Wnt1 ligand increased the rhythm of the time-dependent reduction in sperm viability during the achievement of both IVC and IVAE. This finding was concomitant with an increase in the percentage of spermatozoa with altered membrane fluidity and permeability determined through both merocyanine-540 and YO-PRO-1 stains. While the Wnt1 ligand did not affect total sperm motility during the achievement of the IVC, it induced a fast and transient increase in the overall motility patterns in spermatozoa subjected to IVAE. This IVAE-linked action was related to a decrease in the percentage of cells with high mitochondrial membrane potential and an increase in the percentage of cells with high intracellular Fluo-3-marked Ca(2+) content. In conclusion, our results suggest that the Wnt1 ligand-modulated Wnt/ß-catenin signaling pathway plays a relevant role in the modulation of both IVC and subsequent, progesterone-induced IVAE. Furthermore, our data indicate that the transduction pathways by which the Wnt1 ligand acts on IVC and IVAE are different, and that the Wnt/ß-catenin pathway is independent from GSK-3 activity in the achievement of IVC.

La proteína C reactiva CRP (por sus siglas en inglés) es una proteína de fase aguda que se utiliza para el seguimiento de enfermedades inflamatorias tales como artritis reumatoidea, lupus eritematoso o vasculitis y procesos infecciosos tales como sepsis y septicemia; así como también, para evaluar la eficacia de las drogas antiinflamatorias y antimicrobianas indicadas en el tratamiento de estas patologías. Igualmente se ha asociado a daño tisular en diversas especialidades quirúrgicas. El objetivo de este estudio fue relacionar los niveles plasmáticos de CRP con la infección y el edema posterior a la cirugía de los terceros molares. A tal efecto se evaluaron 60 pacientes, distribuidos en 3 grupos A, B y C bajo antibioticoterapia profiláctica con Clindamicina (A: dosis única de 600 mg, B: 300 mg c/6h por 5 días y C: Placebo) y terapia analgésica y antiinflamatoria (Ibuprofeno 400mg c/6h por 3 días). A quienes se tomaron muestras de sangre antes y a las 72 horas de la odontectomía de los terceros molares y fotografías digitales para calcular el área de inflamación. No se demostró la relación de los niveles de CRP con infección ya que ningún paciente presentó proceso infeccioso pero si se demostró la relación cualitativa (sensibilidad) de CRP y cuantitativa mediante correlación de Spearman (p<0,05) ya que mientras mayor fue el área de la inflamación, mayores fueron los niveles plasmáticos de CRP.


The C reactive protein (CRP) is an unspecific acute phase reaction used for the follow-up of such inflammatory diseases such as rheumatoid arthritis, lupus, or vasculitis and such infectious processes like sepsis; as well as also, to evaluate the efficiency of the anti-inflammatory and antimicrobial drugs indicated in the treatment of this pathologies. Equally it has associated to tissue damage in diverse surgical specialties. The aim of this study was to evaluate the relation between CRP levels as indicator of postoperative infection and edema after third molar surgery. We evaluated 60 patients distributed in three groups A, B and C under antibiotic prophylaxis with Clindamycin (A: single dosis 600 mgs, B: 300 mgs each 6/h by 5 days and C: placebo) and analgesic and anti-inflammatory therapy with Ibuprofen 400 mg. each 6/h by 3 days. Who were taken blood samples to measure the CRP before and 72 hours after surgery and digital photographs to calculate the edema area. We did not demonstrated relation between CRP and infection because no one patient was infected in any group but we demonstrated (By Searman (p<0,05) the value of CRP as indicator of edema in the third molar surgery.

This study investigated the dynamical process of chromosome condensation after colcemid treatment. Two pairs of human chromosomes, #2 and #3, were highlighted for the accurate identification by fluorescence in situ hybridization (FISH). A computerized image analysis system was used to measure the lengths of the two pairs of chromosomes averaged over 50 metaphases of different cultures with colcemid (0.5 µg/mL) added either at 3 or 48 h of a total 72 h culture period. For determining whether the process of chromosome condensation was chaotic or random, the algorithm of Detrended Fluctuation Analysis (DFA) was used. In order to evaluate the power of the method, the data were shuffled and DFA was performed again. It was found that colcemid prolonged treatment induced a significantly greater chromosome condensation (p<0.05), and the dynamics of this process was determined by the DFA and showed to be chaotic, with scaling exponents with range values 0.5< α<1.0. When the data were shuffled, the scaling exponent αreduced around to 0.5, which was characteristic of random events. These findings reinforced the idea that colcemid could interfere in some manner with the structure of chromosomes and the dynamics of chromosome condensation was non-linear.

Species distribution modelling has been a powerful tool to explore the potential distribution of parasites in wildlife, being the basis of studies on biogeography. Vexillata spp. are intestinal nematodes found in several species of mammalian hosts, such as rodents (Geomyoidea) and hares (Leporidae) in the Nearctic and northern Neotropical regions. In the present study, we modelled the potential distribution of Vexillata spp. and their hosts, using exclusively species from the Geomyidae and Heteromyidae families, in order to identify their distributional patterns. Bioclimatic and topographic variables were used to identify and predict suitable habitats for Vexillata and its hosts. Using these models, we identified that temperature seasonality is a significant environmental factor that influences the distribution of the parasite genus and its host. In particular, the geographical distribution is estimated to be larger than that predicted for its hosts. This suggests that the nematode has the potential to extend its geographical range and also its spectrum of host species. Increasing sample size and geographical coverage will contribute to recommendations for conservation of this host-parasite system.
