OBJECTIVES: The objective of the study was to evaluate whether a twice-daily instillation of 0.45% preservative-free ketorolac tromethamine (FKT) or 0.4% benzalkonium chloride-preserved ketorolac tromethamine (BACKT), every 12 h for 30 days may affect tear film parameters and the meibography in healthy dogs. Additionally, we assessed whether the same treatments irritated the ocular surface, affected goblet cell density (GCD), and the levels of oxidative stress biomarkers (OSB) in the conjunctiva of the same dogs. PROCEDURES: Experimental and masked comparison study. In 11 healthy dogs baseline values of the lipid layer thickness, tear meniscus height, non-invasive tear breakup time (NI-TFBT), and the meibomian gland (MG) loss were assessed by OSAvet®. For each dog, one eye received 40 µL of BACKT, while the other received 40 µL FKT, every 12 h for 30 consecutive days. Tear parameters and meibography were repeated 15, 30, and 60 days post-treatments. Conjunctival hyperemia and blepharospasm were monitored at the same time points. At baseline and Day 30, a conjunctival biopsy was collected for GCD and OSB determination. RESULTS: Conjunctival hyperemia and blepharospasm were not observed. At Day 15, the MG loss increased only in FKT-treated eyes (p < .001). On Day 30, both treatment groups showed increased MG loss, shortened NI-TFBT, and reduced GCD and catalase (p < .05). At Day 30, BACKT-treated eyes showed lower levels of superoxide dismutase (SOD) (p = .006) and higher levels of malondialdehyde (MDA) (p = .02). Differences between treatments were not observed for any parameter at any time point (p > .05). 60 days after treatment, OSAvet® parameters tended to return to values assessed at baseline; however, significant differences remained for MG loss (p < .05). CONCLUSIONS: Twice-daily instillation of KT, containing or not BAC, for 30 consecutive days shortened NI-TFBT, decreased GCD, and increased the MG loss in healthy dogs. KT should be used with caution when prescribed for long periods, particularly in patients with tear film abnormalities. However, future controlled studies using KT, BAC, and other topical NSAIDs are indicated to further support this finding.

Background: Episcleral inflammation may be assumed to be primary immune-mediated, secondary to intra- or extraocular diseases, or systemic abnormalities. We aimed to report a confirmed and another suspect case of nodular episclerokeratites (NEK) due to its rarity in the clinical setting and the paucity of case reports in Brazilian literature. Cases: Case 1. Refers to a 7-year-old castrated male, Collie-mixed breed, presenting with epiphora and an irregular ocular surface shape in the left eye (LE). Ophthalmic evaluation of this eye revealed mucoid discharge, conjunctival hyperemia, episcleral injection, and a gelatinous mass in the temporal limbic region. Biomicroscopic evaluation of the anterior chamber, lens, and vitreous was impaired in the LE because of corneal vessels and a mild flare in the aqueous humor. Histopathology of a scleral biopsy revealed the presence of lymphocytes, histiocytes, and some plasma cells. Positive CD3-lymphocytes were observed by immunohistochemistry, confirming the diagnosis of NEK. Case 2. Refers to a 8-year-old, spayed female Border Collie with a history of exophthalmos, conjunctival hyperemia, and inability to close the eyelid of the LE. During ophthalmic examination, an irregular espicleral nodule of approximately 9 mm was also found in the temporal limbic region, along with enlargement of episcleral vessels and scleral thinning at the equatorial region. The cornea showed mild and diffuse edema, and white crystal-like deposits were distributed in a band-like fashion at the dorsal aspect. Ultrasonography revealed scleral thinning without evidence of a mass effect arising from the iris, ciliary body, or retrobulbar space. Based on these findings, NEK was suspected. In both cases, the clinical signs reduced significantly after seven days of topical treatment with corticosteroids and cyclosporine. Discussion: It is assumed that scleral disorders are primarily immune mediated. However, such conditions may develop secondary to ocular trauma (surgery and foreign bodies), Ehrlichia canis, and Onchocerca spp. Infections and situations were ruled out in both cases. In case 1, additional histological and immunohistochemical findings supported a primary and immune-mediated scleral disease. Although the definitive diagnosis was not confirmed by histology in case 2, one can assume that the episcleral inflammation may have arisen due to an immune-mediated disorder once the eye responded positively to corticosteroid therapy. Additionally, secondary glaucoma was excluded as a possible diagnosis in case 2, because the intraocular pressure of the affected eye was below the reference range for dogs, coupled with the irregular appearance of the episclera, which is not characteristic of canine glaucoma. Moreover, in case 2, because remission of the masses of neoplastic origin after corticotherapy was not expected, the tumor was discarded. NEK has a characteristic ploriferative behavior and resistance to topical immunosuppression; clinical recurrence was not observed in the LE of either patient who remained on treatment after 60 days of follow-up. Regarding prognosis, one study showed a correlation between cellular contingent and therapeutic responses. The percentage of positive CD79a cells (B-lymphocytes) was significantly higher in cases of epicleritis and NEK, in which a poor response was achieved after topical immunosuppressive treatment. As shown by the veterinary literature and the cases described here, the complete remission of NEK is more common in unilateral cases, as confirmed after a 12-month follow-up. The 2 reported cases are useful for clarifying the common findings, diagnosis, and long-term management of NEK. Scleral abnormalities, such as NEK, must be included in the list of differential diagnoses of glaucoma, neoplasia, and endophthalmitis during ophtalmic examination.

Objetivou-se avaliar os efeitos cardiorrespiratórios da dexmedetomidina isolada e associada à morfina em pacientes submetidas à anestesia geral inalatória com isoflurano e submetidas à ovariohisterectomia eletiva. Vinte cadelas saudáveis foram selecionadas a partir de exames físico e laboratoriais. Os animais foram distribuídos aleatoriamente em dois grupos: dexmedetomidina (10 µg/Kg) (GD) e dexmedetomidina (10 µg/Kg) associado com morfina 0,3 mg/Kg (GDM), administrados pela via intramuscular (IM). Ato contínuo, o paciente foi induzido com propofol (à efeito) e mantido com isoflurano diluído em oxigênio 100% e administrado através de vaporizador calibrado. Foram aferidos os parâmetros fisiológicos: frequências cardíaca (FC) e respiratória (f), temperatura corporal (T°C), pressões arteriais sistólica (PAS), diastólica (PAD) e média (PAM), saturação de oxigênio na hemoglobina (SpO2), pressão parcial de gás carbônico no final da expiração (ETCO2) e isoflurano expirado (ETISO). A hemogasometria arterial foi utilizada para a análise do potencial hidrogeniônico (pH), bicarbonato (HCO3-) e pressão parcial de dióxido de carbono (PaCO2). O procedimento cirúrgico teve duração máxima de 45 minutos. Os parâmetros demonstraram diferença estatística entre os tempos e grupos avaliados, de acordo com os testes de Tukey e Bonferroni (p<0,05). Observou-se bradicardia 30 minutos após a aplicação da medicação pré-anestésica e acidemia no período trans-anestésico. Apesar disto, tanto a dexmedetomidina isolada quanto associada, conferiram estabilidade hemodinâmica e respiratória, apesar da bradicardia e acidemia observada.(AU)


This study aimed to evaluate the cardiorespiratory effects of dexmedetomidine alone and associated with morphine. Twenty healthy dogs were selected from physical and laboratory exams. The animals were randomly distributed into two groups: 10 µg/Kg dexmedetomidine (DG) and 10 µg/Kg dexmedetomidine group associated with 0.3 mg/Kg morphine (GDM), administered intramuscularly (IM). The patients were induced with propofol (to effect) and maintained with isoflurane diluted in 100% oxygen and administered through a calibrated vaporizer. Physiological parameters were measured: heart rate (HR), respiratory rate (f), body temperature (T°C), systolic arterial pressure (SAP), diastolic arterial pressure (DAP) and mean arterial pressure (MAP), oxygen saturation of hemoglobin (SpO2), end-tidal carbon dioxide partial pressure (ETCO2) and end-tidal isoflurane concentration (ETISO). Arterial blood gas analysis was used to analyze hydrogen potential (pH), bicarbonate (HCO3-) and partial pressure of carbon dioxide (PaCO2). The surgical procedure lasted up to 45 minutes. The parameters showed a statistical difference between the times and groups evaluated, according to the Tukey and Bonferroni tests (p<0.05). Bradycardia was observed 30 minutes after the application of pre-anesthesic medication and acidemia in the trans-anesthetic period. Despite this, both dexmedetomidine alone and associated, provided good hemodynamic and respiratory stability, despite the bradycardia and acidemia observed.(AU)


Este estudio tuvo como objetivo evaluar los efectos cardiorrespiratorios de la dexmedetomidina sola y asociada con la morfina en pacientes sometidas a anestesia general por inhalación con isoflurano y sometidas a ovariohisterectomía electiva. Se seleccionaron veinte perras sanas a partir de exámenes físicos y de laboratorio. Los animales se dividieron aleatoriamente en dos grupos: dexmedetomidina (10 µg/kg) (GD) y dexmedetomidina (10 µg/kg) asociada con 0,3 mg/kg de morfina (GDM), administrada por vía intramuscular (IM). Posteriormente, el paciente fue inducido con propofol (en efecto) y mantenido con isoflurano diluido en oxígeno al 100% y administrado mediante de un vaporizador calibrado. Se midieron parámetros fisiológicos: frecuencia cardíaca (FC) y respiratoria (f), temperatura corporal (T°C), presión arterial sistólica (PAS), diastólica (PAD) y media (PAM), saturación de oxígeno de hemoglobina (SpO2), presión parcial de carbono dióxido al final de la expiración (ETCO2) e isoflurano expirado (ETISO). Se utilizó gasometría arterial para analizar el potencial de hidrógeno (pH), el bicarbonato (HCO3-) y la presión parcial de dióxido de carbono (PaCO2). El procedimiento quirúrgico tuvo una duración máxima de 45 minutos. Los parámetros mostraron diferencia estadística entre los tiempos y grupos evaluados, según las pruebas de Tukey y Bonferroni (p <0.05). Se observó bradicardia 30 minutos después de la aplicación de medicación preanestésica y acidemia en el período transnestésico. A pesar de esto, tanto la dexmedetomidina aislada como la asociada proporcionaron estabilidad hemodinámica y respiratoria, a pesar de la bradicardia y acidemia observadas.(AU)

This study aimed to evaluate the postoperative analgesic efficacy of dexmedetomidine alone and when associated with morphine in patients under general inhalational anesthesia with isoflurane and undergoing elective ovariohysterectomy (OH). Twenty healthy bitches were selected via physical and laboratory examinations. Prior to the study, they underwent a 24-h period of adaptation to the environment and observers. They were then randomly divided into two groups: the dexmedetomidine group receiving a dose of 10 µg/kg, and dexmedetomidine group (10 µg/kg) associated with morphine receiving a dose of 0.3 mg/kg, administered via the intramuscular route. Thereafter, patients were induced with propofol and maintained with isoflurane diluted in 100% oxygen administered through a calibrated vaporizer. To verify postoperative analgesia, pain assessments were performed using the modified Glasgow (EGM) and Melbourne (EM) scales. For sedation assessment, the Dobbins scale was used at different times: before the administration of pre-anesthetic medication (T0) and at another six times in the postoperative period, 1 (T1), 2 (T2), 4 (T3), 8 (T4), 12 (T5), and 24 (T6) h after orotracheal extubation. No statistical differences were observed between groups in the scales according to Dunn's Kruskal-Wallis post hoc test, and between the times a statistical difference was noticed by the Friedman test (p<0.05) for the EGM and EM scale scores, but there was no need for analgesic rescue. Therefore, we found that isolated dexmedetomidine and morphine produced adequate analgesic effects in the postoperative period of bitches submitted for OH.

Não foram observadas diferenças estatísticas entre os grupos nas escalas de acordo com o teste de Kruskal-Wallis post-hoc de Dunn; e entre os tempos notou-se diferença estatística pelo teste de Friedman (p<0,05) para EGM e EM, porém não houve necessidade de resgate analgésico. Portanto, conclui-se que a dexmedetomidina isolada e associada a morfina produziu efeito analgésico adequado no período pós-operatório de cadelas submetidas a OH.

O objetivo deste trabalho foi avaliar os efeitos cardiorrespiratórios de duas doses de fentanil associadas à lidocaína e cetamina em fêmeas caninas anestesiadas com sevoflurano e submetidas à ovariohisterectomia eletiva. Foram utilizados 18 animais distribuídos aleatoriamente em dois grupos. Os animais do grupo A (GA) receberam pela via intravenosa um bolus de fentanil de 0,0018 mg/kg e os do grupo B (GB) 0,0036 mg/kg, ambos associados a lidocaína 3 mg/kg e cetamina 0,6 mg/kg. Imediatamente após o bolus realizou-se a indução com propofol seguido do início da infusão contínua (IC) de fentanil na dose de 0,0018 mg/kg/h para o GA e 0,0036 mg/kg/h para GB, ambos associados a 3 e 0,6 mg/kg/h de lidocaína e cetamina. A anestesia foi mantida com sevoflurano diluído em oxigênio 100% a 1,5V% por meio de vaporizador calibrado que foi ajustado para a manutenção do plano anestésico cirúrgico. Os animais foram posicionados em decúbito dorsal e permaneceram sob ventilação espontânea. Foram avaliados os valores basais (T0), após indução (T1) e 5 (T5), 20 (T20) e 35 (T35) minutos de IC dos seguintes parâmetros: frequência cardíaca (FC), frequência respiratória (f), pressão arterial sistólica (PAS), diastólica (PAD) e média (PAM), saturação de oxigênio na hemoglobina (SatO2), pressão parcial de dióxido de carbono (EtCO2) e o sevoflurano expirado (EtSevo). A análise estatística foi realizada através da análise de variância seguida do teste de Scott-knott. As diferenças foram consideradas significativas quando P< 0,05. A FC reduziu após 20 minutos de IC e a f, PAS, PAD e PAM diminuíram após indução anestésica. Tais diferenças não foram relevantes clinicamente e os valores se mantiveram dentro do limite fisiológico. Pode-se concluir que as duas doses de infusão contínua de fentanil produziram estabilidade cardiovascular e respiratória, além de permitirem a diminuição do requerimento de sevoflurano para a realização da ovariohisterectomia eletiva.(AU)


The aim of this study was to evaluate the cardiorespiratory effects of two doses of fentanyl associated with lidocaine and ketamine in canine females anesthetized with sevoflurane and submitted to elective ovariohysterectomy. 18 animals were randomly assigned to two groups. Group A (GA) animals received a loading dose intravenously of fentanyl 0.0018 mg/kg and those of group B (GB) 0.0036 mg/kg, both associated with lidocaine 3 mg/kg and ketamine 0, 6 mg/kg. Immediately after the loading dose, induction with propofol was realized followed by continuous infusion (CI) of fentanyl at the dose of 0.0018 mg/kg/h for GA and 0.0036 mg/kg/h for GB, both associated to 3 and 0.6 mg/kg/h of lidocaine and ketamine. The anesthesia was maintained with sevoflurane diluted in 100% oxygen at 1.5% by a calibrated vaporizer that was adjusted for the maintenance of the surgical anesthetic plane. The animals were placed in dorsal decubitus position and remained under spontaneous ventilation. Was evaluated the baseline values (T0), after induction (T1) and 5 (T5), 20 (T20) and 35 (T35) minutes of following parameters: heart rate (HR), respiratory rate (f), systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean blood pressure (MAP), oxygen saturation in hemoglobin (SatO2), partial carbon dioxide (EtCO2) pressure and expired sevoflurane (EtSevo). Statistical analysis was performed through analysis of variance followed by the Scott-knott test. Differences were considered significant when P< 0.05. HR decreased after 20 minutes of CI and f, SBP, DBP and MAP decreased after anesthetic induction. These differences were not clinically relevant and the values remained within the physiological limit. It can be concluded that the two doses of continuous infusion of fentanyl produced cardiovascular and respiratory stability, besides allowing the reduction of the sevoflurane requirement for elective ovariohysterectomy.(AU)


El objetivo de este trabajo fue evaluar los efectos cardiorrespiratorios de dos dosis de fentanil asociadas a la lidocaína y cetamina en perras anestesiadas con sevoflurane y sometidas a la ovariohisterectomía electiva. Se utilizaron 18 animales distribuidos aleatoriamente en dos grupos. Los animales del grupo A (GA) recibieron por vía intravenosa un bolus de fentanil de 0,0018 mg/kg y los del grupo B (GB) 0,0036 mg/kg, ambos asociados a lidocaína 3 mg/kg y cetamina 0,6 mg/kg. Inmediatamente después del bolus se realizó la inducción con propofol seguido del inicio de la infusión continua (IC) de fentanil a la dosis de 0,0018 mg/kg/h para el GA y 0,0036 mg/kg/h para GB, ambos asociados a 3 y 0,6 mg/kg/h de lidocaína y cetamina. La anestesia fue mantenida con sevoflurane diluido en oxígeno 100% a 1,5V% a través de vaporizador calibrado que fue ajustado para el mantenimiento del plano anestésico quirúrgico. Los animales fueron colocados en decúbito dorsal y permanecieron bajo ventilación espontánea. La FC redujo después de 20 minutos de IC y la f, PAS, PAD y PAM disminuyeron después de la inducción anestésica. Estas diferencias no fueron relevantes clínicamente y los valores se mantuvieron dentro del límite fisiológico. Se puede concluir que las dos dosis de infusión continua de fentanil produjeron estabilidad cardiovascular y respiratoria, además de permitir la disminución del requerimiento de sevoflurane para la realización de la ovariohisterectomía electiva.(AU)

Background: Dioctophimosis is a parasitic disease caused by Dioctophyma renale, more commonly known as “giantKidney worm,” and can be found in other organs or free in the abdominal cavity, which is related to the patients eatinghabits. Most of the animals affected are asymptomatic. The diagnosis is made through ultrasonography, finding eggs ofthe parasite in the urine, or necropsy. The treatment of choice is nephrectomy of the affected kidney. We aimed to reportthe accidental diagnosis of parasitism by D. renale in an 8-year-old dog, which was surgically treated by nephrectomy ofthe right kidney after evident destruction of the renal parenchyma on ultrasonography.Case: An 8-year-old Boxer female dog underwent a physical examination and showed a mass in the region of the left caudal thoracic breast, bilateral alopecia on flanks, and purulent ocular secretions. Laboratory tests of complete blood count(CBC) and serum biochemical profile, X-ray examination, and ultrasonography were requested. Anemia was observedon the erythrogram, and enzymes (creatinine, urea, alanine aminotransferase [ALT], and albumin) were within normallimits. There were no abnormalities on X-ray examination, and abdominal ultrasonography showed slight splenomegaly,enlarged left kidney, reduced corticomedullary region, measuring approximately 7.63 cm longitudinally. In the rightkidney, loss of the renal parenchyma, measuring approximately 5.49 cm in diameter and the presence of a cylindrical androunded structure, typical of D. renale infection. After this, a urinalysis was suggested, which revealed the presence ofthe helminthic eggs, confirming the diagnosis. The owner was informed about the need for nephrectomy of the affectedkidney as treatment. The kidney showed destruction of the renal parenchyma. O objetivo deste trabalho foi avaliar os efeitos cardiorrespiratórios de duas doses de fentanil associadas à lidocaína e cetamina em fêmeas caninas anestesiadas com sevoflurano e submetidas à ovariohisterectomia eletiva. Foram utilizados 18 animais distribuídos aleatoriamente em dois grupos. Os animais do grupo A (GA) receberam pela via intravenosa um bolus de fentanil de 0,0018 mg/kg e os do grupo B (GB) 0,0036 mg/kg, ambos associados a lidocaína 3 mg/kg e cetamina 0,6 mg/kg. Imediatamente após o bolus realizou-se a indução com propofol seguido do início da infusão contínua (IC) de fentanil na dose de 0,0018 mg/kg/h para o GA e 0,0036 mg/kg/h para GB, ambos associados a 3 e 0,6 mg/kg/h de lidocaína e cetamina. A anestesia foi mantida com sevoflurano diluído em oxigênio 100% a 1,5V% por meio de vaporizador calibrado que foi ajustado para a manutenção do plano anestésico cirúrgico. Os animais foram posicionados em decúbito dorsal e permaneceram sob ventilação espontânea. Was evaluated the baseline values (T0), after induction (T1) and 5 (T5), 20 (T20) and 35 (T35) minutes of following parameters: heart rate (HR), respiratory rate (f), systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean blood pressure (MAP), oxygen saturation in hemoglobin (SatO2), partial carbon dioxide (EtCO2) pressure and expired sevoflurane (EtSevo). Statistical analysis was performed through analysis of variance followed by the Scott-knott test. Differences were considered significant when P< 0.05. HR decreased after 20 minutes of CI and f, SBP, DBP and MAP decreased after anesthetic induction. These differences were not clinically relevant and the values remained within the physiological limit. It can be concluded that the two doses of continuous infusion of fentanyl produced cardiovascular and respiratory stability, besides allowing the reduction of the sevoflurane requirement for elective ovariohysterectomy.

Background: Elective ovariossalpingohysterectomy is a common surgery that causes mild to moderate pain. Aiming at the treatment of pain in these patients, analgesia should be balanced from pre-anesthetic, transoperative and postoperative medication, with-2-agonists and opioids being commonly used, especially dexmedetomidine that has sedation, relaxation muscle and analgesia.The aim of the present study was to evaluate the postoperative analgesic efficacy of dexmedetomidine alone and associated withmorphine when used in the preanesthetic medication of bitches submitted to elective ovarian oysterectomy using the modifiedGlasgow pain scale and the Melbourne pain scale.Materials, Methods & Results: Twenty bitches were used in this study, all of them submitted to ovarian oysterectomy, and were clinically healthy according to laboratory and clinical exams. During the experiment, bitches were randomly distributed into two groups,which received preanesthetic medication (MPA) with 5 μg/kg dexmedetomidine alone (GD) or associated with 0.3 mg/kg morphine(GDM) intramuscularly (IM). In both groups, anesthetic induction with dose-effect propofol was performed and maintained withisoflurane diluted in 100% oxygen administered through a calibrated vaporizer and appropriate anesthetic system based on the weightof the animal and maintained on spontaneous ventilation. After stabilization of the patient the surgery was started, which lasted 45min. The bitches were evaluated by the Modified Glasgow Scale (GSM), the Melbourne Scale (MS) and the Dobbins Sedation Scale(DS), previously administered MPA (T0) and the others performed in 1h (T1), 2h (T2), 4h (T3), 8h (T4), 12h (T5) and 24h (T6) afterextubation. Background: Congenital uterine malformations are rarely identified in domestic and human animals and are related to problems during the embryonic formation of the paramesonephric ducts. Cases of agenesis and segmental aplasia or unicorn uterus have been described in bitches, but there are no reports of total uterine aplasia in this specie. The report of a case of total uterine aplasia and segmental atrophy of the cranial vagina, accidentally diagnosed is reported herein.Case: An adult female bitch without defined breed in bad general condition was attended in the emergency service in a Veterinary Hospital. During anamnesis the information was restricted because it was an errant animal rescued for the consultation, but accompanied for at least 12 months in the environment in which it lived. Data related to the estrous cycle were unknown, but no gestation was observed in the last year. Investigations detected anemia and intense thrombocytopenia, azotemia and increased alanine aminotransferase. Two days later, with no progression, images suggestive of hemometra/ mucometra were observed during abdominal ultrasonography, and the patient underwent emergency ovariohysterectomy despite the general poor condition. Before the procedure the uterus was not routinely identified, but a fibromuscular, nontubular and thin structure occupied the region corresponding to the horns and uterine body, thickening in the cervix region. Death four days after surgery and sent to necropsy. During necropsy it was observed that the vulva and the vestibule of the vagina did not present macroscopic alterations, however there was a marked digitiform narrowing of the cranial region of the vagina, which ended in blind bottom. The mucosa in this narrowing was smooth, without folds. The microscopic structure of the ovaries, uterine tubes and caudal vagina were preserved.[...](AU)

Background: Elective ovariossalpingohysterectomy is one of the most accomplished surgeries in veterinary practice, presenting moderate degree of pain. Aiming at balanced anesthesia, α2-agonists have been increasingly used, with dexmedetomidine being characterized by its sympatholytic, sedative, analgesic properties and synergism with opioids, benzodiazepines and other drugs. The objective of this study was to evaluate the effects of dexmedetomidine alone and associated with morphine under the cardiovascular, respiratory and body temperature variables in the pre and trans-operative periods in bitches submitted to elective ovariossalpingohisterectomy under general inhalation anesthesia.Materials, Methods & Results: Sixteen bitches were used in this study, which were submitted to elective ovariossalpingohisterectomy, all of which were clinically healthy based on clinical and hematological results. The animals were randomized into two groups, in which intramuscular dexmedetomidine alone and 5 µg/kg (DG) and 0.3 mg/kg morphine (GDM). In both groups, anesthesia was induced with propofol administered and maintained with isoflurane diluted in 100% oxygen administered through a calibrated vaporizer and appropriate anesthetic system based on the weight of the animal and kept under spontaneous ventilation. Background: Breast tumors are common and require surgical treatment. A mastectomy causes edema, inflammation, and moderate to severe pain; therefore, analgesics should be used efficiently during the trans and postoperative periods. Tumescence anesthesia has been studied in veterinary medicine; however, there is limited literature on the comparison of the constituents of the different solutions and the most suitable protocol. The objective of this study was to evaluate the residual postoperative analgesia of two solutions through the Melbourne, Modified Glasgow for dogs (EGM), and Visual Analogue (EVA) scales in bitches who underwent a unilateral mastectomy.Materials, Methods & Results: Twelve bitches, weighing between 5 and 15 kg and aged between 5 and 13 years old, were included in the study. To determine if the animals were medically fit to undergo the procedure, they were evaluated by clinical examination, laboratory testing (complete blood count, serum biochemistry [urea, creatinine and alanine aminotransferase/ALT], and imaging (thorax x-ray and abdominal ultrasonography). Patients were randomly divided into two groups. One group received a lidocaine-containing tumescent solution (GTL) that consisted of 210 mL of lactated Ringers solution (at a temperature between 8 and 12°C), 40 mL of 2% lidocaine hydrochloride without vasoconstrictor, and 0.5 mL of adrenaline (1 mg/mL). The other group received ropivacaine (GTR) with 233.3 mL of lactated Ringers solution (at the same temperature as the previous groups), 16.7 mL of ropivacaine (7.5 mg/mL), and 0.5 mL of adrenaline (1 mg/mL). Both groups received a combination of acepromazine (0.04 mg/kg) and meperidine (2 mg/kg) as preanesthetic medication (MPA), followed by induction using propofol (to effect) and maintenance using isoflurane. Background: Elective ovariossalpingohysterectomy is one of the most accomplished surgeries in veterinary practice, presenting moderate degree of pain. Aiming at balanced anesthesia, α2-agonists have been increasingly used, with dexmedetomidine being characterized by its sympatholytic, sedative, analgesic properties and synergism with opioids, benzodiazepines and other drugs. The objective of this study was to evaluate the effects of dexmedetomidine alone and associated with morphine under the cardiovascular, respiratory and body temperature variables in the pre and trans-operative periods in bitches submitted to elective ovariossalpingohisterectomy under general inhalation anesthesia.Materials, Methods & Results: Sixteen bitches were used in this study, which were submitted to elective ovariossalpingohisterectomy, all of which were clinically healthy based on clinical and hematological results. The animals were randomized into two groups, in which intramuscular dexmedetomidine alone and 5 µg/kg (DG) and 0.3 mg/kg morphine (GDM). In both groups, anesthesia was induced with propofol administered and maintained with isoflurane diluted in 100% oxygen administered through a calibrated vaporizer and appropriate anesthetic system based on the weight of the animal and kept under spontaneous ventilation. After stabilization of the patient, the surgical procedure started, which lasted 45 min. Background: Congenital uterine malformations are rarely identified in domestic and human animals and are related to problems during the embryonic formation of the paramesonephric ducts. Cases of agenesis and segmental aplasia or unicorn uterus have been described in bitches, but there are no reports of total uterine aplasia in this specie. The report of a case of total uterine aplasia and segmental atrophy of the cranial vagina, accidentally diagnosed is reported herein.Case: An adult female bitch without defined breed in bad general condition was attended in the emergency service in a Veterinary Hospital. During anamnesis the information was restricted because it was an errant animal rescued for the consultation, but accompanied for at least 12 months in the environment in which it lived. Data related to the estrous cycle were unknown, but no gestation was observed in the last year. Investigations detected anemia and intense thrombocytopenia, azotemia and increased alanine aminotransferase. Two days later, with no progression, images suggestive of hemometra/ mucometra were observed during abdominal ultrasonography, and the patient underwent emergency ovariohysterectomy despite the general poor condition. Before the procedure the uterus was not routinely identified, but a fibromuscular, nontubular and thin structure occupied the region corresponding to the horns and uterine body, thickening in the cervix region. Death four days after surgery and sent to necropsy. During necropsy it was observed that the vulva and the vestibule of the vagina did not present macroscopic alterations, however there was a marked digitiform narrowing of the cranial region of the vagina, which ended in blind bottom. The mucosa in this narrowing was smooth, without folds. The microscopic structure of the ovaries, uterine tubes and caudal vagina were preserved.[...]

A Osteoartrite (OA) é uma denominação clínica para uma combinação de condições patológicas que envolvem a degeneração progressiva da cartilagem articular e remodelação de osso subcondral. A curcumina, um potente agente anti-inflamatório, têm sido extensivamente estudada, no entanto não oferece boa biodisponibilidade sistêmica. Nanopartículas de ouro (AuNPs) apresentam aplicações potenciais na administração de substâncias terapêuticas aumentando a eficiência do transporte de fármacos. O objetivo deste estudo foi realizar a síntese e caracterização de um sistema conjugando as AuNPs à curcumina e avaliar seu potencial terapêutico em um modelo experimental de OA em camundongos por desestabilização do menisco medial (DMM). As AuNPs foram conjugadas com curcumina e os sistemas foram caracterizados por espectroscopia no UV-VIS, espalhamento de luz dinâmico (DLS) e determinação do potencial zeta. Formou-se 4 grupos de oito animais cada, denominados A, B, C, D que receberam injeção intra-articular de AuNPs, curcumina, AuNP-curcumina e solução fisiológica, respectivamente. Após 7 semanas, a cartilagem da articulação-femoro- tibio-patelar (AFTP) foi avaliada em uma variação de escore de 0 a 24. A conjugação de AuNP-curcumina mostrou boa estabilidade e aplicação terapêutica, mas não diferiu significativamente (P>0,05) dos grupos A e B, no entanto, mostrou menor valor de escore e significância (P<0,001) em relação ao grupo controle. Os resultados deste trabalho mostram a importância do desenvolvimento de novos nanofármacos. Neste caso a conjugação de AuNPs com a curcumina permitiu a obtenção de um nanofármaco com sugestivo potencial para aplicação no tratamento da OA.(AU)

Curcumin, a potent anti-inflammatory agent, has been extensively studied; however, it does not provide good systemic bioavailability. Gold nanoparticles (AuNPs) have potential applications in the administration of therapeutic substances in order to increase the transport efficiency of drugs. The objectives of this study were to explore the synthesis and characterization of a system combining AuNPs with curcumin and evaluate its therapeutic potential in an experimental model of OA in mice by the destabilization of the medial meniscus (DMM). The AuNPs were conjugated with curcumin and the systems were characterized by UV-VIS spectroscopy, dynamic light scattering (DLS), and zeta potential. Four groups of eight animals each were formed and labelled A, B, C, and D, which received intra-articular injections of AuNPs, curcumin, AuNP-curcumin, and physiologic solution, respectively. After seven weeks, the cartilage of the stifle joint (SJ) was rated on a scale ranging from 0 to 24. Combination of AuNP-curcumin demonstrated good stability and therapeutic applications, but it did not differ significantly (P>0.05) from groups A and B. However, the control group had a significantly lower score (P<0.001). Results of this study demonstrated the importance of developing new nanodrugs. In this case, the combination of AuNPs and curcumin yielded the nanodrug effects suggestive of a potential for application in the treatment of OA.(AU)
