RESUMO

BACKGROUND: To describe an oncolytic adenovirus (OAd) encoding SP-SA-E7-4-1BBL that is capable of inducing tumor regression in therapeutic assays. Herein, we tested whether the antitumor effect is given by the induction of a tumor-specific immune response, as well as the minimum dose needed to elicit antitumor protection and monitor the OAd biodistribution over time. METHODS AND RESULTS: C57BL/6 mice (n = 5) per group were immunized twice with OAds encoding SP-SA-E7-4-1BBL, SA-E7-4-1BBL, or SP-SA-4-1BBL and challenged with TC-1 cancer cells. The DNA construct SP-SA-E7-4-1BBL was employed as a control via biolistic or PBS injection. Groups without tumor development at 47 days were rechallenged with TC-1 cells, and follow-up lasted until day 90. The minimum dose of OAd to induce the antitumor effect was established by immunization using serial dilution doses. The cytometry bead assay and the ELISpot assay were used to evaluate cytokine release in response to ex vivo antigenic stimulation. The distribution profile of the OAd vaccine was evaluated in the different organs by histological, immunohistochemical and qPCR analyses. The OAd SP-SA-E7-4-1BBL-immunized mice did not develop tumors even in a rechallenge. A protective antitumor effect was observed from a dose that is one hundredth of most reports of adenoviral vaccines. Immunization with OAd increases Interferon-gamma-producing cells in response to antigen stimulation. OAd was detected in tumors over time, with significant morphological changes, contrary to nontumor tissues. CONCLUSIONS: The OAd SP-SA-E7-4-1BBL vaccine confers a prophylactic, safe, long-lasting, and antigen-dependent antitumor effect mediated by a Th1 antitumor immune response.

RESUMO

BACKGROUND: Occupational accidents in the plumbing activity in the construction sector in developing countries have high rates of work absenteeism. The productivity of enterprises is heavily influenced by it. OBJECTIVE: To propose a model based on the Plan, Do, Check, and Act cycle and data mining for the prevention of occupational accidents in the plumbing activity in the construction sector. METHODS: This cross-sectional study was administered on a total of 200 male technical workers in plumbing. It considers biological, biomechanical, chemical, and, physical risk factors. Three data mining algorithms were compared: Logistic Regression, Naive Bayes, and Decision Trees, classifying the occurrences occupational accident. The model was validated considering 20% of the data collected, maintaining the same proportion between accidents and non-accidents. The model was applied to data collected from the last 17 years of occupational accidents in the plumbing activity in a Colombian construction company. RESULTS: The results showed that, in 90.5% of the cases, the decision tree classifier (J48) correctly identified the possible cases of occupational accidents with the biological, chemical, and, biomechanical, risk factors training variables applied in the model. CONCLUSION: The results of this study are promising in that the model is efficient in predicting the occurrence of an occupational accident in the plumbing activity in the construction sector. For the accidents identified and the associated causes, a plan of measures to mitigate the risk of occupational accidents is proposed.

RESUMO

Scedosporium apiospermum is an opportunistic pathogen that can cause pulmonary infections in both immunosuppressive and immunocompetent patients. Cytokines are molecules that mediate the immune response to promote or eliminate fungal infections. In this work, we evaluated the cytokines profile in the lung and serum of mice infected with Scedosporium apiospermum. We found early production of IL-6, IL-1ß and TNF-α cytokines in the lung of infected mice during the first 5 days of infection. We suggest that release of pro-inflammatory cytokines could play a role in the control of fungal invasion.

RESUMO

A substantial corpus of experimental research indicates that in many species, long-term habituation appears to depend on context-stimulus associations. Some authors have recently emphasized that this type of outcome supports Wagner's priming theory, which affirms that responding is diminished when the eliciting stimulus is predicted by the context where the animal encountered that stimulus in the past. Although we agree with both the empirical reality of the phenomenon as well as the principled adequacy of the theory, we think that the available evidence is more provocative than conclusive and that there are a few nontrivial empirical and theoretical issues that need to be worked out by researchers in the future. In this paper, we comment on these issues within the framework of a quantitative version of priming theory, the SOP model.

RESUMO

This paper presents an open-source online tool for introducing psychology students to the major theoretical and empirical facts of habituation. The tool was designed in a way that combines theory and data through simulated experiments. The simulations exemplify how the priming theory of Allan R. Wagner accounts for the set of behavioral characteristics of habituation proposed by Richard F. Thompson and W. Alden Spencer in 1966. Through this interactive platform, the user can learn the basics of the theory and examine how it accounts for the empirical facts with different parameters. Instructions and commands are provided in three languages: English, Spanish, and Portuguese.

ABSTRACT This work aims to determine the way in which social responsible investments has the potential of providing dynamism to the entrepreneurial development in agribusiness initiatives. At the same time, it seeks to integrate this concept with sustainable development objectives, and with conventional financial purposes within this particular industry. To do it, this work proposes an ideal theoretical model that exposes the dynamics of the social responsible investments and its positive influence on the entrepreneurial development. Methodologically, by using relevant literature, a conceptual analysis is performed from a theoretical perspective, followed by a framework analysis through the lens of seven Colombian case studies in order to perform a theory-building approach. Specifically, this work examines several concepts related with entrepreneurial development and the influence of double- and triple-bottom-line agribusiness models, together with conditioning factors for obtaining responsible financing. Together with the model at issue, this work provides a duly supported conceptual approach with five associative propositions, which ultimately fosters the prospective testing of the model trough deductive methods in both ways: qualitative and quantitative.

Chylothorax is a serious disease, even more if it is bilateral and of a malignant cause; but it has treatment and the patient’s condition may be stable. Objective: to present two patients of medical interest and prove that a good response from the treatment may be expected.Clinical cases: two male patients with bilateral chylothorax and lymphoma were simultaneously attended at the hospital. Complementary diagnostic tests were made. The diagnosis of both patients turned out to be non-Hodgkin’s and Hodgkin’s lymphomas respectively. They were put on a diet in which fat was cut; the oncospecific treatment and repeated thoracocentesis were applied in both hemithoraxes. The first patient, after getting better from the bilateral effusion and initial symptoms, passed away 11 months later for not controlling the base disease. The second patient presented a stable condition of both complaints 18 months later. In both cases chylotorax was eased and it was not necessary to repeat the drainage. Analyses compared to other reports are made mainly when the lymphoma is the etiological cause; the adopted conduct coincides with other current studies. Conclusions: the patients got better from bilateral chylothorax; one of them did not get a stable condition of his base disease and died and the second one is asymptomatic. A good therapeutic response may be obtained...

RESUMO

The solid-pseudopapillary tumors of the pancreas (TSPP) are a uncommon neoplasm (1- 2% of exocrine tumors). Are more frequent in young women and presents a low malignant potential (15% develop metastases). Its preoperative diagnosis is very difficult, because these tumors haven`t radiological features that makes distinguish from the other pancreatic cysts tumors. Usually are a histopathological finding. Surgical therapy provides good survival, even in the presence of metastases. We present 3 cases with clinical, imaging, treatment and histopathology to provide more information about this rare disease.

The effect of lysine supplementation in diets for growing pigs on weight gain, carcass and meat characteristics was evaluated. Eighteen castrated pigs (Yorkshire, Hampshire, Duroc y Landrace) of 10.13 kg initial weight and 40 d of T2 = T1 + 0.285 kg of lysine / 100 kg of feed; and T3 = T1 + 0.569100 kg of feed. Basal diet contained 20.81% CP and 3,460 Mcal/kg of ME. Pigs were fed for 33 d. At the end of the feeding trial final weight, cross height (cm) and thoracic circumference (cm), and subcutaneous fat measured with ultrasound were recorded. For carcass and meat characteristics, four pigs per treatment selected at random were slaughtered. Samples of rib, ham and shoulder from each carcass were analyzed for DM, CP, fat and ash. There were no differences (P>0.05) between treatments for weight gain, cross height, thoracic circumference, weight at sacrifice, hot and cold carcass yields, rib eye area and subcutaneous fat. The content of DM, CP, fat and ash was similar (P>0.05) among treatments. It is concluded that lysine as growth promotant of lean tissue was no reflected in weight gain, carcass characteristics or meat composition of the growing pigs.

RESUMO

[reaction: see text] Aromatic O-triisopropylsilyl ketoximes were efficiently rearranged to cyclic and acyclic aniline derivatives on reduction with BF3-ethearate/borane. The bulk of the substituents on the silicon atom, the size of the aliphatic ring, and the presence of alkoxy substituents on the aryl group all play an important role in the aniline.

RESUMO

Statistical analysis of amino acid patterns in approximately 160,000 alpha-helices in experimentally determined structures revealed di-, tri-, and tetrapeptides, whose frequencies deviate most from the statistical model. Importantly, some sequences were never found in alpha- helices. This fact was detected initially with tripeptides, where nearly 1% of the possible sequences were never seen in the helical segments. For tetrapeptides, this effect is very strong and significant; almost 43% of the possible sequences never appear in alpha-helices. It is possible that there are some steric and energetic restrictions that do not allow these tetrameric amino acid sequences to form alpha-helical structure.
