RESUMO

BACKGROUND: Low muscle quality index (MQI) is a potential risk of developing functional impairments in older people. However, considering that individuals with Down syndrome (DS) present with a faster decline in biological aging, an investigation on MQI in individuals with DS is necessary. The aims of this present cross-sectional study were to compare (1) MQI between adolescents with and without DS and (2) evaluate laboratory versus field-based estimates of MQI. METHODS: Fifty-six adolescents were recruited and separated into two groups: DS (n = 30, 13 boys and 17 girls; age: 12.38 ± 3.07 years) and a control (non-DS; n = 26, 9 boys and 17 girls; age: 12.46 ± 2.88 years). Laboratory MQI was derived from the ratio of grip strength to arm muscle mass (in kg) measured by dual-energy X-ray absorptiometry (DXA). Field-based MQI was quantified from the ratio of hand grip strength (HGS) to body mass index (BMI). For statistical analyses, a two-way ANOVA was conducted for group comparisons, and a Pearson correlation was used to test the association between field MQI and laboratory MQI. RESULTS: Adolescents with DS displayed lower field (P = 0.001), laboratory MQI estimates (P = 0.001) and HGS (P = 0.001) as compared non-DS. Also, there was a strong correlation effect between field MQI and laboratory MQI estimates (P < 0.001, R = 0.81). CONCLUSION: Adolescents with DS have lower field and laboratory MQI compared with adolescents without DS. Simpler field MQI might be used in daily clinical practice, with special attention to those with DS.

RESUMO

Porcine respiratory disease complex is a major health concern for the porcine industry, causing significant economic loss. In this study, a total of 156 samples from pigs referred to a diagnostic laboratory in Brazil for 15 months were analyzed by histopathology, bacterial isolation, PCR, and immunohistochemistry. Multiple infections were common, so 42.3% of the pigs had more than one pathogen detected in the lungs. Swine influenza virus was detected in 25.0% of the cases. Porcine circovirus type 2 was detected in 7.1% of the pigs, which was often associated with Pasteurella multocida. In addition, one case of porcine circovirus type 3 infection associated with granulomatous pneumonia was diagnosed. Bacteria were isolated in 125 cases, namely Pasteurella multocida (34.0%), Glaesserella (Haemophilus) parasuis (35.2%), Streptococcus suis (13.5%), and Actinobacillus pleuropneumoniae (7.7%). Mycoplasma hyopneumoniae was identified in 7.0% of the cases, and 18.6% of pigs carried Salmonella sp. The most common patterns of pulmonary inflammation were broncopneumonia, bronchointerstitial pneumonia, and pleuritis, in that order. This study demonstrated that histopathology is an efficient tool along with other laboratorial diagnostic tests for establishing an etiologic diagnosis in cases of porcine respiratory disease complex.

Although thyroid dysfunction occurs frequently in humans and some animal species, the mechanisms by which hypo- and hyperthyroidism affect the corpus luteum have not been thoroughly elucidated. This study evaluated the levels of proliferative activity, angiogenesis, apoptosis and expression of cyclooxygenase-2 in the corpus luteum of female rats with thyroid dysfunction. These processes may be important in understanding the reproductive changes caused by thyroid dysfunction. A total of 18 adult female rats were divided into three groups (control, hypothyroid and hyperthyroid) with six animals per group. Three months after treatment to induce thyroid dysfunction, the rats were euthanized in the dioestrus phase. The ovaries were collected and immunohistochemically analysed for expression of the cell proliferation marker CDC-47, vascular endothelial growth factor (VEGF), VEGF receptor Flk-1 and cyclooxygenase-2 (COX-2). Apoptosis was evaluated using the TUNEL assay. Hypothyroidism reduced the intensity and area of COX-2 expression in the corpus luteum (p < 0.05), while hyperthyroidism did not alter COX-2 expression in the dioestrus phase. Hypothyroidism significantly reduced the expression of CDC-47 in endothelial cells and pericytes in the corpus luteum, whereas hyperthyroidism did not induce a detectable change in CDC-47 expression (p > 0.05). Hypothyroidism reduced the level of apoptosis in luteal cells (p < 0.05) and increased VEGF expression in the corpus luteum. In contrast, hyperthyroidism increased the level of apoptosis in the corpus luteum (p < 0.05). In conclusion, thyroid dysfunction differentially affects the levels of proliferative activity, angiogenesis and apoptosis and COX-2 expression in the corpus luteum of female rats.

RESUMO

A 10-year-old female English pointer was diagnosed with an ovarian tumour with abdominal metastases. Ultrasonography revealed several nodules of 1-5 cm diameter within the abdominal cavity. Fine needle aspiration cytology of the nodules suggested a malignant mesenchymal tumour. On necropsy examination the right ovary and its capsule were enlarged and there were white-red, friable nodular masses distributed over the surface of the pancreas, liver, omentum, mesentery and serosae of the small and large intestines. Microscopical evaluation revealed neoplastic cells with a high degree of pleomorphism and vascular invasion. Immunohistochemically, the neoplastic cells expressed myosin, desmin, vimentin and CD10, but were negative for cytokeratin, placental alkaline phosphatase, inhibin-alpha and smooth muscle actin. Based on these findings a diagnosis of primary ovarian alveolar rhabdomyosarcoma was made.

The aim of this study was to evaluate the effects of thyroxine administration on morphometric parameters, expression of vascular endothelial growth factor (VEGF) and vascularization in the uterus and placenta and reproductive parameters in gilts at 70 days of gestation. At 150 days of age, i.e., before first heat, 20 gilts were randomly divided into two experimental groups: treated (n=10) and control (n=10). The treated group received a daily dose of 400 µg of L-thyroxine (T(4)) in their diet until slaughter and the control group received only typical meals. Before artificial insemination, blood was collected to determine plasma total T(4). The gilts were inseminated in the second oestrus and slaughtered at 70 days of gestation. The foetal thyroid follicular epithelium height, number, size and weight of foetuses; foetal myogenesis, corpora lutea number, embryonic mortality rate, uterine weight, placental weight and placental fluid volume were measured. Histomorphometric and immunohistochemical analysis of uterus and placenta were determined. The averages of all variables were compared by the Student's t-test. The gilts treated with thyroxine showed significant increase of plasma total T(4). At 70 days of gestation, the heights of the trophoblastic epithelium, endometrial epithelium and endometrial gland epithelium were significantly higher in the group treated with T(4). The expression of cytoplasmatic and nuclear VEGF in trophoblastic cells and the number of blood vessels per field in endometrial stroma were significantly higher in the gilts treated with T(4). No other significant differences between groups were obtained with respect to other parameters (p>0.05). We conclude that oral administration of T(4) up to 70 days of pregnancy in gilts affects the morphometric parameters, the expression of placental VEGF and the uterine vascularization but does not affect reproductive parameters in gilts during early gestation.

RESUMO

Conidiobolomycosis is an infection caused by fungi of the genus Conidiobolus spp. that is associated with a granulomatous rhinitis in humans and animals. In sheep, the disease is clinically characterized by respiratory changes, exophthalmia, craniofacial asymmetry, and fever. Microscopically, there are multifocal granulomas containing intralesional hyphae. For a long time, this disease was diagnosed as Enzootic Ethmoid Tumor. This report described three cases of conidiobolomycosis that had been previously diagnosed as Enzootic Ethmoid Tumor. Samples of nasal cavity, lung, parotid gland, and lymph node from three sheep submitted to histopathological examination in 1979 were retrieved in 2010 for a new histological, histochemical, and immunohistochemical evaluations. Paraffin-embedded tissue sections were stained with Hematoxylin-Eosin, Grocott's Metanamine Silver, and Periodic Acid Schiff. Immunohistochemistry was performed on paraffin embedded tissues using anti-cytokeratin primary antibody. Based on histopathology, histochemistry, and immunohistochemistry, the diagnosis of conidiobolomycosis was confirmed in three sheep, which had been previously diagnosed as Enzootic Ethmoid Tumor in 1979.

A case of partial segmental aplasia of the uterine body in a sexually mature mongrel bitch in anestrous phase and normal clinical condition is reported. At necropsy, no changes in the genital organs were observed with the exception of the uterine body that had a segmental (2mm in length) decrease in diameter in its medial-caudal portion, with absence of lumen and hydrometra in the cranial portion of the uterine body and uterine horns. Histopathologic evaluation indicated reduction in thickness of the whole uterine body at the site of stenosis, with all layers preserved including perimetrium, myometrium, and endometrium, but with the absence of a lumen. The walls of the uterine horns were thin and there was a marked decrease in the number of endometrial glands. There were several follicles on different stages of development and corpora lutea in regression, indicating that the bitch had normal cyclic activity.

RESUMO

The effect of the thyroxine administration on body weight and on some physicochemical and histological characteristics of the Longissimus dorsi muscle was evaluated in gilts at 70 days of gestation. Twenty middle-sisters gilts, daughters of F1 females with a ¾ Landrace X » Large White, male were used. At 150 days of age, i.e., before first heat, the animals were randomly distributed into two groups: treated (n = 10) and control (n = 10). The treated group received a daily dose of 400μg of L-thyroxine (T4) in the diet until three days before slaughter and the control group received only diet. Before artificial insemination, blood was collected for determination of plasma total T4. The gilts were inseminated in the second estrus and slaughtered at 70 days of gestation. The initial and final body weight, daily weight gain and pH, water retention, and percentage of muscle fibers and stroma in the Longissimus dorsi muscle were studied. The means of all variables were compared by the Student t-test. Before insemination, the gilts showed clinical signs of hyperthyroidism, characterized by significant increase in plasma levels of T4, mild exophthalmos, and behavioral change characterized by aggression and agitation. There was not significant difference among groups according to the studied parameters. It was concluded that the daily administration of 400μg of thyroxine does not change the daily weight gain during the gestation, as well the pH, the water retention, and the percentage of muscle fibers and stroma of the Longissimus dorsi muscle of gilts at 70 days of gestation.(AU)

The electrocardiographical and echocardiographical alterations produced by intraruminal administration of Mascagnia rigida griseb. (Malpighiaceae) in sheep were studied. Fifteen male Santa Inês sheep were randomly allotted to three groups: G1 - animals only received water (control); G2 - animals were treated with M. rigida, 20g/kg/day, for three days; and G3 - animals were treated with M. rigida, 20g/kg/day, for seven days. All treatments were administered by an orogastric tube. M. rigida administration produced increase in heart rate during rest and exercise, and decreased in fractional ejection, and fractional shortening.
