RESUMO

The present work evaluated the immunomodulatory effect of thalidomide (Thal) at different doses on tumor-associated macrophages (TAMs) using a mouse model of human breast cancer. Mice were inoculated with 4T1 cells in the left flank and treated with Thal once a day at concentrations of 50, 100, and 150mg/kg body weight from the 5th day until the 28th day of tumor inoculation. The tumors were sized, proliferation index and TAMs count were evaluated in primary tumors and metastatic lungs. In addition, the metastasis rate was evaluated in the lungs. Thal at 150mg/kg significantly decreased tumor growth, proliferation index, and TAMs infiltration in primary tumors. Conversely, a higher number of TAMs and lower proliferation index were observed in metastatic lungs in mice treated with 150mg/kg of Thal. Furthermore, Thal at 150mg/kg significantly decreased the metastatic nodules in the lungs. Our findings demonstrated that Thal treatment considerably decreased the primary tumor and lung metastasis in mice associated with different TAM infiltration effects in these sites.(AU)


No presente trabalho, foi avaliado o efeito imunomodulador de diferentes doses de talidomida em macrófagos associados ao tumor (TAMs), em um modelo murino de câncer de mama. Camundongos foram inoculados com células 4T1, na região do flanco esquerdo, e tratados com talidomida, uma vez ao dia, nas doses de 50, 100 e 150mg/k, por massa corporal, do quinto dia ao 28º dia de inoculação tumoral. Os tumores foram medidos, o índice de proliferação celular e a contagem de TAMs foram avaliados nos tumores primários e nos pulmões com metástases. Além disso, a taxa de metástases pulmonares também foi avaliada. A talidomida na dose de 150mg/kg diminuiu significativamente o crescimento tumoral, o índice de proliferação celular e a infiltração de TAMs nos tumores primários. Por outro lado, maior número de TAMs e menor índice de proliferação celular foram observados nos pulmões metastáticos, em camundongos tratados com 150mg/kg de talidomida. Ademais, a talidomida na dose de 150mg/kg diminuiu significativamente os nódulos metastáticos nos pulmões. Os resultados demonstraram que o tratamento com talidomida diminuiu o crescimento tumoral e as metástases pulmonares em camundongos, associado com diferentes efeitos na infiltração de TAMs nesses locais.(AU)

Objetivou-se avaliar o comportamento ingestivo de cordeiros Santa Inês alimentados com resíduo de cervejaria desidratado (RCD). Foram utilizados 35 cordeiros, machos, não castrados, com peso médio inicial de 16,00±1,69kg e, aproximadamente, 70 dias de idade. Adotou-se o delineamento inteiramente ao acaso, com cinco tratamentos e sete repetições, consistindo os tratamentos em: 0; 20; 40; 60 e 80% de inclusão de RCD na porção concentrada da ração. A inclusão de RCD na ração não influenciou o tempo de alimentação (TAL; h/dia) e a eficiência de alimentação (gFDN/h; P>0,05). O TAL obtido neste estudo apresentou valor médio de 4,90h/dia. Observou-se efeito linear decrescente (P<0,05) com a inclusão do RCD, para as eficiências de alimentação (gMS/h) e ruminação (gMS/h e gFDN/h). Contudo, efeito linear crescente (P<0,05) foi constatado para tempo de ruminação e mastigação total, bem como para o número de mastigações merícicas por dia para os animais alimentados com o subproduto. A inclusão de resíduo de cervejaria desidratado influencia o comportamento ingestivo de cordeiros Santa Inês, diminuindo a eficiência de alimentação, quando relacionada ao consumo de matéria seca por hora, e aumentando o tempo de ruminação, podendo ser adicionado em até 20% na porção concentrada da ração.(AU)

Thirty-five male lambs were used, with an initial mean weight of 16.00±1.69kg and, approximately, 70 days of age. A completely randomized design was used, with five treatments and seven replicates, the treatments being: 0; 20; 40; 60 and 80% of inclusion of DBR in the concentrated portion of the ration. The inclusion of DBR in the ration did not influence feeding time (FT; h/day) and feeding efficiency (gNDF/h; P> 0.05). The FT obtained in this study had an average value of 4.90h/day. There was a decreasing linear effect (P< 0.05) for feed efficiency (gDM/h) and rumination efficiency (gDM/h and gNDF/h). However, linear increasing effect (P< 0.05) was observed for rumination and total chewing time, as well as for the number of chews per day for animals fed with the byproduct. The inclusion of dehydrated brewer's residue in the concentrate can influence the ingestive behavior of Santa Ines lambs, reducing feed efficiency, when related to the dry matter intake per hour, and increasing the total rumination time in Santa Ines lambs, it can be added up to 20% in the concentrated portion of the ration.(AU)

Cetose subclínica é uma grande preocupação em rebanhos leiteiros, e seu diagnóstico e prevenção podem ter um grande impacto na saúde animal. Este estudo comparou quatro locais diferentes para a coleta de amostras de sangue (jugular, veias mamárias e coccígeas e ponta da cauda) para medição de ß-hidroxibutirato (BHBA), usando-se um medidor portátil automático. Foram utilizadas seis vacas Holandesas, e a coleta de sangue foi feita no segundo, quinto, 10º, 15º e 21º dias pós-parto. Os resultados do medidor portátil foram semelhantes aos resultados do laboratório e apresentaram uma correlação forte de 0,83. As concentrações séricas de BHBA nas amostras de sangue coletadas na ponta da cauda, na jugular e na coccígea foram semelhantes. No entanto, o sangue retirado da veia mamária tinha uma concentração mais baixa. Portanto, as amostras de sangue para aferição de BHBA podem ser recolhidas nas veias jugular e coccígeas e na ponta da cauda, sendo as duas últimas as opções mais fáceis para monitorar o BHBA em rebanho leiteiro.(AU)

A 3 year old female feline of mixed breed was sent to the diagnostic imaging sector under suspicion of bleeding due to ovariohysterectomy. An abdominal ultrasonography was performed to confirm the initial suspicion. However, no signs of bleeding were found, instead it was observed that the left renal silhouette had two pelvises and was elongated and larger than normal. The right kidney was not found. Excretory urography was requested to evaluate the condition of the ureters. The final diagnosis was crossed renal ectopia with fusion in an asymptomatic cat with no changes in renal function.(AU)


Uma gata, sem raça definida, com três anos de idade, foi encaminhada para o setor de diagnóstico por imagem sob suspeita de hemorragia devido à ovário-histerectomia. Foi realizada ultrasonografia abdominal para confirmar a suspeita inicial, mas nenhum sinal de hemorragia foi encontrado; visibilizou-se, entretanto, a silhueta renal esquerda alongada, com presença de duas pelves. O rim direito não foi encontrado. Solicitou-se exame de urografia excretora para avaliação de ureteres. O diagnóstico foi de ectopia renal cruzada com fusão em um felino assintomático e sem alterações na função renal.(AU)

BACKGROUND: After 5 years' sick leave in Brazil, employees must retire due to disability. The duration from breast cancer surgery to the end of treatment should be ~9 months. However, diagnosis alone can take 6 months. Surveys administered soon after returning to work have highlighted problems regarding the slow speed of the treatment process and lack of protective legislation. AIMS: To assess the barriers and facilitators experienced and the coping strategies adopted by Brazilian women 30 days after return to work following breast cancer treatment. METHODS: A qualitative study of 12 women treated for breast cancer. The interviews were recorded, transcribed verbatim and independently analysed by two researchers using a standardized method of analysis. RESULTS: Women took an average of 583 days to return to work following breast cancer treatment. The return-to-work experience was considered good, with the physical barriers being fatigue and problems with the arms, and the work environmental barriers being related to discrimination from employers and overprotection from colleagues. Facilitators included social and emotional support given by colleagues/relatives/employers and jobs requiring more cognitive effort than physical exertion. Coping strategies were related to job role adjustments and reduction in tasks and working hours. CONCLUSIONS: Results were similar to those reported by previous studies, with the exception of the facilitators. Cognitive effort is commonly considered a barrier. However, the present study had an unusually long duration before return to work, possibly reducing the acute effects of chemotherapy on cognition.

Spodoptera marima (Schaus, 1904) and Spodoptera ornithogalli (Guenée, 1852) are noctuid moths recognized for their economic importance in the New World. Historically, they have been considered cryptic species, being supposedly differentiated from each other by the absence of sexual dimorphism in S. marima, subtle differences in coloration on the hindwings and scale tufts of the ovipositor and the shape of male valva. The species are allopatric: North and Central America for S. ornithogalli and South America for S. marima. Previous phylogenetic studies using multilocus DNA sequences including a few specimens of S. marima suggested these species merge into a single clade. Here, we performed a comparative analysis to elucidate further their taxonomic status, using specimens of S. marima from various localities in Brazil. These species were recovered in the phylogenetic analysis as consistently mixed with each other in a major well-supported clade within Spodoptera. The existence of a spatial pattern for genetic structure was not clearly supported when two biogeographic regions (Neotropics and Neartic) were taken in separation; however, an individual-level isolation by distance was significant. The morphological characters traditionally used to differentiate the species were also not consistent. Therefore, we now consider S. marima a new junior synonym of S. ornithogalli. Results on host plant use demonstrated that it is a polyphagous species using more than 200 plants, which may mediate phenotypic variation along its geographical range.

Genomic regions under high selective pressure present specific runs of homozygosity (ROH), which provide valuable information on the genetic mechanisms underlying the adaptation to environment imposed challenges. In broiler chickens, the adaptation to conventional production systems in tropical environments lead the animals with favorable genotypes to be naturally selected, increasing the frequency of these alleles in the next generations. In this study, ~1400 chickens from a paternal broiler line were genotyped with the 600 K Affymetrix® Axiom® high-density (HD) genotyping array for estimation of linkage disequilibrium (LD), effective population size (N e ), inbreeding and ROH. The average LD between adjacent single nucleotide polymorphisms (SNPs) in all autosomes was 0.37, and the LD decay was higher in microchromosomes followed by intermediate and macrochromosomes. The N e of the ancestral population was high and declined over time maintaining a sufficient number of animals to keep the inbreeding coefficient of this population at low levels. The ROH analysis revealed genomic regions that harbor genes associated with homeostasis maintenance and immune system mechanisms, which may have been selected in response to heat stress. Our results give a comprehensive insight into the relationship between shared ROH regions and putative regions related to survival and production traits in a paternal broiler line selected for over 20 years. These findings contribute to the understanding of the effects of environmental and artificial selection in shaping the distribution of functional variants in the chicken genome.

This study consisted of two experiments with the following objectives: to evaluate the effects of tannins from the tropical legume macrotiloma (Macrotyloma axillare) on total gas and methane (CH4 ) production, as well as on ruminal fermentation parameters by performing an in vitro bioassay, with samples incubated with and without polyethylene glycol (PEG) in a semi-automatic system; and secondly in a 17 day in vivo experiment, to determine apparent total tract digestibility (ATTD) of dietary nutrients and ruminal fermentation parameters of 12 intact 8- to 9-month-old Santa Inês (averaging 24.95 ± 1.8 kg body weight) ewes fed tropical grass hay supplemented with macrotiloma hay. The ewes were divided into two treatment groups depending on their diet: chopped aruana grass hay (Panicum maximum cv. Aruana) (control-CON); and aruana grass hay supplemented with chopped macrotiloma hay (macrotiloma-MAC). The animals were kept for 5 consecutive days in metabolic cages for the ATTD assay, and at the end of this period, samples of rumen fluid were collected from each ewe to determine ammoniacal nitrogen (NH3 -N) and short-chain fatty acid (SCFA) production, and protozoa count. For the in vitro assay, a decrease in total gas and CH4 production was observed for samples incubated without PEG (p < .05). No differences were observed for the other parameters evaluated (p > .05). In the in vivo experiment, increased intake and ATTD of crude protein were observed for the animals fed MAC when compared to CON (p < .05). For rumen fermentation parameters, increased NH3 -N, total SCFA and isobutyrate concentrations, as well as reduced protozoa count were observed for MAC when compared to CON (p < .05). The results observed here indicated the potential of macrotiloma for use as a ruminant feed, and antimethanogenic potential of this plant was noted.

Adult foretarsi of Heliconius erato Linnaeus (Lepidoptera, Nymphalidae) are reduced in size and are not used for walking. Foretarsi of the female have specialized sensilla that are presumably used to identify the host plant, by drumming. The mid- and hind tarsi also bear sensilla in both sexes, but these have not been described in detail, nor has their chemosensory function been determined. We described and compared the tarsi of H. erato under light and scanning electron microscopy. Behavioral experiments showed that differences in the shape, number, and size of sensilla were related to feeding and oviposition behaviors. Two types of sensillum (chaeticum and trichodeum) were found in similar numbers and size on the mid- and hind tarsi of both sexes. Sensilla on the female foretarsi act in host-plant site selection, strongly affecting oviposition rates when isolated. Male foretarsi lack sensilla, which may have been selected against due to the absence of function and thus lost. Sensilla on the mid- and hind tarsi are involved in sugar detection in both sexes, responding to an effective dose of sucrose (ED50) near 0.01 M, and therefore might be used to identify food resources when the butterflies settle on flowers.

Abstract Pollinators provide an essential service to natural ecosystems and agriculture. In tomatoes flowers, anthers are poricidal, pollen may drop from their pore when flowers are shaken by the wind. However, bees that vibrate these anthers increase pollen load on the stigma and in fruit production. The present study aimed to identify the pollinator richness of tomato flowers and investigate their morphological and functional traits related to the plant-pollinator interaction in plantations of Central Brazil. The time of anthesis, flower duration, and the number and viability of pollen grains and ovules were recorded. Floral visitors were observed and collected. Flower buds opened around 6h30 and closed around 18h00. They reopened on the following day at the same time in the morning, lasting on average 48 hours. The highest pollen availability occurred during the first hours of anthesis. Afterwards, the number of pollen grains declined, especially between 10h00 to 12h00, which is consistent with the pollinator visitation pattern. Forty bee species were found in the tomato fields, 30 of which were considered pollinators. We found that during the flowering period, plants offered an enormous amount of pollen to their visitors. These may explain the high richness and amount of bees that visit the tomato flowers in the study areas. The period of pollen availability and depletion throughout the day overlapped with the bees foraging period, suggesting that bees are highly effective in removing pollen grains from anthers. Many of these grains probably land on the stigma of the same flower, leading to self-pollination and subsequent fruit development. Native bees (Exomalopsis spp.) are effective pollinators of tomato flowers and are likely to contribute to increasing crop productivity. Resumo Polinizadores fornecer um serviço essencial para os ecossistemas naturais e para agricultura. Em tomateiros, as anteras são poricidas e o pólen pode sair a partir dos poros quando as flores são agitadas pelo vento. No entanto, as abelhas que vibram as anteras aumentam a carga de pólen no estigma e na produção de frutos. O presente estudo teve como objetivo identificar a riqueza dos polinizadores das flores de tomate e investigar suas características morfológicas e funcionais relacionadas com a interação planta-polinizador em plantações do Brasil central. Foram registrados o tempo de antese, duração flor, bem como o número e viabilidade de grãos de pólen e óvulos. Os visitantes florais foram observados e coletados. Os botões florais abriram-se em torno 06h30 e fechou em torno de 18h00. As flores reabrem no dia seguinte ao mesmo tempo na parte da manhã, com longevidade média de 48 horas. A maior disponibilidade de pólen ocorreu durante as primeiras horas da antese. Depois disso, o número de grãos de pólen diminuiu, especialmente entre as 10h00 às 12h00, o que é consistente com os padrões de visitação de polinizadores. Quarenta espécies de abelhas foram encontradas nos campos de tomate, 30 das quais foram consideradas polinizadores. Durante o período de floração, as plantas oferecem enorme quantidade de pólen para os seus visitantes. Isto pode explicar a alta riqueza e quantidade de abelhas que visitam as flores de tomate nas áreas de estudo. O período de disponibilidade e redução de pólen durante todo o período do dia é sobreposto com o período de alimentação das abelhas, o que sugere que elas são altamente eficazes na remoção de grãos de pólen das anteras. Muitos desses grãos provavelmente são depositados no estigma da mesma flor, levando à auto-polinização e o desenvolvimento de frutos. Abelhas nativas (Exomalopsis spp.) são polinizadores efetivos de flores de tomate, podendo contribuir para o aumento da produtividade das culturas. Por outro lado, as flores de tomate oferecem grandes quantidades de pólen de recursos para uma alta riqueza e quantidade de abelhas, que mostram um estudo forte interação planta-polinizador nos agroecossistemas.

PURPOSE: Non-alcoholic fatty liver disease (NAFLD) is an insidious pathologic condition that can manifest from simple steatosis to steatohepatitis (NASH) with potential progression to cirrhosis. Like the polycystic ovary syndrome (PCOS), NAFLD is associated with obesity, diabetes mellitus, insulin resistance and metabolic syndrome. PCOS women have an increased risk of NAFLD, but it is debatable which features of PCOS, either specific (androgen excess) or unspecific (metabolic derangements) affect the NAFLD risk. METHODS: We performed a systematic review and meta-analysis of studies that addressed the association of PCOS and NAFLD. We selected 17 studies published between 2007 and 2017 that included 2734 PCOS patients and 2561 controls of similar age and body mass index (BMI). RESULTS: PCOS patients have increased prevalence of NAFLD (odds ratio 2.54, 95% confidence interval 2.19-2.95). PCOS women with hyperandrogenism (classic phenotype) have a higher prevalence of NAFLD compared to women with PCOS without hyperandrogenism, even after correction for confounding variables. Among women with PCOS, those with NAFLD have higher serum total testosterone (mean difference 0.40 nmol/L, 95% CI 0.29-0.50 nmol/L) and free androgen index (mean difference 4.46, 95% CI 3.53-5.39) than those without NAFLD. The studies that used multivariate analysis controlling for age, BMI, triglycerides, and insulin resistance index confirmed that serum androgens are independent predictors of NAFLD in women with PCOS. CONCLUSION: The prevalence of NAFLD is increased in women with PCOS and the presence of NAFLD is associated with high serum androgen levels, in addition to obesity and insulin resistance.

Afterwards, the number of pollen grains declined, especially between 10h00 to 12h00, which is consistent with the pollinator visitation pattern. Forty bee species were found in the tomato fields, 30 of which were considered pollinators. We found that during the flowering period, plants offered an enormous amount of pollen to their visitors. These may explain the high richness and amount of bees that visit the tomato flowers in the study areas. The period of pollen availability and depletion throughout the day overlapped with the bees foraging period, suggesting that bees are highly effective in removing pollen grains from anthers. Many of these grains probably land on the stigma of the same flower, leading to self-pollination and subsequent fruit development. Native bees (Exomalopsis spp.) are effective pollinators of tomato flowers and are likely to contribute to increasing crop productivity. The present study investigated the effect of birthweight on testicular development and spermatogenesis in boars. Twenty-four pairs of littermate boars were selected: one piglet with the highest birthweight (HW) and the other with the lowest birthweight (LW) within the litter. Two subsets of 12 pairs of male littermates from each birthweight group were obtained after selection: one subset was orchiectomised at 8 days and the other at 8 months of age. HW boars had higher body and testicular weights at both ages (P<0.05). Testosterone concentrations and the relative expression of 17α-hydroxylase in the testis were similar between birthweight groups. Birthweight affected somatic and germ cell numbers in the neonatal testis, which were higher in HW boars (P<0.05). Moreover, a significant reduction in the number of pachytene spermatocytes and round spermatids was observed in LW boars (P<0.05) at 8 months of age, which caused a decrease in the total number of elongated spermatids and daily sperm production (P<0.05). Hence, HW boars have the potential to produce more spermatozoa and consequently more semen doses per ejaculate, and would be very valuable to an industry that relies on AI.

The objective of this study was to evaluate differential expression of innate and adaptive immune genes, including immunoglobulin, immune cell receptor, cytokine, inflammatory protein, toll-like receptors (TLR) and recombination-activating gene (RAG) in skin from channel catfish, Ictalurus punctatus after immunization with live theronts of Ichthyophthirius multifiliis (Ich) by intraperitoneal injection. The immunized catfish showed significantly higher survival rate (95%) than those of mock-immunized control fish (0% survival) after the theront challenge. The gene expression of innate immune system, such as cytokines (IL-1ß type a, IL-1ß type b, IFN-γ, TGF1-ß and TNF-α) and inflammatory proteins (NF-kB and iNOS 2), showed significant upregulation at day 1 (D1) post-immunization. Expression of TLR genes exhibited a rapid increase from hour 4 (h4) to D10 post-immunization. Genes of the adaptive response, such as the cell receptor MHC I, CD8+ , CD4+ and TCR-α, showed upregulation at D1, D6 and D10. The TCR-ß expression increased rapidly at h4 and remained upregulated until D10. Immunoglobulin IgM upregulation was detected from h4 until D2 while IgD expression was increased from D1 until D10. Rapid upregulation of innate and adaptive immune genes in skin of catfish following live theront vaccination was demonstrated in this study ultimately resulting in significant protection against Ich infection.

Crespa goats are phenotypically similar to the Angora breed, and are traditionally reared in small, low-tech farms in southernmost Brazil. Whether they represent degenerated remnants of pure Angora goats or result from foreign breeds introduced during colonial times and recently mixed with commercial breeds is unknown. Since the degree of relatedness of Crespa in relation to other goats is completely unknown, we performed a comparative assessment of the genetic similarity between Crespa and foreign commercial breeds reared in the region (Angora, Alpine, Anglo-Nubian, Boer, and Saanen), particularly the Angora. We used 11 microsatellites to score alleles in 148 individuals and performed a Bayesian assignment test, which revealed six clusters (K = 6; Ln likelihood = -5047.6). In addition, a segment of the mitochondrial DNA (mtDNA) control region was sequenced to investigate the relatedness of Crespa goats to Portuguese autochthonous breeds (Algarvia, Bravia, Charnequeira, Serpentina, and Serrana). The origin of the Crespa breed could not be ascertained from the mtDNA, but it does not only descend from the Angora. It is probably related to other introduced and autochthonous Portuguese breeds, in particular the Algarvia. Therefore, our results indicate that this distinctive source of genetic diversity is partly a remnant of animals that were introduced during the colonial period. By recognizing it as genetically distinct, we provide further support for the protection of this particular gene pool.

RESUMO

The little-known Neotropical tortricid moth Lasiothyris luminosa (Razowski & Becker) is reported for the first time as a grapevine pest in northeastern Brazil. A diagnosis based on morphological characters including the genitalia is provided for both sexes, together with a preliminary description of economic damage caused by the larval stage on flowers and fruits. In addition, we sequenced a partial region of the cytochrome oxidase subunit I gene from specimens from the region and reconstructed phylogenetic relationships using representative lineages of closely related tortricids as terminals.
