RESUMO

The aim of this study was to perform a proteomic analysis to isolate and identify proteins from the swine sperm nuclear matrix to contribute to a database of swine sperm nuclear proteins. We used pre-chilled diluted semen from seven boars (19 to 24 week old) from the commercial lineLandrace x Large Whitex Pietran. The semen was processed to separate the sperm heads and extract the chromatin and nuclear matrix for protein quantification and analysis by mass spectrometry, by LTQ Orbitrap ELITE Mass spectrometer (Thermo-Finnigan) coupled to a nanoflow chromatography system (LC-MS/MS). We identified 222 different proteins in the sample; a total of 159 (71.6%) were previously described as present in the somatic or sperm nuclei of other species, 41 (18.5%) did not have a previously reported nuclear presence and 22(9.9%) had not been characterized. The most abundant family of proteins corresponded to ribosomal (13.1%), followed by cytoskeleton (12.2%), uncharacterized (9.9%), histones (5.4%), proteasome subunits (3.6%)and heat shock (1.8%). The other proteins clustered in other families accounted for 54% of the total proteins. The protein isolation of the nuclear matrix of the swine spermatozoa was satisfactory, thus demonstrating that the protocol used was efficient. Several proteins were identified and described. However, it was not possible to identify some protein structures. Therefore,this study helps to establish a starting point for future proteomic studies comparing fertile and sub-fertile animals.

RESUMO

Com o intuito de avaliar os fatores etários e sexuais sobre as concentrações séricas de lipídios e enzimas avaliadoras da função hepática, foram coletadas amostras de sangue de 34 bezerros mestiços (machos e fêmeas) em intervalos mensais, da semana do nascimento até seis meses de idade. Foram encontrados os seguintes valores: CT 86,20 mg/dL, HDL-C 29,28 mg/dL, VLDL-C 5,61mg/dL, LDL-C 51,31mg/dL, TG 28,07mg/dL, AST 74,43U/L, ALT 24,18U/L, FAL 152,67U/L e GGT 111,10U/L. O fator etário influenciou no CT, HDL-C, LDL-C, ALT e GGT. Verificou-se também uma correlação positiva e significativa entre CT e HDL-C, CT e LDL-C e entre VLDL-C e TG. A partir dos resultados obtidos, tais achados poderão contribuir para futuros estudos nesta população de animais.(AU)


Aiming to evaluate the age and sex factors on serum lipids and enzymes evaluators of liver function, blood samples were collected from 34 crossbread calves (males and females) at monthly intervals, week of birth to six months of age. Were found the following valeus: CT 86.20 mg/dL, HDL-C 29.28 mg/dL, VLDL-C 5.61 mg/dL, LDL-C 51.31 mg/dL, TG 28.07 mg/dL, AST 74.43 U/L, ALT 24.18 U/L, ALP 152.67 U/L and GGT 111.10 U/L. The age factor showed difference, and influence can be seen on CT, HDL-C, LDL-C, ALT, FAL and GGT. There was also a significant positive correlation between CT and HDL-C, TC and LDL-C and VLDL-C and TG. From the results obtained, these findings may contribute to future studies in this population of animals.(AU)

Todo movimento é um evento mecânico que implica em uma força sobre a coluna. Sempre que tal força exceder a tolerância da coluna lesões irão ocorrer, podendo ser de diferentes magnitudes. Foram analisadas radiografias de 33 cães com suspeita clínica de alteração vertebral, atendidos no Hospital Veterinário da Universidade Federal de Uberlândia entre janeiro de 2007 e setembro de 2010. Notou-se que 20 animais (60,60 %) apresentaram alterações no perfil anátomo-funcional da coluna vertebral (colunas em cifose, lordose ou excessivamente retilíneas). Com relação à presença de lesões, foi verificado que 30 cães (90,90%) tiveram diagnóstico positivo para algum processo patológico. Os resultados não apresentaram diferença (p > 0,05). Concluiu-se que alterações morfológicas da coluna vertebral são contundentes para o desenvolvimento de distúrbios biomecânicos e posteriores manifestações clínicas, pois levam a um aumento na magnitude, estresse e freqüência de cargas aplicadas, com consequente comprometimento na síntese dos tecidos, sendo associadas às mudanças nas estruturas dos discos intervertebrais.(AU)


Every movement is a mechanical event that requires a force on the spine. Whenever this force exceeds the tolerance of the spine, injuries will occur, and may be of different magnitudes. From it were analyzed radiographs of 33 dogs with clinical suspicion of vertebral change, consulted in Veterinary Hospital of the Federal University of Uberlândia between january 2007 and september 2010. It was noted that 20 animals (60.60%) showed changes in anatomical and functional profile of the spine (columns in kyphosis, lordosis or too straight). With respect to lesions, it was found that 30 dogs (90.90%) had a positive diagnosis for any disease process. These results showed no differences (p > 0.05). Therefore, the morphology of the spine are remarkable for the development of biomechanical changes and subsequent clinical manifestations, since they lead to an increase in the magnitude, frequency and stress of applied loads, with consequent changes in tissues synthesis, being associated with changes in structures of intervertebral discs.(AU)
