RESUMO

Rumen-protected choline (RPC) promotes benefits in milk production, immunity, and health in dairy cows by optimizing lipid metabolism during transition period management and early lactation. However, the RPC success in dairy cows depends on choline bioavailability, which is affected by the type of protection used in rumen-protected choline. Therefore, our objectives were to determine the effects of a novel RPC on dry matter intake (DMI), identify markers of metabolism and immunity, and evaluate lactation performance. Dry Holstein (n = 48) cows at 245 ± 3 d of gestation were blocked by parity and assigned to control or RPC treatment within each block. Cows enrolled in the RPC treatment received 15 g/d of CholiGEM (Kemin Industries, Cavriago RE, Italy) from 21 d prepartum and 30 g/d of CholiGEM from calving to 21 d postpartum. During the transition period, DMI was measured daily, and blood was sampled weekly for energy-related metabolites such as ß-hydroxybutyrate (BHB), glucose, and nonesterified fatty acids (NEFA), as well as immune function markers such as haptoglobin (Hp) and lipopolysaccharide-binding protein (LPB). Vaginal discharge samples were collected at the calving and 7 d postpartum and stored in microcentrifuge tubes at -80°C until 16S rRNA sequencing. The main responses of body condition score, body weight, DMI, milk yield, milk components, and immune function markers were analyzed using the GLIMMIX procedure of SAS with the effects of treatment, time, parity, and relevant covariates added to the models. The relative abundance of microbiome α-diversity was evaluated by 3 indexes (Chao1, Shannon, and Simpson) and ß-diversity by principal coordinate analysis and permutational multivariate ANOVA. We found no differences in DMI in the pre- and postpartum periods. Cows fed RPC increased the yields of energy- and 3.5% fat-corrected milk and fat yield in primiparous and multiparous cows, with an interaction between treatment and parity for these lactation variables. However, we found no differences in milk protein and lactose up to 150 DIM between treatments. Glucose, NEFA, and BHB had no differences between the treatments. However, RPC decreased BHB numerically (control = 1.07 ± 0.13 vs. RPC = 0.63 ± 0.13) in multiparous on the third week postpartum and tended to reduce the incidence of subclinical ketosis (12.7% vs. 4.2%). No effects for Hp and LPB were found in cows fed RPC. Chao1, Shannon, and Simpson indexes were lower at calving in the RPC treatment than in the Control. However, no differences were found 7 d later for Chao1, Shannon, and Simpson indexes. The vaginal discharge microbiome was altered in cows fed RPC at 7 d postpartum. Fusobacterium, a common pathogen associated with metritis, was reduced in cows fed RPC. Rumen-protected choline enhanced lactation performance and health and altered the vaginal discharge microbiome which is a potential proxy for uterine healthy in dairy cows. The current study's findings corroborate that RPC is a tool to support adaptation to lactation and shed light on opportunities for further research in reproductive health.

RESUMO

The present study aimed to evaluate and compare the palmar and plantar digital arteries of 10 horses and 10 mules through B-mode and spectral Doppler ultrasound. Likewise, compare the plantar arteries of the pelvic limbs with the palmar arteries of the thoracic limbs, in addition to verifying the differences of the digital arteries in the contralateral limbs (right and left) in horses and mules using B-mode and spectral Doppler ultrasonography. The evaluated parameters were the diameter and the intima-media thickness (IMT), resistivity index (RI), pulsatility index (PI), peak of systolic velocity (pSV), final diastolic velocity (fDV) and mean velocity (MV) of the lateral and medial palmar and plantar arteries digital in healthy horses and mules. The horses presented higher diameters values, IMT, pSV, fDV and MV in comparison to the mules. The variables RI and PI also presented differences between horses and mules. Also, both species showed higher values in the variables of B-mode and Doppler ultrasonography in the thoracic limbs. Differences were also detected in the comparison between the limb sides (right and left) in the two modalities. The B-Mode and spectral Doppler ultrasound techniques are viable tools to identify differences in the palmar and plantar digital arteries in healthy horses and mules, highlight the differences between the thoracic and pelvic limbs and their respective sides (right and left) as well.

Seven forelimb hooves from healthy horses were submitted to regular trimming and fourteen days later, submitted to toe trimming. Toe angle and length, lateral and medial heels, frog and sole length and width, distal, proximal, and metacarpal phalangeal interphalangeal angles were measured, as well as locomotion evaluation through cinematographic analysis. The measurements were performed ten, 14, 15, 30 and 45 days after the regular trimming, and at 14 days two measurements, one before and one after the toe trimming, were carried out. For cinematography, the sequence of supports and time spent in each support were taken. The comparison of the means obtained from the individuals regarding the variables, between limbs, limbs for each individual, time - points, moments for each individual and between limbs for each moment, as well as the time spent in each in the supports, was performed using ANOVA. Results lower than those of statistical significance (p<0.05) were submitted to Tukey's test. The toe trimming promotes changes in toe length, angle, lateral heel angle, medial and lateral heel length, frog length, width, and sole length, and changes the pattern of the trot of horses.

The aim of this study was to evaluate the effects of Psyllium (PSY) and Carboxymethylcellulose (CMC) administration on fecal elimination of sand in horses with asymptomatic sand accumulations. Eight horses were selected from sandy areas and randomly divided into 2 groups of four animals. The subjects were treated either with CMC or PSY. The presence of intestinal sand was confirmed through radiography and glove sedimentation test. The study was performed in two phases, with a 7-day interval. In phase I, all the animals received 8 liters of warm water; in phase II, the CMC group received 8 liters of water + 1g/kg of CMC, whereas the PSY group received 8 liters of water + 1g/kg of PSY. All administrations were performed through nasogastric intubation and fractionated in 2 equal volume administrations with an interval of two hours. General and specific physical examination of the digestive system were performed in conjunction with abdominal ultrasonography before the administrations and after 6, 12, 24, 36 and 48 hours, aiming to evaluate intestinal motility and presence of sand. All the feces eliminated by the animals within the 72 hours following the administrations were quantified, diluted and sedimented in order to calculate the sand output (g/kg of feces). All the animals were also subjected to radiographic examination to quantify sand accumulation prior to phase I and after 72 hours of phases I and II. No adverse effects were observed after the treatments. It was possible to notice higher sand elimination in both groups during the phase I, whereas no difference was observed in sand elimination rates between the groups in phase II. The radiographic scores presented differences between the initial timepoint and 72h in phases I and II for both groups. Based on the sand elimination rates and radiographic score, this study demonstrated that sand output was greater after administration of water alone, compared to CMC and Psyllium, leading to the inference that removal of the sandy environment and prevention of sand re-ingestion are effective measures for the elimination of sand from the colon of horses with asymptomatic sand accumulations.(AU)

The IR, IP, VS and VM differed between horses and mules, being higher in the horses, but the VD was higher in mules. No difference in the variables in B-mode and Doppler between gender were observed in horses, different from mules, in which the diameters, IR and IP values were higher in males and the VS and VM was higher in females. Body mass did not influence B-Mode, both for horses and mules. A significant correlation was observed for Doppler. The common carotid arteries of horses and mules are different by ultrasound scans B-mode and spectral Doppler. Gender does not influence the B-Mode and Doppler variables in horses, but can in mules. The body mass of horses and mules, regardless of gender, is not associated with B-mode variables, but with Doppler variables.(AU)

RESUMO

The objective of this study was to investigate the associations between natural antibodies (NAbs) and total serum immunoglobulins (Igs) concentrations (both from IgG and IgM isotypes), with variables of polymorphonuclear leukocytes (PMN) function of dairy cows in the early postpartum period. Seventy-five healthy postpartum dairy cows at 2 ± 1 days in milk were enrolled in a cross-sectional study. Natural and total antibodies in serum samples were measured by ELISA. Flow cytometry was used to determine the phagocytosis and oxidative burst capacities of PMN and the quantification of the adhesion molecule l-selectin. Leukocyte count and differentials, and serum haptoglobin were also measured. A positive correlation between NAbsIgM and total serum IgM and between NAbsIgM and NAbsIgG were found. Before performing the associations between circulating Igs concentrations and PMN function variables, cows were categorized into having low, medium or high circulating antibodies based on their NAbs and total Igs serum concentrations. None of the PMN variables assessed differed between low, medium, and high cows for both NAbsIgM and NAbsIgG. While associations between PMNs function and total IgG were not observed, some associations between total IgM concentrations and PMN activity were found. Cows with high serum IgM had greater phagocytic activity compared to cows in the low IgM group. Finally, the proportion of PMN that performed oxidative burst and PMN surface expression of l-selectin intensity was greater in high total IgM group when compared to medium and low groups. In conclusion, association between NAbs, and PMN activity variables were not observed, but total serum IgM was associated to some PMN function variables in early post-partum dairy cows.

RESUMO

A 15-year-old, mixed breed, male horse was attended with a history of multiple abscesses in the cervical region with a three-year evolution. Upon admission, three fistulous tracts with drainage of purulent secretions in the cervical region, low body score, restriction of cervical movements, and painful sensitivity to palpation were observed. The horse was diagnosed with osteomyelitis secondary to Streptococcus equi infection. The initial treatment was antibiotic therapy and local curative. Owing to the lack of response, surgical debridement was performed. An initial favorable response was observed; however, after 4 months, drainage recurred, and the animal was euthanized. A post-mortem computed tomography scan was performed to obtain details of the injury. Cervical osteomyelitis is rare, and its occurrence through hematogenous spread in adult horses and the tomographic findings had not been reported previously. The long period of evolution, difficulty in performing an aggressive debridement, and the presence of multi-drug resistant bacteria contributed to the negative outcome.(AU)

Dyslipidemia has been described in sickle cell anemia (SCA) but its association with increased disease severity is unknown. Here, we examined 55 children and adolescents with SCA as well as 41 healthy controls to test the association between the lipid profiles in peripheral blood and markers of hemolysis, inflammation, endothelial function, and SCA-related clinical outcomes. SCA patients exhibited lower levels of total cholesterol (P<0.001), low-density lipoprotein cholesterol (LDL-c) (P<0.001), and high-density lipoprotein cholesterol (HDL-c) (P<0.001), while displaying higher triglyceride (TG) levels and TG/HDL-c ratio values (P<0.001). TG/HDL-c values were positively correlated with lactate dehydrogenase (P=0.047), leukocyte count (P=0.006), and blood flow velocity in the right (P=0.02) and left (P=0.05) cerebral artery, while being negatively correlated with hemoglobin levels (P<0.04). Acute chest syndrome (ACS) and vaso-occlusive events (VOE) were more frequent in SCA patients exhibiting higher TG/HDL-c values (odds ratio: 3.77, P=0.027). Multivariate logistic regression analysis confirmed independent associations between elevated TG/HDL-c values and SCA. Thus, children and adolescents with SCA exhibited a lipid profile associated with hemolysis and inflammatory parameters, with increased risk of ACS and VOE. TG/HDL-c is a potential biomarker of severity of disease.

RESUMO

Background Complex percutaneous coronary intervention (PCI) is associated with higher ischemic risk, which can be mitigated by long-term dual antiplatelet therapy (DAPT). However, concomitant high bleeding risk (HBR) may be present, making it unclear whether short- or long-term DAPT should be prioritized. Objectives This study investigated the effects of ischemic (by PCI complexity) and bleeding (by PRECISE-DAPT [PRE dicting bleeding Complications in patients undergoing stent Implantation and Sub sequent Dual Anti Platelet Therapy] score) risks on clinical outcomes and on the impact of DAPT duration after coronary stenting. Methods Complex PCI was defined as ≥3 stents implanted and/or ≥3 lesions treated, bifurcation stenting and/or stent length >60 mm, and/or chronic total occlusion revascularization. Ischemic and bleeding outcomes in high (≥25) or non-high (<25) PRECISE-DAPT strata were evaluated based on randomly allocated duration of DAPT. Results Among 14,963 patients from 8 randomized trials, 3,118 underwent complex PCI and experienced a higher rate of ischemic, but not bleeding, events. Long-term DAPT in non-HBR patients reduced ischemic events in both complex (absolute risk difference: −3.86%; 95% confidence interval: −7.71 to +0.06) and noncomplex PCI strata (absolute risk difference: −1.14%; 95% confidence interval: −2.26 to −0.02), but not among HBR patients, regardless of complex PCI features. The bleeding risk according to the Thrombolysis In Myocardial Infarction scale was increased by long-term DAPT only in HBR patients, regardless of PCI complexity. Conclusions Patients who underwent complex PCI had a higher risk of ischemic events, but benefitted from long-term DAPT only if HBR features were not present. These data suggested that when concordant, bleeding, more than ischemic risk, should inform decision-making on the duration of DAPT. (AU)

RESUMO

Malassezia pachydermatis is an important opportunistic agent of dermatitis and otitis in dogs. M. pachydermatis is generally treated with topical therapies using combinations of antifungal, antimicrobial and anti-inflammatory agents. We investigated the in vitro activities of carvacrol (CRV), cinnamaldehyde (CIN) and thymol (THY) alone and in combination with antifungal agents (fluconazole, itraconazole, ketoconazole, clotrimazole, miconazole, terbinafine and nystatin) against M. pachydermatis. The assays were performed according to the Clinical and Laboratory Standards Institute (CLSI), using Sabouraud dextrose broth and checkerboard microdilution. The mean fractional inhibitory concentration index (FICI) showed primary synergies for the combinations carvacrol+nystatin, thymol+nystatin, and carvacrol+miconazole (80%). In conclusion, the results obtained indicate that the phytochemicals tested showed relevant in vitro anti-M. pachydermatis activity. Future in vivo experiments are needed to elucidate the safety and therapeutic potential of these combinations.

RESUMO

RESUMO

Polycationic polymers and DNA form soluble complexes in aqueous solution, which allows the transfer of genetic material into cells. Therefore, these chemically-modified polymers are of interest for use in studies aimed at better transfection efficiency and gene expression along with reduced cytotoxicity. In this study, branched poly(ethylene imine) (PEI) was modified by alkylation with n-alkyl groups (n = 4, 6, 8 and 12 carbons). The polyplexes formed through interaction of the modified PEIs and calf thymus DNA (ctDNA) were investigated using UV-Vis spectrophotometry, ethidium bromide fluorescence emission, circular dichroism spectroscopy, dynamic light scattering, and small-angle X-ray scattering techniques along with the determination of the zeta potential and viscosity. According to the results obtained, the formation of ctDNA-PEI polyplexes occurs in three steps. Firstly, when a small amount of polyelectrolyte is present the ctDNA chains are partially compacted. Subsequently, with the addition of more polyelectrolyte, the complexes have a null charge density and micrometric size. Lastly, with a higher concentration of PEI, the ctDNA is fully compacted by the PEI chains, leading to positively charged complexes with Rh values in the range of 52.0-86.0 nm. The viscosity and SAXS analysis suggested that the unmodified PEI exhibits the strongest interaction and promotes the best ctDNA condensation.

RESUMO

The environmental conditions which contributed to the formation of the notorious quaternary deposits of freshwater sponge spicules in karstic lentic environments in Brazil have been subject of some speculation. No investigation has yet been conducted to test whether these deposits currently originate in karstic lakes. To provide for such an investigation, Serra Negra Lake, which is formed on an ultramafic-alkaline-carbonatite dome at central western Brazil, close to the area of occurrence of the paleo-deposits was selected for the study. Bottom sediments were sampled at 10 stations across the lake, and water was sampled at five of the stations, in June/2011 (rainy season) and October/2011 (dry season). Analysis of granulometry, organic matter and presence of spicules were carried out in the sediments. Lake water was analysed for the main physical and chemical characteristics. Deposit of spicules was restricted to the northern area of the lake, which is rich in macrophyte. The taxonomic analysis of the spicules indicated the contribution of five sponge species, Dosilia pydanieli, Metania spinata, Radiospongilla amazonensis, Trochospongilla variabilis and Heterorotula fistula, which formed large deposits in neighbouring areas. The high silica concentration, derived from the dome volcanic rocks, constant water level and available substrate are credited for the continuous production of sponges and spicules, confirmed by the rare presence of gemmoscleres. The lake is classed as a minerotrophic fen type of bog with a heavy contribution from the surrounding creeks. Lake sediments are fine with high levels of organic matter and peat, which contributed to the trapping of spicules in the sediments.

In modern society the work and athletic performance of horses has led to a very important animal production sector in which Brazil possesses the third largest horse stock. Among all equine lesions described, metacarpophalangeal (fetlock) joint lesions are considered one of the main causes of lameness. Consequently, there is a need to improve the understanding and diagnosis of these injuries. The most efficient imaging diagnostic methods for the fetlock region are computed tomography, radiography and ultrasound. Imaging studies of the anatomical structures involving this joint are extremely important to obtain a more precise diagnose. The present study was performed in order to evaluate the capacity of different imaging diagnostic modalities to detect a variety of lesions in different fetlock structures. Twenty horses (Equus caballus) used for horsemanship activities were referred to the Department of Animal Reproduction and Veterinary Radiology of São Paulo State University, Botucatu campus, with clinical signs of metacarpophalangeal joint injuries. Horses were submitted to radiographic and ultrasonographic exam and computed tomography scan. Image analysis revealed a significant capacity of these methods to characterize lesions in this region. However, computed tomography provided broader and better evaluation of lesions in bones and adjacent structures, because it allows the analysis to be performed on three-dimensional projections, with attenuation coefficients (window selections) and tissue density measurement through Hounsfield Units (HU).
