RESUMO

The spread of Zika virus (ZIKV) from the African continent to the Americas promoted its molecular evolution, as reflected by mutations in its RNA genome. Most of the ZIKV genome sequences in the GenBank database have incomplete 5' and 3' UTR sequences, reflecting the deficiency of whole-genome sequencing technologies to resolve the sequences of the genome ends. We modified a protocol for rapid amplification of cDNA ends (RACE) to determine the complete sequences of the 5' and 3' UTRs of a previously reported ZIKV isolate (GenBank no. MH544701.1). This strategy is useful for determining 5' and 3' UTR sequences of ZIKV isolates and will be useful for comparative genomics applications.

RESUMO

Ante la pandemia de Covid-19 causada por el vírus SARS-CoV-2 que puso a los trabajadores de la región lationamericana en una condición de mayor vulnerabildad, en este artículo se consideran algunas de las respuestas proporcionadas por la psicología convencional de corte industrial-organizacional, las cuales han estado caracterizadas por su enfasis técnico y acrítico dirigido a responsabilizar a los trabajadores de la actual situación. Tambien son analizadas las propuestas construidas por la psicología crítica descritas como soluciones colectivas que convocan instancias Estatales, organizacionales, sindicales y gremiales para diseñar intervencinoes que contribuyan a mitigar la situacion de mayor vulnerabilidad laboral. Por último, son consideradas las respuestas derivadas de perspectivas externas a la psicologia que examinan criticamente esta disciplina para mostrar las operaciones de gobierno que son posibles gracias a las intervenciones psicológicas referidas a la contencion de la pandemia. Se señala que la llamada crisis de la crítica estaría permeándose tanto en aquellas psicologías que asumen perspectivas no hegemónicas, como en la crítica misma de la psicología cuando consideran la situación actual.(AU)


Diante da pandemia da Covid-19 causada pelo vírus SARS-CoV-2, que colocou os trabalhadores da região latino-americana em uma condição mais vulnerável, neste artigo se considera algumas das respostas fornecidas pela psicologia convencional, que tem se caracterizado por sua ênfase técnica e acrítica no sentido de responsabilizar os trabalhadores pela situação atual. Também se analisam as propostas construídas pela psicologia crítica descritas como soluções coletivas que apelam às instâncias estatais, organizacionais, sindicais e grupais para conceber intervenções que ajudem a mitigar a situação de maior vulnerabilidade no trabalho. Por fim, são consideradas as respostas derivadas de perspectivas externas à psicologia que examinam criticamente essa disciplina para mostrar as operações de governo que são possíveis por meio de intervenções psicológicas relacionadas à contenção da pandemia. Ressalta-se que a chamada crise da crítica estaria permeando tanto aquelas psicologias que assumem perspectivas não hegemônicas, como a própria crítica da psicologia em si ao se considerar a situação atual.(AU)


Faced with the Covid-19 pandemic caused by the SARS-CoV-2 virus that put workers in the Latin American region in a more vulnerable condition, this article considers some of the responses provided by conventional industrial-organizational psychology, which have been characterized by their technical and uncritical emphasis aiming at holding workers accountable for the current situation. Also, the proposals constructed by critical psychology are analyzed to indicate that these collective solutions appeal to State, organizational, union and group instances to design interventions that contribute to mitigate the situation of greater vulnerability at work. Finally, the responses derived from perspectives external to psychology that critically examine this discipline are considered to show the government operations that are possible through psychological interventions related to the containment of the pandemic. It is pointed out how the so-called crisis of criticism is permeating those psychologies that assume non-hegemonic perspectives, as well as in the criticism of psychology itself when they consider the current situation.(AU)

OBJETIVO: Evaluar el efecto de la refrigeración en la apoptosis y la viabilidad del lipoaspirado en las primeras 2 horas después de la toma. MÉTODO: Se incluyeron 20 pacientes que fueron sometidas a una liposucción del abdomen por razones estéticas. Se obtuvieron 5 ml de grasa y se procesaron para su estudio. La viabilidad se determinó usando azul tripano. La apoptosis se determinó usando el ensayo TUNEL. RESULTADOS: Todas las pacientes eran mujeres, con una edad media de 36.5 años (rango: 21-67). Con respecto a la viabilidad, en el tiempo 0, en el grupo control fue del 59.08 ± 24% y en el grupo de refrigeración fue del 60.96 ± 22%; a los 60 minutos, los valores fueron del 50.82 ± 21% y el 55 ± 32.6%, respectivamente (p = 0.74); y a los 120 minutos, fueron del 42.69 ± 20.85% y el 50.33 ± 21%, respectivamente. En cuanto a la apoptosis, el porcentaje de células apoptóticas en el tiempo 0 fue del 37.87 ± 9.7% para el grupo de control y del 34.28 ± 9.74% para las muestras refrigeradas; a los 60 minutos, del 51.11 ± 8.64% y el 45.94 ± 9.15%, respectivamente; y a los 120 minutos, del 62.97 ± 13.33% y el 55.81 ± 9.45%, respectivamente. CONCLUSIONES: Refrigerar el lipoaspirado a 4 °C disminuyó la mortalidad y la apoptosis de los adipocitos en menos del 10% en las primeras 2 horas desde la toma.


OBJECTIVE: To evaluate the effect of refrigeration in the apoptosis and viability of the lipoaspirate in the first 2 h after harvesting. METHODS: 20 consecutive patients who underwent liposuction from the abdomen for esthetic reasons were included. 5 ml of fat were obtained and processed for study. The viability was obtained using trypan blue. Apoptosis was determined using TUNEL assay. RESULTS: All patients were female with a median age of 36.5 (21-67) years. On regard of the viability, at time 0, the viability in the control group was 59.08 ± 24% and 60.96 ± 22% in the refrigeration group. At 60 min, the values were 50.82 ± 21% versus 55 ± 32.6% (p = 0.74) and a 120 min, 42.69 ± 20.85% and 50.33 ± 21% respectively. On regard of apoptosis, the percentage of apoptotic cells at time 0 was 37.87 ± 9.7% for the control group and 34.28 ± 9.74% for refrigerated samples. At 60 min 51.11 ± 8.64% versus 45.94 ± 9.15% and at 120 min, 62.97 ± 13.33% versus 55.81 ± 9.45% respectively. CONCLUSIONS: Refrigerating the lipoaspirate at 4 °C decreased the mortality and apoptosis of the adipocytes in <10% within the first 2 h from harvesting.

Many secretory cells increase the synthesis and secretion of cargo proteins in response to specific stimuli. How cells couple increased cargo load with a coordinate rise in secretory capacity to ensure efficient transport is not well understood. We used thyroid cells stimulated with thyrotropin (TSH) to demonstrate a coordinate increase in the production of thyroid-specific cargo proteins and ER-Golgi transport factors, and a parallel expansion of the Golgi complex. TSH also increased expression of the CREB3L1 transcription factor, which alone caused amplified transport factor levels and Golgi enlargement. Furthermore, CREB3L1 potentiated the TSH-induced increase in Golgi volume. A dominant-negative CREB3L1 construct hampered the ability of TSH to induce Golgi expansion, implying that this transcription factor contributes to Golgi expansion. Our findings support a model in which CREB3L1 acts as a downstream effector of TSH to regulate the expression of cargo proteins, and simultaneously increases the synthesis of transport factors and the expansion of the Golgi to synchronize the rise in cargo load with the amplified capacity of the secretory pathway.

Resumen Las herramientas propuestas en los marcos poscoloniales y decoloniales no han sido de mayor interés para los investigadores que se ocupan de la interrelación entre el conocimiento psicológico y el mundo laboral. Entonces, los estudios críticos han dejado de lado las implicaciones que tiene este conocimiento cuando "viaja" más allá de las fronteras de los países en donde se produce la Psicología. En este artículo se examinan algunas "dimensiones coloniales" que emergen de las articulaciones del "complejo psi" cuando se vincula con el mundo del trabajo en lugares en donde no se producen los objetos psicológicos, sino en donde estos son apropiados, replicados o hibridados para su uso cotidiano en los ámbitos laborales.


Abstract The tools proposed in the postcolonial and de-colonial frameworks have not been of interest to researchers preoccupied with the interrelation between the psychological knowledge and the world of work. Thus, critical studies have left aside the implications that this knowledge has when it goes beyond the borders in which Psychology is produced. This article examines some "colonial dimensions" that emerged from the articulations of the "psy complex" in places that do not produce psychological objects, but which are appropriate, replicated and hybridized for being used in workplaces.

The GTPase Rab1b is involved in ER to Golgi transport, with multiple Rab1b effectors (located at ERES, VTCs and the Golgi complex) being required for its function. In this study, we performed live-cell dual-expression studies to analyze the dynamics of Rab1b and some effectors located at the ERES-Golgi interface. Rab1b occupied widely distributed mobile punctate and tubular structures, displaying a transient overlaps with its effectors and showing that these overlaps occurred at the same time in spatially distinct steps of ER to Golgi transport. In addition, we assessed Rab1b dynamics during cargo sorting by analyzing the concentration at ERES of a Golgi protein (SialT2-CFP) during Brefeldin A washout (BFA WO). Rab1b was associated to most of the ERES structures, but at different times during BFA WO, and recurrently SialT2-CFP was sorted in the ERES-Rab1b positive structures. Furthermore, we reveal for first time that Rab1b localization time at ERES depended on GBF1, a Rab1b effector that acts as the guanine nucleotide exchange factor of Arf1, and that Rab1b membrane association/dissociation dynamics at ERES was dependent on the GBF1 membrane association and activity, which strongly suggests that GBF1 activity modulates Rab1b membrane cycling dynamic.

En este artículo se considera la re-organización del "complejo psi", en relación a las transformaciones que ocurren contemporáneamente en el mundo del trabajo. Para esto se hace un recorrido examinando los cambios en el lugar y las operaciones de la psicología con respecto a las formas sociales empleo y post-empleo. Se argumenta que el papel de la psicología en relación a la producción de la subjetividad laboral se ha ido transformando en medio a los cambios que ávidamente han ido socavando lo que ahora se conoce como el "buen trabajo". Estas mutaciones se caracterizan por el aumento acelerado de incertidumbres relacionadas con los distintos aspectos de las condiciones laborales tales como, la contratación, los horarios, los espacios laborales, y las relaciones interpersonales. Se concluye que, en este contexto, la psicología juega un papel preponderante para la producción de los nuevos sujetos que requiere la organización capitalista del trabajo.


In this article the re-organization of the "psy complex" is considered as regards the contemporary transformations that are taking place in the world of work. Toward this end, a review of the changes in the place and operations of psychology with respect to the employment and post-employment social forms is conducted. It is argued that the role of psychology in the production of workers' subjectivity is transforming amidst the mutations that have been avidly debilitating what is at present known as "good work".These mutations are characterized by an increasing rate of incertitude in aspects such as work contracts, rosters, labor spaces, and interpersonal relationships. It is concluded that in this context, psychology plays a central role for the production of new subjects that the capitalist organization of work requires.


Neste artigo, a reorganização do "complexo psi" é considerada em relação às transformações que estão ocorrendo no mundo do trabalho contemporâneo. Para isso, examinam-se as mudanças ocorridas com o ambiente e as operações da psicologia em relação às formas sociais de emprego e pós-emprego. Argumenta-se que o papel da psicologia em relação à produção da subjetividade laboral modifica-se em meio às transformações que, avidamente, vêm debilitando o que agora se conhece como o "bom trabalho". Essas mutações são caracterizadas pelo aumento acelerado das incertezas relacionadas com diferentes aspectos das condições de trabalho, como contratação, horários, espaços de trabalho e relações interpessoais. Conclui-se que, nesse contexto, a psicologia desempenha papel importante para a produção de novos sujeitos requeridos pela organização capitalista do trabalho.

RESUMO

En este artículo, se presenta la construcción de un "museo de la psicología" como estrategia para la elaboración de la historia oral de la disciplina. En primer lugar, el debate entre psicología e historia constituye el marco para considerar el puesto que pueden ocupar los relatos de la experiencia de la práctica psicológica, en relación con la constitución de la historia de esta disciplina. A continuación, se describen los elementos fundamentales de la estrategia llamada "museo de la psicología", diseñada para elicitar historias orales. Para finalizar, a manera de ilustración, se consideran ventajas y limitaciones que emergen de la aplicación de la estrategia.


In this article the construction of a "museum of psychology" is presented as a strategy for the elaboration of the oral history of the discipline. First of all, the debate between history and psychology constitutes the framework to consider the place of psychological practices as regards the construction of the history of psychology. Then, the fundamental elements of the strategy called "the museum of psychology" are introduced. Finally, as an illustration, the advantages and the limitations of the strategy are presented.

RESUMO

En este artículo se consideran diversas maneras en las cuales se han formulado objeciones a la relación que la psicología ha establecido con el mundo del trabajo. Se revisan las principales corrientes críticas, para establecer el foco de análisis de sus propuestas y los objetivos que persiguen al examinar dicha relación. La discusión aporta elementos para constituir una actitud de balance que difiera de aquellas posiciones catastróficas que ven en la psicología un saber que promueve el control y la subordinación y aquellas posiciones optimistas que encuentran en este conocimiento estrategias para el progreso, el bienestar y la emancipación.


This article considers different ways in which have been formulated objections to the relationship established between psychology and the world of work. The main perspectives that propose critiques are revisited to establish the core analyses and their main objectives pursued. The discussion aims to contribute to the construction of an attitude of balance that differs from those catastrophic perspectives which evaluate psychology as a knowledge that promotes control and subordination and those optimistic positions which find in this knowledge some strategies for progress, wellbeing and emancipation.

Rab1b belongs to the Rab-GTPase family that regulates membrane trafficking and signal transduction systems able to control diverse cellular activities, including gene expression. Rab1b is essential for endoplasmic reticulum-Golgi transport. Although it is ubiquitously expressed, its mRNA levels vary among different tissues. This work aims to characterize the role of the high Rab1b levels detected in some secretory tissues. We report that, in HeLa cells, an increase in Rab1b levels induces changes in Golgi size and gene expression. Significantly, analyses applied to selected genes, KDELR3, GM130 (involved in membrane transport), and the proto-oncogene JUN, indicate that the Rab1b increase acts as a molecular switch to control the expression of these genes at the transcriptional level, resulting in changes at the protein level. These Rab1b-dependent changes require the activity of p38 mitogen-activated protein kinase and the cAMP-responsive element-binding protein consensus binding site in those target promoter regions. Moreover, our results reveal that, in a secretory thyroid cell line (FRTL5), Rab1b expression increases in response to thyroid-stimulating hormone (TSH). Additionally, changes in Rab1b expression in FRTL5 cells modify the specific TSH response. Our results show, for the first time, that changes in Rab1b levels modulate gene transcription and strongly suggest that a Rab1b increase is required to elicit a secretory response.

Progressive facial hemiatrophy, also known as Parry-Romberg syndrome, is a progressive and self-limited deformation of the subcutaneous tissue volume on one side of the face that creates craniofacial asymmetry. We present the case of a patient with a five-year history of progressive right facial hemiatrophy, who underwent facial volumetric restoration using cell-assisted lipotransfer (CAL), which consists of an autologous fat graft enriched with adipose-derived stem cells (ASCs) extracted from the same patient. ASCs have the capacity to differentiate into adipocytes. They also promote angiogenesis, release angiogenic growth factors, and some can survive as stem cells. The use of autologous fat as a filler in soft tissue atrophy has been satisfactory in patients with mild and moderate Parry-Romberg syndrome. Currently, CAL has showed promising results in the long term by decreasing the rate of fat reabsorption. The permanence and stability of the graft in all the injected areas has showed that autologous fat grafts enriched with stem cells could be a promising technique for the correction of defects caused by this syndrome.

Alrededor del mundo, las organizaciones del sector salud se han tenido que adaptar a las prescripciones nacionales e internacionales que regulan tanto el trabajo como la salud. Este proceso ha sido acompañado de la introducción, en las áreas de recursos humanos, de conocimientos y prácticas guiadas por los principios provenientes de las Ciencias Sociales y Administrativas. En este encuentro entre las reformas liberales y las maneras de administrar la fuerza laboral, juegan un papel fundamental las significaciones que circulan en las organizaciones, puesto que de ellas depende, en gran medida, el éxito de agendas, programas e intervenciones dirigidas a la administración de los trabajadores. En este artículo, se presentan significaciones que están circulando acerca de trabajo, trabajador, organización, gestión humana y contratación, en un área de recursos humanos, compuesta por catorce trabajadores, de un hospital universitario en la ciudad de Bogotá, Colombia. El diseño cualitativo utilizado buscó recoger tanto las significaciones circulantes como su correlato emocional.


Around the world, healthcare organizations have had to accommodate to national and international regulations governing work and health. This process has been accompanied by the introduction of knowledge and practices coming from social and administrative sciences. In this encounter between the liberal reforms and the particular ways of managing the workforce, the circulating meanings play an important role because, to a large extent, the agendas, programs and interventions geared to towards workforce management depend on these circulating meanings to succeed. In this paper, meanings about work, worker, organization, human resources management and hiring that are currently circulating in a human resources department of a university hospital in Bogotá, Colombia, are presented. The qualitative design used in this paper aimed to collect the circulating meanings as well as their emotional correlative elements.

In eukaryotic cells, proteins destined for secretion are translocated into the endoplasmic reticulum (ER) and packaged into so-called COPII-coated vesicles. In the ER exit sites (ERES), COPII has the capacity of deforming the lipid bilayer, where it modulates the selective sorting and concentration of cargo proteins. In this study, we analyze the involvement of Rab1b in COPII dynamics and function by expressing either the Rab1b negative-mutant (Rab1N121I) or the Rab1b GTP restricted mutant (Rab1Q67L), or performing short interference RNA-based knockdown. We show that Rab1b interacts with the COPII components Sec23, Sec24 and Sec31 and that Rab1b inhibition changes the COPII phenotype. FRAP assays reveal that Rab1b modulates COPII association/dissociation kinetics at the ERES interface. Furthermore, Rab1b inhibition delays cargo sorting at the ER exit sites. We postulate that Rab1b is a key regulatory component of COPII dynamics and function.

Rabs GTPases are key regulatory factors that specifically associate to organelles that integrate membrane transport pathways. Rabs, through their interactions with diverse effector proteins, regulate the formation, movement, tethering and fusion of transport carriers (vesicles and/or tubules). The mammalian Rab1b GTPase is required for ER to Golgi transport and interacts with multiple effectors localized at the ER-Golgi interface. Here, we focus on interactions between Rab1b and effectors that play essential roles in COPII and COPI vesicle formation/function. Based on evidence to date, we propose a model of Rab1b action at the ER exit sites.

Dumps are sites where the presence of high heavy metal (HM) concentration is a common occurrence, creating the need for implementing restoration processes immediately after their closure. In the 7.6 ha and 45 m high Morro de Moravia dump, arose from the disposal of Medellín solid wastes from 1974 to 1984, previous studies have demonstrated high contents of contaminants, including HM, prompting the need to identify effective mechanisms to implement its restoration. The objective of this study was to evaluate the adaptation, growth and phytoremediation capacity of Bidens pilosa, Lepidium virginicum, Brachiaria decumbens and Arachis pintoi. Content of HM (mg/kg) in Moravia residue matrix went from 17 to 8193 for Pb, 44 to 564 for Cr, 0.2 to 339 for Cd and 77 to 1679 for Ni. Measurements of plant cover, plant height and dry matter production at all plant species studied suggested adequate growth and adaptation to the Moravia dump conditions. Plant absorption of HM showed the pattern Cr > Cd > Ni > Pb. Estimated bioconcentration factors were generally low, and maximum values were 0.36 in A. pintoi (Cr), 2.96 in B. pilosa (Cd) and 0.26 in B. decumbens (Ni). However, our estimations of the phytoremediation potential of the assayed species, suggested they possess low remediation efficiency. Further investigation should be carried out in order to identify more efficient HM accumulators, and to test the use of technologies such as modification of pH, rhizoremediation or the use of genetically enhanced accumulators to increase HM availability to plants.


En los basureros se observan altas concentraciones de metales pesados (MP), creando la necesidad de conducir procesos de restauración en dichos lugares. En el Morro de basuras de Moravia, con 7,6 ha y 45 m de altura, conformado por la disposición de los residuos sólidos de la ciudad de Medellín entre 1974 y 1984, estudios anteriores demostraron alto contenido MP, evidenciando la necesidad de identificar mecanismos efectivos para su restauración. El objetivo de este estudio fue evaluar la adaptación, crecimiento y capaci-dad fitorremediadora de Bidens pilosa, Lepidium virginicum, Brachiaria decumbens y Arachis pintoi. El contenido de MP (mg/kg) en la matriz de residuos de Moravia varió entre 17 y 8.193 para Pb, 44 a 564 para Cr, 0,2 a 339 para Cd y 77 a 1.679 para Ni. Las mediciones de cobertura, altura y producción de materia seca mostraron que todas las especies evaluadas tuvieron un nivel adecuado de adaptación y crecimiento a las condiciones del basurero de Moravia. La absorción de MP presentó el orden Cr > Cd> Ni > Pb. Los factores de bioconcentración estimados fueron bajos, siendo los valores máximos: 0,36 (A. pintoi, Cr), 2,96 (B. pilosa, Cd) y 0,26 (B. decumbens, Ni). Sin embargo, nuestras estimaciones del potencial de fitorremediación de las plantas evaluadas sugieren que éstas poseen baja eficiencia fitorremediadora. Debe dirigirse investigación con el fin de identificar especies acumuladoras de MP más eficientes o introducir tecnologías que aumenten la disponi-bilidad de los MP, tales como la modificación del pH, rizorremediación o el uso de plantas acumuladoras genéticamente modificadas.

RESUMO

En esta entrevista Teresa Cabruja considera los orígenes y el desarrollo de la Psicología crítica en España, así como el lugar de su trabajo en relación con la constitución de este campo. A lo largo de la entrevista emergen momentos claves en los cuales se va configurando un conocimiento psicológico, que se aparta de las visiones convencionales de la disciplina. Se resalta la importancia que tienen los nodos locales para la producción de las perspectivas críticas, así como los encuentros internacionales para la expansión y cimentación de estas visiones alternativas al conocimiento psicológico convencional.


In this interview Teresa Cabruja traces the origins and development of critical psychology in Spain and considers the place of her own work as regards the constitution of this field. During the interview some key moments that configured crítical psychology emerged. The importance of local nodes for the production of knowledge, as well as the international meetings for the expansion and consolidation of alternative views to the mainstream psychology are pointed out.

BACKGROUND: The mestizo patient usually has a small nose with a wide base, round nostrils, and a convex dorsum. The alar cartilages are weak, thin, and short, providing deficient structural support and poor definition of the nasal tip. Cartilage grafts in the nasal tip are very often used to correct this condition, but a common problem of this procedure is the cephalic or lateral rotation of these grafts. OBJECTIVE: We used an angulated extended columellar graft to give columellar support and projection to nasal tip grafts for better control and prediction of the position and effect of these grafts. METHODS: The surgical protocol included a medical history, development of a surgical plan by analysis of the deformity, and the use of pre- and postoperative photographs taken at both intermediate and long-term follow-up visits for evaluation of the results. RESULTS: Sixty-seven patients underwent surgery using this procedure, 56 with an open technique and 11 with a closed technique. Follow-up ranged from 6 months to 4 years. The results obtained were satisfactory, showing better control and prediction of the shape of the nasal tip. CONCLUSIONS: The angulated extended columellar graft provides better control of the projection and angularity of cartilage grafts placed in the nasal tip.
