RESUMO

Immune imprinting is now evident in COVID-19 vaccinated people. This phenomenon may impair the development of effective neutralizing antibodies against variants of concern (VoCs), mainly Omicron and its subvariants. Consequently, the boost doses with bivalent vaccines have not shown a significant gain of function regarding the neutralization of Omicron. The approach to design COVID-19 vaccines must be revised to improve the effectiveness against VoCs. Here, we took advantage of the self-amplifying characteristic of RepRNA and developed a polyvalent formulation composed of mRNA from five VoCs. LION/RepRNA Polyvalent induced neutralizing antibodies in mice previously immunized with LION/RepRNA D614G and reduced the imprinted phenotype associated with low neutralization capacity of Omicron B.1.1.529 pseudoviruses. The polyvalent vaccine can be a strategy to handle the low neutralization of Omicron VoC, despite booster doses with either monovalent or bivalent vaccines.

Nucleic acid-based therapies have the potential to address numerous diseases that pose significant challenges to more traditional methods. RNA-based therapies have emerged as a promising avenue, utilizing nanoformulation treatments to target a range of pathologies. Nanoformulation offers several advantages compared to other treatment modalities, including targeted delivery, low toxicity, and bioactivity suitable for drug loading. At present, various types of nanoformulations are available, such as liposomes, polymeric nanoparticles (NPs), magnetic NPs, nanoshells, and solid lipid nanoparticles (SLNs). RNA-based therapy utilizes intracellular gene nanoparticles with messenger RNA (mRNA) emerging prominently in cancer therapy and immunotechnology against infectious diseases. The approval of mRNA-based technology opens doors for future technological advancements, particularly self-amplifying replicon RNA (repRNA). RepRNA is a novel platform in gene therapy, comprising viral RNA with a unique molecular property that enables the amplification of all encoded genetic information countless times. As a result, repRNA-based therapies have achieved significant levels of gene expression. In this context, the primary objective of this study is to furnish a comprehensive review of repRNA and its applications in nanoformulation treatments, with a specific focus on encapsulated nanoparticles. The overarching goal is to provide an extensive overview of the use of repRNA in conjunction with nanoformulations across a range of treatments and therapies.

Norepinephrine plays an important role in modulating memory through its beta-adrenergic receptors (Adrß: ß1, ß2 and ß3). Here, we hypothesized that multisensory stimulation would reverse memory impairment caused by the inactivation of Adrß3 (Adrß3KO) with consequent inhibition of sustained glial-mediated inflammation. To test this, 21- and 86-day-old Adrß3KO mice were exposed to an 8-week multisensory stimulation (MS) protocol that comprised gustatory and olfactory stimuli of positive and negative valence; intellectual challenges to reach food; the use of hidden objects; and the presentation of food in ways that prompted foraging, which was followed by analysis of GFAP, Iba-1 and EAAT2 protein expression in the hippocampus (HC) and amygdala (AMY). The MS protocol reduced GFAP and Iba-1 expression in the HC of young mice but not in older mice. While this protocol restored memory impairment when applied to Adrß3KO animals immediately after weaning, it had no effect when applied to adult animals. In fact, we observed that aging worsened the memory of Adrß3KO mice. In the AMY of Adrß3KO older mice, we observed an increase in GFAP and EAAT2 expression when compared to wild-type (WT) mice that MS was unable to reduce. These results suggest that a richer and more diverse environment helps to correct memory impairment when applied immediately after weaning in Adrß3KO animals and indicates that the control of neuroinflammation mediates this response.

RESUMO

Objetivo: relatar a experiência sobre a fundação, implementação, consolidação e ações de uma liga acadêmica interdisciplinar de gerontologia. Método: relato de experiência construído durante os meses de abril a maio de 2022 acerca da história e trajetória da Liga Acadêmica Interdisciplinar de Gerontologia da URI Santiago. Os dados que embasaram a escrita do relato de experiência foram extraídos do livro ata da Liga e das verbalizações dos integrantes. Resultados: fundar, implementar e consolidar uma Liga em tempos pandêmicos ascendeu uma maior responsabilidade na escolha da fundamentação teórica; fontes seguras para seleção; e na diversidade de conteúdos (complexidade do envelhecimento), em atenção às questões econômicas, sociais, culturais e de gênero que possuem relação direta com a saúde e a longevidade, sem esquecer da inclusão social (acesso e acessibilidade à tecnologia digital, literacia em saúde, territórios vulneráveis, condições individuais e familiares dos idosos). Conclusão: as atividades realizadas durante o percurso inicial da Liga, além de proporcionar aos discentes a autonomia em atuar de forma prática no contexto da gerontologia, despertou outros membros da comunidade acadêmica para o trabalho interdisciplinar e de engajamento.

RESUMO

Este artigo tem como objetivo descrever a experiência de estudantes dos cursos de Nutrição, Fisioterapia e Enfermagem por meio de atividades educativas de articulação interdisciplinar no Programa Saúde na Escola na zona rural do município de Petrolina-PE. As turmas selecionadas foram o quinto, sexto e sétimo anos do Ensino Fundamental, com alunos da faixa etária de dez a 15 anos, e os temas foram trabalhados em seis encontros com abordagens das metodologias ativas. A aproximação dos escolares com as graduandas dos cursos de saúde favoreceu o desenvolvimento de uma aprendizagem significativa entre os mesmos, além possibilitar o desenvolvimento de conhecimentos, habilidades e atitudes que contribuirão para o desempenho profissional das graduandas. A experiência com o trabalho interprofissional é motivadora para o desenvolvimento de competências na criação de práticas colaborativas, além de abranger um conjunto de dimensões que vão desde experiências prévias, até maior flexibilidade e criatividade para novas vivências.


It aims to reach the experience of students in nutrition, physiotherapy and nursing courses through educational activities of interdisciplinary articulation in the School Health Program in the rural area of the city of Petrolina-PE. The selected groups were the fifth, sixth and seventh years of elementary school, with students aged between ten and 15 years old, and the themes were worked on in six meetings with approaches to active methodologies. The approximation of the students with the undergraduates of the health courses favored the development of a close learning process for them, in addition to enabling the development of knowledge, skills and attitudes that will contribute to the professional performance of graduates. The experience with interprofessional work is motivating for the development of skills in the creation of collaborative practices, in addition to covering a set of dimensions that range from previous experiences to greater flexibility and creativity for new experiences.

The objective of this study was to evaluate carcass characteristics and meat quality of surgically castrated and immunocastrated male pigs. Data were collected from 24 surgically castrated pigs and 24 immunocastrated male entire originating from the commercial line (Agroceres x Topigs), receiving isonutritive diets and were housed in the same experimental shed, slaughtered at 177 days old, with a slaughter weight of 127.8 (± 6.5) and 135.1 (± 7.8) kg, respectively. Carcass and meat quality characteristics such as carcass yield, lean meat yield, fat content, backfat thickness, muscle depth, pH, meat coloring and, drip, thawing and cooking loss, were evaluated. Immunocastrated pigs have heavier carcass, higher meat yield and lower fat thickness when compared to surgically castrated animals. Regarding meat quality, the immunocastrated presented a lighter meat, less reddness and less water holding capacity. In conclusion, this study confirmed that the carcass of male pigs, submitted immunocastration, has lower yield, however, more meat and less fat content, when compared to carcass of male pigs surgically castrated. Meat quality showed peculiar characteristics for immunocastrated pigs, and can be used more for sausage production, since it retains less water and have smaller redness color compared to surgically castrated pigs.

RESUMO

Abstract Higher levels of reactive species of oxygen are harmful to plant tissues. This study evaluated the action of different doses of thymol on soybean seed germination, biometric analysis and enzymatic parameters; both involved in germination process. High doses of thymol affected the plantlet growth, but not hampered the germination.

Destros e canhotos diferem quando comparados em algumas tarefas motoras, parecendo os canhotos usufruir de alguma vantagem em tarefas visuo-motoras. Neste estudo foi analisado, em cada grupo de preferência manual, o efeito da velocidade do estímulo, do sexo e da mão de execução no desempenho de uma tarefa simples de antecipação-coincidência. Participaram 12 destros e 12 canhotos de ambos os sexos, estudantes universitários de Desporto. Empregou-se o "Bassin Anticipation Timer" para avaliar a capacidade de antecipação-coincidência em três velocidades: 268 cm/s, 402,3 cm/s e 536,4 cm/s (6, 9 e 12 mph, respectivamente). Os sujeitos executaram a tarefa tanto com a mão preferida como com a mão não preferida. Principais resultados: 1) apenas os destros foram afetados pela variável velocidade do estímulo, apresentando antecipação das respostas e maior variabilidade na velocidade 268 cm/s, enquanto nas velocidades 402,3 cm/s e 536,4 cm/s as respostas foram enviesadas no sentido do atraso da resposta e com variabilidade menos acentuada na velocidade mais alta; 2) o sexo teve um efeito significativo apenas nos canhotos, sendo o sexo masculino mais preciso e menos enviesado nas suas respostas do que o sexo feminino; 3) a assimetria manual manifestou-se apenas nos canhotos na velocidade de 268 cm/s e no Erro Variável. Concluímos que cada grupo de preferência manual parece comportar-se de forma diferenciada em tarefas perceptivas de Antecipação-Coincidência onde a velocidade do estímulo é manipulada.


Right- and left-handers differ when compared in some motor tasks, and left-handers enjoy some advantage over right-handers in visuo-motor tasks. In this study we intend to analyze the effect of stimulus speed, in a simple coincidence-anticipation task, depending on handedness. Twelve right-handers and twelve left-handers of both sexes, students of Physical Education, participated in this study. The Bassin Anticipation Timer was used to evaluate the coincidence-anticipation ability at three different and random velocities: 268 cm/s, 402.3 cm/s e 536.4 cm/s (6, 9 e 12 mph, respectively). The subjects were evaluated performing the task with the preferred hand and non-preferred hand. The results revealed that: 1) only right-handers were affected by the variable stimulus speed, anticipating responses and being more variable at 268cm/s, while at 402.3 cm/s and 536.4 cm/s responses were late; moreover they were less variable at 536.4cm/s; 2) sex as a main factor was significant only in the left-handed group, males being more accurate and less biased in their responses than females; 3) manual asymmetry was only apparent in the left-handed group at the 268cm/s and in the variable error. The perceptual component of the task seems to be crucial in distinguishing right-handers and left-handers where the speed of the stimulus is manipulated.
