BACKGROUND The objective of this study was to develop and validate machine learning (ML) algorithms to predict the 30-day and 6-month risk of deteriorating functional status following surgical treatment for thoracic spinal stenosis (TSS). We aimed to provide surgeons with tools to identify patients with TSS who have a higher risk of postoperative functional decline. MATERIAL AND METHODS The records of 327 patients with TSS who completed both follow-up visits were analyzed. Our primary endpoint was the dichotomized change in the perioperative Japanese Orthopedic Association (JOA) score, categorized based on whether it deteriorated or not. The models were developed using Naïve Bays, LightGBM, XGBoost, logistic regression, and random forest classification models. The model performance was assessed by accuracy and the c-statistic. ML algorithms were trained, optimized, and tested. RESULTS The best-performing algorithms for predicting functional decline at 30 days and 6 months after TSS surgery were XGBoost (accuracy=88.17%, c-statistic=0.83) and Naïve Bays (accuracy=86.03%, c-statistic=0.80). Both algorithms presented good calibration and discrimination in our testing data. We identified several significant predictors, including poor quality of intraoperative SSEP/MEP baseline, poor quality of preoperative SSEP, duration of symptoms, operated level, and motor dysfunction of the lower extremity. CONCLUSIONS The best-performing algorithms for predicting functional decline at 30 days and 6 months after TSS surgery were XGBoost (accuracy=88.17%, c-statistic=0.83) and Naïve Bays (accuracy=86.03%, c-statistic=0.80). In this study, high-pressure torsion (HPT) processing is applied to the as-cast Al0.5CoCrFeNi high-entropy alloy (HEA) for 1, 3, and 5 turns. Microstructural observations reveal a significant refinement of the second phase after HPT processing. This refinement effect is influenced by the number of processing turns and the distance of the processing position from the center. As the number of processing turns or the distance of the processing position from the center increases, the fragmentation effect on the second phase becomes more pronounced. The hardness of the alloy is greatly enhanced after HPT processing, but there is an upper limit to this enhancement. After increasing the number of processing turns to 5, the increase in hardness at the edge becomes less significant, while the overall hardness becomes more uniform. Additionally, the strength of the processed alloy is significantly enhanced, while its ductility undergoes a noticeable decrease. With an increase in the number of processing turns, the second phase is further refined, resulting in improvement of strength and ductility.

Plant endophytic microbial communities consist of many latent plant pathogens and, also, many pathogen-related species with reduced virulence. Though with a long history of co-evolution, the diversity and composition of the endophytic mycobiota, especially the pathogen-related fungal groups, has been under-investigated in Citrus (C.). Based on the amplicon sequencing of fungal internal transcribed spacer (ITS), the leaf endophytic mycobiota were profiled on citrus varieties from different citrus-producing regions. The pomelo variety shared significantly distinctive leaf mycobiota when compared to the mandarin and sweet orange; these conform to their host genetic relationships. In addition, a data set of 241 citrus-related fungi, including 171 (71%) pathogens and potential pathogens, was summarized from previous studies. Under the criteria of local BLAST (covered ITS nucleotide ≥ 150 bp, sequence identity ≥ 99%), a total of 935 fungal operational taxonomic units (OTUs) were assigned to 62 pathogen-related fungal groups, representing 14.9% of the relative abundance in the whole community. Of which, the top groups consisted of Colletotrichum gloeosporioides (mean relative abundance, 4.3%), Co. citricola and Co. karstii (2.7%), Zasmidium citri-griseum (2.4%), and Z. fructigenum (1.4%). At the genus level, the ratio of the pathogen-related fungal groups in 64% of fungal genera (16 out of 25) exceeded 50%, which are the solely or mainly occurring fungi of their genus in citrus. Our study suggests that the leaf endophytic compartment may be an important place for the growth of latent pathogens.

Neuroendocrine regulation is essential for maintaining metabolic homeostasis. However, whether neuroendocrine pathway influence bone metabolism and skeletal senescence is unelucidated. Here, a central neuroendocrine circuit is identified that directly controls osteogenesis. Using virus based tracing, this study is identified that melanin concentrating hormone (MCH) expressing neurons in the lateral hypothalamus (LH) are connected to the bone. Chemogenetic activation of MCH neurons in the LH induces osteogenesis, whereas inhibiting these neurons reduces osteogenesis. Meanwhile, MCH is released into the circulation upon chemogenetic activation of these neurons. Single cell sequencing reveals that blocking MCH neurons in the LH diminishes osteogenic differentiation of bone marrow stromal cells (BMSCs) and induces senescence. Mechanistically, MCH promotes BMSC differentiation by activating MCHR1 via PKA signaling, and activating MCHR1 by MCH agonists attenuate skeletal senescence in mice. By elucidating a brain-bone connection that autonomously enhances osteogenesis, these findings uncover the neuroendocrinological mechanisms governing bone mass regulation and protect against skeletal senescence.

Currently, the application scope of big data (BD for short here) technology is relatively narrow, mostly used in the medical field, and the degree of application is relatively superficial, mostly for data statistical record analysis. Therefore, By combining the literature review, this article has decided to construct a system based on BD technology for analyzing the advanced neural activity status of athletes. The system is mainly divided into two parts, one is the biological information collection part. As an important source of system data, it is necessary to use professional equipment to collect ECG and EEG data and ensure the accuracy of the data through signal filtering, Gaussian noise elimination, salt and pepper noise, and exponential noise de-noising technology. The other is the algorithm problem of BD systems. Considering that the traditional algorithm can not deal with a large amount of data effectively, this paper chooses the BD spectral clustering algorithm based on core points as the main algorithm to cluster the data. By evaluating the efficiency of system learning, data collection and classification, system scheme construction, and error rate, this article ultimately determined the practical feasibility and effectiveness of the system. After completing the construction of the system, considering the gap between the system's performance and traditional data, this article analyzed the improvement data of various aspects of sports training. This paper compares the performance differences between the system based on BD technology and the traditional data analysis method under different indicators. In terms of data collection and classification, the accuracy of the system based on BD technology in the collection and classification of ECG and EEG data reached 100 % and 90 %, respectively, which was significantly higher than 60 % and 30 % of the traditional methods. By comparing the data from five training courses, it is found that the training efficiency of the conventional method has increased by 60 % in the first course, while the efficiency of the training method based on the BD system has increased by 85 % in the fifth course. For the activation efficiency of nerve function, the activation efficiency of brain nerve function reached 60 % and 90 % respectively in the two nerve function activation training based on the BD system, which was much higher than 30 % and 45 % of the traditional methods. Through a series of tests and comparative analysis of data, the effectiveness of the BD system is finally determined, which can achieve the goal of improving athletes' training efficiency.

BACKGROUND: Ustekinumab is an interleukin (IL)-12/IL-23 inhibitor for the treatment of moderate-to-severe psoriasis. OBJECTIVE: This real-world study compared ustekinumab and tumor necrosis factor-alpha inhibitors (TNFis) in Chinese moderate-to-severe psoriasis patients. METHODS: Patient health records of 110 moderate-to-severe psoriasis patients initiating or switching biologics were reviewed, with 31 patients receiving ustekinumab (ustekinumab group) and 79 patients receiving TNFis (TNFi group). RESULTS: Compared with TNFi group, psoriasis area and severity index (PASI)-75 response rate at month 6 (M6) were elevated (87.1% versus 65.8%, p = 0.026) in the ustekinumab group, whereas the rates at month 1 (M1) and month 3 (M3) and PASI-90 response rates at M1, M3, and M6 only showed an increasing trend (all p > 0.050) in the ustekinumab group than the TNFi group. By subgroup analyses, ustekinumab (versus TNFi) was more effective in patients with biologics therapy history than those without. Compared with the TNFi group, the ustekinumab group had lower dermatology life quality index scores and higher patient satisfaction scores at M3 and M6 (all p < 0.050). CONCLUSION: Chinese moderate-to-severe psoriasis patients treated with ustekinumab have a better treatment response at 6 months with improved quality of life and patient satisfaction after 3-6 months of treatment when compared to TNFi.

Tuning the strength of intramolecular electric field (IEF) in conjugated molecules has emerged as an effective approach to boost charge transfer. While direction manipulation of IEF would be a potential way that is still unclear. Here, we leverage the control of peripheral substituents of conjugated phthalocyanines to chemically tune the spatial orientation of IEF. By analyzing the spatial swing of side chains using the Kolmogorov-Arnold representation and least squares algorithm, a comprehensive mathematical-physical model has been established. This model enables rapid evaluation of the IEF and maximum hole transport performance induced by spatial swings. The champion phthalocyanine as dopant-free hole transport material in perovskite solar cell realizes a record performance of 23.41%. Greatly device stability is also exhibited. This work affords a new way to enhance hole transport capabilities of conjugated molecules by optimizing their IEF vector for photovoltaic devices.

Traditional fungi ß-glucan commonly possesses high molecular weight with poor water solubility, which remains significant challenge in the drug development and medical application. Water-soluble ß-glucan with high molecular weight (dHSCG) of 560 kDa, low molecular weight (dLSCG) of 60 kDa, and sulfated derivative (SCGS) with a molecular weight of 146 kDa and sulfate degree at 2.04 were obtained through well-controlled degradation and sulfated modification from Saccharomyces cerevisiae in this study. The structural characteristics were confirmed as ß-1,3/6-glucan by FT-IR and NMR spectroscopy. Carbohydrate microarrays and surface plasmon resonance revealed distinct and contrasting binding affinities between the natural ß-glucans and sulfated derivatives. SCGS exhibited strong binding to FGF and VEGF, while natural ß-glucan showed no response, suggesting its potential as a novel antitumor agent. Moreover, SCGS significantly inhibited the migration rate of the highly metastatic melanoma (B16F10) cells. The lung metastasis mouse model also demonstrated that SCGS significantly reduced and eliminated the nodules, achieving an inhibition rate of 86.7% in vivo, with a dramatic improvement in IFN-α, TNF-α, and IL-1ß levels. Through analysis of protein content and distribution in lung tissues, the anti-tumor and anti-metastasis mechanism of SCGS involves the regulation of degrading enzymes to protect extracellular matrix (ECM), as well as the reduction of angiogenic factor release. These findings provide a foundation for exploring the potential of SCGS in the development of new anti-tumor and anti-metastasis drugs and open up a new field in cancer research.

Arbuscular mycorrhizal (AM) fungi are well known for enhancing phosphorus uptake in plants; however, their regulating roles in cation transporting gene family, such as natural resistance-associated macrophage protein (NRAMP), are still limited. Here, we performed bioinformatics analysis and quantitative expression assays of tomato SlNRAMP 1 to 5 genes under nutrient deficiency and cadmium (Cd) stress in response to AM symbiosis. These five SlNRAMP members are mainly located in the plasma or vacuolar membrane and can be divided into two subfamilies. Cis-element analysis revealed several motifs involved in phytohormonal and abiotic regulation in their promoters. SlNRAMP2 was downregulated by iron deficiency, while SlNRAMP1, SlNRAMP3, SlNRAMP4, and SlNRAMP5 responded positively to copper-, zinc-, and manganese-deficient conditions. AM colonization reduced Cd accumulation and expression of SlNRAMP3 but enhanced SlNRAMP1, SlNRAMP2, and SlNRMAP4 in plants under Cd stress. These findings provide valuable genetic information for improving tomato resilience to nutrient deficiency and heavy metal stress by developing AM symbiosis.

Residents' participation is crucial for sustainable old neighborhood regeneration. Residents' intentions to participate in the regeneration are low in China, resulting in unsustainable neighborhood development problems. Therefore, it is essential to investigate the drivers of residents' participation. Residents' behavioral mechanisms are deeply embedded in their social networks, especially in acquaintance social networks typical of old neighborhoods in China. By constructing a mediated moderation model based on the Theory of Planned Behavior (TPB) and danwei system and taking the old neighborhood regeneration in Xi'an as a case study, this paper investigates the paths and mechanisms of the social network embeddedness, TPB, and danwei system on residents' participation intention. The results show that social network embeddedness can improve residents' intention to participate in regeneration through both direct and indirect pathways, and its indirect effect is much larger than the direct effect. Attitude, subjective norm, and perceived behavioral control significantly and positively mediated the contribution of social network embeddedness on participation intention. However, the specific mediating effect of attitude is significantly smaller than the other two. The danwei system has a significant moderating effect in the first stage of the relationship between social network embeddedness and participation intention. These imply that the extrinsic social factor of social network embeddedness is internalized as a driving force for convergence and motivation of residents' participation intentions mainly through the normative pressure and exchange of experiences between acquaintances and that the danwei system reinforces this extrinsic-intrinsic facilitation process through the danwei's "physical presence." These findings provide revelatory countermeasures for exploring the factors that motivate residents to participate actively in neighborhood regeneration and encourage their involvement at the intervention level.

Butyl benzyl phthalate (BBP), a common phthalate plasticizer, is frequently detected in aquatic environments. However, there has been relatively little research on its effects on gill-related responses. This study exposed adult zebrafish to BBP concentrations ranging from 5 to 500 µg/L for 28 days, specifically investigating its toxicity in the gills. Assessment of oxidative stress biomarkers and gene expression related to apoptosis and mitochondria was conducted. Results demonstrated that exposure to 500 µg/L of BBP disrupted the antioxidant defense system, leading to lipid peroxidation and DNA damage. Moreover, the expression level of the caspase-3 gene exhibited an approximate two-fold increase, whereas the expression of 18rs-rrn decreased by 50 % on day 28. Gene Ontology enrichment analysis indicated suppressed expression of antioxidant and metabolic process terms, alongside inhibition of metabolism, immune, and signal transduction-related pathways. This study offers novel insights into the toxic effects and mechanisms of BBP on fish, providing valuable data for assessing environmental risks linked to BBP contamination and advocating for its management in aquatic ecosystems.

BACKGROUND: Sirtuin 7 (SIRT7) is pivotal in diverse diseases progression. Importantly, SIRT7 is associated with melanin production. However, whether SIRT7 regulates vitiligo is unclear. Therefore, we aimed to investigate the effects of SIRT7 on pigmentation and the modification of glucose 6-phosphate dehydrogenase (G6PD). METHODS: After knockdown SIRT7 and G6PD, pigmentation of melanocytes was evaluated using commercial kits, immunofluorescence, and Western blot analysis. The succinylation of G6PD mediated by SIRT7 was analyzed using co-immunoprecipitation, immunofluorescence, Western blot analysis, and cycloheximide-chase experiment. RESULTS: We found that SIRT7 was highly expressed in vitiligo skin lesions. Knockdown of SIRT7 increased tyrosinase activity, melanin content, and the levels of α-melanocyte-stimulating hormone, MITF, TYR, TRP1, and TRP2. Additionally, SIRT7 directly interacted with G6PD. Silenced SIRT7 promoted the succinylation of G6PD and enhanced its protein stability. G6PD knockdown reversed the effect of reduced SIRT7 expression on melanin production. CONCLSUION: Silencing of SIRT7 promotes pigmentation of melanocytes by succinylating G6PD, suggesting that SIRT7-mediated G6PD desuccinylation may promote vitiligo progression.

RESUMO

Rosai Dorfman disease is a rare non Langerhans histiocytic disease with intranodal and/or extranodal manifestations, most commonly affecting cervical lymph nodes. There is no treatment guideline for this disease. This paper reports a case of infraglottic portion, which did not show typical symptoms, such as fever or lymphadenopathy, and was easily misdiagnosed as malignant tumor preoperatively. The patient underwent low-temperature plasma minimally invasive surgery combined with hormone therapy and achieved good clinical effect. By reviewing the literature, we can better understand the clinical manifestations, diagnosis and treatment of this rare disease.

Populus tomentosa, an indigenous tree species, is widely distributed and cultivated over 1,000,000 km2 in China, contributing significantly to forest production, ecological conservation and urban-rural greening. Although a reference genome is available for P. tomentosa, the intricate interspecific hybrid origins, chromosome structural variations (SVs) and sex determination mechanisms remain confusion and unclear due to its broad and even overlapping geographical distribution, extensive morphological variations and cross infiltration among white poplar species. We conducted a haplotype-resolved de novo assembly of P. tomentosa elite individual GM107, which comprises subgenomes a and b with a total genome size of 714.9 Mb. We then analysed the formation of hybrid species and the phylogenetic evolution and sex differentiation across the entire genus. Phylogenomic analyses suggested that GM107 likely originated from a hybridisation event between P. alba (♀) and P. davidiana (♂) approximately 3.8 Mya. A total of 1551 chromosome SVs were identified between the two subgenomes. More noteworthily, a distinctive inversion structure spanning 2.15-2.95 Mb was unveiled among Populus, Tacamahaca, Turaga, Aigeiros poplar species and Salix, highlighting a unique evolutionary feature. Intriguingly, a novel sex genotype of the ZY type, which represents a crossover between XY and ZW systems, was identified and confirmed through both natural and artificial hybrids populations. These novel insights offer significant theoretical value for the study of the species' evolutionary origins and serve as a valuable resource for ecological genetics and forest biotechnology.

Objective: This investigation aims to elucidate the correlations between dietary intakes of vitamin E, B6, and niacin and the incidence of cataracts, utilizing the comprehensive NHANES 2005-2008 dataset to affirm the prophylactic roles of these nutrients against cataract formation. Methods: Using data from the NHANES 2005-2008 cycles, this analysis concentrated on 7,247 subjects after exclusion based on incomplete dietary or cataract data. The identification of cataracts was determined through participants' self-reported ophthalmic surgical history. Nutritional intake was gauged using the automated multiple pass method, and the data were analyzed using logistic and quantile regression analyses to investigate the relationship between vitamin consumption and cataract prevalence. Results: Our analysis identified significant inverse associations between the intake of vitamins E, B6, and niacin and the risk of cataract development. Specifically, higher intakes of vitamin B6 (OR = 0.85, 95% CI = 0.76-0.96, p = 0.0073) and niacin (OR = 0.98, 95% CI = 0.97-1.00, p = 0.0067) in the top quartile were significantly associated with a reduced likelihood of cataract occurrence. Vitamin E intake showed a consistent reduction in cataract risk across different intake levels (OR = 0.96, 95% CI = 0.94-0.99, p = 0.0087), demonstrating a nonlinear inverse correlation. Conclusion: The outcomes indicate that elevated consumption of vitamin B6 and niacin, in conjunction with regular vitamin E intake, may have the potential to delay or prevent cataract genesis. These results suggest a novel nutritional strategy for cataract prevention and management, advocating that focused nutrient supplementation could be instrumental in preserving eye health and reducing the risk of cataracts. Further research is recommended to validate these findings and establish optimal dosages for maximum benefit.

BACKGROUND: Diffusion-weighted imaging (DWI), a pivotal component of multiparametric magnetic resonance imaging (mpMRI), plays a pivotal role in the detection, diagnosis, and evaluation of gastric cancer. Despite its potential, DWI is often marred by substantial anatomical distortions and sensitivity artifacts, which can hinder its practical utility. Presently, enhancing DWI's image quality necessitates reliance on cutting-edge hardware and extended scanning durations. The development of a rapid technique that optimally balances shortened acquisition time with improved image quality would have substantial clinical relevance. OBJECTIVES: This study aims to construct and evaluate the unsupervised learning framework called attention dual contrast vision transformer cyclegan (ADCVCGAN) for enhancing image quality and reducing scanning time in gastric DWI. METHODS: The ADCVCGAN framework, proposed in this study, employs high b-value DWI (b = 1200 s/mm2) as a reference for generating synthetic b-value DWI (s-DWI) from acquired lower b-value DWI (a-DWI, b = 800 s/mm2). Specifically, ADCVCGAN incorporates an attention mechanism CBAM module into the CycleGAN generator to enhance feature extraction from the input a-DWI in both the channel and spatial dimensions. Subsequently, a vision transformer module, based on the U-net framework, is introduced to refine detailed features, aiming to produce s-DWI with image quality comparable to that of b-DWI. Finally, images from the source domain are added as negative samples to the discriminator, encouraging the discriminator to steer the generator towards synthesizing images distant from the source domain in the latent space, with the goal of generating more realistic s-DWI. The image quality of the s-DWI is quantitatively assessed using metrics such as the peak signal-to-noise ratio (PSNR), structural similarity index (SSIM), feature similarity index (FSIM), mean squared error (MSE), weighted peak signal-to-noise ratio (WPSNR), and weighted mean squared error (WMSE). Subjective evaluations of different DWI images were conducted using the Wilcoxon signed-rank test. The reproducibility and consistency of b-ADC and s-ADC, calculated from b-DWI and s-DWI, respectively, were assessed using the intraclass correlation coefficient (ICC). A statistical significance level of p < 0.05 was considered. RESULTS: The s-DWI generated by the unsupervised learning framework ADCVCGAN scored significantly higher than a-DWI in quantitative metrics such as PSNR, SSIM, FSIM, MSE, WPSNR, and WMSE, with statistical significance (p < 0.001). This performance is comparable to the optimal level achieved by the latest synthetic algorithms. Subjective scores for lesion visibility, image anatomical details, image distortion, and overall image quality were significantly higher for s-DWI and b-DWI compared to a-DWI (p < 0.001). At the same time, there was no significant difference between the scores of s-DWI and b-DWI (p > 0.05). The consistency of b-ADC and s-ADC readings was comparable among different readers (ICC: b-ADC 0.87-0.90; s-ADC 0.88-0.89, respectively). The repeatability of b-ADC and s-ADC readings by the same reader was also comparable (Reader1 ICC: b-ADC 0.85-0.86, s-ADC 0.85-0.93; Reader2 ICC: b-ADC 0.86-0.87, s-ADC 0.89-0.92, respectively). CONCLUSIONS: ADCVCGAN shows excellent promise in generating gastric cancer DWI images. It effectively reduces scanning time, improves image quality, and ensures the authenticity of s-DWI images and their s-ADC values, thus providing a basis for assisting clinical decision making.

African American (AA) kidney transplant recipients (KTRs) have poor outcomes, which may in-part be due to tacrolimus (TAC) sub-optimal immunosuppression. We previously determined the common genetic regulators of TAC pharmacokinetics in AAs which were CYP3A5 *3, *6, and *7. To identify low-frequency variants that impact TAC pharmacokinetics, we used extreme phenotype sampling and compared individuals with extreme high (n = 58) and low (n = 60) TAC troughs (N = 515 AA KTRs). Targeted next generation sequencing was conducted in these two groups. Median TAC troughs in the high group were 7.7 ng/ml compared with 6.3 ng/ml in the low group, despite lower daily doses of 5 versus 12 mg, respectively. Of 34,542 identified variants across 99 genes, 1406 variants were suggestively associated with TAC troughs in univariate models (p-value < 0.05), however none were significant after multiple testing correction. We suggest future studies investigate additional sources of TAC pharmacokinetic variability such as drug-drug-gene interactions and pharmacomicrobiome.

The salinization of soil constitutes a substantial hindrance to the advancement of sustainable agriculture. Our research seeks to elucidate the role of a Rab GTPase-activating protein (RabGAP) family member, SlRabGAP22, in salt tolerance and its translational regulation under salt stress in tomatoes, employing gene-editing techniques and ribosome profiling methodologies. Findings demonstrate that SlRabGAP22 acts as a positive regulator of tomato salt tolerance, with four predicted upstream open reading frames (uORFs) classified into three categories. Functional uORFs were found to be negative regulation. Editing these uORFs along with altering their classifications and characteristics mitigated the inhibitory effects on primary ORFs and fine-tuned gene expression. Enhanced tomato salt tolerance was attributed to improved scavenging of reactive oxygen species, reduced toxicity Na+, and diminished osmotic stress effects. Furthermore, we conducted genome-wide analysis of ORFs to lay the foundation for further research on uORFs in tomatoes. In summary, our findings offer novel perspectives and important data for the enhancement of genetic traits via uORF-based strategies and translational regulation against the backdrop of salt stress.
