Background: Canine hypercortisolism (HC) is the most prevalent endocrinopathy in dogs in southern Brazil. The prognosis depends on several factors including the general health status, owners´ commitment, and the development of disease complications and comorbidities occurrence, such as cardiovascular complications including mitral valve disease (MVD), systemic arterial hypertension, and left ventricular hypertrophy. The main objective of the present study was to assess cardiovascular parameters in canine HC, based on investigating survival-related variables. The study also aimed to evaluate the influence of concurrent preclinical (MVD) on dogs` survival and the impact of HC on MVD progression. Materials, Methods & Results: A total of 25 dogs with spontaneous HC were enrolled and divided into 2 subgroups accordingly to their echocardiographic findings: group HC (normal echocardiography at first evaluation, n = 16); and group HC + MVD (concurrent presence of stages B1 and B2 MVD diagnosed at the first evaluation, n = 9). The patients were evaluated at diagnosis (T0); 6 months after treatment begging (T1); and after 12 months of treatment (T2). The owners were further contacted by phone or e-mail for 1 more year after T2 regarding survival information. A control group (CG, n = 20) was also evaluated at T0 and T1. At each evaluation, dogs were submitted to a complete clinical evaluation and physical exam, associated with a minimum database (CBC, serum biochemistry, and urine analysis) and cardiovascular evaluation composed of systolic blood pressure determination, electrocardiogram (ECG), and echodopplercardiography. In the HC group, 11/16 dogs underwent the evaluation at T2, while 4/9 dogs from the HC + MVD group and 17/20 dogs from the CG underwent the evaluation at T2. Five dogs (31.25%) from the HC group and 4 dogs (44.44%) from the HC + MVD group died before the end of the follow-up period. In the control group, only 1 dog (5.26%) died before the end of the study. Despite the higher mortality in the HC + MVD group during the follow-up period, there was no significant difference (P = 0.632) in survival when compared with the HC group. The MVD of 4 dogs included in the HC + MVD group was classified as stage B1, while the other 5 dogs were classified as stage B2. Only 1 dog from the CG developed stage B1 MVD in the period studied; however, progression of the MVD stage was documented in 1/4 of dogs in the HC + MVD group and MVD development was documented in 3/11 of dogs from the HC group from T0 to T2. The odds ratio (OR) and respective 95% confidence interval (95%CI) for HC as a risk factor for MVD progression were 4.267 (0.4268 - 42.65; P = 0.342). Exercise intolerance (12/16 dogs) and dyspnea (6/16 dogs) were the cardiorespiratory clinical signs with the highest incidence in the HC group at T0. When compared to the control group, both exercise intolerance (P < 0.001) and dyspnea (P = 0.03) occurrence were significantly higher in the HC group. The age (P = 0.001) and the occurrence of dyspnea (P = 0.036) at diagnosis were significantly higher in dogs with HC that died during the follow-up than those that remained alive. Regarding the occurrence of cardiac arrhythmias verified by ECG, no statistically significant differences were observed among groups. Discussion: The study suggests that systemic hypertension and altered echocardiographic measurements did not interfere with dogs' survival; however, dyspnea was associated with a worse prognosis. Finally, it is possible to conclude that mitral valve degeneration is a common comorbidity in dogs with HC, however, it was not evidenced their interference in the survival of dogs with this endocrine disease or even a role of the HC in the progression of the MVD.

Background: Assessing data from metabolic profile is a complex task depending on individual factors such as breed, age,sex and physiological status and environmental factors such as climate and management. Therefore, reference intervalsproduced in herds from different geographic regions are not always precise in the local reality. The aim of this study isto compile results of values of the most used parameters in the metabolic profile of healthy Holstein cows, grouped bylactation stage, obtained from studies in Rio Grande do Sul, Brazil, as an effort for the elaboration of regional referenceintervals for dairy cattle in this geographic area.Materials, Methods & Results: The research was defined through the PICO strategy, considering multiparous Holsteincows from scientific studies carried out in Rio Grande do Sul, during the period 1996-2021, in which the nitrogen, glycidic, lipidic, enzymatic or mineral metabolic profile was determined. Google Scholar, Scientific Electronic Library andPubMed were the electronic databases selected for the search, using the following descriptors: “profile” and “metabolic”and “metabolic profile” and “dairy” or “cattle” or “cow” and “Brazil” and “southern” or “Rio Grande do Sul”. Studiesin Portuguese, English and Spanish languages were considered. The references of the articles initially selected were alsoverified, through ResearchGate and Google Scholar, in addition to unpublished data, unfinished studies or data in the socalled “gray literature”. It was possible to identify a total of 34 publications, which were tabulated to assess the scientificquality and bias risk. Studies were evaluated by a pair of independent reviewers, in order to compare results. Study's aims were to characterize subclinical bacteriuria (SB) and sporadic bacterial cystitis (SBC) in dogs with spontaneous hypercortisolism (HC). Prospective cross-sectional design divided patients as newly diagnosed (n = 27), poorly controlled (n  = 21), well controlled (n  = 34), and controls (n  = 19). Urine culture positive results were identified by MALDI-TOF and submitted to antibiogram. Escherichia coli was the most common microorganism (36%). The majority of positive cultures in HC were SB (12.2%). All 4.1% SBC cases were in well controlled HC cases. Bacteriuria correlated with low urine specific gravity and low lymphocyte count. HC degree of control correlated with leukocyturia. SB/SBC cases were treated based in antimicrobial susceptibility leading to microbiological cure in 75% of HC cases. Persistent infections occurred only in SB cases, all by E. coli which became more resistant. SB/SBC prevalence in canine HC is actually lower. Further evidence for current ISCAID guideline contraindication for SB treatment due to HC were provided.

BACKGROUND: Platelets undergo structural, biochemical and functional alterations when stored, and platelet storage lesions reduce platelet function and half-life after transfusion. The objective of this study was to evaluate stored canine platelet concentrates with platelet aggregation, flow cytometry and biochemistry assays. Twenty-two bags of canine platelet concentrates were obtained by the platelet-rich plasma method and were assessed on days 1, 3 and 5 after collection. Parameters such as platelet counts, residual leukocytes, platelet swirling, glucose, lactate, pH, CD62P expression (platelet activation), JC-1 (mitochondrial function) and annexin V (apoptosis and cell death) were assessed. RESULTS: Over the five days of storage there was a significant decrease in glucose, HCO3, pCO2, ATP, pH, swirling and mitochondrial function, associated with a significant increase in lactate levels and pO2. At the end of storage pH was 5.9 ± 0.6 and lactate levels were 2.8 ± 1.2 mmol/L. Results of the quality parameters evaluated were similar to those reported in human platelets studies. The deleterious effects of storage were more pronounced in bags with higher platelet counts (> 7.49 × 1010/unit), suggesting that canine platelet concentrates should not contain an excessive number of platelets. CONCLUSIONS: Quality parameters of canine platelets under standard storage conditions were similar to those observed in human platelets. Our results have potential to be used for the routine evaluation and quality control in veterinary blood banks.

Background: Retained placenta (RP) is characterized by a failure to remove the fetal membranes within the first 12-24h after calving. This condition appears to be related to a decrease in neutrophil activity and to the suppression of the immune response in the prepartum period. The specific reasons for some cows to retain the placenta after parturition is stillnot fully understood, but numerous predisposing factors have been related, which may include mechanical, nutritional,infectious and handling factors. The aim of this study was to analyze the occurrence of retained placenta in dairy cowsand to correlate the main predisposing factors related.Materials, Methods & Results: This study was conducted in nine dairy farms located in the Rio Grande do Sul state,Brazil, with an average of 45 lactating dairy cows producing 10,100 kg / dairy cow in a period of 305 days. The total dietfor postpartum cows was estimated to meet or exceed the requirements of dairy cows according to previously establishedguidelines (NRC 2001). A total of 393 calving Holstein cows (126 primiparous and 267 multiparous) were analyzed, ofwhich 203 were kept in a semi-confined production system (free-stall and pasture system) and 190 animals were kept ina free-stall production system. Statistically, the cows were the experimental unit, and the results were analyzed using thePearsons Chi-squared test or Fishers exact test for the comparisons of occurrence of peripartum disorders. In addition,linear and logistic regression models were constructed to determine the effect of the dependent variable on the other indicators, which may be continuous or categorical. Mitochondria perform crucial roles in many biochemical processes, and mitochondrial depolarization is an early sign of platelet apoptosis. The mitochondrial membrane potential is usually evaluated through JC-1 probe, but it can also be assessed with MitoTracker probes. Our aim was to evaluate mitochondrial viability in stored canine platelet concentrates (PCs) with the fluorescent probes JC-1 and MitoTracker. Platelets from 22 canine PCs were stained with JC-1 and MitoTracker probes on days 1, 3, and 5 of storage. Data on metabolic parameters were also collected for correlation studies. Results of JC-1 and MitoTracker revealed a decrease in mitochondrial membrane potential in day 5 of storage compared to days 1 and 3, providing evidence of mitochondrial depolarization, a finding that was confirmed by the data on metabolic parameters. MitoTracker probes also added information regarding platelet swelling. In conclusion, MitoTracker probes offered a more complete mitochondrial analysis in the evaluation of stored canine PCs. © 2018 International Society for Advancement of Cytometry.

This study vised to detect possible changes in chemical and cellular composition of cows milk at different milking sessions and to test if proportional or composite milk sampling can be used instead of separate milk analysis. Two experiments were conducted: one in cows milked thrice daily and the other one in cows milked twice daily. In both experiments, three milk samples were collected from each cow in each milking session in order to determine milk composition at each milking; to have a composite sample, i.e., a single sample from the same aliquots of milk from each milking session; and to have a proportional sample, i.e., a single sample from each milking session in a volume proportional to milk volume yielded. Fat content and fat to protein ratio were higher in the afternoon on both experiments. Protein content was larger in the afternoon on cows milked twice daily, but not on cows milked thrice daily. Lactose level was not different across the milking sessions in any of the experiments. The urea level was higher in the afternoon and evening milking on cows milked thrice daily, and in the morning milking on cows milked twice daily. Somatic cell count did not differ between milking in any experiment. No difference in milk composition was noted between proportional and composite samples on cows milked thrice or twice daily. The collection of proportional and composite samples is recommended in twice or thrice daily milking sessions so that the analysis of milk components becomes more easy and cheap.(AU)


Este estudo visou detectar possíveis mudanças na composição química e celular do leite de vaca em diferentes sessões de ordenha, e testar se a amostragem de leite proporcional ou composta pode ser usada em vez de uma análise separada do leite. Dois experimentos foram conduzidos: um em vacas ordenhadas três vezes ao dia e outro em vacas ordenhadas duas vezes ao dia. Em ambos os experimentos, três amostras de leite foram coletadas de cada vaca em cada ordenha: para determinar a composição do leite em cada ordenha; ter uma amostra composta, isto é, uma única amostra das mesmas alíquotas de leite de cada sessão de ordenha; e ter uma amostra proporcional, ou seja, uma única amostra de cada sessão de ordenha em um volume proporcional ao volume de leite produzido. O conteúdo de gordura e a relação gordura:proteína foram maiores no período da tarde nos dois experimentos. O conteúdo de proteína foi maior à tarde nas vacas ordenhadas duas vezes ao dia, mas não nas vacas ordenhadas três vezes ao dia. O nível de lactose não foi diferente nas sessões de ordenha em nenhum dos experimentos. O nível de ureia foi maior à tarde e à noite, nas vacas ordenhadas três vezes ao dia e, pela manhã, nas vacas ordenhadas duas vezes ao dia. A contagem de células somáticas não diferiu entre as ordenhas em nenhum experimento. Não houve diferença na composição do leite entre as amostras proporcionais e compostas em vacas ordenhadas três vezes ao dia. Pre-loading handling and conditions of transport are related to welfare, disease risk and product quality of production animals. These steps continue to be one of the major animal management problems in Brazil. This study evaluated the effects of different types of pre-loading handling and road transport times on the haematological and biochemical traits of cattle. Eighteen male cattle were submitted to three travel times (24, 48 and 72 h) in a truck soon after load using different types of pre-loading handling: traditional (rough handling), training (gentle handling) and use of flags to movement cattle. Haematological traits, blood biochemical measures as well as blood and faecal cortisol were analysed in order to assess animal welfare and physiological status. The traditional management showed to be more stressful, also had animals with a greater number of neutrophils and lower numbers of lymphocytes than handling with flags, showing that animals submitted to more stressful situations can have compromised immune system. Serum aspartate aminotransferase concentrations were within the reference levels and when taken together with increased creatine kinase patterns observed indicate muscle damage in traditional management. Decrease in glucose concentrations over time from traditional management to flag management was observed, while fructosamine was increased in traditional management with 72 h of travel. When taken together, all reported factors, immune, enzymatic, energetic and hormonal, indicate that the quality of pre-loading handling and time of transport were determinant for animal welfare, its homeostatic balance and sanitary conditions.

Background: Abomasal diseases of dairy cattle are mainly associated with stress conditions, nutritional disorders and metabolic disorders. Dairy cattle that develop left or right abomasal displacement generally lose their appetite and have 30 to 50% drop in milk production. The objective of this work is to report the clinical and laboratorial aspects of a high yielding Holstein cow suffering left abomasal displacement in Rio Grande do Sul, southern Brazil.Case: The cow was part of a research project of the Veterinary Faculty of the Federal University of Rio Grande do Sul and was monitored weekly. The animal showed decreased milk production, anorexia, prostration, tremors, diarrhea, smelly metritis and a body score of 3 in a scale of 1-5. In the clinical examination, the animal presented respiratory rate of 26 movements per minute (RV: 26-35), heart rate of 71 beats per minute (RV: 48-84), body temperature of 38.5°C (RV: 38- 39.3), normocorate mucosa, moderate dehydration and no ruminal movements. In the abdominal auscultation the animal presented metallic sound in the left flank. Blood concentration of beta-hydroxybutyrate was 8.3 mmol/L (RV: < 1.2 mmol/L). The cow underwent surgery for correction of left abomasum displacement. After surgery, the animal showed improvement of the clinical condition and ate hay, pasture and silage. Feeding with concentrate was avoided for a week. The animal had satisfactory evolution, being monitored for more 4 weeks and afterward was discharge.Discussion: Genetic improvement in milk cattle has led to the selection of animals with higher milk production through the selection of animals with deeper body capacity, which may allow more space in the abdomen for abomasal movement. In the present case, clinical ketosis was confirmed through the presence of ketonuria and ketonemia.[...]

The cow underwent surgery for correction of left abomasum displacement. After surgery, the animal showed improvement of the clinical condition and ate hay, pasture and silage. Feeding with concentrate was avoided for a week. The animal had satisfactory evolution, being monitored for more 4 weeks and afterward was discharge.Discussion: Genetic improvement in milk cattle has led to the selection of animals with higher milk production through the selection of animals with deeper body capacity, which may allow more space in the abdomen for abomasal movement. Worldwide, milk analysis is an satisfactory measurement of milk quality on dairy farms. Milk composition is the base for payment systems, so knowledge of its characteristics at different periods is essential for modern dairy producer. The aim of the study was to evaluate the effects of year, season, stage of lactation, and lactation number on variations in milk yield and composition of Holstein cows (n=148,604). Milk yield estimated by multiple regression was 25,50L/cow/day. Winter was the season with the highest milk yield and the highest protein and lactose content. Milk fat was highest in the fall, in cows in late lactation and also in primiparous cows. Lactating cows from 6 to 60 days in lactation and those in 2nd or 3rd lactation showed the highest milk yield. Lactose concentration was positively correlated with milk yield. Somatic cell score was negatively correlated with milk yield and with lactose concentration. Most important variations in milk characteristics occur among seasons, lactation stages and parities. Mammary gland health is yet the greatest challenge to be overcome.(AU)


A determinação da qualidade do leite nas fazendas é um parâmetro mundialmente aceito e utilizado como base para os sistemas de pagamento. Os produtores modernos utilizam o conhecimento das características do leite para adotar medidas que visam melhorar produtividade e qualidade. O objetivo do estudo foi avaliar os efeitos do ano, estação, estágio de lactação e número de lactação sobre as variáveis produção e composição do leite de vacas Holandesas (n=148,604). A produção de leite estimada por regressão múltipla foi de 25,50L/vaca/dia. O inverno foi à estação com a maior produtividade e os maiores teores de proteína e lactose. A gordura de leite foi maior no outono em vacas primíparas e no final da lactação. As vacas com estágio de lactação entre 6 a 60 dias e aquelas que se encontram na segunda ou terceira lactação apresentaram a maior produtividade. A concentração de lactose correlacionou-se positivamente com a produção de leite. O escore das células somáticas correlacionou-se negativamente com a produtividade e com a concentração de lactose. As variações mais importantes nas características do leite ocorrem entre estações, estágios de lactação e número de partos. A saúde da glândula mamária ainda é o maior desafio a superar.(AU)

The cowside determination of blood β-hydroxybutyrate (BHB) is an important tool for diagnosing subclinical ketosis in dairy cattle. Portable methods to measure BHB have been introduced in the past years. This study evaluated the accuracy of two hand-held electronic devices for blood determination of BHB in dairy cows at early postpartum. A total of 98 blood samples were collected from dairy cows in the first month of lactation and tested with FreeStyle Optium (FSO, Abbott) and KetoVet (KVE, TaiDoc) portable devices according to the manufacturer instructions. Spectrophotometric BHB analysis (Ranbut, Randox) was used as standard method. The incidence of subclinical ketosis was 37.7 % determined by the standard method, 40.8 % determined by the FSO system and 42.8 % detected with the KVE system. The sensitivity and specificity indexes were 88.1% and 98.4% for FSO and 78.7% and 92.4% for KVE, respectively. The Pearson´s correlation coefficients comparing the portable devices to the standard technique were 0.96 for FSO and 0.93 for KVE. No significant difference in BHB values was found between the two portable tests and the standard method. Predictive values (PV) were better using FSO (positive PV 97.3 %, negative PV 92.4%) than using KVE (positive PV 88.1 %, negative PV 85.9 %). Passing-Bablok regressions revealed good agreement between methods. Though FSO system had a better performance than KVE system, the results suggest that the two portable systems have good accuracy and are reliable for measuring BHB.(AU)

Este estudo objetivou avaliar a acurácia de dois aparelhos eletrônicos portáteis para dosagem sanguínea de BHB de vacas leiteiras no início do pós-parto. Background: Feline Leukemia Virus (FeLV) is an oncogenic virus that usually invades bone marrow causing non-regenerative anemia and thrombocytopenia. In FeLV positive cats, the myeloid leukemia is characterized by intense proliferation of blast cells in a bone marrow with myeloid precursor predominance and several malignancies degree. The leukocyte alkaline phosphatase is found secondary granules of mature neutrophils of humans, horses and cows but not in dogs and cats, which demonstrate the enzyme activity in myeloblast and promyeloblasts cells. In this case is describing a FeLV-positive case with leukemic disorder accompanied by extremely high levels of serum alkaline phosphatase. Case: A 8-year-old castrated male Domestic Short Hair cat with a 3-month history of convulsions, pica, vomits, lethargy, hypodipsia and anorexia was examined and showed mild dehydration, severe jaundice and hyperpnoea. Laboratory results evidenced severe normocytic hypochromic non-regenerative anemia, moderated leukocytosis with severe degenerative neutrophil left shift, moderated lymphocytosis and severe thrombocytopenia. Serum biochemical profile showed mild hyperalbuminemia, moderated increment in alanine transaminase and severe increment in alkaline phosphatese. The cat was positive for ELISA point of care FeLV antigen and negative for FIV antibodies. Urinalysis revealed moderated bilirubinuria [...](AU)

This study observes the effects of manipulating the diet in its physical form and in cereal composition of the concentrate on sorting behaviour, feed intake, composition of milk, and degree of discomfort in dairy cows. In a commercial dairy farm with 360 mid-lactation Holstein cows (averaged 105 days), three treatments of feeding handling were applied sequentially during 120 days, each separated by periods of 30 days of resting: (1) Time of chopping of the total mixed ration (TMR): 15min (N= 180) and 23min (N= 180), (2) Physical form of the concentrate: meal (N= 180) and pellet (N= 180), and (3) Cereal composition in the concentrate: mix of corn, barley and wheat in traditional proportions (3.6:1:1) (N= 120), barley only (N= 120) and corn only (N= 120). Animals feeding TMR chopped for 23min selected less and consumed more ration, and had more milk protein and less discomfort than cows feeding TMR chopped for 15min. The physical form of the concentrate, meal or pellet, did not affect feed sorting, but the meal form resulted in greater feed intake and milk fat percentage. The type of cereal in the concentrate did not affect feed sorting but affected feed intake, milk yield and milk composition. In conclusion, it is recommended using TMR chopped during 23min instead of 15min and using the traditional mix of cereals in the concentrate (corn, barn and wheat) instead of using only one cereal.


O estudo observou os efeitos da manipulação da dieta na sua forma física e na composição de cereais do concentrado sobre comportamento de escolha e consumo do alimento, composição do leite e grau de desconforto em vacas leiteiras. Em um rebanho comercial com 360 vacas de raça Holandesa no terço médio da lactação (média 105 dias em lactação), foram aplicados três tratamentos de manejo alimentar de forma sequencial durante 120 dias, separados por 30 dias de repouso: (1) Tempo de picado da ração totalmente misturada (TMR): 15min (N= 180) e 23min (N= 180), (2) Forma física do concentrado: farelo (N= 180) e pellet (N= 180); e (3) Composição do cereal no concentrado: mistura tradicional de milho, cevada e trigo em proporção 3,6:1:1 (N= 120), apenas cevada (N= 120) e apenas milho (N= 120). Os animais consumindo TMR picada por 23min selecionaram menos e consumiram mais ração e tiveram mais proteína láctea e menos desconforto. A forma física do concentrado, farelo ou pellet, não influenciou o comportamento de escolha da ração. O farelo resultou em maior consumo e maior percentual de gordura láctea. O tipo de cereal no concentrado não afetou a escolha da ração, mas afetou o consumo de alimento, a produção e a composição de leite, com maior produção e melhor conforto usando cevada. Em conclusão, recomenda-se usar TMR picada por 23min ao invés de 15min e usar a mistura tradicional de cereais (milho, cevada e trigo) ao invés de usar apenas um cereal.
