RESUMO

The wheat leaf rust fungus, Puccinia triticina, is found in the major wheat growing regions of the world and is a leading cause of yield loss in wheat. Populations of P. triticina are highly variable for virulence to resistance genes in wheat and adapt quickly to resistance genes in wheat cultivars. The objectives of this study were to determine the genetic relatedness of worldwide collections of P. triticina using restriction site associated genotyping by sequencing. A total of 558 isolates of P. triticina from wheat producing regions in North America, South America, Europe, the Middle East, Ethiopia, Russia, Pakistan, Central Asia, China, New Zealand, and South Africa were characterized at 6745 single nucleotide loci. Isolates were also tested for virulence to 20 near-isogenic lines that differ for leaf rust resistance genes. Populations that were geographically proximal were also more closely related for genotypes. In addition, groups of isolates within regions that varied for genotype were similar to groups from other regions, which indicated past and recent migration across regions. Isolates from tetraploid durum wheat in five different regions were highly related with distinct genotypes compared to isolates from hexaploid common wheat. Based on a molecular clock, isolates from durum wheat found only in Ethiopia were the first to diverge from a common ancestor form of P. triticina that is found on the wild wheat relative Aegilops speltoides, followed by the divergence of isolates found worldwide that are virulent to durum wheat, and then by isolates found on common wheat.

Many plant pathogenic fungi have a global distribution across diverse ecological zones and agricultural production systems. Puccinia triticina, the wheat leaf rust fungus, is a major pathogen in many wheat production areas of the world. The objective of this research was to determine the genetic relatedness of P. triticina in different worldwide regions. A total of 831 single-uredinial isolates collected from 11 regions were characterized for multilocus genotype at 23 simple sequence repeat loci and for virulence to 20 lines of wheat with single genes for leaf rust resistance. A total of 424 multilocus genotypes and 497 virulence phenotypes were found. All populations had high heterozygosity and significant correlation between virulence and molecular variation, which indicated clonal reproduction. The populations from North America and South America, Central Asia and Russia, and the Middle East and Europe were closely related for multilocus genotypes and many individual isolates from other continental regions were closely related. Twenty-seven multilocus genotypes were found in more than one continental region, and 13 of these had isolates with identical virulence phenotypes. The wide geographic distribution of identical and highly related multilocus genotypes of P. triticina indicated past and more recent migration events facilitated by the spread of clonally produced urediniospores.

The self-assembly of thiomalic acid (TMA) on Au(111) and on preformed Au nanoparticles (AuNPs) protected by weak ligands has been studied by X-ray photoelectron spectroscopy (XPS) and electrochemical techniques. Results show that TMA is adsorbed on the Au(111) surface as thiolate species with a small amount of atomic sulfur (∼10%) and a surface coverage lower than that found for alkanethiols due to steric factors. The amount of atomic sulfur markedly increases when the TMA is adsorbed on AuNPs by the ligand exchange method. We propose that the atomic sulfur is produced as a consequence of C-S bond cleavage, a process that is more favorable at defective sites of the AuNPs surface. The bond scission is also assisted by the presence of the electron-withdrawing carboxy moiety in the α-position relative to the C-S bond. Moreover, the high local concentration of positively charged species increases the stability of the negatively charged leaving group, leading to a higher amount of coadsorbed atomic sulfur. Our results demonstrate that the terminal functionalities of thiols are conditioning factors in the final structure and composition of the adlayers.

A doubled haploid (DH) barley (Hordeum vulgare L.) population of 334 lines (ND24260 × Flagship) genotyped with DArT markers was used to map genes for adult plant resistance (APR) to leaf rust (Puccinia hordei Otth) under field conditions in Australia and Uruguay. The Australian barley cultivar Flagship carries an APR gene (qRphFlag) derived from the cultivar Vada. Association analysis and composite interval mapping identified two genes conferring APR in this DH population. qRphFlag was mapped to the short arm of chromosome 5H (5HS), accounting for 64-85% of the phenotypic variation across four field environments and 56% under controlled environmental conditions (CEC). A second quantitative trait locus (QTL) from ND24260 (qRphND) with smaller effect was mapped to chromosome 6HL. In the absence of qRphFlag, qRphND conferred only a low level of resistance. DH lines displaying the highest level of APR carried both genes. Sequence information for the critical DArT marker bPb-0837 (positioned at 21.2 cM on chromosome 5HS) was used to develop bPb-0837-PCR, a simple PCR-based marker for qRphFlag. The 245 bp fragment for bPb-0837-PCR was detected in a range of barley cultivars known to possess APR, which was consistent with previous tests of allelism, demonstrating that the qRphFlag resistant allele is common in leaf rust resistant cultivars derived from Vada and Emir. qRphFlag has been designated Rph20, the first gene conferring APR to P. hordei to be characterised in barley. The PCR marker will likely be effective in marker-assisted selection for Rph20.

Leaf rust, caused by Puccinia triticina, is the most prevalent and widespread disease of wheat in South America. The objective of this study was to determine whether genetically differentiated groups of P. triticina are currently present in South America and to compare the South American population with the previously characterized population in North America. In total, 130 isolates of P. triticina from the wheat-growing regions of Argentina, Brazil, Chile, Peru, and Uruguay, mostly from the 1990s to 2008, were tested for virulence on 20 lines of wheat with single genes for leaf rust resistance and for molecular genotypes with 23 simple-sequence repeat (SSR) markers. After removal of isolates with identical virulence and SSR genotypes, 99 isolates were included for further analysis. Principal coordinate analysis plots indicated five different groups of isolates based on SSR genotypes that also differed for virulence to leaf rust resistance genes. All pairs of groups, except for one pair, were significantly differentiated for SSR genotypes according to R(ST) statistics. All but two pairs of groups were significantly differentiated for virulence phenotype according to Phi(PT) statistics. Isolates in all five groups had high values of fixation index for SSR alleles and linkage disequilibrium was high across all isolates that indicated the clonal reproduction of urediniospores. Only one of the five P. triticina groups from South America was differentiated for SSR genotypes from all of the six P. triticina groups from North America. The high degree of similarity for SSR genotype of isolates from both South America and North America suggested a common European origin of P. triticina that was introduced to both continents. The emergence of the same P. triticina virulence phenotypes with highly related SSR genotypes in the United States in 1996 and in Uruguay in 1999 indicated the likely intercontinental migration of these genotypes from Mexico to both South America and North America.

En este artículo se presenta el estado actual del tratamiento con endoprótesis de las patologías de aorta, así como las indicaciones, las contraindicaciones y el futuro del tratamiento con este tipo de dispositivos.

Endovascular treatment of aortic pathologies is actually an alternative to open surgery. It has proven to be safe, showing similar or better results to those achieved by surgery. In this article, treatment of aortic pathologies by means of endoprosthesis is presented, as well as its indications, contraindications and future treatment with this kind of devices.

In September 2003, leaves exhibiting spot-type lesions similar to those produced by Cochliobolus sativus Drechs. ex Dastur were widely observed in six commercial barley crops of cvs. Norteña Daymán, Norteña Carumbé, and MUSA 936 in Soriano and Río Negro provinces, the main barley production region in western Uruguay. Spot lesions were tan to dark brown, circular to elliptical, and 3 to 10 mm in diameter. Larger lesions were surrounded by a chlorotic margin of varying width. Affected leaf pieces (10 to 15) from each field were placed in a moist chamber for 2 days to promote sporulation. A fungus identified morphologically as Pyrenophora teres (Died.) Drechs. (1) was consistently isolated from infected leaves. However, symptoms did not correspond to the net-type lesions of net blotch commonly produced by P. teres f. sp. teres in Uruguay. Three monoconidial cultures were obtained by transferring single conidia to potato dextrose agar and then to 10% V8 juice agar and incubated at 20 to 22°C with a 12-h photoperiod for 10 days. Adding sterile water to each plate and gently rubbing the surface with a microscope slide prepared inoculum for pathogenicity tests. Conidia concentration was adjusted to 1 × 104 conidia per ml. Sixty-eight barley genotypes from Uruguay, ICARDA/CIMMYT, and North Dakota were grown in the greenhouse for 2 weeks at 20 to 22°C with a 14-h photoperiod. For each monoconidial isolate, three seedlings of each genotype were inoculated at the three-leaf stage 15 to 16 days after seeding with 0.4 ml of the inoculum suspension with an airbrush inoculator. A drop of Tween 20 was added per 40 ml of inoculum suspension. One set of each genotype was inoculated with sterile water as a control. Seedlings were placed in a dew chamber at 20°C and 100% relative humidity in the dark for 24 h and then returned to prior conditions. The first lesions developed after 7 to 9 days. Leaves two and three of the plants were visually rated for disease (3) 13 days after inoculation. Control plants were disease free. The most susceptible reactions were observed on cvs. Norteña Daymán, MUSA 936, and line CLE 230 (Uruguay). Symptoms were similar in shape and size to those observed in the fields. The most resistant infection types were observed on several Uruguayan and North Dakota advanced lines. The fungus was consistently reisolated from inoculated plants. On the basis of morphology and symptoms produced, the pathogen was identified (2) as P. teres. f. sp. maculata Smedeg. La técnica de la oclusión rápida de la vía aérea al final de la inspiración permite medir separadamente el componente viscoelástico del pulmón y la caja torácica. En el presente estudio se analizan los efectos del isofluorano sobre la mecánica ventilatoria con esta técnica. Se evaluaron 10 ratas ventiladas mecanicamente y curarizadas. Se separaron en dos grupos, el 1 (n=5) recibió pentobarbital sódico (20 mg/kg) previa sedación con diazepam (5 mg) El grupo 2 recibió isofluorano a una concentración del 2 por ciento. Se midió volumen corriente, flujo aéreo y presiones esofágica, traqueal y transpulmonar. Se realizaron oclusiones rápidas de la vía aérea al final de la inspiración. No hubo diferencias significativas en el peso ni en el flujo y volumen corriente al que fueron ventiladas. La DPtot,rs y DP2,rs fueron significativamente mayores en el grupo anestesiado con isofluorano (p<0,05). La DP1,rs y la DP1,L no presentaron diferencias significativas. Las DP2,L fueron mayores en el grupo 2 (x = 2,041 cm H2O ES = ñ 0,191) que en el grupo 1 (x =1,190 cm H2O; ES = ñ 0,092). También hubo diferencias en el DE,L, siendo menor para el grupo 1 que para el 2 (x = 0,578 cm H2O.ml-1 ES = ñ 0,048, y x =1,007 cm H2Oml-1 ES = ñ 0,085, respectivamente). La elasticidad estática y dinámica del pulmón no presentaron diferencias significativas. Concluimos que el isofluorano determina un aumento de las presiones viscoelásticas del pulmón. Esto lleva a un incremento de la DE, L. No se produjeron modificaciones en las presiones de la vía aérea central ni en la elasticidad(AU)

Se estudiaron 27 pacientes con Enfermedad Pulmonar Obstructiva Crónica (EPOC) con diagnóstico clínico-radiológico y espirométrico. Durante la respiración tranquila se midieron: la complacencia pulmonar estática (CE), la complacencia pulmonar dinámica (CD) y la resistencia pulmonar total (RPT). Se compararon los resultados con los de una población normal. La CE del grupo de pacientes con EPOC fue de 0.515ñ0.385 L/cm H²O y la del grupo control 0.192ñ0.039 L/cm H²O (p<0.05). La RPT de los pacientes con EPOC fue de 5.6ñ4.3 cm H²O/L/s, significativamente mayor que la del grupo control: 2.64ñ1.07 cm H²O/L/s (p<0.05). No existió diferencia significativa en los valores de la CD entre ambos grupos. No se encontró correlación entre la complacencia estática y la capacidad vital. Tampoco existió correlación de la CD o la RPT con los parámetros espirométricos indicadores de limitación del flujo espiratorio (CVF, VEF1s, VEF1s/CVF Y FEF 25-75 por ciento). Se concluye que el estudio de la mecánica ventilatoria durante la respiración tranquila en el EPOC, aporta información sobre la distensibilidad pulmonar y el estado de la vía aérea que es complementaria de la obtenida mediante la espirometría forzada (AU)
