Infectious Bronchitis Virus (IBV) is a major threat to the poultry industry worldwide, causing significant economic losses. While the virus's genetic structure is well understood, the specific strains circulating in Bolivia have remained uncharacterized until now. This study aimed to identify and characterize new IBV strains in Bolivia. Tissue samples from broilers exhibiting clinical signs of Infectious Bronchitis were screened to detect IBV using real-time RT-PCR (RT-qPCR). Positive samples with low cycle threshold (Ct) values were selected for sequencing the full S1 gene. Of the 12 samples analyzed, 10 were determined to be positive for IBV. However, only four samples yielded sufficient genetic material for sequencing and subsequent phylogenetic analysis. The results revealed the presence of GI-1 and GI-23 lineages, both belonging to genotype I (GI). The GI-1 lineage showed >99% sequence identity to the H120 and Massachusetts vaccine strains, suggesting a close relationship. In contrast, the GI-23 lineage clustered with other IBV strains, showing a distinct subclade that is genetically distant from Brazilian strains. No evidence of recombination was found. Furthermore, amino acid substitution analysis identified specific mutations in the S1 subunit, particularly in the hypervariable regions 1, 2, and 3. These mutations could potentially alter the virus's antigenicity, leading to reduced vaccine efficacy. The findings of this study highlight the importance of continued and broad genomic surveillance of circulating IBV strains and the need to improve vaccination strategies in Bolivia.

Background: Syphilis is a serious global public health challenge. Despite prior progress in syphilis control, incidence has been increasing in recent years. Syphilis is a common coinfection among people living with HIV (PLHIV). In Panama, few data describe syphilis prevalence among PLHIV. We describe syphilis antibody and high-titer (⩾1:8) active syphilis prevalence and associated factors among individuals who attended an antiretroviral clinic. Methods: A cross-sectional study was undertaken during February-March 2022 and September-October 2022 for adults (⩾18 year) assigned male and female at birth, respectively. Participants provided peripheral blood samples and self-administered a questionnaire. Samples were screened using immunochromatography; antibody-positive samples were tested using rapid plasma regain to 1:512 dilutions. Logistic regression was used to identify factors associated with syphilis antibody and high-titer active syphilis. Results: In all, 378 participants gave blood samples; 377 individuals participated in the questionnaire (216 self-reported male sex [males], 158 female [females], and three intersex individuals). Median age was 36 years (interquartile range: 28-45 years). Overall, syphilis antibody prevalence was 32.3% (122/378) (males, 50.7% [108/2013]; females, 5.7% [9/158]; intersex individuals, 100.0% (3/3)], p < 0.01. High-titer active syphilis was found among 24.6% (n = 30) of samples with positive antibody test (males 27.8% [n = 30], females 0.0% [0/9], intersex individuals 0.0% [0/3]). Antibody positivity was associated in the multivariable model with males (50.7%, AOR = 24.6, 95%CI: 1.57-384.53). High-titer active syphilis was associated with younger participant age (18-30 years, 13.2%, OR = 4.82, 95%CI: 1.17-19.83); 31-40 years, 7.8%, OR = 4.24, 95%CI: 1.04-17.21 versus 3.2% >40 years), homosexual identity (16.0% OR = 34.2, 95%CI: 4.50-259.27 versus 0.6% among heterosexual identity); in the multivariable model, associated with sexual identity (bisexual 19.1%, AOR = 10.89, 95%CI: 1.00-119.06) compared to heterosexual identity (0.6%) and weakly associated with concurrency (⩾1 ongoing sexual relationships, 15.9%, AOR = 3.09, 95%CI: 0.94-10.14). Conclusion: This study found very high prevalence of syphilis antibodies and high-titer syphilis among PLHIV in Panama. Those most affected are males, younger in age, those who practice concurrent sexual relationships, and those who reported homosexual and bisexual identity. Targeted interventions should include repetitive testing and treatment, especially among individuals who may be at increased infection risk.
Prevalence of syphilis among people living with HIV who attend a large antiretroviral therapy clinic, Panama, 2022 Syphilis is a significant health challenge worldwide. On a global scale, yearly syphilis incidence is increasing, including in Panama. However, there are no current data to explain syphilis prevalence and who is most affected among people living with HIV in Panama. In order to understand syphilis in Panama and create targeted interventions among specific groups of people, it is important to describe how many people are infected, and who is most affected by this infection. Therefore, we conducted a study among 378 people living with HIV at a treatment clinic in Panama City, Panama. Blood samples and demographic data were collected. In all, syphilis antibodies were found in 32.3% of individuals (50.7% of those who self-report as males, 5.7% as females, and 100% of those who self-report as intersex. Of those with positive antibody tests, 24.6% of individuals also had active syphilis. Only individuals who identify as male had active syphilis. Our findings show high syphilis prevalence among people with HIV in Panama City, particularly among males, those who are younger, those who report sexual identity as homosexual or bisexual, and those with ongoing sexual relationships with more than one individual. Targeted interventions are needed among people living with HIV, especially among the groups most affected. These interventions could include testing more often for syphilis and providing timely treatment, especially among individuals who may be at increased risk of infection.

Genome sequencing of highly virulent Salmonella enterica subsp. enterica serovar Javiana strain FARPER-220 (ST-1674) isolated from broiler chickens in Peru revealed multiple virulence factors, antibiotic resistance genes, and invasion-related subcategories. The results provide insights into the potential importance of this strain in causing infections in various animals.

BACKGROUND: Newcastle disease (ND) is a major threat to the poultry industry, leading to significant economic losses. The current ND vaccines, usually based on active or attenuated strains, are only partially effective and can cause adverse effects post-vaccination. Therefore, the development of safer and more efficient vaccines is necessary. Epitopes represent the antigenic portion of the pathogen and their identification and use for immunization could lead to safer and more effective vaccines. However, the prediction of protective epitopes for a pathogen is a major challenge, especially taking into account the immune system of the target species. RESULTS: In this study, we utilized an artificial intelligence algorithm to predict ND virus (NDV) peptides that exhibit high affinity to the chicken MHC-I complex. We selected the peptides that are conserved across different NDV genotypes and absent in the chicken proteome. From the filtered peptides, we synthesized the five peptides with the highest affinities for the L, HN, and F proteins of NDV. We evaluated these peptides in-vitro for their ability to elicit cell-mediated immunity, which was measured by the lymphocyte proliferation in spleen cells of chickens previously immunized with NDV. CONCLUSIONS: Our study identified five peptides with high affinity to MHC-I that have the potential to serve as protective epitopes and could be utilized for the development of multi-epitope NDV vaccines. This approach can provide a safer and more efficient method for NDV immunization.

Introducción: El desarrollo psicomotor es un proceso gradual y continuo; las familias cubanas necesitan herramientas que les permitan vigilarlo. Objetivo: Describir las percepciones que poseen las familias de la vigilancia del desarrollo psicomotor de sus hijos menores de un año. Métodos: Se realizó una investigación cualitativa, de tipo fenomenológica, en el Consultorio del Médico y la Enfermera de la Familia No. 9 del Policlínico Universitario Héroes del Moncada, La Habana, de noviembre 2019 a febrero 2022. El referente teórico fue el Modelo de Traducción del Conocimiento. Las unidades de análisis fueron ocho familias que tenían un niño menor de un año, a las que se aplicó el Método Creativo Sensible, mediante la dinámica de arte del Árbol del conocimiento. Se grabaron las sesiones, y las narrativas fueron transcritas en un cuerpo textual a las que se les realizó análisis de contenido. Resultados: Las familias expresaron que la llegada del niño significó alegría y unión familiar; así como la necesidad de contar con diferentes materiales que ayudaran para la vigilancia del desarrollo psicomotor. De las 33 pautas analizadas fue necesario traducir tres al lenguaje familiar. Conclusiones: El nacimiento de un hijo tuvo un efecto favorable. La dinámica empleada fue efectiva para el propósito de la descripción de las pautas del desarrollo psicomotor para niños cubanos por las familias estudiadas. Introduction: Psychomotor development is a gradual and continuous process; Cuban families need tools that allow them to monitor it. Objective: To describe the families' perceptions with respect to monitoring the psychomotor development of their infants aged less than one year. Methods: A qualitative research, of phenomenological type, was carried out in the family doctor and nurse's office 9, belonging to Policlínico Universitario Heroes del Moncada, of Havana, from November 2019 to February 2022. The theoretical reference was the knowledge translation model. The units of analysis were 8 families with an infant aged less than one year and applied the creative sensitive method, using the art dynamics of the tree of knowledge. The sessions were recorded, and the narratives were transcribed into a textual body later subjected to content analysis. Results: The families expressed that the arrival of the child meant joy and family unity, as well as the need to have different materials that help monitor the child's psychomotor development. Of the 33 guidelines analyzed, it was necessary to translate three into a family language. Conclusions: The birth of a child had a favorable effect. The used dynamics was effective for the purpose that the studied families describe psychomotor development guidelines for their Cuban children. The need for some kind of tool for monitoring of psychomotor development was evidenced(AU)

Por medio de la presente y motivados por la lectura y análisis del artículo Características clínicas y microbiológicas de la infección de vías urinarias bajas en población ambulatoria,1 quisiéramos llamar la atención sobre un tema que últimamente tiene poca visibilidad en las publicaciones. Se trata de los factores de riesgo presentes en la edad reproductiva y que inciden negativamente en futuros embarazos, lo que se conoce como factores de riesgo preconcepcional. Al revisar lo publicado en los últimos cinco años apenas se aborda directamente el tema, que por demás mantiene vigencia capital cuando de salud materno infantil se trata. Surveillance helps us identify and monitor strains with zoonotic potential. A tracheal swab from a pelican on a Peruvian beach was H5N1 positive (clade 2.3.4.4b) using Oxford Nanopore's MinION platform. The near-complete genome sequence of strain VFAR-140 will aid us in understanding avian influenza epidemiology and spread.

This study presents a draft genome sequence of a Newcastle disease virus (NDV) strain (VFAR-136) isolated from a fighting cock (Gallus gallus) in the south of Peru. Strain VFAR-136 is a new report of NDV genotype VII circulating in Peru.

Within the framework of the current COVID-19 pandemic, there is a race against time to find therapies for the outbreak to be controlled. Since vaccines are still tedious to develop and partially available for low-income countries, passive immunity based on egg-yolk antibodies (IgY) is presented as a suitable approach to preclude potential death of infected patients, based on its high specificity/avidity/production yield, cost-effective manufacture, and ease of administration. In the present study, IgY antibodies against a recombinant RBD protein of SARS-CoV-2 were produced in specific-pathogen-free chickens and purified from eggs using a biocompatible method. In vitro immunoreactivity was tested, finding high recognition and neutralization values. Safety was also demonstrated prior to efficacy evaluation, in which body weight, kinematics, and histopathological assessments of hamsters challenged with SARS-CoV-2 were performed, showing a protective effect administering IgY intranasally both as a prophylactic treatment or a post-infection treatment. The results of this study showed that intranasally delivered IgY has the potential to both aid in prevention and in overcoming COVID-19 infection, which should be very useful to control the advance of the current pandemic and the associated mortality.

In this study, we developed a new recombinant virus rHVT-F using a Turkey herpesvirus (HVT) vector, expressing the fusion (F) protein of the genotype XII Newcastle disease virus (NDV) circulating in Peru. We evaluated the viral shedding and efficacy against the NDV genotype XII challenge in specific pathogen-free (SPF) chickens. The F protein expression cassette was inserted in the unique long (UL) UL45-UL46 intergenic locus of the HVT genome by utilizing a clustered regularly interspaced short palindromic repeat (CRISPR)/Cas9 gene-editing technology via a non-homologous end joining (NHEJ) repair pathway. The rHVT-F virus, which expressed the F protein stably in vitro and in vivo, showed similar growth kinetics to the wild-type HVT (wtHVT) virus. The F protein expression of the rHVT-F virus was detected by an indirect immunofluorescence assay (IFA), Western blotting, and a flow cytometry assay. The presence of an NDV-specific IgY antibody was detected in serum samples by an enzyme-linked immunosorbent assay (ELISA) in SPF chickens vaccinated with the rHVT-F virus. In the challenge experiment, the rHVT-F vaccine fully protects a high, and significantly reduced, virus shedding in oral at 5 days post-challenge (dpc). In conclusion, this new rHVT-F vaccine candidate is capable of fully protecting SPF chickens against the genotype XII challenge.

City plans symbolizing cosmologies have long been recognized as a defining element of Mesoamerican civilizations. The origins of formal spatial configurations are thus the key to understanding early civilizations in the region. Assessment of this issue, however, has been hindered by the lack of systematic studies of site plans over broad areas. Here, we report the identification of 478 formal rectangular and square complexes, probably dating from 1,050 to 400 BC, through a lidar (laser imaging, detection and ranging) survey across the Olmec region and the western Maya lowlands. Our analysis of lidar data also revealed that the earlier Olmec centre of San Lorenzo had a central rectangular space, which possibly provided the spatial template for later sites. This format was probably formalized and spread after the decline of San Lorenzo through intensive interaction across various regions. These observations highlight the legacy of San Lorenzo and the critical role of inter-regional interaction.

As humans are the primary geomorphic agents on the landscape, it is essential to assess the magnitude, chronological span, and future effects of artificial ground that is expanding under modern urbanization at an alarming rate. We argue humans have been primary geomorphic agents of landscapes since the rise of early urbanism that continue to structure our everyday lives. Past and present anthropogenic actions mold a dynamic "taskscape" (not just a landscape) onto the physical environment. For example, one of the largest Pre-Columbian metropolitan centers of the New World, the UNESCO world heritage site of Teotihuacan, demonstrates how past anthropogenic actions continue to inform the modern taskscape, including modern street and land alignments. This paper applies a multi-scalar, long durée approach to urban landscapes utilizing the first lidar map of the Teotihuacan Valley to create a geospatial database that links modern and topographic features visible on the lidar map with ground survey, historic survey, and excavation data. Already, we have recorded not only new features previously unrecognized by historic surveys, but also the complete erasure of archaeological features due to modern (post-2015) mining operations. The lidar map database will continue to evolve with the dynamic landscape, able to assess continuity and changes on the Teotihuacan Valley, which can benefit decision makers contemplating the stewardship, transformation, or destruction of this heritage landscape.

SARS-CoV-2 main protease is a common target for inhibition assays due to its high conservation among coronaviruses. Since flavonoids show antiviral activity, several in silico works have proposed them as potential SARS-CoV-2 main protease inhibitors. Nonetheless, there is reason to doubt certain results given the lack of consideration for flavonoid promiscuity or main protease plasticity, usage of short library sizes, absence of control molecules and/or the limitation of the methodology to a single target site. Here, we report a virtual screening study where dorsilurin E, euchrenone a11, sanggenol O and CHEMBL2171598 are proposed to inhibit main protease through different pathways. Remarkably, novel structural mechanisms were observed after sanggenol O and CHEMBL2171598 bound to experimentally proven allosteric sites. The former drastically affected the active site, while the latter triggered a hinge movement which has been previously reported for an inactive SARS-CoV main protease mutant. The use of a curated database of 4.8 k flavonoids, combining two well-known docking software (AutoDock Vina and AutoDock4.2), molecular dynamics and MMPBSA, guaranteed an adequate analysis and robust interpretation. These criteria can be considered for future screening campaigns against SARS-CoV-2 main protease.

The application of lidar remote-sensing technology has revolutionized the practice of settlement and landscape archaeology, perhaps nowhere more so than in the Maya lowlands. This contribution presents a substantial lidar dataset from the Puuc region of Yucatan, Mexico, a cultural subregion of the ancient Maya and a distinct physiographic zone within the Yucatan peninsula. Despite the high density of known sites, no large site has been fully surveyed, and little is known about intersite demography. Lidar technology allows determination of settlement distribution for the first time, showing that population was elevated but nucleated, although without any evidence of defensive features. Population estimates suggest a region among the most densely settled within the Maya lowlands, though hinterland levels are modest. Lacking natural bodies of surface water, the ancient Puuc inhabitants relied upon various storage technologies, primarily chultuns (cisterns) and aguadas (natural or modified reservoirs for potable water). Both are visible in the lidar imagery, allowing calculation of aguada capacities by means of GIS software. The imagery also demonstrates an intensive and widespread stone working industry. Ovens visible in the imagery were probably used for the production of lime, used for construction purposes and perhaps also as a softening agent for maize. Quarries can also be discerned, including in some cases substantial portions of entire hills. With respect to agriculture, terrain classification permits identification of patches of prime cultivable land and calculation of their extents. Lidar imagery also provides the first unequivocal evidence for terracing in the Puuc, indeed in all northern Yucatan. Finally, several types of civic architecture and architectural complexes are visible, including four large acropolises probably dating to the Middle Formative period (700-450 B.C.). Later instances of civic architecture include numerous Early Puuc Civic Complexes, suggesting a common form of civic organization at the beginning of the Late Classic demographic surge, (A.D. 600-750).

Introducción: En el mundo moderno, el traumatismo es la primera causa de mortalidad y discapacidad en menores de 35 años. Múltiples son los esfuerzos para tratar de definir su pronóstico desde el momento en que se produce. Con el decurso de los años se han creado varias escalas, para describir la gravedad de las lesiones, las alteraciones fisiológicas que se producen, así como para evaluar los sistemas de atención. Objetivo: Describir el comportamiento de los lesionados en el Hospital "Calixto García" según variables. Métodos: Se realizó un estudio observacional descriptivo, prospectivo y longitudinal en 1582 lesionados atendidos en el servicio de Cirugía General. Resultados: El intervalo trauma/ingreso superó los 60 minutos en un número elevado y la mitad de ellos presentó parámetros clínicos de inestabilidad hemodinámica en la recepción; se logró su compensación en la primera hora de admitidos en 7 de cada 10 lesionados. Más de la mitad desarrolló alguna complicación y predominó la estadía hospitalaria entre 15 y 21 días. Conclusiones: Algunas de las variables estudiadas se comportaron similar a la literatura consultada. Los resultados sugieren la necesidad de implementar un programa de atención prehospitalaria al lesionado más efectivo para mejorar la atención y el índice de sobrevida en los mismos(AU)
Introduction: In the modern world, trauma is the leading cause of death and disability in people under 35 years of age. Multiple efforts are made to define its prognosis from the moment it occurs. Over the years, several scales have been created to describe the severity of injuries, the physiological changes that occur, as well as to assess care systems. Objective: To describe the behavior of injured patients in Calixto García Hospital according to variables. Methods: A observational, descriptive, prospective and longitudinal study was carried out with 1582 injured patients treated in the general surgery service. Results: The trauma/admission interval exceeded sixty minutes in a high number of cases. Half of them presented clinical parameters of hemodynamic instability at admission. Compensation was achieved at the first hour of admission in seven out of ten injured patients. More than half the patients developed some complication. There was a predomination of hospital stay between 15 and 21 days. Conclusions: Some of the variables studied behaved similarly to the way described in the consulted literature. The results suggest the need to implement a more effective prehospital care program for the injured patients to improve care and survival rate(AU)

INTRODUCCIÓN: Desde la terapia ocupacional se identifica una serie de labores imprescindibles en la intermediación laboral de personas en situación de discapacidad (PeSD). La incorporación de estos elementos, en la Ley chilena N°21.015, es de especial importancia para lograr una inclusión laboral efectiva. OBJETIVO: Relevar el rol que cumplen los terapeutas ocupacionales en la inclusión laboral de PeSD en el marco de la Ley 21.015. MÉTODO: Esta investigación es de tipo cualitativa, específicamente un estudio descriptivo y prospectivo, en el que se utilizó un diseño fenomenológico hermenéutico. La muestra fue no probabilística por redes, en la que se entrevistó a siete terapeutas ocupacionales con experiencia en inclusión laboral de PeSD. RESULTADOS: Los terapeutas ocupacionales reconocen tareas de su rol en cada una de las etapas del proceso de intermediación laboral, las cuales varían según el usuario objetivo del profesional y el origen de la discapacidad de la persona incluida. Los entrevistados consideran que la Ley 21.015 no contempla la mayoría de estas tareas y tampoco establece cuáles son los profesionales capacitados para la realización de este proceso. Los terapeutas ocupacionales justifican la relevancia de su rol en base a la mirada holística que poseen de la persona y el conocimiento especializado de la discapacidad. CONCLUSIONES: La terapia ocupacional es una profesión relevante en todo el proceso de intermediación laboral. Este proceso es una labor que desempeña un equipo transdisciplinario y, en el que, además, es de suma importancia que sea un trabajo intersectorial para que se logren resultados eficaces y satisfactorios.
INTRODUCTION: Due to occupational therapy, a series of essential tasks in the labour intermediation of people with disabilities are identified. The incorporation of the elements listed in the Chilean Law 21.015 is of vital importance to achieve an effective labour inclusion. OBJECTIVE: Demonstrate the role that occupational therapists play in relation to the labour inclusion of people with disabilities, regarding the Law 21.015. METHOD: This is a qualitative investigation, specifically a descriptive and prospective study, in which a hermeneutics phenomological design was used. The sample was not probabilistic in networks, in which seven experienced occupational therapists in labour inclusion of people with disabilities were interviewed RESULTS: Occupational therapists recognized the importance of their role in each stage of the labour intermediation, which varies between the target users of the professional and the origen of the disability of the person. The persons interviewed recognized that most of these tasks are not included in the Law 21.015, nor does it establish which are the qualified professionals for the realization of this process. Based on the holistic view of the person and their specific knowledge about disability, occupational therapists justify the importance of their role. CONCLUSION: The occupational therapist is of vital importance in all the processes involving labour intermediation. The process is carried by a transdisciplinary team, where by effective and satisfactory results are achieved thanks to intersectorial work.

RESUMEN Introducción: La perforación del esófago constituye una de las urgencias más graves y difíciles que ha de afrontar un cirujano por las características y ubicación del órgano. El pronóstico depende sobre todo de la rapidez del diagnóstico y de la elección del tratamiento instaurado en principio. Objetivo: Presentar un caso portador del Síndrome de Boerhaave. Caso clínico: Paciente masculino en la 5ta década de vida que acudió por dolor torácico posterior a cuadro emético. Luego de estudio radiográfico se diagnosticó ruptura espontánea de esófago o síndrome de Boerhaave. Conclusiones: El enfoque terapéutico adecuado asociado al diagnóstico oportuno y precoz del síndrome garantiza mejores índices de sobrevida(AU)
ABSTRACT Introduction: Esophageal perforation is one of the most serious and difficult emergencies that a surgeon has to face due to the characteristics and location of the organ. Such prognosis depends mainly on the speed of the diagnosis and the choice of treatment established initially. Objectives: To present a case with such syndrome and to review the literature to update the therapeutic approach of this entity given its high mortality. Clinical case: Male patient in the fifth decade of life who presented for chest pain after an emetic condition. After a radiographic study, a spontaneous rupture of the esophagus or Boerhaave syndrome was diagnosed. Conclusions: The appropriate therapeutic approach associated with the early and timely diagnosis of the syndrome guarantees better survival rates(AU)

Infectious coryza (IC), an upper respiratory tract disease affecting chickens, is caused by Avibacterium paragallinarum. The clinical manifestations of IC include nasal discharge, facial swelling, and lacrimation. This acute disease results in high morbidity and low mortality, while the course of the disease is prolonged and mortality rates are increased in cases with secondary infections. Studies regarding the immune response in infected chickens are scarce, and the local immune response is the focal point of investigation. However, a large body of work has demonstrated that severe infections can impact the systemic immune response. The objective of this study was to evaluate the systemic effects of Avibacterium paragallinarum (serovar B-1) infection on immune cells in specific pathogen-free (SPF) chickens. The current study revealed the presence of a transient circulating monocyte population endowed with high phagocytic ability and clear downregulation of major histocompatibility complex class II (MHC-II) surface expression. In human and mouse studies, this monocyte population (identified as tolerant monocytes) has been correlated with a dysfunctional immune response, increasing the risk of secondary infections and mortality. Consistent with this dysfunctional immune response, we demonstrate that B cells from infected chickens produced fewer antibodies than those from control chickens. Moreover, T cells isolated from the peripheral blood of infected chickens had a lower ability to proliferate in response to concanavalin A than those isolated from control chickens. These findings could be related to the severe clinical signs observed in complicated IC caused by the presence of secondary infections.

A series of defect pyrochlores of the composition (H3O)1+pSb1+pTe1-pO6 have been prepared by ion exchange from K-containing pyrochlores K1+pSb1+pTe1-pO6 in sulfuric acid at 280 °C for 24 h. The structural characterization of the hydronium-containing pyrochlores, including the location of the H3O+ units within the three-dimensional framework, was possible from neutron powder diffraction data in undeuterated samples. The crystal structure for all the compounds is defined in the Fd3[combining macron]m space group, and consists of a covalent framework of SbVO6 and TeVIO6 octahedra distributed at random and connected by their vertices with (Sb,Te)-O1-(Sb,Te) angles close to 136°, conforming to large cages where the hydronium species are located off-center. The absence of K+ ions in the ion-exchanged pyrochlores was confirmed by inductively coupled plasma optical emission spectroscopy and scanning electron microscopy coupled with energy dispersive X-ray spectroscopy. The shape and size of the hydronium units evolve along with the series, becoming more compact as the framework covalence and Lewis-basicity decrease upon Sb enrichment of the structure (for greater p values). The amount and lability of the H3O+ species also increase throughout the series, as wanted: a straightforward correlation of the catalytic activity in the fructose dehydration reaction to 5-hydroxymethylfurfural has been observed, reaching conversion rates up to 88.5% of concentrated fructose solution for the p = 0.25 catalyst. Moreover, a pseudo-first-order kinetic mechanism was simulated, and the kinetic constants obtained from diluted and concentrated enhanced reaction systems were determined and compared.

BACKGROUND: In the poultry industry, quantitative analysis of chicken T cell proliferation is important in many biological applications such as drug screening, vaccine production, and cytotoxicity assessment. Several assays have been established to evaluate this immunological response in chicken cells. However, these assays have some disadvantages including use of radioactive labels ([3H]-Thymidine assay), necessity of DNA denaturation or digestion (BrdU incorporation assay), lack of sensitivity and underestimation of anti-proliferative effects (MTT assay), and modulation of activation molecules and cell viability reduction (CFSE assay). Overcoming these limitations, the EdU proliferation assay is sensitive and advantageous compared to [3H]-Thymidine radioactive labels in studies on cell proliferation in vitro and allows simultaneous identification of T cell populations. However, this assay has not been established using primary chicken cells to evaluate T cell proliferation by flow cytometry. RESULTS: Here, we established an assay to evaluate the proliferation of primary chicken splenocytes based on the incorporation of a thymidine analog (EdU) and a click reaction with a fluorescent azide, detected by a flow cytometer. We also established a protocol that combines EdU incorporation and immunostaining to detect CD4+ and CD8+ proliferating T cells. By inducing cell proliferation with increasing concentrations of a mitogen (Concanavalin A), we observed a linear increase in EdU positive cells, indicating that our protocol does not present any deficiency in the quantity and quality of reagents that were used to perform the click reaction. CONCLUSIONS: In summary, we established a reliable protocol to evaluate the proliferation of CD4+ and CD8+ chicken T cells by flow cytometry. Moreover, as this is an in-house protocol, the cost per sample using this protocol is low, allowing its implementation in laboratories that process a large number of samples.
