In the present work, two mathematical diffusion models have been used to estimate the growth of the iron monoboride and diiron boride coating formed on AISI 420 steel. The boronizing of the steel was carried out with the solid diffusion packing method at a boronizing temperature of 1123 K-1273 K. Experimental results show the two-coating system consists of an outer monoboride and an inner diiron boride coating with a predominantly planar structure at the propagation front. The depth of the boride coating increases according to temperature and treatment time. A parabolic curve characterizes the propagation of the boride coatings. The two proposed mathematical models of mass transfer diffusion are founded on the solution corresponding to Fick's second fundamental law. The first is based on a linear boron concentration-penetration profile without time dependence, and the second model with time dependence (exact solution). For both models, the theoretical law of parabolic propagation and the average flux of boron atoms (Fick's first fundamental law) at the growth interfaces (monoboride/diiron boride and diiron boride/substrate) are considered to estimate the propagation of the boride coatings (monoboride and diiron boride). To validate the mathematical models, a programming code is written in the MATLAB program (adaptation 7.5) designed to simulate the growth of the boride coatings (monoboride and diiron boride). The following parameters are used as input data for this computer code: (the layer thicknesses of the FeB and Fe2B phases, the operating temperature, the boronizing time, initial formation time of the boride coating, the surface boron concentration limits, FeB/Fe2B and Fe2B/Fe growth interfaces, and the mass transfer diffusion coefficient of boron in the iron monoboride and diiron boride phases). The outputs of the computer code are the constants εFeB and εFe2B. The assessment of activation energies of AISI 420 steel for the two mathematical models of mass transfer is coincident (QFeB=221.9 kJ∙mol-1 and QFe2B=209.1 kJ∙mol-1). A numerical analysis was performed using a standard Taylor series for clarification of the proximity between the two models. SEM micrographs exhibited a strong propensity toward a flat-fronted composition at expansion interfaces of the iron monoboride and diiron boride coating, confirmed by XRD analysis. Tribological characterizations included the Vickers hardness test method, pin-on-disc, and Daimler-Benz Rockwell-C indentation adhesion tests. After thorough analysis, the energies were compared to the existing literature to validate our experiment. We found that our models and experimental results agreed. The diffusion models we utilized were crucial in gaining a deeper understanding of the boronizing behavior of AISI 420 steel, and they also allowed us to predict the thicknesses of the iron monoboride and diiron boride coating. These models provide helpful approaches for predicting the behavior of these steels.

Hard-coated surfacing of a few micrometers is widely applied to increase the efficiency of tools, e.g., for cutting, forming, and casting applications. Therefore, the base thermodiffusion surface treatment is a practical solution to these issues by hardening surface layers with interstitial elements such as carbon, nitrogen, and boron. In particular, within this study, the growth kinetics of an iron boride layer on ASTM 283 steel were investigated with two diffusion models of the powder-pack boriding technique in the temperature range of 1123-1273 K with different treatment periods. The first model, called the steady-state diffusion model, used the modified version of the mass balance equations at the Fe2B/substrate growth interface, the parabolic growth law, and the solution of Fick's second law without time dependence. At the same time, the second diffusion model was based on Goodman's method, also called the integral heat balance method. Afterward, the diffusion coefficient of boron in the Fe2B phase was calculated by fitting the experimental data to the models. Nevertheless, the estimated value for the activation energy of ASTM A238 steel in both diffusion models was coincident (168.2 kJ∙mol-1). A mathematical analysis was implemented by means of a power series (Taylor series) to explain this similarity. The SEM examinations showed a solid tendency to saw-tooth morphology at the growth interface with the formation of the Fe2B layer, whose presence was verified by XRD analysis. The tribological characterizations, including the tests of Rockwell-C indentation, pin-on-disc, and Vickers hardness test method, were used to analyze the antiwear features of the Fe2B layers. Finally, this value of energy was compared to the literature for its experimental validation.

In addition to the UPR pathway, yeast cells require components of the HOG pathway to respond to ER stress. In this work, we found that unphosphorylated Sln1 and Ssk1 are required to mount an appropriate response to Tn. We also found that the MAPKKKs Ssk2 participates in the Tn response, but its osmo-redundant protein Ssk22 does not. We also found that the Pbs2 docking sites for Ssk2 (RDS-I and KD) are partially dispensable when mutated separately; however, the prevention of Ssk2 binding to Pbs2, by the simultaneous mutation of RDS-I and KD, caused strong sensitivity to Tn. In agreement with the lack of Hog1 phosphorylation during Tn treatment, a moderate resistance to Tn is obtained when a Pbs2 version lacking its kinase activity is expressed; however, the presence of mutual Pbs2-Hog1 docking sites is essential for the Tn response. Finally, we detected that Tn induced a transcriptional activation of some components of the SLN1 branch. These results indicate that the Tn response requires a complex formed by the MAPK module and components of the SLN1 branch but not their canonical osmoregulatory activities.

Composting and vermicomposting before addition to the soil is a viable alternative to the disposal of cattle manure. However, this residue has been used in the untreated form for seedling production. This study evaluated the use of cattle manure in natura, and the organic composted or vermicomposted variants of cattle manure on substrates used in the production of Eucalyptus urograndis seedlings. The treatments consisted of substrates formulated by mixing the organic fertilizers with washed sand in 20, 40, 60, 80 or 100 (v/v) percentages, compared to a commercial substrate. The E. urograndis seedlings were grown in tubes in a greenhouse and evaluated after 120 days for shoot and root dry weights, plant height, stem diameter, shoot/root ratio, and Dickson quality index. The substrate formulated from a mixture of 80% (v/v) of cattle manure in natura and washed sand allows for better development in Eucalyptus seedlings. Substrate containing 100% bovine manure vermicompost or organic compost produced seedlings with lower quality than in natura bovine manure but superior to the commercial substrate. Due to the health risks associated with use of untreated bovine manure, organic compost and vermicompost are good alternatives for the production of E. urograndis seedlings.


Uma alternativa para destinar corretamente o esterco bovino é realizar a compostagem e/ou vermicompostagem e depois adicioná-lo ao solo como fonte de nutrientes às plantas. Porém, este resíduo tem sido utilizado na produção de mudas sem tratamento. O estudo teve por objetivo avaliar o uso de esterco bovino in natura, de composto orgânico e de vermicomposto, produzidos a partir do esterco bovino, em substratos para a produção de mudas de Eucalyptus urograndis. Os tratamentos constituíram-se dos substratos formulados pela mistura dos fertilizantes orgânicos com areia lavada nas porcentagens de 20, 40, 60, 80 e 100 (v/v), em comparação com um substrato comercial (SC). As mudas de Eucalyptus urograndis foram cultivadas em tubetes, em casa de vegetação, e avaliadas aos 120 dias quanto a massa seca da parte aérea e raiz, altura da planta, diâmetro do caule, relação parte aérea/raiz e índice de qualidade de Dickson. O substrato formulado pela mistura de 80% (v/v) de esterco bovino in natura com areia lavada proporcionou o melhor desenvolvimento das mudas de eucalipto. O substrato contendo 100% de vermicomposto de esterco bovino ou composto orgânico produziu mudas com qualidade inferior ao esterco bovino in natura, mas superior ao substrato comercial. Devido aos riscos à saúde associados ao uso de esterco bovino não tratado, o composto orgânico e o vermicomposto são boas alternativas para a produção de mudas de E. urograndis.

In the yeast Saccharomyces cerevisiae, components of the High Osmolarity Glycerol (HOG) pathway are important for the response to diverse stresses including response to endoplasmic reticulum stress (ER stress), which is produced by the accumulation of unfolded proteins in the lumen of this organelle. Accumulation of unfolded proteins may be due to the inhibition of protein N-glycosylation, which can be achieved by treatment with the antibiotic tunicamycin (Tn). In this work we were interested in finding proteins involved in the ER stress response regulated by Hog1, the mitogen activated protein kinase (MAPK) of the HOG pathway. A high gene dosage suppression screening allowed us to identify genes that suppressed the sensitivity to Tn shown by a hog1Δ mutant. The suppressors participate in a limited number of cellular processes, including lipid/carbohydrate biosynthesis and protein glycosylation, vesicle-mediated transport and exocytosis, cell wall organization and biogenesis, and cell detoxification processes. The finding of suppressors Rer2 and Srt1, which participate in the dolichol biosynthesis pathway revealed that the hog1Δ strain has a defective polyprenol metabolism. This work uncovers new genetic and functional interactors of Hog1 and contributes to a better understanding of the participation of this MAPK in the ER stress response.

Eukaryotic cells have evolved signalling pathways that allow adaptation to harmful conditions that disrupt endoplasmic reticulum (ER) homeostasis. When the function of the ER is compromised in a condition known as ER stress, the cell triggers the unfolded protein response (UPR) in order to restore ER homeostasis. Accumulation of misfolded proteins due to stress conditions activates the UPR pathway. In mammalian cells, the UPR is composed of three branches, each containing an ER sensor (PERK, ATF6 and IRE1). However, in yeast species, the only sensor present is the inositol-requiring enzyme Ire1. To cope with unfolded protein accumulation, Ire1 triggers either a transcriptional response mediated by a transcriptional factor that belongs to the bZIP transcription factor family or an mRNA degradation process. In this review, we address the current knowledge of the UPR pathway in several yeast species: Saccharomyces cerevisiae, Schizosaccharomyces pombe, Candida glabrata, Cryptococcus neoformans, and Candida albicans. We also include unpublished data on the UPR pathway of the budding yeast Kluyveromyces lactis. We describe the basic components of the UPR pathway along with similarities and differences in the UPR mechanism that are present in these yeast species.

It is known that the sleep-waking cycle is modulated by several molecules that may also regulate food intake, among them several neuropeptides. The cocaine-and-amphetamine-regulated transcript has been studied in relation to food ingestion, but it seems to have several other functions that may include sleep regulation. In this context, we studied the effect of the intracerebroventricular administration of the cocaine-and-amphetamine-regulated transcript (0.15, 0.3, 0.6, 0.9nmol) on the sleep-waking cycle (12-h recordings), as well as its effect on food intake in rats. Additionally, we analyzed the neuronal activity as measured by c-Fos expression induced by the cocaine-and-amphetamine-regulated transcript in neurons of nuclei involved in the regulation of sleep and feeding behavior. Our main finding is that 0.3nmol of the cocaine-and-amphetamine-regulated transcript increases rapid-eye-movement sleep. In addition, our results further support that this neuropeptide triggers satiety; c-Fos expression suggested that the cocaine-and-amphetamine-regulated transcript activates specific hypothalamic nuclei without affecting other brain structures known to be involved in sleep regulation. These data further support the notion that a few neuropeptides are involved in the regulation of both the sleep-waking and the hunger-satiety cycles.

Los objetivos de este estudio fueron establecer la existencia y eventual disposición de las conexciones ligamentosas entre la articulación temporomandibular (ATM) y el oído medio en el feto a término y determinar si son capaces de trasmitir movimientos desde l articulación al martillo. Se estudiaron 11 hemicabezas pertenecientes a seis fetos a término preservados en formalina al 10 por ciento. Siete hemicabezas se mantuvieron en formalina durante toda la investigación, mientras que cuatro fueron sometidas a un proceso de conservación en medio acuoso. Ocho hemicabezas (grupo I) fueron microdisecadas a través de la fosa craneal media; las tres restantes (grupo II) fueron primero microdisecadas y luego cortadas sagitalmente a lo largo de laATM y el oído medio. En todas las hemicabezas se traccionó del disco en forma controlada y manteniendo intactos los labios de la fisura escamotimpánica con el fin de observar si se producían movimientos en el martillo. En todos los especímenes estudiados se observó una banda fibrosa trapezoidal (ligamento discomaleolar) que se extiende a través de la fisura escamotimpánica. Desde la parte posteromedial del disco hasta el proceso anterior del martillo. La mayor parte de las fibras de este ligamento conectaban directamente al disco con el martillo, pero algunas se insertaban en la fisura escamotimpánica. La tracción del disco provocó desplazamientos del martillo en cinco especímenes pertenecientes a tres individuos, lo que sugiere que la trasmisión de movimientos de la ATM al oído medio es una variable interindividual en el feto a término
