RESUMO

The objective of the present study was to evaluate the effects of increasing doses of protease on broilers from 1 to 42 days of age. A total of 1290 Ross AP broilers were used, distributed among five treatments: positive control diet, negative control diet (NC), NC + 50 ppm of protease, NC + 100 ppm of protease, and NC + 200 ppm of protease. Each treatment contained six replicates of 43 animals each. The inclusion of proteases in the diet had effects (P < 0.05) on body weight, feed intake, weight gain, and feed conversion in the 12 to 21 day period; body weight, weight gain, and feed intake in the 29 to 42 day period; nutrient digestibility (energy metabolizability coefficient and crude protein at 28 days); and intestinal parameters (crypt and muscle width of jejunum and ileum at 28 days and villus length, crypt length, and jejunum thickness muscle layer at 42 days). These results indicate that the inclusion of protease in broiler feed can improve production parameters when the amount of crude protein in the diet is reduced.

Three experiments were carried out to evaluate the levels of digestible valine in diets with reduced crude protein on the performance, carcass yield and muscle fiber diameter of male broilers during the pre-starter (1 to 8 d of age), starter (9 to 21 d of age) and grower phases (21 to 42 d of age). A total of 1,080 chickens in the pre-starter phase, 900 in the starter phase and 864 in the grower phase were distributed in a completely randomized design with six treatments, consisting of a control positive diet (C+) and diets with 2 percentage points (p.p.) reduction of crude protein level and five graded levels of digestible valine (Val), with six replicates of 30, 25 and 24 birds per experimental unit, respectively. The recommended level of TID Val in the low-protein diets for broilers in the pre-starter, starter and grower stages were 1.028; 0.905 and 0.789%, respectively. The reduction of 2 percentage points of the crude protein level in diets based on corn and soybean meal impaired (p <= 0.05) the feed conversion ratio in the starter and grower stages. Likewise, the reduced-protein diets decreased (p <= 0.05) muscle fiber diameter, but did not affect (p> 0.05) carcass and parts yields, or abdominal fat percentage at 42 days.(AU)

AIM: To assess the functional consequence of the hepatocyte nuclear factor 1alpha gene (HNF-1alpha) G574S variant previously proposed as a diabetes susceptibility allele, in a group of Mexican Type 2 diabetic patients with end-stage renal disease (ESRD). METHODS: The transcriptional activity of the HNF-1alpha G574S recombinant protein on the human insulin promoter was assessed by transfection assays in RINm5f and HepG2 cell lines. RESULTS: Two unrelated Mexican diabetic patients with no known African ancestry were found to carry the G574S variant. This substitution was not found among unrelated healthy control subjects. Whereas the G574S HNF-1alpha transcription activation of the human insulin promoter was 40% lower than that of the wild-type protein in RINm5f beta cells, no difference was found in a hepatic cell line (HepG2). CONCLUSIONS: G574S affects the transactivation potential of HNF-1alpha on the insulin promoter in pancreatic beta-cells. Although it has been difficult to prove its role in the development of diabetes in case-control association studies, this variant exhibits functional effects consistent with it being a potential diabetes susceptibility allele.
