RESUMO

This study aimed to investigate the liquid-liquid equilibrium (LLE) behavior of sesame fatty acid ethyl ester (FAEE) and methyl ester (FAME) in combination with glycerol and the co-solvents ethanol and methanol. FAEE and FAME were produced through the transesterification of mechanically extracted and purified sesame oil, using potassium hydroxide (KOH) as a homogeneous base catalyst. The reactions were conducted in ethanol and methanol to produce FAEE and FAME, respectively. Post-reaction, the products were separated and purified, followed by an analysis of the LLE behavior at 313.15 K and 323.15 K under atmospheric pressure (101.3 kPa). The experimental process for the miscibility analysis utilized a jacketed glass cell adapted for this study. Miscibility limits or binodal curves were determined using the turbidity-point method. Tie lines were constructed by preparing mixtures of known concentrations within the two-phase region, which allowed the phases to separate after agitation. Samples from both phases were analyzed to determine their composition. This study revealed that higher temperatures promoted greater phase separation and enhanced the biodiesel purification process. The NRTL model effectively correlated the activity coefficients with the experimental data, showing good agreement, with a root-mean-square deviation of 3.5%. Additionally, the data quality was validated using Marcilla's method, which yielded an R2 value close to 1. Attraction factors and distribution coefficients were also calculated to evaluate the efficiency of the co-solvents as extraction agents. The findings indicated higher selectivity for methanol than for ethanol, with varying degrees of distribution among the co-solvents. These results offer significant insights into enhancing biodiesel production processes by considering the effects of co-solvents on the LLE properties of mixtures, ultimately contributing to more efficient and cost-effective biodiesel production.

RESUMO

The synthesis by coprecipitation of Layered Double Hydroxides (LDHs) is governed by the stages of nucleation and crystal growth associated with the efficiency of the mixing and dispersion process of the reagents. Mixing efficiency is related to process variables, such as agitation speed, type of impeller and baffles presence, among others. In this context, this work proposes an analysis of these variables in a batch reactor, using a 23 factorial design employing the factors: acceleration speed (200 and 1000 rpm), mixing time (2 and 18 h) and presence or absence of baffles. The results were evaluated quantitatively (amount of LDH produced, time and amount of base for the formation of LDHs to begin) and qualitatively (mixing aspects, sedimentation ad grinding). The significant factors affecting the amount of LDH produced (51.94-80.81 g) were agitation speed and aging time. These factors were also correlated with the structural characteristics of the materials produced, such as crystallinity, crystallite size (70.99-174.79 nm), surface area (69.81-97.62 m2/g), pore volume (0.28-0.59 cm3/g), and pore diameter (11.40-34.66 nm). LDHs produced at higher agitation rates (1000 rpm) and longer aging times (18 h) yielded higher quantities of materials (80.81 g) with improved structural characteristics. The study highlights the importance of systematically exploring the synergistic effect between process variables, emphasizing the research potential in this area.

RESUMO

Leishmaniasis is a neglected tropical disease; there is currently no vaccine and treatment is reliant upon a handful of drugs suffering from multiple issues including toxicity and resistance. There is a critical need for development of new fit-for-purpose therapeutics, with reduced toxicity and targeting new mechanisms to overcome resistance. One enzyme meriting investigation as a potential drug target in Leishmania is M17 leucyl-aminopeptidase (LAP). Here, we aimed to chemically validate LAP as a drug target in L. major through identification of potent and selective inhibitors. Using RapidFire mass spectrometry, the compounds DDD00057570 and DDD00097924 were identified as selective inhibitors of recombinant Leishmania major LAP activity. Both compounds inhibited in vitro growth of L. major and L. donovani intracellular amastigotes, and overexpression of LmLAP in L. major led to reduced susceptibility to DDD00057570 and DDD00097924, suggesting that these compounds specifically target LmLAP. Thermal proteome profiling revealed that these inhibitors thermally stabilized two M17 LAPs, indicating that these compounds selectively bind to enzymes of this class. Additionally, the selectivity of the inhibitors to act on LmLAP and not against the human ortholog was demonstrated, despite the high sequence similarities LAPs of this family share. Collectively, these data confirm LmLAP as a promising therapeutic target for Leishmania spp. that can be selectively inhibited by drug-like small molecules.

RESUMO

Abstract The Pyroligneous extract is a product from the combustion of plant biomass with applications in the fields of health, industrial chemistry, and agriculture. The discovery of new molecules with therapeutic potential and of natural origin continues to be one of the great challenges for research centres around the world. The following work aims to analyze, through a technological prospection, the use of pyroligneous extracts for therapeutic purposes. To carry out the study, searches were carried out in documents deposited in Brazil, Europe, and the United States and searched on platforms specialized in patents. The number of inventions using pyroligneous extract with therapeutic applications is still quite small, however, innovations have been observed for the treatment of diseases of great clinical relevance such as cancer and hypertension. The systematic mapping of innovations is of great importance for the development of new technologies.

RESUMO

The disposal of coconut wastes is costly and damaging to the environment, but its uses are advantageous activated carbons production. Coconut leaves waste were used for activated carbon production by pyrolysis at 500° C and activation with potassium carbonate. The activated carbon was used for caffeine removal from aqueous solution. The coconut leaves activated carbon showed a predominantly amorphous structure from X-ray diffraction analysis and a pH at the zero charge point of 7.9. From the N2 adsorption/desorption method, the adsorbent showed a predominance of mesopores, with average pore size of 45.48 ηm and a surface area of 678.03 m2/g. From kinetic studies the data followed the pseudo-second order, where the intraparticle diffusion can be neglected. The adsorption isotherms were satisfactorily adjusted for the Redlich-Peterson model and a type curve L was identified. The thermodynamic parameters showed that adsorption occurred spontaneously, was exothermic and governed by physical adsorption. The artificial neural networks developed were capable of predicting both kinetics and equilibrium adsorption data under different operating conditions and was comparable to the traditional models available in literature in the training experiments, encouraging its use for data generalization when an efficient dataset is used. In conclusion, coconut leaves waste showed to be a promising feedstock to produce activated carbon aiming caffeine removal from water and wastewater.

RESUMO

OBJECTIVE: To characterize the clinical-epidemiological profile of multisystem inflammatory syndrome in children temporally associated with COVID-19 (MIS-C), and to identify factors associated with MIS-C deaths in Brazil, 2020. METHODS: This was a cross-sectional study, using national MIS-C monitoring data. Logistical regression was performed to estimate crude and adjusted odds ratios (OR). RESULTS: Median case (n=652) age was 5 years, 57.1% were male, 52.0% were of brown race/skin color and 6.4% died. Likelihood of death was greater among those who presented O2 saturation <95% (ORa=4.35 - 95%CI 1.69;11.20) and altered urea results (ORa=5.18 - 95%CI 1.91;14.04); likelihood of death was lower when red skin blotches were not present (ORa=0.23 - 95%CI 0.09;0.62), when anticoagulants were used (ORa=0.32 - 95%CI 0.12;0.89) and when immunoglobulins were used (ORa=0.38 - 95%CI 0.15;1.01). CONCLUSION: Fatality ratios were higher among cases that presented O2 saturation <95% and altered urea results. Fatality ratios were lower among those with red skin blotches, and those who used immunoglobulins and anticoagulants.

RESUMO

This study assessed the fracture resistance of simulated immature teeth reinforced with calcium aluminate cement (CAC) or mineral trioxide aggregate (MTA) containing calcium carbonate nanoparticles (nano-CaCO3). The microstructural arrangement of the cements and their chemical constitution were also evaluated. Forty-eight canines simulating immature teeth were distributed into 6 groups (n=8): Negative control - no apical plug or root canal filling; CAC - apical plug with CAC; CAC/nano-CaCO3 - apical plug with CAC+5% nano-CaCO3; MTA - apical plug with MTA; MTA/nano-CaCO3 - apical plug with MTA+5% nano-CaCO3; and Positive control - root canal filling with MTA. The fracture resistance was evaluated in a universal testing machine. Samples of the cements were analyzed under Scanning Electron Microscope (SEM) to determine their microstructural arrangement. Chemical analysis of the cements was performed by Energy Dispersive X-ray Spectroscopy (EDS). The fracture resistance of CAC/nano-CaCO3 was significantly higher than the negative control (p<0.05). There was no significant difference among the other groups (p>0.05). Both cements had a more regular microstructure with the addition of nano-CaCO3. MTA samples had more calcium available in soluble forms than CAC. The addition of nano-CaCO3 to CAC increased the fracture resistance of teeth in comparison with the non-reinforced teeth. The microstructure of both cements containing nano-CaCO3 was similar, with a more homogeneous distribution of lamellar- and prismatic-shaped crystals. MTA had more calcium available in soluble forms than CAC.

RESUMO

The oil spill off the coast of the Brazilian Northeast region is one of the most significant global events regarding contamination and environmental impact in recent years. This work evaluates the effects of oil spills on the Northeast coasts between Alagoas and Sergipe states from October 2019 to January 2020. Analysis of some sampling points of seawater revealed the presence of Hg, Cd, Pb, and Cu in levels above the maximum concentration limits established by the Brazilian legislation. For water quality parameters, phosphorus, nitrite, and turbidity showed statistically different values. However, the chromatographic profiles of oil obtained from different beaches were quite similar. Seawater, fishes, and massunins (bivalve) presented the main polycyclic aromatic hydrocarbons: naphthalene, phenanthrene, fluoranthene, fluorene, and acenaphthalene. Therefore, the concentration of organic and inorganic contaminants determined in different environmental locations served as a subsidy to assess the effect of the preliminary oil spill on the Brazilian coast.

The objective of the present study was to evaluate the influence of the socioeconomic profile of 338 people who answered questions about knowledge, consumption, determining factors in the acquisition, frequency of consumption, place of purchase, evaluation of general aspects by the consumer and repetition of the purchase of beef packed with modified atmosphere packaging (MAP). The generalized linear model methodology, GENMOD, was used with a probit link function to test the interaction of the questions with the socioeconomic profile and for all significant variables. The risk was calculated using the odd ratios (OR) with a confidence interval of 95%. Only age and education influenced consumers' disposition regarding the questions raised in the questionnaire. Most of the people who answered the questionnaire had never consumed any product packaged with MAP, but they are very interested in trying it out. Among consumers who had already consumed some product packaged with MAP, the vast majority consumed beef. Consumers over the age of 50 showed a greater tendency to consume beef with MAP more than once a week. Appearance was the main factor for decision making during the purchase and the supermarket is the main place of purchase of the product. The vast majority of consumers liked and would buy beef packed with MAP again.(AU)

OBJECTIVE: to analyze characteristics, incidence and factors associated with serious adverse events (SAEs) following yellow fever vaccination during an outbreak of the disease in Brazil (2016-2017). METHODS: this was a case-control study using data from the National Immunization Program Information System (SI-PNI); SAE were considered to be cases, and non-serious adverse events (NSAE) were considered to be controls. RESULTS: we analyzed 135 SAE cases and 1,058 controls; of the 135 SAE, 79 (58.5%) were males and median age was 28 years [09-49]; incidence in January 2017 reached 1.3 case per 100,000 vaccine doses administered; there was statistical association with males (Odds Ratio [OR]=1.73 - 95%CI 1.20;2.48), primary vaccination (OR=1.65 - 95%CI 1.01;2.71), and being 60 years of age or older taking as reference those aged under 5 (OR=4.4; p-value <0.02). CONCLUSION: SAE owing to yellow fever vaccine showed a greater chance of occurring in men, the elderly and primary vaccination.

RESUMO

BACKGROUND: After renal transplantation (RTx) hepatitis C virus (HCV) is associated with higher morbidity and mortality resulting in lower patient and graft survival. Few studies have investigated the evolution of renal transplant patients with cirrhosis owing to HCV. The objectives were to evaluate the post-transplant evolution of cirrhotic patients and to compare them with noncirrhotic patients considering the outcomes, including hepatic decompensation, graft loss, and death. METHODS: The retrospective-cohort study analyzed the data of patients undergoing RTx between 1993 and 2014, positive anti-HCV, HCV-RNA before RTx, and availability of data for assessment of cirrhosis. Demographic, clinical, and laboratory variables were compared between the groups according to the outcomes. The same were made between cirrhotic patients with and without portal hypertension (PH). Survival curves were constructed by the Kaplan-Meier test and compared by the log-rank test. Variables associated with the outcomes were analyzed using Cox regression. RESULTS: This study included noncirrhotic (n = 201) and cirrhotic patients (n = 23). In cirrhotic patients, they were significantly older (49 vs 41.6 years) and mostly male (87% vs 65%), with a greater number of previous RTx (48% vs 18%), less frequent use of azathioprine (26% vs 54%), cyclosporine (13% vs 46.5%), more frequent use of tacrolimus (87% vs 55%), lower count of platelets × 1000 cells/mm3(110 vs 187), and higher pre-RTx international normalized ratio (1.20 vs 1.1).The Kaplan-Meier survival differed in cirrhotic vs noncirrhotic patients only in hepatic decompensation. Cox regression analysis identified pretransplant cirrhosis (hazard ratio 6.64, 95% confidence interval, 2.59-17.06) and tacrolimus (hazard ratio 3.17,95% confidence interval, 1.05-9.58) as variables independently associated with decompensation. CONCLUSIONS: Patients with HCV and cirrhosis exhibit higher morbidity when submitted to RTx than noncirrhotic patients, with a higher risk of hepatic decompensation. However, no difference was observed in liver-related mortality, suggesting that RTx is a feasible option in cirrhotic patients without decompensation, even if they have PH.

RESUMO

The vast majority of consumers liked and would buy beef packed with MAP again.

The endocarp of Wodyetia bifurcata was used to produce biochar by vacuum pyrolysis as an alternative adsorbent for methylene blue (MB) removal. The influence of different pyrolysis temperatures, particle diameters and activation agents in the adsorption process was studied. Kinetics and adsorption equilibrium were also evaluated. Biochar obtained at higher pyrolysis temperatures and activated with H3PO4 showed the best adsorptive capacities, achieving 83% of MB removal. The experimental data fitted better with pseudo-second order model. Isotherms performed at 25, 40, 50 and 60 °C showed that the adsorption of MB onto the activated biochar had no concentration dependence in the range studied. Experimental isotherms fitted well with the Freundlich and Sips models and the thermodynamic parameters suggested a physical adsorption mechanism in a heterogeneous surface, spontaneous at all temperatures evaluated. In brief, the activated carbon obtained from Wodyetia bifurcata can be a promising material for MB removal from aqueous solutions.

RESUMO

This work aims to produce activated carbon from the endocarp of Syagrus oleracea by vacuum pyrolysis and evaluate its potential as an adsorbent. The effects of pyrolysis temperature (400 °C, 500 °C, 600 °C, and 700 °C), particle diameter (0.467 mm, 0.267 mm, and lower than 0.234 mm), and activation agent (H2SO4, H2PO3, and KOH) on the potential for methylene blue adsorption were investigated. In addition, kinetics and adsorption equilibrium were evaluated. The best condition found was particle diameter < 0.234 mm, final pyrolysis temperature of 700 °C, and using KOH. Adsorption kinetics followed pseudo-second order, with equilibrium reached within 20 min. Isotherms followed the Freundlich model. Values of adsorption capacity were in the order of 30 mg/g. Thermodynamic parameters indicated that adsorption occurred spontaneously with a reduction in the heterogeneity of the solid interface/solution. The results obtained demonstrate the great adsorption capacity of the activated carbon prepared from endocarp of Syagrus oleracea via vacuum pyrolysis for the removal of up to 98% methylene blue from aqueous solutions.

RESUMO

The aim of this paper was to prepare, by the freeze-drying method, ionically crosslinked chitosan membranes with different contents of pentasodium tripolyphosphate (TPP) and loaded with 1,4-naphthoquinone (NQ14) drug, in order to evaluate how the physical crosslinking affects NQ14 release from chitosan membranes for cancer therapy application. The membranes were characterized by Fourier transform infrared spectroscopy (FTIR), wide-angle X-ray diffraction (WAXD), scanning electron microscopy (SEM), swelling degree, and through in vitro drug release and cytotoxicity studies. According to the results, the molecular structure, porosity and hydrophilicity of the chitosan membranes were affected by TPP concentration and, consequently, the NQ14 drug release behavior from the membranes was also affected. The release of NQ14 from crosslinked chitosan membranes decreased when the cross-linker TPP quantity increased. Thus, depending on the TPP amount, the crosslinked chitosan membranes would be a potential delivery system to control the release of NQ14 for cancer therapy application. Lastly, the inhibitory potential of chitosan membranes ionically crosslinked with TPP and loaded with NQ14 against the B16F10 melanoma cell line was confirmed through in vitro cytotoxicity studies assessed via MTT assay. The anti-proliferative effect of prepared membranes was directly related to the amount of cross-linker and among all membranes prepared, such that one crosslinked with 0.3% of TPP may become a potential delivery system for releasing NQ14 drug for cancer therapy.
