This study aimed to investigate the effect of temperature on gonadal differentiation, growth, survival, and sex ratio of Leporinus friderici reared at 25 °C or 29 °C from 50 to 240 days after eclosion (DAE) in a water recirculation system. A total of 110 fish at 50 DAE (6.7 ± 0.1 cm and 6.1 ± 0.3 g) were equally and randomly distributed in 10 boxes (90 L) (11 fish/box, 5 boxes/temperature). One fish from each experimental unit was randomly sampled at 50, 70, 90, 110, 130, 150, 170, 190, 210 and 240 DAE. Female gonadal differentiation started at 150 DAE (11.4 ± 0.0 cm and 16.4 ± 0.0 g) at 25 °C and at 170 DAE (10.7 ± 0.7 cm and 27.7 ± 8.5 g) at 29 ºC, while testes differentiation only occurred at 29 °C from 190 DAE (12.1 ± 0.0 cm and 38.0 ± 0.0 g). Of 50 fishes sampled in each condition, 17 (12 females and five males) and three (three females) displayed gonadal differentiation at 29 °C and 25 °C, respectively. Final biometric values at 29 °C were twice those obtained at 25 °C, reaching 13.9 ± 0.65 cm and 57.3 ± 10.12 g versus 11.2 ± 0.39 cm and 28.5 ± 2.95 g, respectively. While temperature clearly influenced gonadal differentiation and growth, it had inconclusive effects on sex ratio. The higher temperature (29 °C) has direct implications for the production of this species, as it accelerates growth without causing mortality.

In this study, we aimed to propose changes in the protocol of cultured Astyanax altiparanae hypophysation to increase the maximum ovulation rate of 60% registered previously. To that two consecutive experiments were conducted. In the first experiment, three carp pituitary homogenate (CPH) doses (3, 6, and 9 mg/kg) were administered in a single injection, while in the second experiment, the 6 mg/kg CPH dose was tested either in single or double injections. In the first experiment, a single injection of 3 mg/kg CPH did not induce final oocyte maturation or spawning, while a dose of 6 mg/kg CPH resulted in an increase in the plasma level of prostaglandin (PGF2α) at ovulation. The single higher dose of 9 mg/kg CPH did not improve reproductive performance and even though anticipated the resumption of meiosis it was detrimental to the spawning rate. In the second experiment, the dose of 6 mg/kg CPH fractionated into two injections led to a higher spawning rate, spawning volume per female body mass, frequency of post-ovulatory complexes, and PGF2α concentration at ovulation compared to the single injection. The most effective treatment remained the 6 mg/kg of CPH fractionated into two injections, but still providing very low proportion of ovulated females (∼40 %). Overall, this study indicates that the spawning protocols for this species need to be improved to induce ovulation in a larger number of females and be more potent in those females that respond positively.

The mapará, Hypophthalmus marginatus, is a rheophilic and planktophagous catfish and it is one of the main fishing resources in the Amazon region. To understand its nutritional characteristics, this work described the morphology and histochemistry of the digestive tract of H. marginatus. The oropharyngeal cavity has abundant, long and thin gill rakers, responsible for retaining plankton, while the short and muscular oesophagus promotes the transport of these items to the stomach and prevents water ingestion. The stratified oesophageal epithelium has goblet cells with mucins predominantly neutral that could facilitate the food passage the food passage. The U-shaped siphonal stomach shows a columnar epithelium with neutral mucins to protect against autodigestion. The cardiac and fundic regions have gastric glands, while the pyloric region has a thick muscular layer with a sphincter. The coiled intestine with intestinal quotient of 2.14 ± 0.5 shows long longitudinal folds in the anterior region, decreasing in height in the aboral direction, and highlighting its function in digestion and nutrient absorption. Goblet cells are abundant in posterior intestine and rectum, and rectum shows epithelial cells with mucins in the apical cytoplasm, for protection and defecation. Intraepithelial lymphocytes also are abundant in posterior intestine and rectum, and it acts in immunological defence.

In this review, we present a background on the Brazilian Federal Legislation on the environmental licen-sing of hydroelectric plants focusing on the procedure of the repair of environmental damage caused by dams to migratory fish. To that end, the Brazilian electrical matrix was first addressed, thus highlighting the significant contribution of the energy produced by hydroelectric projects. To better contextualize the characteristics of the legislation, separate sections concerning illustrative reports of the effects of dams on migratory fish and the current panorama of aquaculture in Brazil and in the world were included. In this review, we also present a discussion on the specific legislation concerning a mitigation measure, the "fish restocking programs," which have the potential to promote fishing and aquaculture, but still lack a scien-tific basis on their effectiveness and correct application. An assessment of this historical process in Brazil indicates that different mitigation measures imposed to obtain operating licenses by hydroelectric plants vary among different hydroelectric projects and that this heterogeneity in the conditions imposed may have effects (of unknown proportions) on local fish communities. Considering the absence of a specific device foreseen in the law that requires the owner or concessionaire of dams in watercourses to provide for fish restocking programs or specific ichthyofauna conservation programs, the issue seems to depend on the discretion of the licensing agency to demand that the hydroelectric plant operators carry out the rein-troduction of fish in their reservoirs. This review concludes that there are political and scientific issues to be debated and explored in order to improve public policies on this topic of extreme relevance for society.(AU)


Nesta revisão, apresentamos um histórico da legislação brasileira sobre licenciamento ambiental de usi-nas hidrelétricas com foco no procedimento de reparação de danos ambientais aos peixes migratórios causados por barramentos. Para tanto, a matriz elétrica brasileira foi inicialmente abordada, destacando a significativa contribuição da energia produzida por projetos hidrelétricos. A fim de melhor contextua-lizar as características da legislação sobre o tema, foram incluídas seções referentes a relatos ilustrativos acerca dos efeitos das barragens para os peixes migratórios no Brasil e o panorama atual da aquicultura desenvolvida no país e no mundo. Apresentamos também uma discussão sobre a legislação específica relativa a uma medida de mitigação, os "programas de repovoamento de peixes", que têm potencial para promover a pesca e a aquicultura, mas parecem carecer de base científica sobre sua eficácia e correta aplicação. Uma avaliação desse processo histórico no Brasil indicou que as diversas medidas de mitigação impostas a usinas hidrelétricas, para obterem licenças de operação, variam entre os projetos hidrelétricos e que essa heterogeneidade nas condições impostas pode ter efeitos (de proporções desconhecidas) nas comunidades piscícolas locais. Considerando, então, a inexistência de dispositivo específico previsto em lei, que obrigue o proprietário ou a concessionária de barragens em cursos d'água a prever programas de repovoamento de peixes ou de conservação da ictiofauna, a questão parece depender da discricio-nariedade do órgão licenciador de exigir que os operadores das usinas reintroduzam os peixes em seus reservatórios. Esta revisão mostra que há questões políticas e científicas a serem debatidas e exploradas, visando ao aprimoramento de políticas públicas sobre esse tema de extrema relevância para a sociedade.(AU)

Breeding technology is of utmost importance for reproduction of wild fish in captivity for the reintroduction and selective breeding programs purposes. The main challenge is that when applied to wild undomesticated specimens, conventional protocols often cause breeders and/or embryo mortality and spawning failure. In this study, we evaluated the reproductive performance of wild Leporinus friderici, a great importance fish for subsistence fishing in South American rivers, applying conventional and lower-dose hormonal therapies by means of two consecutive experiments. In the first, a conventional (0.5 and 5.5 mg/kg) and a lower carp pituitary extract (CPE) dose (0.5 and 1.0 mg/kg) were applied. In the second, a conventional mammalian GnRH analogue associated with metoclopramide (mGnRHa + MET) (40 µg mGnRHa + 20 mg MET/kg) and a lower dose (4 µg mGnRHa + 2 mg MET/kg and 8 µg + 4 mg of mGnRHa + MET/kg) were applied. Ovulation was observed in all treatments, however, only lower CPE protocol provided viable embryos. High levels of 17α,20ß-dihydroxy-4-pregnen-3-one (DHP) and 17ß estradiol (E2) detected in conventional, but not in lower CPE dose, at ovulation, might be associated to the mortality of the embryos. The use of lower CPE dose applied here was the best way to obtain L. friderici viable embryos. These results directly contribute to the knowledge about poorly explored effects of reproductive management and hormonal therapies in wild-type breeders, showing that the use of reduced doses may be an alternative to reproductive success.

The aim of the study was to establish an effective feminization protocol for Leporinus macrocephalus using the 17-estradiol (E2). Thus, one hundred and fifty fingerlings with 50 days old post-hatch were randomly distributed in fifteen experimental tanks of 90L and fed for 60 days on a diet supplemented with 50 or 100mg Kg-1 of E2. At the end of the experiment, sex ratios were determined through histological and macroscopic observations. Histologically, the differentiated ovaries were evidenced by the presence of numerous nests of oogonia and oocytes in primary growth stage. The female ratio (73%) for the group treated with 100mg Kg-1 E2 was significantly higher than those of control (52%) and 50mg Kg-1 treatment (48%) groups. These results indicate that 100 mg Kg-1 E2 administered for 60 days was the most effective treatment for 50 days old L. macrocephalus post larval feminization. In conclusion, the successful sex control can be achieved through dietary hormonal manipulation. However, future studies should be conducted to evaluate the economic feasibility of this technique.(AU)


O objetivo do estudo foi avaliar o efeito do 17-estradiol (E2) na feminização de Leporinus macrocephalus. Dessa forma, 150 alevinos com 50 dias de idade foram distribuídos aleatoriamente em 15 tanques experimentais de 90 L e alimentados por 60 dias com dieta suplementada com 50 ou 100 mg kg-1 de E2. No final do experimento, as proporções sexuais foram determinadas por meio de observações histológicas e macroscópicas. Histologicamente, os ovários diferenciados foram evidenciados pela presença de numerosos ninhos de oogonia e oócitos em fase crescimento primário. O percentual de fêmeas (77%) do grupo tratado com 100 mg kg-1 E2 foi significativamente maior comparado ao grupo controle (52%) e ao grupo 50 mg kg-1 (48%). Os resultados obtidos neste estudo inicial indicaram que 100 mg kg-1 de E2, administrados durante 60 dias, foi o tratamento mais efetivo na feminização de L. macrocephalus com 50 dias de idade. No entanto, estudos futuros com variações no intervalo de aplicações podem trazer ainda melhores resultados.(AU)

In this study, we evaluated the possibility of obtaining successive Astyanax altiparanae spawns under laboratory conditions. In order to do so, 104 specimens were randomly distributed into four boxes (10 females and 16 males each) and kept in a recirculation system at an average temperature of 29.24 ± 0.42 °C, under natural photoperiod, for 30 days. On the onset of the experiment, males and females were induced for reproduction with a 6 mg kg-1 carp pituitary extract dose. After that, ovary (for gonadosomatic index and stereological assessment) and blood samples (for steroid evaluation) were collected from eight females (two per box) at the following moments: immediately after hormonal induction (day 0) and on days 1, 6, 16 and 30 after spawning. On day 6, spawned females presented complete mature ovaries similar to those on day 0 and, in this period, we did not observe postovulatory complexes, indicating that their resorption happened very fast. Concomitantly, the steroid levels increased gradually up to day 6, which corroborated an intense vitellogenic activity in this period. This study has demonstrated that A. altiparanae females are suitable for induced spawning within six days after spawning, when kept at 29.24 ± 0.42 °C, in a system maintained with recirculated water.(AU)


Neste estudo, avaliamos a possibilidade de obtenção de desovas sucessivas de Astyanax altiparanae mantidos em biotério. Para isso, 104 indivíduos foram distribuídos aleatoriamente em quatro caixas (10 fêmeas e 16 machos em cada) e mantidos a uma temperatura média de 29,24 ± 0,42 °C, sob fotoperíodo natural, por 30 dias. No início do experimento, machos e fêmeas foram induzidos à reprodução com uma dose de extrato de hipófise de carpa (6 mg kg-1). Posteriormente, amostras de ovário (para cálculo do índice gonadosomático e avaliação estereológica) e de sangue (para avaliação de esteroides) foram coletadas de oito fêmeas (duas por caixa) nos seguintes períodos: imediatamente após a indução hormonal (dia 0) e 1, 6, 16 e 30 dias após a desova. No dia 6, as fêmeas desovadas já apresentavam ovários maduros completos similares aos descritos no dia 0. No dia 6 não foram mais observados complexos pós-ovulatórios, indicando que a sua reabsorção foi relativamente rápida. Concomitantemente, os níveis de esteroides aumentaram gradativamente até o dia 6, corroborando uma atividade vitelogênica intensa neste período. Neste estudo demonstramos que fêmeas de A. altiparanae apresentam-se aptas para indução hormonal seis dias após a reprodução, quando mantida a 29,24 ± 0,42 °C, em um sistema mantido com água recirculada.(AU)

Several studies have been developed to support the replacement of the crude carp pituitary extract (CPE) by synthetic products for induced reproduction of South American rheophilic species. However, results have been quite heterogeneous and there is no consensus or a routine use of synthetic products in these species. Thus, the aim of this study was to evaluate the ovulatory process in L. elongatus using different protocols of hormonal induction. Thus, fifteen wild mature females maintained at the Experimental Fish Station, Salto Grande, SP, Brazil were submitted to three different hormonal treatments: CPE (fractioned dose: 0.5 and 5.0 mg kg-1 ); mGnRHa (single dose: 3.5 µg kg-1 ) and mGnRHa (single dose: 5.0 µg kg-1 ). The spawning rate and absolute fecundity were similar among the treatments, but fertility rates were higher for CPE treatment (23.60 ± 9.40) then for mGnRHa treatments (close to or zero zero). Although females ovulated in all treatments, none of them provided viable embryos, showing hatching rates close to zero or zero. Several studies have been developed to support the replacement of the crude carp pituitary extract (CPE) by synthetic products for induced reproduction of South American rheophilic species. However, results have been quite heterogeneous and there is no consensus or a routine use of synthetic products in these species. Thus, the aim of this study was to evaluate the ovulatory process in L. elongatus using different protocols of hormonal induction. Thus, fifteen wild mature females maintained at the Experimental Fish Station, Salto Grande, SP, Brazil were submitted to three different hormonal treatments: CPE (fractioned dose: 0.5 and 5.0 mg kg-1 ); mGnRHa (single dose: 3.5 µg kg-1 ) and mGnRHa (single dose: 5.0 µg kg-1 ). The spawning rate and absolute fecundity were similar among the treatments, but fertility rates were higher for CPE treatment (23.60 ± 9.40) then for mGnRHa treatments (close to or zero zero). Although females ovulated in all treatments, none of them provided viable embryos, showing hatching rates close to zero or zero. Tebuthiuron is a phenylurea herbicide widely used in agriculture that can reach the aquatic environments, possibly posing negative effects to the aquatic biota. Phenylurea herbicides, such as diuron, are known to cause estrogenic and anti-androgenic effects in fish, but no such effects were yet reported for tebuthiuron exposure. Thus, the aim of this study was to evaluate if tebuthiuron, at environmentally relevant concentrations (100 and 200ng/L) and after 25days of exposure have estrogenic and/or anti-androgenic effects on male of Nile tilapia (Oreochromis niloticus), through the evaluation of plasmatic testosterone (T) and estradiol (E2) levels, brain aromatase (CYP19) levels (western-blot), and by evaluating the histology of the testicles. When compared to the control group, plasmatic T levels decreased about 76% in the animals exposed to 200ng/L of tebuthiuron, while E2 levels increased about 94%, which could be related to a significant increase (77%) in CYP19A1 levels, an enzyme that catalyzes the conversion of androgens into estrogens. Histological analyses of the testicles also demonstrated that tebuthiuron at both tested concentrations caused a decrease in the diameter of the seminiferous tubules and in the diameter of the lumen. Therefore, the gonadosomatic index (GSI) was reduced by 36% % in the animals exposed 200ng/L to tebuthiuron. Indeed, the relative frequency of spermatocytes and spermatids increased respectively 73% (200ng/L) and 61% (100ng/L) in the tebuthiuron exposed animals, possibly due to the impairment of sperm release into the lumen, that was decreased 93% (200ng/L) in the treated animals compared to the control. These results confirm that tebuthiuron causes estrogenic and anti-androgenic effects in Nile tilapias at environmentally relevant concentrations.

Prochilodus lineatus is an important representative of the order Characiformes and a species that offers great advantages to fish farming. Therefore, detailed knowledge of its reproductive biology can be applied to various fields of production and biotechnology. In this study, we have identified testicular germ cells during spermatogenesis and have evaluated the volumetric proportion of the testes occupied by structures of the tubular and intertubular compartments. In addition, the individual volume of type A spermatogonia was measured and used to estimate the mean number of these cells per testis. Gonads of adult P. lineatus males were extracted and fixed. Light and transmission electron microscopy were applied to fragments of three testicular regions. Histological, stereological, and morphometric analyses were performed. The stereological data suggest that components of the tubular and intertubular compartments of the P. lineatus testes present a uniform distribution in all three regions and therefore reflect regions with similar distributions of cell types. In addition, P. lineatus testes showed ∼0.6% of type A spermatogonia, as well as a predominance of cysts of primary spermatocytes and spermatids during the reproductive phase evaluated. The results from this study provide a better understanding of the morphology and structure of the testis and of the characterization of the type A spermatogonia in P. lineatus. The nuclear diameter of germ cells also decreases significantly during spermatogenesis. The data presented herein are the first of its kind for the order Characiformes and may be useful for future biotechnology studies on fish reproduction. The aim of this study was to remove the adhesiveness of Pseudoplatystoma fasciatum eggs using solutions of urea for 10, 30, 60 or 90 min (treatments T0 to T3) or in urea for 10 min and washed in tannin (T4). In the control group, eggs were kept in water. After two experiments, we observed that eggs of control group presented the best embryo viability rates, even maintaining egg adhesiveness, being better than the other treatments. The T4 had the worst embryo viability rates. We observed that embryos of the T4 treatment presented reduced growth and formed a separate group when analyzing morphological parameters (egg diameter, total egg area, embryo area and yolk sac area) by multivariate analysis. Concluding, the control group, free of chemicals, provided the best results and is considered the best alternative for the immediate conservation and aquaculture production of P. fasciatum.


O objetivo deste estudo foi neutralizar a adesividade de ovos de Pseudoplatystoma fasciatum a partir da utilização de soluções de ureia por 10, 30, 60 ou 90 min (tratamentos T0 a T3) ou em ureia por 10 min e lavados em tanino (T4). No controle, os ovos foram mantidos apenas em água. Após dois experimentos realizados, no grupo controle foram observadas as melhores taxas de viabilidade embrionária, mesmo mantendo a adesividade dos ovos, sendo melhor que os demais tratamentos. No T4 foram verificadas as piores taxas de viabilidade embrionária. In this study, we investigated the reproductive cycle and spawning season of old (OFs) and young (YFs) P. mesopotamicus females, aiming to improve the process of selection of suitable fish for induced spawning. We also evaluated the accuracy of using external body characteristics in selecting suitable females. To that, 60 OFs and 60 YFs (10 and four years old, respectively) were submitted to seven samplings during two reproductive cycles. In each sampling, females (five to 10 per treatment) were randomly chosen to evaluation of plasma concentrations of gonadal steroids and ovary composition (biopsies and stereology). Females were also submitted to an external evaluation concerning the length of abdominal thickness, the degree of the abdominal rigidity and the aspect of urogenital papilla. In spite of OFs have shown reduced concentrations of 17α- hydroxyprogesterone during the spawning season, we observed that OFs and YFs kept in captivity had similar reproductive cycles and spawning seasons (in December for both groups). We do not recommend the exclusive use of external evaluation for selecting breeders to spawning induction due to a great inaccuracy of data and lack of association with histology and ovarian biopsy, which are more accurate and reliable.(AU)


Neste estudo, investigamos o ciclo reprodutivo e a época de desova de fêmeas “velhas” (FVs) e fêmeas “jovens” (FJs) de P. mesopotamicus, visando melhorar o processo de seleção de peixes para a indução hormonal. Avaliamos também a acurácia do uso de características corporais externas para a seleção de fêmeas para indução hormonal. Para isso, 60 FVs e 60 FJs (10 e quatro anos, respectivamente) foram submetidas a sete amostragens durante dois ciclos reprodutivos. Em cada amostra, as fêmeas (cinco a 10 por tratamento) foram capturadas para avaliação das concentrações plasmáticas de esteroides gonadais e composição dos ovários (biópsias e estereologia). As fêmeas foram também submetidas à uma avaliação externa relativa à espessura abdominal, o grau de rigidez abdominal e o aspecto da papila urogenital. Exceto pela ocorrência de concentrações reduzidas de 17α-hidroxiprogesterona nas FVs, durante a época de desova, observamos que FVs e FJs, mantidas em cativeiro, apresentam ciclos reprodutivos e épocas de desova similares (em dezembro para ambos os grupos). Não recomendamos o uso exclusivo da avaliação externa para a seleção de reprodutores para indução a desova, devido a uma imprecisão dos dados e falta de associação destes com a histologia e biópsia de ovário, os quais são mais precisos e confiáveis.(AU)
