RESUMO

The secondary metabolism products of plants have influenced great economic interest, given their chemical diversity and biological activities. Because of this, this study evaluates the phytochemical composition, antimicrobial activity, insecticidal, and antioxidant activity of plant extracts and oil of Myrcia oblongata. Saponins, steroids, triterpenoids, tannins, and flavonoids were detected. The extracts showed antimicrobial capacity on the tested microorganisms, except for the methanolic extract, which showed no activity for P. mirabilis and S. enteritidis. Regarding the analysis of antioxidant compounds, the hexanic, ethyl acetate and acetone extracts showed higher antioxidant activities and also higher insecticidal performance on Alphitobius diaperinus larvae, resulting in 80% adult mortality. The results reported here show that there may be a relationship between antioxidant potential and the insecticidal effect of Myrcia oblongata DC. The components present in both the extract and the oil can be used as natural alternative to synthetic compounds in the biological control of parasites and pathogenic microorganisms.

The objective of this study was to evaluate the effects of natural phytosanitary products (NPs) on spores and crystals of Bacillus thuringiensis subsp. kurstaki S-1905 (Btk S-1905). For the spore assay, NPs and bacteria were applied in combination and individually. For the combined application, Btk S-1905 + NP mixtures were inoculated on nutrient agar (NA), and for the separate applications, the NPs were spread on NA plates, which were later inoculated with the pathogen. The number of colony-forming units (CFU) per milliliter was quantified after 18 h of incubation. For the crystal protein degradation assay, the Btk S-1905 + NP mixtures were added to the diet of Anticarsia gemmatalis (Lepidoptera: Erebidae), and mortality was evaluated at the following time points: 12, 24, 48, and 72 h. Scanning electron microscopy and agarose gel electrophoresis were carried out. Biogermex and Ecolife® reduced the CFU ml-1 in both combined and separate applications. Biogermex, Ecolife®, and Planta Clean were antagonistic to the action of bacterial toxins, and no product affected the morphology or resulted in the degradation of the crystal proteins. The remaining products evaluated did not reduce the CFU ml-1 and had additive effect when combined with the crystal toxin.

RESUMO

The purpose was to evaluate the side effects of strains Metarhizium anisopliae (Metsch.) Sorokin sensu lato Unioeste 43 and M. anisopliae sensu stricto ESALQ 1641 on Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae) under controlled conditions. A free-choice test for parasitism was performed, with the confinement of T. pretiosum females mated with cards (1 × 5 cm) containing age-standardized Anagasta kuehniella Zeller eggs, either sprayed with a fungal strain (109 conidia/ml) or 0.01% v/v Tween 80 (control). For the no-choice tests, T. pretiosum females mated were confined with cards sprayed with fungal strains before or after parasitism, and cards with fungal applications at different times. The number of parasitized eggs, percentage of emergence, longevity, egg-to-adult period, sex ratio, total and confirmed mortality by the fungus, and longevity of females that parasitized previously sprayed eggs, were assessed. Histological analysis of immature phases was also performed. The fungus was repellent to T. pretiosum in the free-choice test, while in the no-choice test, fungal applications before and after parasitism did not affect the number of eggs parasitized by T. pretiosum or the sex ratio of emerging adults. However, both strains affected adult emergence rates, the egg-to-adult period, and longevity. Overall, both M. anisopliae strains had minor effects on these biological parameters of T. pretiosum under controlled conditions. Hyphae were not detected in histological observations of immature stages of the parasitoid.

RESUMO

Control of the lesser mealworm (Alphitobius diaperinus Panzer, 1797 [Coleoptera: Tenebrionidae]) is usually conducted with chemical insecticides, which have limited efficacy as well as environmental and health risks. Diatomaceous earth (DE) is safe and effective in controlling many insects including lesser mealworm. However, little is known of DE efficacy against this pest. Thus, an assessment of DE was conducted for the first time in commercial broiler houses in Brazil, with the aim of developing a strategy for the use of DE in the control of this insect. The efficacy of DE was determinate to a minimum concentration, and the application was directed to the most infested sites. The DE applied to the entire poultry house (280 g/m2) was verified to control the insects. DE was also comparable to chemical treatments, with a reduction of 80% in the insect population. DE controlled the lesser mealworm and is recommended for application at a concentration of 280 g/m2 when applied to the surface of new poultry litter, before the first lot of birds is housed. Thereafter, DE should be reapplied before the housing of each lot of birds, in the same concentration, only in the housing area and under the feeders and near the walls and pillars. In addition to being a control strategy, DE can be an important tool in the management of lesser mealworm insecticide resistance.

RESUMO

Gyropsylla spegazziniana (Paraguay tea ampul) is one of the most important pests of Paraguay tea plants, and prohibition of synthetic insecticide use for control of this pest has led to the search for alternative methods. This laboratory study aimed to compare different control strategies for G. spegazziniana, utilizing a commercial neem seed oil product. Paraguay tea seedlings were treated with neem oil solution both pre- and post-infestation with 5th instar nymphs. The systemic action of neem oil was also evaluated by treating plant soil with the neem oil solution, followed by transfer of the insects to plants 24 h post-treatment. Spray treatments were effective against the pest, especially post-infestation (80% mortality), demonstrating the potential of neem oil for control of the Paraguay tea ampul. No significant effects were observed with respect to systemic activity.(AU)

Poultry red mite Dermanyssus gallinae is a cosmopolitan and hematophagous species commonly found in layer houses around the world. Poultry mite infestations may cause anemia, stress, low body weight and egg production, and mortality. Mite control is typically based on chemical products, but they are not effective and leave residues in eggs; therefore, alternative control methods, such as entomopathogenic fungi, need to researched. This study aimed at evaluating, in the laboratory, the activity of Brazilian isolates of entomopathogenic fungi against D. gallinae. The mites were collected from a commercial layer house and were sprayed with conidial suspensions (1 × 108 conidia/mL) of five isolates of Beauveria bassiana and Metarhizium anisopliae. All tested isolates were pathogenic for the red mite, with confirmed mortality ranging from 22.9 to 52.4%. This demonstrate the potential of the tested entomopathogenic fungi isolates for mite control, and reinforces the need for further studies with other isolates, application strategies, and with fungal formulations.(AU)

RESUMO

This study was performed under laboratory conditions to identify isolates of the fungus Beauveria spp. that can control Polyphagotarsonemus latus in the greenhouse and field. Thirty Beauveria spp. isolates were tested by spraying 1 mL conidia (1 × 108 conidia/mL) on pepper leaf discs containing 15 mites. Evaluations were performed on the 3rd and 6th day post application by counting the number of dead mites. Vegetative growth and conidial production were measured from the selected isolates, and bioassays were conducted in the greenhouse on bean seedlings in plastic pots. The isolate Unioeste 53 was selected, and a conidial suspension (1 × 108 conidia/mL) was applied with a backpack sprayer. The evaluation consisted of pre- and post-treatment counts of the number of live mites on ten leaflets in both the plots treated with the fungus and control plots, and the same procedure was followed for the field experiment. In the laboratory, the Unioeste 53 isolate resulted in total and confirmed mortality rates of 70% and 57.7%, respectively. In the greenhouse, the population decreased by 76.71% by the 16th day after application. In the field, the population decreased by 66% by the 12th day after application, demonstrating the potential of this fungus for mite management.(AU)

Abstract The fungus Beauveria bassiana is naturally found in poultry houses and causes high rates of mortality in Alphitobius diaperinus. Laboratory and field experiments have shown the potential of this fungus as an insect control agent. However, in poultry houses, bacteria as Salmonella, can be found and have been studied alternative control methods for this pathogen. Thus, this study aimed to evaluate the effect of plant extracts and a disinfectant on the fungus Beauveria bassiana (strain Unioeste 4). Conidial viability, colony-forming unit (CFU) counts, vegetative growth, conidia production, insecticidal activity of the fungus and compatibility were used as parameters in the evaluation of the effect of these products on the fungus. Alcoholic and aqueous extracts of jabuticaba (Myrciaria cauliflora (Mart.), guava (Psidium guajava (L.)), and jambolan (Syzygium cumini (L.), at concentrations of 10% as well as the commercial disinfectant, Peroxitane® 1512 AL, were evaluated at the recommended concentrations (RC), 1:200 (RC), 0.5 RC and 2 RC. There was a negative influence of alcoholic and aqueous extracts of jabuticaba, guava and three dilutions of Peroxitane on the viability of conidia. The CFUs and vegetative growth of the fungus were affected only by the Peroxitane (all dilutions). For conidial production, the aqueous extract of guava had a positive effect, increasing production, while the Peroxitane at the R and RC concentrations resulted in a negative influence. The mortality of A. diaperinus, caused by the fungus after exposure to these products, was 60% for the peracetic acid at 0.5 RC, and above 80% for the extracts. Thus, the results showed that all the extracts and Peroxitane at RC 0.5 are compatible with the fungus B. bassiana Unioeste 4, however only the extracts had a low impact on inoculum potential.(AU)

No significant effects were observed with respect to systemic activity.

RESUMO

This study aimed at evaluating the effect of the use of disinfectants and insecticides recommended for the control of pathogens and insects in poultry houses on the biological parameters of the entomopathogenic fungus Beauveria bassiana strain Unioeste 4. Disinfectants and insecticides were used at recommended concentration (RC), half the recommended concentration (0.5 RC), and twice there commended concentration (2RC). All treatments were sprayed on the fungus previously inoculated on PDA culture media. Germination, colony forming unit (CFU), vegetative growth and conidial yield were evaluated. Germination was the most affected parameter by insecticides, with reduction of up to 25% using Couro Limpo and Cypermil products. Cypermil also reduced the vegetative growth in all tested concentrations. Conidial production was reduced when products were used at the highest concentration. On the other hand, the confirmed mortality of the lesser mealworm by the fungus was not affected by none of the tested products. All disinfectants at the highest concentration reduced germination and conidia production, but did not affect fungus vegetative growth. The confirmed mortality by the fungus was most influenced by the product TH4. All tested products tested were considered compatible with the fungus.
