RESUMO

The drug release behavior of pH-sensitive starch-based hydrogels was systematically studied. Hydrogels were synthesized by copolymerization of acrylic acid (AA) and other acrylate comonomers onto the starch backbone. The hydrophilic agents 2-hydroxy ethyl methacrylate (HEMA), and acrylamide (AAm), as well as the hydrophobic butyl-methacrylate (BMA), were utilized as comonomers. Methylene-bisacrylamide (MBA) was employed as a crosslinking agent. The synthesized hydrogels were loaded with caffeine as a model drug. The effects of the hydrophobic/hydrophilic character of the comonomers and chemical crosslinking on the swelling capacity and the release rate of caffeine were investigated. The use of the crosslinking agent and hydrophobic monomers decreased the swelling capacity of the hydrogels. The release rate of caffeine increased with the presence of a hydrophobic monomer. The fastest release was obtained with the AA/BMA/AAm formulation, and the slowest release was observed with the AA/HEMA/AAm formulation. The transport mechanism was controlled by Fickian diffusion in formulations containing AAm, and controlled by the polymer-relaxation mechanism in formulations containing MBA. Overall, our results showed that the swelling and drug delivery behavior can be tuned by varying the chemical composition of the copolymer formulations. These starch-based hydrogels can be useful as drug delivery devices in many biomedical applications.

RESUMO

Three-dimensional (3D) printing technologies have become an attractive manufacturing process to fabricate scaffolds in tissue engineering. Recent research has focused on the fabrication of alginate complex shaped structures that closely mimic biological organs or tissues. Alginates can be effectively manufactured into porous three-dimensional networks for tissue engineering applications. However, the structure, mechanical properties, and shape fidelity of 3D-printed alginate hydrogels used for preparing tissue-engineered scaffolds is difficult to control. In this work, the use of alginate/gelatin hydrogels reinforced with TiO2 and ß-tricalcium phosphate was studied to tailor the mechanical properties of 3D-printed hydrogels. The hydrogels reinforced with TiO2 and ß-TCP showed enhanced mechanical properties up to 20 MPa of elastic modulus. Furthermore, the pores of the crosslinked printed structures were measured with an average pore size of 200 µm. Additionally, it was found that as more layers of the design were printed, there was an increase of the line width of the bottom layers due to its viscous deformation. Shrinkage of the design when the hydrogel is crosslinked and freeze dried was also measured and found to be up to 27% from the printed design. Overall, the proposed approach enabled fabrication of 3D-printed alginate scaffolds with adequate physical properties for tissue engineering applications.

RESUMO

Fermentation of sugar for production of ethanol was carried out using Saccharomyces cerevisiae cells immobilized in calcium alginate films. Thin films of calcium alginate casted on a microchannel surface were used instead of the typical spherical bead configuration. Yeast immobilized on alginate films produced a higher ethanol yield than free yeast cells under the same fermentation conditions. Also, a silicalite-1/poly dimethyl siloxane composite pervaporation membrane was synthesized for ethanol separation, and characterized with flux and separation factor. The composite membrane synthesized with a 3-1 ratio of silicalite-1 to poly dimethyl siloxane showed promising results, with a flux of 140.6g/m2h±19.3 and a separation factor of 37.52±3.55. Thus, the performance of both the alginate film with immobilized cells and the customized hybrid membrane suggests they could have an interesting potential application in an integrated reaction-separation device for the production and purification of bioethanol.

RESUMO

Many complex systems can be described as networks exhibiting inner organization as communities of nodes. The identification of communities is a key factor to understand community-based functionality. We propose a family of measures based on the weighted sum of two dissimilarity quantifiers that facilitates efficient classification of communities by tuning the quantifiers' relative weight to the network's particularities. Additionally, two new dissimilarities are introduced and incorporated in our analysis. The effectiveness of our approach is tested by examining the Zachary's Karate Club Network and the Caenorhabditis elegans reactions network. The analysis reveals the method's classification power as confirmed by the efficient detection of intrapathway metabolic functions in C. elegans.
