Glioblastoma (GB) is the worst and most common primary brain tumor. Temozolomide (TMZ), an alkylating agent, is widely used for treating primary and recurrent high-grade gliomas. However, at least 50% of TMZ treated patients do not respond to TMZ and the development of chemoresistance is a major problem. Here, we designed a lipid nanoemulsion containing a thermoresponsive polymer (poloxamer 407) aiming to improve TMZ release into the brain via nasal delivery. Increasing amounts of poloxamer 407 were added to preformed nanoemulsions (250 nm-range) obtained by spontaneous emulsification. The influence of the polymer concentration (from 2.5% to 12.5%) and temperature on viscosity was clearly evidenced. Such effect was also noticed on the mucoadhesiveness of formulations, as well as TMZ release rate and retention/permeation through nasal porcine mucosa using Franz-type diffusion cells. From these results, a formulation containing 10% of poloxamer (NTMZ-P10) was selected for further experiments by nasal route. A significantly higher TMZ amount was observed in the brain of rats from NTMZ-P10 in comparison with controls. Finally, our results show that formulation reduced significantly tumor growth by three-fold: 103.88 ± 43.67 mm3 (for NTMZ-P10) and 303.28 ± 95.27 mm3 (control). Overall, these results suggest the potential of the thermoresponsive formulation, administered by the non-invasive nasal route, as a future effective glioblastoma treatment.

Treatment for visceral leishmaniasis (VL) is hampered mainly by drug toxicity, their high cost, and parasite resistance. Drug development is a long and pricey process, and therefore, drug repositioning may be an alternative worth pursuing. Cardenolides are used to treat cardiac diseases, especially those obtained from Digitalis species. In the present study, cardenolide digitoxigenin (DIGI) obtained from a methanolic extract of Digitalis lanata leaves was tested for its antileishmanial activity against Leishmania infantum species. Results showed that 50% Leishmania and murine macrophage inhibitory concentrations (IC50 and CC50, respectively) were of 6.9 ± 1.5 and 295.3 ± 14.5 µg/mL, respectively. With amphotericin B (AmpB) deoxycholate, used as a control drug, values of 0.13 ± 0.02 and 0.79 ± 0.12 µg/mL, respectively, were observed. Selectivity index (SI) values were of 42.8 and 6.1 for DIGI and AmpB, respectively. Preliminary studies suggested that the mechanism of action for DIGI is to cause alterations in the mitochondrial membrane potential, to increase the levels of reactive oxygen species and induce accumulation of lipid bodies in the parasites. DIGI was incorporated into Pluronic® F127-based polymeric micelles, and the formula (DIGI/Mic) was used to treat L. infantum-infected mice. Miltefosine was used as a control drug. Results showed that animals treated with either miltefosine, DIGI, or DIGI/Mic presented significant reductions in the parasite load in their spleens, livers, bone marrows, and draining lymph nodes, as well as the development of a specific Th1-type response, when compared with the controls. Results obtained 1 day after treatment were corroborated with data corresponding to 15 days after therapy. Importantly, treatment with DIGI/Mic induced better parasitological and immunological responses when compared with miltefosine- and DIGI-treated mice. In conclusion, DIGI/Mic has the potential to be used as a therapeutic agent to protect against L. infantum infection, and it is therefore worth of consideration in future studies addressing VL treatment.

A clioquinol (ICHQ)-containing Pluronic® F127 polymeric micelle system (ICHQ/Mic) was recently shown to be effective against Leishmania amazonensis infection in a murine model. In the present study, ICHQ/Mic was tested against L. infantum infection. BALB/c mice (n = 12 per group) were infected with L. infantum stationary promastigotes through subcutaneous injection and, 45 days after challenge, received saline or were treated via the subcutaneous route with empty micelles, ICHQ or ICHQ/Mic. In addition, animals were treated with miltefosine by the oral route, as a drug control. Half of the animals were euthanized 1 and 15 days after treatment, aiming to evaluate two endpoints after therapy, when parasitological and immunological parameters were investigated. Results showed that the treatment using miltefosine, ICHQ or ICHQ/Mic induced significantly higher anti-parasite IFN-γ, IL-12, GM-CSF, nitrite and IgG2a isotype antibody levels, which were associated with low IL-4 and IL-10 production. In addition, a higher frequency of IFN-γ and TNF-α-producing CD4+ and CD8+ T-cells was found in these animals. The parasite load was evaluated in distinct organs, and results showed that the treatment using miltefosine, ICHQ or ICHQ/Mic induced significant reductions in organic parasitism in the treated and infected mice. A comparison between the treatments suggested that ICHQ/Mic was the most effective in inducing a highly polarized Th1-type response, as well as reducing the parasite load in significant levels in the treated and infected animals. Data obtained 15 days after treatment suggested maintenance of the immunological and parasitological responses. In conclusion, ICHQ/Mic could be considered in future studies for the treatment of visceral leishmaniasis.
TITLE: Un système à micelles polymériques Pluronic® F127 contenant du clioquinol est efficace pour le traitement de la leishmaniose viscérale dans un modèle murin. ABSTRACT: Un système à micelles polymériques Pluronic® F127 (ICHQ/Mic) contenant du clioquinol (ICHQ) s'est récemment révélé efficace contre l'infection à Leishmania amazonensis dans un modèle murin. Dans la présente étude, l'ICHQ/Mic a été testé contre l'infection à L. infantum. Les souris BALB/c (n = 12 par groupe) ont été infectées par des promastigotes stationnaires de L. infantum par injection sous-cutanée et ont reçu 45 jours après l'épreuve une solution saline ou ont été traitées par voie sous-cutanée avec des micelles vides, ICHQ ou ICHQ/Mic. De plus, les animaux ont été traités avec de la miltefosine par voie orale, comme contrôle médicamenteux. La moitié des animaux ont été euthanasiés 1 et 15 jours après le traitement, dans le but de mesurer deux critères d'évaluation après la thérapie, lorsque les paramètres parasitologiques et immunologiques ont été étudiés. Les résultats ont montré que le traitement par miltefosine, ICHQ ou ICHQ/Mic induisait des niveaux d'anticorps anti-parasite IFN-γ, IL-12, GM-CSF, nitrite et IgG2a significativement plus élevés, associés à de faibles productions d'IL-4 et IL-10. De plus, une fréquence plus élevée de cellules T CD4+ et CD8+ produisant de l'IFN-γ and TNF-α a été trouvée chez ces animaux. La charge parasitaire a été évaluée dans des organes distincts et les résultats ont montré que le traitement utilisant la miltefosine, ICHQ ou ICHQ/Mic induisait des réductions significatives du parasitisme des organes chez les souris traitées et infectées. Une comparaison entre les traitements a suggéré qu'ICHQ/Mic était le plus efficace pour induire une réponse de type Th1 polarisée, ainsi que pour réduire la charge parasitaire à des niveaux significatifs chez les animaux traités et infectés. Les données obtenues 15 jours après le traitement suggèrent le maintien des réponses immunologiques et parasitologiques. En conclusion, ICHQ/Mic pourrait être envisagé dans de futures études pour le traitement contre la leishmaniose viscérale.

Benznidazole and nifurtimox are the only drugs specifically approved for the treatment of Chagas disease. Both compounds are given orally in tablets, but occasionally are ineffective and cause adverse effects. Benznidazole, the first-line treatment in many countries, is a compound with low solubility in water that is administered at high doses for long periods of time. To improve its solubility, we developed a new liquid formulation on the basis of solid dispersions (SD) using the amphiphilic polymer poloxamer 407. Herein we present data on its trypanocidal performance in mouse models of acute and chronic Trypanosoma cruzi infection. SD at doses of 60 or 15 mg/kg per day given with different administration schedules were compared with the commercial formulation (CF; 50 mg/kg per day) and vehicle. The SD performance was assessed by direct parasitemia, total anti-T. cruzi antibodies, and parasitic burden in tissues after 4 or 6 mo posttreatment. The efficacy of the SD was equivalent to the CF but without manifest side effects and hepatotoxicity. Considering our previous data on solubility, together with these on efficacy, this new liquid formulation represents a promising alternative for the treatment of Chagas disease, particularly in cases when dosing poses a challenge, as in infants.

New therapeutic strategies against leishmaniasis are desirable, since the treatment against disease presents problems, such as the toxicity, high cost and/or parasite resistance. As consequence, new antileishmanial compounds are necessary to be identified, as presenting high activity against Leishmania parasites, but low toxicity in mammalian hosts. Flau-A is a naphthoquinone derivative recently showed to presents an in vitro effective action against Leishmania amazonensis and L. infantum species. In the present work, the in vivo efficacy of Flau-A, which was incorporated into a Poloxamer 407-based micelle system, was evaluated in a murine model against L. amazonensis infection. Amphotericin B (AmB) and Ambisome® were used as controls. The animals were infected and later treated with the compounds. Thirty days after the treatment, parasitological and immunological parameters were evaluated. Results showed that AmB, Ambisome®, Flau-A or Flau-A/M-treated animals presented significantly lower average lesion diameter and parasite burden in tissue and organs evaluated, when compared to the control (saline and micelle) groups. Flau-A or Flau-A/M-treated mice were those presenting the most significant reductions in the parasite burden, when compared to the others. These animals developed also a more polarized antileishmanial Th1 immune response, which was based on significantly higher levels of IFN-γ, IL-12, TNF-α, GM-CSF, and parasite-specific IgG2a isotype; associated with low levels of IL-4, IL-10, and IgG1 antibody. The absence of toxicity was found in these animals, although mice receiving AmB have showed high levels of renal and hepatic damage markers. In conclusion, results suggested that the Flau-A/M compound may be considered as a possible therapeutic target to be evaluated against human leishmaniasis.

Miltefosine (hexadecylphosphocholine, HePC), a synthetic antitumor designed from natural phospholipids, is clinically approved for cutaneous metastases of breast cancer and cutaneous lymphoma. This drug acts mainly at cellular membrane level, where it accumulates and interferes with lipid metabolism and lipid-dependent signaling pathways leading the cells to apoptosis. However, HePC systemic and peroral administration induces hemolysis and mucosal toxicity, respectively. To overcome these limitations, we investigated the protective properties of colloidal polymeric micelles (PM) composed by Pluronics, triblock copolymers of poly(ethylene oxide) and poly(propylene oxide). We found that both Pluronic composition and concentration modulate the hemolytic profile of incorporated drug (HePC-PM) by increasing the drug amount to cause in vitro hemolysis. Moreover, small-angle X-ray scattering (SAXS) was used to assess structural information of interactions between HePC and PM. Additionally, we showed that HePC-PM prevented mucosal irritation, decreasing bleeding and lysis of blood vessels in a chicken chorioallantoic membrane model. Interestingly, HePC-PM increased the in vitro selective cytotoxicity against cervix tumor cells rather healthy fibroblasts, suggesting a differential uptake of these nanostructures by tumor cells. Furthermore, we also found that HePC induces cytotoxicity and decrease cell survival, migration and proliferation in osteosarcoma cells in vitro. We showed that cytotoxicity by HePC is associated with caspase-3 activation, DNA fragmentation, apoptotic-like bodys formation and inhibition of both constitutive and cytokine-stimulated Akt/PKB phosphorylation. HePC-PM clearly reduces the drug cytotoxic effects. Finally, we demonstrated that Pluronic F127 polymeric micelles are efficient for cargo delivering the encapsulated drug preferentially into tumor cells rather than healthy cells. These findings together suggest that Pluronic F127 PM reduce drug side effects and provide a potential alternative for systemic delivery of HePC, as well as other amphiphilic drugs
Miltefosina (hexadecilfosfocolina, HePC), um fármaco antitumoral sintético desenvolvido a partir de fosfolipídios naturais, é clinicamente aprovada para o tratamento tópico de metástases de câncer de mama e linfomas cutâneos. Atua principalmente nas membranas celulares, onde se acumula e interfere no metabolismo lipídico e nas vias de sinalização dependentes de lipídios levando as células à apoptose. No entanto, quando administrada sistemicamente ou oralmente a HePC induz hemólise e toxicidade de mucosas, respectivamente. Para superar estas reações adversas investigamos os efeitos protetores conferidos por micelas poliméricas coloidais (PM) compostas por Pluronics, copolímeros tribloco de poli(óxido de etileno) e poli(óxido de propileno). Inicialmente, encontramos que a composição e concentração do Pluronic modulam o perfil hemolítico do fármaco encapsulado (HePC-PM), aumentando a quantidade necessária de HePC para causar hemólise in vitro. Além disso, utilizamos o espalhamento de raios-X a baixo ângulo (SAXS) para obter informações estruturais das interações entre HePC e PM. Em seguida, mostramos que HePC-PM preveniu a irritação da mucosa, diminuindo a hemorragia e a vasoconstricção em membrana corioalantóica de ovos embrionados. Estudos in vitro demonstraram que a HePC-PM aumentou seletivamente a citotoxicidade contra células de carcinoma HeLa em relação a fibroblastos saudáveis, sugerindo captação diferencial dessas nanoestruturas pelas células tumorais. Além disso, relatamos que, in vitro, a HePC induz citotoxicidade, diminui a sobrevivência, migração e proliferação osteossarcomas. Esta citotoxicidade está associada à ativação da caspase-3, fragmentação do DNA, formação de corpos apoptóticos e inibição da fosforilação de Akt/PKB. Adicionalmente, HePC-PM reduz os efeitos citotóxicos nestas linhagens. Finalmente, demonstramos que as micelas poliméricas de Pluronic F127 são eficientes para a entrega intracelular fármacos preferencialmente em células tumorais, e em menor grau em células saudáveis. Em conjunto, os dados sugerem que este sistema nanoestruturado reduz a toxicidade da HePC e representa uma alternativa potencial para a administração sistêmica deste e de outros fármacos anfifílicos

The bone morphogenetic protein type 2, is an osteoinduction protein, but new carriers to improve its actions are being studied in recent researches. The objective of this study was to evaluate the poloxamer gel as potential carrier of the rhBMP-2 during the bone healing process and verify if the collagen sponge can improve this process. Fifty-six male Wistar rats were used, which were divided into two groups, considering two periods of time. Thus, each one of these groups was divided in four groups with seven animals each, according to the treatment and the defect filled by 4 microg rhBMP-2 in aqueous solution, 4 microg rhBMP-2 in aqueous solution+collagen sponge, 4 microg rhBMP-2 combined with poloxamer gel, and 4 microg rhBMP-2 combined with poloxamer gel+collagen sponge. After 2 and 4 weeks, respectively, animals were perfused and the hemi-mandibles removed for histological analysis. Statistical analysis of the data showed significant difference between all analyzed groups (p<0.01), and between the periods of time (p=0.0118). There was no interaction between the applied treatment and considered periods of time (p=0.642). Results showed that the rhBMP-2 used in this study was able to induce bone regeneration and had its action optimized when combined to the used carriers, being the bone repair time-dependent.
