Mastitis, an inflammation of the mammary gland affecting milk production and quality in dairy herds, is often associated with Staphylococcus spp. in goats. Neutrophils are crucial in combating infections by migrating into milk and deploying various defense strategies, including the release of neutrophil extracellular traps (NETs) composed of DNA, histones, and bactericidal proteins. This study investigated whether NETs are released by goat neutrophils stimulated in vitro by Staphylococcus aureus and Staphylococcus warneri, two common pathogens of goat mastitis. PMNs were isolated from blood from healthy adult goats. We evaluated goat NET formation by stimulating cells with: phorbol 12-myristate 13-acetate (PMA) as a positive control, cytochalasin for inhibition of actin polymerization, S. aureus, and S. warneri. NET formation was observed in response to chemical stimulation and bacterial presence, effectively trapping pathogens. Variations in NET formation between S. aureus and S. warneri suggest pathogen-specific responses. These findings suggest that the formation of NETs may be an important complementary mechanism in the defense against mastitis in goats. In conclusion, this study unveils a novel defense mechanism in goats, indicating the role of NETs against S. aureus and S. warneri in mastitis.

In herds of dairy goats, mastitis represents a major health and economic problem due to the multiresistance of some microorganisms. In this context, the study aimed to determine the potential of antimicrobial action and antibiofilm of the crude ethanolic extract (CEE) of Hymenaea martiana (jatobá) leaves, as well its fractions, on Staphylococcus sp isolated from bacterial cultures of goat milk. In vitro assays were performed to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC), as well as tests of the effect of CEE on biofilm formation and quantification and the consolidated biofilm. The experimental infection was performed in two groups, each consisting of five goat. Experimental Group 1 (G1) consisted of five females treated with an intramammary ointment based on the CEE, at a concentration of 5%. Experimental Group 2 (G2) consisted of five females treated with a commercial intramammary ointment based on gentamicin, once a day, for six consecutive days. The diagnosis of mastitis was performed using a bacterial culture. The dichloromethane fraction of CEE was the one with the lowest concentrations of MBC, ranging from 195.3 to 781 µg / ml. Concerning to the biofilm, interference of the tested extract was observed for two isolates. In the present study, the ointment prepared from H. martiana extract (jatobá) was able to reduce bacterial infection in mammary glands experimentally infected with S. aureus. Antibacterial activity may be related to the classes of secondary metabolites found.

Objetivo:analizar la evidencia de estudios previos sobre las diferentes alternativas de tratamientos con el uso de agentes físicos y técnicas manuales utilizados en la fisioterapia para la ingurgitación mamaria a nivel internacional. Material y método:estudio de revisión sistemática con meta-análisis según el pro-tocolo prisma. Búsqueda en las bases de datos de Scopus y Medline a través de PubMed, publicados desde el 01 de enero de 2015 hasta el 31 de diciembre de 2021. La estrategia de búsqueda empleó los siguien-tes términos: breastengorgement, treatment, breastfeeding, six-pointengorgementscale, physiotherapy, breastcancer, physicaltherapy. Los 5 estudios elegidos para esta revisión sistemática fueron valorados con la Escala pedro para conocer la calidad metodológica. Resultados:diferencias medias estandarizadas oscilaron entre 0,5959 y 2,7373, la mayoría de las estimaciones positivas. El resultado promedio difirió significativamente de cero (z = 3,5686, p = 0,0004). Según prueba Q, los resultados reales parecen ser heterogéneos (Q (5) = 23,2212, p = 0,0003, tau² = 0,4759, I² = 83,6163%). Intervalo de predicción del 95 % para los resultados reales viene dado por -0,3733 a 2,5931. Un intervalo de predicción del 95 % para los resultados reales viene dado por -3,6762 a 11,5933. Conclusiones: las técnicas estudiadas disminuyen el dolor y la congestión mamaria, sin embargo, se ha visto que la combinación entre ellas puede favorecer aún más la mejora de los mismos. Al aplicarse en las mujeres con ingurgitación generaron beneficios a corto y largo plazo para disminuir el dolor y la ingurgitación mamaria
Objective: To analyze the evidence of previous studies on the different treatment alternatives with the use of physical agents and manual techniques used in physiotherapy for breast engorgement at an international level. Method: Systematic review study with meta-analysis according to the prisma pro-tocol. Search in Scopus and Medline databases through PubMed, published from January 1, 2015 to December 31, 2021. The search strategy employed the following terms: breast engorgement, treatment, breastfeeding, six-point engorgement scale, physiotherapy, breast cancer, Physical therapy. The 5 stu-dies chosen for this systematic review were assessed with the PEDro Scale for methodological quality. Results: Standardized mean differences ranged from 0.5959 to 2.7373, most of the positive estimates. The average result differed significantly from zero (z = 3.5686; p = 0.0004). According to Q test, the actual results appear to be heterogeneous (Q(5) = 23.2212; p = 0.0003; tau² = 0.4759; I² = 83.6163%). The 95% prediction interval for the actual results is given by −0.3733 to 2.5931. A 95% prediction interval for the actual results is given by −3.6762 to 11.5933. Conclusions: The techniques studied decrease breast pain and engorgement; however, it has been seen that the combination between them can further favor their improvement. When applied in women with engorgement, they generated short and long term benefits in reducing pain and breast engorgement

La mastitis linfocítica o mastitis diabética es una patología benigna rara, caracterizada por una inflamación fibrótica de la mama, causando gran preocupación por la simulación de un carcinoma. Está asociada por lo general a la diabetes tipo 2, aunque pueden presentarse en otras patologías autoinmunes y además en pacientes sin comorbilidades. Se presenta en caso clínico de una paciente joven sin comorbilidades que consulto en consultorio de patología mamaria por tumor mamario. Se realizó una biopsia percutánea por trucut con confirmación histológica de mastitis linfocítica, se realizó control cercano observándose remisión clínica e imagenológica de la lesión en 6 meses de seguimiento. Se presenta el caso a fin de recalcar la importancia del diagnóstico histológico percutáneo de la lesión para determinar la naturaliza de la misma, constatándose una patología benigna que no requiere resección, evitando así causar deformidades que podrían afectar psicológicamente a la paciente.
Lymphocytic mastitis or diabetic mastitis is a rare benign pathology, characterized by fibrotic inflammation of the breast, causing great concern due to the simulation of carcinoma. It is generally associated with type 2 diabetes, although it can occur in other autoimmune pathologies and also in patients without comorbidities. The clinical case of a young patient without comorbidities who consulted in the breast pathology clinic due to a breast tumor. A percutaneous trucut biopsy was performed with histological confirmation of lymphocytic mastitis, close control was performed, observing clinical and imaging remission of the lesion in 6 months of follow-up. The case is presented in order to emphasize the importance of percutaneous histological diagnosis of the lesion, to determine its nature, confirming a benign pathology that does not require resection, thus avoiding causing deformities that could psychologically affect the patient.

Using on-farm microbiological culture (OFC), based on chromogenic culture media, enables the identification of mastitis causing pathogens in about 24 h, allows rapid decision making on selective treatment and control management measures of cows with clinical mastitis (CM). However, accurate interpretation of OFC results requires trained and experienced operators, which could be a limitation for the use of OFC in dairy farms. Our hypothesis was that AI-based automated plate reading mobile application can analyze images of microorganisms' colonies in chromogenic culture media with similar diagnostic performance as a trained specialist evaluator. Therefore, the aim of the present study was to evaluate the diagnostic accuracy of an AI-based application (Rumi; OnFarm, Piracicaba, São Paulo, Brazil) for interpreting images of mastitis causing microorganism colonies grown in chromogenic culture media. For this study two trials were organized to compare the results obtained using an AI-based application Rumi with the interpretation of: (1) a trained specialist, using MALDI-TOF MS as the gold standard; (2) farm personnel users (FPU). In trial 1, a total of 476 CM milk samples, from 11 farms located in São Paulo (n = 7) and Minas Gerais (n = 4), southeast Brazil, were inoculated in chromogenic culture media plates (Smartcolor 2, OnFarm, Piracicaba, São Paulo, Brazil) by specialists under lab conditions, and digital images were recorded 24 h after incubation at 37 °C. After that, all the 476 digital images were analyzed by the Rumi and by another specialist (who only had access to the digital images) and the diagnostic accuracy indicators sensitivity (Se) and specificity (Sp) were calculated using MALDI-TOF MS microbiological identification of the isolates as the reference. In Trial 2, a total of 208 CM milk samples, from 150 farms from Brazil, were inoculated in chromogenic culture media plates by FPU, and the results of microbiological growth were visually interpreted by FPU under on-farm conditions. After visual interpretation, results were recorded using an OnFarmApp application (herd manage application for mastitis by OnFarm, Piracicaba, São Paulo, Brazil), and the images of the chromogenic culture plates were captured by the OnFarmApp to be evaluated by Rumi and Bayesian Latent Class Models were performed to compare Rumi and the FPU. In Trial 1, Rumi presented high and intermediate accuracy results, with the only exception of the low Enterococcus spp.'s Se. In comparison with the specialist, Rumi performed similarly in Se and Sp for most groups of pathogens, with the only exception of non-aureus staphylococci where Se results were lower. Both Rumi and the specialist achieved Sp results > 0.96. In Trial 2, Rumi had similar results as the FPU in the Bayesian Latent Class Model analysis. In conclusion, the use of the AI-based automated plate reading mobile application can be an alternative for visual interpretation of OFC results, simplifying the procedures for selective treatment decisions for CM based on OFC.

Two cases of mammary tuberculosis (TB) are presented, one of them with additional pleural and lymph node involvement. Both were HIV-negative, with no history of previous TB, with long-standing breast lesions. Both presented a successful outcome with antituberculosis treatment. Breast TB is a very rare pathology that is difficult to diagnose. The cases are presented to consider their differential diagnosis in patients with chronic mastitis and/or nodular or ulcerated lesions of the breast. Multidisciplinary management is recommended.
Se presentan los casos de dos pacientes con tuberculosis (TB) mamaria, una de ellas también con compromiso pleural y ganglionar. Ambas HIV negativas, sin antecedentes de TB previa, con lesiones mamarias de largo tiempo de evolución. Las dos presentaron buena evolución con tratamiento antifímico. La TB mamaria es una afección muy poco frecuente de difícil diagnóstico. Se presentan los casos con el fin de plantear su diagnóstico diferencial en pacientes con mastitis crónicas y/o lesiones nodulares o ulceradas de la mama. Se recomienda el manejo multidisciplinario.

The increase in bacterial resistance to antibiotics is a global problem for public health. In our previous works, it was shown that the application of AgNPs in cow mastitis treatment increased S. aureus and S. dysgalactiae susceptibility to 31 antibiotics due to a decrease in the bacterial efflux effect. The aim of the present work was to shed light on whether the change in adhesive and anti-lysozyme activities caused by AgNPs also contribute to the restoration of bacterial susceptibility to antibiotics. In vivo sampling was performed before and after cow mastitis treatments with antibiotics or AgNPs. The isolates were identified, and the adhesive and anti-lysozyme activities were assessed. These data were compared with the results obtained for in vitro pre-treatment of reference bacteria with AgNPs or antibiotics. The present study revealed that bacterial treatments in vitro and in vivo with AgNPs: (1) decrease the bacterial ability to adhere to cells to start an infection and (2) decrease bacterial anti-lysozyme activity, thereby enhancing the activity of lysozyme, a natural "antibiotic" present in living organisms. The obtained data contribute to the perspective of the future application of AgNPs for recovering the activity of antibiotics rapidly disappearing from the market.

Selenium is an important element in nutrition, showing great potential in the udder health of dairy goats and in the control of subclinical mastitis. However, there are few studies that evaluated the influence of selenium supplementation on subclinical mastitis in goats. The aim of this study was to evaluate the incidence of subclinical mastitis in dairy goats supplemented with organic selenium (Se yeast) in a semi-arid region. Sixteen Saanen × Toggenburg crossbred lactating goats were allocated randomly into two treatments: with and without addition of organic selenium (Se) to the concentrate. Milk samples were collected every 20 days from each udder half to determine the somatic cell count (SSC), chloride content, pH, electrical conductivity, microbiological isolation, composition, and selenium contents. The highest serum selenium concentrations in the blood of these goats occurred at 42 days of supplementation (P < 0.001). Goats which received organic selenium supplementation had higher serum selenium concentrations (P < 0.05). The milk composition variables did not differ (P > 0.05) between the tested treatments, teats, and collections. After 60 days of supplementation, a difference was observed (P < 0.05) between treatments for SSC, chloride content, and pH. Addition of organic selenium to the diet of dairy goats after 60 days of supplementation was promising in reducing the somatic cell count, consequently improving milk quality.

Background: We report the case of 3 cows, 1 ewe, and 1 doe, with chronic mastitis and which underwent a mastectomy. All animals had a history of mastitis, edema with purulent secretion in the udder region, areas of abscess formation, fibrosis, hypogalactia, and agalactia. The animals had already been treated with systemic antibiotic therapy and anti-inflammatory drugs without response. The objective of this work is to describe the surgical and clinicopathological aspects of performing a radical mastectomy in the field in 3 cows and 1 ewe, and a partial mastectomy in a doe, with both procedures performed successfully and considered efficient to maintain the survival of the affected animals, in addition to being economically viable. Cases: In all animals reported in the study, a general and a specific clinical examination were performed, where on palpation of the udder were observed an increase in the region of the mammary apparatus, fluid fluctuation, areas of fibrosis with increased stiffness, and loss of udder functionality. In the case of the doe, there was a functional teat, which was preserved during the surgical procedure. Radical mastectomy was performed in 3 cows and 1 ewe, and the doe underwent a partial mastectomy, with preservation of the functional teat. All animals have fasted for food and water for 48 h. In cows, the anesthetic protocol consisted of 2% xylazine hydrochloride [0.15 mg/kg, intramuscularly (IM)], followed by an epidural block in the lumbosacral region with 2% lidocaine hydrochloride [10 mL]. The midazolam hydrochloride [0.1 mg/kg, intravenously (IV)] and 2% xylazine hydrochloride [0.05 mg/kg, IM] were administered to the ewe and the doe, also associated with lumbosacral epidural anesthesia with 2% lidocaine hydrochloride [5 mL]. The animals were positioned in the right lateral decubitus position and the entire abdominal and udder region were shaved and sanitized for the surgical procedures of radical mastectomy and partial mastectomy, with the use of degerming and topical povidone-iodine. The postoperative period consisted of penicillin-based antibiotic therapy associated with streptomycin [24,000 IU/kg, IM, 10 consecutive days] and flunixin meglumine [2 mg/kg, IM, 3 consecutive days] in cows; enrofloxacin [2.5 mg/kg, IM, 7 consecutive days] and flunixin meglumine [0.5 mg/kg, IM, 5 consecutive days] in the ewe; oxytetracycline [10 mg/kg, IM, 5 applications on alternate days] and flunixin meglumine [0.5 mg/kg, IM, 3 consecutive days] in the doe. In all animals, the cleaning of the surgical wound was performed daily with 10% topical Povidone-iodine and Permethrin ointment, for 14 days, when the stitches were removed. Discussion: The choice of the surgical procedure resulted in improvement of the clinical conditions and increased survival of all animals, and in the case of the doe, partial mastectomy contributed to the preservation of the functional teat, preserving its productive capacity. All anesthetic protocols used and the anatomical positioning of the animals were satisfactory for performing the procedures. Although they are techniques described in the literature with great blood loss, especially in cows, blood transfusion was not required in any of the animals. The stitches were removed 14 days after the surgical procedures, and the surgical wound was completely healed.

This study evaluated if the milk ring test (MRT) used to the brucellosis diagnostic at dairy herd level can be done in milk samples with conservative (Bronopol®) collected to the somatic cell count (SCC) that is used to mastitis monitoring. It were analyzed 38 bulk tank milk samples (BTMS) from 19 dairy herds artisanal cheese producers and 36 BTMS from a brucellosis free dairy herd, used as negative control (NC) and positive control (PC), when was added serum from vaccinated animals. The milk samples were collected in two different bottles (with or without Bronopol®). All the 38 BTMS from artisanal cheese producers (samples with and without Bronopol®) were non-reagent to MRT, as all NC. All PC were reagent to MRT, without interference of Bronopol®. Results showed that the milk sample used to SCC presented the proportion of agreement of results in all milk samples (100%), that is, false positive and false negative results were not observed. Results indicated that Bronopol® did not interfere with the color in the time of MRT reading suggesting that the same milk samples could be used to monitoring mastitis and brucellosis.
O objetivo deste estudo foi avaliar se o teste do anel do leite (TAL), utilizado para o diagnóstico da brucelose em nível de rebanho leiteiro, pode ser realizado em amostras de leite com conservador (Bronopol®) coletado para contagem de células somáticas (CCS) que é utilizado para monitoramento de mastite. Foram analisadas 38 amostras de leite de tanques de expansão (ALTE) de 19 rebanhos leiteiros produtores de queijo artesanal e 36 ALTE de rebanho leiteiro livre de brucelose, utilizadas como controle negativo (CN) e controle positivo (CP), quando foi adicionado soro de animais vacinados. As amostras de leite foram coletadas em dois frascos diferentes (com e sem Bronopol®). Todos as 38 ALTE de produtores artesanais de queijo (amostras com e sem Bronopol®) foram não reagentes ao TAL, como todos os CN. Todos os CP foram reagentes ao TAL, sem interferência do Bronopol®. Os resultados mostraram que a amostra de leite utilizada para CCS apresentaram a proporção de concordância dos resultados em todas as amostras de leite (100%), ou seja, não foram observados resultados falso-positivos e falso-negativos. Os resultados indicam que o Bronopol® não interferiu na cor no momento da leitura do TAL, sugerindo que as mesmas amostras de leite poderiam ser usadas para monitorar mastite e brucelose.

Background: Staphylococcus spp. are the most frequently isolated microorganisms in mastitis cases of small ruminants. The virulence factors of Staphylococcus spp. are critical in the treatment. Therefore, preventive medicine and mastitis control programs, especially herd vaccinations are of great importance in the prevention of mastitis. However, it is not always easy to obtain these vaccines under field conditions. This study, it was aimed to compare the effects of different commercial Staphylococcus spp. vaccines licensed for bovines and species-specific mastitis vaccines on mastitis rates and somatic cell count (SCC) on Saanen goats on field conditions. Materials, Methods & Results: The animal material consisted of 115 (230 udder halves) nulliparous Saanen goats. Goats were randomly grouped as bovine vaccine 1 (BV1 , n = 58), bovine vaccine 2 (BV2 , n = 58), small ruminant vaccine (SRV, n = 56), and control (n = 56). Vaccines were administered to goats in 2 doses according to the label regimen. First milk samples were collected between 0-5 days in milk (DIM) for microbiological analysis and 25-35 DIM for SCC. The other milk samples were collected at 25-35 (1st month) DIM, 60-65 (2nd month) DIM, 85-95 (3rd month) DIM, 115-125 (4th month) DIM, 145-155 (5th month) DIM for microbiological analysis and SCC. Non-aureus staphylococci (NAS) and Staphylococcus aureus were the most frequently isolated microorganism. It was found that the total mastitis rate decreased in vaccine groups compared to the control group. A significant difference was found only in the BV2 and SRV groups. The significant difference in S. aureus infection was found only in the SRV group. Mastitis vaccines used in this study decreased the NAS mastitis rate, but no significant difference was observed. It was found that the clinical mastitis incidence decreased in all vaccine groups compared to the control group, and a significant difference was found between the BV2 and SRV groups compared to the control group (P < 0.05). Somatic cell count was lower in the SRV and BV2 groups compared to the control group (P < 0.05). Discussion: In this study, compatible with the previous reports NAS and S. aureus were the most frequently isolated microorganism. The diversity of virulence factors of Staphylococcus spp. also plays an important role in its high incidence. In some countries, mastitis vaccines used in cows are also administered to small ruminants for reducing infection rates. Similarly, in this study, it was found that the mastitis rate decreased in all vaccine groups compared to the control group. A significant difference was found only in the BV2 and SRV groups. It is thought that the reason for the statistical difference may be due to the biofilm antigen in the BV2 and SRV. In addition, J5 strain in the BV2 is estimated to be effective in reducing the prevalence of gram-negative mastitis. It was observed that the infection rates decreased in the vaccine groups, especially due to S. aureus and NAS. Spontaneous treatment rates were very close to each other between the groups. The reason for the high rate of spontaneous treatment in this study can be explained by the fact that the animals were young and in their 1st lactation. SCC was lower in all vaccine groups compared to the control group. This situation is associated with the decrease in infection rates related to the use of vaccines. It was observed that SCC was lower in the vaccine groups. In addition, SCC was found to be lower in this study compared to similar studies. However, it is evident that the use of species-specific vaccines in the SRV group significantly reduced the rates of total S. aureus mastitis, subclinical NAS mastitis, and new infections by NAS compared to other vaccines. Furthermore, the species-specific vaccine significantly increased the rate of spontaneous treatment for S. aureus mastitis.

This research aims to describe the cost-benefit analysis of selective therapy adoption for clinical mastitis based on the identification of pathogens using on-farm culture system. A survey of clinical mastitis cases was carried out in the database of a dairy herd with 500 lactating Holstein cows, during the period of September 2017 to October 2019. Variables such as frequency of cases, main pathogens identified, costs with antibiotic therapies and milk disposal were evaluated and compared between one year before (P1) and after (P2), the farm's selective therapy adoption. During the study period, 599 cases of clinical mastitis were identified, 361 in P1 and 238 in P2. The average cost of therapies was US74.33 and US91.82 for P1 and P2. Considering the two years of the study, the estimated cost of treating clinical mastitis was US45,444.31, of which US$27,559.97 were spent in P1 and US17,884.34 in P2. Based on the result of the identification of the pathogen types associated with clinical mastitis, selective therapy demonstrated cost reduction already in the first year of implementation.(AU)
Esta pesquisa objetivou descrever a análise custo-benefício da adoção de terapia seletiva para mastite clínica a partir da identificação de patógenos por meio da cultura em placas na fazenda. Foi realizado um levantamento de casos clínicos de mastite no banco de dados de uma fazenda com 500 vacas Holandesas em lactação, durante o período de setembro de 2017 à outubro de 2019. Foram avaliadas as variáveis frequência de casos, principais patógenos identificados, custos com antibioticoterapia e descarte de leite foram, as quais foram comparados entre um ano antes (P1) e após (P2), à adoção da terapia seletiva na fazenda. Durante o período do estudo, foram identificados 599 casos de mastite clínica, sendo 361 em P1 e 238 em P2. O custo médio das terapias foi de US74,33 e US91,82 para P1 e P2. Considerando os dois anos de estudo, o custo estimado do tratamento da mastite clínica foi de US45.444,31, dos quais US27.559,97 foram gastos em P1 e US17.884,34 em P2. Com base na identificação dos tipos de patógenos associados à mastite clínica, a terapia seletiva demonstrou redução de custos já no primeiro ano de implantação.(AU)

This study aimed to characterize the production and management systems of white Morada Novasheep herds and to present data on the occurrence of caseous lymphadenitis (CLA) and clinical mastitis (CM) in a population maintained in a semi-arid region. Blood samples were collected from adult females from herds of the Brazilian northeastern region. The first venous blood sample was used for a complete blood examination. The second blood sample was used to detect the presence of anti-C. pseudotuberculosis specific antibodies. The production and management systems of the herds were characterized using questionnaires, divided into three blocks of indicators: Facilities; Characterization of the production system; Health management. Body weight and hematological results were submitted to an analysis of variance, on which the fixed effect was clinical diagnosis, and the principal component analysis was performed to determine the main variables. For CLA, three herds stood out as having the highest rates of seropositivity (85.71, 79.41 and 60.45%). The same herds also had the highest occurrences of CM (9.30, 8.82 and 5.71%). The main variables that showed differences according the diagnosis of both diseases were blood granulocytes and lymphocytes. The study made it possible to observe the relationship between the best sanitary practices and the lowest occurrence rates of both CLA and CM. Este estudo teve como objetivo caracterizar os sistemas de produção e manejo de rebanhos de ovelhas Morada Nova da variedade branca e apresentar dados sobre a ocorrência de linfadenite caseosa (CLA) e mastite clínica (MC) em uma população mantida em região semiárida. Amostras de sangue foram coletadas de fêmeas adultas de rebanhos da região nordeste do Brasil. A primeira amostra de sangue venoso foi utilizada para um exame de sangue completo. A segunda amostra de sangue foi utilizada para detectar a presença de anticorpos anti-C. anticorpos específicos de pseudotuberculosis. Os sistemas de produção e manejo dos rebanhos foram caracterizados por meio de questionários, divididos em três blocos de indicadores: Instalações; Caracterização do sistema de produção; Gestão de saúde do rebanho. O peso corporal e os resultados hematológicos foram submetidos a uma análise de variância, na qual o efeito fixo foi o diagnóstico clínico, e a análise de componentes principais foi realizada para determinar as variáveis principais. Para o CLA, três rebanhos se destacaram com os maiores índices de soropositividade (85,71, 79,41 e 60,45%). Os mesmos rebanhos também apresentaram as maiores ocorrências de MC (9,30, 8,82 e 5,71%). As principais variáveis que apresentaram diferenças de acordo com o diagnóstico de ambas as doenças foram granulócitos e linfócitos. O estudo possibilitou observar a relação entre as melhores práticas sanitárias e as menores taxas de ocorrência tanto de CLA quanto de MC. INTRODUCCIÓN. La mastitis granulomatosa idiopática es una patología inflamatoria benigna de mama con clínica y hallazgos imagenológicos no específicos; usualmente confundida con cáncer de mama. El síntoma más frecuente es una masa mamaria palpable. El diagnóstico es histopatológico. OBJETIVO. Describir el perfil demográfico, presentación clínica y hallazgos radiográficos de pacientes con diagnóstico histopatológico de mastitis granulomatosa idiopática. MATERIALES Y MÉTODOS. Estudio observacional, descriptivo, retrospectivo. Población de 1130 y muestra de 49 datos de historias clínicas electrónicas de pacientes con diagnóstico histológico de mastitis granulomatosa idiopática con el código CIE10 N61x Trastornos Inflamatorios de la mama, atendidas en la Unidad Técnica de Imagenología del Hospital de Especialidades Carlos Andrade Marín en la ciudad de Quito entre enero 2019 hasta diciembre 2021. El criterio de inclusión fue la confirmación histopatológica de mastitis granulomatosa idiopática. Los criterios de exclusión: antecedentes de neoplasia maligna de mama, antecedentes de HIV, patología inflamatoria sistémica como granulomatosis de Wegener, sarcoidosis, infecciones granulomatosas crónicas como tuberculosis, brucelosis, histoplasmosis, sífilis y reacciones a cuerpos extraños como material de implantes mamarios. Se analizaron datos demográficos, presentación clínica, hallazgos mamográficos, ecográficos y la categorización BIRADS. Se efectuó un análisis univarial; para las variables cualitativas se realizó frecuencias y porcentajes; para las variables cuantitativas se realizó medidas de tendencia central. La información recolectada fue analizada en el programa estadístico International Business Machines Statistical Package for the Social Sciences. RESULTADOS La mediana de la edad fue 36 años. El 94,00% de pacientes tenían por lo menos un hijo; 77,50% presentaron con una masa palpable; 55,10% se acompañaron de signos inflamatorios; 16,00% asociaron fístulas y 24,40% presentaron secreción. Solo 1 caso presentó afectación bilateral. CONCLUSIÓN En este estudio, la mastitis granulomatosa idiopática afecta a mujeres en edad reproductiva sin antecedentes de malignidad quienes presentan una masa mamaria palpable que puede estar acompañada de signos inflamatorios, colecciones y fístulas. La realización de una biopsia core eco guiada, para confirmar su diagnóstico.
INTRODUCTION. Idiopathic granulomatous mastitis is a benign inflammatory breast pathology with nonspecific clinical and imaging findings; usually mistaken for breast cancer. The most frequent symptom is a palpable breast mass. The diagnosis is histopathologic. OBJECTIVE. To describe the demographic profile, clinical presentation and radiographic findings of patients with histopathologic diagnosis of idiopathic granulomatous mastitis. MATERIALS AND METHODS. Observational, descriptive, retrospective study. Population of 1130 and sample of 49 data from electronic medical records of patients with histological diagnosis of idiopathic granulomatous mastitis with ICD10 code N61x Inflammatory disorders of the breast, attended at the Technical Imaging Unit of the Carlos Andrade Marín Specialties Hospital in the city of Quito between January 2019 and December 2021. The inclusion criterion was histopathological confirmation of idiopathic granulomatous mastitis. Exclusion criteria: history of malignant breast neoplasia, history of HIV, systemic inflammatory pathology such as Wegener's granulomatosis, sarcoidosis, chronic granulomatous infections such as tuberculosis, brucellosis, histoplasmosis, syphilis and reactions to foreign bodies such as breast implant material. Demographic data, clinical presentation, mammographic and ultrasound findings and BIRADS categorization were analyzed. Univariate analysis was performed; frequencies and percentages were used for qualitative variables; measures of central tendency were used for quantitative variables. RESULTS. The median age was 36 years. 94,00% of patients had at least one child; 77,50% presented with a palpable mass; 55,10% were accompanied by inflammatory signs; 16,00% were associated with fistulas and 24,40% presented with discharge. Only 1 case presented bilateral involvement. CONCLUSION. In this study, idiopathic granulomatous mastitis affects women of reproductive age with no history of malignancy who present with a palpable breast mass that may be accompanied by inflammatory signs, collections and fistulas. The performance of an echo-guided core biopsy to confirm the diagnosis.

Extended therapy is an alternative approach to treat nonresponsive staphylococcal mastitis, although it has been used mainly in adult cows. This study investigated the efficacy of intramammary ceftiofur in the treatment of staphylococcal subclinical mastitis in primiparous cows. Sixty primiparous cows Holstein were divided into four groups (G1 = Group negative to staphylococci infection, G2 = Group positive to staphylococci infection untreated with intramammary ceftiofur, Group, G3 = Group treated with intramammary ceftiofur early lactation, G4 = Group treated with intramammary ceftiofur late lactation group) with milk samples collected at five moments (M0 = diagnosis of subclinical mastitis, M1 = 7 days after diagnosis of subclinical mastitis and early extended therapy, M2 = 14 after the diagnosis of subclinical mastitis, M3 = 21 days after diagnosis of mastitis) and M4 = 28 days after diagnosis of mastitis). In groups G3 and G4, which were treated with intramammary ceftiofur, the microbiological cure was evidenced by a reduction in SCC of 73.3% and 46.7%, respectively. Thus, in the treated groups (G3 + G4) and untreated (G2), cure rates were 60% and 26.7%, respectively (P <0.05). Forty-five strains of staphylococci were Motta, R. G. et al.1958Semina: Ciênc. Agrár. Londrina, v. 43, n. 5, p. 1957-1974, set./out. 2022isolated, with a predominance of S. aureus (51,1%) and S. intermedius (48,8%). The in vitro antimicrobial susceptibility pattern showed higher sensitivity indices of isolates for oxacillin (80%), ceftiofur (77.8%), cephalexin (77.8%), ciprofloxacin (66.7%), and gentamicin (60%). In contrast, cloxacillin (24.4%), penicillin (35.6%), and tetracycline (44.5%) were ineffective antimicrobials for staphylococci isolates. Among the milk samples taken when M0 and M1 (7 days after isolation), 13.3% were positive for the detection of the inhibiting substances (antimicrobials) in Delvotest™ and all negative in the Snap test™, whereas in the M2 (14 days after isolation), 55% were positive in Delvotest™ and 46.7% in the Snap test™. Twenty-one days after diagnosis (M3), 16.7% revealed positive results in Delvotest™ and 11.7% in the Snap test™. For M4 (28 days after isolation), 11.7% showed positive results in Delvotest™, and 8.3% revealed positive results in the Snap test™. There was a reduction in SCC among all cured animals. Significant results were identified for milk production in the M0, so G1 produced more milk than the other three groups, G2, G3, and G4. A statistical association was observed between the reduction in the volume of daily milk and production in animals with subclinical mastitis, denoting losses in milk production in primiparous bovines infected during lactation by staphylococci. Extended intramammary therapy with ceftiofur in bovine primiparous cows subclinically affected by staphylococcal mastitis is effective mainly in cases treated at the beginning of lactation, showing microbiological cure and reduction in SCC.(AU)
O estudo presente investigou a eficácia do ceftiofur intramamário no tratamento da mastite subclínica por estafilococos em primíparas bovinas. Foram utilizados 60 animais da raça holandesa, divididos em quadro grupos (G1 = Grupo negativo, G2 = Grupo positivo não tratado com ceftiofur intramamário, G3 = Grupo tratado com ceftiofur intramamário no início da lactação, G4 = Grupo tratado com ceftiofur intramamário no final da lactação), com amostras de leite coletadas em cinco momentos (M0 = diagnóstico da mastite subclínica, M1 = 7 dias após diagnóstico e início da terapia estendida, M2 = dia 14 após o diagnóstico da mastite subclínica, M3 = dia 21 após o diagnóstico da mastite, M4 = dia 28 após o diagnóstico da mastite). Nos grupos tratados com ceftiofur intramamário, foi evidenciada a cura microbiológica com redução na CCS em 73,3% no G3 e 46,7% no G4. Nos grupos tratados (G3+G4) e não tratado (G2), as taxas de cura foram, respectivamente, 60% e 26,7% (p<0,05). Foram isoladas 45 linhagens de estafilococos, com predomínio de S. aureus (51,1%) e S. intermedius (48,8%). O perfil de sensibilidade microbiana "in vitro" dos isolados apresentou maiores índices de sensibilidade para oxacilina (80%), ceftiofur (77,8%) e cefalexina (77,8%), ciprofloxacino (66,7%) e gentamicina (60%). Em contraste, cloxacilina (24,4%), penicilina (35,6%) e tetraciclina (44,5%) foram os antimicrobianos menos efetivos diante dos estafilococos isolados. Nas amostras de leite colhidas nos momentos M0 e M1, 13,3% foram positivas para o teste de detecção de substâncias inibidoras na prova de Delvotest™ e todas negativas no Snap test™. Já, no M2, 55% foram positivas no Delvotest™ e 46,7% no Snap test™. No M3, 16,7% positivas no Delvotest™ e no 11,7% no Snap test™. No M4, 11,7% positivas no Delvotest™ e 8,3% no Snaptest™, dois dias após o término do tratamento (M2). Foram identificados nos grupos G3 e G4 a presença de animais positivos em 96,7% para o Delvotest™ e 90% para o Snap test™. Ocorreu redução na CCS em todos os animais curados. Foram identificados resultados significativos para a produção de leite no MO, posto que o G1 produziu mais leite se comparado aos outros três grupos G2, G3 e G4. Foi observada associação estatística entre a redução no volume de leite diário e a produção nos animais com mastite subclínica, denotando prejuízos na produção de leite em primíparas bovinas infectadas na lactação por estafilococos. A terapia intramamária estendida com ceftiofur em primíparas com mastite estafilocócica é eficaz principalmente nos casos tratados no início da lactação, mostrando cura microbiológica e redução da CCS, indicando que esta abordagem terapêutica alternativa pode ser utilizada para o tratamento de novilhas.(AU)

The purpose of this study was to evaluate the effect of retained placenta (RP) and clinical mastitis (CM) on the reproductive efficiency of crossbred dairy cows during the postpartum period and the effect in some innate immune system indicators. For this, two experiments were carried out. In the first, a total of 232 cows were evaluated and divided as: healthy control (n = 184), RP (n = 22), and CM (n = 26) groups. The RP and CM was evaluated until 30 days postpartum (DPP) and reproductive rates were measured. In experiment 2, cows were divided in control (n = 10), RP (n = 10), and CM (n = 30) groups. Between 40 and 50 DPP, clinical, gynecological examination and endometrial cytobrush were performed to evaluate subclinical endometritis (SE) and gene expression of interleukins 1ß (IL-1ß) and 6 (IL-6), chemokine ligand 5 (CCL5), estrogen α (ESR1), and progesterone (PGR) receptors by qRT-PCR analysis. In experiment 1, the conception rate at 1st artificial insemination (AI) was lower in RP and CM groups and pregnancy rate at 150 days decreased in CM group. Calving-to-1st AI interval and days open were shorter in healthy cows. In experiment 2, the occurrence of SE was 26.7% and higher in RP and CM groups. The expression of IL-1ß increased in RP and CM groups, while IL-6 was less expressed in RP group. The CCL5, ESR1, and PGR were similar between groups. In conclusion, cows with RP and CM had their reproductive efficiency negatively affected and had they initial pro-inflammatory response improved by the increase of IL-ß.

Several studies have assessed the therapeutic potential of natural products against mastitis, but only a few have evaluated the impacts of this alternative therapy on the main properties of milk and dairy products. In this study, we observed how the treatment of mastitis with ethanolic extract of Jatobá (Hymenaea martiana Hayne) influenced the physicochemical and sensory characteristics of coalho cheese. An ointment containing the ethanolic extract was prepared for intramammary use in six dairy goats. The experiment was conducted in three experimental moments. Milking was performed, manually, and both milk and cheese were subjected to physicochemical and sensory tests. No difference was observed (p>0.05) in the physicochemical aspects of milk between the studied groups. The solids-non-fat showed a statistical difference between experimental moments M1 and M2. The protein means varied from 3.33 to 3.62, and there was a statistical difference between the two moments, while the lactose means varied from 4.79 to 5.38%. The physicochemical aspects of cheese remained similar with both treatments. Except for appearance, the sensory characteristics showed no statistical difference. In conclusion, the use of Jatobá extract ointment did not influence the physicochemical and sensory characteristics of goat milk or cheese.
A mastite representa uma das principais enfermidades nos rebanhos de caprinos leiteiros, com implicações na produção de leite e na qualidade final dos derivados. Objetivou-se observar a influência do tratamento da mastite com o extrato etanólico da Hymenaea martiana Hayne sobre as características físico-químicas e sensoriais do queijo tipo coalho. Uma pomada contendo extrato foi elaborada para uso intramamário em seis fêmeas caprinas especializadas na produção de leite. As metades esquerdas dos tetos das cabras receberam essa infusão, enquanto a metade direita era controle. O experimento foi conduzido em três momentos experimentais. Realizou-se ordenha manual e análise físico-química do leite. Para o queijo, foram realizados testes físico-químicos e teste sensorial. Quantos aos aspectos físico-químicos do leite, é possível afirmar que não houve diferença (P>0,05) entre os grupos. Houve diferença estatística para sólidos não gordurosos nos momentos experimentais M1 e M2. As médias de proteína variaram entre 3,33 e 3,62, e houve diferença estatística em dois momentos. As médias de lactose variaram entre 4,79% e 5,38%. Os aspectos físico-químicos do queijo mantiveram resultados similares entre os dois tratamentos. Os parâmetros sensoriais para o queijo coalho de cabra não tiveram diferença estatística entre os tratamentos, contudo a aparência se destacou em relação aos demais parâmetros sensoriais. O uso da pomada elaborada com extrato de jatobá não interferiu sobre as características físico-químicas do leite de cabra e sobre as características físico-químicas e sensoriais do queijo tipo coalho de cabra.

Evaluate the effects of antimicrobial photodynamic therapy (aPDT) and photobiomodulation (PBM) as alternatives in the treatment of mastitis in sheep. A total of 100 sheep were evaluated, and four teats with clinical mastitis and 16 teats with subclinical mastitis were selected. Milk was collected for isolation and identification of microorganisms. They were grown on TSA, EMB, and MacConkey agar for 24 h, and the microorganisms were identified by Gram stain and biochemical tests. The ceilings were subdivided into four groups: G1, treatment with photosensitizer; G2, treatment with PBM (diode laser λ = 660 nm); G3, aPDT with methylene blue, and G4, control group. Milk samples were collected before, 24 and 48 h after treatments. Cases of subclinical mastitis presented coagulase-negative Staphylococcus and Streptococcus spp, and clinical mastitis had Escherichia coli grow from the samples. The treatments decrease the total bacterial count of negative coagulase Staphylococcus, Streptococcus spp, and Escherichia coli. Comparing the treatments, aPDT stood out, as it was able to photoinactivate all bacteria. Treatment with methylene blue photosensitizer, PBM, and aPDT induced the initial microbial reduction, but aPDT was more effective 48 h after treatment.
