This study aimed to evaluate the internal and microbiological quality of eggs submitted to different types of shell treatments. One hundred and forty-four fresh red eggs were distributed in a scheme of 4 treatments (no washing; washing and immersion in chlorine; washing and immersion in peracetic acid; and washing and spraying of propolis extract) x 5 storage periods (7, 14, 21, 28, and 35 days), stored at 25°C, in each period 6 eggs per treatment were analyzed. The parameters to assess were: weight loss; shell weight; yolk weight; albumen weight, yolk diameter; yolk height; albumen height and Haugh unit (HU). The microbiological quality of eggs was evaluated at 35 days through analysis for aerobic mesophilic bacteria, thermotolerant coliforms, Staphylococcus spp., Salmonella spp. and molds and yeasts. The treatment with propolis extract was the only effective one to maintain the high HU quality of the eggs until 21 days of storage at 25°C and was effective against microbiological contamination of all bacterial groups. The results presented showed greater effectiveness of the propolis extract for maintenance of internal and microbiological quality of eggs, it can be an alternative product to chemical sanitizers.(AU)

Salmonellosis is a disease that worries poultry farmers and can seriously impact the food safety of the population that consumes products of animal origin. The present study sought to detect the presence of Salmonella spp. in eggs intended for consumption from family poultry farms in municipalities of Alagoas State. The study was carried out from eight farms, where the sample from each farm was obtained by making pools of shell and internal contents of six randomly selected eggs. The pools were submitted to microbiological culture and the colonies were characterized and evaluated by means of laboratory tests. To this end, 50% (4/8) of the shell samples and 75% (6/8) of the internal contents of the eggs were positive for Salmonella spp. In addition, Klebsiella pneumoniae 12.5% (1/8) and Proteus spp. 25% (2/8) were found in the shell samples, and Yersinia spp. 12.5% (1/8) in the internal contents of the eggs. Salmonella spp. and other enterobacteria were confirmed to occur in eggs intended for consumption. The way the birds were raised did not seem to have a significant influence on the results obtained, and the presence of passerines on the farms may have contributed to the existence of bacteria there. Being aware of the risk to public health that some of these bacteria can present, it is necessary to take decisions that support the small producer in search of food safety for all.(AU)


A Salmonelose é uma doença que preocupa os criadores de aves e pode impactar seriamente a segurança alimentar da população que consome produtos de origem animal. O presente estudo buscou detectar a presença de Salmonella spp. em ovos destinados ao consumo provindos de aviculturas familiares de municípios do Estado de Alagoas. O estudo foi realizado a partir de oito granjas, onde a amostra de cada uma delas foi obtida pela confecção de pools de casca e de conteúdo interno de seis ovos selecionados aleatoriamente. Os pools foram submetidos a cultivo microbiológico e as colônias caracterizadas e avaliadas por meio de testes laboratoriais. Para tanto, 50% (4/8) das amostras de casca e 75% (6/8) das amostras de conteúdo interno dos ovos foram positivos para Salmonella spp. Além disso, também fora constatada a presença de Klebsiella pneumoniae12.5% (1/8) e Proteus spp. 25% (2/8) nas amostras de casca, e a presença de Yersinia spp. 12.5% (1/8) no conteúdo interno dos ovos. Foi confirmada a ocorrência de Salmonella spp. e outras enterobactérias em ovos destinados ao consumo. O modo de criação das aves não parece ter influenciado significativamente nos resultados obtidos e a presença de passeriformes nas granjas podem ter contribuído com a existência das bactérias no local. Tendo ciência do risco à saúde pública que algumas dessas bactérias podem apresentar, se faz necessária a tomada de decisões que apoiem o pequeno produtor em busca de uma segurança alimentar para todos.(AU)

This study aimed to evaluate the effectiveness of ultraviolet light in reducing bacterial load of eggshells and the impact of experimental disinfection on hatching, embryo mortality, and time-borne distribution using broiler breeder hens of different ages (38, 42, and 48 weeks old). Fertile eggs were subjected to different exposure periods (5, 7, and 9 minutes) of UV light (UV-C) with a 254 nm wavelength. For controls, eggs disinfected with paraformaldehyde (5.3 g/m3) and eggs not disinfected (NC). After subjection to disinfection protocols, the eggs were placed into sterile plastic bags containing 20 mL of peptone saline solution (0.1% m:v) and massaged for 1 minute to release the bacterial load. Aliquots of this solution were incubated in specific medium for bacterial growth for 48 hours at 37ºC for subsequent CFU counts. To evaluate the effects of disinfection on production, eggs previously disinfected by UV-C (9 min) and paraformaldehyde and NC eggs were candled between incubation days 10 and 13 and at the end of the incubation period to assess embryonic mortality. Hatchability distribution was performed every 8 hours. The 9 minutes 254nm UV-C light exposure was able to disinfect viable eggs and matched the effectiveness of the paraformaldehyde technique.(AU)


Objetivou-se avaliar a eficácia da luz ultravioleta na redução da carga bacteriana de cascas de ovos e o impacto na eclosão e na mortalidade embrionária observando-se a idade das matrizes (38, 42 e 48 semanas). Os ovos foram submetidos a diferentes períodos de exposição (cinco, sete e nove minutos) à luz UV (UV-C) com comprimento de onda de 254nm. Os controles foram ovos desinfetados com paraformaldeído (5,3g/m³) e ovos não desinfetados (NC). Após a desinfecção, os ovos foram colocados em sacos plásticos estéreis contendo 20mL de solução salina peptonada (0,1% m:v) e massageados por um (1) minuto para descolamento das bactérias. Alíquotas dessa solução foram incubadas em meio para crescimento bacteriano por 48 horas a 37ºC e contagem de UFC. Para avaliar os efeitos da desinfecção, ovos previamente desinfetados por UV-C (nove minutos) e ovos com paraformaldeído e NC foram submetidos à ovoscopia entre os dias 10 e 13 de incubação e ao final do período de incubação, para avaliação da mortalidade embrionária. A distribuição da eclodibilidade foi realizada a cada oito horas. A exposição à luz UV-C de 25nm de nove minutos desinfetou os ovos férteis e coincidiu com a eficácia do paraformaldeído.(AU)

Background: Food contamination is an important and growing public health concern due to the risk of foodborne illnesses.In this context, the egg, consumed all over the world, stands out. This food has been pointed out as a carrier of severalbacteria, causing outbreaks of food toxinfection. The production of free-range chicken (Gallus gallus domesticus) eggs hasbeen an alternative for generating income for producers in Brazil; however, there is no monitoring of the sanitary qualityof this product. The objective of this study was to identify microorganisms in 128 free-range chicken eggs sold in openmarkets in the semiarid region of Northeastern Brazil.Materials, Methods & Results: The study was carried out at commercial fairs in the state of Rio Grande do Norte, Braziliansemiarid region. The minimum number of eggs to be used was determined by the formula for simple random samples considering the following parameters: expected frequency of positive eggs of 50% (for sample maximization), 95% confidencelevel and sampling error of 10%. The minimum number of eggs to be sampled was 97; however, 128 eggs were obtained inthe period from August 2018 to April 2019. The internal content of the eggs was subjected to bacteriological culture usingan adapted methodology, and for the in vitro susceptibility assessment the disk diffusion method on Müller-Hintonagarwasused. In addition to biochemical tests, Gram-negative bacteria were subjected to microbiological diagnosis using theMALDI-TOF (Matrix Associated Laser Desorption-Ionization - Time of Flight) technique. Bacterial growth was found in40 (31.3%) eggs. The microorganisms most frequently isolated were Staphylococcus spp. (27.5%), Bacillus spp. (15%) andEnterobacter cloacae (25%). It can be used for a wide variety of products, including poultry, eggs, pet foods, and environmental samples, and results are obtained within about 24 h. In this chapter, all steps of the 3M MDS™ method for detection of Salmonella are described and detailed.

Salmonella spp. remains the most significant foodborne pathogen in south Brazil, but its epidemiology tends to change over time. Using official and surrogate data, a microbial subtyping model attributed different Salmonella serovars to laying hens, pigs, broilers, and turkeys from 2005 to 2015 in Rio Grande do Sul (RS). Additional to the subtyping model, three sub-analyses of outbreak data attributed Salmonella spp. in humans to animal and non-animal food. Laying hens/eggs was the most important source of human salmonellosis in RS, with almost 40% (159 cases; 95% credibility interval, 43-247) attribution proportion, followed by pigs reared in Santa Catarina, a neighbor state (34.5%). The Salmonella serovars Enteritidis and Typhimurium were the most common serovars involved. Source-related parameters had wide credibility intervals but showed a higher risk of illness from contaminated eggs than from the other three animal-food sources. Analysis of the outbreak data corroborated the findings and indicated signs of decreasing importance for eggs and increasing importance for pork consumption.

A total of 240 white Shaver laying hens from 22 to 34 weeks of age were assigned to 3 treatments and 5 replications. The treatments included:i) Conventional (hens were kept in experimental building without access to outdoor area and fed with the conventional diet), ii) Semi-organic (hens were kept in experimental building with access to outdoor area and fed with the organic diet plus amino acids and vitamin-minerals supplement), and iii) Organic (hens were kept in experimental building with access to outdoor and fed with the organic diet). The results showed statistically significant differences in the feed intake, egg production, egg mass, egg weight, and change body weight(g) means among the treatments (p <0.05). The hens kept in the organic treatment had fecal highest contain of oocytes coccidia (p <0.05). The yolk color index and shell strength in the organic treatment significantly increased in comparison with that of other treatments (p <0.05). The highest HDL was in the semi-organic and organic treatments (p <0.05). The lowest egg yolk cholesterol concentration was found in hens kept in the semi-organic and organic treatments (p <0.05). It is concluded that organic production system is useful for improving egg quality.

Antimicrobial resistance is a worldwide public health threat. Farm animals are important sources of bacteria containing antimicrobial resistance genes (ARGs). Although the use of antimicrobials in aquaculture and livestock has been reduced in several countries, these compounds are still routinely applied in animal production, and contribute to ARGs emergence and spread among bacteria. ARGs are transmitted to humans mainly through the consumption of products of animal origin (PAO). Bacteria can present intrinsic resistance, and once antimicrobials are administered, this resistance may be selected and multiply. The exchange of genetic material is another mechanism used by bacteria to acquire resistance. Some of the main ARGs found in bacteria present in PAO are the bla, mcr-1, cfr and tet genes, which are directly associated to antibiotic resistance in the human clinic.

Curcumin is an herbal component with several biological properties, and we highlight here the thermal stability, antioxidant and anti-inflammatory activity. We determined whether curcumin supplementation in the diets of laying hens under cold stress and naturally infected with Escherichia coli would control infection, and would have positive effects on overall health, as well as egg production and quality. We allocated 36 chickens that had been naturally infected with E. coli into two groups, with six replicates and three chickens per repetition: control group and curcumin group (200 mg curcumin/kg). The experimental period lasted 42 days, and fecal, blood and egg samples were collected at 1, 21 and 42 days We found that feces and eggs had lower total bacterial counts, E. coli counts and total coliform counts in the curcumin group at 21 and 42 days. In fresh eggs, the brightness and yellow intensity (b+) were significantly higher in the curcumin group. In stored eggs, higher specific gravity, albumen height and lower yolk pH were observed in the curcumin group. Fresh eggs collected and stored on day 42 showed lower levels of lipid peroxidation in the curcumin group, while the total antioxidant capacity in the stored eggs was significantly higher in the curcumin group. The curcumin group showed lower total leukocyte counts as a result of lower numbers of neutrophils and lymphocytes, as well as lower levels of total protein, alkaline phosphatase and alanine aminotransferase. Lower serum lipoperoxidation at 42 days was observed in the curcumin group, probably because of the higher activity of glutathione peroxidase and glutathione transferase in other words, because of antioxidant stimulation. Taken together, our findings suggest that curcumin supplementation in laying hens under cold stress and with colibacillosis has positive effects on infection control because of antioxidant stimulation.

Neutral Electrolyzed Water (NEW) was tested in vitro and on artificially contaminated eggs against Salmonella enterica subsp. enterica or Escherichia coli. The antibacterial effect was measured 30 s after treatment. NEW microbicide activity results were compared against 2% citric acid and 0.9% saline solutions. NEW caused an in vitro decrease in Salmonella titers by ˃5.56 Log10 CFU mL-1 and in artificially contaminated eggs by ˃1.45 Log10 CFU/egg. When it was tested against E. coli, it decreased in vitro bacterial titers by ˃3.28 Log10 CFU mL-1 and on artificially contaminated eggs by ˃6.39 Log10 CFU/egg. The 2% citric acid solution caused an in vitro decrease of 0.4 Log10 CFU mL-1 of Salmonella and E. coli and on eggs artificially contaminated with E. coli or Salmonella there was a decrease of 0.06 and 0.62 Log10 CFU/egg respectively. We evaluated egg cuticle integrity by scanning electron microscopy after treatments with evaluated solutions; the 2% citric acid solution caused damage to the cuticle and exposed eggshell pores and no interaction of NEW or NaCl with the cuticle was observed. NEW treatment showed a fast-bactericidal effect in vitro and table eggs.

The aim of this study was to develop a chitosan biofilm against Salmonella enteritidis, for the conservation of fertile and table eggs. Two experiments were performed. Experiment 1: 400 specific pathogen-free table eggs were divided in a completely randomized design into four treatments, five replicates and each replicate with 20 table eggs. Experimental groups were assigned to control and 1, 5 and 10% chitosan treatment. The eggs were immersed in the chitosan solution. They were then exposed to Salmonella enteritidis and stored for 1, 24, 96 and 168h at 4ºC. The eggs were then washed with 10mL of physiological saline solution. Experiment 2: 80 specific pathogen-free fertile eggs were tested, the assays were assigned to control and 1, 5 and 10% chitosan treatment. Each treatment had 20 fertile eggs. The eggs were immersed in the chitosan solution. They were individually weighed and incubated. Egg weight, humidity loss, and hatchability (weight and length of newly hatched chicks) characteristics were assessed. In Experiment 1, comparison between treatments showed differences (P< 0.05) in the total recovered of Salmonella enteritidis on eggshell, with the lower values in 5 y 10% chitosan treatment at 96 y 168h respectively. In Experiment 2, chitosan did not show any effect on the egg weight and chick weight, where the average was 57.44 and 38.23g respectively. The humidity loss and chick length showed differences (P< 0.05), with the lower values in 5 y 10% chitosan treatment. The antibacterial activity of chitosan biofilm provide a practical tool against Salmonella enteritidis in fertile and table eggs because the chitosan did not affect egg weight and chick weight, relevant parameters in the poultry industry.(AU)


O presente estudo teve como objetivo desenvolver um biofilme de quitosana contra Salmonella enteritidis, para conservação de ovos férteis e de mesa. Dois experimentos foram realizados. Experimento 1: 400 ovos de mesa livres de patógenos especificados foram divididos em delineamento inteiramente casualizado em quatro tratamentos, cinco repetições e cada réplica contendo 20 ovos de mesa. Grupos experimentais foram designados para controle e 1, 5 e 10% de tratamento com quitosana. Os ovos foram imersos em solução de quitosana. Em seguida foram expostos a Salmonella enteritidis, e armazenados por 1, 24, 96 e 168h a 4ºC. Após, os ovos foram lavados com 10mL de solução salina fisiológica. Experimento 2: 80 ovos férteis livres de patógenos especificados foram testados. Os ensaios foram atribuídos a controle e 1, 5 e 10% de tratamento com quitosana. Cada tratamento teve 20 ovos férteis. Os ovos foram imersos em solução de quitosana. Em seguida foram individualmente pesados e incubados. Peso dos ovos, perda de umidade e características de eclodibilidade (peso e comprimento dos pintinhos recém-nascidos) foram avaliados. The aim of this study was to develop a chitosan biofilm against Salmonella enteritidis, for the conservation of fertile and table eggs. Two experiments were performed. Experiment 1: 400 specific pathogen-free table eggs were divided in a completely randomized design into four treatments, five replicates and each replicate with 20 table eggs. Experimental groups were assigned to control and 1, 5 and 10% chitosan treatment. The eggs were immersed in the chitosan solution. They were then exposed to Salmonella enteritidis and stored for 1, 24, 96 and 168h at 4ºC. The eggs were then washed with 10mL of physiological saline solution. Experiment 2: 80 specific pathogen-free fertile eggs were tested, the assays were assigned to control and 1, 5 and 10% chitosan treatment. Each treatment had 20 fertile eggs. The eggs were immersed in the chitosan solution. They were individually weighed and incubated. Egg weight, humidity loss, and hatchability (weight and length of newly hatched chicks) characteristics were assessed. In Experiment 1, comparison between treatments showed differences (P< 0.05) in the total recovered of Salmonella enteritidis on eggshell, with the lower values in 5 y 10% chitosan treatment at 96 y 168h respectively. In Experiment 2, chitosan did not show any effect on the egg weight and chick weight, where the average was 57.44 and 38.23g respectively. The humidity loss and chick length showed differences (P< 0.05), with the lower values in 5 y 10% chitosan treatment. The antibacterial activity of chitosan biofilm provide a practical tool against Salmonella enteritidis in fertile and table eggs because the chitosan did not affect egg weight and chick weight, relevant parameters in the poultry industry.(AU)


O presente estudo teve como objetivo desenvolver um biofilme de quitosana contra Salmonella enteritidis, para conservação de ovos férteis e de mesa. Dois experimentos foram realizados. Experimento 1: 400 ovos de mesa livres de patógenos especificados foram divididos em delineamento inteiramente casualizado em quatro tratamentos, cinco repetições e cada réplica contendo 20 ovos de mesa. Grupos experimentais foram designados para controle e 1, 5 e 10% de tratamento com quitosana. Os ovos foram imersos em solução de quitosana. Em seguida foram expostos a Salmonella enteritidis, e armazenados por 1, 24, 96 e 168h a 4ºC. Após, os ovos foram lavados com 10mL de solução salina fisiológica. Experimento 2: 80 ovos férteis livres de patógenos especificados foram testados. Os ensaios foram atribuídos a controle e 1, 5 e 10% de tratamento com quitosana. Cada tratamento teve 20 ovos férteis. Os ovos foram imersos em solução de quitosana. Em seguida foram individualmente pesados e incubados. Peso dos ovos, perda de umidade e características de eclodibilidade (peso e comprimento dos pintinhos recém-nascidos) foram avaliados. Two experiments were conducted to evaluate the effects of -mannanase on egg production performance, egg quality, intestinal microbiota, viscosity, and ammonia concentration in laying hens. In Exp. 1, two hundred and seventy 30-wk-old Hy-Line Brown laying hens were assigned to 6 diets arranged in a 3 × 2 factorial of three levels of MEn and CP [(a corn-soybean meal based diet (HEHP), a diet containing 50 kcal of MEn/kg and 1.0% less energy and CP than the HEHP (MEMP), and a diet containing 80 kcal of MEn/kg and 1.5% less energy and CP than HEHP (LELP)], and -mannanase supplementation (0 or 0.04%). In Exp. 2, A total of two hundred and sixteen62-wk-old Hy-Line Brown laying hens were assigned to 6 dietary treatments in a 3 × 2 factorial arrangement. In Exp. 1, -mannanase supplementation increased egg production rate in hens fed LELP diet, but not in those fed HEHP or MEMP diet (interaction, p 0.01), and the interaction was significant (p 0.01) for egg mass. -mannanase supplementation decreased (p 0.05) ammonia concentration. In Exp. 2, the supplementation of -mannanase increased egg production rate and egg mass in hens fed LELP diet, whereas no differences were found in those fed HEHP or MELP diet (interaction, p 0.01). The supplementation of -mannanase in a lower energy and protein diet resulted in similar production performance when compared to high-energy and high-protein diet during early and late stages of egg production.

Egg quality has been widely studied, mainly because defects in quality can pose risks to public health, as well as economic losses. Nevertheless, studies about fungi in eggs are scarce. The objective was to compare the fungal microbiota from washed and unwashed eggs in the rainy season and dry season of the year. This exploratory research consisted in the analysis of large size white table eggs acquired from 48 different lots. Two manufacturers were sampled considering the main characteristic of washed or unwashed eggs. From each lot, a 30-egg pack were purchased and six of those eggs were used for mycological analyzes. The eggs were analyzed externally with 0.1% peptone salt solution wash of the eggshells and internally with aliquots being sampled from a pool made from the six eggs content. Samples were inoculated in Potato Dextrose Agar and isolated colonies were passed to test tubes. When sporulated, the isolates were subjected to decimal dilutions using 0.1% Tween 80 to dissociate the conidia. Microcultures were carried out for optical microscopy observation of the reproductive structures of fungi, stained with lactophenol. Aspergillus spp. was the most frequently isolated fungi isolated, with A. niger and A. flavus predominant in the dry season, while A. fumigatus and A. terreus in the rainy season. Entretanto, estudos sobre fungos em ovos são escassos. O objetivo foi comparar a microbiota fúngica de ovos lavados e ovos não lavados nas estações chuvosa e seca do ano. As amostras consistiram de ovos comerciais, do tipo branco e tamanho grande, com caráter exploratório pela análise de 48 lotes. Dois fabricantes foram amostrados considerando a característica principal de ovos lavados ou não lavados. De cada lote, foram adquiridos 30 ovos, sendo seis ovos utilizados nas análises micológicas. Os ovos foram analisados externamente fazendo lavagem com solução salina peptonada e internamentecom alíquotas retiradas de pools de seis ovos. Amostras foram inoculadas em Ágar Dextrose Batata e os isolados foram passados para tubos de ensaio. Posteriormente as colônias foram replicadas para tubos de ensaio contendo meio Ágar Dextrose Batata. Quandojá esporulados, os isolados foram submetidos a diluições utilizando Tween 80 0,1% para desagregar os conídios. Foram realizados microcultivos para observação em microscópio óptico das estruturas de reprodução dos fungos, corados com lactofenol. Aspergillusspp. foi a espécie mais frequentemente isolada, com predominância de A. niger e A. flavus na estação seca e A. fumigatuseA. terreusna chuvosa. O experimento foi realizado no setor de avicultura/UFRRJ, utilizando 348 galinhas semipesadas (linhagem Dekalb Brown), com 52 semanas de idade, criadas sob dois sistemas de produção: cage-free e em gaiolas. Os dados obtidos pela análise físico-química e microbiológica dos ovos e a resistência óssea à quebra foram submetidos à análise de variância. No caso de ocorrerem efeitos dos diferentes sistemas de produção, foi aplicado o teste de Tukey a 5% de probabilidade para comparação das médias. A qualidade físico-química foi igualmente favorecida pelos dois sistemas de produção, indicando que as circunstâncias experimentais propiciaram condições adequadas para a formação de ovos de boa qualidade. O sistema de gaiola não desfavoreceu as características ósseas das galinhas, apontando que, em densidades adequadas, a gaiola pode não exercer um fator prejudicial para a qualidade óssea. O sistema de produção cage-free piorou a contaminação da casca, comprovando que ovos postos em ninhos são mais contaminados em comparação aos produzidos em gaiolas.(AU)

The data obtained by the physical-chemical and microbiological analysis of the eggs and the bone resistance to the break were submitted to analysis of variance, in case of effects of the different production systems, the Tukey's test was applied at 5% of probability for comparison of the means. The physical-chemical quality was also favored by the two production systems, indicating that the experimental circumstances provided adequate conditions for the formation of good quality eggs. The cage system did not disfavor the bony characteristics of the hens, indicating that at suitable densities, the cage may not exert a detrimental factor to bone quality. The cage-free production system worsened shell contamination by proving that nesting eggs are more contaminated compared to those produced in cages.(AU)
